Abstract Dental diseases such as caries and periodontitis have been common public health problems. Dental disease treatment can be achieved through stem cell-based dental regeneration. Biophysical cues determine the fate of stem cells and govern the success of dental regeneration. Some studies have manifested exosomes derived from stem cells could not only inherit biophysical signals in microenvironment but also evade some issues in the treatment with stem cells. Nowadays, biophysical cue-regulated exosomes become another promising therapy in dental regenerative medicine. However, methods to improve the efficacy of exosome therapy and the underlying mechanisms are still unresolved. In this review, the association between biophysical cues and dental diseases was summarized. We retrospected the role of exosomes regulated by biophysical cues in curing dental diseases and promoting dental regeneration. Our research also delved into the mechanisms by which biophysical cues control the biogenesis, release, and uptake of exosomes, as well as potential methods to enhance the effectiveness of exosomes. The aim of this review was to underscore the important place biophysical cue-regulated exosomes occupy in the realm of dentistry, and to explore novel targets for dental diseases.

Purpose Dentin hypersensitivity (DH) is a prevalent condition, but long-term effective treatments are scarce. Differentiation of odontoblast-like cells is promising for inducing tertiary dentinogenesis and ensuring sustained therapeutic efficacy against DH. This study examined the effects and mechanism of action of mild heat stress (MHS) on the differentiation of odontoblast-like MDPC-23 cells.Methods We used a heating device to accurately control the temperature and duration, mimicking the thermal microenvironment of odontoblast-like cells. Using this device, the effects of MHS on cell viability and differentiation were examined. Cell viability was assessed using the MTT assay. The expression and nucleoplasmic ratio of the yes-associated protein (YAP) were examined by western blotting and immunofluorescence. The gene expression levels of heat shock proteins (HSPs) and dentin matrix protein-1 (DMP1) were measured using qPCR. Dentin sialophosphoprotein (DSPP) expression was evaluated using immunofluorescence and immunoblotting. Verteporfin was used to inhibit YAP activity.Results Mild heat stress (MHS) enhanced the odontoblast differentiation of MDPC-23 cells while maintaining cell viability. MHS also increased YAP activity, as well as the levels of HSP25 mRNA, HSP70 mRNA, HSP90α mRNA, DMP1 mRNA, and DSPP protein. However, after YAP inhibition, both cell viability and the levels of HSP90α mRNA, DMP1 mRNA, and DSPP protein were reduced.Conclusion YAP plays a crucial role in maintaining cell viability and promoting odontoblast differentiation of MDPC-23 cells under MHS. Consequently, MHS is a potential therapeutic strategy for DH, and boosting YAP activity could be beneficial for maintaining cell viability and promoting odontoblast differentiation.

In-situ XRD, 31P NMR and 23Na NMR were used to analyze the interaction behavior of Na3V2(PO4)3 at low voltage, and then a new intercalation model was proposed. During the transition from Na3V2(PO4)3 to Na4V2(PO4)3, Na ions insert into M1, M2 and M3 sites simultaneously. Afterwards, during the transition of Na4V2(PO4)3 to Na5V2(PO4)3, Na ions mainly insert into M3 site.

Introduction: Urbanization has affected the quality of the living environments. It is important to improve residents’ living environments and promote their happiness.Methods: We use a national and representative dataset targeting the labor force in China, collecting basic information about the work environments, the social environments, and the urban environments. This work employed a linear regression model to investigate the influencing factors of residents’ SWB.Results: The three major findings are as follows: 1) At the national level, residents’ SWB exhibits a notable spatial variability, being higher in the northern regions and lower in the southern areas. 2) The dimensions of the urban environments (housing prices, POI density, NPP, land development intensity and the number of tertiary hospitals), the work environments (work pressure and job satisfaction), and the social environments (community trust, social justice and sense of security), along with sociodemographic characteristics, significantly influence SWB. 3) In China, the impacts on SWB exhibit pronounced regional heterogeneity. The relationship between environmental pollution and SWB is characterized by an inverted U-shaped pattern. Intriguingly, while housing prices negatively affect SWB in the eastern and central regions, the enforcement of housing purchase limits has been observed to enhance residents’ SWB in the western region.Discussion: First, this work show the overall spatial level of SWB in China geographically. And second, this research found the SWB heterogeneity on a regional level (eastern, central, and western regions). Furthermore, this methodological framework provides a novel perspective among the urban, work and social environments on SWB. This work also contributes to inform policy to improve residents’ SWB in China.

Accurately extracting pixel-level buildings from high-resolution remote sensing images is significant for various geographical information applications. Influenced by different natural, cultural, and social development levels, buildings may vary in shape and distribution, making it difficult for the network to maintain a stable segmentation effect of buildings in different areas of the image. In addition, the complex spectra of features in remote sensing images can affect the extracted details of multi-scale buildings in different ways. To this end, this study selects parts of Xi’an City, Shaanxi Province, China, as the study area. A parallel encoded building extraction network (MARS-Net) incorporating multiple attention mechanisms is proposed. MARS-Net builds its parallel encoder through DCNN and transformer to take advantage of their extraction of local and global features. According to the different depth positions of the network, coordinate attention (CA) and convolutional block attention module (CBAM) are introduced to bridge the encoder and decoder to retain richer spatial and semantic information during the encoding process, and adding the dense atrous spatial pyramid pooling (DenseASPP) captures multi-scale contextual information during the upsampling of the layers of the decoder. In addition, a spectral information enhancement module (SIEM) is designed in this study. SIEM further enhances building segmentation by blending and enhancing multi-band building information with relationships between bands. The experimental results show that MARS-Net performs better extraction results and obtains more effective enhancement after adding SIEM. The IoU on the self-built Xi’an and WHU building datasets are 87.53% and 89.62%, respectively, while the respective F1 scores are 93.34% and 94.52%.

Identifying the grade of Gastrodia elata in the market has low efficiency and accuracy. To address this issue, an I-YOLOX object detection algorithm based on deep learning and computer vision is proposed in this paper. First, six types of Gastrodia elata images of different grades in the Gastrodia elata planting cooperative were collected for image enhancement and labeling as the model training dataset. Second, to improve feature information extraction, an ECA attention mechanism module was inserted between the backbone network CSPDarknet and the neck enhancement feature extraction network FPN in the YOLOX model. Then, the impact of the attention mechanism and application position on model improvement was investigated. Third, the 3 × 3 convolution in the neck enhancement feature extraction network FPN and the head network was replaced by depthwise separable convolution (DS Conv) to reduce the model size and computation amount. Finally, the EIoU loss function was used to predict boundary frame regression at the output prediction end to improve the convergence speed of the model. The experimental results indicated that compared with the original YOLOX model, the mean average precision of the improved I-YOLOX network model was increased by 4.86% (97.83%), the model computation was reduced by 5.422 M (reaching 3.518 M), the model size was reduced by 20.6 MB (reaching 13.7 MB), and the image frames detected per second increased by 3 (reaching 69). Compared with other target detection algorithms, the improved model outperformed Faster R-CNN, SSD-VGG, YOLOv3s, YOLOv4s, YOLOv5s, and YOLOv7 algorithms in terms of mean average precision, model size, computation amount, and frames per second. The lightweight model improved the detection accuracy and speed of different grades of Gastrodia elata and provided a theoretical basis for the development of online identification systems of different grades of Gastrodia elata in practical production.

BackgroundWe hypothesized that subluxating patellar during minimally invasive total knee arthroplasty (MIS-TKA) would affect intraoperative soft tissue balance and postoperative clinical outcome.MethodsFrom December 2018 to May 2020, 189 patients receiving primary MIS-TKA were enrolled. The gap-balance technique was used, with patients randomly assigned to undergo osteotomy and balance of soft tissue with patella reduced (group A; n = 93) or subluxated (group B; n = 96). The gap and varus?valgus angle were compared between groups in both extension and flexion position. The gap and varus?valgus angle were also compared before and after reducing patellar in group B. Femoral prosthesis rotation, mechanical femoral axis–to–tibial axis angle, Knee Society Score (KSS), visual analog scale (VAS), and range of motion (ROM) were compared postoperatively between two groups. Follow-up was 12 months.ResultsThe flexion gap and the varus angle were significantly greater (0.4 mm and 0.7 degree) after patella reduction than before reduction, but the extension joint gap and varus angle were comparable before and after patella reduction. The femoral prosthesis tended to be internally rotated (0.65 degree) in group B. ROM and VAS was better in the group A than in group B at 1 month after surgery, but the differences were not significant at 3, 6 and 12 months. Under the burgeoning development of urbanization in China, the low-income groups have received attention recently. By applying a linear regression model and utilizing the date from the 2016 China Labor-force Dynamics Survey, this study has explored the effects of urban environments on the community trust in low-income groups, paying particular attention to the difference between local residents and migrants in the Pearl River Delta (PRD). The empirical findings suggest the following: (1) community trust in low-income groups is influenced by social environment dimension, urban space dimension, and sociodemographic characteristics. Specifically, urbanization rate, population density, POl density, land development intensity, social contact, self-rated health, and age have significant effects on the community trust of low-income groups. (2) For local residents, social environment dimension (social contact), urban space dimension (urbanization rate), and sociodemographic characteristics (political status, hukou status, age, and self-rated health) have significant effects on community trust. (3) In the case of migrants, only the sociodemographic characteristics (working in private enterprises or organizations and in agriculture) have a significant impact on community trust. According to the empirical results, the optimization of physical space and social space should consider low-income groups’ needs in livable community planning.

A high‐performance liquid chromatography‐tandem mass spectrometry method has been developed for the determination of irinotecan, 7‐ethyl‐10‐hydroxycamptothecin (SN38), and SN38 glucuronide (SN38G) in rat plasma in the present study. The analytes were separated on a C18 column and a triple‐quadrupole mass spectrometry equipped with an electrospray ionization source was applied for detection. Irinotecan, SN38 and SN38G could be well retained in the C18 column after optimization of mobile phase. A simple protein precipitation was used to pretreat the plasma. The extraction recovery was above 90% and the matrix effect could be negligible. The method was linear over the concentration ranges of 3.46–3458.8 ng/mL for irinotecan, 2.53–2530.0 ng/mL for SN38 and 2.5–2500.0 ng/mL for SN38G. The precision and accuracy was within the acceptable limits. The simple and convenient method was validated and successfully applied to support the pharmacokinetic study and elucidate the mechanism of irinotecan‐induced diarrhea after irinotecan was intravenously injected to the Sprague‐Dawley rats. Osteosarcoma (OS) is recognized as a common malignant tumor with a high trend of metastasis and diffusion. Despite the progresses that have been made in surgery, chemotherapy, and radiotherapy in the recent decades, the prognosis of patients with OS still remains poor. MiRNAs are being increasingly considered as new therapeutic targets for OS treatment. Our research aims to investigate the regulatory impact of miR-92a in the development of OS. Quantitative real-time PCR (qRT-PCR) results revealed that the expression of miR-92a was aberrantly overexpressed in human OS cell lines. By using cell counting kit-8 (CCK-8) assays, colony formation assays, flow cytometric analyses and Transwell assays, our data suggested that up-regulation of miR-92a promoted the proliferation, migration, and invasion of MNNG and U2OS cells, while inhibiting their apoptosis. In contrast, the knockdown of miR-92a effectively reversed these cellular biological behaviors. Furthermore, bioinformatics analysis indicated that Dickkopf-related protein 3 (DKK3) was a possible target of miR-92a. Subsequently, negative regulation of miR-92a on DKK3 was observed, which further supported the direct binding between them. In addition, silencing DKK3 rescued the inhibitory effect of miR-92a inhibitor on the development of OS. To sum up, our study revealed that miR-92a played a carcinogenic role in the growth of OS by promoting the tumorigenesis of OS cells via targeting of DKK3, thus revealing a new therapeutic target for OS. Monocyte chemotactic protein-1 (MCP-1/CCL2) is an important immune factor, which may be important in cancer progression by promoting proliferation, invasion, metastasis and the tumor microenvironment. Previous studies have demonstrated that CCL2 affects the proliferation of osteosarcoma cells via the RANKL signaling pathway. However, the underlying mechanisms remain to be elucidated. To investigate the role of CCL2 in osteosarcoma cells, MTT, spheroid forming, wound healing and transwell assays were performed to examine the proliferation and invasion abilities of the osteosarcoma cells. It was revealed that the high-grade osteosarcoma cells exhibited increased expression levels of CCL2 compared with the low-grade osteosarcoma cells (P<0.001). Furthermore, knockdown of CCL2 decreased the proliferation and invasion abilities of the osteosarcoma cells (P<0.01). These results suggested that the expression of CCL2 is high in high-grade osteosarcoma cells and promotes the proliferation and invasion of osteosarcoma cells. [ABSTRACT FROM AUTHOR]

The objective of this study is to do a meta-analysis of the literature and compare the safety and efficacy of endoscopic carpal tunnel release (ECTR) and open carpal tunnel release (OCTR) for idiopathic carpal tunnel syndrome (CTS). A comprehensive literature search of the electronic databases MEDLINE, EMBASE, Google Scholar, and the Cochrane Controlled Trial Register was undertaken for randomized studies reporting carpal tunnel syndrome treated with ECTR or OCTR. The quality of randomized trials was critically assessed. Pooled relative risk (RR) and 95% confidence intervals (CIs) for safety and efficacy outcome variables were calculated by fixed-effect or random-effect methods with RevMan v.5.1 provided by the Cochrane Collaboration. A total of 13 randomized trials were included by total retrieve and riddling. The results of our meta-analysis showed no significant difference in the overall complication rate (RR = 1.34, 95% CI [0.74, 2.43], P = 0.34), subjective satisfaction (RR = 1.0, 95% CI [0.93, 1.08], P=0.92), time to return to work (mean difference = −3.52 [−8.15, 1.10], P = 0.14), hand grip and pinch strength, and the operative time (mean difference = −1.89, 95% CI [−5.84, 2.06]) between patients in the ECTR and OCTR groups (P = 0.16, 0.70, and 0.35, respectively). The rate of hand pain (RR = 0.73, 95% CI [0.53, 0.93], P = 0.02) in the ECTR group was significantly lower than that in the OCTR group. ECTR treatment seemed to cause more reversible postoperative nerve injuries as compared with OCTR (RR = 2.38, 95% CI [0.98, 5.77], P = 0.05). Although ECTR significantly reduced postoperative hand pain, it increased the possibility of reversible postoperative nerve injury in patients with idiopathic CTS. No statistical difference in the overall complication rate, subjective satisfaction, the time to return to work, postoperative grip and pinch strength, and operative time was observed between the two groups of patients. Background: Osteosarcoma is the most frequent primary malignant bone tumor, notorious for its lung metastasis. Shikonin, an effective constituent extracted from Chinese medicinal herb, was demonstrated to induce necroptosis in some cancers.Methods: MTT assay was performed to detect cell survival rate in vitro. Flow cytometry was used to analyze cell cycle and cell death. Western blot was performed to determine the expression levels of RIP1, RIP3, caspase-3, caspase-6 and PARP. The tibial primary and lung metastatic osteosarcoma models were used to evaluate the anti-tumor effect of shikonin in vivo.Results: The cell survival rate was decreased in a dose and time dependent manner when treated with shikonin. No major change in cell cycle was observed after shikonin treatment. The cell death induced by shikonin could be mostly rescued by specific necroptosis inhibitor necrostatin-1, but not by general caspase inhibitor Z-VAD-FMK. The number of necrotic cells caused by shikonin was decreased after being pretreated with Nec-1 detected by flow cytometry in K7 cells. After 8-hour treatment of shikonin, the expression levels of RIP1 and RIP3 were increased while caspase-3, caspase-6 and PARP were not activated in K7 and U2OS cells determined by Western blot. Size of primary tumor and lung metastasis in shikonin treated group were significantly reduced. The protein levels of RIP1 and RIP3 in primary tumor tissues were increased by shikonin. The overall survival of lung metastatic models was longer compared with control group (p < 0.001).Conclusions: Shikonin had prompt but profound anti-tumor effect on both primary and metastatic osteosarcoma, probably by inducing RIP1 and RIP3 dependent necroptosis.
