Universidade de Costa Rica A cultura do café tem apresentado uma série de doenças que provocam severos danos na cafeicultura de diversos países. Visando minimizar esses problemas, programas de melhoramento genético têm desenvolvido cultivares com resistência à fatores bióticos e abióticos. No entanto, o uso dessa cultivares associado a autogamia e origem do cafeeiro arábica tem resultado em estreita relação genética entre os genótipos e, consequentemente, maior vulnerabilidade a doenças e pragas. Técnicas tradicionais de inoculação do patógeno complementadas com a biologia molecular permitem verificar a presença de genes que conferem resistência a ferrugem (Hemileia vastatrix), principal doença da cultura, e ao Colletotrichum kahawae causador da antracnose verde dos grãos, além de auxiliar o estudo de diversidade das cultivares. Marcadores ligados a genes que conferem resistência a doenças, complementam os trabalhos clássicos de fenotipagem das plantas de café na determinação da resistência ou a susceptibilidade a patógenos. Diante desse panorama, esse trabalho teve como objetivo a verificação da diversidade genética das cultivares de café da América Central, utilizando 16 marcadores microssatélites distribuídos aleatoriamente no genoma de C. arabica. Além disso, a resistência dessas cultivares foi verificada por meio da fenotipagem com as raças II e XXXIII de H. vastatrix e genotipagem com marcadores moleculares ligados a genes de resistência a H. vastatrix e a C. kahawae. As análises moleculares com marcadores aleatórios revelaram diversidade entre e dentro das cultivares analisadas, sendo possível obter o padrão molecular da maioria delas. Essas análises permitiram também observar erros de identificação de cultivar, misturas ou hibridações nas cultivares. Das 36 cultivares fenotipadas para resistência, 30 foram resistentes a raça II e 26 a raça XXXIII de H. vastatrix. Na genotipagem, os cafeeiros apresentaram variabilidade e alelos que conferem resistência a H. vastatrix, complementado os resultados fenotípicos, sugerindo uma piramidação de genes de resistência nos cafeeiros da América Central. Several diseases can cause severe damages to coffee plants in many coffee- producing countries. In order to minimize those problems, plant breeding has developed plants with resistance to biotic and abiotic factors. However, the use of Arabica coffee cultivars coupled with self-fertilization of the plant and the origin of the crop resulted in a close genetic relationship between cultivars, leading to susceptibility to diseases and pests. Traditional techniques complemented by molecular biology allow the verification of genes that confer resistance to coffee leaf rust (Hemileia vastatrix), the main disease of the crop, and to Colletotrichum kahawae (CBD), the causal agent of green anthracnose in grains. Microsatellite markers (SSR) are extensively employed to obtain unique molecular patterns (fingerprinting). Specific markers linked to genes that confer resistance to diseases complement classic phenotyping and help determine plant resistance or susceptibility to pathogens. In that sense, this work aimed to verify the genetic diversity of coffee cultivars from Central America using 16 SSR markers randomly distributed in C. arabica genome. Moreover, the resistance of these cultivars was verified through phenotyping with races II and XXXIII of H. vastatrix and genotyping with molecular markers linked to resistance genes to H. vastatrix and C. kahawae. Fingerprinting analysis also revealed cultivars with the same molecular pattern, suggesting misidentification, mixtures, or hybridization of the analyzed cultivars. Of the 36 cultivars phenotyped for disease resistance, 30 were resistant to H. vastatrix race II and 26 to race XXXII. In genotyping, coffee plants demonstrated variability and alleles that confer resistance to H. vastatrix, suggesting a pyramiding of resistance genes in Central American cultivars. Conselho Nacional de Desenvolvimento Científico e Tecnológico O café é a segunda principal commodity no mercado mundial. A espécie Coffea arabica é uma das principais espécies cultivadas no mundo. No entanto, a cultura hospeda uma grande variedade de patógenos e pragas. Os patógenos Hemileia vastatrix e Colletotrichum kahawae são as doenças mais importantes da cultura, e a praga bicho-mineiro, impacta severamente a produtividade e qualidade de bebida. Assim, como forma de controle, o desenvolvimento de cultivares resistentes, com a utilização de marcadores moleculares auxiliam no progresso de melhoramento genético da cultura. O objetivo do trabalho foi identificar cafeeiros portadores de genes de resistência para as principais doenças do café; determinar a diversidade genética por meio de características morfológicas e estimar os parâmetros genéticos e os ganhos com a seleção utilizando a metodologia de modelos mistos (REML/BLUP). O estudo foi composto por duas populações, a primeira, os cafeeiros em geração F 2 são provenientes do cruzamento entre cultivar resistente a ferrugem e CBD e outra ao bicho-mineiro, enquanto que a segunda, são originados de cruzamentos de cafeeiros arábicas com fontes de resistência (seleção indiana). A identificação de cafeeiros resistentes foi feita fenotipicamente e/ou molecularmente. A análise de diversidade genética foi baseada na distância generalizada de Mahalanobis e a seleção fenotípica, por meio do procedimento REML/BLUP. Ambas populações apresentaram resistência múltipla para as principais doenças com diferentes combinações. Com base nas análises utilizando o método REML/BLUP, foi possível selecionar cafeeiros promissores por serem geneticamente divergentes e complementares pela análise de agrupamento e por proporcionar ganhos com a seleção principalmente para a característica infestação de bicho- mineiro. Além disso, foi possível observar que os materiais em desenvolvimento apresentam superioridade em relação as cultivares comerciais. Palavras-chave: Hemileia vastatrix. Colletotrichum kahawae. Melhoramento preventivo. Piramidação de genes. Coffee is the second main commodity on the world market. The Coffea arabica species is one of the main cultivated species in the world. However, the crop hosts a wide variety of pathogens and pests. The pathogens Hemileia vastatrix and Colletotrichum kahawae are the most important diseases of the crop, and the leaf miner pest severely impacts productivity and beverage quality. Thus, as a form of control, the development of resistant cultivars, with the use of molecular markers, helps in the progress of genetic improvement of the crop. The objective of this work was to identify coffee plants carrying resistance genes for the main coffee diseases; determine genetic diversity through morphological traits and estimate genetic parameters and selection gains using the mixed model methodology (REML/BLUP). The study was composed of two populations, the first, the coffee trees in the F 2 generation, come from the cross between a cultivar resistant to rust and CBD and the other to the leaf miner, while the second, are originated from crosses of arabica coffee with sources of resistance. (Indian Seletion). The identification of resistant coffee trees was made phenotypically and/or molecularly. Genetic diversity analysis was based on generalized Mahalanobis distance and phenotypic selection, using the REML/BLUP procedure. Both populations showed multiple resistance to the main diseases with ivomercial combinations. Based on the analysis using the REML/BLUP method, it was possible to select promising coffee plants because ivome are genetically ivomercialiv and complementary by cluster analysis and because ivome provide gains with selection mainly for the leaf miner infestation characteristic. Coordenação de Aperfeiçoamento de Pessoal de Nível Superior Neste trabalho estão descritas as sínteses e caracterizações de 67 compostos derivados de sulfonamidas, sendo 32 inéditos, com potencial atividade antifúngico para o controle de doenças de plantas. A primeira etapa consistiu na síntese de 55 sais complexos de zinco com diferentes ligantes N-R-sulfonilditiocarbimatos derivados de sulfonamidas. Estes diânions foram isolados na forma de sais de amônio e fosfônio. Os sais foram caracterizados por espectroscopias no infravermelho, de ressonância magnética nuclear de 1 H e 13 C, por análises elementares. Também foram realizados estudos estruturais por difração de raio-X. Estes sais foram submetidos a ensaios biológicos para avaliação de sua atividade inibidora da germinação de Hemileia vastatrix, apresentando uma excelente atividade in vitro. Os mais ativos foram selecionados para testes in vivo e foram capazes de controlar o desenvolvimento da ferrugem do café, sendo um deles mais eficaz que o fungicida mancozeb na dose de 300 µmolL -1 . Estes mesmos sais foram submetidos a testes de inibição de crescimento micelial de fungos dos gêneros Botrytis e Rhizoctonia, in vitro, nas concentrações de 50 e 150 µmolL -1 , onde apresentaram boa atividade. O composto mais ativo foi submetido a testes de toxicidade com abelha (Apis mellifera) e foi verificada baixa toxicidade. A segunda etapa consistiu na síntese de alilsulfonamidas derivadas de adutos de Morita-Baylis-Hillman (MBH). Dois adutos MBH foram sintetizados através do acoplamento de um aldeído e alquenos contendo os grupos retiradores de elétrons CN e CO 2 CH 3 . Os adutos de MBH foram utilizados como intermediários para a obtenção de doze alilsulfonamidas por meio de reações com sulfonamidas primárias. As alilsulfonamidas foram totalmente caracterizadas por métodos espectroscópicos. Estudos por difração de Raios-X confirmaram a estereoquímica E para alilsulfonamidas contendo um grupo éster, e Z quando possuem o grupo ciano. Testes in vitro mostraram que as alilsulfonamidas apresentam uma ação antifúngica moderada, inibindo o crescimento micelial de B. cinerea.Palavras-chave: Alilsulfonamida. Ditiocarbimato de zinco. This work describes the synthesis and characterization of 67 compounds derived from sulfonamides, 32 of which were unpublished, with potential antifungal activity for the control of plant diseases. The first step consisted on the synthesis of 55 complex zinc salts with different N-R-sulfonyldithiocarbimates ligands derived from sulfonamides. These complex-dianions were isolated in the form of salts of ammonium and phosphonium. The salts were characterized by infrared, 1 H and 13 C nuclear magnetic resonance spectroscopies, elemental analyses and by X-ray diffraction studies. These salts were submitted to biological tests to evaluate their influence on the germination of Hemileia vastatrix, showing an excellent in vitro inhibitory activity. The most active salts were selected for in vivo tests and were able to control the development of the coffee leaf rust, one of them being more effective than the fungicide mancozeb at 300 µmolL -1 . These salts were submitted to mycelial growth inhibition tests with Botrytis cinerea and Rhizoctonia solani, in vitro. The zinc-dithiocarbimates presented good activity against B. cinerea, being less active against R. solani, at the concentrations of 50 and 150 µmolL -1 . The most active compound was submitted to toxicity tests with honeybees (A. mellifera) and low toxicity was verified. The second step consisted on the synthesis and characterization of allylsulfonamides derived from Morita-Baylis- Hillman (MBH) adducts. Two MBH adducts were synthesized through the coupling of benzaldehyde and two alkenes containing the electron withdrawing groups CN and CO 2 CH 3 . The MBH adducts were used as intermediates to obtain twelve allylsulfonamides, which were fully characterized and submitted to in vitro tests against B. cinerea. X-ray diffraction studies confirmed the E stereochemistry for the allylsulfonamides containing the ester group, and the Z configuration when they present the cyano group. In vitro tests showed that the allylsulfonamides have a moderate antifungal action, inhibiting the mycelial growth of B. cinerea. Keywords: Allylsulfonamide. Zinc-dithiocarbimate. Fungicide. Coordenação de Aperfeiçoamento de Pessoal de Nível Superior As bis(piridilmetileno)ciclohexanonas (14C-15C), a 125 µmol L -1, inibiram totalmente a germinação de H. vastatrix e apresentaram IC 50 estimados em 1,3 e 2,6 vezes menores que o obtido para o fungicida comercial oxicloreto de cobre. A bis(2-piridilmetileno)ciclohexanona 13C foi ativa contra Rhizoctonia solani com IC 50 = 155,5 nmol por plugue. Esses compostos mais ativos foram submetidos a teste de citotoxicidade contra a linhagem de células Vero e apresentaram melhor perfil toxicológico quando comparado ao oxicloreto de cobre. Além disso, foram realizados cálculos in silico de parâmetros físico-químicos para bis(ilideno)ciclohexanonas, e os valores obtidos foram considerados adequados para agroquímicos. As 2-amino-4H-pirano-3-carbonitrilas (1D-14D) foram sintetizadas com bons rendimentos (29-94%). A maioria destes compostos, quando testados na concentração de 200 nmol por plugue, inibiu em pelo menos 50% o crescimento micelial de Botrytis cinerea. No entanto, foi observado que as soluções estoque preparadas para testes biológicos apresentavam alteração de cor quando mantidas na bancada do laboratório, iluminadas por lâmpadas de LED comuns. Isso nos levou a investigar a possível degradação fotoinduzida destes compostos. Experimentos de FT-IR-ATR e HPLC confirmaram a fotodegradação. O produto da reação fotoquímica do composto 7D, que apresentou o maior percentual de conversão fotoquímica, foi isolado e identificado como (E)-7-ciano-5-(4-nitrobenzilideno)-8-(4- nitrofenil)biciclo[4.2.0]oct-1(6)-eno-7- carboxamida (7D*). Este composto foi testado contra B. cinerea, mas nenhum efeito foi observado. Enfim, as bis(ilideno) cicloalcanonas e 2-amino- 4H-pirano-3-carbonitrilas foram avaliadas quanto ao seu efeito fitotóxico sobre folhas de plantas daninhas e efeito inibidor da AChE. Os compostos não mostraram efeito fitotóxico apreciável. A 2-amino-4H-pirano-3-carbonitrila (12D) produziu a maior inibição enzimática AChE (52,3%) a 50 µmol L -1 . Com o objetivo de se obterem compostos com maiores atividades anti-AChE, foram sintetizados análogos ao composto 12D. Entretanto, nenhum dos compostos produziu aumento da atividade inibitória. Desta forma, a principal atividade observada para os compostos foi a ação contra fungos fitopatogênicos, o que indica que tais compostos apresentam potencial para o desenvolvimento de novos fungicidas de interesse agrícola. In the present work, extracts from leaves of the tree Bacupari (Garcinia gardneriana) and a series of compounds derived from triterpene (lupeol), compounds analogues to curcumin and chalcones (bis(ylidene) cyclohexanones) and 4H-pyrans (2-amino-4H-pyran-3-carbonitriles) were produced and evaluated for their antifungal, phytotoxic, and anti-acetylcholinesterase (AChE). The extracts of G. gardneriana leaves were obtained using the solvents hexane, ethyl acetate and ethanol, successively. The hexane extract inhibited the germination of Hemileia vastatrix urediniospores by 98% with an IC 50 value of 211.3 µg mL -1 . The hexane extract showed low phytotoxic effect on cucumber and sorghum seed germination, and low anti-AChE activity at the concentrations evaluated. GC-MS analysis revealed lupeol as the main constituent of the hexane extract. Lupeol was isolated and tested on H. vastatrix and presented low antifungal activity. Thus, the effect produced by the extract was attributed to the synergistic action involving lupeol and minor components of the extract. Lupeol was used to synthesize a series of esters (1B-8B) and to perform the evaluation of their biological activities. However, no appreciable effects were observed in the tests performed. The bis(ylidene) cyclohexanones (1C-20C) were synthesized with good yields (28-97%). bis(pyridylmethylene) cyclohexanones (14C-15C), at 125 µmol L -1 , totally inhibited the germination of H. vastatrix, and presented IC 50 values estimated as 1.3 and 2.6 times lower than that obtained for the commercial fungicide copper oxychloride. Bis(2-pyridylmethylene) cyclohexanone 13C was active against Rhizoctonia solani with IC 50 = 155.5 nmol per plug. These more active compounds were submitted to cytotoxicity test against a Vero cell line and presented a better toxicological profile when compared to copper oxychloride. In addition, in silico calculations of the physicochemical parameters for bis(ylidene) cyclohexanones were performed, and the values were considered suitable for agrochemicals. The-amino-4H-pyran-3-carbonitriles (1D-14D) were synthesized in good yields (29-94%). Almost all compounds, when tested at 200 nmol per plug, inhibited at least 50% of the mycelial growth of Botrytis cinerea. However, we observed that stock solutions prepared for biological tests sunderwent color changing when kept on the laboratory bench, illuminated by common LED daylight tubes. This prompted us to investigate the possible photoinduced degradation of these compounds. FT-IR-ATR and HPLC experiments were carried out and confirmed the photodegradation. The product of the photochemical reaction of compound 7D, which showed the highest percentage of photochemical conversion, was isolated and identified as (E)-7-cyano-5-(4-nitrobenzylidene)-8-(4-nitrophenyl)bicyclo[4.2.0]oct-1(6)- ene-7-carboxamide (7D*). This compound was tested for antifungal activity against B. cinerea, but no effect was observed. Finally, as bis(ylidene) cycloalkanones and 2-amino-4H-pyran-3- carbonitriles were evaluated for their phytotoxic effect on leaves of weeds and for their AChE inhibitory activity. None of the produced phytotoxic activity. 2-amino-4H-pyran-3-carbonitrile (12D) produced the highest AChE enzymatic inhibition (52.3%) at 50 µmol L -1 . In order to obtain compounds with greater anti-AChE activities, analogs to compound 12D were synthesized. However, none of the compounds produced an increase in inhibitory activity. Thus, the main activity observed for the compounds was the action against phytopathogenic fungi, which indicates that such compounds present potential for the development of new fungicides of agricultural interest. Conselho Nacional de Desenvolvimento Científico e Tecnológico Os principais problemas fitossanitários nas lavouras de café arábica são a ferrugem causada pelo fungo Hemileia vastatrix e Coffee Berry Disease (CBD) causada pelo fungo Colletotrichum kahawae, sendo uns dos principais responsáveis pela redução da produtividade do cafeeiro e qualidade de bebida. Uma forma socioambiental eficiente para o controle dessas doenças é o uso de cultivares resistentes obtidas por meio de melhoramento genético. No entanto, o melhoramento genético do cafeeiro é um processo demorado, que pode levar cerca de 25 anos para se obter uma nova cultivar. Nesse sentido, a utilização de métodos de melhoramento associados a marcadores moleculares é uma alternativa viável para acelerar o processo de desenvolvimento de novas cultivares de café. Uma das aplicações de extrema relevância dos marcadores moleculares no melhoramento genético de plantas consiste em sua utilização como ferramenta auxiliar nos procedimentos de seleção assistida por marcadores moleculares (SAM). A SAM no processo de piramidação de alelos de resistência a doenças é amplamente eficiente, uma vez que permite a seleção na ausência do patógeno, anula o efeito do ambiente, de pleiotropia e epistasia e permite a eliminação dos genótipos indesejáveis nas primeiras gerações de seleção. Desse modo, o objetivo do presente estudo foi implementar a seleção assistida no melhoramento de duas populações de C. arabica, C6T-10 e C6T-24, em geração F 2 de seleção recorrente. Estudo da diversidade genética e análises morfoagronômicas dessas populações também foram realizadas para auxiliar na seleção dos cafeeiros. Para tanto, DNA de 148 cafeeiros pertencente às duas populações foi extraído e amplificado com marcadores moleculares associados a genes de resistência a ferrugem e ao CBD, bem como marcadores microssatélites distribuídos aleatoriamente no genoma. Para resistência a ferrugem foram utilizados marcadores ligados a três locos, sendo dois QTL que correspondem a genes maiores de resistência as raças I, II e patótipo 001 de H. vastatrix e um gene RGA (Disease Resistance Gene Analogs). Para CBD foram utilizados dois marcadores que se encontram flanqueando o gene Ck-1. Os marcadores microssatélites foram usados para o estudo de diversidade genética. Os dados dos marcadores associados aos dois QTL sugerem que todos cafeeiro da população C6T-24 possuem resistência para as raças I, II e patótipo 001 de H. vastatrix, pois a presença de um alelo dominante em um dos dois locos é suficiente para que o indivíduo seja resistente. O gene RGA associado a resistência a esse patógeno foi identificado em 117 cafeeiros. Para resistência a C. kahawae, 46 cafeeiros da população C6T-10 e 59 da população C6T-24 apresentaram o gene Ck-1. Foram identificados 37 cafeeiros da população C6T-10 e 23 da população C6T-24 com a piramidação dos quatro locos de resistência a H. vastatrix e C. kahawae. Esses mesmos cafeeiros foram avaliados como resistentes à ferrugem em nível de campo no ano de 2019. Outras características morfoagronômicas foram analisadas, permitindo a seleção de genótipos contendo piramidação de genes de resistência e características de interesse agronômico. Para auxiliar na seleção de genótipos a serem cruzado para formar novo ciclo de retrocruzamento, foi realizado estudo da diversidade dos cafeeiros, usando a análise de agrupamento UPGMA. Observou-se a formação de quatro grupos principais, permitindo evidenciar a diversidade dentro da população C6T-10. Com base nesse trabalho, foi possível selecionar cafeeiros em geração F 2 contendo a piramidação de alelos de resistência a H. vastatrix e C kahawae, bem como genótipos contendo diversidade que podem ser usados para cruzamento e formação de nova população para seleção recorrente. The main phytosanitary problems in Arabica coffee washers are rust that is caused by the fungus Hemileia vastatrix and Coffee Berry Disease (CBD), which is caused by the fungus Colletotrichum kahawae, being one of the main responsible for the reduction of coffee and drink quality. An efficient socioenvironmental way to control these diseases is the use of resistant cultivars, preventing the means of genetic breeding. However, the coffee plant breeding is a long process, which can take about 25 years to obtain a new cultivar. Thus, the use of breeding methods associated with molecular markers is a viable alternative to accelerate the process of developing new coffee cultivars. One of the extremely relevant applications of molecular markers in the plant breeding method consists of its use as an auxiliary tool in the selection procedures assisted by molecular markers (SAM). In SAM, the process of gene stacking disease-resistant alleles is efficient, as it allows selection in the absence of pathogens, cancels the environmental effects, pleiotropy, and epistasis and allows the selection of undesirable genotypes in the first selection tests. Now the objective of the present study was to implement a selection assisted in the improvement of two species of C. arabica, C6T-10, and C6T-24, in the F2 generation of recurrent selection. Genetic diversity studies and morpho- agronomic analyzes of these were also carried out for auxiliary coffee selection. For this purpose, the DNA of 148 coffee trees from two species was extracted and amplified with molecular markers associated with rust resistance genes and CBD, as well as microsatellite markers distributed randomly in the genome. For resistance to rust, markers linked to three sites were used, two QTL that use the genes with the highest resistance such as races I, II, and model 001 of H. vastatrix and one RGA (Disease Resistance Gene Analogs) gene. For CBD, two markers were used that separate the Ck-1 gene. Microsatellite markers were used to study genetic diversity. The data from the markers associated with the two QTLs suggest that all coffee trees in the C6T-24 population show resistance to races I, II and model 001 of H. vastatrix, since a dominant allele is present in one of the two locations is sufficient for anyone to be tough. The RGA gene associated with resistance to this pathogen was identified in 117 coffee trees. For resistance to C. kahawae, 46 coffee trees from the C6T-10 population and 59 from the C6T-24 population had the Ck-1 gene. 37 coffees fromthe C6T-10 population and 23 from the C6T-24 population were selected with a gene stacking of four resistance sites to H. vastatrix and C. kahawae. These same coffee trees have been rated as rust-resistant at the field level in 2019. Other morpho-agronomic characteristics were analyzed, allowing the selection of genotypes that use resistance gene stacking and characteristics ofagronomic interest. For an auxiliary selection of genotypes to be crossed to form a new backcross cycle, a study of coffee diversity was carried out, using a UPGMA cluster analysis. Observe the formation of four main groups, allowing us to demonstrate the diversity in the C6T-10 population. Based on this work, it was possible to select coffee trees in the F2 generation, including the removal of resistance alleles to H. vastatrix and C kahawae, as well as the selected genotypes that can be used for monitoring and training new populations for selection. RESUMO O objetivo deste trabalho foi avaliar o comportamento de 54 clones de cafeeiro conilon oriundos do Programa de Melhoramento Genético do Incaper à ferrugem em condições controladas de temperatura e fotoperíodo. Utilizou-se o delineamento inteiramente casualizado com três repetições, sendo cada repetição composta por 16 discos de folha cada, acondicionados em gerbox e inoculados com 104 esporos.mL-1 de H. vastatrix. Os gerbox foram colocados sob ausência de luz e 22ºC por 48 horas e então em fotoperíodo de 12 horas até o término do experimento. Foram avaliados os componentes de resistência: período de incubação, período latente, incidência, porcentagem de discos com esporulação, número de esporos e severidade. Com base nos resultados verificou-se a formação de três grupos de genótipos de café conilon: Resistentes, Moderadamente Resistentes e Suscetíveis. No grupo Resistente foram alocados 19 genótipos, no grupo Moderadamente Resistente foram alocados 19 genótipos e no grupo Suscetível foram alocados 16 genótipos. Assim, com base nos resultados, conclui-se que há variação no nível de resistência dos genótipos de C. canephora à H. vastatrix. Tal informação subsidia os programas de melhoramento na seleção adequada de progênies de C. canephora quanto à resistência à ferrugem.

Conselho Nacional de Desenvolvimento Científico e Tecnológico Para alcançar o atual patamar de importância, a cafeicultura brasileira de um modo geral e a cafeicultura mineira contaram com árduo trabalho técnico e científico ao longo de vários anos. Entre as principais ações científicas para o desenvolvimento da cafeicultura, certamente está na instalação de programas de melhoramento genético, responsáveis pelo desenvolvimento de materiais genéticos mais produtivos, tolerantes à seca, com arquitetura adequada e resistentes às moléstias agrícolas que assolam o café, sobretudo a ferrugem, causada pelo fungo Hemileia vastatrix Berk. et Br., que é a principal doença da cafeicultura mundial. Assim como a ferrugem, a bienalidade de produção, ou seja, a variação de uma alta produção seguida de safra com uma baixa produção, é outro problema que assola cafeicultores cm todo Brasil. A seleção de genótipos com potencial produtivo, resistência a ferrugem, que sofram pouco com o efeito da bienalidade e apresente alta adaptabilidade contudo, não é tarefa fácil, principalmente devido a existência de interação entre genótipos e ambiente, ou seja, os genótipos respondem de forma diferenciada aos diferentes ambientes. Várias metodologias estão disponíveis para se avaliar os genótipos e classificá-los quanto à estabilidade e adaptabilidade, de maneira que possam ser selecionados ou recomendados para diferentes ambientes. Neste contexto, o objetivo deste trabalho foi avaliar o comportamento de 32 genótipos de cafeeiros Arábica, portadores de genes de resistência à ferrugem, bem como estudar a bienalidade, adaptabilidade e estabilidade mesmos, em Viçosa e Patrocínio, duas importantes regiões produtoras de café de Minas Gerais. Foram instalados, em fevereiro de 2006, um ensaio no município de Viçosa, em área experimental da Universidade Federal de Viçosa e outro no Campo Experimental de Patrocínio/Epamig, no município de Patrocínio. Os experimentos foram montados no delineamento experimental de blocos ao acaso, com 32 tratamentos, quatro repetições e parcelas de seis plantas. A produtividade para as colheitas (2008 a 2013) em sacas de café beneficiado por hectare (sc/ha) foi estimada, considerando-se um rendimento médio de 480 litros de café da roça por saca de 60 kg de café beneficiado. Para análise da adaptabilidade e estabilidade foram utilizadas quatro diferentes metodologias estatísticas. A bienalidade foi menor em todos os tratamentos nas condições de Patrocínio — MG. Esse resultado demonstra que a região de Patrocínio detém melhores condições que Viçosa para a regeneração dos ramos plagiotrópicos. O genótipo IPR 103 apresentou a maior média geral de produtividade (31,86 sacas de 60 kg/há) ao longo das seis colheitas nos dois locais de estudo. O material genético IPR 100 demonstrou ser um genótipo muito responsivo em produtividade, para um manejo de alto nível tecnológico, ao passo que o genótipo IPR 103 apresentou adaptabilidade geral para os ambientes em estudo. Outros genótipos como Acauã e Catucaí Amarelo 24/137 apesar da produtividade elevada apresentaram baixa previsibilidade de desempenho. De forma geral os genótipos com fontes de resistência a ferrugem apresentaram bom desempenho e são, portanto, materiais genéticos recomendados para as condições de estudo. In February 2006, an experiment was carried out in the municipality of Viçosa, in an experimental area of the Federal University of Viçosa and another in the Experimental Field Patrocinio / Epamig, in the city of Patrocínio. The experiments were Set up in a randomized complete block design, with 32 treatments, four replications and plots of six plants. Productivity for harvests (2008 to 2013) in coffee sacks benefited per hectare (sc / ha) was estimated, considering an average yield of 480 liters of coffee per acre of 60 kg of coffee benefited. The genetic material IPR 100 proved to be a very responsive genotype in productivity, for a high technological level management, while the IPR 103 genotype presented general adaptability to the environments under study. Other genotypes such as Acauã and Catucaí Amarelo 24/137, despite high yields, presented low predictability of performance. In general, the genotypes with rust resistance sources showed good performance and are, therefore, genetic material recommended for the study conditions.

A ferrugem do cafeeiro, causada pelo fungo Hemileia vastatrix Berk. & Br., é a principal doença em todos os países produtores de café no mundo. No Brasil, a doença causa perdas entre 35% e 50% e, na América Central e Colômbia, entre 20% e 80%. O Brasil possui o maior banco de germoplasma com acessos derivados do Híbrido de Timor (HDT), na Universidade Federal de Viçosa (UFV), o qual vem sendo utilizado e, ao longo dos anos, tem sido valioso para os programas de melhoramento, visando encontrar cultivares com resistência durável à ferrugem. Cultivares com resistência à ferrugem geradas por diferentes institutos de melhoramento genético de café no Brasil foram avaliadas em campo para conhecer seu comportamento em diferentes ambientes. As epidemias registradas na América Central de 2011 a 2013 foram decorrentes de diferentes fatores econômicos, ambientais e, possivelmente, do surgimento de novas raças de H. vastatrix; a suplantação da resistência de cultivares de café lançadas como resistentes à ferrugem no mundo todo também constitui um problema para os programas de melhoramento genético. Diante desses fatos, este estudo teve como objetivos: (i) avaliar fenotipicamente os acessos de HDT; (ii) estudar o comportamento de cultivares lançadas como resistentes pelos diferentes programas de melhoramento genético do Brasil; (III) avaliar a resistência de 12 cultivares às raças II (v5) e XXXIII (v5, 7, 9) de H. vastatrix; (IV) avaliar o teor de clorofila das cultivares resistentes à ferrugem; e (V) caracterizar as raças de H. vastatrix Berk. & Br. presentes em Honduras. A avaliação dos HDTs foi realizada no viveiro de café do Departamento de Fitopatologia da UFV. Para avaliação das cultivares com resistência à ferrugem, foram conduzidos experimentos em campo, em Varginha e Viçosa, no Estado de Minas Gerais. A caracterização de raças de H. vastatrix foi desenvolvida no Instituto Hondurenho do Café (IHCAFE), em Honduras. Vinte e oito cultivares lançadas como resistentes foram estudadas durante oito anos. Os testes visando avaliar a resistência de 12 cultivares às duas raças de H. vastatrix foram conduzidos em câmaras de incubação. O teor de clorofila a, b, total e carotenoides foi determinado nas 28 cultivares. Para caracterização de raças de H. vastatrix, isolados do patógeno foram coletados em lavouras de café em Honduras, nas cultivares Lempira e Catuaí vermelho, em diversas altitudes. A caracterização fisiológica de raças do patógeno foi baseada na ausência (resistente) ou presença (suscetível) de uredósporos nos discos foliares dos clones diferenciadores. Os resultados da avaliação dos acessos de HDT, utilizando a rede de correlações, mostrou que houve correlações significativas diferentes de zero nas características vigor, produção, diâmetro de copa e altura das plantas. Nos testes de cultivares resistentes à ferrugem, verificou-se predominância da resistência quantitativa, além de algumas cultivares terem apresentado tolerância à ferrugem. Cerca de 38,4% das cultivares avaliadas foram resistentes à raça II (v5) e 100% suscetíveis à raça XXXIII (v5, 7, 9) de H. vastatrix. Quanto aos teores de clorofilas e carotenoides, houve grande variação dos valores entre os genótipos, porém não foi observada correlação com a produtividade e a incidência da ferrugem. Diante da rápida suplantação da resistência qualitativa das cultivares de café lançadas, recomenda-se buscar nos genótipos de café características de tolerância à doença, pelo fato de ser duradoura. Em se tratando da caracterização de raças de H. vastatrix, não foi possível enquadrar nenhuma das combinações dos genes de virulências dos isolados em raças patogênicas. Comprovou-se, portanto, a suplantação da resistência da variedade Lempira pelo fungo e foi feita a identificação de 16 patótipos do patógeno. The coffee rust, caused by the fungus Hemileia vastatrix Berk. & Br., is the main disease in all coffee producing countries in the world. In Brazil, the disease causes losses between 35 and 50% and, in Central America and Colombia, losses from 20 to 80%. Brazil has the largest germplasm bank with accesses derived from the Timor Hybrid (HDT) at the Federal University of Viçosa (UFV), which has been used and, over the years, has been valuable for breeding programs, aiming at to find cultivars with durable resistance to rust. Cultivars with resistance to rust, generated by the different institutes of coffee genetic improvement in Brazil, were evaluated in the field to know their behavior in different environments. The epidemics recorded in Central America from 2011-2013 were due to different economic, environmental and possibly the emergence of new races of H. vastatrix; the overriding resistance of coffee cultivars launched as rust resistant worldwide is also a problem for breeding programs. In view of these facts, the work had as objectives: (i) to evaluate phenotypically the accesses of (HDT); (ii) study the behavior of cultivars launched as resistant by the different breeding programs in Brazil; (III) to evaluate the resistance of twelve cultivars to H. vastatrix races II (v 5) and XXXIII (v5, 7, 9); (IV ) to evaluate the chlorophyll content of rust-resistant cultivars; (V) characterize the H. vastatrix in Honduras. The evaluation of the (HDT) was carried out in the coffee nursery of the Department of Plant Pathology of the UFV. To evaluate the cultivars with resistance to rust were conducted field experiments, in Varginha and Viçosa, Minas Gerais state cities. The H. vastatrix isolates characterization was developed at the Honduran Coffee Institute (IHCAFE) in Honduras. Twenty-eight cultivars released as resistant were studied for eight years. The tests to evaluate the resistance of 12 cultivars of the two races of H. vastatrix were conducted in incubation chambers. The content of chlorophyll a, b, total and carotenoids was performed in 28 cultivars. For characterization of H. vastatrix races isolated from the pathogen were collected in coffee fields in Honduras, in the cultivars Lempira and Red Catuaí, at different elevations. The physiological characterization the pathogen isolates was based on the absence (resistant) or presence (susceptible) of uredospores in the leaf disks of the differentiating clones. The data access coffee genotypes evaluation (HDT), using a network of correlations, were correlated to different levels in the following variables: vigor, yield, crown diameter and plant height. In the tests of rust resistant cultivars, they showed that there was predominance of quantitative resistance, besides some cultivars presented tolerance to rust. About 38.4% of the evaluated cultivars were resistant to race II (v 5) and, 100% susceptible to race XXXIII (v5, 7, 9), of H. vastatrix. In the chlorophyll and carotenoid contents, there was a great variation of the values among the genotypes, however, no correlation was observed with productivity and the rust incidence. In view of the rapid substitutability of the qualitative resistance of the coffee cultivars, it is recommended that coffee genotypes should be considered as tolerant to the disease, because it is durable. In the characterization of H. vastatrix races it was not possible to fit any of the combinations of the virulence genes of the isolates into pathogenic races. It was verified, therefore, the substitution of resistance of the variety Lempira by the fungus and the identification of 16 pathotypes of the pathogen.

Com este trabalho, objetivou-se verificar a influência da aplicação de ciproconazol+tiametoxam, via solo, no crescimento de mudas clonais de cafeeiro conilon que compõem cultivar clonal 'Vitória Incaper 8142'. O experimento foi conduzido em vasos e sob condições de ambiente controlado. Utilizou-se delineamento inteiramente casualizado, com distribuição fatorial de 13x2, com quatro repetições, sendo os fatores: 13 clones que compõem a cultivar clonal 'Vitória Incaper 8142'; tratados e não tratados com ciproconazol+tiametoxam. O objetivo foi avaliar o efeito de fungicidas no controle da ferrugem, no crescimento vegetativo e reprodutivo do café conilon. O delineamento experimental utilizado foi em blocos casualizados com seis tratamentos e quatro repetições (duas aplicações de piraclostrobina e epoxiconazol + boscalida e duas aplicações de piraclostrobina e epoxiconazol - T1; três aplicações de piraclostrobina e epoxiconazol - T2; três de piraclostrobina, epoxiconazol e fluxapiroxade - T3; três de epoxiconazol - T4 e três de piraclostrobina - T5 e, sem aplicação de fungicida - T6). Foram realizadas avaliações do progresso da ferrugem do cafeeiro; do crescimento de ramos, número de nós por ramo e comprimento dos internódios; índice relativo de clorofila; número médio de rosetas com frutos, de frutos por ramo e de frutos por roseta; uniformidade de maturação dos frutos; porcentagem de frutos chochos, peso dos frutos e produtividade. Com base nos resultados obtidos no trabalho, concluiu-se que a aplicação com epoxiconazol proporcionou menor crescimento acumulado e número de nós por ramo plagiotrópico e menor número de rosetas com frutos. Os tratamentos com piraclostrobina influenciaram positivamente os índices relativo de clorofila e proporcionaram valores inferiores da relação clorofila a/b. Não houve efeito dos produtos na cultura do café conilon nas demais variáveis monitoradas.Palavras-chave: Coffea canephora, estrobilurinas, Hemileia vastatrix, respostas fisiológicas, triazóis. EFFECT OF FUNGICIDES ON CONILON COFFEE ABSTRACT:The aim of it was evaluate the effect of fungicides in control of rust, vegetative growth and reproductive conilon coffee. The experiment was design in radom blocks with six treatments and four replicates (two applications of pyraclostrobin and epoxiconazole + boscalid and two applications of pyraclostrobin and epoxiconazole - T1; three applications of pyraclostrobin and epoxiconazole – T2; three of pyraclostrobin, epoxiconazole and fluxapyroxad - T3; three of epoxiconazole - T4 and three of pyraclostrobin - T5 and, without application of fungicide - T6) and four replicates. Evaluations of rust progress were carried out; growth of branches, number of nodes per branch and length of internodes; relative index of chlorophyll; average number of rosettes with fruits, fruits per branch and fruits per rosette and productivity. Based on the results obtained in the work, it was concluded that the application with epoxiconazole provided lower cumulative growth and number of nodes per plagiotropic branch, and lower number of rosettes with fruits. The treatments with pyraclostrobin positively influenced the relative chlorophyll indices and provided lower values of the chlorophyll a/b ratio. There was no effect of the products in conilon coffee culture in the other monitored variables.Keywords: Coffea canephora, strobilurins, Hemileia vastatrix, physiological responses, triazoles.

A ferrugem do cafeeiro causada pelo fungo Hemileia vastatrix vem causando grandes perdas na cafeicultura ao longo dos anos. O controle químico envolvendo fungicidas sistêmicos e protetores tem sido uma ferramenta importante, visando controle eficiente e evitando o surgimento de mutantes resistentes, na população do fungo aos fungicidas sistêmicos. Dentro os produtos mais utilizados, destacam-se os fungicidas cúpricos, preventivamente por sua eficiência, na inibição da germinação dos esporos; quando a incidência atinge 5 %, nos anos de alta carga de frutos pendentes nas plantas, torna-se necessário a utilização de fungicidas sistêmicos. A ocorrência de chuva tem efeito direto no sucesso do controle, pois pode alterar a dinâmica dos fungicidas na superfície foliar. Neste trabalho foi avaliado os efeitos e ocorrências de chuvas após aplicação de fungicidas sistêmicos e protetores, em folhas de cafeeiro, no controle da ferrugem. Foram realizados dois experimentos em casa de vegetação, onde as folhas destacadas de Coffea arabica var. Caturra foram submetidas a uma chuva de 30 mm. No primeiro teste avaliou-se fungicidas sistêmicos (epoxiconazole + piraclostrobina), hidróxido de cobre, sulfato de zinco e ácido bórico, misturados e sozinhos, utilizando a dose comercial. As folhas foram atomizadas e em seguida submetidas a chuva. No primeiro experimento aguardou-se 30 minutos para a aplicação da chuva. Os melhores resultados foram aqueles que continham (epoxiconazole + piraclostrobina) + hidróxido de cobre, onde a severidade foi praticamente zero Independiente do tempo de chuva após pulverização dos tratamentos. Concluiu-se que a aplicação destes fungicidas, mesmo após serem submetidos a chuva, teve controle eficiente da doença. No segundo experimento foi avaliado o oxicloreto de cobre adicionado adjuvantes. Com ocorrência de chuva de 30 mm após os tempos zero, 30, 120 e 480 minutos, depois da pulverização. Os tratamentos mais eficientes foram onde houve a mistura de oxicloreto de cobre 3,0 kg há -1 + OM a 0,75%, demostrando maior retenção na superfície da folha e maior controle da doença. Os menos eficientes foram aqueles que tinham oxicloreto de cobre + PAFE a 0,10% e 0,20%. Conclui-se que o uso de adjuvantes, pode favorecer a tenacidade e eficiência dos produtos, tendo em consideração a formulação do fungicida. Coffee rust, caused by the fungus Hemileia vastatrix, has been causing significant yield losses of coffee crop over the years. Chemical control involving systemic curative and protective fungicides have been considered important tools, aiming the effective control and avoiding the emergence of resistant mutants in the fungal population to systemic fungicides. Among the most commonly used products, copper fungicides are prominent due to their preventative and efficient mode of action in inhibiting spore germination; when the incidence reaches 5% in the years of high pending fruits’ load, the use of systemic fungicides becomes necessary. In addition, rain has a direct impact on the management success due to the removal of deposits of products on the leaf surface. Thus, the objective of this study was to evaluate the retention time of systemic and protective fungicide residues on coffee leaves submitted to rain, aiming the control of rust. Two experiments were carried out, in which leaves of Coffea arabica var. Caturra were subjected to a 30-mm rainfall treatment. The first assay employed systemic fungicides (epoxiconazole + pyraclostrobin), copper hydroxide, zinc sulfate, and boric acid, mixed and alone, using the commercial dose. The leaves were atomized and then exposed to precipitation regime. In the first experiment, we allowed 30 minutes before the rainfall application. The best results were those containing (epoxiconazole + pyraclostrobin) + copper hydroxide, in which the severity was practically zero. Therefore, the application of these fungicides, even after being submitted to rain, showed effective control over the disease. Copper oxychloride added to the OM and PAFE was used in the second trial. 30-mm rainfall was applied after the times zero, 30, 120, and 480 minutes, respectively after spraying. The most efficient treatments were the mixture of copper oxychloride + OM at 0.75 %, showing greater retention at the leaf surface and greater control of the disease. The least efficient were those having copper oxychloride + 0.10% and 0.20% PAFE. To conclude, the use of mineral oil adjuvant may favor the tenacity and efficiency of the product, taking into account the formulation of the fungicide.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior O Brasil é o país com a maior produção e exportação de café e as espécies mais cultivadas mundialmente são o Coffea arabica e o Coffea canephora. A doença do cafeeiro de maior relevância no cenário mundial é a ferrugem alaranjada, causada pelo fungo biotrófico Hemileia vastatrix. O principal sintoma é o aparecimento de manchas amareladas na parte abaxial das folhas e, em condições climáticas favoráveis, as perdas na lavoura podem chegar a 50% da produção. O cultivar Híbrido de Timor CIFC 832/2 tem sido uma importante fonte de genes de resistência para os programas de melhoramento. Entretanto, o alto potencial adaptativo do fungo e a própria pressão seletiva causada pela utilização de cultivares resistentes vem culminando no surgimento de diversas raças fisiológicas do fungo capazes de suplantar a resistência obtida através dos métodos clássicos de melhoramento. Uma nova abordagem no estudo da relação parasito-hospedeiro é a integração de dados de genômica e outras ômicas para a construção de um modelo metabólico. Através da análise de balanço do fluxo (FBA) é possível simular o papel de cada gene na célula auxiliando na compreensão da interação entre o cafeeiro e o fungo. Com isto, o presente trabalho propôs a construção de um modelo metabólico baseado no genoma seguido da análise de FBA para cultivares resistente e susceptível após a infecção por H. vastatrix. Os dados de transcriptomica foram integrados ao modelo original gerando modelos contexto- específicos. Essas análises indicaram um acúmulo de glicerol-3-fosfato (G3P) no Hibrido de Timor CIFC 832/2 sugerindo que o metabolismo primário contribui para a resistência do cafeeiro, uma vez que a participação deste metabólito na resposta imune adquirida de plantas já foi demonstrada. The major coffee disease in the world is the Coffee Leaf Rust, caused by the biotrophic fungus Hemileia vastatrix. The main symptom is the appearance of yellow spots on the abaxial part of the leaves and, under favorable climatic conditions; crop losses can reach 50% of production. The Híbrido de Timor CIFC 832/2 has been an important source of resistance genes for breeding programs, however, the high adaptive potential of the fungus and the selective pressure caused by the use of resistant cultivars has culminated in the appearance of several rust races capable of overcoming the resistance obtained through classical breeding methods. A new approach in the study of the host-parasite relationship is the integration of genomic and other omic data for the construction of a metabolic model. Through flux balance analysis (FBA) it is possible to simulate the role of each gene in the cell, contributing to understand the interaction between coffee and fungus. With this, the present work proposed the construction of a genome-scale metabolic model followed by FBA analysis for resistant and susceptible cultivars after H. vastatrix infection. Transcriptomic data were integrated into the original model generating context- specific models. These analyzes indicated an accumulation of glycerol-3- phosphate (G3P) in the Híbrido de Timor CIFC 832/2 suggesting that the primary metabolism contributes to coffee resistance, since the participation of this metabolite in the systemic acquired resistance of plants has already been demonstrated.

O fungo está amplamente distribuído nas regiões produtoras de café, causando severos danos econômicos. Novas raças fisiológicas de H. vastatrix tem surgido, com frequência em todo mundo, infectando algumas cultivares de café lançadas comercialmente como resistentes à ferrugem. Dessa forma, a obtenção de cafeeiros resistentes tem sido um desafio devido ao alto potencial adaptativo do fungo e tem se tornado cada vez mais difícil a caracterização dos patótipos por meio da coleção de cafeeiros diferenciadores de raça. Além disso, as informações acerca da interação de cafeeiros com as raças existentes são escassas, embora o processo infeccioso de H. vastatrix seja bem estudado. Estudos genômicos estão ajudando no entendimento dos mecanismos moleculares envolvidos no processo de interação entre planta e patógeno e no desenvolvimento de técnicas moleculares para a identificação de isolados. Nosso objetivo foi caracterizar as raças de H. vastatrix encontradas nas principais regiões produtoras de café do Brasil, e estudar as proteínas candidatas a efetoras preditas para H. vastatrix que possam contribuir para patogenicidade nas diferentes raças do fungo. Foram utilizados 56 isolados monopustular coletados de diferentes espécies de café, Coffea canephora, C. arabica e Híbrido de Timor (C. canephora x C. arabica) e multiplicados em cafeeiro suscetível C. arabica variedade Caturra. Cada isolado foi inoculado em cafeeiros que compõe a série diferenciadora de raças, com três repetições. O DNA dos 56 isolados monopustulares foram extraídos para os estudos genômico. Foram utilizadas 47 combinações de primer para amplificar genes de candidatos a efetores. Sete genes foram amplificados, sequenciados e tiveram sua expressão validada em cafeeiros resistente e suscetível. As sequências obtidas foram alinhadas para os 46 isolados, usando o programa DNA Baser. Estudo de domínio conservado foram realizados no programa Pfam e a categorização das proteínas no Blast2GO. O alinhamento das sequências de nucleotídeos, proteínas e agrupamento dos isolados foi feito no programa Clustalw e as diferença de nucleotídeo foi analisada no Weblogo. Os dados obtidos foram usados para o estudo de diversidade. Por meio da inoculação na série diferenciada, foram identificadas sete raças, sendo cinco já descritas no Brasil e duas pela primeira vez (raça XXIX e XXX). Não foi possível caracterizar 15 combinações dos genes de virulência, sendo assim consideradas novas raças. A análise da coleção de isolados permitiu inferir a presença de pelo menos seis novos genes na série diferenciadora, o que possibilitou diferenciar dois isolados descritos como raças III. Na análise genômica, os 47 genes candidatos a efetores estudados se mostraram conservados em todos os isolados e estão ligados a 15 processos biológicos do desenvolvimento do fungo. Foram identificados 14 domínios nas proteínas provenientes desses genes. Para os sete genes sequenciados nos diferentes isolados, observou-se diferença na sequência dos nucleotídeos e presença de indivíduos heterozigoto e homozigoto. Com análise de diversidade foi possível discriminar os indivíduos, sendo o isolado Hv-09 o mais divergente. As informações geradas nesta pesquisa poderão ampliar o entendimento da interação entre H. vastatrix e o cafeeiro. O conhecimento dos genes que estão envolvidos no processo infeccioso auxiliará o entendimento dos mecanismos moleculares que levam à suplantação da resistência por novas raças do fungo. Coffee rust, caused by the fungus Hemileia vastatrix Berkeley & Broome, is considered the main disease of the crop. The fungus is widely distributed in the coffee producing regions, causing severe economic damage. New physiological races of H. vastatrix have arisen, often all over the world, infecting some commercially released coffee cultivars as resistant to rust. Thus, obtaining resistant coffee trees has been a challenge due to the high adaptive potential of the fungus and it has become increasingly difficult to characterize the pathotypes through the collection of coffee races. In addition, information on the interaction of coffee trees with the existing races is scarce, although the infectious process of H. vastatrix is well studied. Genomic studies are helping to understand the molecular mechanisms involved in the process of interaction between plant and pathogen and in the development of molecular techniques for the identification of isolates. Our objective was to characterize the H. vastatrix race found in the main coffee producing regions of Brazil and to study the effector candidate proteins predicted for H. vastatrix that may contribute to pathogenicity in the different fungus races. Fifty six monopustular isolates collected from different coffee species, Coffea canephora, C. arabica and Hybrid of Timor (C. canephora x C. arabica) were used and multiplied in susceptible coffee C. arabica variety Caturra. Each isolate was inoculated in coffee plants that make up the differentiating series of races, with three replicates. The DNA of the 56 monopustular isolates were extracted for genomic studies. We used 47 primer combinations to amplify effector candidate genes. Seven genes were amplified, sequenced and had their validated expression in resistant and susceptible coffee trees. The sequences obtained were aligned for the 46 isolates using the DNA Baser program. A conserved domain study was performed on the Pfam program and the protein categorization in Blast2GO. The alignment of the nucleotide sequences, proteins and clustering of the isolates was done in the Clustalw program and the nucleotide differences were analyzed in Weblogo. The data obtained were used for the study of diversity. Through inoculation in the differentiated series, seven races were identified, five of which were already described in Brazil and two for the first time (races XXIX and XXX). It was not possible to characterize 15 combinations of the virulence genes, thus being considered new races. The analysis of the collection of isolates allowed to infer the presence of at least six new genes in the differentiator series, which made it possible to differentiate two isolates described as breeds III. In genomic analysis, the 47 candidate effector genes studied have been conserved in all isolates and are linked to 15 biological processes of fungus development. 14 domains were identified in proteins from these genes. For the seven genes sequenced in the different isolates, we observed a difference in nucleotide sequence and presence of heterozygous and homozygous individuals. With diversity analysis it was possible to discriminate individuals, being the Hv-09 isolate the most divergent. The information generated in this research may broaden the understanding of the interaction between H. vastatrix and the coffee tree. The knowledge of the genes that are involved in the infectious process will help the understanding of the molecular mechanisms that lead to the supplanting of resistance by new races of the fungus.

Para aumentar a eficiência de seleção de cafeeiros superiores no melhoramento genético deve-se realizar a caracterização morfoagronômica dos materiais genéticos a serem selecionados e a estimação dos valores genéticos dos indivíduos. Além disso, aliar essas metodologias à seleção assistida por marcadores moleculares (SAM) é uma forma de minimizar as limitações da seleção puramente fenotípica. Essa estratégia reduz o tempo e o custo para seleção e, para a resistência a doenças, permite a busca de resistência duradoura e de amplo espectro por meio da piramidação de genes. Portanto, objetivou-se identificar cafeeiros para avanço de geração no programa de melhoramento por meio de seleção baseada na metodologia de modelos mistos (REML/BLUP) aliada à seleção assistida por marcadores. Para isso, sete características morfoagronômicas foram utilizadas para avaliar a diversidade genética, estimar os parâmetros genéticos e quantificar os ganhos com a seleção de famílias em melhoramento. Cafeeiros resistentes a doenças em campo foram analisados com marcadores moleculares para identificar indivíduos portadores de genes que conferem resistência a diferentes raças de H. vastatrix e resistência a C. kahawae, visando a piramidação de genes e obtenção de resistência múltipla. As populações analisadas neste trabalho fazem parte do Programa de Melhoramento Genético do Cafeeiro desenvolvido pela Empresa de Pesquisa Agropecuária de Minas Gerais (EPAMIG), em parceria com a Universidade Federal de Viçosa (UFV), a Universidade Federal de Lavras (UFLA) e a Empresa Brasileira de Pesquisa Agropecuária (Embrapa Café). As plantas que deram origem a essas populações, por serem potenciais fontes de resistência à H. vastatrix, foram introduzidas pela UFV, oriundas do Centro de Investigação das Ferrugens do Cafeeiro (CIFC), localizado em Oeiras/Portugal. Com base nas características morfoagronômicas e análise de agrupamento, observou-se alto grau de parentesco entre as famílias. O índice de seleção permitiu selecionar nove famílias, com ganhos de seleção superior a 26% em relação à população original. Com base em marcadores moleculares, das nove famílias selecionadas, oito apresentaram em suas progênies o gene S H 3 de resistência a H. vastatrix e a outra o gene Ck-1 de resistência a C. kahawae. A SAM permitiu, ainda, identificar uma família que embora não tenham sido selecionada nas análises morfoagronômicas, é portadora do gene para resistência às raças I, II e patótipo 001 de H. vastatrix. Os cafeeiros selecionados pelas características morfoagronômicas e SAM foram superiores às testemunhas e apresentaram genes em indivíduos diferentes, que conferem resistência às principais doenças do cafeeiro, ferrugem e C. kahawae. Esses cafeeiros podem ser utilizados como fonte de resistência para novos cruzamentos. Além disso, buscou-se avaliar novas fontes de resistência em outra população, no qual a SAM permitiu identificar 31 famílias portadoras do gene S H 3, duas com o gene Ck-1, seis para o QTL ligado ao grupo de ligação 2, e duas para o QTL ligado ao grupo de ligação 5. Esses diferentes genes foram encontrados isolados em alguns cafeeiros e também piramidados em outros, com diferentes combinações. Os cafeeiros selecionados poderão ser utilizados para avanço de geração no melhoramento e como fontes para piramidação de genes nos programas de melhoramento com vista a obter cultivar com resistência múltipla e durável. Coffea arabica L. is the most economically important species and the development of resistant cultivars to the main coffee plants diseases, such as rust (Hemileia vastatrix) and fruit anthracnose (Colletotrichum kahawae), is important to obtain productive stability and production costs decrease. In order to increase the selection efficiency for superior coffee plants in genetic improvement, the morphological analysis of the genetic material to be selected and the estimation of the individuals’ genetic values should be carried out. In addition, combining these methodologies with molecular marker-assisted selection (MAS) is a way of minimizing the purely phenotypic selection limitations. This strategy reduces time and selection cost and, for disease resistance, allows the search for long- lasting and wide spectrum resistance through gene pyramidation. Therefore, this study’s objective was to identify coffee plants for the breeding program advance generation, based on mixed models selection (REML/BLUP), combined with markers assisted selection. For this, seven morphoagronomic characteristics were used to evaluate genetic diversity, to estimate the genetic parameters and to quantify the gains with families’ selection in breeding. Field-resistant coffee plants were analyzed with molecular markers to identify individuals carrying genes that confer resistance to different strains of H. vastatrix and resistance to C. kahawae, aiming at genes pyramiding of and obtaining multiple resistance. The populations analyzed in this work are part of the Coffee Breeding Program developed by the Empresa de Pesquisa Agropecuária de Minas Gerais (EPAMIG), in partnership with the Universidade Federal de Viçosa (UFV), the Universidade Federal de Lavras (UFLA) and the Empresa Brasileira de Pesquisa Agropecuária (Embrapa Café). The plants that gave origin to these populations, being potential resistance sources to H. vastatrix, were introduced by the UFV, from the Centro de Investigação das Ferrugens do Cafeeiro (CIFC), located in Oeiras/Portugal. Based on the morphoagronomic characteristics and cluster analysis, a high kinship degree between families was observed. The selection index allowed nine families to be selected, with selection gains higher than 26% in relation to the original population. Based on molecular markers, eight out of the nine families selected had H. vastatrix resistance S H 3 gene and the Ck-1 gene resistance to C. kahawae in their progenies. The MAS also allowed to identify a family that although has not been selected by the morphoagronomic analyzes, it is a carrier of the resistance gene to the races I, II and to H. vastatrix 001 pathotype. The coffee plants selected by MAS and morphological characteristics were superior to the controls and presented genes in different individuals, which confer resistance to the main coffee plants diseases, rust and C. kahawae. These coffee plants can be used as a resistance source for new crossings. In addition, new resistance sources in another population were evaluated, in which MAS allowed to identify 31 families carrying S H 3 gene, two with Ck-1 gene, six for the QTL linked to the linkage group 2, and two for the QTL linked to the linking group 5. These different genes were found isolated in some coffee plants and also pyramidal in others, under different combinations. The selected coffee plants may be used for generation advance and as gene pyramiding sources in breeding programs aiming to obtain a durable and multiple resistant crop.

Rust is the most devastating disease of coffee. The rust intensity is affected by water supply and nutritional balance of plants. Thus the aim of this study was to evaluate the incidence of rust in coffee plantations with different management of irrigation and phosphorus fertilization. The study was carried out in Lavras-MG, with coffee cultivar MGS Crossing. Evaluations of the disease were carried out from March 2012 to November 2014. The experimental design was a randomized complete block design with twelve treatments, 3 fertilization managements with 4 drip irrigation management, in a fatorial scheme of variance analysis, with three replicates, totaling 36 plots. Considerando que o CO2 é o gás de efeito estufa que tem maior importância devido ao maior volume de emissões, os efeitos do enriquecimento do ar com CO₂[duas condições: concentração ambiente (nas faixas 360-450ppm) e concentração enriquecida (entre 490 ppm e 620 ppm)] foram avaliados sobre o desenvolvimento de plantas de café e sobre a ferrugem do cafeeiro e bicho-mineiro na cultivar “Catuaí Vermelho IAC 144” em experimento do tipo FACE (“Free Air Carbon Dioxide Enrichment”). O experimento foi composto por 12 parcelas octogonais com 10 metros de diâmetro, das quais seis foram enriquecidas com CO2 e seis permaneceram na concentração ambiente. As avaliações foram realizadas nos anos de 2013, 2014 e 2015. Para o desenvolvimento da cultura foram avaliadas a altura das plantas, o diâmetro do colo das plantas e o número de folhas nos ramos. Para os problemas fitossanitários foram avaliados a ferrugem do cafeeiro e o bicho-mineiro, ambos por meio da incidência. Em condições de enriquecimento do ar com CO2 a altura das plantas e o diâmetro do caule foram maiores, nos três anos avaliados. Não houve diferença estatística para o número de folhas em todo o período avaliado. A incidência de bicho-mineiro foi maior em condições ambientes, enquanto a incidência da ferrugem do cafeeiro foi semelhante em ambas as condições. In recent decades the planet's climate has been gradually changing as a result of climate change, mainly due to the intensification of anthropogenic activities, such as the burning of fossil fuels and changes in land use, which are responsible for changes in various components of the environment, such as carbon dioxide (CO2), methane, nitrous oxide and other greenhouse gases. Considering that CO2 is the most important greenhouse gas due to higher emissions, the effects of CO2 enrichment [two conditions: ambient concentration (between 360ppm and 490ppm) and enriched concentration (between 490ppm and 620ppm)] were evaluated on the development of coffee plants and on coffee leaf rust and leaf miner in the "Catuaí Vermelho IAC 144" cultivar in a Free Air Carbon Dioxide Enrichment (FACE) experiment. The experiment was composed of 12 octagonal plots with 10 meters in diameter, of which six were enriched with CO2 and six remained in the ambient concentration. The evaluations were carried out in the years of 2013, 2014 and 2015. For the development of the crop were evaluated height of plants, stem diameter and number of leaves in the branches. For the phytosanitary problems, was evaluated the incidence of coffee leaf rust and leafminer incidence. In CO2 enrichment conditions, plant height and stem diameter were higher in the three years evaluated. There was no statistical difference for the number of leaves throughout the evaluated period. The incidence of leaf miner was higher in environmental conditions, while the incidence of coffee leaf rust was similar in both conditions.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior O cafeeiro é uma das culturas mais importantes no mundo, porém, é atacada por diversas doenças, sendo a ferrugem, causada pelo fungo Hemileia vastatrix, considerada a principal doença em todas as regiões produtivas. O estudo do transcriptoma do cafeeiro durante a interação com H. vastatrix pode auxiliar no controle dessa doença pelo entendimento de quais mecanismos de defesa a planta esta ativando. Em um patossistema, entender a expressão gênica é essencial para a identificação dos genes relacionados com os mecanismos de defesa e resistência da planta, que são ativados pelos genes de patogenicidade do agente causal. Esses genes identificados podem ser utilizados para obtenção de cultivares resistentes. Para estudar o transcriptoma de um organismo, o método que tem sido utilizado é o sequenciamento de mRNA, denominado RNA-Seq. Esta abordagem permite monitorar a expressão de genes da planta e do patógeno em diferentes etapas ao longo do processo infeccioso. Desta forma, o objetivo deste trabalho foi estudar o transcriptoma do cafeeiro durante a interação com H. vastatrix, fungo causador da ferrugem, a fim de identificar os genes que são ativados ou reprimidos em resposta à infecção. Folhas dos cafeeiros C. arabica cv. Caturra CIFC 19/1 (suscetível) e Híbrido de Timor CIFC 832/1 (resistente) foram inoculadas com urediósporos da raça XXXIII de H. vastatrix e coletadas em diferentes tempos após a inoculação (12, 24, 96 horas e 17 dias). Como controle, foram utilizadas folhas dos dois genótipos de café inoculados com água. Após a extração do RNA das amostras, foram obtidas as bibliotecas de cDNA, que foram sequenciadas usando a plataforma Illumina Miseq. As análises dos dados foram realizadas utilizando ferramentas de bioinformática e consistiram em: i) avaliação da qualidade das sequências com o programa FastQC; ii) limpeza dos dados e sobreposição de reads de acordo com os critérios de qualidade, usando Pear e Clean Solexa; iii) mapeamento dos transcritos com o genoma de referência de Coffea canephora e montagem de novo como estratégia complementar; iv) analise de expressão diferencial de genes; v) anotação; e vi) validação de genes candidatos por PCR em tempo real. Um total de 43.159 contigs foram obtidos após o mapeamento contra C. canephora e a montagem de novo. Os resultados sugerem que durante a infecção inicial (12 e 24 hai), o Híbrido de Timor (HdT) foi mais responsivo ao ataque de H. vastatrix em relação ao Caturra por apresentar maior número de genes up-regulated. Foram selecionados treze genes (up- regulated) exclusivos do HdT, para avaliar o padrão de expressão com o uso de qPCR. As estratégias utilizadas para a montagem do transcriptoma foram eficientes e permitiram a obtenção de um banco de dados com qualidade, o qual poderá ser utilizado para mineração de genes expressos no patossistema C. arabica-H. vastatrix durante a infecção. Coffee is one of the most important crops in the world; however, several diseases, among which coffee rust, caused by the fungus Hemileia vastatrix, is considered the main disease in all producing regions. The study of coffee transcriptome during the interaction with H. vastatrix can help in the control of this disease as it reveals the mechanism of activation of genes involved in defense. In a pathosystem, understanding gene expression is essential for the identification of genes related to plant defense and resistance mechanisms, which are activated by the presence of the pathogen as well as the pathogenicity genes of the microorganism. To study the transcriptome of an organism, the method that has been used is the sequencing of next generation sequencing of mRNA, called RNA-Seq. This approach allows monitoring the gene expression of an organism at different stages throughout the infection process. Thus, the objective of this work was to study the coffee transcriptome during interaction with Hemileia vastatrix, rust-causing fungus, in order to identify genes that are activated or repressed in response to infection. Leaves of two Arabica coffee cultivars, Caturra CIFC 19/1 (susceptible) and Híbrido de Timor CIFC 832/1 (resistant), were inoculated with H. vastatrix race XXXIII and collected at different times after inoculation (12, 24, 96 hours and 17 days). As a control, leaves of the two coffee cultivars were inoculated with water. After extraction of the RNA from the samples, cDNA libraries were constructed and sequenced using the Illumina Miseq platform. Data analyzes were performed using bioinformatics tools and consisted of: i) evaluation of sequence quality with FastQC program; ii) data cleaning and overlapping of reads according to the quality criteria, using Pear and Clean Solexa; iii) mapping of transcripts with the reference genome of Coffea canephora and de novo assembly as a complementary strategy; iv) analysis of differential gene expression; v) annotation; and vi) validation of candidate genes by real-time PCR. A total of 43,159 contigs were obtained after mapping against C. canephora and de novo assembly. The results suggest that during the initial infection (12 and 24 hai), Híbrido de Timor (HdT) was more responsive to H. vastatrix attack than Caturra because it had a higher number of up-regulated genes. Thirteen (up-regulated) genes unique to HdT were selected to evaluate the expression pattern by qPCR. The strategies used to assemble the transcriptome were efficient and allowed to obtain a high quality database, which will be used for mining of genes expressed in the C. arabica-H. vastatrix interaction.

Selecting superior genotypes is facilitated by marker-assisted selection (MAS), which is particularly suitable for transferring disease resistance alleles because it nullifies environmental effects and allows selection of resistant individuals in the absence of the pathogen or race, enabling preventive breeding. Molecular markers linked to two major genes (SH3 and SH?), conferring resistance to coffee rust, and those linked to the Ck-1 gene, conferring resistance to coffee berry disease (CBD), have previously been identified. These markers were validated and used in a progeny of crosses between Indian selections with Coffea arabica cultivars. Eleven resistant individuals homozygous for SH3 were identified by MAS. Of these, seven carry SH? from Híbrido de Timor and the gene introduced from Coffea liberica (SH3). SH? was characterized as derived from Coffea canephora. Thus, it was possible to identify C. arabica genotypes carrying important genes for rust resistance introgressed from other coffee species. MAS also allowed identification of sources of CBD resistance for use in preventive breeding for resistance to this serious disease. Using two validated molecular markers, two coffee plants carrying Ck-1 were identified: the UFV 328-60 genotype (F2) was resistant and homozygous based on both molecular markers but exhibited no markers related to SH3 and SH?, and the UFV 317- 12 genotype (F1) was resistant and homozygous but resistant and heterozygous based on CBD-Sat207 and CBD-Sat235, respectively. Along with possessing Ck- 1, the latter carries SH?. Overall, plants carrying different genes for resistance to rust and CBD were identified. These plants are important sources for gene pyramiding in breeding programs aimed at multiple and durable resistance.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior Durante a interação com as plantas hospedeiras, os fungos causadores de ferrugens secretam e translocam diversas proteínas efetoras para o citoplasma das células vegetais, onde atuam suprimindo as respostas de defesa e promovendo o parasitismo. Em estudos prévios foram identificados genes candidatos a efetores em Hemileia vastatrix (HvECs) que codificam proteínas secretadas com funções desconhecidas. Visando entender o papel dessas proteínas secretadas na patogênese da ferrugem do cafeeiro, o presente trabalho teve como objetivo o estudo funcional de 54 candidatos a efetores do isolado monopostular de H. vastatrix HV-01 (raça II), avaliando-se a sua capacidade de suprimir a imunidade ativada por padrões moleculares conservados (PAMPs) da bactéria não patogênica Pseudomonas fluorescens em Nicotiana benthamiana. Essa resposta de imunidade, denominada PTI (PAMP-Triggered Imunnity), é a primeira linha de defesa induzida que as plantas utilizam para resistir ao ataque de patógenos. As sequências dos 54 genes que codificam as potenciais proteínas efetoras foram clonadas individualmente no vetor de expressão pEDV6 (Effector Delivery Vector) e os plasmídeos recombinantes foram transferidos para P. fluorescens EtHAn (Effector to Host Analyser) (Pf EtHan), que possui um sistema de secreção tipo III (SST3) capaz de translocar as proteínas codificadas pelos genes candidatos para o citoplasma de N. benthamiana. Desesseis proteínas efetoras apresentaram capacidade de suprimir PTI com alta reprodutibilidade em diferentes experimentos de co-infiltração de Pf EtHAn com a bactéria patogênica P. syringae pv. tomato DC3000 em N. benthamiana. A supressão de PTI por um gene candidato (HvEC_064) foi confirmada analisando-se o crescimento populacional de Pst DC3000 e morte celular nos tecidos infiltrados onde ocorreu, ou não, a supressão de PTI e ativação de imunidade desencadeada pelo reconhecimento de efetores dessa bactéria. Pode-se postular, com base nesses estudos, que os efetores de H. vastatrix com atividade supressora de PTI em N. benthamiana são prováveis efetores citoplasmáticos. During the interaction with host plants, rust fungi secrete and translocate several effector proteins into the cytoplasm of plant cells to suppress defence responses and promote parasitism. Previous studies have identified genes in Hemileia vastatrix that encode effectors with unknown functions. In order to understand the role of these effectors in the pathogenesis of coffee rust, this study aimed the functional analysis of 54 candidate effectors from the monopustular isolate of Hemileia vastatrix HV 01 (race II), by evaluating their ability to suppress Pathogen Associated Molecular Patterns (PAMPs) responses triggered by the nonpathogenic bacteria Pseudomonas fluorescens in Nicotiana benthamiana. This immune response, known as PTI (PAMP-Triggered Immunity), is the first line of induced defenses used by plants to resist pathogen attack. Gene sequences encoding the 54 putative effectors were individually cloned into pEDV6 (Effector Delivery Vector) vector. The recombinant plasmids were transferred to P. fluorescens EtHAn (Effector to Host Analyser) (Pf EtHan), which has a type III secretion system (T3SS) able to translocate the proteins encoded by the candidate genes into the cytoplasm of N. benthamiana. Sixteen candidate effector genes suppressed PTI with high reproducibility in different co-infiltration experiments of Pf EtHAn with the pathogenic bacterium P. syringae pv. tomato DC3000 in N. benthamiana. The suppression of PTI by a candidate gene (HvEC_064) was confirmed by analysing Pst DC3000 population growth and cell death in the infiltrated tissues where PTI was suppressed. The effectors of H. vastatrix with suppressive activity of PTI identified are likely cytoplasmic effectors. These effectors will be further investigated in future studies aimed to identify avirulence genes of this fungus and its targets in coffee plant cells.

Resumo: O objetivo deste trabalho foi avaliar o efeito do aumento da concentração de dióxido de carbono atmosférico sobre o crescimento de plantas e a severidade da ferrugem (causada por Hemileia vastatrix) em cafeeiro (Coffea arabica). Utilizaram-se as cultivares 'Obatã IAC 1669-20' e 'Catuaí Vermelho IAC 144', moderadamente resistente e suscetível à doença, respectivamente. Os tratamentos consistiram de 400 e 508 μmol mol-1 de CO2 atmosférico, em dois tipos de estufas de topo aberto (OTC). As plantas das duas cultivares, cultivadas nas OTCs com adição de CO2, apresentaram aumento de área foliar, e da taxa de crescimento em altura e em diâmetro do caule, em comparação às plantas cultivadas em estufas à concentração ambiente de CO2; porém, as cultivares não diferiram quanto ao teor de cera das folhas. Em 'Catuaí Vermelho IAC 144', a severidade da ferrugem diminuiu com o aumento da concentração de CO2, quanto ao número de lesões, área foliar lesionada, número de lesões esporuladas, área lesionada esporulada, percentagem de área foliar lesionada e área abaixo da curva de progresso da doença; no entanto, em 'Obatã IAC 1669-20', a severidade da doença não variou. Conselho Nacional de Desenvolvimento Científico e Tecnológico A raça XXXIII de Hemileia vastatrix foi recentemente identificada no Brasil, infectando algumas cultivares resistentes à ferrugem do cafeeiro. Embora o processo infeccioso de H. vastatrix seja bem estudado, não existem informações acerca da interação de cafeeiros com esta raça do fungo. Diante disso, um dos objetivos deste trabalho foi avaliar o processo de colonização do fungo e as respostas de defesa da planta em genótipos de cafeeiro resistente (Híbrido de Timor – HDT CIFC 832/1) e suscetível (C. arabica cv. Caturra CIFC 19/1) infectados pela raça XXXIII. As primeiras respostas citológicas induzidas pelo fungo foram observadas particularmente nas células estomáticas de ambos os genótipos, às 17 horas após inoculação (hai), e corresponderam à morte celular e ao acúmulo de compostos fenólicos. No genótipo suscetível, o fungo foi capaz de colonizar os tecidos do hospedeiro e produziu um grande número de haustórios, culminando na esporulação cerca de 20 dias após a inoculação (dai). Ao contrário, no genótipo resistente, o crescimento do fungo foi impedido, com alta frequência no estádio pré-haustorial. Estas observações citológicas indicam que a resistência de HDT CIFC 832/1 à raça XXXIII de H. vastatrix é pré-haustorial, ao contrário da resistência pós-haustorial que é geralmente descrita para interações cafeeiro - H. vastatrix. Com base nesta análise, foram definidos os tempos mais adequados para o estudo de genes expressos ao longo do processo infeccioso. Embora o sequenciamento do transcriptoma tenha sido bastante utilizado em estudos de interação planta-patógeno, uma das maiores dificuldades consiste na separação dos transcritos provenientes da planta e do fungo, principalmente quando não existe genoma de referência. Por esta razão, foi realizado o sequenciamento e a montagem do transcriptoma de esporos hidratados e germinados da raça XXXIII de H. vastatrix. A montagem resultou em 32.882 contigs, a partir dos quais foi gerado um conjunto de 11.989 genes preditos. Plantas de C. arabica cv. Caturra Vermelho (CIFC 19/1) e Híbrido de Timor (CIFC 832/1) foram inoculadas com esporos frescos de H. vastatrix (raça XXXIII) para estabelecer uma interação compatível e incompatível, respectivamente. Como controle, foram utilizadas folhas não inoculadas. Nos tempos de coleta pré-definidos (12, 24, 96 horas e 17 dias após a inoculação), as folhas foram removidas e utilizadas para extração de RNA. O sequenciamento das dez bibliotecas de cDNA foi realizado na plataforma Illumina MiSeq, gerando um total de 206 milhões de reads. Após tratamento dos reads, foi realizado o mapeamento das bibliotecas da interação, utilizando como referência o transcriptoma de H. vastatrix previamente montado. Os dados obtidos mostraram que muitos genes de H. vastatrix são similares aos genes do cafeeiro, o que dificulta o estudo da expressão de genes na interação. Por outro lado, genes não conservados entre os dois organismos puderam ser avaliados quanto à sua expressão. Foi verificada a presença de muitos genes comuns às duas interações (compatível e incompatível), sendo que a grande maioria não apresentou expressão diferencial, principalmente nas fases iniciais do processo infeccioso. As maiores diferenças entre as interações foram encontradas aos 17 dai, consistindo principalmente de genes “no hit”, ou seja, genes que não apresentam similaridade com sequências dos bancos de dados de proteínas. Estas sequências podem corresponder a genes específicos de H. vastatrix. As informações geradas nesta pesquisa poderão auxiliar no melhor entendimento da interação entre H. vastatrix e o cafeeiro. O conhecimento dos genes expressos durante a infecção poderá auxiliar no entendimento dos mecanismos moleculares que levam à suplantação da resistência por novas raças do fungo. The race XXXIII of Hemileia vastatrix was recently identified in Brazil, infecting some cultivars that had been released as resistant to coffee leaf rust. To gain new insights into the interaction between coffee plants and this race of the fungus, we evaluated the fungal growth and the plant defence responses in coffee genotypes resistant (Híbrido de Timor – HDT CIFC 832/1) and susceptible (C. arabica cv. Caturra CIFC 19/1) to race XXXIII. The first cytological responses induced by the fungus were observed particularly in stomatal cells of both coffee genotypes by 17 hours post-inoculation (hpi) and corresponded to hypersensitive-like response (HR) and accumulation of phenolic-like compounds. In the susceptible genotype, the fungus was able to colonize the host tissues, produced a large number of haustoria and sporulated around 20 days post-inoculation (dpi). Reversely, in the leaves of resistant genotype, the fungus stopped its growth with higher frequency at pre-haustorial stages. These cytological observations indicate that the resistance of HDT CIFC 832/1 to race XXXIII of H. vastatrix is pre-haustorial, contrary to the post-haustorial resistance that is generally described for coffee - H. vastatrix interactions. Based on this analysis, the most suitable key time points of the infection process were defined, aiming at the study of differentially expressed genes in the interaction. Although the transcriptome sequencing has been widely used in plant-pathogen interaction studies, a major challenge is to separate transcripts from the plant and the fungus, especially in the absence of reference genomic sequences. For this reason, we performed the transcriptome sequencing and assembly of H. vastatrix (race XXXIII) hydrated and germinated spores. A total of 32.882 contigs were assembled, from which 11.989 genes were predicted. Plants of C. arabica cv. Caturra (CIFC 19/1) and Híbrido de Timor (CIFC 832/1) were inoculated with fresh spores of H. vastatrix race XXXIII to establish a compatible and an incompatible interaction, respectively. No inoculated leaves were used as control. In each time point (12, 24, 96 hours and 17 days post- inoculation), the leaves were removed and used for RNA extraction. Illumina MiSeq sequencing of ten cDNA libraries generated 206,000,000 reads. High quality reads of the H. vastatrix and coffee interacting transcriptomes were mapped against the H. vastatrix (race XXXIII) hydrated and germinated spores transcriptome previously assembled. The data showed that many genes of H. vastatrix are similar to coffee genes, which makes difficult the study of gene expression in the interaction. On the other hand, genes not conserved between the organisms could be evaluated for their expression. The presence of many common genes to both interactions (compatible and incompatible) was verified, and the majority did not show differential expression, especially in the early stages of the infection process. The major differences between interactions were found at 17 dpi, consisting mainly of genes that showed no similarity to protein databases. Those sequences can correspond to H. vastatrix specific genes. This investigation provides new insights into the coffee-rust interaction. The knowledge of gene expression profiling during the infection process may contribute to understanding the molecular mechanisms that lead to the breakdown of resistance by new physiological races of the fungus.

The aim of this study was to select F1 hybrids with high yield, high vegetative vigor and simultaneous resistance to coffee leaf rust, bacterial blight and leaf miner. The experiments were set up in three different Paraná State municipalities: Londrina, at the experimental station of the Instituto Agronômico do Paraná (IAPAR); Congonhinhas and Itaguajé. Yield and plant growth traits were assessed, as well as resistance to coffee leaf rust, bacterial blight and leaf miner in 13 F1 hybrids and four commercial cultivars type pure lines used as control. Relative yield in relation to the mean value of the best line was estimated for all the hybrids. In all locations, hybrids 7, 11 and 12 had average yield significantly higher than the controls. Hybrids 6 and 9 were simultaneous resistant to leaf rust, bacterial blight and leaf miner. Hybrid 12 was resistant to leaf rust and bacterial blight. The hybrids 6, 7, 9, 11 and 12 have potential to become coffee cultivars. O objetivo deste estudo foi selecionar híbridos F1 com alta produtividade, elevado vigor vegetativo e com resistência simultânea à ferrugem, mancha aureolada e ao bicho-mineiro. Os experimentos foram instalados em três diferentes municípios do estado do Paraná: Londrina, na estação experimental do Instituto Agronômico do Paraná (IAPAR); Congonhinhas e Itaguajé. Foram avaliadas as características produtividade e vigor vegetativo, além da resistência à ferrugem, mancha aureolada e bicho mineiro, em 13 híbridos e em quatro cultivares comerciais do tipo linhagem, usadas como controle. Produtividade relativa, em relação à média da melhor testemunha foi estimada para todos os híbridos. Os híbridos 7, 11 e 12 apresentaram produtividade média significativamente maior do que as testemunhas em todos os locais. Os híbridos 6 e 9 foram simultaneamente resistentes à ferrugem, mancha aureolada e ao bicho- mineiro. O híbrido 12 foi resistente à ferrugem e à mancha aureolada. Os híbridos 6, 7, 9, 11 e 12 têm potencial para se tornarem cultivares de café.

Conselho Nacional de Desenvolvimento Científico e Tecnológico A ferrugem alaranjada causada pelo fungo biotrófico Hemileia vastatrix Berk. et Br. é considerada em todo o mundo uma séria doença do cafeeiro que tem causado vários prejuízos para a cafeicultura. O uso de fungicidas é o método mais empregado para o controle da doença, porém sua aplicação deve ser feita de forma racional, para não inviabilizar a cultura e agredir o meio ambiente. Na busca por cultivares resistentes, já foram identificados nove genes dominantes de resitência a H.vastatrix presentes em cafeeiros de diferentes espécies. Uma potencial estratégia para introgredir esses genes de resistência no cafeeiro é o uso de marcadores moleculares. Logo, objetivou-se com esse trabalho, converter marcadores AFLPs ligados a locos de resistência do cafeeiro à raça I, II e ao patótipo 001 de H. vastatrix em marcador SCAR (Sequence Characterized Amplified Region) e avaliar a eficiência do uso desses marcadores e de um microssatélite previamente identificado na Seleção Assistida por Marcadores (SAM). Para isso, quatro combinações de primers AFLPs ligados à resistência do cafeeiro à raça I, II e ao patótipo 001 de H. vastatrix foram clonados e sequenciados. Os marcadores SCAR desenvolvidos foram denominados de SCAFs e, posteriormente, validados nos genitores resistente Híbrido de Timor UFV 443-03; genitor suscetível Catuaí Amarelo IAC 64 (UFV 2148-57) e em 247 indivíduos da população F2 de mapeamento. Os dados foram codificados e analisados no programa GQMOL. Com as sequências dos SCARs foi feita uma busca local no banco de dados do genoma de referência de Coffea canephora utilizando a ferramenta BLAST, e-value de -27. A fim de identificar quais genes estão envolvidos nas regiões dos cromossomos onde os SCARs apresentaram maior similaridade, foi realizada uma busca local utilizando a ferramenta Gbrowse, e-value de -10. Para a realização da SAM foram genotipados o marcador SCAF2 validado e um marcador microssatélite SSR16 em indivíduos das populações F3 e de retrocruzamentos suscetíveis. Na realização da fenotipagem dos 16 discos de folhas de 1,5 cm de diâmetro de cada planta dos genitores, da geração F3 e dos retrocruzamentos foram inoculados com uredósporos da raça II. Após as inoculações, os discos foram colocados sobre uma tela de nylon e espuma, saturada com água e acondicionados no interior de um gerbox. Os gerbox contendo os discos de folhas, foram fechados e mantidos na ausência de luz durante 48 horas, a 22oC e, em seguida, transferidos para uma câmara sob condições controladas de temperatura de 22°C e 12 horas de luz. No momento em que os discos inoculados do genitor suscetível Catuaí Amarelo IAC 64 (UFV 2148-57) iniciou a esporulação, em torno dos 25 dias após a inoculação (dpi), foi iniciada a avaliação de resistência/suscetibilidade. Na validação dos marcadores SCARs apenas o marcador SCAF2 apresentou polimorfismo entre os cafeeiros resistentes e suscetíveis, se comportando como marcador dominante em repulsão. Os outros marcadores SCARs desenvolvidos não apresentaram polimorfismo entre os cafeeiros quando os fragmentos foram analisados em gel de agarose. Logo, somente o marcador SCAF2 foi validado mantendo o mesmo ordenamento no grupo de ligação 2 (GL2) do mapa genético de ligação, ficando ligado a 0 cM do AFLP que lhe deu origem. Na busca local no genoma de C. canephora encontrou-se maior similaridade dos marcadores SCAF1, SCAF2 com o cromossomo 0, chamado de ChrUn, e maior similaridade dos marcadores SCAF3 e SCAF4 com o cromossomo 10. Na identificação dos genes, os marcadores SCAF1 e SCAF2 pertencentes ao GL2 do mapa genético apresentaram similaridade com alguns genes que codificam proteínas relacionadas à resistência a patógenos. Na SAM, foi verificado que o uso dos marcadores moleculares permitiu selecionar indivíduos homozigotos dominantes para um dos locos e para os dois locos, sendo mais eficiente que a fenotipagem. Logo, com o desenvolvimento de marcadores SCAR e SSR16 intimamente ligados aos locos de resistência à ferrugem do cafeeiro e seu uso na SAM possibilitaram um avanço nos programas de melhoramento com a seleção precoce de indivíduos e também uma posterior clonagem posicional de genes ligados à resistência a essa doença. Coffee leaf rust (CLR) caused by the biotrophic fungus Hemileia vastatrix (L.) Berk. & Broome is widely considered as a serious disease causing various damages to coffee production. The use of fungicides is the employed control method, though their application should be made in a rational manner to avoid hampering the culture and the environment. In search for resistant cultivars, nine dominant resistant genes to H. vastatrix have been identified in different coffee species. One potential strategy for the introgression of these resistance genes is the use of molecular markers. Therefore, the objective of this work was to convert AFLP markers linked coffee loci resistant to race I, II and pathotype 001 of H. vastatrix in SCAR (Sequence Characterized Amplified Region) marker and evaluate the efficiency of these markers and a microsatellite marker previously identified in Marker Assisted selection (MAS). For this purpose, four primer combinations of AFLPs linked to resistant genes to race I, II and pathotype 001 of H. vastatrix were cloned and sequenced. Developed SCAR markers were named SCAF and later validated in the resistant parent Híbrido de Timor (UFV 443-03); susceptible parent Catuaí Amarelo IAC 64 (UFV 2148-57) and 247 individuals of the F2 mapping population. Data were coded and analyzed in GQMOL software. With the sequences of the SCAR, a local search was made in the reference genome of C. canephora database using the BLAST tool with e-value of 10-27. In order to identify genes in the regions of the chromosome where the SCAR showed greatest similarity, a local search was performed using the tool GBrowse with 10-10 e-value. To perform the MAS, validated SCAF2 and microsatellite SSR16 markers were analysed in individuals of F3 population and susceptible backcrossing. As part of phenotyping, 16 leaf discs of each parent plant, and the F3 backcross generation were inoculated with race II uredospores. After inoculation, the discs were placed on a germination box with a nylon mesh and foam, saturated with water. The germination boxes containing the leaf discs were sealed and kept in the dark for 48 hours at 22 °C and then transferred into a chamber under 22 ° C and 12 hours of photoperiod. The susceptible parent Catuai Amarelo IAC 64 (UFV 2148-57) sporulation started around 25 days after inoculation (dpi), from there began the evaulation of individual from other pouplations. In the validation of SCAR marker, only SCAF2 marker showed polymorphism between the resistant and susceptible coffee, behaving as a dominant marker in repulsion. The other developed SCAR markers did not show polymorphism between coffee varieties when the fragments were analyzed on agarose gel. Yet, only the marker SCAF2 was validated keeping the same order in the Linkage Group 2 (GL2) of the genetic linkage map, being linked at 0 cM of AFLP from which it was developed. The local search in the C. canephora genome found greater similarity of SCAF1 and SCAF2 markers to chromosome 0, called ChrUn, and greater similarity of SCAF3 and SCAF4 markers to chromosome 10. In identifying genes, SCAF1 and SCAF2 markers belong to GL2 genetic map with some similarity to genes encoding proteins related to resistance to pathogens. In the MAS, it was found that the use of molecular markers allowing the selection of homozygous dominant for one of the loci for the two loci, being more efficient than phenotyping. Therefore, with the development of SCAR and SSR16 markers closely linked to coffee rust resistance loci and their use in the MAS enabled an advance in breeding programs with early selection of individuals and also a subsequent positional cloning of genes linked to resistance to this disease.

Durante a interação com a planta hospedeira, os fungos causadores de ferrugem secretam um arsenal de proteínas efetoras que modificam a estrutura e a função da célula hospedeira, permitindo o estabelecimento da interação biotrófica. Algumas dessas proteínas efetoras, denominadas proteínas de avirulência (Avr), são reconhecidas por proteínas codificadas por genes de resistência, o que desencadeia uma resposta de defesa da planta contra a infecção pelo patógeno. Estudos prévios demonstraram que a proteína HvEC-016 secretada por H. vastatrix apresenta características de proteína de avirulência quando expressa transientemente em genótipos de cafeeiros portadores do gene de resistência S H 1. No entanto, análises funcionais mais detalhadas ainda são necessárias para confirmar o papel dessa proteína durante a interação do patógeno com o cafeeiro. Desse modo, os objetivos deste estudo foram: (i) confirmar a atividade efetora da proteína HvEC_016 como fator de avirulência e/ou virulência em genótipos de cafeeiro com a presença ou ausência do gene de resistência S H 1; (ii) avaliar a estabilidade dos plasmídeos nos clones bacterianos utilizados nos estudos da expressão trasiente nos tecidos foliares de cafeeiro; (iii) identificar novas proteínas secretadas pela raça II de H. vastatrix (isolado Hv-01) que apresentam atividade de avirulência em cafeeiros com diferentes combinações de genes S H , utilizando o vetor de expressão transiente pEDV6 e o Sistema de Secreção Tipo III de Pseudomonas syringae pv. garcae (estirpe 1723). Confirmaram-se as atividades funcionais de avirulência e virulência da proteína HvEC_016 por meio da avaliação do crescimento populacional do clone Psg 1723 pEDV::HvEC_016 em plantas de cafeeiro com e sem o gene S H 1, respectivamente. A análise por PCR de colônia das células bacterianas isoladas a partir dos tecidos foliares infiltrados mostrou que o vetor pEDV6 apresentou estabilidade nos clones bacterianos viiianalisados. A expressão transiente de cinco genes candidatos a efetores (HvEC_006, HvEC_054, HvEC_057, HvEC_081 e HvEC_101) suprimiu o desenvolvimento de sintomas da doença em cafeeiros portadores do gene S H 1. A análise do crescimento das populações desses clones em genótipos com ou sem o gene S H 1 deverá ser futuramente efetuada para comprovar os resultados obtidos. A expressão transiente de proteínas efetoras putativas no citoplasma do cafeeiro mediante a aplicação do sistema EDV constitui uma metodologia importante para a identificação de genes de avirulência e/ou virulência de H. vastatrix. During the interaction with the host plant, rust fungi secrete a range of effector proteins, which modify the structure and function of the host cell allowing the establishment of the biotrophic interaction. The proteins encoded by resistance genes recognize some of these effector proteins, in this case also known as protein avirulence (Avr), and triggers a defense response in the plant that restricts further development of the pathogen. At least nine dominant genes (S H 1-S H 9) which provide resistance to the coffee rust have been genetically identified. In addition, genes from Hemileia vastatrix that encode secreted proteins have also been characterized. A protein secreted by H. vastatrix, HvEC_016, with potential avirulence activity when expressed transiently in coffee genotype carrying the resistance gene S H 1 was previously identified. This transient expression was obtained using the pEDV6 expression vector and the type III secretion system (T3SS) of Pseudomonas syringae pv. garcae. However, further functional analyses are required to confirm the role of HvEC_016 protein during the H. vastatrix interaction with coffee. Therefore, the purposes of this study were: (i) to confirm the HvEC_016 avirulence and/or virulence function in coffee genotypes with the presence or absence of the S H 1 resistance gene ; (ii) to analyse the stability of pEDV6 vector in the bacteria cells infiltrated into coffee leaves; (iii) to identify new proteins secreted by H. vastatrix (isolated HV-01) which have virulence activity in coffee plants with different combinations of S H genes, by analyzing five effector candidate genes (HvECs) using pEDV6 and the T3SS of Pseudomonas syringae pv. garcae (strain 1723). By analysing the population growth of the Psg 1723 PEDv::HvEC_016 clone in coffee plants, it was possible to confirm the avirulence and virulence activities of the HvEC_016 protein in the presence and absence of S H 1 gene, respectively. PCR analysis of bacteria colonies isolated from the infiltrated leaf tissue showed that pEDV6 vector is stable during the growth of the analysed clones in coffee leaves without selection. xTransient expression of five genes HvECs (HvEC_006, HvEC_054, HvEC_057, HvEC_081 and HvEC_101) suppressed the development of disease symptoms in coffee genotypes with the S H 1 gene. Additional experiments are underway to confirm these results and to verify if these effectors have virulence activity in coffee genotypes without resistance genes. These results also demonstrate the potencial of pEDV6 and Pseudomonas syringae pv. garcae for functional studies of candidate effectors from H. vastatrix.
