Nervensysteme basieren auf der Zusammenarbeit zahlreicher Nervenzellen. Um ihre Arbeitsweise aufklären zu können, benötigt man eine Methode, die es erlaubt, die Aktivität einer großen Zahl von Nervenzellen auf der Ebene einzelner Zellen zu beobachten. In der vorliegenden Arbeit wurden hierfür geeignete Sensoren entwickelt. Mit rauscharmen Elektrolyt-Oxid-Silizium-Feldeffekttransistoren (EOS-FETs) konnten die beim Feuern von Säugetierneuronen entstehenden sehr kleinen extrazellulären Spannungen detektiert werden. Ausgehend von den Skalierungsgesetzen der Zell-Transistor-Kopplung und der Rauschmechanismen wurde eine optimale Sensorgröße abgeleitet. Möglichkeiten zur Reduzierung des Verstärker- und Transistorrauschens wurden untersucht und eine Prozeßführung entwickelt, die rauscharme, glatte und optisch homogene Chips liefert. Die Charakterisierung der nach diesem Prozeß gebauten Transistoren zeigte sowohl die erwartete Abhängigkeit des Rauschens von den Parametern des Transistors als auch die gute Übereinstimmung der Kennlinien mit dem Ergebnis eines numerischen Simulationsprogramms für Feldeffekttransistoren. Nervenzellen aus dem Hippocampus der Ratte wurden auf den Chips kultiviert. Die spontanen Aktionspotentiale einzelner Neurone konnten verletzungsfrei und mit gutem Signal-Rausch-Verhältnis detektiert werden. Das Transistorrauschen erwies sich als so gering, daß bereits die thermodynamische Grenze extrazellulärer Messungen erreicht wurde: Das Rauschen wurde nicht mehr von den Transistoren, sondern von den Zellen dominiert. Ein zur Beschreibung dieses Abdichtungsrauschens entwickeltes Modell konnte die Beobachtungen, insbesondere den ungewöhnlichen Exponenten der Frequenzabhängigkeit, erklären. Zum Vergleich mit der traditionellen Methode für extrazelluläre Messungen, planaren Metallelektroden, wurden dichte, erwachsene Kulturen aus Rattenneuronen untersucht. Die dabei beobachteten Signale waren um knapp eine Größenordnung stärker als es von Metallelektroden berichtet wird. Die Diskrepanz wird diskutiert und auf die Andersartigkeit der Meßmethoden und der Kontaktausbildung zurückgeführt. Die hier vorgestellten Transistoren weisen mehrere für Sensoren wünschenswerte Eigenschaften auf: Ihr Rauschen bewegt sich nahe an der thermodynamischen Grenze, ihre glasartige Oberfläche ist biokompatibel, chemisch homogen und extrem glatt, so daß das biologische System durch die Anwesenheit der Sensoren nicht beeinflußt wird. Sie sind in der Lage, neben den elektrischen auch chemische Signale der Zellen zu detektieren, und kommen in gewissem Sinne sogar ohne jegliche Zellsignale aus, können sei doch bereits die bloße Adhäsion von Zellen nachweisen. Diese Merkmale machen die Transistoren interessant für zahlreiche weitere Anwendungen in Neurobiologie, Zellbiologie und Pharmakologie. Und mit der Beobachtung des Abdichtungsrauschens steht eine neue Methode zur Untersuchung der Zelladhäsion zur Verfügung, die sich zudem auch in vielen nichtbiologischen Situationen anwenden läßt. Nervous systems are based on the cooperation of numerous neurons. In order to investigate their functioning a method is required which allows the observation of the activity of a large number of neurons at the level of individual cells. In this thesis, suitable sensors for this application have been developed. With low-noise electrolyte-oxide-silicon field-effect transistors the small extracellular voltages arising during the elicitation of action potentials could be detected. Starting from the scaling laws of the cell-transistor-coupling and the noise mechanisms an optimal sensor size was determined. Measures to reduce the noise of amplifier and transitors were examined and a process was developed which yields chips with low noise and a smooth and optical homogeneous surface. The characterisation of the transistors built with this process showed the expected dependency of the noise on the transistors' parameters and a good agreement of the device behaviour with the results of a numerical simulation package for field-effect transitors. Neurons from the rat's hippocampus were cultivated on the chips. Spontaneous action potentials were recorded with good signal-to-noise ratios and without injuring the neurons. The transistor noise proved to be so low that the thermodynamic limit of extracellular measurements became visible: The noise was not dominated any more by the transitors but by the cells. A model developed to describe this sealing noise was able to to explain the observations, in particular the unusual exponent of frequency dependence. In order to draw a comparison with the traditional method for extracellular recordings (planar metal electrodes) dense mature cultures of rat neurons were examined. The observed signals were about a magnitude larger than those reported from metal electrodes. The discrepancy is discussed and attributed to the differences of the measurement methods and the contact types. The transistors presented in this work have several features desirable for sensors: Their noise is close to the thermodynamic limit, their glass-like surface ist biocompatible, chemical homogeneous and extremly flat, which prevents influencing the biological system by the sensors' presence. They are able to record not just the electrical, but also the chemical signals of the cells and can even do without any cell signals at all, as they are able to detect the mere adhesion of cells. These features render the transistors attractive for many additional applications in neurobiology, cell biology and pharmacology. The observation of adhesion noise provides a new method to investigate cell adhesion and in addition can be applied in many non-biological situations.

Die Aktivität neuronaler Netze kann durch Feldeffekttransistoren mit hoher Auflösung nicht invasiv verfolgt werden. Bis jetzt war die Messung mit Feldeffekttransistoren auf elektrische Signale der Zellen beschränkt. Ein weiterer wichtiger Bereich der neuronalen Informationsverarbeitung, der chemisch an speziellen Stellen, den Synapsen, durch das Ausschütten von Vesikeln stattfindet, wurde mit Feldeffekttransistoren bislang nicht untersucht. Diese Arbeit zeigt, daß es möglich ist, die Ausschüttung von Vesikeln einer Zelle mit dem Feldeffekttransistor zu messen. Neuroendocrine Zellen aus der Rindernebenniere, die ein etabliertes Modellsystem für die synaptische Übertragung sind, werden auf der Oberfläche eines linearen Arrays von Feldeffekttransistoren kultiviert. Unter Stimulation der Zellen mißt der Transistor Signale, welche mit der Referenzmethode zum Detektieren der Vesikel, der Amperometrie, verglichen werden. Dieser Vergleich zeigt, daß der Transistor die Exozytose einzelner Vesikel mißt und daß fast alle ausgeschütteten Vesikel vom Transistor detektiert werden. Im Gegensatz zu den Signalen der Amperometrie besitzen die Signale der Transistoren eine starke Abhängigkeit von der extrazellulären Pufferkonzentration; ein Indiz, daß die Transistorsignale auf der Ausschüttung von Protonen durch die Vesikel beruhen. Dieses wird durch das Zerstören der granularen pH-Gradienten mit der Pore Nigericin sowie den Carrier Valinomycin bestätigt. Während die amperometrischen Messungen von diesen Substanzen unbeeinflußt bleiben, erlöschen die Signale im Transistor nach ihrem Wirken fast gänzlich. Mittels Transistoren der Gategröße von 2x2 m2 und eines Abstandes von 4,6 m in einer linearen Anordnung ist es möglich, mit mehreren Transistoren die Exozytose unterhalb einer Zelle zu messen. So kann eine räumliche Karte der Ausschüttung, die physiologisch an bestimmte Stellen in der Zelle gekoppelt ist, erstellt werden. Ein Modell der Transistorsignale berücksichtigt die Freisetzung von Protonen in den engen Spalt zwischen Zelle und Transistoroberfläche, ihre Diffusion entlang des Spaltes und ihre Reaktion mit den durch den Transistor bereitgestellten dissoziierten Oberflächengruppen. Ein Vergleich des Modells mit dem Experiment zeigt, daß die Transistorsignale nur in dem Kontext der Komplexität der Vesikelfusion und des Zerfalls der granularen Matrix erklärt werden können. Da eine Vielzahl an Vesikeln eine erhöhte Protonenkonzentration besitzt, ist in dieser Arbeit nicht nur eine neue fundamentale Kopplung einer Zelle an einen Feldeffekttransistor und damit ein neuartiges hybrides System experimentell und theoretisch erstellt, sondern es wird auch eine neue Methode zum Detektieren der Vesikelausschüttung begründet. The activity of neuronal nets can be resolved non invasive and at a high spatial resolution by field effect transistors. Up to now measurements were restricted to electric signals of cells. Another important way of information processing takes place at special sites, the synapses, where vesicles fuse with the cell membrane and extrude their content. This way of information flow has not been examined with field-effect transistors so far. Here we show that it is possible to detect the release of vesicles by field-effect transistors. Neuroendocrine cells from the bovine adrenal medulla are an established model system for the synaptic transmission. We culture these cells on linear arrays of field-effect transistors. Under stimulation of the cells the transistors detect sharp signals. We compare these data with the reference method amperometry and conclude that the transistors measure the release of individual vesicles at a high resolution. Contrary to the amperometric signals the transistor signals exhibit a dependence of the extracellular buffer concentration, a first evidence, that the transistor signals are based on the release of protons by the vesicles. This is validated by the destruction of the vesicular pH gradients through the pore nigericin and the carrier valinomycin. While the amperometric measurement remains unaffected, there is a massive reduction of transistor signals after the application of these substances. Arrays of transistors with a gate size of 2x2 m2 and a pitch of 4.6 m make it possible to record the release of vesicles beneath a single cell with several transistors simultaneously. Thus a map of the release sites of a cell can be generated. A model of the transistor signals includes the release of the protons within the narrow cleft between the cell and the transistor surface, the proton diffusion along this cleft and the binding of the protons to the surface groups provided by the transistor. A comparison of this model to the experiment makes clear that the transistor signals can only be understood in the context of the complexity of the vesicle fusion and the dissociation of the granular matrix. Since many more vesicles than only those of chromaffin cells are acidic, we could establish a novel method of vesicle release detection and described a fundamental coupling of a cell to a computer chip. We present a theoretical study of a spin field-effect transistor realized in a quantum well formed in a p--doped ferromagnetic-semiconductor- nonmagnetic-semiconductor-ferromagnetic-semiconductor hybrid structure. Based on an envelope-function approach for the hole bands in the various regions of the transistor, we derive the complete theory of coherent transport through the device, which includes both heavy- and light-hole subbands, proper modeling of the mode matching at interfaces, integration over injection angles, Rashba spin precession, interference effects due to multiple reflections, and gate-voltage dependences. Numerical results for the device current as a function of externally tunable parameters are in excellent agreement with approximate analytical formulae., 9 pages, 11 figures
