Although defects in protein crystals have been studied mainly by x-ray diffraction, optical methods, for the first time, have started to be employed widely to reveal not only lattice defects in protein crystals but also surface topography of the crystal at a molecular level. This observation became possible by developing various optical microscopy and new phase-shift interferometry.
