The purpose of work to study the conditions of prooxidant andantioxidant system in the skin of guinea pigs in different periods of contact dermatitis formation.Materials and methods. Researches were conducted on guinea pigs, divided into five groups: I - control, II – 4th day of expirement, III – 8th day of contact dermatitis development, IV – 10th day and V - 18 th day of model process. Early period included groups of animals on the 4th and 8th days of experiment. The late one – guinea pigs on the 10th and 18th days of contact dermatitis. Experimental contact dermatitis was simulated by method of Volkovoj V. A. (2010). Condition of free radical lipid oxidization in the skin was determined on maintenance content of malonic dialdehyde by method of Corobeynikov E. G. (1989) and diene conjugates by method of Gavrylov V. G., Myshkorudna M. I. (1989). The degree of activity of antioxidant defence was estimated on maintenance enzymes - superoxidedismutase by method of R. Fried (1975), catalase by the method of B. Holmes, C. Masters (1970), glutationperoxidase method of Arkhipova O. G. (1988) and glutationreductase method of Moina V. M. (1986).Results and discussion. Gradual increasing of lipid peroxydations products - malonic dialdehyde, diene conjugate had been determined on the 4th, 8th, 10th and 18th days of experimental contact dermatitis development in this research. Primary increasing of the indices of superoxidedismutase, glutationreductase, glutationperoxidase and catalase has been investigated in the skin on the 4th day with the following decreasing of these enzymes from the 8th day of experiment. It testified about imbalance in antioxidant and prooxydant systems, especially on the 10th and 18th days of the study, which exacerbates the inflammatory process in the skin and also triggers a cascade of other mechanisms of damage.

As a result of the conducted researches it is established that with the development of experimental contact dermatitis and experimental pneumonia there is an increase in the level of stable metabolites of nitric oxide, increase of activity of total NO synthase and, at the same time, inhibition of L-arginine activity in the blood, thus these indicators were most expressed on the late stages of her forming of combine pathology(18th day). The use of thiotriazoline in curative aims stipulated the decline of stable NO metabolites on 31.9% (p≤0.05), NOS in blood on 26.6% (р1 ≤0.05) and increase of content L-arginine on 34.5% (р1 ≤0.05) at experimental contact dermatitis and experimental pneumonia as compared to the group of animals that did not yield to influence of this preparation.

The aim of our research was to determine the character of the disturbances content level of pro-inflammatory interleukin-6, tumor necrosis factor-α and anti-inflammatory cytokine interleukin-10 (IL-10) in the serum of guinea pigs and estimation of thiotriazoline influence on the cytokine profile in the conditions of formation of combined pathology - experimental contact dermatitis and experimental pneumonia.It has been determined significant advantage of pro-inflammatory cytokine activity, which increasing according the development of the combined pathological process and, simultaneously, reducing the anti-inflammatory IL-10. It has been detected corrective action of thiotriazoline on the investigated indices of cytokine profile.

Literary sources indicate that various pathological processes are accompanied by the development of a violation of the immune system.The aim of the study was to determine the pathogenetic justification for the use of thiotriazoline on the basis of immune system disorders in experimental pneumonia (EP) and contact dermatitis (CD).The researches were carried out on 55 guinea-pigs. The study material was collected on the 4th, 8th, 10th and 18th days of EP and ECD under ether anesthesia. EP was called by the method of VN Shlyapnikov, TL Solodov. ECD was simulated by method of VA Volkovoj. The content of T and B lymphocytes in the blood was determined by the method of Chernushenko KF, Kogosova LS, and the level of the circulating immune complexes (CIC) by the method of V. Haskova, J. Kaslik. We have established a gradual increase in the level of B-lymphocytes and CIC against the background of a decrease in T-lymphocytes in the blood on the 4th, 8th, 10th and 18th days of the development of EP and CD, with a predominance on the 18th day of the experiment before treatment. The use of thiotriazoline caused an immunocorrective effect in CD and EP.

Тhe aim of our work was to find out the peculiarities of changes ofendogenous intoxication indices in guinea pig blood during theformation of combined pathology - experimental contact dermatitis and experimental pneumonia and correction of their disorders with thiotriazoline.The paper found a gradual increase in the molecules of mediummass and erythrocyte index of intoxication in guinea pig serum with experimental contact dermatitis and experimental pneumonia with the highest degree of expression on the 18thday of the experiment, and this may be may indicate the development of endogenous intoxication of the animal body. At the same time, the use of the drug thiotriazoline caused the decrease of the studied indicators, which gives grounds to stateits positive effect on certain markers of endogenous intoxicationand the feasibility of further research.

The work demonstrated, that polymorho-nuclear leucocytes take an active part in formation of experimental contact dermatitis and experimental pneumonia before and after administration of the antioxidant thiotriazoline and also testify to metabolic processes stimulation in leucocytes. The immune-modulating thiotriazoline preparation application, beginning from 8 th day of the experiment, and during 10 days, showed the renewal of indexes of phagocytar leucocytes' activity, the test of nitroblue tetrasolium, what testifies to its corregating action – on nonspecific resistance condition.

The aim of our research was to to investigate the features of the proteinase-inhibitory system in guinea pig lungs in the dynamics of the formation of experimental contact dermatitis (ECD) and experimental pneumonia (EP) and to establish the correction of their disorders by thiotriazoline.Materials and methods. The researches were carried out on 60 guinea-pigs which were divided into 6 groups. I group (control) were intact guinea-pigs, II group – were animals with an ECD and EP (4th day), III group – were guinea-pigs on the 8th day of the experiment, IV group consisted of animals with an experimental CD and EP (10th day) and V group included guinea-pigs with an combined pathology (18th day, without thiotriazoline using). The last sixth group included animals with an ECD and EP that were treated by thiotriazoline from the 8thto the 18th days of the experiment. Two periods of development of ECD and EP were distinguished: early (4th and 8th days of experiment) and late (10th and 18th days). Experimental contact dermatitis was simulated by method of VA Volkovoj (2010). EP was called by the method of VN Shlyapnikov, TL Solodov (1998). Thiotriazoline was administered intramuscularly at a dose of 100 mg per 1 kg of weight daily from the 8th to the 18th days of the experiment. The condition of proteinase-inhibitory system was determined by lysis of the azoalbumin, azokasein and azokolagen and maintenance content of α1-protease inhibitor (α1-PI), α2-macroglobulin (α2-М) by method of Veremeenko K.N., Goloborodko O.P. (1988). Results and discussion. It is established that under the conditions of development of experimental contact dermatitis and experimental pneumonia there are changes in the proteinase-inhibitory system, which is manifested by activation of proteolysis. There was an increase in the level of azoalbumin, azocasein and azocollagen, especially on the 18th day of the experiment and depression of protease inhibitors. This led to the formation of proteinase-inhibitory imbalance before treatment. The use of thiotriazoline led to the correction of proteolysis (their reduction) and the growth of protease inhibitors in the lungs with the development of contact dermatitis and experimental pneumonia, which indicated its corrective effect on impaired proteinase-inhibitory system markers and its appropriate pathogenetic justification.
