Myopericytoma is defined as a rare benign myxoid tumour that is usually found in distal extremities. However, it may rarely affect the oral cavity. Few cases of oral myopericytoma were reported in the literature. A 56‐year‐old female patient presented with a mouth floor lesion. A wedge biopsy performed from the lesion revealed an atypical round cell proliferation, which may be plasmablastic lymphoma or plasmacytoma. Post‐contrast neck MRI described a well‐defined lobulated lesion at the right sublingual region measuring 14 × 21 mm. Transoral wide local excision of the ulcer was performed, and reconstruction of the defect was done using an inferiorly‐based nasolabial flap. Microscopic examination of the excised swelling revealed a tumour proliferation formed of bland‐looking spindle cells alternating with fibroblasts and intervening vascular spaces. No detected mitosis or necrosis in the examined material. IHC staining of the tumour cells was positive for Caldesmon and alpha smooth muscle actin, while Ki67 was positive in 3% of cellularity. Desmin, ALK, B‐catenin, ERG, CD31, and CD138 showed a negative reaction. This led to the diagnosis of bland spindle cell proliferation suggestive of oral myopericytoma with free all surgical margins. Myoepricytoma is a rare benign tumour that rarely affects the oral cavity. Surgical excision is the mainstay of treatment. One radiotherapy (RT) protocol used for canine oral melanoma (OM) gives 36 Gy total, in six weekly or biweekly fractions (6 Gy × 6). This retrospective study characterizes oncologic outcomes for a relatively large group of dogs treated with this protocol and determines whether radiation dose intensity (weekly vs. biweekly) affected either progression‐free or overall survival (PFS and OS). Dogs were included if 6 Gy × 6 was used to treat grossly evident OM, or if RT was used postoperatively in the subclinical disease setting. Kaplan–Meier statistics and Cox regression modelling were used to determine the predictive or prognostic value of mitotic count, bony lysis, World Health Organization (WHO) stage (I, II, III, or IV), using systemic anti‐cancer therapies, tumour burden at the time of RT (macroscopic vs. subclinical), radiation dose intensity (weekly vs. biweekly), and treatment planning type (manual vs. computerized). The median PFS and OS times for all dogs (n = 101) were 171 and 232 days, respectively. On univariate analysis PFS and OS were significantly longer (p = <.05) with subclinical tumour burden, WHO stages I or II, and weekly irradiation. On multivariable analysis, only tumour stage remained significant; therefore, cases were grouped by WHO stage (I/II vs. III/IV). With low WHO stage (I/II), PFS and OS were longer when irradiating subclinical disease (PFS: risk ratio = 0.449, p =.032; OS: risk ratio = 0.422, p =.022); this was not true for high WHO stage (III/IV). Tumour stage is a prognostic indicator for canine malignant head and neck tumours (MHNT). However, consensus is lacking on nodal staging in the absence of clinically apparent nodal disease (cN0 neck). This prospective observational study aims to determine the diagnostic accuracy of radiopharmaceutical and blue dye for sentinel lymph node biopsy (SLNB), to assess the correspondence between sentinel lymph node (SLN) and clinically expected regional lymph node (RLN) and the impact on staging of the procedure in dogs with MHNT and cN0 neck. Twenty-three dogs with MHNT and cN0 neck underwent tumour excision and SLNB guided by preoperative lymphoscintigraphy and intraoperative gamma-probe and blue dye. Diagnostic performances and detection rate were calculated. Correspondence between SLN and RLN, number of nodes excised, histopathological status of the SLN and complications related to the procedure were recorded. The mapping technique identified at least one SLN in 19/23 dogs, with a detection rate of 83%. The SLN did not correspond to the RLN in 52% of dogs. Multiple nodes were removed in 61% of dogs. At histopathology, eight (42%) dogs had SLN+, of which four differed from the RLN. Only minor self-limiting complications occurred in five (22%) dogs. Radiopharmaceutical and blue dye guidance is accurate (sensitivity 88.9%; specificity 100%) for SLNB in dogs with MHNT and cN0 and allowed the extirpation of unpredictable and/or multiple SLN with minimal morbidity. Incorporation of SLNB in the management of MHNT is desirable to correctly stage the cN0 neck, owing the unpredictability of the lymphatic drainage. [ABSTRACT FROM AUTHOR]

Background: Myeloid sarcoma (MS) is a solid malignant tumour associated with infiltration of immature myeloid precursor cells in an extramedullary site. The term MS has replaced the term granulocytic sarcoma and chloroma, which were used in the past. MS in the oral cavity is very uncommon, with less of 40 cases reported until recently. Case Report: We report the first case, the features, and the diagnostic sequence, of intraoral MS with bilateral palatal involvement, which presented as an initial manifestation, and preceded the appearance of acute myeloid leukaemia (AML). Diagnostic confirmation of such oral mucosal lesions usually requires biopsy, histopathological examination with additional immunohistochemical investigation. MS can occur during the course of acute or chronic myelogenous leukaemia, and myelodysplastic syndromes. In the vast majority of the reported cases, only one site was involved with a single intraoral MS lesion, and the cases predominantly associated with AML. Conclusion: The majority of intraoral MS occurs in patients with known AML, but in some of them, presented as an initial manifestation, and preceded the appearance of the disease. Therefore, clinicians should carefully evaluate all unusual oral lesions of unknown origin.

This patient we report was therefore treated with surgical excision and post-operative radiotherapy.

Case report: A 6‐year‐old speyed female Bull Arab‐cross dog was found to have a small tonsillar nodule. Histological examination revealed a well‐differentiated mast cell tumour (MCT). At initial staging, no evidence of concurrent cutaneous or visceral MCTs was found on a complete blood count, a single lateral thoracic radiograph, abdominal ultrasound or cytology of the spleen and regional lymph nodes. A diagnosis of primary tonsillar MCT was made. At 40 months postoperatively, the dog is alive with no evidence of gross tumour progression, in contrast to some previous reports of rapid disease progression and metastasis in dogs with primary oral MCTs. Mucoepidermoid carcinoma is a malignant epithelial tumour of the salivary glands. It is an unusual type of a tumour in that it varies in its aggressiveness from a low-grade slow growing to high-grade rapidly growing behaviour. Low grade mucoepidermoid carcinomas maybe masqueraded in their clinical presentation, thus need to be looked at with a suspicious eye. We present two intraoral cases of this unique tumour.

Abstract: In the present paper, we compared the histopathological and vibrational analyses of different tissue sections of Oral Squamous Cell Carcinoma (OSCC) at various malignancy grades, in order to unambiguously identify them. To achieve reliable results, healthy and dysplastic samples were also taken into account. FT-IR microspectroscopy is considered an effective tool for studying different molecular structures occurring in tumoral tissues and offers an interesting alternative to detect biochemical changes in a non-subjective way. In particular, on an adequate number of tissue sections affected by three different grades of OSSC (well G1, moderately G2, and poorly G3 differentiated), as well as on dysplastic and healthy tissues (all obtained from surgical resection), the chemical maps were acquired on meaningful areas containing both epithelial and connective structures. The multivariate analysis (Hierarchical Cluster Analysis, HCA, and Principal Component Analysis, PCA), performed separately on epithelial and connective spectral data, afforded to a good segregation for the different morphological structures. By analysing the representative spectra of healthy, dysplastic and tumoral epithelia and connectives, modifications were pin-pointed in the position of bands and absorbance band ratios usually associated with carcinogenesis. Above all, the changes in the protein pattern (with modifications in the length of side chains and in secondary structures), and in carbohydrates and nucleic acids moieties were associated with specific spectral markers of this pathology. The vibrational investigation led to a satisfactory understanding of these lesions so contributing to an early diagnosis, when the sole morphological inspection may result troublesome. Introduction. The oral cavity is one of the largest reservoirs of microorganisms and many pathogenic bacteria have been shown to be associated with the aetiology of oral cancers. Gap Statement. Owing to the complexity of oral microbial communities and their unclear relationship with oral cancer, identification of specific bacteria which contribute to oral cancer is a key imperative. Aim. To compare and investigate the variations in the composition of the bacterial microbiome and its functions between patients with oral tumorous lesions and healthy subjects. Methodology. Twenty-seven samples from individuals with oral tumours (five oral benign tumours and 22 oral squamous cell carcinomas) and 15 samples from healthy subjects were collected. Genomic DNA was extracted and the V3-V5 region of the 16S rRNA gene was sequenced. Subsequently, bioinformatic assessment was conducted using QIIME2, PICRUSt and linear discriminant analysis effect size analyses (LEfSe). Results. The oral microbiota was composed mainly of the phyla Proteobacteria (31.76 %, 35.00 %), Bacteroidetes (30.13 %, 25.13 %) and Firmicutes (23.92 %, 17.07 %) in tumorous and healthy individuals, respectively. Neisseria , Prevotella , Fusobacterium , Streptococcus , Capnocytophaga , Veillonella , Haemophilus , Prevotella , Porphyromonas and Leptotrichia were the most abundant genera. Alpha diversity in the tumour group was significantly greater than that in the healthy group ( P <0.05). Differential analysis of microbes between groups demonstrated a significantly higher number of Neisseria , Veillonella , Streptococcus , Leptotrichia , Lautropia , Sphingopyxis , Sphingobium , Tannerella , Actinomyces and Rothia in healthy controls compared with the tumour group. However, the genera Treponema , Micrococcus , Pseudomonas , Janthinobacterium , Parvimos , Loktanella , Staphylococcus , Acinetobacter , Catonella , Aggregatibacter and Propionibacterium were significantly higher in the tumour group. Pathways related to cancers, cell motility, environmental adaptation, metabolism and signal transduction were enhanced in the tumour group, while functions associated with immune system diseases, replication, repair and translation were significantly enhanced in the healthy group. Conclusion. Variations in the oral microbiota and its functions showed a correlation with oral tumours. The tumour group showed an increased abundance of some multi-drug-resistant and periodontitis-related pathogens. The significantly altered microbiotas may serve as potential biomarkers or inform combination therapy for oral tumours.

Summary: The aim of this study was to investigate the predisposition of the FGFR4 Gly/Arg polymorphism for development of head and neck squamous cell carcinoma (HNSCC) and, furthermore, to examine if the FGFR4 Arg388 allele can be associated with resistance to chemo- and radiotherapy. When analysing 110 tumour biopsies a significant 1.7-fold increased risk to develop HNSCC in individuals carrying the Gly388 allele (p =0.026) was found. Moreover a 2-fold increased risk for males harbouring the Gly388 allele (p =0.031) to develop HNSCC was detected. In 39 HNSCC cell lines the role of the Arg388 allele for radiation and cisplatin sensitivity was investigated. Our results show no role of the Arg388 allele for the radiosensitivity (p =0.996) but indicate a tendency to increased cisplatin sensitivity (p =0.141). When screening the transmembrane and kinase domains in the FGFR4 gene a novel mutation, probably generating a truncated protein lacking exons 14–18, was found in six of eight selected cell lines. Taken together, we have here identified a marker that predicts the risk to develop HNSCC and possibly the sensitivity to cisplatin as well as a novel mutation in the FGFR4 gene. The efficacy of radiotherapy prior to surgery for the treatment of maxillofacial tumours is compared with and without sensitization with haematoporphyrin derivative (HPD). Residual tumour was found in the resected tissue in 92% of unsensitized but only 52% of sensitized cases, suggesting considerable enhancement of tumour destruction by use of the HPD. The article discusses a study conducted by researchers in India to evaluate 23 cases of oral neoplasms in dogs using cytological and histopathological methods. The results showed that five of 23 conditions were epithelial tumours including three papillomas in male, non-descript Rottweiller and Spitz dogs, and one each of basal cell carcinoma and squamous cell carcinoma. It also mentions the four mesenchymal tumours which included two fibrosarcomas and one each of osteoma and liposarcoma.
