ABSTRACT Microbially produced electrically conductive protein filaments are of interest because they can function as conduits for long-range biological electron transfer. They also show promise as sustainably produced electronic materials. Until now, microbially produced conductive protein filaments have been reported only for bacteria. We report here that the archaellum of Methanospirillum hungatei is electrically conductive. This is the first demonstration that electrically conductive protein filaments have evolved in Archaea. Furthermore, the structure of the M. hungatei archaellum was previously determined (N. Poweleit, P. Ge, H. N. Nguyen, R. R. O. Loo, et al., Nat Microbiol 2:16222, 2016, https://doi.org/10.1038/nmicrobiol.2016.222). Thus, the archaellum of M. hungatei is the first microbially produced electrically conductive protein filament for which a structure is known. We analyzed the previously published structure and identified a core of tightly packed phenylalanines that is one likely route for electron conductance. The availability of the M. hungatei archaellum structure is expected to substantially advance mechanistic evaluation of long-range electron transport in microbially produced electrically conductive filaments and to aid in the design of “green” electronic materials that can be microbially produced with renewable feedstocks. IMPORTANCE Microbially produced electrically conductive protein filaments are a revolutionary, sustainably produced, electronic material with broad potential applications. The design of new protein nanowires based on the known M. hungatei archaellum structure could be a major advance over the current empirical design of synthetic protein nanowires from electrically conductive bacterial pili. An understanding of the diversity of outer-surface protein structures capable of electron transfer is important for developing models for microbial electrical communication with other cells and minerals in natural anaerobic environments. Extracellular electron exchange is also essential in engineered environments such as bioelectrochemical devices and anaerobic digesters converting wastes to methane. The finding that the archaellum of M. hungatei is electrically conductive suggests that some archaea might be able to make long-range electrical connections with their external environment.

Successful synthesis of ordered porous, multi-component complex materials requires a series of coordinated processes, typically including fabrication of a master template, deposition of materials within the pores to form a negative structure, and a third deposition or etching process to create the final, functional template. Translating the utility and the simplicity of the ordered nanoporous geometry of binary oxide templates to those comprising complex functional oxides used in energy, electronic, and biology applications has been met with numerous critical challenges. This review surveys the current state of commonly used complex material nanoporous template synthesis techniques derived from the base anodic aluminum oxide (AAO) geometry.

The recent availability of digital traces generated by cellphone calls has significantly increased the scientific understanding of human mobility. Until now, however, based on low time resolution measurements, previous works have ignored to study human mobility under various time scales due to sparse and irregular calls, particularly in the era of mobile Internet. In this paper, we introduced Mobile Flow Records, flow-level data access records of online activity of smartphone users, to explore human mobility. Mobile Flow Records collect high-resolution information of large populations. By exploiting this kind of data, we show the models and statistics of human mobility at a large-scale (3,542,235 individuals) and finer-granularity (7.5min). Next, we investigated statistical variations and biases of mobility models caused by different time scales (from 7.5min to 32h), and found that the time scale does influence the mobility model, which indicates a deep coupling of human mobility and time. We further show that mobility behaviors like transportation modes contribute to the diversity of human mobility, by exploring several novel and refined features (e.g., motion speed, duration, and trajectory distance). Particularly, we point out that 2-hour sampling adopted in previous works is insufficient to study detailed motion behaviors. Our work not only offers a macroscopic and microscopic view of spatial-temporal human mobility, but also applies previously unavailable features, both of which are beneficial to the studies on phenomena driven by human mobility.

There has been a dramatic growth of shared mobility applications such as ride-sharing, food delivery, and crowdsourced parcel delivery. Shared mobility refers to transportation services that are shared among users, where a central issue is route planning. Given a set of workers and requests, route planning finds for each worker a route, i.e., a sequence of locations to pick up and drop off passengers/parcels that arrive from time to time, with different optimization objectives. Previous studies lack practicability due to their conflicted objectives and inefficiency in inserting a new request into a route, a basic operation called insertion. In addition, previous route planning solutions fail to exploit the appearance patterns of future requests hidden in historical data for optimization. In this paper, we present a unified formulation of route planning called URPSM. It has a well-defined parameterized objective function which eliminates the contradicted objectives in previous studies and enables flexible multi-objective route planning for shared mobility. We propose two insertion-based frameworks to solve the URPSM problem. The first is built upon the plain-insertion widely used in prior studies, which processes online requests only, whereas the second relies on a new insertion operator called prophet-insertion that handles both online and predicted requests. Novel dynamic programming algorithms are designed to accelerate both insertions to only linear time. Theoretical analysis shows that no online algorithm can have a constant competitive ratio for the URPSM problem under the competitive analysis model, yet our prophet-insertion-based framework can achieve a constant optimality ratio under the instance-optimality model. Extensive experimental results on real datasets show that our insertion-based solutions outperform the state-of-the-art algorithms in both effectiveness and efficiency by a large margin (e.g., up to 30× more effective in the objective and up to 20× faster). Many object localization applications need the relative locations of a set of objects as oppose to their absolute locations. Although many schemes for object localization using radio frequency identification (RFID) tags have been proposed, they mostly focus on absolute object localization and are not suitable for relative object localization because of large error margins and the special hardware that they require. In this paper, we propose an approach called spatial-temporal phase profiling (STPP) to RFID-based relative object localization. The basic idea of STPP is that by moving a reader over a set of tags during which the reader continuously interrogating the tags, for each tag, the reader obtains a sequence of RF phase values, which we call a phase profile, from the tag's responses over time. By analyzing the spatial-temporal dynamics in the phase profiles, STPP can calculate the spatial ordering among the tags. In comparison with prior absolute object localization schemes, STPP requires neither dedicated infrastructure nor special hardware. We implemented STPP and evaluated its performance in two real-world applications: locating misplaced books in a library and determining the baggage order in an airport. The experimental results show that STPP achieves about 84% ordering accuracy for misplaced books and 95% ordering accuracy for baggage handling. We further leverage the controllable reader antenna and upgrade STPP to infer the spacing between each pair of tags. With the marvelous development of wireless techniques and ubiquitous deployment of wireless systems indoors, myriad indoor location-based services (ILBSs) have permeated into numerous aspects of modern life. The most fundamental functionality is to pinpoint the location of the target via wireless devices. According to how wireless devices interact with the target, wireless indoor localization schemes roughly fall into two categories: device based and device free. In device-based localization, a wireless device (e.g., a smartphone) is attached to the target and computes its location through cooperation with other deployed wireless devices. In device-free localization, the target carries no wireless devices, while the wireless infrastructure deployed in the environment determines the target's location by analyzing its impact on wireless signals. This article is intended to offer a comprehensive state-of-the-art survey on wireless indoor localization from the device perspective. In this survey, we review the recent advances in both modes by elaborating on the underlying wireless modalities, basic localization principles, and data fusion techniques, with special emphasis on emerging trends in (1) leveraging smartphones to integrate wireless and sensor capabilities and extend to the social context for device-based localization, and (2) extracting specific wireless features to trigger novel human-centric device-free localization. We comprehensively compare each scheme in terms of accuracy, cost, scalability, and energy efficiency. Furthermore, we take a first look at intrinsic technical challenges in both categories and identify several open research issues associated with these new challenges. [ABSTRACT FROM AUTHOR]

Counting or estimating the number of tags is crucial for RFID system. Researchers have proposed several fast cardinality estimation schemes to estimate the quantity of a batch of tags within a short time frame. However, existing estimation schemes scarcely consider the privacy issue. Without effective protection, the adversary can utilize the responding signals to estimate the number of tags as accurately as the valid reader. To address this issue, we propose a novel privacy-preserving estimation scheme, termed as MEAS, which provides an active RF countermeasure against the estimation from invalid readers. MEAS comprises of two components, an Estimation Interference Device (EID) and two well-designed Interference Blanking Estimators (IBE). EID is deployed with the tags to actively generate interfering signals, which introduce sufficiently large estimation errors to invalid or malicious readers. Using a secret interference factor shared with EID, a valid reader can perform accurate estimation via two IBEs. Our theoretical analysis and simulation results show the effectiveness of MEAS. The spatial features of emitted wireless signals are the basis of location distinction and determination for wireless indoor localization. Available in mainstream wireless signal measurements, the Received Signal Strength Indicator (RSSI) has been adopted in vast indoor localization systems. However, it suffers from dramatic performance degradation in complex situations due to multipath fading and temporal dynamics. Break-through techniques resort to finer-grained wireless channel measurement than RSSI. Different from RSSI, the PHY layer power feature, channel response, is able to discriminate multipath characteristics, and thus holds the potential for the convergence of accurate and pervasive indoor localization. Channel State Information (CSI, reflecting channel response in 802.11 a/g/n) has attracted many research efforts and some pioneer works have demonstrated submeter or even centimeter-level accuracy. In this article, we survey this new trend of channel response in localization. The differences between CSI and RSSI are highlighted with respect to network layering, time resolution, frequency resolution, stability, and accessibility. Furthermore, we investigate a large body of recent works and classify them overall into three categories according to how to use CSI. For each category, we emphasize the basic principles and address future directions of research in this new and largely open area. [ABSTRACT FROM AUTHOR]

Gliomas are the most severe brain tumours with a poor prognosis. Although surgery, postoperative radiotherapy and chemotherapy can improve the survival rate of glioma patients, the prognosis of most glioma patients is still poor. In recent years, the influence of gene-targeted therapy on gliomas has been gradually discovered, and intervening the occurrence and development of brain gliomas from the perspective of the gene will significantly improve treatment prognosis. Protein Kinase C and Casein Kinase Substrate in Neurons 1 (PACSIN1) is a member of the conserved peripheral membrane protein family in eukaryotes. Improper expression of PACSIN1 can lead to neurological diseases such as Huntington's disease and schizophrenia. However, its relationship with tumours or even gliomas has not been explored. The study aims to explore PACSIN1 as a prognostic factor that can predict overall survival (OS) for gliomas. We collected the data from CGGA, TCGA, GEO databases and the pathological glioma tissue specimens from 15 clinical glioma patients surgically resected. The differential expression of PACSIN1 in various clinical indicators, the genes related to PACSIN1 expression, the prognostic value of PACSIN1 and the functional annotations and pathway analysis of differently expressed genes (DEGs) were analysed. The results revealed that PACSIN1 had low expression levels in grade IV, IDH1 wild-type and 1p/19q non-codel group gliomas, and PACSIN1 was considered a mesenchymal molecular subtype marker. PACSIN1 expression is positively correlated with OS in all gliomas and it was found that PACSIN1 influenced the occurrence and development of gliomas through synaptic transmission.
