This dissertation covers the history of municipal collecting in Lyon during the years 1900 - 1914. Chapter One deals with the tenure of Édouard Aynard (1837 - 1913), the prominent Lyonnais statesman and Président of the Conseil d'Administration des Musées (1878 - 1897), who sought to elevate the status of the museum in France and Europe by improving the quality of the collection with acquisitions made by purchase. Although he was unable to achieve results that reflected the scope of his ambition, Aynard nevertheless developed strategic policies that appear to have had an immediate and lasting influence on collecting in Lyon. In Chapter Two, the reasons why the Commission Consultative des Musées was established in 1897 are examined. This restructure of the management and organisation of the group was significant because it led to greater diversification of its membership and resulted in a firm emphasis being placed on consensus decision-making. The five key individuals responsible for shaping attitudes after 1900 are also introduced in this chapter: Victor Augagneur (1855 - 1931), Édouard Herriot (1872 - 1957), Raymond Tripier (1838 - 1916), Émile Bertaux (1869 - 1917) and Richard Cantinelli (1870 - 1932). The respective backgrounds of these men are outlined, particularly in the context of their outlooks on the Dreyfus Affair and the Separation of Church and State. In Chapter Three, the issue of regional pride is considered through an examination of the acquisition of works by artists from Lyon and the wider region. Chapter Four consists of a series of six case studies which show that a main priority for the Commission was to actively seek out paintings by major French nineteenth-century artists. This area of collecting activity was important and it is examined in detail in order to determine how, and why, the museum became the first in France to purchase works by Claude Monet (1840 - 1926) and Paul Gauguin (1848 - 1903), in 1902 and 1913 respectively. Chapter Five treats the long collaboration between the Commission and Auguste Rodin (1840 - 1917) which spanned the period 1903 - 1912. It analyses the circumstances that gave rise to Tripier, Herriot and Cantinelli consciously seeking to develop and maintain a strong working relationship with the artist. The success of the approach taken was such that it resulted in Lyon accumulating the most comprehensive collection of works by the sculptor held by any museum in France outside Paris.

3. The Impact of a High School-Based Positive Youth Development Program on Sexual Health Outcomes: Results from a Randomized Controlled Trial

Background: Although positive youth development (PYD) programs have demonstrated effectiveness in improving adolescent reproductive health outcomes, there is a lack of evidence on effective school-based interventions designed especially for high school settings. This study examined the efficacy of Peer Group Connection (PGC-HS), a school-based PYD program, in improving sexual health outcomes for high school participants. Methods: A total of 1523 ninth-grade students at 18 schools were randomly assigned to be offered PGC-HS or a classes-as-usual control condition during 2016 to 2017 and 2017 to 2018 school years. Impacts were assessed on three confirmatory and 6 exploratory outcomes via self-reported participant questionnaire data collected at the beginning of 10th grade. Results: Although the offer of PGC-HS had no statistically detectable effect on confirmatory behavioral outcomes (sexual initiation, frequency of sex, and number of sexual partners) at 10th grade follow-up, causal impact estimates indicate that PGC-HS participants were less likely than control participants to ever have had vaginal sex. PGC-HS participants also scored higher on decision-making skills and perceived connection to peer connectedness. Conclusions: Results suggest that by building social and emotional skills and helping students form supportive peer relationships, PGC-HS may encourage students to make healthier choices and avoid risky behaviors during a critical period in high school, thus, reducing the risk of pregnancy.

Fluorogenic oligonucleotide-based systems are important tools for sequence-specific DNA targeting. Fluorescence systems use the emission of a photon from an excited molecule (fluorophore) to produce a detectable signal. It is a highly sensitive process and its versatility has expanded its use to DNA targeting probes. The aim of the work herein is to investigate the design and synthesis of fluorogenic oligonucleotide-based systems specifically for targeting double-stranded DNA. Triplex-forming oligonucleotides can bind in the major groove of dsDNA through Hoogsteen hydrogen bonding to form triplexes (triple helixes). While other methods of dsDNA detection rely on denaturation of the double-stranded target, triplexes do not, and, hence, they provide a versatile approach to DNA recognition. However, the instability of triplexes, especially at neutral pH, limits their applications. The incorporation of a fluorogenic intercalator, thiazole orange, covalently attached to the TFO provides a triplex-stabilising moiety. The stabilising effect of thiazole orange is additive; multiple incorporations can produce triplexes with increased stability of +50 °C even at neutral pH. The extreme stability provided from the modified fluorogenic TFOs allows increased duplex binding affinity and can compensate for the limited target range of TFOs. CRISPR-Cas is a programmable biological research tool that is invaluable in gene- editing and genomic imaging technologies. The introduction of fluorogenic moieties both on the dCas9 enzyme and in guide RNA is crucial for imaging but often leads to high background fluorescence in biological systems. By modifying the gRNA to contain an 'on/off' fluorogenic system, genomic imaging in live cells of repetitive elements is improved and is cross-validated by GFP-tagged dCas9. The efficient synthesis of long, modified RNA is fundamental, particularly in CRISPR research. The introduction of an azide moiety into RNA to provide a means of 'clicking' modified RNA strands together is an important potential approach. However, it is limited by the protecting groups used for RNA and, in turn, this hinders the length of efficient RNA synthesis. The synthesis of a clickable moiety compatible with 2'-O-TC RNA chemistry is investigated as it could provide a simple and cost- effective route to modified guide RNA and could be instrumental in applications such as modified guide RNA library generation. Overall, the aims of this study were to synthesise and develop fluorogenic oligonucleotide-based systems and investigate their potential applications.

This article discusses the diagnosis and treatment of Drug Reaction with Eosinophilia and Systemic Symptoms (DRESS) syndrome, a rare and severe drug hypersensitivity reaction. The incidence of DRESS is low, ranging from two to five cases per million per year, with a mortality rate of 5-10%. The diagnosis of DRESS is challenging due to its complex clinical presentation and overlap with other conditions. The article provides a diagnostic algorithm and treatment recommendations based on a comprehensive literature review and expert consensus. Corticosteroids are the mainstay of treatment, and second-line therapies may be considered for refractory cases. Prospective clinical trials are needed to further validate treatment approaches. This article examines the Roto Chileno war memorial in downtown Santiago as a complex site of Chilean identity making in the republican era. Conflating two separate nineteenth-century military victories over the combined forces of Bolivia and Peru, the memorial was a physical manifestation of an intricate process of identity construction that celebrated the mestizo heritage of average citizen-soldiers while also emphasizing the idea that Chileans were racially superior compared to other South American nations. The memorial being dedicated specifically to the roto, a mutable racialized term that can be perceived as pejorative even in the present, is therefore an especially evocative example of how this conceptual combination was visually rendered to access elements of white identity. Using fine art media coverage, this article highlights the entangled relationship between memory, nationalism, and race in late nineteenth-century Latin America to consider the role that militarism and masculinity play in the construction of whiteness. It has recently become clear that some language-specific traits previously thought to be unique to humans (such as the capacity to combine sounds) are widespread in the animal kingdom. Despite the increase in studies documenting the presence of call combinations in non-human animals, factors promoting this vocal trait are unclear. One leading hypothesis proposes that communicative complexity co-evolved with social complexity owing to the need to transmit a diversity of information to a wider range of social partners. The Western Australian magpie (Gymnorhina tibicen dorsalis) provides a unique model to investigate this proposed link because it is a group-living, vocal learning species that is capable of multi-level combinatoriality (independently produced calls contain vocal segments and comprise combinations). Here, we compare variations in the production of call combinations across magpie groups ranging in size from 2 to 11 birds. We found that callers in larger groups give call combinations: (i) in greater diversity and (ii) more frequently than callers in smaller groups. Significantly, these observations support the hypothesis that combinatorial complexity may be related to social complexity in an open-ended vocal learner, providing an important step in understanding the role that sociality may have played in the development of vocal combinatorial complexity. Higher education policies regarding COVID-19 had sweeping implications for campuses and their surrounding communities. This study explored the role of relevant health and health policy expertise in shaping campus responses to COVID-19 using data from 1,983 4-year colleges and universities in the United States. The primary outcomes of interest were the various COVID-19 policies that postsecondary institutions adopted during the 2020-2021 academic year, including instructional modality and testing and vaccine requirements. Overall, after controlling for state-level politics and other factors, the presence of faculty expertise in health administration, public health, or medicine had a statistically significant influence on the COVID policies adopted by colleges and universities. Public health faculty in conjunction with health administration faculty influenced changes in instruction modality between semesters. Public health faculty in conjunction with medical faculty influenced changes in testing policies between semesters. Objective: Male allies play an important role in sexual assault prevention, yet many college sexual assault prevention programs struggle to recruit and retain men. This study aims to identify barriers to recruitment and retention of male sexual assault prevention peer educators. Participants: Seventeen undergraduate male student leaders participated in this study during summer 2018. Methods: Semi-structured phone interviews were conducted, recorded, and transcribed. Response data were thematically analyzed. Results: Barriers to recruitment include perceived gender norms and discomfort with the topic of sexual assault. Barriers to retention include male peer educators' perception that women are resistant to men discussing sexual assault. Suggestions for improving recruitment and retention efforts are also identified. Conclusions: Recruitment and retention of male sexual assault prevention peer educators may require recruitment approaches that are tailored to men and programmatic changes that position men as allies in sexual assault prevention. Using early twentieth century Chile as a case study, this article examines the interaction between Catholicism and eugenic social reform in Latin America. Examining articles that appeared in popular and Catholic periodicals between 1891 and 1940, it argues that both Catholic and secular eugenicists advocated for the modernization of patriarchal social structures to protect the racial health of the Chilean population. The article therefore illustrates how Latin American eugenics, though less concerned with biological heritage than its North American counterparts, still supported the notion that organizing societies around gender difference was scientifically sound. Introduction: Etranacogene dezaparvovec gene therapy for haemophilia B demonstrated superior efficacy at 24 months in reducing bleeds versus a ≥6‐month lead‐in period of prophylaxis with FIX products in the phase 3 trial, HOPE‐B. In the absence of head‐to‐head comparisons of etranacogene dezaparvovec versus FIX products, indirect treatment comparisons (ITC) can be used. Aim: To compare the efficacy of etranacogene dezaparvovec versus rIX‐FP, rFIXFc and N9‐GP using ITC, and support HOPE‐B results. Methods: Data were leveraged from Phase 3 pivotal trials: HOPE‐B, PROLONG‐9FP, B‐LONG and Paradigm 2. Annualised bleeding rates (ABR), spontaneous (AsBR) and joint (AjBR) bleeding rates, percentage of patients with no bleeds, and FIX consumption were assessed using inverse probability of treatment weighting and matching adjusted indirect comparisons. Results: Etranacogene dezaparvovec demonstrated statistically significantly lower bleeding rates versus all comparators. Rate ratios for ABR, AsBR and AjBR versus rIX‐FP were 0.19 (p <.0001), 0.08 (p <.0001) and 0.09 (p <.0001), respectively. Rate ratios for ABR, AsBR and AjBR versus rFIXFc were 0.14 (p <.0001), 0.13 (p =.0083) and 0.15 (p =.0111), respectively. Rate ratios for ABR and AsBR, versus N9‐GP were 0.24 (p =.0231) and 0.13 (p =.0071), respectively. Etranacogene dezaparvovec demonstrated significantly higher percentage of patients with no bleeds versus rIX‐FP and rFIXFc; odds ratios: 17.60 (p <.0001) and 5.65 (p =.0037), respectively. Etranacogene dezaparvovec resulted in significantly lower FIX consumption than all comparators. Conclusions: ITC suggests that etranacogene dezaparvovec offers patients with haemophilia B (≤2% of normal FIX expression) a single dose treatment that can significantly reduce bleeding rates and eliminate routine infusions associated with FIX therapies. The objectives of this study were to investigate the effect of level and timing of silage supplementation during early lactation on animal performance and dry matter intake (DMI). Two farm-lets were established with a high (1253 kg DM/ha) and low (862 kg DM/ha) grass availability at turnout. In spring, cows were assigned to one of two treatments as they calved over 2 years; high grass (HG) and low grass (LG). During period 1 (week 1–6), cows on the HG treatment were offered a high daily herbage allowance (DHA) with low silage and the LG treatment were offered a low DHA with high silage. In period 2 (week 7–12), half of the cows from the HG treatment in P1 switched to the LG treatment in P2 and vice versa as 20 LG cows in P1 switched to the HG treatment in P2. Cows on the HG treatment in P2 received a high DHA with no silage and the LG treatment received a low DHA with 3 kg DM/cow silage. Grass DMI was significantly higher for the HG treatment during both periods (+1.6 and +3.4 kg DM/cow/day, respectively). The HG treatment produced +0.9 kg milk/cow/day and had a higher protein concentration (+1.1 g/kg milk) compared to cows on the LG treatment during period 2. Virus host shifts, where a virus transmits to and infects a novel host species, are a major source of emerging infectious disease. Genetic similarity between eukaryotic host species has been shown to be an important determinant of the outcome of virus host shifts, but it is unclear if this is the case for prokaryotes where anti-virus defences can be transmitted by horizontal gene transfer and evolve rapidly. Here, we measure the susceptibility of 64 strains of Staphylococcaceae bacteria (48 strains of Staphylococcus aureus and 16 non-S. aureus species spanning 2 genera) to the bacteriophage ISP, which is currently under investigation for use in phage therapy. Using three methods-plaque assays, optical density (OD) assays, and quantitative (q)PCR-we find that the host phylogeny explains a large proportion of the variation in susceptibility to ISP across the host panel. These patterns were consistent in models of only S. aureus strains and models with a single representative from each Staphylococcaceae species, suggesting that these phylogenetic effects are conserved both within and among host species. We find positive correlations between susceptibility assessed using OD and qPCR and variable correlations between plaque assays and either OD or qPCR, suggesting that plaque assays alone may be inadequate to assess host range. Furthermore, we demonstrate that the phylogenetic relationships between bacterial hosts can generally be used to predict the susceptibility of bacterial strains to phage infection when the susceptibility of closely related hosts is known, although this approach produced large prediction errors in multiple strains where phylogeny was uninformative. Together, our results demonstrate the ability of bacterial host evolutionary relatedness to explain differences in susceptibility to phage infection, with implications for the development of ISP both as a phage therapy treatment and as an experimental system for the study of virus host shifts.

BACKGROUND: Single-center studies have shown that single photon emission computed tomography myocardial blood flow (MBF) measurement is accurate compared with MBF measured with microspheres in a porcine model, positron emission tomography, and angiography. Clinical implementation requires consistency across multiple sites. The study goal is to determine the intersite processing repeatability of single photon emission computed tomography MBF and the additional camera time required. METHODS: Five sites (Canada, Italy, Japan, Germany, and Singapore) each acquired 25 to 35 MBF studies at rest and with pharmacological stress using technetium-99m-tetrofosmin on a pinhole-collimated cadmium-zinc-telluride–based cardiac single photon emission computed tomography camera with standardized list-mode imaging and processing protocols. Patients had intermediate to high pretest probability of coronary artery disease. MBF was measured locally and at a core laboratory using commercially available software. The time a room was occupied for an MBF study was compared with that for a standard rest/stress myocardial perfusion study. With motion correction, the overall correlation in MBF between core laboratory and local site was 0.93 (range, 0.87–0.97) at rest, 0.90 (range, 0.84–0.96) at stress, and 0.84 (range, 0.70–0.92) for myocardial flow reserve. The local-to-core difference in global MBF (bias
-MBF ) was 5.4% (−3.8% to 14.8%; median [interquartile range]) at rest and 5.4% (−6.2% to 19.4%) at stress. Between the 5 sites, bias-MBF ranged from −1.6% to 11.0% at rest and from −1.9% to 16.3% at stress; the interquartile range in bias-MBF was between 9.3% (4.8%–14.0%) and 22.3% (−10.3% to 12.0%) at rest and between 17.0% (−11.3% to 5.6%) and 33.3% (−10.4% to 22.9%) at stress and was not significantly different between most sites. Both bias and interquartile range were like previously reported interobserver variability and less than the SD of the test-retest difference of 30%. The overall difference in myocardial flow reserve was 1.52% (−10.6% to 11.3%). There were no significant differences between with and without motion correction. The average additional acquisition time varied between sites from 44 to 79 minutes. CONCLUSIONS: The average bias-MBF and bias-MFR values were small with standard deviations substantially less than the test-retest variability. This demonstrates that MBF can be measured consistently across multiple sites and further supports that this technique can be reliably implemented. [ABSTRACT FROM AUTHOR]- Published

Background: Frailty and sarcopenia are common age-related conditions associated with adverse outcomes. Physical activity has been identified as a potential preventive strategy for both frailty and sarcopenia. The authors aimed to investigate the association between physical activity and prevention of frailty and sarcopenia in people aged 65 years and older. Methods: The authors searched for systematic reviews (January 2008 to November 2019) and individual studies (January 2010 to March 2020) in PubMed. Eligible studies were randomized controlled trials and longitudinal studies that investigated the effect of physical activity on frailty and/or sarcopenia in people aged 65 years and older. The Grading of Recommendations Assessment, Development and Evaluation approach was used to rate certainty of evidence. Results: Meta-analysis showed that physical activity probably prevents frailty (4 studies; frailty score pooled standardized mean difference, 0.24; 95% confidence interval, 0.04–0.43; P =.017, I
2 = 57%, moderate certainty evidence). Only one trial investigated physical activity for sarcopenia prevention and did not provide conclusive evidence (risk ratio 1.08; 95% confidence interval, 0.10–12.19). Five observational studies showed positive associations between physical activity and frailty or sarcopenia prevention. Conclusions: Physical activity probably prevents frailty among people aged 65 years and older.
