1. Lagoon hydrodynamics of pearl farming islands: the case of Gambier (French Polynesia)

Abstract

Between 2019 and 2020, the Gambier lagoon was instrumented over a period of 9 months with a large array of autonomous oceanographic instruments measuring temperature, pressure, and current. Two deployments were conducted from June to October 2019 (Leg1) and from late October 2019 to late February 2020 (Leg2). A total of 16 instrumented locations were spread across the lagoon and on the forereef. Physical parameters were measured to characterize the wave climate, tide and surges, lagoonal circulation, and spatial and vertical temperature variabilities. Those observations were part of the ANR-funded MANA project (2017–2022) and its derivatives that were aimed at improving knowledge of processes influencing the spat collection of Pinctada margaritifera oysters that are used for black pearl farming and production. This dataset was a prerequisite for the development of a high-resolution biophysical model on the Gambier lagoon that is aimed at understanding the connectivity of oyster larvae (Bruyère et al., 2023a). The sampling strategy focused on the northern region of the Gambier lagoon and especially on the sub-lagoon of Rikitea, which is a prime spat collection site. The dataset was post processed, quality controlled, and is archived in a dedicated repository with a permanent DOI via the SEANOE marine data platform (https://doi.org/10.17882/94148, Andréfouët et al., 2023b).

Abstract

Between 2018 and 2022, four pearl farming Tuamotu atolls of French Polynesia were monitored with autonomous oceanographic instruments to measure the hydrodynamics of atoll lagoons and the ocean–lagoon water exchanges. These surveys were conducted within the framework of the ANR-funded MANA (Management of Atolls) project and its extensions to additional sites. The overarching goal was to improve knowledge on the processes influencing the spat collection of the pearl oyster Pinctada margaritifera, the oyster species used to produce black pearls. These data sets are also critical for the calibration and validation of 3D high spatial resolution hydrodynamic models used to study oyster larval dispersal within lagoons. The observational strategies focused on the characterization of ocean–lagoon exchanges through passes and hoa (i.e., shallow reef flats), lagoon circulation, incident waves breaking on the forereef, water elevation inside the lagoon as well as spatial temperature variability. Chronologically, the investigated atolls were first Raroia Atoll with 9 months of measurements between May 2018 and March 2019 during which the MALIS1 and MALIS2 cruises on-board the R/V ALIS took place. It was followed by a 4 month deployment in Takapoto Atoll (November 2021 to March 2022). In late April 2022, Apataki Atoll was instrumented until end of July, followed by Takaroa measurements between July and October 2022. Apataki (Leg2) and Takaroa Atoll were conjointly instrumented during the MALIS 3 oceanographic cruise. Altogether, those multi-atoll data bring a worldwide unique oceanographic atoll data set that is useful for addressing local pearl farming questions but potentially beneficial for other fundamental and applied investigations. Each data set was post-processed, quality controlled and converted into NetCDF format. Files are available in open source into dedicated repositories in the SEANOE marine data platform. Abstract

Scaffolds are models designed to aid the interaction between cells and extracellular bone matrix, providing structural support for newly formed bone tissue. In this work, wollastonite with β‐TCP porous ceramic scaffolds was developed by the polymer sponge replication. Their microstructure, cell viability and bioactivity were tested. in vivo was performed to evaluate the use of a calcium silicate‐based implant in the repair of rabbit tibias. Holes were made in the both proximal and distal tibial metaphysis of each animal and filled with calcium silicate‐based implant, and in the left tibia, no implant were used, serving as control group. Animals underwent euthanasia after 30 and 60 days of study. The animals were submitted to clinical‐radiographic evaluations and their histology was analyzed by optical and scanning electron microscope. The studied calcium silicate implant provided biocompatibility and promoted bone formation, stimulating the process of bone repair in rabbits, features observed by gradual radiopacity shown in the radiographic evaluations. yes

Abstract

Colorectal cancer is the second leading cause of cancer-related deaths in the United States and accounts for an estimated 1 million deaths annually worldwide. The liver is the most common site of metastatic spread from colorectal cancer, significantly driving both morbidity and mortality. Although remarkable advances have been made in recent years in the management for patients with colorectal cancer liver metastases, significant challenges remain in early detection, prevention of progression and recurrence, and in the development of more effective therapeutics. In 2017, our group held a multidisciplinary state-of-the-science symposium to discuss the rapidly evolving clinical and scientific advances in the field of colorectal liver metastases, including novel early detection and prognostic liquid biomarkers, identification of high-risk cohorts, advances in tumor-immune therapy, and different regional and systemic therapeutic strategies. Since that time, there have been scientific discoveries translating into therapeutic innovations addressing the current management challenges. These innovations are currently reshaping the treatment paradigms and spurring further scientific discovery. Herein, we present an updated discussion of both the scientific and clinical advances and future directions in the management of colorectal liver metastases, including adoptive T-cell therapies, novel blood-based biomarkers, and the role of the tumor microbiome. In addition, we provide a comprehensive overview detailing the role of modern multidisciplinary clinical approaches used in the management of patients with colorectal liver metastases, including considerations toward specific molecular tumor profiles identified on next generation sequencing, as well as quality of life implications for these innovative treatments.

Abstract

The authors present a measurement of the left-right cross-section asymmetry (ALR) for Z boson production by e{sup +}e{sup {minus}} collisions. The measurement includes the final data taken with the SLD detector at the SLAC Linear Collider (SLC) during the period 1996--1998. Using a sample of 383,487 Z decays collected during the 1996-1998 runs they measure the pole-value of the asymmetry, A{sub LR}{sup 0}, to be 0.15056 {+-} 0.00239 which is equivalent to an effective weak mixing angle of sin{sup 2}Sigma{sub W}{sup eff} = 0.23107 {+-} 0.00030. The result for the complete 1992--1998 dataset comprising 537 thousand Z decays is sin{sup 2}Sigma{sub W}{sup eff} = 0.23097 {+-} 0.00027.

Abstract

A hidroxiapatita [Ca10(PO4)6(OH)2, HA] foi sintetizada utilizando-se a rota sol-gel partindo-se de ácido fosfórico e nitrato de cálcio como precursores de cálcio e fósforo, respectivamente e como solvente utilizou-se o metanol na preparação do sol que posteriormente será utilizado na obtenção de recobrimentos de hidroxiapatita sobre substratos de ligas de titânio. O sol permaneceu estável e não ocorreu gelatinização em temperatura ambiente durante sete dias. O sol transformou-se em um gel branco somente após a remoção do solvente a 100ºC. O produto assim obtido foi calcinado em 300°C, 500°C e 700°C e caracterizou-se por DRX, FT-IR, MEV/EDS e TGA/DSC. As fases de HA sintetizada tornaram-se estáveis sem sub-produtos a 700°C. A difração de raios X mostrou que a estrutura apatita é aparente em 300°C. O tamanho do cristal e o teor de HA aumentaram com o aumento da temperatura de calcinação. A análise por MEV mostrou a presença de poros que são importantes para aplicações biomédicas, favorecendo a adesão entre o tecido ósseo neoformado e a apatita sintética, ou seja, osseointegração.Hydroxyapatite [Ca10(PO4)6(OH)2, HA] was synthesized using sol-gel route were used phosphoric acid and calcium nitrate as calcium and phosphorous precursors, respectively and as solvent was using the methanol in the preparation of the sol that later will be used in the obtention apatites coatings on titanium alloys substrates. The sol remained stable and no gelling occurred in ambient environment for over 7 days. The sol became a white gel only after the removal of the solvent at 100ºC. The product was calcined in 300°C, 500°C and 700°C and had been characterized by DRX, FT-IR, SEM/EDS and TGA/DSC. The synthesized HA became stable 700°C without any another product. X ray diffraction showed that structure apatite initially appeared at 300°C. The crystal size and the HA content increase with increasing calcination temperature. SEM analysis indicated the presence of pores that are important for biomedical applications, improving to the adhesion between neoformed bone tissue and synthesized apatite, i.e, osseointegration.

7. Lower thyroid stimulating hormone concentrations linked to suicidal ideations among individuals with anxiety and mood disorders

Abstract

Introduction Suicidal behavior is quite common in individuals with anxiety and mood disorders (AMD). One of the coexistence factors in suicidal behavior is thyroid dysfunction, however the results are still controversial (Shen et al. J Affect Disord 2019;(1)180-185; Zhou et al., Transl Psychiatry. 2021;11(1):97). The role of thyroid function in suicidal ideation among individuals with comorbid AMD have not been thoroughly investigated. Objectives The aim of this cross-sectional study was to identify potential associations between thyroid function and suicidal ideation in a sample of individuals with anxiety and mood disorders. Methods This exploratory study comprised 77 consecutive individuals with AMD (age range 18-73 years, 76% were females) attending the Psychiatry Day care unit. All individuals have been evaluated for current psychiatric diagnoses, suicidal ideation using the Mini International Neuropsychiatric Interview [M.I.N.I. 7.0.2]) as well as for socio-demographic factors and for current psychotropic medication use. Severity of depression and anxiety symptoms have been evaluated using the Patient Health Questionnaire-9 (PHQ-9) and the General Anxiety Disorder-7 (GAD-7). The biochemical blood tests were performed for the concentrations of thyroxine (FT4), triiodothyronine (FT3) and thyroid stimulating hormone (TSH). The univariate and multivariable logistic regression analyses were used to assess the association between biochemical parameters and suicidal ideation. Results Of all study individuals with AMD – 56% have been identified as having current SI. There were not significant differences according to age, gender, education, BMI, smoking, depression and anxiety symptoms and current psychotropics use between SI and non-SI individuals. Serum FT4, FT3 and TSH concentrations were within normal range. However individuals with SI had significantly lower TSH concentrations in comparison to the non-SI (1.54 (0.77) vs. 2.04 (1.22) IU/L, respectively; p = 0.049), without significantly differences in FT4 and FT3 concentrations. A multiple logistic regression, adjusting for sociodemographic factors and severity of mental symptoms revealed, that non-SI individuals with AMD were likely to have higher TSH levels than SI (odds ratio = 2.15 (95% CI 1.10–4.22; p = 0.027). Conclusions Among individuals with AMD, lower levels of TSH concentrations have been associated with presence of suicidal ideation, independently of sociodemographic factors and severity of depression and anxiety. Abstract

We report a new measurement of Rb = Gamma(Z0->b-bbar) / Gamma(Z0->hadrons) using a double tag technique, where the b hemisphere selection is based on the reconstructed mass of the B hadron decay vertex. The measurement was performed using a sample of 130k hadronic Z0 events, collected with the SLD at the SLC. The method utilizes the 3-D vertexing abilities of the CCD pixel vertex detector and the small stable SLC beams to obtain a high b-tagging efficiency and purity. We obtain Rb=0.2142+/-0.0034(stat.)+/-0.0015(syst.)+/-0.0002(R_c)., 11 pages including 3 eps files

Abstract

We have measured the distributions of the jet energies in {ital e{sup +}e{sup {minus}}{r_arrow}q{bar q}g} events, and of the three orientation angles of the event plane, using hadronic {ital Z{sup 0}} decays collected in the SLD experiment at SLAC. We find that the data are well described by perturbative QCD incorporating vector gluons. Abstract

Abstract Metastatic progression defines the final stages of tumor evolution and underlies the majority of cancer-related deaths. The heterogeneity in disseminated tumor cell populations capable of seeding and growing in distant organ sites contributes to the development of treatment resistant disease. We recently reported the identification of a novel tumor-derived cell population, circulating hybrid cells (CHCs), harboring attributes from both macrophages and neoplastic cells, including functional characteristics important to metastatic spread. These disseminated hybrids outnumber conventionally defined circulating tumor cells (CTCs) in cancer patients. It is unknown if CHCs represent a generalized cancer mechanism for cell dissemination, or if this population is relevant to the metastatic cascade. Herein, we detect CHCs in the peripheral blood of patients with cancer in myriad disease sites encompassing epithelial and non-epithelial malignancies. Further, we demonstrate that in vivo-derived hybrid cells harbor tumor-initiating capacity in murine cancer models and that CHCs from human breast cancer patients express stem cell antigens, features consistent with the potential to seed and grow at metastatic sites. Finally, we reveal heterogeneity of CHC phenotypes reflect key tumor features, including oncogenic mutations and functional protein expression. Importantly, this novel population of disseminated neoplastic cells opens a new area in cancer biology and renewed opportunity for battling metastatic disease.

Abstract

We measure the hadronic branching ratios of the Z0 boson into heavy quarks: Rb=Gamma(Z0->bb)/Gamma(Z0->hadrons) and Rc=Gamma(Z0->cc/Gamma(Z0->hadrons) using a multi-tag technique. The measurement was performed using about 400,000 hadronic Z0 events recorded in the SLD experiment at SLAC between 1996 and 1998. The small and stable SLC beam spot and the CCD-based vertex detector were used to reconstruct bottom and charm hadron decay vertices with high efficiency and purity, which enables us to measure most efficiencies from data. We obtain, Rb=0.21604 +- 0.00098(stat.) +- 0.00073(syst.) -+ 0.00012(Rc) and, Rc= 0.1744 +- 0.0031(stat.) +- 0.0020(syst.) -+ 0.0006(Rb), 37 pages, 8 figures, to be submitted to Phys. Rev. Abstract

The high burden of kidney disease, global disparities in kidney care, and the poor outcomes of kidney failure place a growing burden on affected individuals and their families, caregivers, and the community at large. Health literacy is the degree to which individuals and organizations have, or equitably enable individuals to have, the ability to find, understand, and use information and services to make informed health-related decisions and actions for themselves and others. Rather than viewing health literacy as a patient deficit, improving health literacy lies primarily with health care providers communicating and educating effectively in codesigned partnership with those with kidney disease. For kidney policy makers, health literacy is a prerequisite for organizations to transition to a culture that places the person at the center of health care. The growing capability of and access to technology provides new opportunities to enhance education and awareness of kidney disease for all stakeholders. Advances in telecommunication, including social media platforms, can be leveraged to enhance persons’ and providers’ education. The World Kidney Day declares 2022 as the year of “Kidney Health for All” to promote global teamwork in advancing strategies in bridging the gap in kidney health education and literacy. Kidney organizations should work toward shifting the patient-deficit health literacy narrative to that of being the responsibility of health care providers and health policy makers. By engaging in and supporting kidney health-centered policy making, community health planning, and health literacy approaches for all, the kidney communities strive to prevent kidney diseases and enable living well with kidney disease.

Abstract

We have measured gluon splitting into bottom quarks, g -> b bbar, in hadronic Z0 decays collected by SLD between 1996 and 1998. The analysis was performed by looking for secondary bottom production in 4-jet events of any primary flavor. 4-jet events were identified, and in each event a topological vertex-mass technique was applied to the two jets closest in angle in order to identify them as b or bbar jets. The upgraded CCD vertex detector gives very high B-tagging efficiency, especially for B hadrons with the low energies typical of this process. We measured the rate of g -> b bbar production per hadronic event, g_bb, to be (2.44+-0.59 (stat) +-0.34 (syst)) X10**-3., 21 pages, 11 figures; submitted to Physics Letters B

14. Are Surgical Guideline Changes in Diverticulitis Care Associated With Decreased Inpatient Healthcare Expenditure?

Abstract

Objective:. To examine possible associations in inpatient healthcare expenditure and guideline changes in the surgical management of diverticulitis, in terms of both cost per discharge and total aggregate costs of care. Background:. Medical costs throughout the healthcare system continue to rise due to increased prices for services, increased quantities of high-priced technologies, and an increase in the amount of overall services. Methods:. We used a retrospective case-control design using the Healthcare Cost and Utilization Project National Inpatient Sample to evaluate cost per discharge and total aggregate costs of diverticulitis management between 2004 and 2015. The year 2010 was selected as the transition between the pre and postguideline implementation period. Results:. The sample consisted of 450,122 unweighted (2,227,765 weighted) inpatient discharges for diverticulitis. Before the implementation period, inpatient costs per discharge increased 1.13% in 2015 dollars (95% confidence intervals [CI] 0.76% to 1.49%) per quarter. In the postimplementation period, the costs per discharge decreased 0.27% (95% CI –0.39% to –0.15%) per quarter. In aggregate, costs of care for diverticulitis increased 0.61% (95% CI 0.28% to 0.95%) per quarter prior to the guideline change, and decreased 0.52% (95% CI –0.87% to –0.17) following the guideline change. Conclusions:. This is the first study to investigate any associations between evidence-based guidelines meant to decrease surgical utilization and inpatient healthcare costs. Decreased inpatient costs of diverticulitis management may be associated with guideline changes to reduce surgical intervention for diverticulitis, both in regards to cost per discharge and aggregate costs of care.

Abstract

Introduction In our study, we aimed to understand how an individual perceives everyday life, as well as, which role the features of metacognitions and personality play in this process. Everyone is immersed in society and therefore exposed to external influences. Attention has already been focused on the relationship between metacognition and social context. “Internal orientation” depends largely on our personal and family history, socio-economic situation, group membership, and cultural context. Objectives The sample consisted of 30 participants (women and men, M=25,7, Sd=3,6), selected by the criterion of personal interest in this research. Methods The study consisted of several stages. The first stage was devoted to the theoretical analysis of everyday life in modern psychology. The data were processed using descriptive qualitative analysis using the phenomenological method, where we identified categories (Central topics) among the participants ‘ responses. To study the features of metacognition, we selected methods for assessing metacognitive involvement in awareness of internal activity, as well as metacognitive beliefs. The metacognitive awareness inventory questionnaire (MAIL) (Schraw & Dennison, 1994) in adaptation (Karpov & Skiteva, 2005) allows you to assess the level of metacognitive engagement, answer the question about the level of metacognitive awareness of the participant. Results Pearson correlation is revealed a significant relationship between MAI and cognitive self-consciousness (p=0.003), positive beliefs (p=0.002), and needs of controling the thoughts (p=0.076). Conclusions Our research opens the study of the subjective dimensions of person-situation-activity and offers a way of linking research on personality with research on the social processes whereby persons conduct their everyday lives.

Abstract

Introduction The problem of homosexuality is constantly in the spotlight of the mass media, social media and politicians. At the same time, the cultural and national specificity of attitudes towards the phenomenon of homosexuality seems obvious, as well as a significant polarization of opinions within Russian society itself. With significant attention to this issue, there are not many attempts to analyze the socio-psychological basis of representations about homosexuality. At the same time, in a number of foreign studies it was revealed that the modern Z Gen is distinguished by greater tolerance and freedom of views in terms of attitude towards traditionally segregated social groups. Objectives The purpose of this study was to identify representations about homosexuality among different generations of modern Russians. Methods The methodological basis of the research was the study of the structure of social representations (Vergesse methodique). The research methods implied the author’s questionnaire aimed at identifying representations about homosexuality and a modified version of the RAHI questionnaire. The sample was N = 444 (residents of Russia, age 16-65). Results There was shown a significant difference between the Z Gen in terms of tolerance of representations about homosexuality. So called ‘double standards’ were identified in terms of attitudes towards male and female homosexuality. The rooted concept of homosexuality as a relationship based, rather, on a sexual rather than a romantic-spiritual level of relationships, was stated. Conclusions Main hypothesis was confirmed: an inverse relationship between age and perceptions of homosexuality as normative was revealed. Disclosure No significant relationships.

Abstract

Introduction The article is devoted to the main methodological problems of qualitative research in the psychology of culture. Methods of ethnographic (field) observation were borrowed by psychology from social and cultural anthropologists in the first half of the twentieth century, and in modern research practices there is a special ethnographic direction that claims to analyze behavior and lifestyle in various subcultures and communities. Objectives The purpose of this study was to identify the key cross-cultural difference in coping behavior of respondents from Moscow and Tashkent. Methods The methodological basis of the research was the thematic analysis of narratives and free-form interviews. The sample was N = 60 (residents of Russia and Uzbekistan age 17-39). Results The results of thematic analysis of narratives and free-form interviews of respondents from Moscow and Tashkent allow us to conclude that the key cross-cultural difference in coping behavior is the degree of its individualization: representatives of Uzbek culture are focused on receiving support and care from significant Others, and not on independent internal work (unlike Russian respondents). At the same time, they are not satisfied with the traditional prescriptions that come from the family environment, which forces them to coping practices that go beyond the boundaries of normative social (often religious) ideas. Conclusions This can be interpreted from the point of view of the process of modernization of Uzbek culture, which is gradually becoming individualistic, and the latter circumstance requires the construction of flexible coping strategies in the situation of social and cultural changes. Abstract

An association study was conducted to investigate the relation between 14 variants of glucose transporter 1 gene (SLC2A1) and the risk of type 2 diabetes (T2DM) leading to nephropathy. We also performed a meta-analysis of 11 studies investigating association between diabetic nephropathy (DN) and SLC2A1 variants. The cohort included 197 cases (T2DM with nephropathy), 155 diseased controls (T2DM without nephropathy) and 246 healthy controls. The association of variants with disease progression was tested using generalized odds ratio (ORG). The risk of type 2 diabetes leading to nephropathy was estimated by the OR of additive and co-dominant models. The mode of inheritance was assessed using the degree of dominance index (h-index). We synthesized results of 11 studies examining association between 5 SLC2A1 variants and DN. ORG was used to assess the association between variants and DN using random effects models. Significant results were derived for co-dominant model of rs12407920 [OR = 2.01 (1.17–3.45)], rs841847 [OR = 1.73 (1.17–2.56)] and rs841853 [OR = 1.74 (1.18–2.55)] and for additive model of rs3729548 [OR = 0.52 (0.29–0.90)]. The mode of inheritance for rs12407920, rs841847 and rs841853 was ‘dominance of each minor allele’ and for rs3729548 ‘non-dominance’. Frequency of one haplotype (C-G-G-A-T-C-C-T-G-T-C-C-A-G) differed significantly between cases and healthy controls [p = .014]. Regarding meta-analysis, rs841853 contributed to an increased risk of DN [(ORG = 1.43 (1.09–1.88); ORG = 1.58 (1.01–2.48)] between diseased controls versus cases and healthy controls versus cases, respectively. Further studies confirm the association of rs12407920, rs841847, rs841853, as well as rs3729548 and the risk of T2DM leading to nephropathy.

Abstract Background Colorectal cancer (CRC) is a leading cause of cancer-related death. The biologic response of CRC to standard of care adjuvant therapies such as chemotherapy and radiation are poorly understood. MicroRNAs (miRs) have been shown to affect CRC progression and metastasis. Therefore, we hypothesized that specific miRs modulate CRC response to chemoradiation. Methods In this study, we used miR expression profiling and discovered a set of microRNAs upregulated rapidly in response to either a single 2 Gy dose fraction or a 10 Gy dose of γ-radiation in mouse colorectal carcinoma models. We used gain and loss-of-function studies in 2D and 3Dcell proliferation assays and colony formation assays to understand the role of the top miR candidate from our profiling. We used Student’s T-tests for simple comparisons and two-factor ANOVA for evaluating significance. Results The most upregulated candidate at early time points in our signature, miR-451a inhibited tumor cell proliferation and attenuated surviving fraction in longer-term cultures. Conversely, inhibition of miR-451a increased proliferation, tumorsphere formation, and surviving fraction of tumor cells. Using a bioinformatics approach, we identified four genes, CAB39, EMSY, MEX3C, and EREG, as targets of miR-451a. Transfection of miR-451a decreased both mRNA and protein levels of these targets. Importantly, we found miR-451a expression was high and CAB39, EMSY levels were low in a small subset of rectal cancer patients who had a partial response to chemoradiation when compared to patients that had no response. Finally, analysis of a TCGA colorectal cancer dataset revealed that CAB39 and EMSY are upregulated at the protein level in a significant number of CRC patients. Higher levels of CAB39 and EMSY correlated with poorer overall survival. Conclusions Taken together, our data indicates miR-451a is induced by radiation and may influence colorectal carcinoma proliferation via CAB39 and EMSY pathways.

Abstract

New modification of nanosphere lithography has been realized to obtain multilayered systems of ordered gold nanopartciles (NP). NP have been formed using vacuum deposition of 5…60-nm layer of gold on ionic etched multilayered regular coating consisted of several layers of 200-nm polystyrene spheres. Optical study shows that spectra of NP depend on their thickness and may be changed by heat treatment. Increasing the NP thickness within the 5…20-nm range leads to a shortwave displacement of the plasmon resonance peak position, while the longwave shift is observed in 20…60-nm range. Heat treatment of NP brings narrowing and displacement of spectral bands, rising the extinction. It has been supposed that variation of the NP shape is the most substantial factor for changes of optical properties in the 5…20 nm thickness region, while electromagnetic coupling between NP in different layers becomes more important for thicknesses larger than 40 nm. Optical properties inherent to the obtained system of NP can be tuned by changing the polystyrene spheres diameter, extent of etching, thickness of gold layer and with the heat treatment. It may be used in design of nanophotonic devices.

Abstract

The global burden of chronic kidney disease (CKD) is rapidly increasing with a projection of becoming the 5th most common cause of years of life lost globally by 2040. CKD is a major cause of catastrophic health expenditure. The costs of dialysis and transplantation consume up to 3% of the annual healthcare budget in high-income countries. However, the onset and progression of CKD is often preventable. In 2020, the World Kidney Day campaign highlights the importance of preventive interventions – be it primary, secondary, or tertiary. This article focuses on outlining and analyzing measures that can be implemented in every country to promote and advance CKD prevention. Primary prevention of kidney disease should focus on the modification of risk factors and addressing structural abnormalities of the kidney and urinary tracts, as well as exposure to environmental risk factors and nephrotoxins. In persons with pre-existing kidney disease, secondary prevention, including blood pressure optimization and glycemic control, should be the main goal of education and clinical interventions. In patients with advanced CKD, management of co-morbidities such as uremia and cardiovascular disease is a highly recommended preventative intervention to avoid or delay dialysis or kidney transplantation. Political efforts are needed to proliferate the preventive approach. While national policies and strategies for non-communicable diseases might be present in a country, specific policies directed toward education and awareness about CKD screening, management, and treatment are often lacking. Hence, there is an urgent need to increase the awareness of preventive measures throughout populations, professionals, and policy makers.

In patients with colorectal cancer (CRC) that metastasizes to the liver, there are several key goals for improving outcomes including early detection, effective prognostic indicators of treatment response, and accurate identification of patients at high risk for recurrence. Although new therapeutic regimens developed over the past decade have increased survival, there is substantial room for improvement in selecting targeted treatment regimens for the patients who will derive the most benefit. Recently, there have been exciting developments in identifying high-risk patient cohorts, refinements in the understanding of systemic vs localized drug delivery to metastatic niches, liquid biomarker development, and dramatic advances in tumor immune therapy, all of which promise new and innovative approaches to tackling the problem of detecting and treating the metastatic spread of CRC to the liver. Our multidisciplinary group held a state-of-the-science symposium this past year to review advances in this rapidly evolving field. Herein, we present a discussion around the issues facing treatment of patients with CRC liver metastases, including the relationship of discrete gene signatures with prognosis. We also discuss the latest advances to maximize regional and systemic therapies aimed at decreasing intrahepatic recurrence, review recent insights into the tumor microenvironment, and summarize advances in noninvasive multimodal biomarkers for early detection of primary and recurrent disease. As we continue to advance clinically and technologically in the field of colorectal tumor biology, our goal should be continued refinement of predictive and prognostic studies to decrease recurrence after curative resection and minimize treatment toxicity to patients through a tailored multidisciplinary approach to cancer care. Abstract

We have made the first measurement of the double-inclusive B/Bbar energy distribution in e+e- annihilations, using a sample of 400,000 hadronic Z0 decay events recorded in the SLD experiment at SLAC between 1996 and 1998. The small and stable SLC beam spot and the CCD-based vertex detector were used to reconstruct B/Bbar decay vertices with high efficiency and purity, and to provide measurements of the kinematic quantities used to calculate the B energies in this novel technique. We measured the B/Bbar energies with good efficiency and resolution over the full kinematic range. We measured moments of the scaled energies of the B and Bbar hadrons vs. the opening angle between them. By comparing these results with perturbative QCD predictions we tested the ansatz of factorisation in heavy-quark production. This article aims to study the non-Fickian water absorption process in vegetable fiber-reinforced polymer composite using the Langmuir-type model, evaluating the influence of mass diffusivity on the process. The numerical solutions of the governing equations were obtained using the finite-volume method. Transient results of the local and average moisture content, free and entrapped water molecules concentration considering the constant diffusivity and as a function of the average and local moisture content were presented and analyzed. It was observed that the mass diffusivity effectively influences the water absorption behavior, especially in the initial time of the process, where higher differences in the water migration rates into the material are found. Abstract

Seasonal dimorphism (summer/winter) has been so far studied only in a few plants and has been focused on summer drought stress. However, Thymus sibthorpii in the study area appears to be affected by winter chilling stress and not by summer drought stress. Thus, the winter leaves were thicker and more compact compared to the summer leaves and they had more stomata and peltate hairs, more sclerenchymatous fibers, vacuoles with phenolics, and chloroplasts than the summer leaves. In addition, their chloroplasts possessed large grana and starch grains. In the summer leaves, cell vacuoles in mesophyll did not contain phenolics, and chloroplasts were devoid of starch grains and had large plastoglobuli. Physiological measurements revealed higher net photosynthetic rate and chlorophyll content in the winter leaves than in the summer leaves. Proline and soluble sugar content along with antioxidative enzyme (superoxide dismutase, peroxidase, ascorbate peroxidase, glutathione reductase) activities were increased in the winter leaves.

Abstract

The evolution of large amplitude internal waves propagating towards the shore and more specifically the run up phase over the "swash" zone is considered. The mathematical model describing the generation, interaction, and decaying of solitary internal waves of the second mode in the interlayer is proposed. The exact solution specifying the shape of solitary waves symmetric with respect to the unperturbed interface is constructed. It is shown that, taking into account the friction on interfaces in the mathematical model, it is possible to describe adequately the change in the phase and amplitude characteristics of two solitary waves moving towards each other before and after their interaction. It is demonstrated that propagation of large amplitude solitary internal waves of depression over a shelf could be simulated in laboratory experiments by internal symmetric solitary waves of the second mode.

Abstract

Dynamics of large amplitude internal waves in two-layers of shallow water is considered. It is demonstrated that in laboratory experiments the subsurface waves of depression over a shelf may be simulated by internal symmetric solitary waves of the mode 2 ("lump-like" waves). The mathematical model describing the propagation and decaying of large internal waves in two-layer fluid is introduced. It is a variant of Choi-Camassa equations with hydrostatic pressure distribution in one of the layers. It is shown that the numerical scheme developed for the Green-Naghdi equations in open channel flows may be applied for the description of large amplitude internal waves over a shelf.

Abstract Sunflower Verticillium Wilt and Leaf Mottle (SVW), caused by Verticillium dahliae (Kleb.; Vd), is a soil-borne disease affecting sunflower worldwide. A single dominant locus, known as V1, was formerly effective in controlling North-American Vd races, whereas races from Argentina, Europe and an emerging race from USA overcome its resistance. This emphasizes the need for identifying broad-spectrum genetic resistance (BSR) sources. Here we characterize two sunflower mapping populations (MPs) for SVW resistance: a biparental MP and the association MP from the National Institute of Agricultural Technology (INTA), under field growing conditions. Nine field-trials (FTs) were conducted in highly infested fields in the most SVW-affected region of Argentina. Several disease descriptors (DDs), including incidence and severity, were scored across four phenological stages. Generalized linear models were fitted according to the nature of each variable, adjusting mean phenotypes for inbred lines across and within FTs. Comparison of these responses allowed the identification of novel BSR sources. Furthermore, we present the first report of SVW resistance heritability, with estimates ranging from 35 to 45% for DDs related to disease incidence and severity, respectively. This study constitutes the largest SVW resistance characterization reported to date in sunflower, identifying valuable genetic resources for BSR-breeding to cope with a pathogen of increasing importance worldwide.

Despite its worldwide occurrence, the genetic determinants of plant resistance are still largely unknown. Here, we investigated the Sclerotinia-sunflower pathosystem by analysing temporal changes in gene expression in one susceptible and two tolerant inbred lines (IL) inoculated with the pathogen under field conditions. Differential expression analysis showed little overlapping among ILs, suggesting genotype-specific control of cell defense responses possibly related to differences in disease resistance strategies. Functional enrichment assessments yielded a similar pattern. However, all three ILs altered the expression of genes involved in the cellular redox state and cell wall remodeling, in agreement with current knowledge about the initiation of plant immune responses. Remarkably, the over-representation of long non-coding RNAs (lncRNA) was another common feature among ILs. Our findings highlight the diversity of transcriptional responses to SHR within sunflower breeding lines and provide evidence of lncRNAs playing a significant role at early stages of defense.

Abstract

Abstract Background Sclerotinia sclerotiorum is a necrotrophic fungus that causes Sclerotinia head rot (SHR) in sunflower, with epidemics leading to severe yield losses. In this work, we present an association mapping (AM) approach to investigate the genetic basis of natural resistance to SHR in cultivated sunflower, the fourth most widely grown oilseed crop in the world. Results Our association mapping population (AMP), which comprises 135 inbred breeding lines (ILs), was genotyped using 27 candidate genes, a panel of 9 Simple Sequence Repeat (SSR) markers previously associated with SHR resistance via bi-parental mapping, and a set of 384 SNPs located in genes with molecular functions related to stress responses. Moreover, given the complexity of the trait, we evaluated four disease descriptors (i.e, disease incidence, disease severity, area under the disease progress curve for disease incidence, and incubation period). As a result, this work constitutes the most exhaustive AM study of disease resistance in sunflower performed to date. Mixed linear models accounting for population structure and kinship relatedness were used for the statistical analysis of phenotype-genotype associations, allowing the identification of 13 markers associated with disease reduction. The number of favourable alleles was negatively correlated to disease incidence, disease severity and area under the disease progress curve for disease incidence, whereas it was positevily correlated to the incubation period. Conclusions Four of the markers identified here as associated with SHR resistance (HA1848, HaCOI_1, G33 and G34) validate previous research, while other four novel markers (SNP117, SNP136, SNP44, SNP128) were consistently associated with SHR resistance, emerging as promising candidates for marker-assisted breeding. From the germplasm point of view, the five ILs carrying the largest combination of resistance alleles provide a valuable resource for sunflower breeding programs worldwide.

Abstract

A hidroxiapatita [Ca 10 (PO 4 ) 6 (OH) 2 , HA] foi sintetizada utilizando-se a rota sol-gel partindo-se de ácido fosfórico e nitrato de cálcio como precursores de cálcio e fósforo, respectivamente e como solvente utilizou-se o metanol na preparação do sol que posteriormente será utilizado na obtenção de recobrimentos de hidroxiapatita sobre substratos de ligas de titânio. O sol permaneceu estável e não ocorreu gelatinização em temperatura ambiente durante sete dias. O sol transformou-se em um gel branco somente após a remoção do solvente a 100oC. O produto assim obtido foi calcinado em 300°C, 500°C e 700°C e caracterizou-se por DRX, FT-IR, MEV/EDS e TGA/DSC. As fases de HA sintetizada tornaram-se estáveis sem sub-produtos a 700°C. A difração de raios X mostrou que a estrutura apatita é aparente em 300°C. O tamanho do cristal e o teor de HA aumentaram com o aumento da temperatura de calcinação. A análise por MEV mostrou a presença de poros que são importantes para aplicações biomédicas, favorecendo a adesão entre o tecido ósseo neoformado e a apatita sintética, ou seja, osseointegração.

Abstract

Titânio comercialmente puro, Ti-cp, e algumas de suas ligas são consideradas muito importantes na área médica devido sua excelente biocompatibilidade e propriedades mecânicas. Recentemente, foi desenvolvido um método químico, relativamente simples, para induzir a bioatividade desses materiais metálicos inertes, cujo princípio é imitar as condições biológica para obtenção do material desejado. Esta técnica denominada biomimético, foi utilizada para modificar a superfície do Ti-cp através do depósito de uma camada de apatita. O objetivo principal deste trabalho foi estudar a influência do tratamento térmico na evolução da cristalinidade das fases depositadas. Os recobrimentos de apatitas, com tratamentos térmicos entre 400 e 600 °C, mostraram através das técnicas de difração de raios-X e microscopia eletrônica de varredura, uma baixa cristalinidade semelhante às apatitas biológicas. Acima de 700 °C, os recobrimentos de apatita mostraram-se mais cristalinos apresentando uma mistura de fases de hidroxiapatita, fosfato octacálcico e fosfato de magnésio.

Abstract

Objective This study aimed to evaluate the association between polypharmacy and delirium, the association of specific drug categories with delirium, and the differences in drug-delirium association between medical and surgical units and according to dementia diagnosis. Methods Data were collected during 2 waves of Delirium Day, a multicenter delirium prevalence study including patients (aged 65 years or older) admitted to acute and long-term care wards in Italy (2015-2016); in this study, only patients enrolled in acute hospital wards were selected (n = 4,133). Delirium was assessed according to score on the 4 "A's" Test. Prescriptions were classified by main drug categories; polypharmacy was defined as a prescription of drugs from 5 or more classes. Results Of 4,133 participants, 969 (23.4%) had delirium. The general prevalence of polypharmacy was higher in patients with delirium (67.6% vs 63.0%, P = .009) but varied according to clinical settings. After adjustment for confounders, polypharmacy was associated with delirium only in patients admitted to surgical units (OR = 2.9; 95% CI, 1.4-6.1). Insulin, antibiotics, antiepileptics, antipsychotics, and atypical antidepressants were associated with delirium, whereas statins and angiotensin receptor blockers exhibited an inverse association. A stronger association was seen between typical and atypical antipsychotics and delirium in subjects free from dementia compared to individuals with dementia (typical: OR = 4.31; 95% CI, 2.94-6.31 without dementia vs OR = 1.64; 95% CI, 1.19-2.26 with dementia; atypical: OR = 5.32; 95% CI, 3.44-8.22 without dementia vs OR = 1.74; 95% CI, 1.26-2.40 with dementia). The absence of antipsychotics among the prescribed drugs was inversely associated with delirium in the whole sample and in both of the hospital settings, but only in patients without dementia. Conclusions Polypharmacy is significantly associated with delirium only in surgical units, raising the issue of the relevance of medication review in different clinical settings. Specific drug classes are associated with delirium depending on the clinical setting and dementia diagnosis, suggesting the need to further explore this relationship.

35. Co-Expression Networks in Sunflower: Harnessing the Power of Multi-Study Transcriptomic Public Data to Identify and Categorize Candidate Genes for Fungal Resistance.

Abstract

Fungal plant diseases are a major threat to food security worldwide. Current efforts to identify and list loci involved in different biological processes are more complicated than originally thought, even when complete genome assemblies are available. Despite numerous experimental and computational efforts to characterize gene functions in plants, about ~40% of protein-coding genes in the model plant Arabidopsis thaliana L. are still not categorized in the Gene Ontology (GO) Biological Process (BP) annotation. In non-model organisms, such as sunflower ( Helianthus annuus L.), the number of BP term annotations is far fewer, ~22%. In the current study, we performed gene co-expression network analysis using eight terabytes of public transcriptome datasets and expression-based functional prediction to categorize and identify loci involved in the response to fungal pathogens. We were able to construct a reference gene network of healthy green tissue (GreenGCN) and a gene network of healthy and stressed root tissues (RootGCN). Both networks achieved robust, high-quality scores on the metrics of guilt-by-association and selective constraints versus gene connectivity. We were able to identify eight modules enriched in defense functions, of which two out of the three modules in the RootGCN were also conserved in the GreenGCN, suggesting similar defense-related expression patterns. We identified 16 WRKY genes involved in defense related functions and 65 previously uncharacterized loci now linked to defense response. In addition, we identified and classified 122 loci previously identified within QTLs or near candidate loci reported in GWAS studies of disease resistance in sunflower linked to defense response. All in all, we have implemented a valuable strategy to better describe genes within specific biological processes.

Abstract

Diseases caused by necrotrophic fungi, such as the cosmopolitan Sclerotinia sclerotiorum and the Diaporthe/Phomopsis complex, are among the most destructive diseases of sunflower worldwide. The lack of complete resistance combined with the inefficiency of chemical control makes assisted breeding the best strategy for disease control. In this work, we present an integrated genome-wide association (GWA) study investigating the response of a diverse panel of sunflower inbred lines to both pathogens. Phenotypic data for Sclerotinia head rot (SHR) consisted of five disease descriptors (disease incidence, DI; disease severity, DS; area under the disease progress curve for DI, AUDPCI, and DS, AUDPCS; and incubation period, IP). Two disease descriptors (DI and DS) were evaluated for two manifestations of Diaporthe/Phomopsis: Phomopsis stem canker (PSC) and Phomopsis head rot (PHR). In addition, a principal component (PC) analysis was used to derive transformed phenotypes as inputs to a univariate GWA (PC-GWA). Genotypic data comprised a panel of 4269 single nucleotide polymorphisms (SNP), generated via genotyping-by-sequencing. The GWA analysis revealed 24 unique marker-trait associations for SHR, 19 unique marker-trait associations for Diaporthe/Phomopsis diseases, and 7 markers associated with PC1 and PC2. No common markers were found for the response to the two pathogens. Nevertheless, epistatic interactions were identified between markers significantly associated with the response to S. sclerotiorum and Diaporthe/Phomopsis. This suggests that, while the main determinants of resistance may differ for the two pathogens, there could be an underlying common genetic basis. The exploration of regions physically close to the associated markers yielded 364 genes, of which 19 were predicted as putative disease resistance genes. This work presents the first simultaneous evaluation of two manifestations of Diaporthe/Phomopsis in sunflower, and undertakes a comprehensive GWA study by integrating PSC, PHR, and SHR data. The multiple regions identified, and their exploration to identify candidate genes, contribute not only to the understanding of the genetic basis of resistance, but also to the development of tools for assisted breeding.

Abstract

Bovine Anaplasmosis caused by Anaplasma marginale is a worldwide disease prevalent in tropical and subtropical regions where Rhipicephalus microplus is considered the most significant biological vector. Molecular markers previously applied for A. marginale typing are efficient for isolate discrimination but they are not a suitable tool for studying population structure and dynamics. Here we report the development of an MLST scheme based on the study of seven genes: dnaA, ftsZ, groEl, lipA, recA, secY and sucB. Five annotated genomes (Saint Maries, Florida, Mississippi, Puerto Rico and Virginia) and 53 bovine blood samples from different world regions were analyzed. High nucleotide diversity and a large proportion of synonymous substitutions, indicative of negative selection resulted from DnaSP 5.00.02 package application. Recombination events were detected in almost all genes, this evidence together with the coexistence of more than one A. marginale strain in the same sample might suggest the superinfection phenomena as a potential source of variation. The allelic profile analysis performed through GoeBURST shown two main CC that did not support geography. In addition, the AMOVA test confirmed the occurrence of at least two main genetically divergent groups. The composition of the emergent groups reflected the impact of both historical and environmental traits on A. marginale population structure. Finally, a web-based platform "Galaxy MLST-Pipeline" was developed to automate DNA sequence editing and data analysis that together with the Data Base are freely available to users. The A. marginale MLST scheme developed here is a valuable tool with a high discrimination power, besides PCR based strategies are still the better choice for epidemiological intracellular pathogens studies. Abstract

Heteroduplex-based genotyping methods have proven to be technologically effective and economically efficient for low- to medium-range throughput single-nucleotide polymorphism (SNP) determination. In this chapter we describe two protocols that were successfully applied for SNP detection and haplotype analysis of candidate genes in association studies. The protocols involve (1) enzymatic mismatch cleavage with endonuclease CEL1 from celery, associated with fragment separation using capillary electrophoresis (CEL1 cleavage), and (2) differential retention of the homo/heteroduplex DNA molecules under partial denaturing conditions on ion pair reversed-phase liquid chromatography (dHPLC). Both methods are complementary since dHPLC is more versatile than CEL1 cleavage for identifying multiple SNP per target region, and the latter is easily optimized for sequences with fewer SNPs or small insertion/deletion polymorphisms. Besides, CEL1 cleavage is a powerful method to localize the position of the mutation when fragment resolution is done using capillary electrophoresis.

Abstract

Bovine babesiosis, caused by the protozoa Babesia bovis and Babesia bigemina, is a tick-borne disease distributed in tropical regions worldwide. Current control measures are based on the use of acaricides and live attenuated vaccines. The major economic impact of babesiosis lies in the cattle industry. In order to gain insight into the extent of genetic diversity in populations of parasites in the field, we developed two MLST schemes for the molecular genotyping of B. bigemina and B. bovis. We have also developed a custom-designed bioinformatic pipeline to facilitate the automated processing of raw sequences and further diversity and phylogenetic analysis. The overall MLST scheme exhibited the maximum discriminatory power (Simpson Index=1) for B. bovis and a high level of discrimination for B. bigemina (Simpson Index=0.9545). Genetic diversity was very high and infections with multiple genotypes were frequently found for both parasites in outbreak samples from the Northeast and Northwest of Argentina. Recombination events, which could have arisen from these multiple infections, were suggested by intra-loci linkage disequilibrium analysis and the lack of congruence in phylogenetic trees from individual genes. The two MLST schemes developed here are a robust, objective and easily adoptable technology to analyze the genetic diversity and population structure of parasites of the genus Babesia., (Copyright © 2013 Elsevier B.V. 40. Isolation of Paenibacillus sp. and Variovorax sp. strains from decaying woods and characterization of their potential for cellulose deconstruction.

Abstract

Prospection of cellulose-degrading bacteria in natural environments allows the identification of novel cellulases and hemicellulases that could be useful in second-generation bioethanol production. In this work, cellulolytic bacteria were isolated from decaying native forest soils by enrichment on cellulose as sole carbon source. There was a predominance of Gram positive isolates that belonged to the phyla Proteobacteria and Firmicutes. Many primary isolates with cellulolytic activity were not pure cultures. From these consortia, isolation of pure constituents was attempted in order to test the hypothesis whether microbial consortia are needed for full degradation of complex substrates. Two isolates, CB1-2-A-5 and VG-4-A-2, were obtained as the pure constituents of CB1-2 and VG-4 consortia, respectively. Based on 16S RNA sequence, they could be classified as Variovorax paradoxus and Paenibacillus alvei. Noteworthy, only VG-4 consortium showed measurable xylan degrading capacity and signs of filter paper degradation. However, no xylan or filter paper degrading capacities were observed for the pure cultures isolated from it, suggesting that other members of this consortium were necessary for these hydrolyzing activities. Our results indicated that Paenibacillus sp. and Variovorax sp. as well as VG-4 consortium, might be a useful source of hydrolytic enzymes. Moreover, although Variovorax sp. had been previously identified in metagenomic studies of cellulolytic communities, this is the first report on the isolation and characterization of this microorganism as a cellulolytic genus.

Abstract

We report a comprehensive primary metabolite profiling of sunflower (Helianthus annuus) genotypes displaying contrasting behavior to Sclerotinia sclerotiorum infection. Applying a GC-MS-based metabolite profiling approach, we were able to identify differential patterns involving a total of 63 metabolites including major and minor sugars and sugar alcohols, organic acids, amino acids, fatty acids and few soluble secondary metabolites in the sunflower capitulum, the main target organ of pathogen attack. Metabolic changes and disease incidence of the two contrasting genotypes were determined throughout the main infection period (R5.2-R6). Both point-by-point and non-parametric statistical analyses showed metabolic differences between genotypes as well as interaction effects between genotype and time after inoculation. Network correlation analyses suggested that these metabolic changes were synchronized in a time-dependent manner in response to the pathogen. Concerted differential metabolic changes were detected to a higher extent in the susceptible, rather than the resistant genotype, thereby allowing differentiation of modules composed by intermediates of the same pathway which are highly interconnected in the susceptible line but not in the resistant one. Evaluation of these data also demonstrated a genotype specific regulation of distinct metabolic pathways, suggesting the importance of detection of metabolic patterns rather than specific metabolite changes when looking for metabolic markers differentially responding to pathogen infection. In summary, the GC-MS strategy developed in this study was suitable for detection of differences in carbon primary metabolism in sunflower capitulum, a tissue which is the main entry point for this and other pathogens which cause great detrimental impact on crop yield., (2009 Elsevier Ltd. Abstract

Exploiting the manipulation of the SLAC Linear Collider electron-beam polarization, we present precise direct measurements of the parity-violation parameters A(c) and A(b) in the Z-boson-c-quark and Z-boson-b-quark coupling. Quark-antiquark discrimination is accomplished via a unique algorithm that takes advantage of the precise SLAC Large Detector charge coupled device vertex detector, employing the net charge of displaced vertices as well as the charge of kaons that emanate from those vertices. From the 1996-1998 sample of 400 000 Z decays, produced with an average beam polarization of 73.4%, we find A(c)=0.673+/-0.029(stat)+/-0.023(syst) and A(b)=0.919+/-0.018(stat)+/-0.017(syst).

Abstract

We present an improved direct measurement of the parity-violation parameter A(b) in the Z boson-b-quark coupling using a self-calibrating track-charge technique applied to a sample enriched in Z-->bb events via the topological reconstruction of the B hadron mass. Manipulation of the Stanford Linear Collider electron-beam polarization permits the measurement of A(b) to be made independently of other Z-pole coupling parameters. From the 1996-1998 sample of 400,000 hadronic Z decays, produced with an average beam polarization of 73.4%, we find A(b)=0.906+/-0.022(stat)+/-0.023(syst).

Abstract

Heteropteran chromosomes are holokinetic; during mitosis, sister chromatids segregate parallel to each other but, during meiosis, kinetic activity is restricted to one pair of telomeric regions. This meiotic behaviour has been corroborated for all rod bivalents. For ring bivalents, we have previously proposed that one of the two chiasmata releases first, and a telokinetic activity is also achieved. In the present work we analyse the meiotic behaviour of ring bivalents in Pachylis argentinus (Coreidae) and Nezara viridula (Pentatomidae) and we describe for the first time the chromosome complement and male meiosis of the former (2n = 12 + 2m + X0, pre-reduction of the X). Both species possess a large chromosome pair with a secondary constriction which is a nucleolus organizer region as revealed by in-situ hybridization. Here we propose a new mode of segregation for ring bivalents: when the chromosome pair bears a secondary constriction, it is not essential that one of the chiasmata releases first since these regions or repetitive DNA sequences adjacent to them become functional as alternative sites for microtubule attachment and they undertake chromosome segregation to the poles during anaphase I.

Abstract

The parity violation parameters A(b) and A(c) of the Zb(b) and Zc(c) couplings have been measured directly, using the polar angle dependence of the polarized cross sections at the Z(0) pole. Bottom and charmed hadrons were tagged via their semileptonic decays. Both the electron and muon analyses take advantage of new multivariate techniques to increase the analyzing power. Based on the 1993-1998 SLD sample of 550,000 Z(0) decays produced with highly polarized electron beams, we measure A(b) = 0.919+/-0.030(stat)+/-0.024(syst), and A(c) = 0.583+/-0.055(stat)+/-0.055(syst).

Abstract

We present final measurements of the Z boson-lepton coupling asymmetry parameters A(e), A(mu), and A(tau) with the complete sample of polarized Z bosons collected by the SLD detector at the SLAC Linear Collider. From the left-right production and decay polar angle asymmetries in leptonic Z decays we measure A(e) = 0.1544+/-0.0060, A(mu) = 0.142+/-0.015, and A(tau) = 0.136+/-0.015. Combined with our left-right asymmetry measured from hadronic decays, we find A(e) = 0.1516+/-0.0021. Assuming lepton universality, we obtain a combined effective weak mixing angle of sin (2)theta(eff)(W) = 0.230 98+/-0.000 26.

Abstract

We have made the first direct symmetry tests in the decays of polarized Z0 bosons into fully identified bbg states, collected in the SLD experiment at SLAC. We searched for evidence of parity violation at the bbg vertex by studying the asymmetries in the b-quark polar- and azimuthal-angle distributions, and for evidence of T-odd, CP-even or CP-odd, final-state interactions by measuring angular correlations between the three-jet plane and the Z0 polarization. We found results consistent with standard model expectations and set 95% C. limits on anomalous contributions.

Abstract

Interleukin-6 (IL-6) belongs to a family of cytokines that use receptors consisting of a common signal-transducing chain (gp130). Baf/3 cells transfected with the human IL-6 receptor (IL-6R) and gp130 (Baf/3-gp130/IL-6R) can only grow in medium containing IL-6. We attempted to interrupt the signal transducing pathway of IL-6 with the help of antisense oligonucleotides (ASOs) designed against the IL-6R. We used 18 different kinds of antisense oligonucleotides of overlapping sequences around the translational start codon of the human IL-6R. Sense ASOs were used as a control. The proliferation of cells was analysed by H-thymidine incorporation. Cell surface expression of the IL-6R was assessed by FACS analysis. We identified three ASOs which strongly inhibited the proliferation of IL-6 dependent transfected Baf/3 cells. Flow cytometric studies on the suppression of surface expression of IL-6R by ASOs showed a similar pattern. These results should help to clarify the structural requirements of functionally effective ASOs in the inhibition of IL-6R., (Copyright 2001 Academic Press.)

49. Isozyme variation in Larrea ameghinoi and Larrea nitida (Zygophyllaceae): genetic diversity and its bearing on their relationship.

Abstract

Electrophoretic variants for 11 isozyme systems were examined by horizontal polyacrilamide gel electrophoresis in natural populations of Larrea ameghinoi and L. nitida (section Larrea). The two taxa presented fixed alternative allelic variants at loci Adh-1, Gdh-1 and Mdh-1. Genetic variability estimates showed no statistically significant differences among populations of section Larrea (He: 0.097-0.167). However, these values were significantly lower than those reported for species of section Bifolium (L. divaricata, diploid L. tridentata) in a previous contribution (He: 0.17-0.29). Positive and highly significant Wright's fixation indices are in agreement with higher amounts of inbreeding for L. nitida and L. ameghinoi, as suggested on the basis of floral morphology by previous authors. Moreover, the higher levels of genetic differentiation obtained for conspecific populations of section Larrea as compared to those of section Bifolium are also consistent with these observations. Nei's genetic identity values obtained for sympatric (I = 0.80) and allopatric (I = 0.63-0.73) L. ameghinoi - L. nitida population pairs are concordant with those expected for different, but closely related congeneric species.
