Cowpea is one of the most important leguminous crops in Sub-Saharan Africa (SSA), and moisture stress is among the constraints affecting its productivity. This study was conducted to understand the response of cowpea accessions to moisture stress. A total of 255 cowpea accessions from Togo and four checks from the International Institute of Tropical Agriculture (IITA), were assessed. The trials were conducted in the glasshouse and an open field (which was divided into moisture-stressed (MS) and non-moisture-stressed fields (NMS)). In the non-moisture-stressed environment compared to the moisture-stressed environment, there was a greater heritability for agronomic traits such as biomass, seed weight, and pod weight. The accessions with the highest seed weights (yield-related traits), surpassing the checks under both moisture-stressed and non-moisture-stressed conditions in the field, were six viz.: RK173 (49.8 g (MS); 90.4 g (NMS)), RP225 (34.6 g (MS); 119.9 g (NMS)), RP232 (33.4 g (MS); 51.9 g (NMS)), RM357 (27.9 g (MS); 62.9 g (NMS)), RK148 (23.9 g (MS); 63.4 g (NMS)), and Vu081_2_2 (21.8 g (MS); 46.7 g (NMS)). The most promising accession was RK173; this was ranked first under the moisture-stressed condition and ranked second under the non-moisture-stressed condition with a loss in weight of 44.9% due to drought stress. Of the top 20 accessions that recovered after watering resumed in the glasshouse screening, only the following 9 had a recovery percentage higher than 5% viz.: RS029 (34.5%), RK014 (14.2%), RS114 (9.6%), RK121 (8.3%), RS007 (7.6%), RK123 (7.3%), RS037 (7.3%), RS101 (5.6%), and RS108 (5.1%). The best line and those with a higher recovery percentage could be exploited further in order to improve them in future drought breeding programs by crossing them with lines susceptible to drought or using other drought breeding techniques.

Abstract Background The establishment of heterotic groups of inbred lines is crucial for hybrid maize breeding programs. Currently, there is no information on the heterotic patterns of the Provitamin A (PVA) inbred lines developed in the maize improvement program of the International Institute of Tropical Agriculture (IITA) to form productive PVA enriched hybrids for areas affected by vitamin A deficiency. This study assessed the feasibility of classifying PVA-enriched inbred lines into heterotic groups based on PVA content without compromising grain yield in hybrids. Sixty PVA inbred lines were crossed to two testers representing two existing heterotic groups. The resulting 120 testcrosses hybrids were evaluated for two years at four locations in Nigeria. Results The two testers effectively classified the inbred lines into two heterotic groups. The PVA-based general combining ability and specific combining ability (HSGCA) method assigned 31 and 27 PVA enriched maize inbred lines into HGB and HGA, respectively, leaving two inbred lines not assigned to any group. The yield-based HSGCA method classified 32 inbred lines into HGB and 28 inbred lines into HGA. Both PVA and yield-based heterotic grouping methods assigned more than 40% of the inbred lines into the same heterotic groups. Even though both PVA and yield-based heterotic grouping of the inbred lines differed from the clusters defined by the DArTag SNP markers, more than 40% of the inbred lines assigned to HGA were present in Cluster-1 and 60% of the inbred lines assigned to HGB were present in Cluster-3. Interestingly, the inbred lines assigned to the same heterotic groups based on PVA content and grain yield were distributed across the three Ward’s clusters. The PVA-based HSGCA was identified as the most effective heterotic grouping method for breeding programs working on PVA biofortification. Conclusions Selecting PVA enriched maize inbred lines with diverse genetic backgrounds from the three marker-based clusters may facilitate the development of productive hybrids with high PVA content and for generating source populations to develop more vigorous maize inbred lines with much higher concentrations of PVA.

Abstract Background Temperate maize inbred lines with expired Plant Variety Protection Act certificates (Ex-PVP) are potential sources of desirable alleles for tropical germplasm improvement. Up to now, the usefulness of the Ex-PVP inbred lines as a potential source of novel beneficial alleles for Striga hermonthica resistance breeding to enhance genetic gain in tropical maize has not been reported. Results This study was thus conducted to characterize the combining ability of 24 Ex-PVP inbred lines in crosses with two tropical Striga resistant inbred testers under Striga-infested and non-infested conditions and across three locations for 2 years. Many testcrosses between Ex-PVP inbred lines and the first tester (T1) produced competitive or significantly higher grain yields compared to the hybrid between the two resistant testers under Striga infested and non-infested conditions and across multiple test locations. Also, most of the testcrosses with positive heterosis for grain yield and negative heterosis for Striga damage and emerged Striga count involved T1 as a tester. Our study identified six Ex-PVP inbred lines with positive GCA effects for grain yield under Striga infested and non-infested conditions and across multiple test locations. Amongst these, inbred lines HB8229-1 and WIL900-1 also displayed negative GCA effects for emerged Striga count and Striga damage rating. The inbred line HB8229-1 showed positive SCA effects for grain yield with T2, whereas WIL900-1 had positive SCA effects for grain yield with T1. Over 70% of the Ex-PVP inbred lines were consistently assigned to specific heterotic groups using yield-based classifying methods (mean grain yield and SCA effects). Conclusions These results could facilitate systematic introgression of the Ex-PVP inbred lines into the existing Striga resistant heterotic groups in IITA. The Ex-PVP inbred lines with positive GCA effects and producing high grain yields in hybrid combinations could be useful parents for enhancing Striga resistance and agronomic performance of tropical maize hybrids.

Development and deployment of high-yielding maize varieties with native resistance to Fall armyworm (FAW), turcicum leaf blight (TLB), and gray leaf spot (GLS) infestation is critical for addressing the food insecurity in sub-Saharan Africa. The objectives of this study were to determine the inheritance of resistance for FAW, identity hybrids which in addition to FAW resistance, also show resistance to TLB and GLS, and investigate the usefulness of models based on general combining ability (GCA) and SNP markers in predicting the performance of new untested hybrids. Half-diallel mating scheme was used to generate 105 F1 hybrids from 15 parents and another 55 F1 hybrids from 11 parents. These were evaluated in two experiments, each with commercial checks in multiple locations under FAW artificial infestation and optimum management in Kenya. Under artificial FAW infestation, significant mean squares among hybrids and hybrids x environment were observed for most traits in both experiments, including at least one of the three assessments carried out for foliar damage caused by FAW. Interaction of GCA x environment and specific combining ability (SCA) x environment interactions were significant for all traits under FAW infestation and optimal conditions. Moderate to high heritability estimates were observed for GY under both management conditions. Correlation between GY and two of the three scorings (one and three weeks after infestation) for foliar damage caused by FAW were negative (-0.27 and -0.38) and significant. Positive and significant correlation (0.84) was observed between FAW-inflicted ear damage and the percentage of rotten ears. We identified many superior-performing hybrids compared to the best commercial checks for both GY and FAW resistance associated traits. Inbred lines CML312, CML567, CML488, DTPYC9-F46-1-2-1-2, CKDHL164288, CKDHL166062, and CLRCY039 had significant and positive GCA for GY (positive) and FAW resistance-associated traits (negative). CML567 was a parent in four of the top ten hybrids under optimum and FAW conditions. Both additive and non-additive gene action were important in the inheritance of FAW resistance. Both GCA and marker-based models showed high correlation with field performance, but marker-based models exhibited considerably higher correlation. The best performing hybrids identified in this study could be used as potential single cross testers in the development of three-way FAW resistance hybrids. Overall, our results provide insights that help breeders to design effective breeding strategies to develop FAW resistant hybrids that are high yielding under FAW and optimum conditions.

Rice yellow mottle virus (RYMV) is the most harmful virus that affects irrigated and lowland rice in Africa. The RBe24 isolate of the virus is the most pathogenic strain in Benin. A total of 79 genotypes including susceptible IR64 (Oryza sativa) and the resistant TOG5681 (O. glaberrima) as checks were screened for their reactions to RBe24 isolate of RYMV and the effects of silicon on the response of host plants to the virus investigated. The experiment was a three-factor factorial consisting of genotypes, inoculation level (inoculated vs. non-inoculated), and silicon dose (0, 5, and 10 g/plant) applied as CaSiO3 with two replications and carried out twice in the screen house. Significant differences were observed among the rice genotypes. Fifteen highly resistant and eight resistant genotypes were identified, and these were mainly O. glaberrima. Silicon application did not affect disease incidence and severity at 21 and 42 days after inoculation (DAI); it, however, significantly increased plant height of inoculated (3.6% for 5 g CaSiO3/plant and 6.3% for 10 g CaSiO3/plant) and non-inoculated (1.9% for 5 g CaSiO3/plant and 4.9% for 10 g CaSiO3/plant) plants at 42 DAI, with a reduction in the number of tillers (12.3% for both 5 and 10 g CaSiO3/plant) and leaves (26.8% for 5 g CaSiO3/plant and 28% for 10 g CaSiO3/plant) under both inoculation treatments. Our results confirm O. glaberrima germplasm as an important source of resistance to RYMV, and critical in developing a comprehensive strategy for the control of RYMV in West Africa.

Vitamin A deficiency poses health risks for children, pregnant women, and nursing mothers in sub-Saharan Africa (SSA) and Southeast Asia. Provitamin A–biofortified maize varieties can contribute to minimizing the adverse effects of vitamin A deficiency in areas where maize is a staple food crop. Identifying suitable testers is important to breed provitamin A–biofortified hybrid maize. This study was therefore conducted to 1) assess the suitability of maize inbred lines with contrasting levels of provitamin A (one with high and one with low provitamin A concentration) to assess the combining ability of maize inbred lines in accumulating provitamin A and other carotenoids, and grain yield, 2) confirm the mode of inheritance of provitamin A and grain yield, and 3) identify promising inbred lines with desirable combining ability effects for use to develop high-yielding provitamin A–biofortified hybrids. The inbreds crossed to the two inbred testers were evaluated in four environments for the carotenoid content and eight environments for the agronomic performance. The combined analysis of variance revealed a significant genetic variation among the testcrosses for all carotenoids, grain yield, and other agronomic traits. The mode of inheritance for grain yield, other agronomic traits, provitamin A, and other carotenoids was regulated by both additive and non-additive gene effects with a prominence of additive gene effects. The high provitamin A tester that displayed positive GCA effects for β-carotene and provitamin A content, broader agronomic performance of testcrosses, and higher levels of provitamin A in testcrosses can be considered suitable for breeding programs developing provitamin A–biofortified hybrids. The inbred lines TZI2012, TZI2142, TZI2130, TZI2065-2, TZI2161, TZI2025, TZI1278, TZI1314, TZI1304, and TZI2032 with positive GCA effects for grain yield and provitamin A content could be used as parental lines to develop source population of new inbred lines and high-yielding hybrids with elevated levels of provitamin A. The best performing hybrids are promising for release as high-yielding provitamin A maize hybrids after further evaluations.

Probiotics and related preparations, including synbiotics and postbiotics, are living and non-living microbial-based multi-components, which are now among the most popular bioactive agents. Such interests mainly arise from the wide range and numerous beneficial effects of their use for various hosts. The current minireview article attempts to provide an overview and discuss in a holistic way the concepts, methodologies, action mechanisms, and applications of probiotic-based multi-components in human, animal, plant, soil, and environment health. Probiotic-based multi-component preparations refer to a mixture of bioactive agents, containing probiotics or postbiotics as main functional ingredients, and prebiotics, protectants, stabilizers, encapsulating agents, and other compounds as additional constituents. Analyzing, characterizing, and monitoring over time the traceability, performance, and stability of such multi-component ingredients require relevant and sensitive analytical tools and methodologies. Two innovative profiling and monitoring methods, the thermophysical fingerprinting thermogravimetry–differential scanning calorimetry technique (TGA-DSC) of the whole multi-component powder preparations, and the Advanced Testing for Genetic Composition (ATGC) strain analysis up to the subspecies level, are presented, illustrated, and discussed in this review to respond to those requirements. Finally, the paper deals with some selected applications of probiotic-based multi-components to human, animal, plant, soil and environment health, while mentioning their possible action mechanisms.

The development of maize varieties with increased concentration of Provitamin A (PVA) is an effective and affordable strategy to combat vitamin A deficiency in developing nations. However, the considerably high cost of carotene analysis poses a major challenge for maize PVA biofortification, prompting the use of marker-assisted selection. Presently, two types of genotyping with PVA trait-linked functional markers have been developed and extensively used in breeding programs. The two systems are low throughput gel-based genotyping and genotyping with Kompetitive Allele-Specific PCR (KASP) single nucleotide polymorphism (SNPs) markers. Although the KASP SNPs genotyping was developed to replace the gel-based genotyping, studies have not been conducted to compare the effectiveness of the KASP SNPs markers with the gel-based markers. This study was conducted to assess the carotenoid content of 64 tropical PVA biofortified maize inbred lines containing temperate germplasm in their genetic backgrounds and screen them with both gel-based and KASP markers of PSY1, LCYE and crtRB1 genes. Many of the 64 inbred lines had PVA concentrations surpassing the 15 µg/g provitamin A breeding target set by the HarvestPlus Challenge Program. Favorable alleles of crtRB1, crtRB1 and the KASP SNPs markers were detected in 25 inbred lines with high PVA concentrations. Inbred lines with the favorable alleles of LCYE had the highest concentrations of non-PVA carotenoids, whereas those with the favorable alleles of crtRB1 had high levels of PVA carotenoids. Data from the sequenced region of LCYE revealed one SNP in the first intron that clearly differentiated the high and low β-carotene maize inbred lines. The results of our study demonstrate that the automated KASP SNPs markers can replace the gel-based genotyping for screening a large number of early generation maize inbred lines for PVA content.

A hemiparasitic plant, Striga hermonthica (Del.) Benth and soil nitrogen stress are the key constraints to maize (Zea mays L.) productivity in sub-Saharan Africa, where commonly cultivated maize is the normal endosperm type that is deficient in provitamin A, tryptophan and lysine (PVATL). Seventy-six extra-early maize inbreds with provitamin A, tryptophan, and lysine qualities (TZEEIORQ) were developed to address these constraints, and four checks were assessed under Striga, low and high nitrogen conditions at three locations in Nigeria. The inbreds were further genotyped with two beta-carotene hydroxylase 1 (crtRB1) markers, and their seeds were quantified for provitamin A content. Significant (p < 0.01) genetic variations were observed for grain yield and other agronomic attributes of the inbreds under varying environmental conditions. Levels of PVATL for the inbreds ranged from 2.21–10.95 µg g−1, 0.04–0.08%, and 0.19–0.39%, respectively. Beta-carotene marker, crtRB1-3′TE, was polymorphic and grouped the inbreds into two. The marker was effective in identifying inbreds with moderate provitamin A content. Inbreds TZEEIORQ 5, TZEEIORQ 52, and TZEEIORQ 55 exhibited resistance to Striga, tolerance to nitrogen stress with moderate levels of PVATL and could be invaluable sources of favorable alleles for breeding nutritionally improved maize varieties with resistance/tolerance to Striga and soil nitrogen stress.

This study determined the gene effects involved in the inheritance of pod length and other yield-related traits and relationships among traits in crosses between two cowpea lines (TVu 2280 and TVu 2027) and a yard-long bean (TVu 6642) line with long pods. Plants of six generations (P1, P2, F1, F2, BC1P1, and BC1P2) derived from TVu 2280 × TVu 6642 and TVu 2027 × TVu 6642 were evaluated under field conditions. Data collected on 14 yield components of each cross were used for generation mean analysis. Gene effects and their magnitudes varied with the crosses; digenic epistatic gene effects were detected for 10 traits in TVu 2280 × TVu 6642 and 11 traits in TVu 2280 × TVu 6642. Only additive gene effect was significant for pod length in TVu 2280 × TVu 6642 while additive, dominant, and two of the three digenic epistatic gene effects were significant in TVu 2027 × TVu 6642. Models that incorporated only significant additive, dominant, and digenic epistasis were adequate for all 14 traits in TVu 2280 × TVu 6642 and eight of the 12 traits in TVu 2027 × TVu 6642 for which model-fitting was possible. Across segregating generation of the two crosses, pod length (PodLNT) was significantly (p < 0.001) correlated with three major yield components viz. pod weight (0.84, 0.77), number of seeds per pod (0.41, 0.30) and seed weight per pod (0.61, 0.29). Significant correlation of PodLNT with seed yield per plant was moderate and significant (p < 0.01–0.001) in the BC1P1 of the two crosses (0.31 and 0.41). An improvement in cowpea seed yield is feasible through selection for long pods in segregating generations involving crosses with yard-long bean.

Maize streak virus (MSV) is responsible for streak disease of maize and poses a serious threat to maize production in sub-Saharan Africa. Polygenic resistance to MSV has become an essential requirement in modern maize cultivars to mitigate yield losses. Many single nucleotide polymorphism (SNP) markers linked to putative MSV resistance loci have been identified for use in forward breeding. This study aimed to validate, using the high-throughput kompetitive allele specific PCR (KASP) assay, the diagnostic ability of the three SNP markers linked to the loci for the Msv1 resistance trait in 151 early generations inbred lines with diverse genetic backgrounds, together with nine MSV-resistant elite lines and a susceptible check (cv. Pool-16). The phenotypic responses were determined by MSV inoculation using viruliferous leafhoppers (Cicadulina triangular) under screenhouse conditions. Based on an established MSV disease rating system, the maize lines were categorized into resistant, moderately resistant, susceptible, and highly susceptible. The three SNPs associated with MSV resistance were detected in 133 lines, which were categorized as resistant (54), moderately resistant (76), and susceptible (1). The 18 early generation lines without these SNPs were classified as moderately resistant (10), susceptible (5), and highly susceptible (3). This study confirms the strong association of SNPs with MSV resistance and their usefulness for forward breeding in maize while emphasizing the need to identify additional markers to screen lines for MSV resistance without any ambiguity.
