PTB-Mitteilungen. Band 132 (2022), Heft 2. ISSN 0030-834X, Mit der Energiewende hat Deutschland sich das Ziel gesetzt, innerhalb weniger Jahrzehnte ein grundlegend neues, nachhaltiges, dezentrales Energiesystem zu schaffen, das nahezu ohne CO2-Emissionen auskommt. Diese Transformation vor dem Hintergrund aktueller geopolitischer Entwicklungen und seiner Auswirkung auf die Energiesicherheit in Deutschland und Europa ist eine Mammutaufgabe. Als nationales Metrologieinstitut will die PTB alle Akteure – von der Industrie über die Politik bis zum Verbraucher – dabei unterstützen. Denn verlässliche Messungen sind die Voraussetzung für Sicherheit, Effizienz und Verbraucherschutz. Bereits heute hat die PTB Mess- und Kalibriermöglichkeiten im Bereich Solar- und Windenergie entwickelt, die zum Teil weltweit einmalig sind. Doch zu einer erfolgreichen Energiewende gehören auch leistungsfähige Übertragungs- und Verteilnetze, Batterien und andere Speicherelemente, der Aufbau von Wasserstofftechnologien sowie Energieeffizienz. Gemeinsam mit Industriepartnern entwickeln PTB-Wissenschaftlerinnen undWissenschaftler auch hier geeignete Messverfahren und Standards. Besonderes Augenmerk liegt auf der Kopplung von Energiesektoren, der Stabilisierung des Stromnetzes und dem Energieträger Wasserstoff, der eine Schlüsselrolle im Energiesystem der Zukunft spielen wird. Das hier vorliegende Heft der PTB-Mitteilungen bietet mit insgesamt acht ausgewählten Fachartikeln einen hervorragenden Überblick über einzelne Aktivitäten und Ergebnisse aktueller Arbeiten der PTB für die Energiewende.

HIV- und Tuberkulose (TB)-koinfizierte Patienten können nach Beginn einer antiretroviralen Therapie (ART) als Komplikation ein Immunrekonstitutionssyndrom (IRIS) entwickeln. Dabei kommt es zu einem Neuauftreten oder einer Verschlechterung von klinischen Symptomen oder radiologischen Befunden im Zusammenhang mit der TB. Präsentieren kann sich ein IRIS entweder als eine plötzliche Verschlechterung der Infektion nach ART-Beginn („paradoxical/paradoxes IRIS“) oder durch ein Demaskieren einer vorher klinisch inapparenten und unbehandelten Infektion („unmasking/demaskierendes IRIS“). Aufgrund nicht einheitlich definierter Diagnosekritierien kann die Diagnosestellung im klinischen Alltag eine Herausforderung darstellen. Ziel dieser Dissertation war es deshalb, klinische Charakteristika, Risikofaktoren und ggf. protektive Faktoren für die Entwicklung eines IRIS bei TB zu identifizieren. Diese Ergebnisse sollten zu besseren Verständnis und Vorhersagen von IRIS im Zusammenhang mit TB beitragen. Dazu wurden retrospektiv Daten von 52 Patienten, die im Zeitraum 01.01.2010 - 30.06.2016 mit einer HIV-Infektion und zur Behandlung einer aktiven Tuberkulose stationär in der Infektiologie des Uniklinikums Frankfurts aufgenommen wurden, pseudonymisiert erfasst. Es wurden u. a. Arztbriefe, Laborbefunde, Fieberkurven und Visitenberichte aus dem Patientenmanagementprogramm „ORBIS“, der Datenbank „epidem“ und des Laborinformationsprogramms „Nexus swisslab“ des Uniklinikums Frankfurt genutzt. Zu den Parametern gehörten neben patientenspezifischen Daten wie Alter und Geschlecht unter anderem auch Routinelaborparameter, Serologien, Art der TB, genaue ART und TB-Therapien und Laborparameter, die zur Beurteilung einer Entwicklung der Immunrekonstitution und der virologischen Suppression hinweisend sind. Dazu zählen insbesondere HI-Viruslastwerte, CD4- und CD8-Zellzahlen für einen Zeitraum von 48 Wochen ab ART-Beginn. Zur Untersuchung der unterschiedlichen IRIS-Arten wurden die Patienten in zwei Gruppen aufgeteilt: bereits mit einer ART vorbehandelte Patienten, bei denen somit die Entwicklung eines demaskierendem IRIS möglich war, und ART-naive Patienten, die theoretisch ein paradoxes IRIS entwickeln konnten. Durch Beurteilung des Krankheitsverlaufes und unter spezieller Berücksichtigung der HI-Viruslast im Verlaufe der ART wurde nach der IRIS-Definition von French et al. (2004) festgelegt, ob ein IRIS vorlag. Bei unklaren Fällen erfolgte eine gemeinsame Besprechung und definitive Einteilung im kliniksinternen Kolloquium. Schließlich wurde die statistische Auswertung mithilfe des Statistikprogramms „bias“ durchgeführt und dabei jeweils die „IRIS“ mit der „Nicht-IRIS“-Gruppe verglichen. Angewandt wurden der Exakte Fisher-Test für kategorische und der Wilcoxon-Mann-Whitney-Test für numerische Variablen. Die paradoxe IRIS-Inzidenz betrug 29,7 %, die demaskierende IRIS-Inzidenz 46,7 %. Am häufigsten präsentierte sich das IRIS in der Frankfurter Kohorte mit Fieber, am zweithäufigsten als Lymphadenopathie oder mit respiratorischen Beschwerden. Für sowohl Patienten mit paradoxem als auch demaskierendem IRIS zeigte sich ein signifikant längerer Krankenhausaufenthalt als für Patienten, die kein IRIS entwickelten. Sonst wurden für das demaskierende IRIS keine weiteren statistisch signifikanten Parameter gefunden, u. a. aufgrund Limitationen wie der sehr kleinen Studienpopulation (15 Patienten). Patienten mit paradoxem IRIS hatten zudem eine signifikant höhere Rehospitalisierungsrate (63,3 % vs. 15,4 %; p= 0,006), was die klinische Relevanz aufzeigt. Außerdem korrelierten extrathorakale TB-Manifestationen (p= 0,025), niedrige CD4+-Lymphozyten-Zellzahl (p= 0,006) und hohe Viruslast (p= 0,017) vor ART-Beginn mit einer paradoxen TB-IRIS-Entwicklung. Diese Patienten sollten folglich nach ART-Beginn besonders engmaschig klinisch kontrolliert werden, da bei ihnen ein IRIS wahrscheinlicher ist. Ebenfalls statistisch signifikant zeigte sich erhöhte Laktatdehydrogenase (LDH) und erniedrigtes Albumin im Serum. In Kombination mit den davorgenannten Parametern könnten die Werte dabei behilflich sein, das individuelle paradoxe IRIS-Risiko bei Tuberkulose einzuschätzen. ART-Bestandteile oder Zeit zwischen dem Beginn der TB-Therapie und ART hatten in der Studie keinen Einfluss. Patients coinfected with HIV and tuberculosis (TB) are at risk of developing immune reconstitution inflammatory syndrome (IRIS) after initiation of antiretroviral therapy (ART). An IRIS is either characterized by the onset or worsening of clinical or radiologic features of an infection after ART initiation (“paradoxical IRIS”) or an unmasking of a prior undiagnosed and untreated, clinical inapparent infection (“unmasking IRIS”). Diagnosis can be challenging in clinical setting due to non-standardized consensus diagnostic criteria. Therefore, the objective of this dissertation was to identify clinical characteristics and risk or protective factors for TB-IRIS development. Results from this study should aid in better understanding and predicting TB- associated IRIS. Charts were extracted retrospectively of 52 pseudonymized patients, who were treated for HIV and active TB from 01/01/2010 – 30/06/2016 in the department of Infectious Diseases of the University Hospital Frankfurt. Medical charts consisted of hospital discharge papers, laboratory findings, temperature charts and ward round notes, recorded in hospital databases “ORBIS”, “epidem” and “Nexus swisslab”. Demographic and clinical characteristics, routine laboratory, ART and tuberculostatic therapy findings, serologies, type of TB, and immunological and virological response for a 48-week interval (viral load, CD4- and CD8-count) were extracted. Prior to analysis, patients were divided into two groups: ART-naïve and non-ART-naïve TB patients. The former group having thus been at risk of developing paradoxical IRIS, while the latter could have developed unmasking IRIS. The course of disease including HIV viral load were evaluated and patients were classified according to IRIS case definition by French et al. (2004) into those developing IRIS and not. Unclassifiable cases were presented to and ultimately assessed by mentors. The statistical program “bias” was used for comparing results between study groups with or without IRIS. Fisher’s exact test was applied for categorial and Mann-Whitney U test for numerical variables. Incidence of paradoxical TB-IRIS was 29.7 %, of unmasking TB-IRIS 46.7 %. The most common clinical feature of IRIS in our Frankfurt cohort was fever, followed by lymphadenopathy and respiratory complaints. Hospital stay was significantly longer for patients developing paradoxical or unmasking IRIS compared to those without IRIS. For unmasking IRIS patients, there were no further significant differences. However, the small sample size (15 non-ART-naïve patients) should be taken into consideration. Patients with paradoxical IRIS had a significantly higher re-admittance rate to hospital (63.3 % vs. 15.4 %; p= 0.006), underscoring the clinical relevance of paradoxical TB-IRIS. Furthermore, associations of paradoxical IRIS and extra-pulmonary TB (p= 0.025), low CD4-count (p= 0.006), and high viral load (p= 0.017) at baseline were found. These patients at risk should be monitored closely after ART initiation. Elevated lactate dehydrogenase activity (LDH) and lower serum albumin level was associated with paradoxical IRIS development as well. In combination with the parameters detailed above, these findings may help to assess the individual paradoxical TB-IRIS risk. Choice of ART components and time between tuberculostatic therapy and ART did not influence IRIS development in our analysis.

Zusammenfassung Mikroorganismen haben im Laufe ihrer Evolution zur Anpassung an ihren Lebensraum eine Vielfalt an strukturell diversen und zum Teil sehr komplexen Metaboliten entwickelt. Diese verschaffen ihnen Vorteile gegenüber anderen Bewohnern ihrer Umwelt. Die Nutzung dieser Substanzen führte zur Entwicklung von Antibiotika, Immunsuppressiva und Zytostatika. Hierbei zeigen sich besonders die Enzymklassen der Polyketidsynthasen (PKS), nichtribosomalen Peptidsynthetasen (NRPS), NRPS/PKS-Hybride und NRPS-ähnliche Enzyme für die Synthese der Grundgerüste der Sekundärmetabolite verantwortlich. Die so entstandenen Substanzen können durch andere Enzyme (tailoring enzymes) wie beispielsweise Methyltransferasen oder Prenyltransferasen weiter modifiziert werden, wodurch oftmals die biologische Aktivität gesteigert wird. Zur fermentativen, semisynthetischen oder chemoenyzmatischen Herstellung dieser Naturstoffe und deren Derivate ist ein detailliertes Wissen der Biosynthese der beteiligten Enzyme Voraussetzung. Obwohl die individuellen Domänen von NRPS und PKS inzwischen biochemisch und strukturell sehr gut untersucht sind, ist das Verständnis für die Interaktion der Domäne und die ablaufenden Mechanismen noch sehr gering. Da sich einfache Systeme wie die NRPS-ähnlichen Enzyme leichter untersuchen lassen, sollten in der vorliegenden Arbeit alle NRPS-ähnlichen Enzyme mit einer A-T-TE Enzymstruktur aus Aspergillus terreus (A. terreus) und ein weiteres aus Chaetomium globosum (C. globosum) charakterisiert werden. Zunächst sollten die NRPS-ähnlichen Gene apvA, melA, atrAAt, pgnA aus A. terreus mit der eigenen Promoter- und Terminatorsequenz heterolog in Aspergillus nidulans exprimiert werden. Obwohl eine Integration für melA und atrAAt ins Genom beobachtet wurde, konnte in den Ethylacetatextrakten der Kulturüberstände kein Produkt nachgewiesen werden. Für die Expression der NRPS-ähnlichen Gene in Saccharomyces cerevisiae (S. cerevisiae) sollten die Gene von cDNA bzw. gDNA amplifiziert werden. Hierdurch konnte die Exon-Intron Struktur für apvA, melA, btyA und atrAAt korrigiert werden. Des Weiteren konnte die Domänen Architektur für das putative NRPS-ähnlich Gen atrAAt, das laut der NCBI-Datenbank nur aus einer A- und einer T-Domänen bestand, durch die Amplifikation von cDNA zu einer A-T-TE-Struktur korrigiert werden. Die NRPS-ähnlichen Gene haben etwa eine Größe von 2763-2874 bp und kodieren entsprechend für 920-958 AS lange Proteine. Eine Expression der sieben NRPS-ähnlichen Gene in S. cerevisiae führte zu den fünf Produkten Aspuvinon E, Butyrolacton IIa, Atromentin, Phenguignarsäure und Didemethylasterrichinon D. Die Produkte wurden extrahiert und die Strukturen mittels Massenspektrometrie und NMR-Spektroskopie aufgeklärt. Auf einem Liter Kultur konnten für Aspulvinon E und Phenguignarsäure eine Produktion von 13 bzw. 15 mg und für Butyrolacton IIa sogar bis zu 35 mg berechnet werden. In anschließenden Versuchen zur Isolierung der NRPS-ähnlichen Proteine stellte sich heraus, dass Affinitäts-Tags am C-Terminus die Enzymaktivität stark hemmt. Ein N terminaler His6-Tag hatte hingegen keinen negativen Einfluss auf die Aktivität. Anhand des Proteinnachweises mit Hilfe eines Western Blotes wurde der optimale Zeitpunkt für die Isolierung auf etwa 16 Stunden nach der Induktion der Proteinexpression durch Galaktose festgestellt. Die rekombinanten Proteine konnten mit einer Konzentration von etwa 1 ml/l Kultur aufgereinigt werden. Allerdings konnte keine katalytische Aktivität nachgewiesen werden. Für ein alternatives Expressionsystem in Escherichia coli wurden die NRPS-ähnlichen Gene sowie das Phosphpantethenyltransferase Gen npgA in die Vektoren pQE60 und pET28a(+) kloniert. Die Expression konnte aus Zeitgründen nicht mehr durchgeführt werden. Die A-T-TE Domänen der NRPS-ähnlichen Enzyme bilden theoretisch autonome Einheiten, die zwischen den Enyzmen ausgetauscht werden können. Um diese Theorie zu überprüfen und um zusätlich für die NRPS-ähnlichen unnatürliche Produkte zu generieren, sollten die das Substrat aktivierenden A-Domänen mit den TE-Domänen, die für Dimerisierung verantwortlich sind, kombiniert werden. Dafür wurden die A-Domänen von Pgna, ApvA, AstA und AtqA, die Phenyl-; 4-Hydroxy-bzw. Indolpyruvat aktivieren, mit der TE-Domäne von AvpA, BtyA, PgnA, AtrAAt bzw. AstA rekombiniert. Die T-Domäne wurde jeweils von einem der ursprünglichen Enzyme übernommen. Insgesamt wurden 34 Konstrukte hergestellt, die zur Expression in S. cerevisiae verwendet wurden. Für 22 konnten die erwarteten Produkte nachgewiesen und anhand der [M+H-] - Ionen und deren Fragmentierungen sowie NMR-Spektroskopie aufgeklärt werden. So konnten Indolbutyrolacton und Indolguignarsäure synthetisiert werden, welche zuvor noch nie in der Literatur beschrieben worden waren. Weiterhin ist bekannt, dass die durch die NRPS-ähnlichen Enyzme hergestellten Produkte oft nur das Grundgerüst für Sekundärmetabolite bilden. Daher wurde die nähere genetische Umgebung der NRPS-ähnlichen Gene untersucht. Es wurden 4 Prenyltransferasegene identifiziert, die zur Ko-Expression mit den NRPS-ähnlichen ausgewählt wurden. Leider konnte das prenylierte Produkt in den Transformaten der Bäckerhefe nicht detektiert werden., Summary In the course of their evolution, microorganisms have developed a variety of structurally diverse and sometimes very complex metabolites to adapt to their habitat. These give them advantages over other inhabitants of their environment. The use of these substances led to the development of antibiotics, immunosuppressants and antitumor agents. The backbone structures of these substances are often synthesized by the enzyme classes of polyketide synthases (PKSs), nonribosomal peptide synthetases (NRPSs), NRPS / PKS hybrids and NRPS-like enzymes. The resulting structures can be further modified by other tailoring enzymes such as methyltransferases or prenyltransferases, which often increases the biological activity. A detailed knowledge of the biosynthesis and of the involved enzymes is a prerequisite for the fermentative, semisynthetic or chemoenzymatic production of these natural substances and their derivatives. Although the individual domains of NRPS and PKS have biochemically and structurally been investigated, understanding of the interaction of the domain and the mechanisms involved is still very poor. Since simple systems like the NRPS-like enzymes can be investigated more easily, the NRPS-like enzymes with an A-T-TE architecture from Aspergillus terreus (A. terreus) and another from Chaetomium globosum (C. globosum) will be characterized in the present work. First, the NRPS-like genes apvA, melA, atrAAt, pgnA from A. terreus with their own promoter- and terminator-sequence should be analysed by heterologous expression in Aspergillus nidulans. Although an integration for melA and atrAAt into the genome was observed, no product could be detected in the ethyl acetate extracts of the culture. For the expression of the NRPS-like genes in Saccharomyces cerevisiae (S. cerevisiae), cDNA or gDNA was used for the amplification of the genes. This made it possible to revise the exon-intron structures for apvA, melA, btyA and atrAAt. Furthermore, the domain architecture of the putative NRPS-like gene atrAAt, which according to the NCBI database only consisted of one A and one T domains, could be corrected by amplifying cDNA to an A-T-TE structure. The NRPS-like genes have a size of 2763-2874 bp and accordingly code for proteins of 920-958 aa. Expression of the seven NRPS-like genes in S. cerevisiae led to the five products aspuvinone E, butyrolactone IIa, atromentine, phenguignardic acid and didemethylasterrichinone D. The products were extracted and the structures were determined by means of mass spectrometry and NRM spectroscopy. For aspulvinone E and phenguignardic acid, a production of 13 and 15 mg/l and for butyrolactone IIa even up to 35 mg/l could be calculated on one liter of culture. In subsequent attempts to isolate the NRPS-like proteins, it turned out that affinity tags at the C-terminus strongly inhibit enzyme activity. In contrast, an N-terminal His6 tag had no negative influence on the activity. On the basis of the protein detection by Western blot, the optimal point in time for isolation was determined to be around 16 hours after the induction of protein expression by galactose. The recombinant proteins could be purified with a concentration of about 1 ml/l culture. However, no catalytic activity could be detected. To test an alternative expression system, the NRPS-like genes and the phosphpantethenyltransferase gene npgA were cloned into the vectors pQE60 and pET28a (+) for the expression in Escherichia. Due to time limits the gene expression and protein isolation could no longer be carried out. The A-T-TE domains of the NRPS-like enzymes theoretically form autonomous units that can be exchanged between the enzymes. In order to test this theory and in addition to generate unnatural products, the substrate-activating A domains were combined with the TE domains, which are responsible for the dimerisation. For this purpose, the A domains of Pgna, ApvA, AstA and AtqA for activation of phenyl-, 4-hydroxy or indole pyruvate, were recombined with the TE domain of AvpA, BtyA, PgnA, AtrAAt and AstA, respectively. The T domain was taken over from one of the original enzymes. A total of 34 constructs were prepared in this way which were used for expression in S. cerevisiae. 22 of the expected products could be detected. The structure was elucidated on the basis of the molecular ions and their fragmentation as well as NMR spectroscopy. In this way, indole butyrolactone and indole guignaric acid could be obtained, which had never before been described in the literature. It is also known that the products of the NRPS-like enzymes often only form the basic structure for secondary metabolites. Therefore, the genetic environment of the NRPS-like genes was investigated. 4 prenyltransferase genes were identified which were selected for co-expression with the NRPS-like ones in S. cerevisiae. Unfortunately, the prenylated product could not be detected in the baker's yeast transformants.

Recent developments of novel in-vehicle interventions show the potential to transform the otherwise routine and mundane task of commuting into opportunities to improve the drivers' health and well-being. Prior research has explored the effectiveness of various in-vehicle interventions and has identified moments in which drivers could be interruptible to interventions. All the previous studies, however, were conducted in either simulated or constrained real-world driving scenarios on a pre-determined route. In this paper, we take a step forward and evaluate when drivers interact with in-vehicle interventions in unconstrained free-living conditions. To this end, we conducted a two-month longitudinal study with 10 participants, in which each participant was provided with a study car for their daily driving needs. We delivered two in-vehicle interventions - each aimed at improving affective well-being - and simultaneously recorded the participants' driving behavior. In our analysis, we found that several pre-trip characteristics (like trip length, traffic flow, and vehicle occupancy) and the pre-trip affective state of the participants had significant associations with whether the participants started an intervention or canceled a started intervention. Next, we found that several in-the-moment driving characteristics (like current road type, past average speed, and future brake behavior) showed significant associations with drivers' responsiveness to the intervention. Further, we identified several driving behaviors that "negated" the effectiveness of interventions and highlight the potential of using such "negative" driving characteristics to better inform intervention delivery. Finally, we compared trips with and without intervention and found that both interventions employed in our study did not have a negative effect on driving behavior. Based on our analyses, we provide solid recommendations on how to deliver interventions to maximize responsiveness and effectiveness and minimize the burden on the drivers.

Naturstoffe, die aus dem Sekundärmetabolismus stammen werden von Organismen nicht unbedingt zum Überleben, der Reproduktion oder der Differenzierung benötigt, bieten aber dennoch einen signifikanten Selektionsvorteil in der Konkurrenz mit anderen Lebewesen. Daraus resultiert, dass viele dieser Naturstoffe in Organismen bestimmte biologische Aktivitäten aufweisen. Durch die biologische Aktivität vieler dieser Substanzen ist beispielsweise der pilzliche Sekundärmetabolismus eine attraktive Quelle für die Entdeckung und Entwicklung neuer Arzneimittel. Bisher wurde nur ein Bruchteil aller existierenden Pilze identifiziert, was bedeutet, dass noch immer ein enormes ungenutztes Potenzial für die Herstellung pharmazeutisch nutzbarer Naturstoffe vorhanden ist. Dieses Potenzial kann durch verschiedene bioinformatische, molekularbiologische und biochemische Ansätze zugänglich gemacht werden. Die Entdeckung biosynthetischer Gene für Sekundärmetabolite in Pilzen wird dadurch erleichtert, dass relevante Gene für die Herstellung einer Verbindung nebeneinander an einem bestimmten Lokus auf dem Chromosom vorliegen, d.h. sie sind geclustert. Diese biosynthetischen Gencluster (BGCs) enthalten ein oder mehrere Gene, deren Proteinprodukte für die Bildung des Kohlenstoffrückgrats des jeweiligen Stoffs verantwortlich ist. Dieses Rückgrat wird durch weitere, im Cluster kodierte, modifizierende Enzyme strukturell diversifiziert. In dieser Arbeit wurden die BGCs für zwei unterschiedliche Naturstoffe und ihre Analoga in dem Ascomyceten Aspergillus ruber identifiziert und bestätigt. Außerdem wurden einzelne Schritte in diesen Stoffwechselwegen näher untersucht. Das Salicylaldehydderivat Flavoglaucin und seine Analoga unterscheiden sich nur in der Anzahl und Position der Doppelbindungen in der Heptyl-Seitenkette. In vorangegangenen Studien wurde gezeigt, dass einige dieser Substanzen biologisch aktiv sind, indem sie beispielsweise als Antioxidantien wirken, antiinflammatorische Effekte vorweisen oder auch an humane Opioid- und Cannabinoidrezeptoren binden. Obwohl Flavoglaucin und seine Analoga schon 1934 zum ersten Mal beschrieben wurden, war bis zur Veröffentlichung der in dieser Arbeit beschriebenen Daten nicht viel mehr über ihre Biosynthese bekannt, als dass sie Produkte des Polyketidstoffwechsels sind. Durch Genome Mining konnte in dieser Arbeit ein Gencluster (fog-Cluster) im Genom von A. ruber identifiziert werden, der alle nötigen Gene für die Biosynthese dieser Stoffe enthält. Die heterologe Expression dieses BGC im Modellorganismus Aspergillus nidulans bestätigte, dass der fog-Cluster für die Bildung von Flavoglaucin und seinen Analoga verantwortlich ist. Gendeletionen in dem heterologen Expressionsstamm, die Überexpression des Gens für die hoch-reduzierende Polyketidsynthase (HR-PKS) fogA in A. nidulans, in vitro-Enzymassays mit der Prenyltransferase FogH und Fütterungsexperimente zur Untersuchung der FAD-abhängigen Oxidoreduktase FogF zeigten einen ungewöhnlichen Weg hin zum finalen Aldehydprodukt. Das Polyketidgrundgerüst wird von FogA in Kooperation mit dem Cupin FogC und den kurzkettigen Dehydrogenasen/Reduktasen FogB und FogD reduktiv als Salicylalkohol entlassen, nur um nach der C3-Hydroxylierung durch das Cytochrom P450 (CYP) FogE und die C5-Prenylierung durch FogH von FogF wieder zu einem Aldehyd oxidiert zu werden. Diese Reoxidation ist bemerkenswert, da die Freisetzung des Produkts von einer HR-PKS für gewöhnlich hydrolytisch als Carbonsäure oder reduktiv allerhöchstens als Aldehyd erfolgt. Die Modifikation der Intermediate vor der Reoxidation erfordert scheinbar die Hydroxymethylgruppe des Salicylalkohols, denn eine Prenylierung des synthetisierten Aldehyd-Substratanalogs durch FogH findet nicht statt. Diese Arbeit wurde in Kooperation mit Frau Dr. Huomiao Ran durchgeführt. Die Biosynthese von Flavoglaucin weist einige Parallelen zu denen von Sordarial in Neurospora crassa und Trichoxid in Trichoderma virens auf, unterscheidet sich aber neben der Prenylierung signifikant durch die finale Reoxidation und die Variabilität des Sättigungsmusters in der Heptyl-Seitenkette. In dieser Arbeit wurde auch die Biosynthese von Indolalkaloiden der Echinulin-Familie näher untersucht. Aus einer vorigen Studie war bekannt, dass zwei Prenyltransferasen EchPT1 und EchPT2 jeweils die reverse C2-Prenylierung und mehrere konsekutive reguläre Prenylierungen des Indolrings des zyklischen Dipeptids aus L-Tryptophan und L-Alanin katalysieren. Die Einführung einer oder zwei exo-Doppelbindungen an den Substituenten um den 2,5-Diketopiperazinring durch eine CYP aus dem identifizierten BGC wurde jedoch nur vermutet. Durch heterologe Expression des postulierten ech-BGCs in A. nidulans konnte diesem Cluster die Bildung von Substanzen der Echinulin-Familie eindeutig zugewiesen werden. Dabei wurden allerdings nur Substanzen ohne Doppelbindung (Echinulin-Serie) und mit einer (∆10) Doppelbindung am Tryptophansubstituenten (Neoechinulin A-Serie) gebildet. Das Enzym für die Einführung der zweiten (∆14) Doppelbindung am Alaninsubstituenten (Neoechinulin B-Serie) scheint nicht durch die Gene des Clusters kodiert zu werden. Auch die Erweiterung des exprimierten BGCs um drei weitere potenzielle Oxidoreduktasen stromabwärts des ech-Clusters resultierte nicht in der Produktion von Substanzen mit beiden Doppelbindungen. Fütterungsexperimente bestätigten die Einführung der ∆10-Doppelbindung durch die im ech-Cluster kodierte CYP EchP450, aber widerlegten die Hypothese, dass EchP450 auch die Bildung der ∆14-Doppelbindung katalysiert. Weitere in vitro-Enzymassays mit der Prenyltransferase EchPT2 zeigten deutlich ihre Präferenz gegenüber Substraten ohne exo-Doppelbindung, was in gewissem Maße zur Erklärung der Verhältnisse zwischen den verschiedenen Substanzen der Echinulin-Familie in A. ruber beiträgt., Natural products which arise from an organism’s secondary metabolism are not necessarily re-quired for survival, reproduction and differentiation but offer a significant selective advantage in their respective environment. As a result, many of these natural products display significant biolog-ical activities in organisms. Due to the biological activity of many of these substances, the fungal secondary metabolism for example is an attractive source for the discovery and development of new drugs. So far, only a limited number of all existing fungi has been identified, which means that there is still an enormous unused potential for the production of pharmaceutically relevant natural products. This potential can be accessed through various bioinformatic, molecular biological and biochemical approaches. The discovery of biosynthetic genes for secondary metabolites in fungi is facilitated by the fact that relevant genes for the production of a given compound are located in close proximity to each other on the chromosome, i.e., they are clustered. These biosynthetic gene clusters (BGCs) contain one or more genes whose protein products are responsible for the formation of the carbon backbone of the respective substance. This backbone is structurally diversified by tailoring enzymes encoded by other genes in the cluster. In this thesis, BGCs for two different natural products and their conge-ners from the ascomycete Aspergillus ruber were identified and confirmed. In addition, individual steps in these metabolic pathways were examined in more detail. The prenylated salicylaldehyde derivative flavoglaucin and its congeners differ from each other only in the number and position of the double bonds of the heptyl side chain. Previous studies have shown that some of these substances are biologically active, for example by acting as antioxi-dants, showing anti-inflammatory effects, or binding to human opioid and cannabinoid receptors. Although flavoglaucin and its congeners have been known since 1934, not much was known about their biosynthesis other than that they are products of polyketide metabolism. This thesis describes the discovery of a gene cluster (fog cluster) in the genome of A. ruber through genome mining, which contains all the genes necessary for the biosynthesis of these substances. Heterologous ex-pression of this BGC in the model ascomycete Aspergillus nidulans confirmed that the fog cluster is responsible for the formation of flavoglaucin and its congeners. Gene deletions in the heterologous expression strain, the overexpression of the gene for the highly reducing polyketide synthase (HR-PKS) FogA in A. nidulans, in vitro enzyme assays with the prenyltransferase FogH and feeding exper-iments to investigate the FAD-dependent oxidoreductase FogF showed an unusual route to the final aldehyde product. The polyketide skeleton is reductively released as a salicyl alcohol by FogA in cooperation with the cupin protein FogC and the short-chain dehydrogenases/reductases FogB and FogD, only to be reoxidized to the salicylaldehyde by FogF after C3-hydroxylation by the cyto-chrome P450 (CYP) FogE and C5-prenylation by FogH. This reoxidation is remarkable, as product release from an HR-PKS usually occurs hydrolytically as a carboxylic acid or reductively as an alde-hyde. Modification of the intermediates apparently requires the hydroxymethyl group of the salicyl alcohol, because prenylation of a synthesized aldehyde substrate analog by FogH does not occur. This work was carried out in cooperation with Dr. Huomiao Ran. The biosynthesis of flavoglaucin shows some parallels to that of sordarial in Neurospora crassa and trichoxide in Trichoderma virens, but differs significantly in terms of prenylation, the final reoxidation of an alcoholic intermediate to an aldehyde and the variability of the saturation pattern in the heptyl side chain. Furthermore, the biosynthesis of indole alkaloids of the echinulin family was investigated in more detail. A previous study showed that two prenyltransferases EchPT1 and EchPT2 catalyze the re-verse C2 prenylation and several consecutive prenylations at different positions of the indole ring of the cyclic dipeptide cyclo-L-tryptophan-L-alanine. It was speculated that a CYP encoded in the identified BGC introduces two exo double bonds on the substituents of the 2,5-diketopiperazine ring. By heterologous expression of the postulated ech BGC in A. nidulans, the formation of echinulin family indole alkaloids could be clearly assigned to this cluster. However, only substances without a double bond (echinulin series) and with a (∆10) double bond on the tryptophan substitu-ent (neoechinulin A series) were formed. The enzyme for the introduction of the second (∆14) dou-ble bond on the alanine substituent (neoechinulin B series) does not seem to be encoded by the genes of the ech cluster. The expansion of the expressed BGC by three additional potential candi-date genes downstream of the ech cluster did not yield the products with both double bonds. Feeding experiments confirmed the introduction of the ∆10 double bond by the CYP EchP450 en-coded in the ech cluster but refuted the hypothesis that EchP450 also catalyzes the formation of the ∆14 double bond. Further in vitro enzyme assays with the prenyltransferase EchPT2 showed its clear preference for substrates without an exo double bond, which contributes to some extent to the explanation of the different product ratios between the various echinulin family alkaloids in A. ruber.

Background Cancer cachexia as a metabolic syndrome can lead to at least 25% of cancer deaths. The inhibition of muscle atrophy is a main strategy to treat cancer cachexia. In this process, myostatin (MSTN) can exert a dual effect on protein metabolism, including inhibition of protein biosynthesis and enhancement of protein degradation. In this study, we will test the effect on muscle atrophy induced by cancer cachexia of IMB0901, a MSTN inhibitor. Methods Two high-throughput screening models against MSTN were developed. By screening, IMB0901, 2-((1-(3,4-dichlorophenyl)-1H-pyrazolo [3,4-d] pyrimidin-4-yl) amino) butan-1-ol, was picked out from the compound library. The in vitro cell model and the C26 animal model of muscle atrophy induced by cancer cachexia were used to determine the pharmacological activity of IMB0901. Whether IMB0901 could inhibit the aggravating effect of doxorubicin on muscle wasting was examined in vitro and in vivo. Results IMB0901 inhibited the MSTN promoter activity, the MSTN signaling pathway, and the MSTN positive feedback regulation. In atrophied C2C12 myotubes, IMB0901 had a potent efficiency of decreasing MSTN expression and modulating MSTN signaling pathway which was activated by C26-conditioned medium (CM). In C2C12 myotubes, the expressions of three common myotube markers, myosin heavy chain (MyHC), myogenic differentiation 1 (MyoD), and myogenin (MyoG), were downregulated by CM, which could be efficiently reversed by IMB0901 via reduction of ubiquitin-mediated proteolysis and enhancement of AKT/mTOR-mediated protein synthesis. In the C26 animal model, IMB0901 mitigated the weight loss of body, quadricep and liver, and protected the quadriceps cell morphology. Furthermore, IMB0901 decreased the expression of two E3 ligases Atrogin-1 and MuRF-1 in the quadriceps in vivo. At the cellular level, IMB0901 had no influence on anti-tumor effect of three chemotherapeutic agents (cisplatin, doxorubicin, and gemcitabine) and lowered doxorubicin-induced upregulation of MSTN in C2C12 myotubes. IMB0901 did not affect the inhibitory effect of doxorubicin on C26 tumor and delayed the weight loss of muscle and adipose tissue caused by C26 tumor and doxorubicin. Conclusions IMB0901 inhibits muscle atrophy induced by cancer cachexia by suppressing ubiquitin-mediated proteolysis and promoting protein synthesis. These findings collectively suggest that IMB0901 is a promising leading compound for the management of muscle atrophy induced by cancer cachexia. The globally increasing prevalence and incident rates of mental diseases is one of the most serious public health challenges according to the World Health Organization. Today, treatment is based on professional therapies which require a high amount of financial resources and personnel effort, however IT-supported interventions in ubiquitous devices promise help and a new leverage beyond traditional therapies. We identify the car as a space for new treatments since drivers often have time and the environment in the automobile is highly controlled. In-vehicle information systems can reach people in their daily routine and could introduce innovative prevention measures. In this research in progress paper, we address the open question how the car can improve a driver’s affective state while driving. First, we thoroughly describe the design of a study we conducted to motivate other researchers for this topic. Second, we analyse 631 completed interventions collected in a 2-month field study with 10 drivers. First analyses indicate that we can positively influence the short-term affective state of drivers with at least one of ourintervention types. We provide first practical examples of how to reach the masses of everyday drivers.

Einleitung: Eine der größten postoperativen Sorgen von Patient*innen ist der Schmerz. Um diesen zu lindern und das Wohlbefinden der Patient*innen zu erhalten bzw. zu erhöhen, sollten nach dem regelmäßigen Schmerz- bzw. Comfort-Assessment nicht nur pharmazeutische, sondern auch nicht pharmazeutische oder komplementäre Interventionen eingesetzt werden. Das zugrunde liegende Ziel dieser Arbeit ist es, zu untersuchen welche komplementären Pflegeinterventionen inwieweit in der postoperativen Phase zur Schmerzlinderung angewendet werden können. Methodik: Im Rahmen dieser Arbeit erfolgte eine systematische Literaturrecherche in den Datenbanken CINAHL und PubMed. Zwischen 2010 und 2020 veröffentlichte englische sowie deutsche Literatur wurde bevorzugt. Letztlich wurden insgesamt 14 Peer-Review-Studien für diese Arbeit herangezogen. Ergebnisse: Komplementäre Pflegemaßnahmen wie Musik, Teilmassage mit Aromaöl, Therapeutic Touch und Entspannungstechniken erwiesen sich in Kombination mit pharmazeutischen Therapien bei der postoperativen Schmerzlinderung als hilfreich. Ebenfalls wurde herausgefunden, dass sich auf diese Weise Angst reduzieren lässt und Vitalzeichen wie der Blutdruck und der Puls stabilisiert werden können. Außerdem führen diese Maßnahmen zu einer erhöhten Zufriedenheit der Patient*innen und tragen zu deren Wohlbefinden bei. Schlussfolgerung: Nicht pharmazeutische Pflegemaßnahmen haben zwar einen positiven Effekt auf den postoperativen Schmerz, aber die langfristige Auswirkung ist aufgrund der geringen Durchführungsanzahl sowie des kurzen Interventionszeitraums unklar. Das Ziel im Rahmen des Schmerzmanagements ist nicht nur Schmerzen zu lindern, sondern auch den Comfort bzw. das Wohlbefinden der Patient*innen zu erhöhen. Weil während der Literaturanalyse diesbezüglich weder eine Standard Operating Procedure noch Leitlinie oder Maßstäbe o. Ä. gefunden wurden, bedarf es im Bereich der komplementären Pflegemaßnahmen mehr Entwicklung und Forschung. Auch sollte das Thema ‚Comfort‘ im Kontext des postoperativen Schmerzmanagements in zukünftige Studien einbezogen werden. Introduction: One of the patient’s biggest concerns is the postoperative pain. In order to decrease the pain experiences and increase patient’s well-being, not only pharmacological, but also non-pharmacological interventions should be used after a continuous pain and comfort assessment. Therefore, the goal of this paper is to identify what kind of and to what extent complementary nursing methods can be used to relieve pain in the postoperative period. Methodology: In this paper, a systematic literature review was performed in the databases CINAHL and PubMed. Moreover, studies in English and German, which were published between 2010 and 2020, were preferred for this research paper. Finally, 14 peer-reviewed studies were chosen to perform its result in this paper. Results: Complementary nursing methods such as music, massage with aroma oil, Therapeutic Touch and relaxation methods have been proved to be helpful with pharmacological methods to relieve postoperative pain. Moreover, complementary interventions can decrease anxiety and stabilize vital signs such as blood pressure and pulse. These methods also increase patient’s satisfaction and comfort. Conclusion: As a result, non-pharmacological nursing intervention indeed have a positive effect on postoperative pain, but the long-term effect is still unknown due to low performance rate of the intervention and duration. The purpose of pain management is not only to decrease pain, but also to increase patient’s comfort. Since there’s few SOPs, Guidelines and standards were found in studies, more researches should be done in complementary nursing intervention. If studies were to be done in context with postoperative pain management in the future, the theme comfort should be included.

Due to the lack of a suitable theoretical model for simulating radar backscatter of soil based on salt content, we investigated a new method to exploit Sentinel-1 radar backscatters and polarimetric decomposition information for dryland soil salinity monitoring. Soil electrical conductivity (EC) was estimated using Sentinel-1 SAR imagery and field survey data combined with five machine learning models in Esperance, located in the southwest of Western Australia (SWWA). The performance of the five machine learning models was assessed and compared using the root-mean-square error (RMSE), the mean absolute error (MAE), and the correlation coefficient ( $r$ ). The results revealed that the Random Forest Regression model (RFR) yielded the highest prediction performance ( $\text{RMSE}=2.89\ S/m,\ \text{MAE}=1.90 S/m$ , and $\mathrm{r}=0.81$ ) and outperformed the other models. It can be concluded that the intensity images of VV and VH polarization of SAR imagery have the potential to predict EC of soils in SWWA.

Einleitung: Die Tätigkeiten, die Pflegepersonen in der Notfallambulanz übernehmen müssen, führen häufig zur physischen und psychischen Belastung. Doch die individuelle Resilienz erwies sich als besonders hilfreich bei der Reduzierung der Arbeitsbelastung. Das Ziel der Arbeit besteht darin, die individuelle Resilienz der Pflegekräfte in der Notfallambulanz im Kontext der Arbeitsbelastung zu identifizieren und zu beschreiben. Methode: Es wurde eine systematische Literaturrecherche in den Datenbanken CINAHL und PubMed durchgeführt. Dabei wurde vorzugsweise anhand der englisch- und deutschsprachigen Literatur zwischen 2008 und 2019 recherchiert. Insgesamt wurden 10 peer-reviewte Studien herangezogen, um die Ergebnisse darzustellen. Ergebnis: Die zwischenmenschliche Beziehung wurde als ein wesentlicher Grund für die Arbeitsbelastung dargestellt. Ebenfalls führen der Personalmangel, die Überfüllung der Notallambulanz, die Erfahrung mit gewaltsamen Ereignissen und die inadäquaten materiellen Ressourcen zu einer erheblichen physischen und psychischen Arbeitsbelastung. Jedoch konnten bei den Pflegepersonen Resilienz- bzw. Schutzfaktoren beobachtet werden. Dabei erwies sich die soziale Unterstützung der Kolleg*innen als besonders hilfreich. Des Weiteren waren Resilienzfaktoren wie Optimismus, Hardiness, Religion und Spiritualität, Hoffnung, Selbstwertgefühl und Selbstwirksamkeitserwartung festgestellt werden Diskussion: Obwohl die zwischenmenschliche Beziehung eine Arbeitsbelastung darstellt, hat diese gleichzeitig eine entlastende Wirkung. Die individuelle Resilienz setzt sich aus mehreren Resilienz- bzw. Schutzfaktoren zusammen. Da es bei der Literaturrecherche überwiegend Referenzen gab, die sich auf der Resilienz bezüglich der Gewalterfahrung konzentrieren, bedarf es weiterer Forschung zur Resilienz in anderen Bereichen der Arbeitsbelastung. Zudem sollte Guidelines bezüglich der Resilienzförderung entwickelt werden, um den physischen und psychischen Auswirkungen entgegenwirken zu können. Introduction: The activities that emergency nurses have to take over, result often physical and psychological stress. However, it appears that individual resilience is very helpful by reducing occupational stress. The aim of this paper is to describe and to identify individual resilience in context with occupational stress. Method: A systemic literature research was performed in the databases CINAHL and Pubmed. Studies in German and English which was published between 2008 and 2019 were included. At the end 10 peer-reviewed Studies were chosen to perform the result of this paper. Result: Interpersonal relationship was shown as a significant factor for occupational stress. Staff- shortage, overcrowded emergency room (ER), experience with violent event and inadequate material resources also lead to enormous physical and psychological occupational stress. However, resilience or protective factors could be observed on the emergency staffs. At this point social support was proved to be most helpful. Resilience factors such as optimism, hardiness, religion and spirituality, hope, self-esteem and self-efficacy could also be seen on emergency nurses. Discussion: Although interpersonal relationship was considered as a factor of occupational stress in the ER, it also has a helpful character at the same time. The individual resilience is a combination of more than one resilience or protective factor. Thus, more research on resilience in other area of occupational stress should be done, as there has been predominantly references in the literature which focused on resilience in context with work-related violent experiences. Guidlines should also be developed to strengthening resilience in order to counteract the negative physical and psychological impact of occupational stress.

Despite efforts to reduce them, traffic accidents continue to increase and bypass reduction targets. The costs of traffic accidents are enormous, killing 1.35 million people every year and costing 3% of most countries’ GDP. Recent research aims to target interventions at high-accident-density locations, called accident hotspots. New methods and technologies can systematically identify hotspots, but it remains unclear whether hotspots contribute to accident costs as well as volume. This paper investigates the monetary and human costs of accident hotspots. We analyze a dataset of all accidents from 2011 - 2017 in Switzerland. We identify hotspots, then analyze their contributions to traffic accident costs. We find that hotspot accidents are not different in monetary costliness or injury rates from non-hotspot accidents, so hotspots drive costs along with accident volume. However, hotspot accidents are less fatal, so hotspot targeting might not be best for fatalities. If hotspots are reduced to normal road conditions, total monetary costs can be reduced by up to 5% per year as a theoretical upper bound. Targeting the top 10% most frequent, costly, injurious, or deadly hotspots yeilds different results for different cost types, with accident number and monetary cost targets creating the highest reductions overall.

Mutterkornalkaloide sind basische Naturstoffe, die zu der Gruppe der Indolalkaloide gehören. Sie zeigen ein breites Spektrum an pharmakologischen und zuweilen auch toxischen Wirkungen aufgrund ihrer strukturellen Ähnlichkeit zu den Neurotransmittern Dopamin, Serotonin und Adrenalin. Die Hauptproduzenten sind filamentöse Pilze aus den Familien Clavicipitaceae und Aspergillaceae wie z.B. Claviceps purpurea, Aspergillus fumigatus oder Penicillium commune. Die ersten Schritte der Biosynthese von Mutterkornalkaloiden, ausgehend von L-Tryptophan, verlaufen bis zur Stufe von Chanoclavin-I Aldehyd in allen bekannten Produzenten gleich. Später verzweigen sich die Biosynthesewege und es entstehen unterschiedliche Endprodukte. Im Rahmen dieser Arbeit sollte der letzte, noch nicht aufgeklärte, gemeinsame Schritt der Biosynthese untersucht werden: die Umwandlung von 4-DMA-L-Abrin zu Chanoclavin-I. Diese Reaktion untergliedert sich in mindestens zwei Oxidations- und einen Decarboxylierungsschritt. Deletions-Experimente haben gezeigt, dass die FAD-abhängige Oxidoreduktase FgaOx1 und die Katalase FgaCat für die Bildung von Chanoclavin-I essenziell sind. Um den Reaktionsmechanismus aufzuklären, sollten beide Enzyme in E. coli oder S. cerevisiae überproduziert und biochemisch charakterisiert werden. Im Falle von fgaCat war die heterologe Expression in E. coli erfolgreich, es konnte jedoch für dieses Enzym alleine keine katalytische Aktivität festgestellt werden. Die Sequenz von FgaOx1 aus der NCBI-Datenbank wurde durch eine Analyse der Intron-Exon-Struktur korrigiert, eine Überproduktion des Proteins in E. coli oder S. cerevisiae konnte jedoch nicht erreicht werden. Durch einen bioinformatischen Vergleich der Mutterkornalkaloid-Gene aus Aspergillus fumigatus, Claviceps purpurea und Arthroderma benhamiae mit den verfügbaren Genomsequenzen der Ascomyceten aus der NCBI-Datenbank konnten 15 weitere Spezies identifiziert werden, die die genetische Ausstattung für die Produktion von Mutterkornalkaloiden besitzen. Dazu zählen auch die beiden Pilze Penicillium roqueforti und Penicillium camemberti, die aufgrund ihrer industriellen Verwendung in der Käseherstellung bekannt sind. Im Rahmen der bioinformatischen Analysen wurden neben der Sequenz von FgaOx1 aus Aspergillus fumigatus auch die Proteinsequenzen von 27 weiteren Proteinen aus der NCBI-Datenbank durch Analyse der Intron-Exon-Struktur korrigiert. Durch die Identifizierung der Gencluster und die Korrektur der Sequenzen leistet diese Arbeit einen wertvollen Beitrag für die zukünftige Aufklärung der Biosynthesewege und die Analyse der strukturellen Vielfalt der Mutterkornalkaloide in den verschiedenen Produzenten. In der Vergangenheit wurde bereits über die Produktion von Isofumigaclavin A in Penicillium roqueforti berichtet, allerdings gab es zu Beginn dieser Arbeit keine Publikationen, die Aufschluss über die Biosynthese dieser Substanz geben. Durch die Identifizierung von zwei Mutternkornalkaloid-Genclustern und zwei weiteren, nicht geclusterten Genen in unterschiedlichen Bereichen des Genoms wurde der Grundstein für die Aufklärung des Biosyntheseweges gelegt. In dieser Arbeit wurden acht Gene, die möglicherweise für die späteren Schritte der Biosynthese von Isofumigaclavin A verantwortlich sind, ausgewählt und für die heterologe Expression in E. coli und S. cerevisiae kloniert. Drei der Gene konnten erfolgreich in E. coli exprimiert werden und die entsprechenden Proteine (FgaDHPr, FgaOx3Pr3 und FgaATPr) wurden mittels Affinitätschromatographie aufgereinigt. Für FgaATPr konnte in den durchgeführten in vitro-Assays keine enzymatische Aktivität nachgewiesen werden. FgaDHPr wurde als kurzkettige Dehydrogenase/Reduktase charakterisiert, welche die Umwandlung von Chanoclavin-I zu Chanoclavin-I Aldehyd in Anwesenheit von NAD+ katalysieren kann. In der aktiven Form liegt das Enzym vermutlich als Pentamer vor. Der KM-Wert für Chanoclavin-I lag bei 573 μM und für NAD+ bei 82 μM. Die mittlere maximale Reaktionsgeschwindigkeit vmax betrug 326 nmol mg-1 min-1 und die Wechselzahl kcat 0,15 s-1. FgaOx3Pr3 gehört zur Gruppe der „Old Yellow Enzymes“ und fungiert als FMN-enthaltende und NAD(P)H-abhängige Oxidoreduktase. Im Rahmen der durchgeführten Enzym-Assays wurde FgaOx3Pr3 als Chanoclavin-I Aldehyd-Reduktase charakterisiert, die zusammen mit FgaFS aus Aspergillus fumigatus oder EasG aus Claviceps purpurea die Bildung von Festuclavin katalysieren kann. Des Weiteren wird in Anwesenheit von FgaOx3Pr3 die Aktivität der untersuchten Chanoclavin-I Dehydrogenasen (FgaDH, ChaDH, FgaDHPr und FgaDHPca) signifikant gesteigert. Es konnte mit Hilfe der durchgeführten Experimente nachgewiesen werden, dass die Aktivitätssteigerung auf eine Aufhebung der Produkthemmung der FgaDH-Reaktion durch FgaOx3Pr3 zurückzuführen ist. Da diese Fähigkeit noch für kein anderes Enzym dieser Klasse beschrieben wurde, handelt es sich bei FgaOx3Pr3 um ein einzigartiges, bifunktionales Enzym, das ein exzellenter Kandidat für die chemoenzymatische Synthese von Mutterkornalkaloiden des Clavin-Typs ist. Über die Produktion von Mutterkornalkaloiden in Penicillium camemberti wurde bisher nicht in der Literatur berichtet. Auch in dieser Arbeit konnten in den Kulturextrakten der untersuchten Stämme keine Mutterkornalkaloide nachgewiesen werden, obwohl ein potenzielles Gencluster für deren Biosynthese identifiziert wurde. Die beiden Gene fgaDHPca und easHPca wurden für die heterologe Expression in E. coli und S. cerevisiae kloniert und konnten in beiden Organismen erfolgreich überproduziert werden. Für EasHPca konnte keine enzymatische Aktivität nachgewiesen werden. FgaDHPca wurde jedoch wie FgaDHPr ebenfalls als Chanoclavin-I Dehydrogenase charakterisiert, welche in ihrer aktiven Form als Tetramer vorliegt. Die ermittelten KM-Werte lagen bei 536 μM für Chanoclavin-I und 528 μM für NAD+. Die mittlere maximale Reaktionsgeschwindigkeit vmax betrug 383 nmol mg-1 min-1 und die Wechselzahl kcat 0,18 s-1. In dieser Arbeit wurde somit erstmals bewiesen, dass P. camemberti die genetische Ausstattung für die Biosynthese von Mutterkornalkaloiden besitzt., Ergot alkaloids are alkaline natural products that belong to the group of indole alkaloids. They have a broad spectrum of pharmacological, but often toxic effects because of their structural similarity with the neurotransmitters dopamine, serotonin and adrenaline. The main producers are filamentous fungi of the families Clavicipitaceae and Aspergillaceae like Claviceps purpurea, Aspergillus fumigatus or Penicillium commune. Starting with L-tryptophan, the first steps of the biosynthesis of ergot alkaloids are similar in all known producing species. After reaching the branch point of the biosynthetic pathways, different end products are formed. This study aims to investigate the remaining common step of the biosynthesis of ergot alkaloids: the conversion of 4-DMA-L-abrine to chanoclavine-I. This step includes at least two oxidation and one decarboxylation reactions. Deletion studies showed that the FAD dependent oxidoreductase FgaOx1 and the catalase FgaCat are essential for the formation of chanoclavine-I. To elucidate the reaction mechanism, both enzymes should be overproduced in E. coli or S. cerevisiae and characterized biochemically. In the case of fgaCat, the heterologous expression in E. coli was successful, but no catalytic activity could be detected for this enzyme alone. The sequence of FgaOx1 from the NCBI database was corrected after an analysis of the intron-exon-structure, but the overproduction of the protein in E. coli or S. cerevisiae was not successful. A bioinformatic comparison of the ergot alkaloid genes from Aspergillus fumigatus, Claviceps purpurea and Arthroderma benhamiae with available genome sequences of Ascomycetes from the NCBI database revealed the presence of ergot alkaloid genes in 15 species that had not been described before. Two of those species are Penicillium roqueforti and Penicillium camemberti which are known for their usage in the industrial production of cheese. In addition to FgaOx1, another 27 protein sequences from the NCBI database were corrected after analysis of their intron-exon-structures in the course of the bioinformatic studies. By identifying the gene clusters and correcting the sequences, this work is a valuable contribution to the future elucidation of the biosynthetic pathways and the analysis of the structural diversity of ergot alkaloids in various producing species. The production of isofumigaclavine A in Penicillium roqueforti has been reported previously, but there were no publications about its biosynthesis at the beginning of this study. The identification of two ergot alkaloid gene clusters and two additional non-clustered genes in different genomic regions laid the foundation for the elucidation of the biosynthetic pathway. Eight genes are very likely involved in the later steps of the isofumigaclavine A biosynthesis and were chosen for cloning and heterologous expression in E. coli and S. cerevisiae. Three of these genes were successfully expressed in E. coli and the recombinant proteins (FgaDHPr, FgaOx3Pr3 and FgaATPr) were purified via affinity chromatography. The in vitro assays showed no enzymatic activity for FgaATPr. FgaDHPr was characterized as a short-chain dehydrogenase/reductase which catalyzes the conversion of chanoclavine-I to chanoclavine-I aldehyde in the presence of NAD+. The native form of the enzyme is likely a pentamer. The KM value for chanoclavine-I was 573 μM and for NAD+ 82 μM. The mean maximum reaction velocity vmax was 326 nmol mg-1 min-1, corresponding to a turnover number kcat 0.15 s-1. FgaOx3Pr3 belongs to the “Old Yellow Enzymes“ and acts as a FMN containing and NAD(P)H dependent oxidoreductase. FgaOx3Pr3 was characterized as a chanoclavine-I aldehyde reductase which catalyzes the formation of festuclavine in the presence of FgaFS from Aspergillus fumigatus or EasG from Claviceps purpurea in vitro. In addition, the enzyme activities of all investigated chanoclavine-I dehydrogenases (FgaDH, ChaDH, FgaDHPr and FgaDHPca) were enhanced significantly by FgaOx3Pr3. These experiments proved that FgaOx3Pr3 overcomes the product inhibition for the FgaDH reaction, resulting in an enhanced product yield. This feature has not been described for other enzymes of the same class, therefore FgaOx3Pr3 is a unique bifunctional enzyme and an excellent candidate for the chemoenzymatic synthesis of clavine-type ergot alkaloids. The production of ergot alkaloids in Penicillium camemberti has not been reported in the literature so far. Although a potential gene cluster for the biosynthesis of ergot alkaloids has been identified in this study, no ergot alkaloids were detected in the culture extracts of the investigated strains. Nevertheless, the two genes fgaDHPca and easHPca were cloned and successfully expressed in E. coli and S. cerevisiae. EasHPca showed no enzymatic activities in the in vitro assays, while FgaDHPca was characterized as a chanoclavine-I dehydrogenase. The native Form of FgaDHPca is likely a tetramer and the KM values were 536 μM for chanoclavine-I and 528 μM for NAD+. The mean maximum reaction velocity vmax was 383 nmol mg-1 min-1 and the turnover number kcat 0.18 s-1.Thus, it was shown for the first time in this work, that P. camemberti has the genetic potential for the biosynthesis of ergot alkaloids.
