The vacuole of the yeast Saccharomyces cerevisiae plays an important role in nutrient storage. Arginine, in particular, accumulates in the vacuole of nitrogen-replete cells and is mobilized to the cytosol under nitrogen starvation. The arginine import and export systems involved remain poorly characterized, however. Furthermore, how their activity is coordinated by nitrogen remains unknown. Here we characterize Vsb1 as a novel vacuolar membrane protein of the APC (amino acid-polyamine-organocation) transporter superfamily which, in nitrogen-replete cells, is essential to active uptake and storage of arginine into the vacuole. A shift to nitrogen starvation causes apparent inhibition of Vsb1-dependent activity and mobilization of stored vacuolar arginine to the cytosol. We further show that this arginine export involves Ypq2, a vacuolar protein homologous to the human lysosomal cationic amino acid exporter PQLC2 and whose activity is detected only in nitrogen-starved cells. Since their discovery in 1975, by Köhler and Milstein, monoclonal antibodies (mAbs) world has significantly evolved and they currently hold a prominent place in cancers care. Today, the mAbs, having a marketing authorization or in clinical trial, are all IgG class (IgG1). However, this Ab class showed limitations on its use, and the study of other isotypes, such as IgA, could be interesting. Unlike IgG, IgA, original isotype particularly because of their heterogeneity in molecular forms, remains understudied. In this work, we propose a radiolabeling of monomeric, polymeric and secretory IgA with 99mTc by an indirect method, involving 2-iminothiolane and tricarbonyl core. Biodistribution of radiolabeled monomeric and polymeric IgA was evaluated, after intravenous administration, in healthy animals and in mucosal tumor-bearing animals. These studies have allowed us to glimpse the IgA diagnostic potential, but also their interest in targeted therapy of tumors with mucosal localization. Moreover, thanks to their enzymatic strength and retranscytosis, a new administration route of mAbs could be developed. In this context, secretory IgA were administered orally in preliminary biodistribution studies., Depuis leur découverte en 1975 par Köhler et Milstein, le monde des Ac monoclonaux a beaucoup évolué. Ils occupent actuellement une place prépondérante dans la prise en charge de nombreux cancers. De nos jours, les Ac monoclonaux, ayant une AMM ou en essai clinique, sont tous de classe IgG voire IgG1. Cette classe d’Ac a cependant montré des limites à son utilisation, et l’étude d’autres isotypes d’Ac, comme les IgA, pourrait être intéressante. Les IgA, isotype d’Ac particulier en raison notamment de leur hétérogénéité dans les formes moléculaires, demeurent peu étudiées à l’instar des IgG. Dans ce travail, nous proposons un radiomarquage des IgA monomériques, polymériques et sécrétoires, avec le 99mTc par une méthode indirecte impliquant le 2-iminothiolane et le cœur tricarbonyl. Par le biais de ce radiomarquage, la biodistribution des IgA monomériques et polymériques après administration i.v. a été évaluée chez l’animal sain et chez l’animal porteur de tumeur à localisation muqueuse. Ces études nous ont permis d’entrevoir le potentiel diagnostique des IgA, mais aussi leur intérêt en thérapie ciblée de tumeurs à localisation muqueuse. D’autre part, grâce à leur résistance enzymatique et au phénomène de retranscytose, une nouvelle voie d’administration des Ac monoclonaux pourrait être développée. Dans cette optique, des IgA sécrétoires ont été administrées par voie orale lors d’études préliminaires de biodistribution.

Development of fluorine 18 labeled radiotracer of 5-HT6 and 5-HT7 receptors for PET imaging (positron emission tomography) allows the study of those receptors in various neurodegenerative diseases such as schizophrenia and Alzheimer disease. Description of structures and pharmacophores in literature led to pyrrolidine derivatives for 5-HT7 receptors and quinolones for 5-HT6. After their synthesis, 7 radioligands of 5-HT7 receptors have been studied by autoradiography and μPET. These radioligands have shown interesting binding on rat, with more or less selectivity for the receptor. 14 ligands of 5-HT6 receptors have been synthesized and 4 have been radiolabeled to select 2FNQ1P as a selective radioligand toward 5-HT2A. First PET images on cat have shown a selective binding in 5- HT6 rich area in brain. Pursue of biological studies, in collaboration with CERMEP – Imagerie du vivant will give more information on those new radioligands, Le développement de radiotraceurs (18F) des récepteurs de la sérotonine 5-HT6 et 5-HT7 pour l'imagerie TEP (tomographie par émission de positons) permettrait d'étudier la fonction et l'implication de ces récepteurs dans des maladies neurodégénératives telles que la schizophrénie ou la maladie d'Alzheimer. A partir des structures et pharmacophores déjà décrits dans la littérature, nous nous sommes orientés vers des dérivés pyrrolidiniques pour les récepteurs 5-HT7 et quinolines pour les récepteurs 5-HT6. 7 radioligands des récepteurs 5-HT7 marqués au fluor 18 ont pu être étudiés par autoradiographie et imagerie μTEP sur le rat et ont montrés des fixations intéressantes, mais avec une sélectivité moyenne du récepteur. 16 ligands du récepteur 5-HT6 ont été synthétisés et 4 d'entre eux ont été radiomarqués afin d'identifier le 2FNQ1P comme radioligand sélectif vis-à-vis du récepteur 5-HT2A (principal récepteur en compétition). Les premières images TEP réalisées sur le chat ont montrées un marquage sélectif dans les zones cérébrales riches en 5-HT6. La poursuite des études biologiques menées en collaboration avec le CERMEP – Imagerie du vivant permettront d'approfondir les caractéristiques de ces nouveaux radioligands synthétisés

Ce projet s'inscrit dans le développement d'une nouvelle stratégie de radiomarquage au fluor-18 de motifs perfluorés incorporés dans des sondes chimiques (EF3, EF5) visant à détecter et quantifier l'hypoxie tumorale de manière non invasive par tomographie par émis-sion de positrons. La méthode sélectionnée s'est portée sur des précurseurs soufrés (dithioester, dithiocétal) ou azotés qui, mis en présence d'un agent de perfluoration et d'un oxydant adéquats, génèrent un groupement trifluorométhyle ou difluorométhyle par fluorodésulfurisation oxydative. Cette méthode est efficace sur des composés comportant une fonction phtalimide. Des travaux antérieurs ayant validé cette stratégie pour le groupement CF3, notre tra-vail a tout d'abord consisté à synthétiser des précurseurs adéquats pour le radiomarquage du motif C2F5. Les premiers essais de radiomarquage ont également pu être effectués. Enfin, la dernière partie de notre travail s'est attachée à valoriser les résultats de synthèse obtenus en série fluorée. Deux procédures ont été envisagées: marquage au niveau du CF3 (précurseur difluo-ré) et marquage au niveau du CF2 (précurseurs dithiocétal et oximes trifluorés). La synthèse du précurseur difluoré (Beta-phtalimido-alpha,alpha-difluoro-dithioester) nous a amené à développer une nouvelle voie d'accès à l'alpha,alpha-difluoro-beta-alanine (3 étapes), ainsi que la première synthèse de 3,3-difluoro-azétidine-2-ones N-protégées non substituées en position C4 (3 étapes). Nous avons également mis au point une nouvelle méthode de synthèse de dithioesters au départ de thio-w-lactames. Le précurseur difluoré a été obtenu en 7 étapes avec un rendement global de 7%. Les précurseurs trifluorés ont pu être obtenus facilement à partir de la trifluoromé-thylcétone correspondante (3 étapes au total). Ces produits ont été obtenus avec des rende-ments globaux de 56 à 84 %. Les radiosynthèses effectuées ne nous ont pas permis d'atteindre le composé penta-fluoré voulu. Nous avons pu identifier des produits marqués tétrafluorés, mais sommes resté impuissant à introduire le dernier atome de fluor. Enfin, nous avons étendu les résultats de synthèse obtenus en série fluorée. Nous avons ainsi développé une nouvelle voie de synthèse pratique et efficace pour la préparation d'w-amino-dithioesters non fluorés, protégés sous formes de dérivés N-benzoyle ou N-p-nitrophényle, au départ de γ-, δ-, and ε-lactames. Cette méthode a ensuite pu être étendue au départ d'un lactame. The aim of this study was to develop a new strategy for the fluorine-18 radiolabelling of perfluorinated groups incorporated in chemical probes (EF3, EF5), in order to assess non-invasively tumoral hypoxia by means of positron emission tomography. This technique uses sulfur (dithioesters and dithioketals) and nitrogen precursor-which, in combination with the right perfluorination agent and the right oxidant, lead to trifluoromethyl or difluoromethyl groups via oxidative fluorination. This strategy has proved efficient on aliphatic compounds bearing a phtalimide group. Since previous studies have already validated this approach for the CF3 group, we first aimed at synthesizing adequate precursors for the radiolabelling of the C2F5 moiety. First attempts of radiolabelling have also been carried out. Finally, the last part of our work was devoted to highlighting the synthesis results obtained for fluorinated compounds. Two strategies have been considered: labelling at the CF3 level (difluorinated precur-sor) and labelling at the CF2 level (trifluorinated dithioketal and oxime precursors). The synthesis of the difluorinated precursor (Beta-phtalimido-alpha,alpha-difluoro-dithioester) brought us to develop a new way to access alpha,alpha-difluoro-beta-alanine (3 steps), and the first synthesis of N-protected 3,3-difluoro-azetidin-2-ones bearing no subtituents in C4 position. We also developped a new synthesis of dithioesters, starting from thiolactams. The difluorinated precursor was obtained in 7 steps, with an overall yield of 7%. The trifluorinated precursors could be easily obtained from the corresponding trifluoromethylketone (3 steps in total). These compounds were produced with overall yields ranging from 56 to 84%. The radiosynthesis performed did not allow us to gain acces to the targetted penta-fluorinated compound. With sulfur precursors, we have been able to identify labelled tetra-fluorinated compounds, but our attempts to introduce the last fluorine atom were unsuccesful. In the end, we have expanded the scope of the dithioester synthesis developped for fluorinated compounds to non-fluorinated products. We thus disclosed a novel and practical route for preparing w-amino dithioesters, protected as N-benzoyl or N-p-nitro-phenyl and trifluoroacetyl derivatives, starting from γ-, δ-, and ε-lactams. The aim of this thesis is to study the reactivity and exchange phenomena between proteins at a solid/liquid interface. It includes a description of the different aspects related to the proteins used (physicochemical characteristics of fibrinogen and immunoglobulin G, structure, function) and to the structure of the adsorbent (silica). It summarises some important properties of adsorption, desorption and exchange of proteins at the interface, and discusses all the experimental techniques and methods used. The last three chapters concern (1) the study of the reactivity of antibodies towards their antigens, when the latter are adsorbed directly on a solid surface, (2) the study of the reactivity of the same antigen/antibody systems, when the antigen is adsorbed by an exchange process on a pre-adsorbed protein layer, and (3) a dynamic and quantitative analysis of the heterogeneous exchange process between adsorbed immunoglobulin G and fibrinogen in solution., Le but de cette thèse est d'étudier les phénomènes de réactivité et d'échange entre des protéines à une interface solide/liquide. Elle comprend une description des différents aspects liés aux protéines utilisées (caractéristiques physicochimiques du fibrinogène et des immunoglobulines G, structure, fonction) et à la structure de l'adsorbant (silice). Elle résume certaines propriétés importantes concernant l'adsorption, la désorption et l'échange de protéines à l'interface, et présente la liste de toutes les techniques et méthodes expérimentales utilisées. Les trois derniers chapitres concernent (1) l'étude de la réactivité d'anticorps vis-à-vis de leurs antigènes, lorsque ces derniers sont adsorbés directement sur une surface solide, (2) l'étude de la réactivité des mêmes systèmes antigène/anticorps, lorsque l'antigène est adsorbé par un processus d'échange sur une couche protéique pré-adsorbée, et (3) une analyse dynamique et quantitative du processus d'échange hétérogène entre des immunoglobulines G adsorbées et du fibrinogène en solution.
