The microbial communities inhabiting polar ecosystems, particularly in Maxwell Bay, Antarctica, play a pivotal role in nutrient cycling and ecosystem dynamics. However, the diversity of these microbial communities remains underexplored. In this study, we aim to address this gap by investigating the distribution, environmental drivers, and metabolic potential of microorganisms in Maxwell Bay. We analyzed the prokaryotic and eukaryotic microbiota at 11 stations, revealing distinctive community structures and diverse phylum dominance by using high-throughput sequencing. Spatial analysis revealed a significant impact of longitude on microbial communities, with microeukaryotes exhibiting greater sensitivity to spatial factors than microprokaryotes. We constructed co-occurrence networks to explore the stability of microbial communities, indicating the complexity and stability of microprokaryotic communities compared with those of microeukaryotes. Our findings suggest that the microeukaryotic communities in Maxwell Bay are more susceptible to disturbances. Additionally, this study revealed the spatial correlations between microbial communities, diversity, and environmental variables. Redundancy analysis highlighted the significance of pH and dissolved oxygen in shaping microprokaryotic and microeukaryotic communities, indicating the anthropogenic influence near the scientific research stations. Functional predictions using Tax4Fun2 and FUNGuild revealed the metabolic potential and trophic modes of the microprokaryotic and microeukaryotic communities, respectively. Finally, this study provides novel insights into the microbial ecology of Maxwell Bay, expanding the understanding of polar microbiomes and their responses to environmental factors.

Bacteria are vital for maintaining ecosystem function in the rapidly changing environment, but limited data was reported regarding the characteristics of bacterial communities along the regional-scale in aquatic ecosystems on the Fildes Peninsula (Antarctica), where vulnerable aquatic habitats have been affected by the dramatic retreat of the Collins glacier. This study revealed differences and hidden connections of bacterial communities in two aquatic ecosystems (periglacial lakes and coves) near Collins glacier using amplicon sequencing. The results showed periglacial lakes and coves harbored 568 and 388 unique OTUs, respectively, but also shared 199 common OTUs, which meant there existed some linkages connections between the two ecosystems. Abstract Background In the ice-free area of maritime Antarctica, fungi are the essential functioning group in terrestrial and marine ecosystems. Until now, no study has been conducted to comprehensively assess fungal communities in various habitats in Antarctica. We aimed to characterize fungal communities in the eleven habitats (i.e., soil, seawater, vascular plant, dung, moss, marine alga, lichen, green alga, freshwater, feather) in the Fildes Region (maritime Antarctica) using next-generation sequencing. Results A total of 12 known phyla, 37 known classes, 85 known orders, 164 known families, 313 known genera, and 320 known species were detected. Habitat specificity rather than habitat overlap determined the composition of fungal communities, suggesting that, although fungal communities were connected by dispersal at the local scale, the environmental filter is a key factor driving fungal assemblages in the ice-free Antarctica. Furthermore, 20 fungal guilds and 6 growth forms were detected. Many significant differences in the functional guild (e.g., lichenized, algal parasite, litter saprotroph) and growth form (e.g., yeast, filamentous mycelium, thallus photosynthetic) existed among different habitat types. Conclusion The present study reveals the high diversity of fungal communities in the eleven ice-free Antarctic habitats and elucidates the ecological traits of fungal communities in this unique ice-free area of maritime Antarctica. The findings will help advance our understanding of fungal diversity and their ecological roles with respect to habitats on a neighbourhood scale in the ice-free area of maritime Antarctica.

Paleo-climate proxy records documenting sea-ice extent are important sources of information to assess the time of emergence and magnitude of on-going changes in the Arctic Ocean and better predict future climate and environmental evolution in that region. In this study, a suite of geochemical tracers including total organic carbon (TOC), total nitrogen (TN), carbon/nitrogen ratio (C/N), stable isotope composition of organic carbon and nitrogen ( δ ^13 C, δ ^15 N), and phytoplankton biomarkers (highly branched isoprenoids (HBIs) and sterols) were measured in a marine sediment core to document the sea-ice variability in the Chukchi Sea since the beginning of the Industrial Era. The downcore profile of the sea-ice proxy HBIs suggests a transition from extensive sea ice in the late 19th century to Marginal Ice Zone (MIZ) in AD 1930–1990s and then moderate sea-ice cover since 1990s. Rising of all HBI abundances between AD 1865–1875 indicate a transient retreat of summer ice edge off the shelf followed by a return to near-perennial sea ice till 1920–1930 as revealed by the absence of HBIs and brassicasterol. Sea ice retreat occurred again in AD 1920–1930 and followed by colder decades in 1940s–1960s before a sustained decline since the 1990s. The downcore profile of C/N, δ ^13 C of organic matter and sterols indicates a gradual increase of terrigenous inputs accelerating during the most recent decades likely due to enhanced fluvial run-off and sediment transport by sediment-laden sea ice. Concomitantly, increasing δ ^15 N values suggest limited nutrient utilization due to enhanced stratification of the surface ocean caused by increased freshening. The role of the Arctic oscillation (AO), the Pacific decadal oscillation (PDO) and Atlantic Multidecadal Oscillation (AMO) are discussed to explore potential drivers of the observed sea-ice changes.

Background: SCTV01E is a tetravalent recombinant COVID-19 vaccine authorized for emergency use in China for adults 18 years and older but not for those under 18. Objective: This Phase 2 trial assessed the safety and immunogenicity of SCTV01E in healthy children and adolescents aged 3 to 17 years, to establish immunobridging with that observed in adults from the efficacy pivotal trial (NCT05308576). Methods: Participants were randomly assigned to receive either 30 µg of SCTV01E or a placebo. Primary endpoints were safety and immunogenicity focused on the geometric mean titer (GMT) and seroresponse rate (SRR) of neutralizing antibodies (nAb) against Omicron BA.5. Results: In total, 268 participants (214 SCTV01E vs. 54 placebo) were included in the safety analysis, with 241 participants (191 vs. 50) in the immunogenicity analysis. Overall, 127 (59.3%) participants receiving SCTV01E and 9 (16.7%) receiving a placebo reported adverse events (AEs), most of which were Grade 1 or 2. No serious adverse events (SAEs) or adverse events of special interest (AESIs) were reported. In the immunogenicity bridging analysis, data from 95 youths were compared with data from 188 adults; the geometric mean ratio (GMR) of the titers was 8.78 (95% CI: 6.05–12.74, p < 0.001), with the lower bound of the 95% CI exceeding 0.67. The difference in the SRR was 6.34% (95% CI: 0.93–11.22%) (p = 0.029), and the lower bound of the 95%CI was >−5%, indicating superiority. Conclusions: SCTV01E was found to be safe and well tolerated in children and adolescents, generating a robust immune response against Omicron BA.5. This supports its potential use in younger populations.

The diversity, co-occurrence patterns and assembly processes of microeukaryotes in Antarctic freshwater lakes are not well understood, despite its wide distribution and ecological importance. This study used Illumina high-throughput sequencing to explore five freshwater lakes' microeukaryotic communities on the Fildes Peninsula during three summer seasons. A total of 33 phyla were detected, with the phytoplankton occupying the highest percentage of sequences (accounting for up to 98%). Meanwhile, the main dominant taxa were Chrysophyta, Chlorophyta, and Cryptophyta. Alpha diversity varied among lakes, with Changhu (CH), Kitec (KT) lake having higher values, and Yue Ya (YY) lake having the lowest value. There were significant differences in microeukaryotic communities between lakes, with spatial and temporal variation in the relative abundance of dominant taxa (P<0.05). Environmental variables only explained about 30% of the variation in community structure. In the co-occurrence network, microeukaryotes tended to be more symbiotic than competitive with each other (positive correlation 82% vs. negative correlation 18%), with only 8% of OTUs significantly associated with environmental factors. The neutral community model found that neutral processes explained more than 56% of the community variation. The stochastic processes (e.g., homogenizing dispersal and undominated process) predominated in community assembly than the deterministic processes. These findings revealed the diversity of the microeukaryotic community and have important implications for understanding the community assembly in the freshwater lakes of the Fildes Peninsula (Antarctica). Objective: In this paper, we aim to show that the immunogenicity of the lyophilized human rabies vaccine (Vero cells) (investigational vaccine) developed by Dalian Aleph Biomedical Co., Ltd. in healthy participants aged 10–60 years old is non-inferior to the lyophilized PVRV (positive control) manufactured by Liaoning Chengda Biotechnology Co., Ltd. (Shenyang, China), and that its safety is clinically acceptable. Method: A total of 2776 participants were enrolled in this study and divided into four groups: a five-dose test group, a five-dose control group, a four-dose test group, and a four-dose control group. The patients in the four-dose groups (Zagreb) were vaccinated on Days 0 (two doses), 7 (one dose), and 21 (one dose), and those in the five-dose groups (Essen) were vaccinated on Days 0, 3, 7, 14, and 28 (one dose each). The rabies-virus-neutralizing antibody assay with the RFFIT was used to assess the immunogenicity, and the adverse events (AEs) and serious adverse events (SAEs) were identified and collated. Results: The positive seroconversion rate was up to 100% on Days 14 and 35/42 after vaccination following any procedures in pre-immunization antibody-negative participants, and the positive seroconversion rate and geometric mean concentration (GMC) of the test groups (Zagreb and Essen vaccination procedures) was not inferior to that of the control groups. On Day 7 after vaccination, the immunogenicity of the Zagreb procedure with two doses of the vaccine on Day 0 was superior to the Essen procedure with one dose of vaccine, that is, the former had a higher seroconversion rate and RVNA titer. The non-inferiority criterion of immunogenicity was met for the whole population, the population aged 10–18 years and ≥18 years, and the pre-immunization antibody-positive population. The incidences of all AEs, solicited AEs, and unsolicited AEs in both groups were not statistically significant, and no vaccination-related SAEs were observed. Conclusion: The investigated vaccine is safe, its immunogenicity is non-inferior to that of the control vaccine, and the efficacy of the Zagreb procedure is superior to that of the Essen procedure 7 days after the first dose.
