Metaphor as non-literal use of a linguistic expression in a particular communication situation serves as the ubiquitous method of expressing of emotions and feelings (s. Skirl/Schwarz-Friesel 2013). The study of emotion metaphors can be seen as a tool to the conceptual systems of emotions (s. Kövecses 2000, Yu 2002, Stefanowitsch 2006 etc.). Based on Critical Cognitive Linguistic (s. Schwarz 2008, Schwarz-Friesel 2013/ 2015, Kertész´ et al. 2012), which divide verbal manifestation from emotion concept and emphasize the importance of corpus study, this study has investigated emotion metaphors (love and sadness) between 2001-2016 in the German and Chinese internet-based love letters and memorial texts. It has contrasted data in three aspects: firstly, whether there are any universal or cultural-specific conceptual correlations of LOVE/SADNESS in both of the data; Secondly, how metaphorical expressions by degree of conventionality (idiom, lexicalized metaphors, cliched metaphors, creative metaphors, innovative metaphors) will be distributed; Thirdly, whether there are any metaphors which denote or/and express two emotions. The result shows that most of conceptualization are common, however, some metaphors are still different on the verbal expression and mapping process. There are also some specific correlations in the both of corpora, Additionally, all of the classifications of metaphorical expressions are discovered in the corpora. Among them the lexicalized and cliched metaphors have the largest proportion. It is difficult to get any similarity of creative and innovative metaphors in the both of the data. Moreover, the study has developed a method to differ emotion-denoted from emotion-expressed metaphors., Metapher als nicht-sprachlicher Sprachgebrauch in einem konkreten Kommunikationszusammenhang gilt als eine wichtige Methode beim Ausdrücken von Emotionen (s. Skirl/Schwarz-Friesel 2013). Demnach können Studien über Emotionsmetaphern einen Beitrag beim Verstehen vom konzeptuellen System von Emotionen leisten (s. Kövecses 2000, Yu 2002, Stefanowitsch 2006 etc.). Basierend auf Kritischer Kognitiver Linguistik (s. Schwarz 2008, Schwarz-Friesel 2013/ 2015, Kertész´ et al. 2012), die verbale Manifestationen von Emotionskonzepten deutlich differenziert und die Bedeutung von korpusbasierten Studien betont, hat die vorliegende Arbeit versucht, Metaphern von LIEBE und Trauer in deutschen und chinesischen Internet-basierten Texten (2001-2016) zu untersuchen. Die Studie hat alle Daten analysiert und miteinander verglichen in Bezug auf die folgenden drei Aspekte: erstens, ob es gemeinsame oder kultur-bedingte Konzeptualisierungsmuster von LIEBE und TRAUER gibt. Zweitens, wie verteilen sich die metaphorischen Äußerungen hinsichtlich Konventionalitätsgrad (Idiome, lexikalisierte Metaphern, klischeehafte Metaphern, Kreative Metaphern, Innovative Metaphern) in beiden Sprachen. Drittens, wie lassen sich emotionsbezeichnende Metaphern von emotionsausdrückenden Metaphern in beiden Sprachen unterscheiden. Es wird in den Ergebnissen dargelegt, dass die Mehrheit der Konzeptualisierungen von LIEBE und TRAUER in den Korpora gemeinsam ist. Allerdings gibt es auch metaphorische Äußerungen, die unterschiedlich auf der Ebene der verbalen Formulierungen und des Übertragsprozesses sind. In jeweiligen Sprachen lassen sich ebenso spezifische Konzeptualisierungsmuster in beiden Korpora finden. In Bezug auf Konventionalitätsgrad werden alle Klassifikationen der Metaphern entdeckt. Davon haben klischeehafte Metaphern das größte Anteil besitzt. Schließlich hat die Arbeit eine Methode entwickelt, emotionsbezeichnende Metaphern von emotionsausdrückenden Metaphern zu unterscheiden.

Formin-like 1 (FMNL1) is a formin-related protein highly expressed in hematopoietic cells and overexpressed in leukemias as well as diverse transformed cell lines. It has been described to play a role in diverse functions of hematopoietic cells such as phagocytosis of macrophages as well as polarization and cytotoxicity of T cells. However, the specific role of FMNL1 in these processes has not been clarified yet and regulation by interaction partners in primary hematopoietic cells has never been investigated. We performed a proteomic screen for investigation of the interactome of FMNL1 in primary hematopoietic cells resulting in the identification of a number of interaction partners. Bioinformatic analysis considering semantic similarity suggested the giant protein AHNAK1 to be an essential interaction partner of FMNL1. We confirmed AHNAK1 as a general binding partner for FMNL1 in diverse hematopoietic cells and demonstrate that the N-terminal part of FMNL1 binds to the C-terminus of AHNAK1. Moreover, we show that the constitutively activated form of FMNL1 (FMNL1γ) induces localization of AHNAK1 to the cell membrane. Finally, we provide evidence that overexpression or knock down of FMNL1 has an impact on the capacitative calcium influx after ionomycin-mediated activation of diverse cell lines and primary cells.
