The article is devoted to the study of isolation processes, chemical structure and rheological properties of chitin-glucan complexes from the biomass of Armillaria mellea. Because of the studies, it was found that the yield of chitin-glucan complexes ranged from 11.0 to 24.9% with a degree of deacetylation from 24 to 55%. The maximum yield was obtained by treating the fruiting bodies of fungi at the stage of deproteinization with a 3% aqueous solution of sodium hydroxide with mechanical stirring at 225 rpm for 60 min at 75±5°C. It is shown that the degree of deacetylation correlates with the values of the alkali concentration used. The IR spectra of the obtained samples of chitin-glucan complexes include absorption bands characteristic of stretching and bending vibrations of the bonds of chitin and glucan links and coincide with the spectra of chitin-glucan complexes of other fungi. It has been established that the viscosity characteristics of a 1% solution of a chitin-glucan complex in 2% hydrochloric acid are 14 and 190 times lower than 5% and 10% solutions, respectively. Increasing the shear rate makes it possible to detect a decrease in the viscosity of all studied solutions. Storage of a 1% solution of chitin-glucan complexes for 7 days at 25±2°C is accompanied by a decrease in viscosity; the viscosity of 5 and 10% solutions remains stable., Статья посвящена исследованию процессов выделения, химической структуры и реологических свойств хитин-глюкановых комплексов из биомассы грибов Armillaria mellea. В результате проведенных исследований было установлено, что выход хитин-глюкановых комплексов составил от 11,0 до 24,9% со степенью деацетилирования от 24 до 55%. Хранение 1% раствора хитин-глюкановых комплексов в течение 7 суток при 25±2°С сопровождается снижением вязкости, вязкость 5 и 10% растворов остается стабильной.

The results of the treatment of acute myeloid leukemia (AML) in pediatric patients remain poor. The modern programs of treatment allow achieving long-term survival in only 6570% of patients and the event-free survival rates are lower and make up approximately 55%. The development of protocols for high-intensity therapy, which are based on the block principle, followed by long-term maintenance therapy has led to the significant progress in the treatment of AML. As supportive care has improved, it became possible to escalate the doses of chemotherapy, which contributed to the increase of the treatment effectiveness. So far, doses and drugs administration regimens have approached the tolerated level, which necessitates the research into new therapeutic targets. An investifation of the epigenetic processes underlying leukemogenesis made it possible to identify those targets DNA methylation and histone deacetylation. This review presents the results of the use of epigenetic drugs in the treatment of AML in children. The possibilities of including epigenetic agents in standard polychemotherapy protocols are presented.

В настоящей работе представлен селективный способ синтеза 3,7,10-триоксо-2,6-диацетил-2,4,6,8,9,11-гексааза[3.3.3]пропеллана через дезацетилирование триацетилпроизводного обработкой последнего серной и азотной кислотами, а также серно-азотной смесью при комнатной температуре с выходом целевого продукта 31 %, 35 % и 80 %, соответственно. При взаимодействии 3,7,10-триоксо-2,6-диацетил-2,4,6,8,9,11-гексааза[3.3.3]пропеллана с серно-азотной смесью, серной или азотной кислотой дальнейшее снятие ацетильной группы до образования синтеза 3,7,10-триоксо-2-ацетил-2,4,6,8,9,11-гексааза[3.3.3]пропеллана не происходит, а нагревание реакционной массы приводит к разрушению исходной молекулы. Однако при взаимодействии 3,7,10-триоксо-2,6-диацетил-2,4,6,8,9,11-гексааза[3.3.3]пропеллана со смесью концентрированной азотной кислоты и уксусного ангидрида образуется нитропродукт диацетилпроизводного ТНАР – 3,7,10-триоксо-2,6-диацетил-9-нитро-2,4,6,8,9,11-гексааза[3.3.3]пропеллан. При этом, в отличие от триацетил-ТНАР, у которого две ацетильные группы заместили сразу два протона NH-группы одного кольца, а другое кольцо осталось без замещения, у нитро-ацетилпроизводного ТНАР функциональные группы стоят при разных кольцах. Было показано, что ацетилирование 3,7,10-триоксо-2,6-диацетил-2,4,6,8,9,11-гексааза[3.3.3]пропеллана в среде уксусного ангидрида приводит к замещению одного протона свободной NH-группы с образованием триацетилзамщенного, а при замене уксусного ангидрида на хлористый ацетил образуется 3,7,10-триоксо-2,4,6,8,9,11-гексаацетил-2,4,6,8,9,11-гексааза[3.3.3]пропеллана., Here we report a selective method for the synthesis of 3,7,10-trioxo-2,6-diacetyl-2,4,6,8,9,11-hexaaza[3.3.3]propellane via deacetylation of a triacetyl derivative by treatment of the latter with sulfuric and nitric acids and with mixed acid at room temperature to give a target product in 31 %, 35 % and 80 % yields, respectively. On the reaction of 3,7,10-trioxo-2,6-diacetyl-2,4,6,8,9,11-hexaaza[3.3.3]propellane with mixed acid, sulfuric acid or nitric acid, no further detachment of the acyl group to the formation of 3,7,10-trioxo-2-acetyl-2,4,6,8,9,11-hexaaza[3.3.3]propellane did not occur, while heating the reaction mass broke down the starting molecule. However, the reaction between 3,7,10-trioxo-2,6-diacetyl-2,4,6,8,9,11-hexaaza[3.3.3]propellane with concentrated mixed nitric acid/acetic anhydride furnished a nitro product of the diacetyl derivative of ТНАР – 3,7,10-trioxo-2,6-diacetyl-9-nitro-2,4,6,8,9,11-hexaaza[3.3.3]propellane. In this case, in contrast to triacetyl-ТНАР which had two acetyl groups, two protons of the NH group in the same ring were substituted at a time, while one ring remained unsubstituted, whereas the functional groups in the THAP nitroacetyl derivative were located on different rings. It was shown that acetylation of 3,7,10-trioxo-2,6-diacetyl-2,4,6,8,9,11-hexaaza[3.3.3]propellane in acetic anhydride leads to the replacement of one proton with free NH-group with the formation of triacetyl-substituted, and when acetic anhydride is replaced by acetyl chloride, 3,7,10-trioxo-2,4,6,8,9,11-hexaacetyl-2,4,6,8,9,11-hexaaza[3.3.3]propellane is formed., Южно-Сибирский научный вестник, Выпуск 6 (34) 2020
