Abstract The architectural heritages of the Peking-Mukden Railway has witnessed the industrialization of China. When it no longer serves its original function, finding a new way for reuse becomes a trend. It requires feedback from reused cases to guide decision-making. This article aims to propose an approach to post-occupancy evaluation of architectural heritages in the Peking-Mukden Railway and apply it to real cases for analysis. The evaluation indicator system used here consists of a hierarchical structure, evaluation indicators, and interpretation indicators. Cosine similarity, entropy weight method, and precedence chart method are utilized to determine final weights. The GRA-TOPSIS method is utilized to identify reuse cases that require additional optimization. The conclusions are as follows: 1. The cosine similarity can be introduced to neutralize individual preferences. The entropy weight method can be used to neutralize subjective factors. 2. Based on the analysis of the sensitivity levels, indicators that need to be improved first can be identified. Priority should be given to spatial accessibility and cultural transmission when making adjustments with limited resources. 3. The GRA-TOPSIS evaluation method can be used to rank and find reuse cases that need further optimization.

Abstract Phytochrome interacting factors (PIFs) serve as crucial regulators in the light signal transduction pathway and also mediate light signals to regulate secondary metabolite synthesis in plants. However, the regulator role of PIFs in secondary metabolites often varies among different plants. Isorhynchophylline (IRN), an iconic secondary metabolite of Uncaria rhynchophylla, holds significant medicinal value. Low light induces the synthesis of IRN in previous research, but PIFs in U. rhynchophylla have not been studied to date. Building on this, we identified a PIF protein, UrPIF3, which possesses the typical conserved domains of the PIFs and is localized in the nucleus. Moreover, the expression level of UrPIF3 is consistently positively correlated with the expression of two key enzyme genes (UrSGD and UrSTR) in the IRN biosynthesis pathway, regardless of whether under low light or restoring light conditions. Yeast one-hybrid and dual-luciferase assays further demonstrated that UrPIF3 can directly upregulate UrSGD. Conversely, silencing UrPIF3 inhibits IRN synthesis, and significantly reduces the expression levels of UrSGD and UrSTR. In summary, our results suggest that under low light conditions, UrPIF3 can directly upregulate UrSGD and indirectly upregulate UrSTR, thereby promoting the synthesis of IRN.

Abstract Iron is an essential biomineral in the human body. Here, we describe a subset of iron-loaded cancer-associated fibroblasts, termed as FerroCAFs, that utilize iron to induce immunosuppression in prostate cancer and predict an unfavorable clinical outcome. FerroCAFs secrete myeloid cell-associated proteins, including CCL2, CSF1 and CXCL1, to recruit immunosuppressive myeloid cells. We report the presence of FerroCAFs in prostate cancer from both mice and human, as well as in human lung and ovarian cancers, and identify a conserved cell surface marker, the poliovirus receptor. Mechanistically, the accumulated iron in FerroCAFs is caused by Hmox1-mediated iron release from heme degradation. The intracellular iron activates the Kdm6b, an iron-dependent epigenetic enzyme, to induce an accessible chromatin state and transcription of myeloid cell-associated protein genes. Targeting the FerroCAFs by inhibiting the Hmox1/iron/Kdm6b signaling axis incurs anti-tumor immunity and tumor suppression. Collectively, we report an iron-loaded FerroCAF cluster that drives immunosuppression through an iron-dependent epigenetic reprogramming mechanism and reveal promising therapeutic targets to boost anti-tumor immunity.

Abstract We examine the accretion process in a thin disk surrounding a supermassive black hole within the framework of Einstein–Maxwell-scalar (EMS) gravity. Our investigation aims to elucidate how variations in model parameters affect different physical properties of the disk. When keeping EMS parameters $$\beta $$ β and q constant, we observe a reduction in radiation flux and temperature as $$\alpha $$ α increases. However, the luminosity and radiative efficiency exhibit relatively minor variation. Conversely, under fixed $$\alpha $$ α and q, an escalation in $$\beta $$ β leads to heightened levels of radiation flux, temperature, luminosity, and radiative efficiency. These results underscore the diverse influences of model parameters on observable metrics, providing valuable insights for the astronomical study of distinct black holes.

Abstract Langerhans cells (LCs) are resident dendritic cells in the epidermis and their roles in presenting antigens derived from microorganisms present in the skin has been well appreciated. However, it is generally thought that incoming neutrophils are mainly responsible for eradicating invading pathogens in the early stage of wounds and a role of LCs in innate immunity is elusive. In the current study, we showed that wounds absent of LCs had a delayed closure. Mechanistically, LCs were the primary cells in warding off bacteria invasion at the early stage of wound healing. Without LCs, commensal bacteria quickly invaded and propagated in the wounded area. keratinocytes surrounding the wounds responded to the excessive bacteria by elevated production of CXCL5, resulting in an over-influx of neutrophils. The over-presence of activated neutrophils, possibly together with the aggravated invasion of bacteria, was detrimental to epidermal progenitor cell propagation and re-epithelialization. These observations underscore an indispensable role of LCs as effective guardians that preclude both bacteria invasion and damages inflicted by secondary inflammation.

A comprehensive understanding of the diverse value of industrial heritage is a prerequisite for preservation and reuse. After studying the composition and evaluation methods of the value of world cultural heritage, this paper puts forward a mixed decision model based on the combination of interval analytic hierarchy process, expert clustering ranking weighting method, fuzzy set and D-S theory. Firstly, an evaluation system that can describe the value of modern silk industry heritage in Hangzhou-Jiaxing-Huzhou area is established, which reflects comprehensiveness and particularity. Then, the interval analytic hierarchy process and expert clustering ranking method are proposed to calculate the weight of various indicators in the evaluation system, which can effectively reduce the influence of subjective experience and extreme experts on group decision-making. In addition, the fuzzy set theory and D-S theory are used to calculate the protection grades of several modern silk industrial heritages, and compared with the actual protection grades recognized by the state, which verifies the scientificity and effectiveness of the mixed decision-making model. Finally, the results show that the mixed decision model can accurately calculate the protection level consistent with the reality, demonstrating high feasibility and providing new ideas for the future evaluation of industrial heritage values.

Abstract Background Obesity and metabolic syndrome (MetS) have become urgent worldwide health problems, predisposing patients to unfavorable myocardial status and thyroid dysfunction. Low-carbohydrate diet (LCD) and time-restricted eating (TRE) have been confirmed to be effective methods for weight management and improving MetS, but their effects on the myocardium and thyroid are unclear. Methods We conducted a secondary analysis in a randomized clinical diet-induced weight-loss trial. Participants (N = 169) diagnosed with MetS were randomized to the LCD group, the 8 h TRE group, or the combination of the LCD and TRE group for 3 months. Myocardial enzymes and thyroid function were tested before and after the intervention. Pearson’s or Spearman’s correlation was assessed between functions of the myocardium and thyroid and cardiometabolic parameters at baseline. Results A total of 162 participants who began the trial were included in the intention-to-treat (ITT) analysis, and 57 participants who adhered to their assigned protocol were involved in the per-protocol (PP) analysis. Relative to baseline, lactate dehydrogenase, creatine kinase MB, hydroxybutyrate dehydrogenase, and free triiodothyronine (FT3) declined, and free thyroxine (FT4) increased after all 3 interventions (both analyses).
