Gravitational wave high-energy Electromagnetic Counterpart All-sky Monitor (GECAM) is a constellation with four instruments (launch date): GECAM-A/B (10 December 2020), GECAM-C (27 July 2022) and GECAM-D (13 March 2024), which are dedicated to monitoring gamma-ray transients in all-sky. The primary science objectives of GECAM include Gamma-Ray Bursts (GRBs), Soft Gamma-ray Repeaters (SGRs), high energy counterparts of Gravitation Wave (GW) and Fast Radio Burst (FRB), Solar Flares (SFLs), as well as Terrestrial Gamma-ray Flashes (TGFs) and Terrestrial Electron Beams (TEBs). A series of observations and research have been made since the launch of GECAM-A/B. GECAM observations provide new insights into these high-energy transients, demonstrating the unique role of GECAM in the “multi-wavelength, multi-messenger” era.

Abstract Atherosclerosis is a chronic inflammatory disease characterized with innate and adaptive immunity but also involves pyroptosis. Few studies have explored the role of pyroptosis in advanced atherosclerotic plaques from different vascular beds. Here we try to identify the different underlying function of pyroptosis in the progression of atherosclerosis between carotid arteries and femoral. arteries. We extracted gene expression levels from 55 advanced carotid or femoral atherosclerotic plaques. The pyroptosis score of each sample was calculated by single-sample-gene-set enrichment analysis (ssGSEA). We then divided the samples into two clusters: high pyroptosis scores cluster (PyroptosisScoreH cluster) and low pyroptosis scores cluster (PyroptosisScoreL cluster), and assessed functional enrichment and immune cell infiltration in the two clusters. Key pyroptosis related genes were identified by the intersection between results of Cytoscape and LASSO (Least Absolute Shrinkage and Selection Operator) regression analysis. Finally, all key pyroptosis related genes were validated in vitro. We found all but one of the 29 carotid plaque samples belonged to the PyroptosisScoreH cluster and the majority (19 out of 26) of femoral plaques were part of the PyroptosisScoreL cluster. Atheromatous plaque samples in the PyroptosisScoreL cluster had higher proportions of gamma delta T cells, M2 macrophages, myeloid dendritic cells (DCs), and cytotoxic lymphocytes (CTLs), but lower proportions of endothelial cells (ECs). Immune full-activation pathways (e.g., NOD-like receptor signaling pathway and NF-kappa B signaling pathway) were highly enriched in the PyroptosisScoreH cluster. The key pyroptosis related genes GSDMD, CASP1, NLRC4, AIM2, and IL18 were upregulated in advanced carotid atherosclerotic plaques. We concluded that compared to advanced femoral atheromatous plaques, advanced carotid atheromatous plaques were of higher grade of pyroptosis. GSDMD, CASP1, NLRC4, AIM2, and IL18 were the key pyroptosis related genes, which might provide a new sight in the prevention of fatal strokes in advanced carotid atherosclerosis.

Polyurea coating and carbon fibre reinforced polymer reinforcing techniques were applied to retrofit a severely damaged urban utility tunnel (UUT). The blast responses of the retrofitted UUT were investigated through in-filed explosion experimental tests, and the displacements, strains, accelerations, and damage were compared. The retrofitted UUT exhibited comparable or even better blast resistance than the intact UUT. Although the polyurea coating was flexible, it exhibited excellent performance in improving the blast resistance of the damaged UUT. As the UUT reinforced with basalt fibre reinforce polymer (BFRP) bars has smaller damage compared with the UUT reinforced with steel bars, its retrofitting is simple, and the cost is low. The roof was simplified as an elastically simple support one-way slab. Euler beam theory was adopted to analyse the dynamic responses of UUT roof considering the interaction between soil and structure, which agree well with the experiments in the first three cases.

With the increasing awareness of health, using wearable sensors to monitor individual activities and accurately estimate energy expenditure has become a current research focus. However, existing research encounters challenges including low estimation accuracy, a deficiency of frequency domain features, and difficulty in integrating time domain and frequency domain features. To address these issues, we propose an innovative framework called the Dual-Stream Fusion Network (DSFN). This framework combines the Time Domain Encoding (TDE) module, the Frequency Domain Hierarchical-Split Encoding (FDHSE) module, and a Two-Stage Feature Fusion (TSF) module. Specifically, the temporal stream of the framework employs the TDE module to capture deep temporal features that reflect the complex dynamic variations in time-series data. The frequency domain stream introduces the FDHSE module, which extracts frequency domain features using a multi-level, multi-scale approach, ensuring a comprehensive and diverse representation of frequency information. Through this dual-stream architecture, our model effectively learns both time and frequency domain features, addressing the limitations of frequency domain features observed in prior studies. Additionally, we propose the TSF module to fully integrate time and frequency domain features, effectively overcoming the challenge of fusing these two types of features. We conducted experiments on two public datasets, namely the GOTOV dataset (elderly people) and the JSI dataset (young people). Experimental results demonstrate that our method achieves excellent performance across different age groups. Background: Cyanobacterial harmful algal blooms (CyanoHABs) originate from the excessive growth or bloom of cyanobacteria often referred to as blue-green algae. They have been on the rise globally in both marine and freshwaters in recently years with increasing frequency and severity owing to the rising temperature associated with climate change and increasing anthropogenic eutrophication from agricultural runoff and urbanization. Humans are at a great risk of exposure to toxins released from CyanoHABs through drinking water, food, and recreational activities, making CyanoHAB toxins a new class of contaminants of emerging concern. Objectives: We investigated the toxic effects and mechanisms of microcystin-LR (MC-LR), the most prevalent CyanoHAB toxin, on the ovary and associated reproductive functions. Methods: Mouse models with either chronic daily oral or acute intraperitoneal exposure, an engineered three-dimensional ovarian follicle culture system, and human primary ovarian granulosa cells were tested with MC-LR of various dose levels. Single-follicle RNA sequencing, reverse transcription-quantitative polymerase chain reaction, enzyme-linked immunosorbent assay, western blotting, immunohistochemistry (IHC), and benchmark dose modeling were used to examine the effects of MC-LR on follicle maturation, hormone secretion, ovulation, and luteinization. Results: Mice exposed long term to low-dose MC-LR did not exhibit any differences in the kinetics of folliculogenesis, but they had significantly fewer corpora lutea compared with control mice. Superovulation models further showed that mice exposed to MC-LR during the follicle maturation window had significantly fewer ovulated oocytes. IHC results revealed ovarian distribution of MC-LR, and mice exposed to MC-LR had significantly lower expression of key follicle maturation mediators. Mechanistically, in both murine and human granulosa cells exposed to MC-LR, there was reduced protein phosphatase 1 (PP1) activity, disrupted PP1-mediated PI3K/AKT/FOXO1 signaling, and less expression of follicle maturation-related genes. Discussion: Using both in vivo and in vitro murine and human model systems, we provide data suggesting that environmentally relevant exposure to the CyanoHAB toxin MC-LR interfered with gonadotropin-dependent follicle maturation and ovulation. Identifying biomarkers for abdominal aortic aneurysms (AAA) is key to understanding their pathogenesis, developing novel targeted therapeutics, and possibly improving patients outcomes and risk of rupture. Here, we identified AAA biomarkers from public databases using single-cell RNA-sequencing, weighted co-expression network (WGCNA), and differential expression analyses. Additionally, we used the multiple machine learning methods to identify biomarkers that differentiated large AAA from small AAA. Biomarkers were validated using GEO datasets. CIBERSORT was used to assess immune cell infiltration into AAA tissues and investigate the relationship between biomarkers and infiltrating immune cells. Therefore, 288 differentially expressed genes (DEGs) were screened for AAA and normal samples. The identified DEGs were mostly related to inflammatory responses, lipids, and atherosclerosis. For the large and small AAA samples, 17 DEGs, mostly related to necroptosis, were screened. As biomarkers for AAA, G0/G1 switch 2 (G0S2) (Area under the curve [AUC] = 0.861, 0.875, and 0.911, in GSE57691, GSE47472, and GSE7284, respectively) and for large AAA, heparinase (HPSE) (AUC = 0.669 and 0.754, in GSE57691 and GSE98278, respectively) were identified and further verified by qRT-PCR. Immune cell infiltration analysis revealed that the AAA process may be mediated by T follicular helper (Tfh) cells and the large AAA process may also be mediated by Tfh cells, M1, and M2 macrophages. Additionally, G0S2 expression was associated with neutrophils, activated and resting mast cells, M0 and M1 macrophages, regulatory T cells (Tregs), resting dendritic cells, and resting CD4 memory T cells. Moreover, HPSE expression was associated with M0 and M1 macrophages, activated and resting mast cells, Tregs, and resting CD4 memory T cells. Additional, G0S2 may be an effective diagnostic biomarker for AAA, whereas HPSE may be used to confer risk of rupture in large AAAs. Immune cells play a role in the onset and progression of AAA, which may improve its diagnosis and treatment.

To improve corrosion-resistance of shallow-buried concrete urban utility tunnels (UUTs), basalt fiber reinforced polymer (BFRP) bars are applied to reinforce UUTs. As the UUT must have excellent survival capability under accidental explosions, a shallow-buried BFRP bars reinforced UUT (BBRU) was designed and constructed. Repetitive blast experiments were carried out on this BBRU. Dynamic responses, damage evolutions and failure styles of the BBRU under repetitive explosions were revealed. The tunnel roof is the most vulnerable component and longitudinal cracks develop along the tunnel. When the scaled distance is larger than 1.10 m/kg1/3, no cracks are observed in the experiments. When the BBRU is severely damaged, there are five cracks forming and developing along the roof. The roof is simplified as a clamped-supported one-way slab, proved by the observation that the maximum strain of the transverse bar is much larger than that of the longitudinal bar. Dynamic responses of the roof slab are predicted by dynamic Euler beam theory, which can consistently predict the roof displacement under large-scaled-distance explosion. Compared with the UUT reinforced with steel bars, the BBRU has advantages in blast resistance with smaller deflections and more evenly-distributed cracks when the scaled distance is smaller than 1.260 m/kg1/3 and the steel bars enter plastic state. Longer elastic defamation of the BFRP bars endows the UUT more excellent blast resistance under small-scaled-distance explosions.

AIM: To study the effect of zymosan, a ligand found on the surface of fungi, on gap junctional intercellular communication (GJIC) in cultured human corneal fibroblasts (HCFs). METHODS: Zymosan was added to the medium of cultured HCFs with or without the administration of mitogen-activated protein kinase (MAPK) inhibitors or the inhibitor kappa B kinase 2 (IKK2) inhibitor IV. The protein and mRNA levels of connexin 43 (Cx43) in HCFs were measured by Western blot, immunofluorescence, and quantitative reverse transcription-polymerase chain reaction (qRT-PCR) analyses. The GJIC activity was tested using a dye-coupling assay. RESULTS: The reduction of Cx43 protein and mRNA levels as well as a significant decrease in GJIC activity were observed in cultured HCFs when zymosan was added into the culture medium.
