The species included in the ESKAPE group (Enterococcus faecium, Staphylococcus aureus, Klebsiella pneumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa and the genus Enterobacter) have a high capacity to develop antimicrobial resistance (AMR), a health problem that is already among the leading causes of death and could kill 10 million people a year by 2050. The generation of new potentially therapeutic molecules has been insufficient to combat the AMR "crisis", and the World Health Organization (WHO) has stated that it will seek to promote the development of rapid diagnostic strategies. The physicochemical properties of metallic nanoparticles (MNPs) have made it possible to design biosensors capable of identifying low concentrations of ESKAPE bacteria in the short term; other systems identify antimicrobial susceptibility, and some have been designed with dual activity in situ (bacterial detection and antimicrobial activity), which suggests that, in the near future, multifunctional biosensors could exist based on MNPs capable of quickly identifying bacterial pathogens in clinical niches might become commercially available. This review focuses on the use of MNP-based systems for the rapid and accurate identification of clinically important bacterial pathogens, exhibiting the necessity for exhaustive research to achieve these objectives. Wearable biosensors (wearables) enable continual, noninvasive physiologic and behavioral monitoring at home for those with pediatric or congenital heart disease. Wearables allow patients to access their personal data and monitor their health. Despite substantial technologic advances in recent years, issues with hardware design, data analysis, and integration into the clinical workflow prevent wearables from reaching their potential in high-risk congenital heart disease populations. This science advisory reviews the use of wearables in patients with congenital heart disease, how to improve these technologies for clinicians and patients, and ethical and regulatory considerations. Aptamers are single-stranded oligonucleotide sequences capable of binding to specific ligands with high affinity. In this manner, they are like antibodies but have advantages such as lower manufacturing costs, lower immunogenicity, fewer batch-to-batch differences, a longer shelf life, high tolerance to different molecular milieus, and a greater number of potential targets. Due to their special features, they have been used in drug delivery, biosensor technology, therapy, and diagnostics. The methodology that allowed its production was the “Systematic Evolution of Ligands by Exponential enrichment” (SELEX). Unfortunately, the traditional protocol is time-consuming and laborious. Therefore, numerous variants with considerable optimization steps have been developed, nonetheless, there are still challenges to achieving real applications in the clinical field. Among them, are control of in vivo activities, fast renal filtration, degradation by nucleases and toxicity testing. This review focuses on current technologies based on SELEX, the critical factors for successful aptamer selection, and its upcoming biomedical and biotechnological applications.

Background/Aims Upper gastrointestinal bleeding (UGIB) is a life-threatening condition that necessitates early identification and intervention and is associated with substantial morbidity, mortality, and socioeconomic burden. However, several diagnostic challenges remain regarding risk stratification and the optimal timing of endoscopy. The PillSense System is a noninvasive device developed to detect blood in patients with UGIB in real time. This study aimed to assess the safety and performance characteristics of PillSense using a simulated bleeding model. Methods A preclinical study was performed using an in vivo porcine model (14 animals). Fourteen PillSense capsules were endoscopically placed in the stomach and blood was injected into the stomach to simulate bleeding. The safety and sensitivity of blood detection and pill excretion were also investigated. Results All the sensors successfully detected the presence or absence of blood. The minimum threshold was 9% blood concentration, with additional detection of increasing concentrations of up to 22.5% blood. All the sensors passed naturally through the gastrointestinal tract. Conclusions This study demonstrated the ability of the PillSense System sensor to detect UGIB across a wide range of blood concentrations. This ingestible device detects UGIB in real time and has the potential to be an effective tool to supplement the current standard of care. These favorable results will be further investigated in future clinical studies.

A novel genosensor was developed for rotavirus specific cDNA sequence detection. The genosensor was comprised of hierarchical flower-like gold nanostructures, MXene, and polypyrrole (HFGNs/MXene/PPY) nanocomposite as a signal amplification tag, specific antisense ssDNA oligonucleotide as a recognition bioelement, and methylene blue (MB) as a redox marker. The morphological and electrochemical features of the biosensor were first tested and optimized and the high performance of the platform was confirmed in terms of sensitivity and reproducibility. Then, 20 rotavirus RNA isolated from clinical and cell-cultured samples (10 positive and 10 negative confirmed by RT-PCR and electrophoresis methods) were evaluated by the genosensor. The analysis results revealed that the genosensor is able to differentiate successfully between the positive and negative control groups. The developed genosensor for rotavirus RNA detection presented an excellent limit of detection of ∼ 0.8 aM and a determination range of 10−18 and 10−7 M. In addition, the ssDNA/HFGNs/MXene/PPY/GCE showed high selectivity and long-term stability of ~ 24 days. Background: Creatinine is a universally important blood parameter used to detect and monitor acute and chronic kidney disease. Reliable measurements at the bedside remain a challenge in human and veterinary medicine. Despite its potential, a trustworthy point‐of‐care creatinine biosensor has yet to be established. Objectives: We aimed to determine the precision and accuracy of the StatSensor (SS) and StatSensor Xpress (SSX) handheld creatinine measurement devices in dogs. Methods: Paired creatinine samples from dogs with normal (creatinine ≤159 μmol/L), moderate (159–354 μmol/L), and marked (>354 μmol/L) azotemia were compared with a commercial enzymatic analyzer. Within‐day precision and linearity studies were performed prior to method comparison studies. Method comparison was evaluated using Bland–Altman, concordance correlation coefficient, Deming, and Passing‐Bablok regression analysis. Results: Seventy‐eight dogs were enrolled in the study, including 28 (35%), 25 (32%), and 26 (33%) with normal, moderate, and marked azotemia. Total error surpassed recommendations for all devices, and linearity deviated from identity for the SS1 and SS2. The concordance correlation coefficients of the SS1, SS2, SSXI, and SSX2, were 0.69, 0.59, 0.82, and 0.44, respectively. Bland–Altman analyses showed a high variation in the differences, and relationships showed high heteroskedasticity with negative systemic bias among high creatinine concentrations. Conclusions: Neither the SS and SSX are considered acceptable for clinical applications in dogs. Further research is indicated for the development of a reliable, cost‐effective, point‐of‐care creatinine analyzer to improve the rapid detection and monitoring human and veterinary patients. Glycosylation, the intricate process of adding carbohydrate motifs to proteins, lipids, and exosomes on the cell surface, is crucial for both physiological and pathological mechanisms. Alterations in glycans significantly affect cancer cell metastasis by mediating cell-cell and cell-matrix interactions. The subtle changes in glycosylation during malignant transformations highlight the importance of analyzing cell and exosome surface glycosylation for prognostic and early treatment strategies in cancer. This review focuses on recent advancements in sensors for detecting surface glycans on cancer cells, exosomes, and circulating cancer-derived glycoproteins. Among various methods, electrochemical biosensors stand out as a promising tool, offering rapid and cost-effective glycan detection. These devices detect glycan interactions by measuring changes in electrical signals resulting from specific binding events. Techniques such as differential pulse voltammetry, impedance spectroscopy, and chrono amperometry are commonly employed for glycan detection using electrochemical biosensors. Researchers are exploring novel electrode materials and surface functionalization strategies to enhance sensor performance. Notably, selective binding probes such as lectins, aptamers, antibodies, and boronic acids are discussed, with lectins being the most prevalent for specific glycan analysis. By highlighting the significance of electrochemical techniques, emphasizing the role of selective binding probes, integrating microfluidics and miniaturized devices could lead to point-of-care applications for cancer diagnosis and monitoring. Background: Foodborne pathogens, particularly Vibrio parahaemolyticus (VP) found in seafood, pose significant health risks, including abdominal pain, nausea, and even death. Rapid, accurate, and sensitive detection of these pathogens is crucial for food safety and public health. However, existing detection methods often require complex sample pretreatment, which limits their practical application. This study aims to overcome these limitations by developing a label-free magnetic relaxation switch (MRS) biosensor for the detection of VP, utilizing a photocleavable sol-gel phase transition system for improved efficiency and accuracy., Results: In this work, a tag-free magnetic relaxation switch (MRS) biosensor was designed for the detection of Vibrio parahaemolyticus (VP), based on a photocleavable sol-gel phase transition system. A large amount of lithium acyl hypophosphite (LAP), gold nanoparticles (AuNPs), and single-stranded DNA (ssDNA) loaded on the surface of Ti 3 C 2 T x MXene acted as the signal unit LAP-MXene@AuNPs-ssDNA. The pipette tip served as a reaction vessel, and when VP was present, Apt specifically captured VP and released the signal units. The released signal units were then injected into the low-field nuclear magnetic resonance (LF-NMR) test solution, a gel formed by crosslinking of disulfide bonds. The gel was cleaved by LAPs on the signal units under ultraviolet (UV) irradiation, triggering a gel-sol phase transition, which increased transverse relaxation time (T 2 ), thus enabling the detection of VP. Under the optimal experimental conditions, the linear range and detection limit for VP were 10 2 ∼ 10 8  CFU/mL and 10 CFU/mL, respectively., Significance and Novelty: The simplified biometric identification process in the pipette tip reduces errors from multiple sample transfers, enhancing efficiency. Agmatine, the decarboxylation product of arginine, is the precursor of putrescine - a harmful biogenic amine (BA) - that can accumulate in dairy products via bacterial metabolism involving the agmatine deiminase (AGDI) pathway. This first requires agmatine be produced via the decarboxylation of arginine and it remains unknown which microorganisms are responsible for this prior decarboxylation step. In addition, agmatine, as other BA, plays different physiological roles including those of co-transmitter and neuromodulator. Preclinical and clinical studies have shown agmatine to have a neuroprotective effect, rendering it of therapeutic interest being agmatine-producing bacteria proposed as psychobiotics. The identification of BA-producing microorganisms is based on the rise in pH due to the consumption of H + during such decarboxylation reactions. However, in the detection of agmatine-producing microorganisms in cheese, this would lead to false positives since many bacteria possess arginine deiminase activity; this produces ornithine and ammonium from arginine, which also increases the pH. To overcome this problem, a whole-cell biosensor based on a previously developed agmatine-inducible transcription system was designed, and a protocol optimized for the successful identification of agmatine-producing microorganisms in cheese. The application of this protocol in cheese samples allowed for the isolation of agmatine-producing microorganisms identified as Hafnia spp. and unravels, for first time, the capacity of Hafnia paralvei to produce agmatine. Background: Methotrexate (MTX) is widely used in clinical practice for the treatment of malignant tumors and autoimmune diseases. High-dose MTX has been shown to be an effective approach for treating various malignant tumors, but it is accompanied by numerous toxic side effects, necessitating therapeutic drug monitoring (TDM) for patients and timely "folinic acid rescue." High-performance liquid chromatography and fluorescent immunoassay (FIA) are currently used to detect MTX, but these methods are limited by complex sample preparation, time consumption, and high cost. Therefore, a simple, rapid, and cost-effective MTX measurement method is required., Results: We developed a flexible and inexpensive electrochemical sensor using a stearyl trimethyl ammonium bromide (STAB)-modified, screen-printed carbon electrode to directly detect MTX in human serum. Assay performance was validated via detection of MTX in spiked buffer. The sensor was capable of measuring MTX concentrations ranging from 0.01 to 1 μM and 1-1500 μM, with a limit of detection of 3.1 nM and a limit of quantitation of 3.5 nM. For the samples simulating combined medication, the sensor exhibited outstanding selectivity, in cross-reactivity, with the maximum response value of interferents reaching only 3.49 %. Additionally, the sensor shows reliable repeatability with a relative standard deviation of 3.8 % and remarkable stability. Recovery in human serum validated the clinical utility of the sensor in point-of-care testing conditions. The sensor's applicability to personal medicine was confirmed by detecting MTX blood concentration in patients with different diseases. The results obtained by the sensor were compared with those obtained by the FIA technique, a method commonly used in hospitals, showing a high level of consistency between both methods., Significance: To meet the requirements of personalized medicine for MTX-patients, we developed a disposable biosensor with wide detection range from 0.01 to 1 μM and 1-1500 μM. Background: Ochratoxin A (OTA) is a serious food contaminant, not easily degradable, and capable of causing irreversible damage to the human body. Therefore, it is of great practical significance to establish a sensitive and efficient OTA detection method. The electrochemical aptasensor has a broad development prospect in OTA detection with its advantages of fast response speed and low cost., Results: Herein, a cascade signal amplification strategy based on AuPt NPs/Zr-MOF and π-structure bipedal DNA walker-triggered hybridization chain reaction (HCR) was designed for the detection of ochratoxin A (OTA). AuPt NPs/Zr-MOF was employed as the electrode modification material, providing a large number of active sites and high conductivity, achieving 1.47 times signal amplification. Interestingly, bipedal DNA walker binds to hairpin 1 (H1) to form the π-structure. Under the activation of Pb 2+ , one bipedal DNA walker can simultaneously bind and cleave two H1. It exhibits a wide walking range and high recognition efficiency. After H1 is cleaved, the trigger sequence was exposed to trigger HCR and a large amount of methylene blue was loaded on the electrode. Under the optimal conditions, the linear range of the determined OTA is 1 × 10 -3 -500 ng/mL, and the limit of detection is as low as 0.525 pg/mL., Significance: The experimental results demonstrate that the constructed electrochemical aptasensor is a sensitive and efficient platform for OTA monitoring. Background: Aminoglycoside antibiotics (AGs) are commonly utilized in both human and veterinary medicine to treat and manage a range of infections. These antibiotics are recognized for their narrow therapeutic window, with an overdose potentially resulting in severe side effects like kidney and ear damage. Hence, the implementation of a quick, precise, and on-the-spot testing method is crucial in clinical settings. In the present investigation, we designed an innovative indirect lateral flow assay (LFA) utilizing aptamers to detect kanamycin, a type of aminoglycoside antibiotics. We prepared mesoporous silica nanoparticles (MSNs) functionalized with amino groups, loaded them with morphine (MOP), and then sealed them with aminoglycoside aptamers (Apt). The complex of gated-MSNs@Apt was tested using the MOP LFA in the presence or absence of kanamycin antibiotics., Results: The indirect LFA system displayed a single colored line in the test line for positive samples, whereas it exhibited double colored lines in both the control line and test line for negative samples. Our custom-designed LFA biosensors demonstrated two linear ranges, from 10 nM to 350 nM and 500 nM-1500 nM, with a limit of detection (LOD) of 5 nM in serum media under optimized conditions. Epidermal growth factor receptor (EGFR) is a transmembrane protein and a key biomarker implicated in the pathogenesis of breast cancer. Early and precise detection of EGFR is crucial for effective diagnosis, prognosis, and therapeutic intervention. However, conventional EGFR detection techniques, such as biopsy and immunohistochemistry, are often invasive, time-consuming, and limited in sensitivity, highlighting the demand for non-invasive, highly sensitive detection methods. In this study, we fabricated a cerium oxide (CeO₂) and graphene oxide (GO) nanocomposite-based aptasensor for the non-invasive detection of EGFR using electrochemical impedance spectroscopy (EIS). The CeO₂-GO nanocomposite was synthesized via the sol-gel method and characterized through UV-Vis spectroscopy, FTIR, TEM, and XRD, confirming the crystalline structure of hexagonal CeO₂ nanoparticles on amorphous GO sheets. The nanocomposite was functionalized with aptamers specific to EGFR using covalent coupling reactions. The EIS analysis of the fabricated aptasensor (GCE/CeO₂-GO/EGFR-Apt/BSA) demonstrated a wide linear detection range from 10 fg mL -1 to 100 ng mL -1 , with an ultralow detection limit of 1.87 fg mL -1 in PBS, 3.16 fg mL -1 in serum, 5.31 fg mL -1 in sweat, and 6.14 fg mL -1 in saliva samples. These results highlight the aptasensor's high sensitivity, specificity, and potential for real-time, non-invasive EGFR monitoring in clinical samples such as serum, sweat, and saliva. Midkine (MDK) is a neurotrophic growth factor highly expressed during embryogenesis, currently recognized as a multifaceted factor in cancer progression and drug resistance. MDK has demonstrated greater accuracy than existing biomarkers. Serum MDK is a valuable indicator for the non-invasive early detection of tumors. It dynamically changes following surgical tumor excision and prior to recurrence, facilitating prognosis and treatment response evaluation. However, existing methods struggle to achieve the sensitivity required for clinical applications. Herein, we developed a triple-mode biosensor with oriented self-construction and built-in error-checking for rapid, sensitive, and convenient MDK detection. The sensor construction adhered to the principle of achieving oriented and strong covalent connections to ensure high sensitivity. Biosynthesized quantum dots (BQDs) were introduced to orient antibodies, enhancing the exploration of active binding sites and significantly increasing antibody-capturing ability. To further enhance sensitivity and signal amplification, Au@Pt nanorods-Ab 2 (MF-Probe) were used as multifunctional probes, incorporating an error-checking mechanism to minimize false results. Detection was feasible using an electrochemical workstation, a microplate reader, and even a mobile phone. The sensor exhibited a wide linear range from 5 fg/mL to 100 ng/mL and a low limit of detection (LOD) of 1.620 fg/mL. It accurately distinguished MDK levels in the serum of healthy donors and cancer patients. Controlling the size and morphology of supramolecular assemblies is still an enormous challenge for the development of novel functional materials. Herein, cyclometalated platinum(II) complexes were designed and used as effective theragnostic supramolecular nano agents. Benzoquinoline was used as the main C^N ligand, and the increased number of aromatic rings in the N^N ligand of Pt complexes could greatly improve lipophilicity and cytotoxicity towards cancer cells over normal cells. Moreover, the morphology of nanoparticles formed by self-assembly of Pta-d could change from one to three-dimensional (1-3D), which can form nanowires and nanospheres. Besides, complexes of Pta-c, the number of aromatic rings in the N^N ligand of which does not exceed 4, all exhibit significant aggregation-induced phosphorescent emission (AIPE) with 100 - 200-fold enhancement in red phosphoresce intensity recorded in the solvent of Pta-1 and Ptb. Pt-Pt interaction induced by coordination and electrostatic interaction between complex and anions, and a new deep red emission improvement was observed in aqueous solution of Pta and Ptb with the presence of ClO 4 - , and two similar deep red emissions induced by two different interactions can be told by new emerging MMLCT absorption band. Ptd of dppz ligand exhibits the highest efficiency in inducing apoptosis of HeLa and its anticancer mechanism was studied. Simultaneous monitoring of key metabolites like dopamine, ascorbic acid, and uric acid is essential for early disease diagnosis and evaluating treatment. Electrochemical techniques are increasingly used for precise, point-of-care testing (POCT) of these metabolites. Herein, a sample pretreatment method called effervescent solid-phase extraction (ESPE) was proposed for efficient enrichment of trace analytes for electrochemical detection. In an ESPE process, effervescent tablets made of gold nanoparticle-decorated graphene oxide (Au/GO) were first self-dispersed in a test solution to promote the enrichment of analytes on the Au/GO adsorbents, followed by the addition of flocculant effervescent tablets to cause Au/GO sheets to form self-assembled aggregates, which then can be efficiently collected by the foam electrodes. The entire sample pretreatment process operates without external power and takes only 5 min. With the assistance of the ESPE method, our electrochemical sensors achieve an ultralow detection limit for dopamine, ascorbic acid, and uric acid of 80 pM, 1.8 nM, and 460 pM, respectively, which are two to three orders of magnitude lower than the results obtained by the drop casting technique. The enhancement mechanism of our approach is based on increasing the contact probability with analytes through dynamic dispersion of the Au/GO adsorbents, in contrast to the static diffusion mechanism relied on Brownian motion. We also show that combining the ESPE solution kit with a portable micro-electrochemical workstation can attain the same detection level as HPLC in real urine samples. Rapid and effective analysis of foodborne bacteria is crucial for preventing and controlling bacterial infections. Here, we present the synthesis of a self-reporting molecularly imprinted polymer (MIP) as an inner reference probe (IR), and the in-situ growth of metal-organic frameworks on transition metal carbon nitrides (MOF/Ti 3 C 2 T X -MXene) as a signaling nanoprobe (SP). These advancements are then applied in a ratiometric electrochemical bioassay for Staphylococcus aureus (S. aureus) using a hybrid recognition mechanism. When S. aureus is present, the aptamer-integrated MIP (MIP@Apt) efficiently captures it, followed by binding with SP to form a sandwich structure. This leads to decreased current response of IR (I IR ) and increased current intensity of SP (I sp ), enabling quantification through utilization of the I SP to I IR ratio. The biosensor shows a wide detection range (10-10 8  CFU mL -1 ) and low detection limit of 1.2 CFU mL -1 . The rapid, precise identification and quantification of specific biomarkers, toxins, or pathogens is currently a key strategy for achieving more efficient diagnoses. Herein a dopamine-biotin monomer was synthetized and oxidized in the presence of hexamethylenediamine, to obtain adhesive coatings based on polydopamine-biotin (PDA-BT) on different materials to be used in targeted molecular therapy. Insight into the structure of the PDA-BT coating was obtained by solid-state 13 C NMR spectroscopy acquired, for the first time, directly onto the coating, deposited on alumina spheres. The receptor binding capacity of the PDA-BT coating toward 4-hydroxyazobenzene-2-carboxylic acid/Avidin complex was verified by means of UV-vis spectroscopy. Different deposition cycles of avidin onto the PDA-BT coating by layer-by-layer assembly showed that the film retains its receptor binding capacity for at least eight consecutive cycles. Finally, the feasibility of PDA-BT coating to recognize cell lines with different grade of overexpression of biotin receptors (BR) was investigated by tumor cell capture experiments by using MCF-7 (BR+) and HL-60 (BR-) cell lines. Here we demonstrate for the first time that an antibody-gold nanoparticles (AuNPs)-polymer conjugate thin-film biosensor can easily be fabricated to selectively capture Tau protein. Gold nanoparticles (AuNPs) are employed as sensing elements, thus capitalizing on their propensity to undergo assembly or disassembly in response to the adsorption or conjugation of various biomolecules on their surface, thereby forming robust interactions with the target analyte. We show that the Tau protein in its different aggregation phases can be detected, by restricting the reaction area on the solid thin polymer film and thus reducing the diffusion effects usually encountered in immunosensors. A limit of detection (LOD) of 460 pg/mL was reached, demonstrating a great potential for detecting Tau in aggregation states. We developed an arginine-responsive biosensor by integrating cascade enzymatic reactions into nanochannels functionalized with weak polyelectrolytes, which serve as "reactive signal amplifiers." This approach enhances device performance and broadens the detectable analyte range by generating high local analyte concentrations. The nanofluidic biosensor operates rapidly (<5 minutes) with a low detection limit of 3 μM.

Herein, a "turn-on" peptide-based electrochemical biosensor was developed for the dual-signal sensing of heparin employing Ag/2D CuTCPP(Fe). Our method with high analytical performance was exploited in the accurate quantification of heparin in rat serum throughout its metabolic process.

Arginine is one of the most metabolically versatile amino acids and plays pivotal roles in diverse biological and pathological processes; however, sensitive tracking of arginine dynamics in situ remains technically challenging. Here, we engineer high-performance fluorescent biosensors, denoted sensitive to arginine (STAR), to illuminate arginine metabolism in cells, mice, and clinical samples. Utilizing STAR, we demonstrate the effects of different amino acids in regulating intra- and extracellular arginine levels. STAR enabled live-cell monitoring of arginine fluctuations during macrophage activation, phagocytosis, efferocytosis, and senescence and revealed cellular senescence depending on arginine availability. Moreover, a simple and fast assay based on STAR revealed that serum arginine levels tended to increase with age, and the elevated serum arginine level is a potential indicator for discriminating the progression and severity of vitiligo. Background: Aflatoxin B1 (AFB1) is a widely distributed toxic contaminant in food and poses a serious threat to public health. Therefore, an accurate, simple, cost-effective and on-site assay method is needed for sensitive detection of AFB1. Aptamer shows great potential in the construction of biosensor due to its high specificity and affinity. Multimodal biosensor based on aptamer is highly suitable for the analysis of AFB1 under complex conditions. And the detection results in different modes can be verified with each other, which greatly improves the accuracy of AFB1 detection., Results: Herein, accurate detection of AFB1 was achieved through the development of a multi-mode biosensor integrating electrochemistry, glucosemeter and smartphone-based colorimetric quantification. Streptavidin-Cu 3 (PO 4 ) 2 hybrid nanoflowers (SA-Cu 3 (PO 4 ) 2 HNFs) were synthesised and then conjugated with biotinylated invertase as a signal probe. The electrochemical signal was achieved via intrinsic redox activity. Simultaneously, sucrose could be converted to glucose by the action of invertase, which can cause changes in the glucosemeter signal as well as in the colour of urine glucose test strips. The glucosemeter could complete the signal response in 7 s, and the urine glucose test strips could complete the colour development in 30 s. The detection range of AFB1 by this system in electrochemical mode is 0.001-100 ng/mL, and in glucosemeter mode and smartphone mode is 0.01-50 ng/mL. The limits of detection were 0.49 pg/mL in electrochemistry mode, 5.4 pg/mL in glucosemeter mode and 3.7 pg/mL in smartphone mode., Significance: The successful construction of this multi-mode biosensor demonstrates the advantages of multifunctional nanomaterials and mobile technology. Microneedles represent a miniaturized mechanical structure with versatile applications, including transdermal drug delivery, vaccination, body-fluid extraction, and bio-sensing. Over the past two decades, microneedle-based devices have garnered considerable attention in the biomedicine field, exhibiting the potential for mitigating patient discomfort, enhancing treatment adherence, avoiding first-pass effects, and facilitating precise therapeutic interventions. As an application-oriented technology, the innovation of microneedles is generally carried out in response to a specific demand. Currently, three most common applications of microneedles are drug delivery, fluid extraction, and bio-sensing. This review focuses on the progress in the materials, fabrication techniques, and design of microneedles in recent years. On this basis, the progress and innovation of microneedles in the current research stage are introduced in terms of their three main applications.

The split trehalase biosensor has potential as a versatile diagnostic technology. Split enzymes are engineered proteins, divided into inactive fragments, which can reassemble and regain activity when brought together by an analyte. The split TreA biosensor requires no sample processing and produces stable signals (in the form of glucose). Split trehalase reagents can function in blood, but periplasmic trehalase of E. coli requires blood acidification for maximal activity. The objective of this study was to obtain split trehalase with near physiological pH optimum. For this purpose, periplasmic trehalases of Cellvibrio spp. with higher activity at neutral pH, were split in analogy with the E. coli TreA into hood and catalytic domains. However, these split trehalases displayed self-complementation due to spontaneous reassembly. In contrast, when catalytic domains of Cellvibrio trehalases were combined with E. coli hood domains, these hybrids displayed conditional complementation capacity when split trehalase fragments fused to immunoglobulin-binding protein G (STIGA) were used to quantify immunoglobulin concentrations. Other hybrid combinations of Cellvibrio spp. had increased activity compared to the cognate pairs, albeit with strong self-complementation. A mutagenesis analysis of residues responsible for self-complementation led to uncoupling of self-complementation from allostery. The Michaelis-Menten kinetics of Cellvibrio enzymes and fragment pairs confirmed improved activity of a mutated hybrid pair of Cellvibrio hood and catalytic domains at physiological pH. Chemotaxis, the directed movement of microbes in response to chemical gradients, plays a crucial role in the biodegradation of xenobiotics, such as pesticides, industrial chemicals, and pharmaceuticals, which pose significant environmental and health risks. Emerging trends in genomics, proteomics, and synthetic biology have advanced our understanding and control of these processes, thereby enabling the development of engineered microorganisms with tailored chemotactic responses and degradation capabilities. This process plays an essential physiological role in processes, such as surface sensing, biofilm formation, quorum detection, pathogenicity, colonization, symbiotic interactions with the host system, and plant growth promotion. Field applications have demonstrated the potential of bioremediation for cleaning contaminated environments. Therefore, it helps to increase the bioavailability of pollutants and enables bacteria to access distantly located pollutants. Despite considerable breakthroughs in decoding the regulatory mechanisms of bacterial chemotaxis, there are still gaps in knowledge that need to be resolved to harness its potential for sensing and degrading pollutants in the environment. This review covers the role of bacterial chemotaxis in the degradation of xenobiotics present in the environment, focusing on chemotaxis-based bacterial and microfluidic biosensors for environmental monitoring. The development of high photoactive cathode materials combined with the formation of a stable interface are considered important factors for the selective and sensitive photoelectrochemical (PEC) detection of tetracycline (TC). Along these lines, in this work, a novel type II heterostructure composed of two-dimensional (2D) covalent organic frameworks confined to zero-dimensional (0D) carbon quantum dots (CDs/COFs) film was successfully synthesized using the rapid in-situ polymerization method at room temperature. The PEC signal of CDs/COFs was significantly amplified by improving the light absorption and electron transfer capabilities. Furthermore, a cathodic molecularly imprinted PEC sensor (MIP-PEC) for the detection of TC was constructed through fast in-situ Ultraviolet (UV) photopolymerization on the electrode. Finally, a "turn-off" PEC cathodic signal was achieved based on the selective recognition of the imprinted cavity and the mechanism of steric hindrance increase. Under optimal conditions, the proposed sensor demonstrated a wide linear relationship with TC in the concentration range of 5.00 × 10 -12 -1.00 × 10 -5  M, with a detection limit as low as 6.00 × 10 -13  M. Meanwhile, excellent stability, selectivity, reproducibility, and applicability in real river samples was recorded. Chinese hamster ovary (CHO) bioprocesses, the dominant platform for therapeutic protein production, are increasingly used to produce complex multispecific proteins. Product quantity and quality are affected by intracellular conditions, but these are challenging to measure and often overlooked during process optimization studies. pH is known to impact quality attributes like protein aggregation across upstream and downstream processes, yet the effects of intracellular pH on cell culture performance are largely unknown. Recently, advances in protein biosensors have enabled investigations of intracellular environments with high spatiotemporal resolution. In this study, we integrated a fluorescent pH-sensitive biosensor into a bispecifc (bisAb)-producing cell line to investigate changes in endoplasmic reticulum pH (pH ER ). We then investigated how changes in lactate metabolism impacted pH ER , cellular redox, and product quality in fed-batch and perfusion bioreactors. Our data show pH ER rapidly increased during exponential growth to a maximum of pH 7.7, followed by a sharp drop in the stationary phase in all perfusion and fed-batch conditions. pH ER decline in the stationary phase was driven by an apparent loss of cellular pH regulation that occurred despite differences in redox profiles. Finally, we found protein aggregate levels correlated most closely with pH ER which provides new insights into product aggregate formation in CHO processes. The human brain is a complex organ with an intricate neuronal connectivity and diverse functional regions. Neurological disorders often disrupt the delicate balance among these anatomical compartments, resulting in severe impairments. The available therapeutic options constitute an incomplete solution as many patients respond partially, highlighting the need for continued research into causes and treatments. Bottom-up approaches, like in vitro models, offer insights into brain functions as they recreate the in vivo microenvironment that allows studying how specific features affect physiological and pathological conditions. In this work, we engineered the cortical-striatal-thalamic (CST) circuit, involved in many brain functions such as action initiation and selection, using a three-compartment polymeric device. We characterized the emerging spontaneous electrophysiological activity by using Micro-Electrode Arrays (MEAs). Cortical neurons exhibited complex bursting activity, which influenced the entire circuit. Striatal and thalamic neurons displayed predominantly tonic firing when isolated, while interconnections with the cortex synchronized and organized their neuronal activity, highlighting the cortical pivotal role in bursting activity and information processing. The CST circuit demonstrated self-organization abilities and displayed high entropy values, indicative of dynamic richness and information encoding potential. Furthermore, we proved the CST's involvement in learning and memory. Metal-semiconductor nanozyme of dumbbell Au-CeO 2 with spatially separated heterostructure has cascade superoxide dismutase (SOD)-like and peroxidase (POD)-like activities for superoxide anions detection. It was synthesized by selective growth of CeO 2 at the ends of Au nanorod (Au NR). Taking advantage of the excellent local surface plasmon resonance (LSPR) effect of Au NR, the spatially separated Au-CeO 2 has a higher photothermal effect than the continuously growing core-shell structure of Au@CeO 2 . Meanwhile, the hot electrons of Au NR could transfer to CeO 2 under 808 nm laser irradiation, changing the ratio of Ce 3+ /Ce 4+ redox couples over CeO 2 and facilitating H 2 O 2 decomposition thus enhancing POD-like activity. Based on the SOD-like activity of Au-CeO 2 , superoxide anion (O 2 ·- ) can be transformed into hydrogen peroxide (H 2 O 2 ). Dual-mode including absorbance and temperature sensing detection of O 2 ·- , with the detection range from nM to μM i.e., 0.1-150 μM and LOD of 0.033 μM (S/N = 3) was achieved through the cascade catalysis and photothermal effect. Accurate, multiplex, and ultrasensitive measurement of different colocalized protein markers on individual tumor-derived extracellular vesicles (EVs) and dimerized proteins with multiple epitopes could provide insights into cancer heterogeneity, therapy management and early diagnostics that cannot be extracted from bulk methods. However, current digital protein assays lack certain features to enable robust colocalization, including multi-color detection capability, large dynamic range, and selectivity against background proteins. Here, we report a lithography-free, inexpensive (< $0.1) and ultrasensitive dual-color Membrane Digital ELISA (Mem-dELISA) platform by using track-etched polycarbonate (PCTE) membranes to overcome these shortcomings. Their through-pores remove air bubbles through wicking before they are sealed on one side by adhesion to form microwells. Immunomagnetic bead-analyte complexes and substrate solution are then loaded into the microwells from the opposite side, with >80% loading efficiency, before sealing with oil. This enables duplex digital protein colorimetric assay with beta galactosidase and alkaline phosphatase enzymes. The platform achieves 5 logs of dynamic range with a limit of detection of 10 aM for both Biotinylated β-galactosidase (B-βG) and Biotin Alkaline Phosphatase Conjugated (B-ALP) proteins. We demonstrate its potential by showing that a higher dosage of paclitaxel suppresses EpCAM-positive EVs but not GPC-1 positive EVs from breast cancer cells, a decline in chemo-resistance that cannot be detected with Western blot analysis of cell lysate. Rapid and accurate detection of tumor markers at extremely low levels is crucial for the early diagnosis of cancers. In this work, we developed a portable label-free sliding electrochemical paper-based analytical device (ePAD) using copper/cuprous sulfide@N-doped C@Au nanoparticles (Cu/Cu 2 S@NC@Au) hollow nanoboxes as the signal amplifier for the ultrasensitive detection of alpha-fetoprotein (AFP). Cu/Cu 2 S@NC nanoboxes were synthesized by sacrificial template and interface reaction methods, on which Au nanoparticles were electrodeposited to construct unique heterostructure for effectively capturing anti-AFP and serving as signal amplifier. The designed ePAD incorporates sliding microfluidic paper chips to form a flexible three-electrode system, enabling highly sensitive detection of AFP with a wide linear range of 0.005-50 ng mL -1 and a low detection limit of 0.62 pg mL -1 . The practicality of the prepared ePAD was validated through AFP detection in clinical human serum, which was consistent with chemiluminescence immunoassay. Biomolecular interaction acts a pivotal part in understanding the mechanisms underlying the development of Alzheimer's disease (AD). Herein, we built a biosensing platform to explore the interaction between gelsolin (GSN) and different β-amyloid protein 1-42 (Aβ 1-42 ) species, including Aβ 1-42 monomer (m-Aβ), Aβ 1-42 oligomers with both low and high levels of aggregation (LLo-Aβ and HLo-Aβ) via dual polarization interferometry (DPI). Real-time molecular interaction process and kinetic analysis showed that m-Aβ had the strongest affinity and specificity with GSN compared with LLo-Aβ and HLo-Aβ. The impact of GSN on inhibiting aggregation of Aβ 1-42 and solubilizing Aβ 1-42 aggregates was evaluated by circular dichroism (CD) spectroscopy. The maintenance of random coil structure of m-Aβ and the reversal of β-sheet structure in HLo-Aβ were observed, demonstrating the beneficial effects of GSN on preventing Aβ from aggregation. In addition, the structure of m-Aβ/GSN complex was analyzed in detail by molecular dynamics (MD) simulation and molecular docking. The specific binding sites and crucial intermolecular forces were identified, which are believed to stabilize m-Aβ in its soluble state and to inhibit the fibrilization of Aβ 1-42 . Combined theoretical simulations and experiment results, we speculate that the success of GSN sequestration mechanism and the balance of GSN levels in cerebrospinal fluid and plasma of AD subjects may contribute to a delay in AD progression. Malathion (MALA), a widely used insecticide, even at trace levels exhibits deleterious effects towards respiratory tracts, and nervous system, necessitating its detection. Herein, we have offered non-enzymatic trace level monitoring of MALA using g-C 3 N 4 supported CuO-derived biochar. The present B-CuO/g-C 3 N 4 based electrochemical sensor is synthesized using hydrothermal approach followed by calcination at high temperature. The result unveiled the strong interactions, high charge separation efficiency, significant porosity leading to excellent electrochemically active surface area 9.88 × 10 -5  cm 2 with least charge transfer resistance (R CT ) value of 35.2 K Ω. The B-CuO/g-C 3 N 4 based nanocomposite offered excellent complex formation ability with MALA and square wave anodic stripping voltametric method (SWASV) generates an enhanced electrochemical signal due to oxidation of MALA. Following all necessary optimizations, the sensor was capable to exhibit limit of detection (LOD) value of 1.2 pg mL -1 with R 2  = 0.968. The modified biosensor offered its potential towards detection of MALA in apple and tomato samples with a recovery ranging from 87.64 to 120.59%. The development of more sensitive, stable, and portable biosensors is crucial for meeting the growing demands of diverse and complex detection environments. MOF-based nanozymes have emerged as excellent optical reporters, making them ideal signal donors for constructing multi-signal lateral flow immunoassays (LFIA). In this study, a ZrFe-MOF@PtNPs nanocomposite was synthesized by uniformly depositing platinum nanoparticles (PtNPs) onto the surface of ZrFe-MOFs using an impregnation-reduction method. The ZrFe-MOF@PtNPs exhibited broad absorption spectra, excellent peroxidase-like activity (SA = 21.77 U/mg), high solvent stability, and efficient antibody binding capability. A portable LFIA platform was developed based on ZrFe-MOF@PtNPs and a smartphone for the targeted detection of carcinogenic aflatoxins. This method enabled the readout of colorimetric, fluorescent, and catalytic signals, significantly enhancing detection sensitivity, ensuring result accuracy, and expanding the dynamic detection range. For aflatoxin M1, the calculation of the detection limit of the three signal modes reached as low as 0.0062 ng/mL, which is two orders of magnitude more sensitive than AuNPs-LFIA (0.1839 ng/mL).
