O ácido chiquímico possui alto valor de mercado, pois é o precursor do fármaco Fosfato de Oseltamivir (Tamiflu®), um dos poucos fármacos disponíveis no mercado para tratamento da gripe A (H1N1). Tal substância é extraída comercialmente apenas do anis estrelado chinês (Illicium verum Hook.f.), sendo necessário buscar novas fontes desta matéria prima. Neste contexto, buscou-se desenvolver um processo utilizando conceito de química verde, onde lagartas predadoras do gênero Automeris sp. foram alimentadas com folhas de Schinus terebinthifolius Raddi, com objetivo de aumentar a concentração do ácido chiquímico na nova matriz, as fezes das lagartas. Para quantificar os ácidos chiquímico e gálico, nas folhas e fezes, nas etapas de otimização da extração e purificação foi desenvolvido um método cromatográfico via Cromatografia Líquida de Alta Eficiência com detector de Arranjo de Diodos (CLAE- DAD). A extração dos ácidos das matrizes foi otimizada ajustando as condições temperatura, tempo, granulometria, diluição e os extratores Soxhlet e autoclave. O método desenvolvido e otimizado, por CLAE-DAD, foi rápido, seletivo, preciso e robusto nas condições avaliadas. Dentre os fatores avaliados na etapa de extração, o que mais influenciou foi a temperatura, com a condição ótima a 110oC, com uso de autoclave. A máxima concentração nesta condição, para o ácido chiquímico foi de 3,7% m/m nas fezes e de 2,9% m/m nas folhas, evidenciando o aumento da concentração do ácido chiquímico nas fezes das lagartas em relação as folhas. Os dois ácidos foram separados com eficiência, através de uma extração em fase sólida, na qual extrato aquoso foi efluído através de uma coluna recheada com uma mistura de carvão ativado e sílica-gel. Para entender o processo foram determinadas as isotermas de adsorção, em fluxo, para os dois ácidos, sendo que a capacidade máxima de adsorção de ácido chiquímico, igual a 7,459 mg.g -1 , e do ácido gálico sendo igual a 55,780 mg.g -1 , calculadas pelo modelo de isoterma de Langmuir. Shikimic acid has a high market value because it is the precursor of the drug Oseltamivir phosphate (Tamiflu®), one of the few drugs available for the treatment of influenza A (H1N1). Such substance is extracted commercially only from Chinese star anise (Illicium verum Hook.f.), and it is necessary to search for new sources of this raw material. In this context, we sought to develop a process using the concept of green chemistry, where predatory caterpillars of the genus Automeris sp. were fed leaves of Schinus terebinthifolius Raddi in order to increase the shikimic acid concentration in the new matrix, caterpillar feces. In order to quantify the shikimic and gallic acids in the leaves and feces, in the extraction and purification optimization stages, a chromatographic method via High Performance Liquid Chromatography with Diodes Array Detector (CLAE-DAD) was developed. The extraction of acids from the matrices was optimized by adjusting the temperature, time, grain size, dilution conditions and the Soxhlet and autoclave extractors. The method developed and optimized by CLAE-DAD was fast, selective, accurate and robust under the evaluated conditions. Among the factors evaluated in the extraction stage, the one that most influenced was the temperature, with the optimal condition at 110oC, with the use of autoclave. The maximum concentration in this condition for shikimic acid was 3.7% w/w in in caterpillar feces and 2.9% w/w in leaves, showing an increase in the concentration of shikimic acid in the feces in relation to the leaves. The two acids were efficiently separated by solid phase extraction, in which aqueous extract was effected through a column filled with a mixture of activated carbon and silica gel. In order to understand the process, the adsorption isotherms in flux were determined for both acids, with the maximum adsorption capacity of shikimic acid equal to 7,459 mg.g -1 and gallic acid being 55,780 mg.g -1 , calculated by the Langmuir isotherm model.
