Die Polyamine Spermidin und Spermin sind wichtige Regulatoren basaler Zellfunktionen, dienen aber darüber hinaus auch hochspezifischen Aufgaben im Säugergehirn. Für die Polyamine und deren Synthese- und Abbauenzyme findet sich in den verschiedenen Gehirnarealen eine unterschiedliche Verteilung. Auch zwischen Neuronen und Gliazellen ist die Verteilung verschieden. Die Synthese des Diamins Putrescin kann über zwei unterschiedliche Stoffwechselwege erfolgen. Beim „klassischen“ Weg wird Arginin zunächst durch Arginase unter Abspaltung von Harnstoff zu Ornithin hydrolysiert und dann in einem weiteren Schritt durch Ornithindecarboxylase zu Putrescin dekarboxyliert. Beim alternativen Weg wird zuerst Kohlendioxid durch Arginindecarboxylase und dann Harnstoff durch Agmatinase entfernt. Beide Stoffwechselwege könnten demselben Ziel dienen. Als Zwischenprodukt des alternativen Weges entsteht jedoch Agmatin, welches ein endogener Ligand für Imidazolinrezeptoren ist und als Neurotransmitter wirkt. Um die Expressionsmuster der beiden Schlüsselenzyme Arginase und Arginindecarboxylase beurteilen und vergleichen zu können, wurden polyklonale, monospezifische Antikörper gegen Arginase-1 und Arginindecarboxylase hergestellt. Mittels dieser Werkzeuge wurde das Rattengehirn immunzytochemisch systematisch untersucht und die Expressionsmuster beider Enzyme in mehreren Gehirnarealen auf der regionalen, zellulären und subzellulären Ebene verglichen. Sowohl Arginase als auch Arginindecarboxylase Enzyme werden in weiten Teilen des Rattengehirns exprimiert. Beide Antikörper markierten im Cortex und Hippokampus Prinzipalneurone, putative Interneurone und Neuropil. In Bezug auf die subzelluläre Lokalisation beider Enzyme zeigte sich jedoch ein auffallender Unterschied in der Markierung der Neurone. Bei dem Antikörper gegen Arginindecarboxylase fand sich eine Verteilung des Immunsignals im gesamten Cytoplasma, die Immunreaktiviät gegen Arginase war bevorzugt im Golgi-Apparat lokalisiert. Die Tatsache, dass die Verteilung von Arginase und Arginindecarboxylase in Bezug auf bestimmte Zellpopulationen große Übereinstimmungen zeigt, und dass für das Zwischenprodukt Agmatin eine Reihe von endogenen Funktionen bekannt sind, legt nahe, dass die Hauptfunktion des alternativen Stoffwechselweges die Synthese von Agmatin und nicht von Putrescin ist, während der klassische Weg Putrescin und Spermidin/Spermin in diesen Neuronen bereitstellt., Polyamines are important regulators of basal cellular functions but also subserve highly specific tasks in the mammalian brain. With this respect, polyamines und the synthesizing und degrading enzymes are clearly differentially distributed in neurons versus glial cells und also in different brain areas. The synthesis of the diamine putrescine may be achieved via two different pathways. In the “classical” pathway initially urea is removed from arginine by arginase. The resulting ornithine is then decarboxylated to putrescine by arginine decaboxylase. With the alternative pathway first carbon dioxide is removed by arginine decarboxlyase and then urea by agmatinase. Both pathways may serve the same purpose. However the intermediate product of the alternative pathway, agmatine, is an endogenous ligand for imidazoline receptors and serves as a neurotransmitter. In order to evaluate and compare the expression patterns of the two gate keeper enzymes arginase und arginine decarboxylase, a polyclonal, monospecific antibodies against arginase-1 und arginine decarboxylase were generated. These tools, were used to immunocytochemically screen the rat brain und compare the expression patterns of both enzymes in several brain areas on the regional, cellular und subcellular level. Arginase as well as arginine decarboxylase are constitutively und widely expressed in the rat brain. In the cerebral cortex und hippocampus, both antibodies labeled principal neurons, putative interneurons and neuropil. Labeling, however, was strikingly different in these neurons with respect to the subcellular localization of the enzymes. While with antibodies against arginine decarboxylase the immunosignal was distributed throughout the cytoplasm, arginase-like immunoreactivity was preferentially localized to Golgi stacks. Given the apparent congruence of arginase und arginine decarboxylase distribution with respect to certain cell populations, and the fact that several endogenous agmatine is known for several endogenous functions are known for agmatine, it seems likely that the synthesis of agmatine rather than putrescine may be the main purpose of the alternative pathway, while the classical pathway supplies putrescine und spermidine/spermine in these neurons.
