Quand science et musique se rencontrent.Musique et sciences ont souvent été liées dans l’histoire de la musique. Cependant, rares sont les compositeurs qui ont eu l’occasion d’interagir avec un scientifique pour s’imprégner de son travail et l’utiliser comme source d’inspiration. Le projet Muse-IC se propose de donner cette opportunité à des compositeurs d’aujourd’hui, en leur commandant une œuvre inspirée par une découverte scientifique récente. À la suite d’un double appel réalisé auprès de chercheurs et de compositeurs, des compositeurs se sont immergés dans l’univers scientifique de chercheurs afin de repousser les limites de leur démarche créative. Entre 2017 et 2019, six compositeurs ont écrit une pièce inspirée de la découverte d’un chercheur avec lequel ils ont interagit directement. L’aboutissement de ce projet a donné lieu à une conférence-concert à la salle Cortot, à Paris, une occasion unique de sensibiliser le public à l’importance de la recherche fondamentale au travers d’une rencontre originale entre compositeurs, musiciens et chercheurs.

As more and more genomes are being explored and annotated, important features of three-dimensional (3D) genome organization are just being uncovered. In the light of what we know about Polycomb group (PcG) proteins, we will present the latest findings on this topic. The PcG proteins are well-conserved chromatin factors that repress transcription of numerous target genes. They bind the genome at specific sites, forming chromatin domains of associated histone modifications as well as higher-order chromatin structures. These 3D chromatin structures involve the interactions between PcG-bound regulatory regions at short- and long-range distances, and may significantly contribute to PcG function. Recent high throughput "Chromosome Conformation Capture" (3C) analyses have revealed many other higher order structures along the chromatin fiber, partitioning the genomes into well demarcated topological domains. This revealed an unprecedented link between linear epigenetic domains and chromosome architecture, which might be intimately connected to genome function.

Several antimicrobials act by inhibiting the synthesis of nucleic acids (rifamycins, sulfamides, diaminopyridines), modifying their conformation (quinolones, coumarins) or causing irreversible lesions (nitroimidazoles, nitrofurans). The resistance mechanisms are: a reduction in intracytoplasmic accumulation, modification of the target or the production of a new low-affinity target and, more rarely, enzyme inactivation. Although the mechanisms affecting the targets are specific to each family and can lead to high-level resistance, the reduced permeability of the membrane and the increased efflux are non-specific and result in low-level cross-resistance between several families. The genetic mediation is usually chromosomal for rifamycins and quinolones, although plasmid-mediated resistant genes have been observed. On the other hand, for sulfamides and trimethoprim, plasmid-borne genes are frequent. Resistance to nitroimidazoles and nitrofurans is still not widely understood.

Most viruses use the mRNA-cap dependent cellular translation machinery to translate their mRNAs into proteins. The addition of a cap structure at the 5' end of mRNA is therefore an essential step for the replication of many virus families. Additionally, the cap protects the viral RNA from degradation by cellular nucleases and prevents viral RNA recognition by innate immunity mechanisms. Viral RNAs acquire their cap structure either by using cellular capping enzymes, by stealing the cap of cellular mRNA in a process named "cap snatching", or using virus-encoded capping enzymes. Many viral enzymes involved in this process have recently been structurally and functionally characterized. These studies have revealed original cap synthesis mechanisms and pave the way towards the development of specific inhibitors bearing antiviral drug potential.

Background: MicroRNAs (miRNAs), small RNA molecules of approximately 22 nucleotides, have been shown to be up- or downregulated in specific cell types and disease states. These molecules have become recognized as one of the major regulatory gatekeepers of coding genes in the human genome. Content: We review the structure, nomenclature, mechanism of action, technologies used for miRNA detection, and associations of miRNAs with human cancer. miRNAs are produced in a tissue-specific manner, and changes in miRNA within a tissue type can be correlated with disease status. miRNAs appear to regulate mRNA translation and degradation via mechanisms that are dependent on the degree of complementarity between the miRNA and mRNA molecules. miRNAs can be detected via several methods, such as microarrays, bead-based arrays, and quantitative real-time PCR. The tissue concentrations of specific miRNAs have been associated with tumor invasiveness, metastatic potential, and other clinical characteristics for several types of cancers, including chronic lymphocytic leukemia, and breast, colorectal, hepatic, lung, pancreatic, and prostate cancers. Summary: By targeting and controlling the expression of mRNA, miRNAs can control highly complex signal-transduction pathways and other biological pathways. The biologic roles of miRNAs in cancer suggest a correlation with prognosis and therapeutic outcome. Further investigation of these roles may lead to new approaches for the categorization, diagnosis, and treatment of human cancers.

Protein synthesis is an efficient and vital mechanism mediated by the ribosomes. In all living organisms, it allows an accurate correspondence between the genetic information and the newly synthesized polypeptides. The process of translation needs accurate quality-control systems to ensure the correct readout of the genetic data at the cellular level. Among them, bacteria did develop a specific mechanism referred to as "trans-translation", ensuring the recycling of stalled translating ribosomes and the degradation of incomplete nascent proteins when incomplete messenger RNAs (mRNAs) are translated. tmRNA (transfer messenger RNA) and SmpB (small protein B) are the main components of that process. Recent biochemical, genetic and structural data provide insights on how the tmRNA-SmpB complex accomplishes its duty, allowing a deeper understanding of the intricate links between trans-translation, bacterial survival and virulence.

It has long been known that gene regulation is mostly achieved via protein-nucleic acid interactions. However, the role of RNA factors in gene control has been recently growing given the implication of new RNA-based gene regulation mechanisms such as microRNAs and related short-interfering RNAs gene expression inactivation mechanisms. Recent studies have demonstrated that the involvement of RNA in fundamental gene-control processes is even more extensive. Prokaryotic messenger RNAs carry highly structured domains known as riboswitches within their 5'-untranslated regions. Each riboswitch is able to bind with high specificity their cellular target metabolite, without the involvement of a protein cofactor. Upon metabolite binding, the messenger RNA undergoes structural change that will ultimately lead to the modulation of its genetic expression. Riboswitches can alter gene expression at the level of transcription attenuation or translation initiation, and can up- or down-regulate gene expression by harnessing appropriate changes in the mRNA structure. Here, we provide an overview of the adenine riboswitch, one of the smallest riboswitch and one of the few that activates gene expression upon ligand binding. Several crystal structures have been obtained for the ligand-binding domain of this riboswitch providing us with an unprecedented glimpse about how riboswitches use their ligand to regulate gene expression. Moreover, mechanistic studies have recently shed light on the transcriptional regulation mechanisms of the adenine riboswitch suggesting that riboswitches may rely on the kinetics of ligand binding and the speed of RNA transcription, rather than simple ligand affinity. Riboswitches are particularly interesting because RNA-ligand interactions are potentially very important in the elaboration of antimicrobial agents.

Bacterial sterilization by the technology of plasma in post-discharge shows a growing interest. The main appeal of this new process resides in its action at dry and low temperature (60 degrees C). This technology would be therefore useful for the complex medical equipment, sensitive to the oxidization, humidity and/or requiring a temperature lower than 60 degrees C. The objective of this survey is to demonstrate the activity of an atomic flux emanating a plasma of pure molecular nitrogen on the bacterial DNA: does the plasma of nitrogen damage the genetic material?The bacteria tested (Bacillus stearothermophilus, Staphylococcus aureus MRSA and Helicobacter pylori) are cultivated on suitable agar, and the bacterial DNA is extracted from every CFU by the technique of the columns (High Pure PCR Template, Roche). Every quantity of DNA extracted is diluted in 1 ml pure water. Then, 50 microl of each of these solutions of DNA are laid down in sterile Nunclon's plates holes, which undergo an advanced emptiness cycle during 60 minutes. The DNA residues will be then introduced during 40 minutes in a plasma sterilization surrounding wall (Plasmalyse), Satelec) where the debit of nitrogen, the pressure and the temperature are adjusted respectively to 1 L x min(-1), 5 Torrs and 60 degrees C. The DNA so ''plasmalysé'', is recovered then by aspiration in 500 mul pure water and processed to undergo an amplification/detection by Real-Time PCR (LightCycler2.0, Roche). The DNA ''plasmalysé'' will be compared to the intact DNA control(1), to the DNA control control(2) having undergone the cycle of emptiness solely, as well as to the DNA control(3) solely heated to 60 degrees C during 40 min.The amplification curves demonstrated that the only advanced emptiness and the only heat don't have any activity on the bacterial DNA. On the other hand, the DNA ''plasmalysé'' shows a deterioration of the amplified sequences.The genomic bacterial DNA, once extracted, is damaged by the gaseous flux of nitrogen plasma. A new sterilization process of the medical material will presumably impose itself in medium-term.

Aptamers are short oligonucleotides selected from large combinatorial pools of sequences for their capacity to bind to many different targets ranging from small molecules (amino acids, antibiotics...) to proteins or nucleic acid structures. Aptamers present the same high specificity and affinity for their targets as antibodies. In addition to efficient binding, aptamers have been shown in many cases to display an inhibitory activity against their targets. Many aptamers are now being developed against biomedical relevant targets, and one aptamer that inhibits the human VEGF165 already received approval for the treatment of age-related macular degeneration. Here we discuss the principles and the practical way of selecting aptamers (SELEX technology) as well as the structural basis for their performance as ligands. A wide scope of applications is described - aptamers have been used as tools for studying nucleic acids/proteins interactions, detecting, purifying or imaging target molecules, regulating gene expression - and includes recent developments of aptamers for therapy and diagnosis.

Fragile X syndrome is the most common form of inherited mental retardation. This X-linked disease is due to transcriptional silencing of the Fragile Mental Retardation 1 (FMR1) gene and the absence of its gene product, FMRP. This protein is an RNA-binding protein present in mRNP complexes associated with the translation machinery and is thought to be a key player in the control of mRNA transport in neurons. However, the exact role of FMRP in translation remains unclear. Two homologous proteins, FXR1P and FXR2P, are also found in RNP complexes containing FMRP, suggesting that FMRP's functions are much more complex than first thought. The molecular mechanisms altered in cells lacking FMRP still remain to be elucidated, as well as the putative roles of FXR1P and FXR2P as compensatory molecules. Here, we review the various possible functions of FMRP in RNA localization and transport in highly differentiated cells containing dendritic extensions such as neurons.

The discovery of DNA's double helix 50 years ago was the founding event of molecular biology. It was also the moment that forged the reputation of two of biology's most compelling figures, no doubt in perpetuity. However, Jim Watson and Francis Crick were not the only players of this outstanding fest whose certain circumstances remain today rather singular.

The peculiar sequence of telomeric DNA, composed of repetitions of the GGTTAG motif allows the formation of an unusual DNA conformation based on guanine-quadruplex (G-quadruplex). Small molecules that bind and stabilize telomeric DNA under its G-quadruplex conformation are able to impair telomerase activity. Several recent reports have shown that G-quadruplex ligands could block telomerase activity in cancer cells and represent a new experimental approach to limit cancer growth. The intracellular existence of G-quadruplex structure is still controversial, since no direct proof allowed to establish its reality. Many sequences of nucleic acids in the mammalian genome are able to form a G-quadruplex in vitro and several proteins have been described to interact in vitro with G-quadruplex. These data indicated that G-quadruplex are members of a family of target structures larger than that initially described at telomeres, and raised the question of the selectivity and therapeutic index of their ligands in the context of an antitumor therapy.

Electron microscopy of DNA, either free or complexed with ligands, allows the analysis of local conformational variations along individual molecules. Electron microscopy is unique, in that it has the capacity to determine the average behaviour of a population of molecules observed individually, and can thus provide a better appreciation of variability within the series of molecules than biophysical or biochemical methods. Very encouraging results have been obtained by cryoelectron and near-field microscopies, especially atomic force microscopy, in parallel with traditional techniques for visualizing DNA molecules adsorbed onto a support film. Differences in sample processing procedures and image formation modes render these 3 types of microscopies complementary. The torsional stress of a DNA molecule together with a local curvature induced by the protein MC1 from archaebacteria, can be detected within minicircles comprising 207 base pairs.

The concept of antigen internal image was applied to the production of catalytic antibodies. An antibody raised in rabbits to DNase (Ab1), acted as a competitive inhibitor of the catalysis, and thus was assumed to contain anti-active site Ab. This Ab1 was used to elicit a polyclonal anti-idiotypic antibody (Ab2). This later exhibited a DNA recognition specificity, suggesting the existence of structural internal images mimicking the conformation of the active site. Moreover, Ab2 were able to hydrolyse DNA, indicating the existence of internal images mimicking the enzymatic activity of DNase. Consequently, a strategy can thus be considered using the idiotypic way, for the production of abzymes in the form of internal images of enzyme active sites.

We have determined the nucleotide sequence of a DNA segment carrying the entire 15S ribosomal RNA gene of yeast mitochondrial genome. Many stretches of sequence are present which are homologous to the E. coli 16S ribosomal RNA gene. The gene sequence can be folded into a secondary structure according to the [1] model on bacterial ribosomal RNAs. The structure reveals a striking similarity between the two RNAs despite the large difference in their base compositions. In the middle of the gene, we found a guanine-cytosine rich sequence that is also present in several other regions of the mitochondrial genome.

This review tentatively recaptitulates most of the known data concerning the genome of oncornaviruses (Retroviruses). The first part deals with the structural features of the 60-70S RNA and its constituting subunits and other cellular RNAs which are present inside the viruses. The second part describes the viral RNA species which have been found in various cell systems including infected producer cells, transformed and non producer cells, normal and inductible cells. A special part is devoted to viral RNAs isolated from human cancer cells.

The hot phenol extraction of nuclic acids reveals the presence of small amounts of nonintegrated SV 40 DNA in transformed syrian hamster or mouse cells. The extractibility of the viral DNA is influenced by its conformation; SV 40 DNA, form I is preferentially extracted by contrast with form III DNA.

Miller Beatty's technique was adapted to the study of definite chromatin fractions (nucleolar and nonnucleolar chromatin) isolated from Mammalian cells. The ultrastructural organization of the transcriptional complexes obtained depended on the nuclear compartment studied. In isolated nucleoli, there were "Christmas-tree"-like figures. In nonnucleolar chromatin, the figures were different from the former by the internal structure of the RNP fibrils being synthesized.

Dihedral angles and atomic coordinates are given together with a stereo view. Curves of the calculated diffracted intensities are compared with experimental data.

Electron microscopy study of RNA extracted from Lumbovirus shows the existence of circular molecules, which are not susceptible to thermal or chemical denaturation. These facts suggest that the RNA is present inside the virion in a covalently closed circular form.

We present a model for the secondary structure of 16S ribosomal RNA from E. coli. This model has been deduced by restricting the total number of theoretical base pairings using the following criteria: (1) susceptibility of residues towards enzymatic probes that are specific for either paired or single stranded regions; (2) reactivity of certain residues to chemical modification; (3) evidence for medium and long range interactions; (4) comparative analysis of ribosomal RNA sequences from other organisms.
