Аннотация. Виноград обладает богатым генетическим разнообразием и традиционно хранится в генбанках, главным образом в виде полевых коллекций, содержание которых дорого и ненадежно, т.к. насаждения могут быть потеряны в результате биологических или экологических катастроф. Принимая во внимание то, что многие сорта — это незаменимый ресурс для виноделия, часть национального наследия и культуры, методы сохранения разнообразия рода Vitis должны быть дополнены коллекциями in vitro.Вопрос о производстве оздоровленного посадочного материала не перестает быть актуальным. Среди способов воспроизводства растений лидирующее место по преимуществам занимает метод клонального микроразмножения растений. В статье представлены результаты исследований по влиянию различных источников углеводов в питательных средах на развитие растений сортов винограда Каберне-Совиньон и Фиолетовый ранний. Углеводы в культуральных средах играют роль основного питания растений, а также влияют на осмотические характеристики жидкости, что можно использовать для регулирования интенсивности протекания физиологических процессов. Исследования проведены в 2018-2022 гг. на растениях винограда из коллекции in vitro лаборатории биотехнологии Всероссийского научно-исследовательского института виноградарства и виноделия имени Я.И. Потапенко. Изучены особенности роста и развития растений винограда сортов Каберне-Совиньон и Фиолетовый ранний на культуральных средах с разными источниками углеводов – сахароза, фруктоза, сорбит в диапазоне концентраций 5-60 г/л. Определены параметры применения углеводов, позволяющих стимулировать и минимизировать скорость роста растений винограда. Полученные результаты позволят усовершенствовать биотехнологию создания и содержания коллекций винограда in vitro.Максимальные показатели сохранности и продолжительности нахождения в культуре зафиксированы у растений на среде с сорбитом (93 % и 316 дней культивирования). The paper presents the results of research on the influence of various sources of carbohydrates in nutrient media on the development of grapevine cultivars ‘Cabernet-Sauvignon’ and ‘Fioletovyi Ranniy’. Carbohydrates in culture media play the role of main plant nutrition, and also affect the osmotic characteristics of the liquid, which can be used to regulate the intensity of physiological processes. The research was carried out in 2018–2022 on the collection of grapevine plants in vitro at the All-Russian Research Institute for Viticulture and Winemaking named after Ya.I. Potapenko. The features of growth and development of ‘Cabernet-Sauvignon’ and ‘Fioletovyi Ranniy’ grapevines on culture media with different sources of carbohydrates – sucrose, fructose and sorbitol in the concentration range of 5-60 g/l were studied. The parameters of using carbohydrates were determined to stimulate and minimize the growth rate of grapevine plants. The results obtained will allow improving the biotechnology of creating and maintaining grapevine collections in vitro. The maximum indicators of preservation and duration of keeping in culture were recorded in plants on a medium with sorbitol (93 % and 316 days of cultivation). Представлено описание наиболее характерных симптомов поражения возбудителем сосудистого микоза дуба Ceratocystis fagacearum (Bretz) J. Hunt, позволяющих визуализировать патоген. В процессе исследования выявлены характерные конидиальные стадии типа Chalara для C. fagacearum, а для Ophiostoma quercus (= O. roboris) (Georgev.) Nannf. и O. piceae (Münch) Syd. & P. Syd. – конидиальная стадия типа Graphium, позволяющие различать между собой североамериканские и европейские виды фитопатогенных грибов, поражающих дубы. Показано, что оптимальными питательными средам для образования данных спороношений являются солодово-дрожжевой агар и морковная среда – для C. fagacearum, 2% КГА и морковная среда – для O. piceae., The description of the most typical symptoms of Ceratocystis fagacearum (Bretz) J. Анализируемыми показателями ростостимулирующей активности штаммов микроорганизмов были – Ð²ÑÑ Ð¾Ð¶ÐµÑÑ‚ÑŒ семян, длина корешков проростков, титры культивирования на Ð¿Ð¸Ñ‚Ð°Ñ‚ÐµÐ»ÑŒÐ½Ñ‹Ñ ÑÑ€ÐµÐ´Ð°Ñ . В Ñ Ð¾Ð´Ðµ исследований показано, что наибольшую ростостимулирующую активность в отношении пшеницы показали штаммы О-12 и Я-2, в отношении свеклы - штамм О-12. Отобранный штамм Pseudomonas sp. При выращивании данного штамма методом глубинного культивирования на полусинтетической питательной среде с добавлением трегалозы изменение скорости мешалки не увеличивает производительность процесса спустя 72 часа после начала культивирования., This work is devoted to the study of the growth-stimulating activity of collection strains of microorganisms, the preparation of nutrient media and the selection of cultivation conditions in a laboratory fermenter. The tasks of the study: - to study the growth-stimulating activity of collection strains of microorganisms (VNIISKhM collection) and select the most promising strain; - compose nutrient media for the cultivation of the most promising strain based on available and technological components; - to offer recommendations for the development of a technological regime for growing a culture in a laboratory bioreactor. The object of the study is strains of collection and promising isolated microorganisms, including Pseudomonas species pcs. PG-5, 2P-5, 2P-7, O-12, 2P-9, I-2, 2P-12, spring wheat variety Sudarynya, beetroot vegetable variety Detroit. The work was carried out on the basis of the laboratory of biotechnological production "Ecos Biopreparations". The analyzed indicators of the growth-stimulating activity of microorganism strains were seed germination, seedling root length, cultivation titers on nutrient media. In the course of the research, it was shown that strains O-12 and Ya-2 showed the greatest growth-stimulating activity in relation to wheat, and strain O-12 in relation to beets. The selected strain of Pseudomonas sp. O-12 is most responsive to the addition of additional amounts of trehalose and mannitol to nutrient media. When growing this strain by submerged cultivation on a semi-synthetic nutrient medium with the addition of trehalose, changing the stirrer speed does not increase the productivity of the process 72 hours after the start of cultivation.

Лабораторная диагностика имеет решающее значение в выявлении туберкулёза животных. Предложенные питательные среды для изолирования возбудителя богаты разнообразием. Изучены практическая значимость часто используемых питательных сред и их модификации. Исследованы образцы сред Петраньяни, Герольда, Евглевского, Мордовского, Павловского, Левенштейна-Йенсена, Финна-II и некоторые их модификации, для первичного выделения и пересева микобактерий. В сравнительном плане изучены среды также Левенштейна-Йенсена и Финна-II в различных вариантах: Левенштейна - Йенсена, по прописи и замороженная; Левенштейна-Йенсена, сухая и замороженная; Финна-II в авторском варианте и четырех модификациях, АФ - I (гликоколь вместо глутамата натрия), АФ-II- (с картофельным экстрактом), АФ-III - (без гликоколя) и вариант «МФ» (с экстрактом M. flei). Для посева использовали биоматериал от больных животных, также искусственно инфицированный. Исследования позволили установить результативность сред Левенштейна-Йенсена и Финна-II для первичного выделения возбудителя. По ростовым свойствам сухая среда Левенштейна-Йенсена превосходит нативную, но значительно уступает нативной среде Финна по ингибирующим постороннюю микрофлору свойствам. Возможность ускорения роста микобактерии углеводородами определяли добавлением н-алканов с широким диапазоном углеводов – от декана до нонадекана (от С10 до С22). Всего испытано 30 разных вариантов: добавление жидкого парафина; спиртового экстракта M. flei; растворов воска; прополиса на гексане; сыворотки крупного рогатого скота и другие. Использование в качестве стимулятора роста н-алканов, в колическтве 2-3 капель на 10 мл питательной среды (1 капля на 4 мл среды), позволило сократить сроки культивирования на 15-20 дней и увеличить бактериальную массу примерно в 2 раза. The proposed nutrient media for isolating of the pathogen are rich in variety. The practical significance of frequently used culture media and their modifications have been studied. Samples of Petragnani, Gerold, Evglevsky, Mordovsky, Pavlovsky, Levenshtein-Jensen, Finn-II and some of their modifications for the primary isolation and subculturing of mycobacteria were studied. Also, in a comparative plan, the Levenshtein-Jensen and Finn-II media were studied in various versions: Levenshtein-Jensen, prescribed and frozen; Levenshtein-Jensen, dry and frozen; Finna-II in the author's version and four modifications, AF-I (glycocol instead of monosodium glutamate), AF-II- (with potato extract), AF-III - (without glycocol) and MF variant (with M. flei extract). For sowing, biomaterial from sick animals, also artificially infected, was used. The studies made it possible to establish the effectiveness of the Levenshtein-Jensen and Finn-II media for the primary isolation of the pathogen. In terms of growth properties, the dry Levenshtein-Jensen medium is superior to the nitive one, but is significantly inferior to Finn's native medium in terms of properties that inhibit foreign microflora. The ability to accelerate the growth of mycobacteria with hydrocarbons was determined by the addition of n-alkanes with a wide range of carbohydrates - from decane to nonadecane (from C10 to C22). A total of 30 different options were tested: by adding liquid paraffin; alcohol extract of M. flei; wax solutions; propolis on hexane; bovine serum and others. The use of n-alkanes as a growth stimulator in the amount of 2-3 drops per 10 ml of nutrient medium (1 drop per 4 ml of medium) made it possible to reduce the cultivation time on 15-20 days and increase the bacterial mass in about 2 times. The paper analyzes the influence of the culture liquid soil microalgae Eustigmatos magnus, Haematococcus pluvialis and Chlorella vulgaris growth and development of seedlings of radish seeds. It was revealed that the culture fluid of microalgae capable of stimulating the growth and development of seeds of radish, mostly affecting the performance length of the aerial part of seedlings and biomass performance and length of roots.

The study of the characteristics of the development of cyanobacteria of the genera Anabaena and Nostoc on various nutrient media was carried out. It has been shown that cyanobacteria have low and medium levels of variability in cultures (6,15–22,7%), and thecharacteristics of colonies, filaments and cells depend on the composition of the nutrient medium. It has been established that the optimal medium for the cultivation of representatives of thegenera Anabaena and Nostoc is the Bold’s Basal Medium without nitrogen. Cultivation of cyanobacteria on this medium leads to an active accumulation of biomass and an increase in theprocess of nitrogen fixation.

Представлена бактериологическая схема выделения и идентификации бактерии A. salmonicida с использованием оригинальных питательных сред и комплекса бактериологических тестов. Разработанные питательные среды специфичны по отношению к бактериям-ассоциантам других видов и родов. Предлагаемая схема предполагает выделение и идентификацию микроорганизма за 60-84 часа. В результате апробирования данной схемы было выделено 17 штаммов A. salmonicida из 97 проб, отобранных из водных объектов г. Ульяновска и Ульяновской области., A diagram for separation and identification of bacterium A. Salmonicida using original nutrient media and a complex of bacteriological tests is presented in the paper. The developed nutrient media are specific with regard to bacteria-associates of other species and genus. The proposed scheme assumes separation and identification of the microorganism in 60-84 hours. As a result of the testing of this scheme, 17 strains of A. Salmonicida were detected in 97 samples taken from water bodies in Ulyanovsk and the Ulyanovsk region.

Dry nutrient media on the basis of hepatic infusion and Siberian roach hydrolyzate are constructed. Experimental data demonstrate that these media possess high sensitivity, completely inhibit growth of S-form Brucella, do not require рН adjustment, filtration and autoclaving. They can be used for isolation, cultivation and accumulation of L-form Brucella for bacteriological diagnostics of brucellosis. As these media are accessible for transportation, they are applicable for stationary and field conditions.

Nine kinds of strawberry pathogen fungi and five different nutrient media were taken to carry out experiments on evaluation of growth characteristics and sporulation of phytopathogen fungi on various nutrient media. The Goryaev camera was taken to evaluate the intensity of sporulation on each medium. Интенсивность спорообразования на каждой из сред учитывалась с помощью камеры Горяева. По результатам опыта для каждого патогена предложены питательные среды, на которых интенсивность спорообразования является максимальной.

5 experimental nutritious bases were prepared out of plant materials (soybeans, soymilk, sugar-beet molasses - sugar syrup), and their physical and chemical parameters and amino-acid composition were studied. Nutrient media developed on experimental basis met the nutritious demands of studied strains of plague, cholera, anthrax microbes and sporogenous saprophytes.
