In der vorliegenden Arbeit wurden zwei Mutanten des Alpha-Proteobakteriums Rhodospirillum rubrum untersucht, die im Bakteriochlorophyll-Biosyntheseweg unterbrochen sind, um einen Beitrag zum genaueren Verständnis der Biosynthese dieser Moleküle und der einzelnen daran beteiligten Schritte zu liefern. Von den beiden Stämmen ST3 und GN11 wurden die ins Kulturmedium ausgeschiedenen Pigmente aufgereinigt und spektroskopisch analysiert. Ebenfalls wurden sowohl von ST3, als auch von GN11 die in intracytoplasmatischen Membranen gebundenen Pigmente extrahiert und aufgereinigt. Anhand der ermittelten spektroskopischen und chromatographischen Eigenschaften der isolierten Pigmente konnte nachgewiesen werden, dass es sich bei denen ins Kulturmedium ausgeschiedenen und in intracytoplasmatischen Membranen gebundenen Pigmenten um dieselben Bakteriochlorophyllvorstufen handelt. Die im ST3-Stamm angehäuften Vorstufen P660 und P667 (benannt nach dem langwelligsten Absorptionsmaximum) konnten als 2-Devinyl-2-alpha-hydroxyethyl-phaeophorbid a und Phaeophorbid a bestimmt werden. Bei denen im GN11-Stamm angehäuften Vorstufen handelt es sich um ein Gemisch verschiedener Vorstufen in dem mit Sicherheit Magnesium 2,4-Divinylphaeoporphyrin a5 Monomethylester als eine Komponente enthalten ist. Von beiden Stämmen wurden Pigment-Proteinkomplexe aus intracytoplasmatischen Membranen isoliert. Die gewonnen Pigment-Proteinkomplexe zeigen Absorptionsspektren, die in Übereinstimmung sind mit Spektren ganzer Zellen und in beiden Fällen ein deutliches CD-Signal aufweisen. Die Komplexe enthalten neun Proteine unterschiedlicher Größe, für die keine eindeutige Zuordnung getroffen werden konnte. Elektronenmikroskopische Aufnahmen der isolierten Pigment-Proteinkomplexe vom ST3-Stamm zeigen partikuläre, ringförmige Strukturen. Sie neigen zur Aggregation und bilden unter diesen Umständen fädige und runde Formen aus., Two mutants of the alpha-proteo-bacterium Rhodospirillum rubrum, that were blocked in the bacteriochlorophyll biosynthesis pathway, were investigated in this study in order to gain a deeper understanding of the biosynthesis of this molecules and details of the involved steps. From the two strains ST3 and GN11 pigments, excreted into the culture medium, were extracted and cleaned up for spectroscopic analyses. As well pigments bound in intracytoplasmatic membranes from both strains were isolated and cleaned up. Based on the spectroscopic and chromatographic properties of the isolated pigments we were able to show that in ST3 and GN11 excreted and membrane bound pigments represent the same bacteriochlorophyll precursors. In ST3 we found the precursors P660 and P667 (named by the red most absorption maximum) that were identified as 2-devenyl-2- alpha-hydroxyethyl-phaeophorbide a and phaeo¬phorbide a, respectively. The precursors accumulated by GN11 were found to be a mixture of different precursors, definitely containing magnesium-2,4-divinyl-phaeoporphyrin a5 monomethylester. Pigment-protein complexes from intracytoplasmatic membranes were isolated from both strains. The gained pigment-protein complexes showed absorption spectra consistent with spectra of complete cells. In both cases a characteristic CD (circular dichroism) signal could be obtained. The complexes contained at least 9 proteins of different molecular weights, that couldn´t be clearly identified. Electron microscopic investigations of the ST3 pigment-protein-complexes showed particles and, ring like structures. They tend to aggregate and built thread like and globular structures in this case.
