Hepatitis C Virus (HCV) is a global health problem with over 170 million infected people worldwide. HCV targets mainly the hepatocytes and its lifecycle is divided into three steps. Viral particle entry leads to the release of viral genomic RNA into the cytoplasm where translation and replication take place. Then, new viral particles are assembled and secreted. During my thesis, we focused on the entry step. Indeed, HCV entry is a complex multistep process that requires numerous cellular factors. First, the virus interacts with attachment factors and then interacts with specific cellular factors including the tetraspanin CD81, the scavenger receptor B1 and the tight junction proteins CLDN-1, -6, -9 and OCLN. The viral particle is next internalized through a clathrin-dependent endocytosis. Finally, fusion at low pH occurs between viral and endosomal membranes leading to the release of the capsid into the cytoplasm. Among cellular factors involved in HCV entry, two members of the tetraspanin superfamily were identified, CD81 and CD63. Tetraspanins are transmembrane proteins able to organize themselves in tetraspanin-enriched microdomains through tetraspanin-tetraspanin interactions and interactions with partner proteins. We tested the involvement in HCV entry of tetraspanins expressed in hepatocytes. Interestingly, we showed that CD151 is also involved in HCV entry. CD151 seems to play an indirect role in HCV entry through regulating membrane organization, especially by preventing CD81 clustering, which is unfavourable to HCV entry. We also studied the effect of some entry factors polymorphisms on viral entry. In a cohort of drug users infected by HIV but not by HCV, we identified two mutations, R209Q in CLDN-6 and P24A in OCLN, not found in control or coinfected patients. It is very rare for drug users patients infected by HIV not to be infected by HCV, suggesting a natural resistance of these patients to HCV infection. We hypothesized that the two mutations identified might impair HCV entry. However, the characterization of these mutations showed that they did not have a functional effect in vitro. Finally, we investigated the effect of Monensin, a polyether ionophore, on viral entry. This molecule increases endosomal pH through its ions transfer properties across cell membranes. We showed that Monensin inhibits HCV entry through impairing the fusion step. Interestingly, HCV cell-to-cell transmission, another poorly characterized entry pathway, was also blocked by Monensin, suggesting that a pH-dependent fusion step is also required for this transmission route. We generated resistant mutants to Monensin, notably the FL-8 mutant carrying two mutations in envelope glycoproteins, namely Y297H in E1 and I399T in E2. Viral particles expressing these two mutations infected cells in a pH-independent manner, displayed different biophysical properties and were not cell-to-cell transmitted. To conclude, these studies highlight the importance of membrane organization for viral entry through cell factors like CD151. We also pointed out that cell-to-cell transmission is a pH-dependent process. In addition, point mutations in E1 and E2 are sufficient to enable HCV to be pH-independent for its entry. However, polymorphisms in CLDN-6 and OCLN seem to have no effect on viral entry., Le Virus de l’Hépatite C (HCV) est un problème majeur de santé publique qui touche plus de 170 millions de personnes dans le monde. Le HCV cible essentiellement les hépatocytes où il effectue son cycle de réplication qui peut-être divisé en trois étapes majeures : l’entrée de la particule virale qui aboutit à la libération de l’ARN viral dans le cytoplasme, la traduction/réplication du génome viral et l’assemblage/sécrétion des particules néosynthétisées. Durant ma thèse, nous nous sommes intéressés à l’étape d’entrée qui est un processus complexe multiséquentiel faisant intervenir de nombreux facteurs cellulaires. Le virus se lie d’abord à la surface cellulaire via des facteurs d’attachement puis interagit avec des facteurs d’entrée spécifiques tels que la tétraspanine CD81, le récepteur scavenger B1 et les protéines de jonctions serrées CLDN-1, -6, -9 et OCLN. La particule virale est ensuite internalisée via une endocytose dépendante de la clathrine, puis sous l’action du pH acide des endosomes les enveloppes virale et cellulaire fusionnent permettant la libération de la capside virale dans le cytoplasme de la cellule infectée. Parmi les facteurs cellulaires impliqués dans l’entrée du HCV, deux protéines de la famille des tétraspanines ont été identifiées, CD81 et CD63. Les tétraspanines sont des protéines transmembranaires capables de former des microdomaines enrichis en tétraspanines en interagissant entre elles ainsi qu’avec des protéines dites partenaires. Nous avons testé l’implication dans l’entrée virale des tétraspanines exprimées dans les hépatocytes. Ainsi, nous avons montré que CD151 semble également important pour l’entrée du HCV. CD151 aurait un rôle indirect sur l’entrée en jouant sur l’organisation membranaire, en empêchant CD81 de former des clusters qui restreignent l’entrée du virus. Nous avons aussi étudié le rôle que pouvait jouer les polymorphismes de facteurs d’entrée sur l’entrée virale. Sur une cohorte de patients toxicomanes infectés par le HIV mais non infectés par le HCV, deux mutations ont été identifiées, R209Q dans CLDN-6 et P24A dans OCLN, qui n’étaient pas présentes dans les populations contrôles ou coinfectées par le HCV. Chez les patients toxicomanes, il est très rare que les patients infectés par le HIV ne le soient pas par le HCV, suggérant une résistance naturelle de ces patients à l’infection. Nous avons émis l’hypothèse que ces deux mutations pouvaient bloquer l’entrée du HCV. Cependant, la caractérisation de ces mutations a montré qu’elles n’avaient pas d’effet fonctionnel in vitro. Enfin, nous avons exploré l’effet de la Monensine, un ionophore polyéther, sur l’infection par le HCV. Cette molécule augmente le pH des endosomes via ses capacités de transfert d’ions à travers les membranes cellulaires. Nous avons montré qu’elle inhibe l’entrée du HCV en bloquant la fusion. De manière intéressante, la transmission cellule-cellule, un mécanisme d’entrée du virus encore mal caractérisé, était également bloquée par la Monensine, suggérant une étape de fusion dépendante du pH pour cette voie. Nous avons généré des mutants de résistance à la Monensine, notamment le clone FL-8 qui portait deux mutations dans les glycoprotéines d’enveloppe, Y297H dans E1 et I399T dans E2. Les particules virales portant ces deux mutations infectaient les cellules de manière indépendante du pH, présentaient des propriétés physicochimiques différentes et ne se transmettaient plus de cellule à cellule. En conclusion ces études soulignent l’importance de l’organisation membranaire pour l’entrée du virus via certains facteurs cellulaires tels que CD151. Par ailleurs, nous avons mis en évidence que la transmission cellule-cellule était dépendante du pH et que des mutations ponctuelles dans E1 et E2 permettent une fusion indépendante du pH. En revanche, des polymorphismes naturels de CLDN-6 et OCLN n’affectent pas leur capacité à supporter l’entrée virale.

Human infrarenal aorta is prone to develop atherosclerotic lesions. Infrarenal aortic blood flow depends on lower limb muscular activity. Blood flow variations cause shear stress variations known to influence endothelial reactivity. The purpose of this study is to compare vascular reactivity of the suprarenal and infrarenal aorta.Endothelium-dependent relaxation to acetylcholine (ACH) and calcium ionophore (Cal), and endothelium-independent relaxation to sodium nitroprussiate (SNP) in 3 aortic segments (thoracic, abdominal suprarenal and infrarenal) harvested from Sprague-Dawley rats (n = 12) were studied in organ chambers.Endothelium-dependent relaxations to ACH were similar for the 3 aortic segments. However, infrarenal aorta endothelial response to Cal was decreased compared to other segments whereas response to SNP was enhanced. Contractility to vasoconstrictor agonist (phenylephrine and KCl) was diminished compared to the suprarenal segments.Endothelial reactivity is not homogeneous throughout the rat aorta. Endothelial reactivity to Cal seems decreased in the infrarenal aorta which suggests a diminished nitric oxide synthase activity. The increased sensitivity of the infrarenal aorta to nitric oxide donor such as SNP suggests a compensatory mechanism.

Recent evidence indicates the presence of p21 Ras and of a protein with characteristics similar to mitogen-activated protein kinases (MAPKs), also known as extracellular signal-regulated kinases (ERKs), in mammalian spermatozoa, suggesting the occurrence of the Ras/ERK cascade in these cells. In the present study we investigated the subcellular localization of ERKs and their biological functions in human spermatozoa. Immunohistochemistry, immunofluorescence, confocal microscopy, and immunoelectron microscopy demonstrated localization of ERKs in the postacrosomal region of spermatozoa. After stimulation of acrosome reaction with the calcium ionophore A23187 and progesterone, ERKs were mostly localized at the level of the equatorial region, indicating redistribution of these proteins in acrosome-reacted spermatozoa. Two proteins of 42 and 44 kDa that are tyrosine phosphorylated in a time-dependent manner during in vitro capacitation were identified as p42 (ERK-2) and p44 (ERK-1) by means of specific antibodies. The increase in tyrosine phosphorylation of these proteins during capacitation was accompanied by increased kinase activity, as determined by the ability of ERK-1 and ERK-2 to phosphorylate the substrate myelin basic protein. The role of this activity in the occurrence of sperm capacitation was also investigated by using PD098059, an inhibitor of the MAPK cascade. The presence of this compound during in vitro capacitation inhibits ERK activation and significantly reduces the ability of spermatozoa to undergo the acrosome reaction in response to progesterone. Since only capacitated spermatozoa are able to respond to progesterone, these data strongly indicate that ERKs are involved in the regulation of capacitation. In summary, our data demonstrate the presence of functional ERKs in human spermatozoa and indicate that these enzymes are involved in activation of these cells during capacitation, providing new insight in clarifying the molecular mechanisms and the signal transduction pathways of this process.

Calcium loading of a rat parotid microsomal fraction is greatly increased by an ATP-regenerating system (phosphocreatine and creatine phosphokinase). This effect is neither a consequence of a rise in the ATP concentration nor of an increased formation of inorganic phosphate originating from hydrolysis of ATP or phosphocreatine. Addition of ADP to the incubation medium provokes an inhibition of Ca2+ influx and a stimulation of Ca2+ efflux by the microsomal fraction. These results suggest that the stimulation of Ca2+ uptake by the ATP-regenerating system is due, at least in part, to an increase of Ca2+ influx and a slowing down of Ca2+ efflux as consequence of a decrease of ADP availability. It is proposed that the effect of ADP on Ca2+ movements could account for the action of certain agonists on intracellular Ca2+ concentration. Moreover, the InsP3 responsive Ca2+ pool was also shown to be enlarged by the ATP-regenerating system without modification of InsP3 sensitivity.

Ces électrodes sélectives permettent la diminution des quantités de prélèvement et sont d'une grande souplesse d'emploi dans les dosages potentiométriques en comparaison avec les photomètres de flamme. Les appareils ont été choisis et modifiés en fonction des exigences et les soins que nécessitent les impédances très élevées ; et les très faibles courants mis en jeu dans ce type de métrologie. Le pont salin a une structure souple permettant entre autre d'isoler l'électrode de référence de l'électrode de mesure afin de laisser libre l'électrode de mesure, ce qui permet de descendre dans les tubes ou sont les solutions échantillons à mesurer. La composition de chacune des membranes sélectives est optimisé en incorporant un ionophore lipophilique électriquement neutre, un sel constitue de cations hydrophiliques et d'anions lipophiliques, un plastifiant de faible constante diélectrique et un solvant apolaire convenablement choisis et à des quantités adéquates dans une matrice de chlorure de polyvinyle de sorte que ces électrodes soient très performantes. Les tests de reproductibilité, de linéarité et de contamination ainsi que les caractéristiques physiques: sélectivité, limite de détection, stabilité de potentiel, temps de réponse, sensibilité et durée de vie sont données pour chacune des électrodes dans les milieux aqueux et les milieux biologiques (sang total, sérum humain et bovin). Les résultats sont comparés à ceux donnés par la photométrie de flamme. Ces électrodes sélectives nous ont permis de mesurer les énergies libres de transfert des chlorures de métaux alcalins dans les mélanges eau + solvant organique (dans le cas présent eau + méthanol)

Adaptation of rumen fermentation to monensin feeding has been studied with rumen-fistulated sheep receiving a daily dose of 30 mg of monensin for a period of 21 d followed by a 28 d period during which 60 mg doses were administered. The ration consisted of 300 g of hay and 300 g of concentrates, fed at 9.00 h and 16.00 h. Monensin was placed in the rumen as an aqueous suspension, just prior to the morning feeding. Monensin infusion was preceded and followed by a period during which no monensin was infused. The following rumen fermentation parameters were determined: methane production, pH, volatile fatty acids (VFA) molar proportions, total volatile fatty acid concentration, lactate and ammonia concentrations and in sacco degradability of hay. Rumen gas expelled through the fistula was collected for 6 h per day and analysed. Total VFA concentration, molar proportions of individual VFA, pH, lactate and ammonia concentration were determined on rumen contents, sampled just prior the administration of monensin and 2 and 6 h later. In vitro incubations of 3 h were carried out with rumen fluid, sampled 1 h after feeding. In vivo and in vitro methane production was decreased by monensin feeding. The molar proportion of propionate in the rumen was increased, while acetate and butyrate percentages were lowered. The total VFA and ammonia concentrations were also decreased by monensin, but pH values were increased. In vitro production of propionate was stimulated by monensin administration and methanogenesis decreased. The organic matter in sacco degradability was not affected, probably because of the time difference between the introduction of bags and monensin in the rumen. These modifications of rumen fermentation persisted as long as monensin was given, indicating that in this experiment, there was no adaptation to the ionophore.

influence of two new ionophore antibiotics, abierixin and cationomycin, on nitrogen digestion in sheep, Le travail presente dans la these a porte sur l'etude de l'action de 2 molecules antibiotiques ionophores (cationomycine et abierixine) sur la digestion de l'azote chez le ruminant. Le choix de ces molecules a ete fait apres que des tests comparatifs aient ete realises in vitro. L'etude a ete realisee a l'aide de techniques in vitro simulant le fonctionnement du rumen, et sur animaux (moutons) porteurs de fistules permanentes a differents niveaux du tube digestif (rumen, duodenum, ileon). Les plans experimentaux choisis ont permis de gommer les eventuels effets remanents des molecules testes et de realiser des analyses statistiques fiables des resultats (methodes des 2 carres latins inverses de gill). La cationomycine, comme le lasalocide, a provoque une baisse de la degradation des proteines alimentaires dans le rumen et une diminution de la synthese nette de proteines microbiennes. En consequence, l'augmentation du flux duodenal de proteines alimentaires compense exactement la baisse du flux de proteines microbiennes. L'apport global d'acides amines dans l'intestin n'est donc pas modifie, en revanche la composition du melange peut etre alteree. L'abierixine a eu un comportement original in vitro en favorisant le flux intestinal de proteines alimentaires sans reduire celui des proteines microbiennes. Cette molecule favorise donc l'apport d'acides amines mis a la disposition de l'animal. Ces resultats n'ont pas ete confirmes in vivo. Des hypotheses sont avancees pour expliquer ces differences dans le comportement de l'abierixine. Elles ouvrent des perspectives interessantes pour des molecules ayant des actions plus ciblees que celles des antibiotiques ionophores traditionnels utilises comme promoteurs de croissance chez les ruminants. Treatment during starvation of D. discoideum amoebae with micromolar amounts of A23187 causes an enhanced aggregation. Cells develop the properties of differentiated, aggregation competent amoebae earlier than untreated populations. Ionophore increases the release of calcium, and prevents the excretion of the phosphodiesterase ihibitor normally released in the media. A23187 suppresses the morphogenetic block of some aggregates mutants, suggesting that the ionophore either activates cAMP synthesis and excretion, or increases the cellular sensitivity to extracellular cAMP signals. This might result from the enhanced mobilisation of intracellular calcium

An in vitro study was conducted to test the action of different ionophore antibiotics and some of their derivatives on the end-products of rumen fermentation. Nigericin and narasin, like monensin and lasalocid, increased the molar proportion of propionate in the V.F.A. mixture and decreased the proportion of acetate and especially that of butyrate. They had no action on total V.F.A. production. The effect observed with derivatives (lasalocid O-acetyl or nigericin-O-acetyl) was generally less. Gas production, chiefly methane, decreased with the addition of antibiotics. This result agrees with the stoichiometric reactions of carbohydrate fermentation in the rumen. The amount of ammonia nitrogen fixed by bacteria was generally lowered by the addition of five antibiotics (nigericin, narasin, monensin, grisorixin and lasalocid) or two derivatives (lasalocid O-acetyl and nigericin O-acetyl), indicating a decrease in bacterial synthesis. In contrast, calcimycin improved the butyrate production at the expense of propionate, and had no effect on gas composition or bacterial synthesis; X 14547 A and alborixin had only a small effect on rumen fermentations. These results, however, must be interpreted with care since the inocula used came from non-adapted sheep.

The benzodiazepines (BZD) are widely used in clinical practice as anxiolytics, muscle relaxants, sedatives and anticonvulsants. Electrophysiological studies have shown a specific interaction of BZD with - aminobutyric acid (GABA), an inhibitory neurotransmitter, of which they enhance the physiological effects. The discovery of saturable and stereospecific binding sites with high affinity for BZD, and their brain distribution indicates a predominantly cortical action of BZD. Furthermore, BZD receptors seem to be linked to GABA receptors--modulating their inhibitory effects--and closely connected to the chloride conductance mechanism associated to the GABA receptor. The discovery of BZD receptors suggests the existence of endogenous ligands. Inosine, hypoxanthine or nicotinamide are reported to have BZD-like activities, in spite of a relative low affinity for their binding sites. Their putative role as endogenous anxiolytics needs to be supported by behavioral studies.

Transport of Na+ and K+ by different carboxylic polyether antibiotics and some of their derivatives was measured in a triphasic water/chloroform/water model system. Monensin, nigericin, narasin and lasalocid proved to be efficient K+ carriers, while grisorixin, alborixin and X 14547A gave lower ionic fluxes. Furthermore, the structural modifications of nigericin, grisorixin and lasalocid reduced the ionophore properties of the corresponding natural metabolite. Monensin was also characterized as a good Na+ carrier. Cation transport results were in keeping with those of a previous study on the end-products of rumen fermentation. In both cases monensin, nigericin, narasin and lasalocid were the most efficient compounds.

In contrast to conventional membranes prepared from a mixture of glyceryl-monooleate (GMO) and decane (GMO-D) which still contain decane in the final state, the membranes formed from a mixture of GMO and squalene (GMO-S) or triolein (GMO-T) contain finally only GMO and are said to be "solventless". The use of the electric current relaxation technique following a voltage jump has allowed us to characterize the effect of the absence of solvent on the various steps of ion transport induced by nonactin. We have measured the rates of formation (kri) of dissociation (kDi) and of translocation (kis) of the ion-ionophore complex as well as the rate of translocation of the ionophore (ks). With the GMO-S and GMO-T membranes, kis is about ten times higher, ks, twice as high, and kDi twice as small than with the GMO-D membranes. The observed effects are always more prominent with the GMO-T membranes. These results can be interpreted in terms of modifications of the energy barriers in the membrane due to the absence of solvent.
