In der vorliegenden Arbeit wird die Röntgenabsorptionsspektroskopie (XAS) an leichten Elementen mit einer optimierten laser-basierten Plasmaquelle (LPQ) im weichen Röntgenbereich hinsichtlich ultrakurzer Belichtungszeiten im ns-Bereich gezeigt. Im Fokus der Untersuchungen stehen dabei organische Moleküle aus den Gruppen der Polymere und der Metalloporphyrine. Die XAS ist eine elementspezifische Messmethode, deren Nahkantenabsorptionsfeinstruktur (NEXAFS) Aussagen über die elektronische Struktur des jeweiligen Moleküls zulässt. NEXAFS-Untersuchungen ermöglichen die Analyse von Molekülen in Lösung, was für Untersuchung von dynamischen Prozessen von großem Interesse ist. Bei Metalloporphyrinen kann dies beispielsweise Aufschluss über die Wechselwirkung zwischen dem kohlenstoffbasierten Porphyrinring und dem zentralen Metallatom geben, welche maßgebliche für ihre jeweilige Funktionsweise verantwortlich ist. Die an Synchrotronstrahlungsquellen etablierte NEXAFS-Spektroskopie im weichen Röntgenbereich erlaubt aufgrund steigender Nachfrage nach Messzeiten nur einen begrenzten Zugang zu dieser Analysemethode. Daher wird eine Optimierung der LPQ des Berlin Laboratory for innovative X-ray Technologies (BLiX) für die NEXAFSSpektroskopie leichter Elemente durchgeführt. Ein an die Quellen- und Laborbedingungen angepasstes Reflexionszonenplatten-Spektrometer ermöglicht hochauflösende NEXAFS-Untersuchungen im sogenannten Einzelschuss-Modus, bei dem eine minimale Strahlenbelastung der Probe vorliegt und eine Überwachung der Probendegradation möglich ist. Die Ergebnisse stellen die Vorstufe zu „pump-probe“-Konzepten im Labor zur Analyse von dynamischen Prozessen dar. Als Hauptkomponente organischer Moleküle steht die Untersuchung der K-Kante von Kohlenstoff (C) im Vordergrund. Einzelschuss NEXAFS-Messung von Polymeren zeigen die Machbarkeit der Methode und ermöglichen die Einordnung zu bereits veröffentlichten Arbeiten. Das unter anderem in den Lebenswissenschaften Einsatz findende Chlorophyllin-Natrium-Kupfer-Salz (Cu-Chl) kann als Modellsystem der Metalloporphyrine angesehen werden. Mit auf Dichtefunktionaltheorie basierenden Berechnungen können die NEXAFS-Spektren hinsichtlich elektronischer Übergänge und der Molekülorbitalverteilung der Cu-Chl Moleküle interpretiert werden. Analog wurden die mit Synchrotronstrahlung durchgeführten Magnesium NEXAFS Untersuchungen des photosynthetischen Moleküls Chlorophyll (Chl) a interpretiert, bei denen es erstmalig gelungen ist, intakte Chl a Moleküle in Lösung mit einer röntgenspektroskopischen Methode zu detektieren. X-ray absorption spectroscopy (XAS) on light elements with an optimized laserproduced plasma (LPP) source in the soft X-ray region is presented facilitating exposure times in the ns regime. A key aspect is the investigation of organic molecules belonging to polymers and metalloporphyrins. With XAS the local probing of specific elements in a molecule is feasible. The interpretation of the characteristic Near Edge X-ray Absorption Fine Structure (NEXAFS) of an absorption edge can lead to information about the electronic molecular structure. NEXAFS spectroscopy renders the investigation of liquid samples possible, which is of great interest especially for monitoring dynamic processes. In the special case of metalloporphyrins this can lead to information about the interaction between the carbon containing porphyrin ring and the central metal ion, which is responsible for the particular function of the molecule. Up to now, NEXAFS spectroscopy in the soft X-ray region is a well-established method at synchrotron radiation facilities but due to the high demand for beamtime, there is only limited access. Therefore, an optimized LPP soft X-ray source at the Berlin Laboratory for innovative X-ray technologies (BLiX) for NEXAFS spectroscopy of light elements is presented as well as a reflection zone plate (RZP) spectrometer for high resolution XAS adapted to the laboratory and source requirements. This RZP spectrometer enables a single shot mode of the LPP source using the emission of a 1 ns single pulse. With this method the sample is exposed to a minimal radiation dose and an online monitoring of sample degradation is possible. The investigations represent the primary stage for future laboratory based „pump-probe“ investigations on dynamic processes of biomolecular systems. As the main component of organic molecules the investigation of the carbon K edge focuses on this work. NEXAFS investigations of the different polymers show the feasibility of the single shot measurement mode and allow a comparison of the laboratory-based investigation method to already published work. The investigated chlorophyllin sodium copper salt (Cu-Chl) which is used in life science applications can be seen as a model system for metalloporphyrins. Based on density functional theory calculations the characteristic NEXAFS structures can be assigned to electronic transitions and the molecular orbital distribution of the Cu-Chl molecule. A comparable investigation of the magnesium K edge performed at the synchrotron facility BESSY II is carried out for Chlorophyll (Chl) a, which is an important molecule for photosynthesis. For the first time, intact Chl a molecules in solution were detected and molecular orbitals can be assigned to occurring NEXAFS-structures.
