Measuring the transit time of a cell forced through a bottleneck is one of the most widely used techniques for the study of cell deformability in flow. It in turn provides an accessible and rapid way of obtaining crucial information regarding cell physiology. Many techniques are currently being investigated to reliably retrieve this time, but their translation to diagnostic-oriented devices is often hampered by their complexity, lack of robustness, and the bulky external equipment required. Herein, we demonstrate the benefits of coupling microfluidics with an optical method, like photocells, to measure the transit time. We exploit the femtosecond laser irradiation followed by chemical etching (FLICE) fabrication technique to build a monolithic 3D device capable of detecting cells flowing through a 3D non-deformable constriction which is fully buried in a fused silica substrate. We validated our chip by measuring the transit times of pristine breast cancer cells (MCF-7) and MCF-7 cells treated with Latrunculin A, a drug typically used to increase their deformability. A difference in transit times can be assessed without the need for complex external instrumentation and/or demanding computational efforts. Multispectral imaging and time-resolved imaging are two common acquisition schemes in fluorescence microscopy, and their combination can be beneficial to increase specificity. The multidimensionality of the dataset (space, time, and spectrum) introduces some challenges, such as the acquisition of big datasets and long measurement times. In this work, we present a time-resolved multispectral fluorescence microscopy system with a short measurement time, achieved by exploiting Compressive Sensing (CS) based on the Single-Pixel Camera (SPC) scheme. Data Fusion (DF) with a high-resolution camera allows us to tackle the problem of low spatial resolution, typical of SPC. The combined use of SPC, CS, and DF, in which hardware and algorithms are integrated, represents a computational imaging framework to reduce the number of measurements while preserving the information content. This approach has been exploited to demonstrate a zoom feature without moving the optical system. Cell membrane perturbation is a common way to stimulate cells by using external actuators. Recently, nanotechnology has added a number of new strategies for doing this, enlarging the scope and the range of mechanisms involved. Here, we describe a number of possible perturbation actions that are driven by light, and we try to capture the underlying phenomena. The non‐covalent affinity of photoresponsive molecules to biotargets represents an attractive tool for achieving effective cell photo‐stimulation. Here, an amphiphilic azobenzene that preferentially dwells within the plasma membrane is studied. In particular, its isomerization dynamics in different media is investigated. It is found that in molecular aggregates formed in water, the isomerization reaction is hindered, while radiative deactivation is favored. However, once protected by a lipid shell, the photochromic molecule reacquires its ultrafast photoisomerization capacity. This behavior is explained considering collective excited states that may form in aggregates, locking the conformational dynamics and redistributing the oscillator strength. By applying the pump probe technique in different media, an isomerization time in the order of 10 ps is identified and the deactivation in the aggregate in water is also characterized. Finally, it is demonstrated that the reversible modulation of membrane potential of HEK293 cells via illumination with visible light can be indeed related to the recovered trans→cis photoreaction in lipid membrane. These data fully account for the recently reported experiments in neurons, showing that the amphiphilic azobenzenes, once partitioned in the cell membrane, are effective light actuators for the modification of the electrical state of the membrane. Current technologies to monitor formation and disruption in in vitro cell cultures are based either on optical techniques or on electrical impedance/resistance measurement, which often rely on cumbersome and time‐consuming measurements and data analyses. In this paper, poly(3,4‐ethylenedioxythiophene) polystyrene sulfonate (PEDOT:PSS)‐based organic electrochemical transistors (OECTs) are proposed with channel areas specifically designed and dimensioned as fast and real‐time monitoring devices for a large variety of cell lines, with a broad range of tissue resistance. In particular, it is investigated how and why two device configurations provide a different response to leaky‐barrier (NIH‐3T3) and strong‐barrier (CaCo‐2) cell lines growth and detachment, achieving a continuous monitoring also for leaky‐barrier cell layer growth and detachment. Data are collected using the transistor dynamic behavior to a DC potential pulse on the gate, providing an excellent time resolution and thus enhancing the amount of information that can be collected for fast biological processes (<5 s). A single-pixel camera combined with compressive sensing techniques is a promising fluorescence microscope scheme for acquiring a multidimensional dataset (space, spectrum, and lifetime) and for reducing the measurement time with respect to conventional microscope schemes. However, upon completing the acquisition, a computational step is necessary for image reconstruction and data analysis, which can be time-consuming, potentially canceling out the beneficial effect of compressive sensing. In this work, we propose and experimentally validate a fast-fit workflow based on global analysis and multiple linear fits, which significantly reduces the computation time from tens of minutes to less than 1 s. Moreover, as the method is interlaced with the measurement flow, it can be applied in parallel with the acquisitions.
