2. Prospects for using CRISPR-Cas9 system in the treatment of human viral diseases

The aim. To analyze the possibility of using the genetic mechanisms of CRISPR-Cas9 technology in the prevention and treatment of certain viral diseases.Materials and methods. The search for publications was carried out in Russian and foreign literature using the following search engines: RSCI, Cyberleninka, eLibrary, PubMed, Cochrane Library, etc. A review of domestic and international scientific papers on the research topic was carried out using search keywords: CRISPR, genetic engineering, genome editing, Cas9, sgRNA.Results. A review of using CRISPR-Cas9 method (“genetic scissors”) as a gene therapy for some viral diseases was carried out, and its main advantages and disadvantages were revealed. An analysis of the data of scientific studies on genetic research methods over the past decade discovers the main aspects of CRISPR-Cas9 technology, modern classification and prospects for using this technology in clinical practice for the treatment and prevention of human viral diseases. The possibilities of creating a more versatile and stable version of the CRISPR-Cas9 technology are considered. Particular attention is paid to the technological difficulties and obstacles that scientists face when implementing this system for targeted use in clinical medicine.Conclusion. One of the rapidly developing areas in science giving promising prospects for modern healthcare is genetic engineering, especially in cases where scientific developments are applied in clinical practice. The discovery of “genetic scissors” technology has revolutionized all medicine. Wide opportunities for developing new treatment methods for many viral diseases and creating conditions for their early prevention opened up for the medical community. In the future, with the introduction of this technology into clinical practice, it will become possible to treat diseases that have not previously responded to ongoing therapy and were considered incurable.

3. The effect of the Pharmaiod preparation in the virus elimination of pear plants using the thermotherapy method

Currently, the main methods of virus elimination of fruit crops are dry-air thermotherapy, meristem culture and chemotherapy. The use of a complex of physical and chemical methods makes it possible to increase the efficiency of virus elimination. Improving the methods of recovery must be carried out in relation to certain types of viruses, the biological characteristics of the crop and variety. The purpose of the research is to study the effect of the Pharmaiod preparation on biometric, physiological parameters and the release of harmful viruses-free pear plants in the process of dry-air thermotherapy. Improvement of pear plants of varieties Letnyaya Zabava, Zolotoy Vityaz, hybrids R-11-9, R-10-3 and R-2-4 from harmful latent viruses of Apple stem grooving virus (ASGV), Apple stem pitting virus (ASPV), Apple chlorotic leaf spot virus (ACLSV), Apple mosaic virus (ApMV) were carried out in 2020-2021 at the temperature of 38 °C for 3 months using a thermal chamber designed by the Federal Horticultural Research Centеr for Breeding, Agrotechnology and Nursery. Biometric parameters in pear plants depended on varietal characteristics, the duration of therapy and the concentration of the Pharmaiod preparation. In most of the varieties and forms studied, after 3 months of thermotherapy, the Pharmaiod preparation at a concentration of 1 ml/l contributed to an increase in the length of 1 shoot of pear plants by 1.4-3.1 times compared with the control. Under the conditions of a thermal chamber, according to a set of indicators, pear hybrids R-11-9 and R-10-3 were characterized by the highest heat resistance; medium heat resistance was shown by Letnyaya Zabava and Zolotoy Vityaz varieties; low – by R-2-4 hybrid. Higher heat-resistant varieties and forms were characterized by a more restrained growth of shoots in length compared to lower heat-resistant ones. An average negative significant correlation at the 5% significance level was established between the water content in the leaves and the length of 1 shoot (r = -0.52). The yield of virus-free pear plants depended on the type of virus. The yield of pear plants free from 4 major harmful viruses as a result of dry-air thermotherapy without the use of the Pharmaiod preparation was 50 %, with the use of the drug it was 60 %.

Secretory phospholipases A2 (sPLA2) represent a large superfamily of enzymes with a molecular weight of 14-19 kDa, including 15 groups and more than 30 isoforms belonging to four types: secretory (sPLA2), cytosolic (cPLA2), calcium-independent (iPLA2) and lipoprotein-associated phospholipase A2 (LP-PLA2, PAF-AH). Eleven species of secretory sPLA2s (IB, IIA, IIC, IID, IIE, IIF, III, V, X, XIIA, and XIIB) have been found in mammals, performing versatile functions and participating in the pathogenesis of a wide range of diseases. On the one hand, sPLA2 may promote elimination of damaged, apoptotic cells by hydrolyzing membrane phospholipids, and exerts a strong bactericidal and antiviral properties, including pronounced effects against antibiotic-resistant strains of microorganisms. In this regard, the use of sPLA2 may represent a new strategy for the treatment of bacterial and viral infections. Moreover, due to the action of sPLA2 on its substrates, a number of biologically active molecules (arachidonic, lysophosphatidic acids, lysophospholipids, fatty acids, prostaglandins, leukotrienes, thromboxanes) are formed, which provide strong inflammatory, detergent, coagulating effects and increase vascular permeability. This pro-inflammatory role of sPLA2 may explain its increase levels and activity in cardiovascular, respiratory, autoimmune, metabolic, oncological, bacterial and viral disorders. The review article presents a classification of sPLA2 isoforms, their substrates, regulatory factors, biological significance, and mechanisms of their strong bactericidal, virucidal, and pro-inflammatory activity in the heart and lung disorders, autoimmune, metabolic, bacterial, and viral diseases. In particular, the mechanisms of the selective action of sPLA2 against Gram-positive and Gram-negative microorganisms are discussed. We consider diagnostic and prognostic significance, correlations between elevated levels and activity of sPLA2 and distinct clinical symptoms, severity and outcome in the patients with coronary heart disease (CAD), acute myocardial infarction (AMI), atherosclerosis, acute inflammatory lung injury (ALI), respiratory distress syndrome (ARDS), chronic obstructive pulmonary disease (COPD), rheumatoid arthritis, bronchial asthma, bacterial infections, septicemia and viral (COVID-19) infections. The opportunity of using sPLA2 as a biomarker of the severity and outcome of patients with chronic obstructive pulmonary disease, bacterial infections, sepsis and viral infections, including COVID-19, is also considered.

5. The results of catching winged aphids with water traps on potato plantings in the southern part of the Arkhangelsk region

Dynamics of the number and species composition of aphids were studied on seed potatoes using yellow water traps in the southern part of the Arkhangelsk region in 2018-2019. 12 species of winged aphids (165 insects) were identified in 2018, and 16 species (115 insects) - in 2019. The following types dominated: black bean aphid A. fabae Scop., bird cherry-oat aphid R. padi L., blackcurrant-sowthistle aphid H. lactucae L., glasshouse-potato aphid A. solani Kalt. The share of aphids as direct and indirect vectors of viral diseases was 91 % of the total number in 2018, and 79.1 % in 2019. The pest population of potato plants depended directly on weather conditions of the growing season. The sum of the average daily temperatures during the period of catching aphids was 1273.3 C in 2018, and 983.3 C - in 2019. During the experimental period, 131.6 mm of precipitation fell in 2018, and 280.4 mm - in 2019. Due to the constant presence of aphids on seed potato crops, annual monitoring of pest population and, in case of necessity, protective measures are required.

6. Influence of Infectious Diseases Incidence on Epidemic Situation with Chronic Diseases of the Gastrointestinal Tract

Aim. To study the relationship of the incidence of infectious and non-infectious diseases of the digestive system.Materials and methods. A retrospective epidemiological analysis of the incidence of the population of the Russian Federation by diseases of the digestive system of an infectious and non-infectious nature for the period of 2002-2019. The data were sampled from state statistics forms No. 2, No. 23; determining the relationships between these disease groups using Pearson correlation methods and linear regression. A case-control study by questioning a sample of 107 people.Results. A parallel increase in the incidence of diseases of the digestive system of a non-infectious and infectious nature is observed. The most pronounced increase is in the incidence of viral infections. Strong correlation exists between rotavirus infection and diseases of the liver, pancreas, intestines; between norovirus infection and diseases of the liver, pancreas; enterovirus infection and pancreatic disease; as well as hepatitis A and gallbladder diseases. Patients suffering from chronic non-infectious diseases of the digestive system, with a reliably high chance, often suffer from acute intestinal infections, rotavirus infection, hepatitis A, and are also better aware of their etiology than those who do not have such diseases.Conclusion. Prevention of infectious diseases of the digestive system will reduce the incidence of noncommunicable diseases associated with them.

7. Influence of the level of mineral nutrition and complex agrotechnical techniques on seed productivity and quality of new potato varieties of different maturity groups in specific soil and climatic conditions

Relevance. The aim was to study the reaction of new potato varieties early ripening group Zabava and middle-ripening group Smolyanochka on the timing of planting and doses of mineral fertilizers for obtaining maximum yield of seed potatoes. As mineral fertilizers used azophoska, superphosphate and potassium chloride.Results. It was found that the variety Zabava compared to the variety Smolyanochka, had a higher stem-forming ability and exceeded the parameters of the stem in all variants of the experiment. This is due to the biological characteristics of the studied varieties, since the Zabava variety, unlike the Smolyanochka variety, had more multiple eyes on the surface of the tuber. Against the background of applying mineral fertilizers N60P72K90, the density of the stem was higher, compared with the background of applying N32P32K32 in both the Zabava variety and the Smolyanochka variety. It should also be noted that there was a tendency to reduce the stem at a later date of planting in all variants of the experiment. The percentage of viral diseases affected increased with later planting dates and was highest with the planting dates of 14 and 20 may. The highest yield of tubers of the seed fraction was obtained in the variant with a planting date of may 14 against the background of balanced doses of mineral fertilizers (N60P72K90) and amounted to 13.3 t/ha in the Zabava variety, 13.0 t/ha in the Smolyanochka variety. It should be noted that different backgrounds of mineral fertilizers did not have a significant impact on the percentage of potato tubers affected by diseases. At the same time, the incidence of tubers with diseases did not exceed the regulatory requirements of GOST R 53136 – 2008 "Seed potatoes. Technical conditions».

8. FEATURES OF THE AGROTECHNOLOGY OF EARLY RIPE GRADES OF POTATOES AT CULTIVATION ON THE SEED PURPOSES IN CONDITIONS OF THE SMOLENSK REGION

The most significant in the technology of growing potatoes are the timing of removal of the tops, as this factor directly affects the physiological state of the tubers and their quality. Therefore, the aim of the study was to determine the effect of the timing of removal of tops on yield, quantitative yield and quality of elite seed of early maturing potato varieties Snegir, Delphin, Udacha and Zhukovskij rannij. Experience, observations and accounting were carried out according to generally accepted methods. The study was conducted in a field experiment of the Smolensk Institute of agriculture (ex. Smolensk GOSHOS). As a result of phenological observations, it was found that the mass flowering did not occur simultaneously in all studied potato varieties. Before all the phase of mass flowering occurred in the Zhukovskij rannij variety. The percentage of plant viral diseases in all studied varieties in the variant with the term of removal of the tops 21 days after mass flowering was higher, compared with the options for removing the tops in earlier flowering periods. The maximum yield of seed fraction of potato tubers in varieties Delphin and Snegir (12.6 and 26.1 t/ha, respectively) was noted in the version with the second term of removal of the tops, in varieties Zhukovskij rannij and Udacha (23.2 and 23.4 t/ha, respectively) – in the version with the term of removal of the tops 21 days after mass flowering. The yield of tubers seed fraction and their mass were the highest varieties of Dеlphin and Zhukovskij rannij. According to the results of the tuberous analysis, an increase in the total number of tubers affected by fungal diseases was observed in variants with later dates of removal of the tops. The most affected disease of all the studied varieties was the grade of bullfinch (1.5-4%), the most resistant to disease – cv. Udacha.

Вирусы – мельчайшие возбудители инфекционных болезней растений, не имеющие клеточного строения и способные размножаться только в живых клетках организма – хозяина. Детальное изучение симптомов вирусных болезней имеет важное значение в борьбе с ними. Поэтому технологическая схема управления фитосанитарным состоянием косточковых культур должна включать этапы мониторинга, оценки его результатов, а также критерии биологической эффективности и экологической безопасности на основе диагностики патогенов. В насаждениях косточковых культур центральной зоны садоводства региона с 2011 по 2017 гг. нами выявлены следующие виды наиболее распространенных вирусных болезней: некротическая кольцевая пятнистость (НКП), хлоротическая кольцевая пятнистость (ХКП), ямчатость древесины, деформирующая мозаика, ленточный узор, звездчатая пятнистость косточковых культур. Уточнены признаки поражения листовой пластинки, плодов, штамбов, ветвей, побегов. Вскрыты основные причины, влияющие на развитие данных болезней. Дан анализ особенностям характера заболеваний и природы патогенов. В предлагаемой статье поднимается актуальная для каждого садовода проблема борьбы с вирусными болезнями косточковых культур., Viruses are the smallest pathogens of plant infectious diseases that do not have a cellular structure and are able to reproduce only in living cells of the host organism. A detailed study of the symptoms of viral diseases is important in combating them. Therefore, the technological scheme for managing the phytosanitary condition of stone fruit crops should include the stages of monitoring, evaluation of its results, as well as criteria for biological effectiveness and environmental safety based on the diagnosis of pathogens. In plantations of stone fruits of the central zone of gardening in the region from 2011 to 2017. We identified the following types of the most common viral diseases: necrotic ring spotting (NCP), chlorotic ring spotting (HKP), wood pitting, deforming mosaic, ribbon pattern, star-shaped spotted stone fruit. The signs of lesion of the lamina, fruits, shtambov, branches, shoots are specified. Revealed the main causes affecting the development of these diseases. An analysis of the nature of the dis-ease and the nature of pathogens is given. 10. Значение оздоровленного от вредоносных вирусов гибридного фонда малины в современной селекции

В мире остро стоит проблема ограничения распространения вредоносных вирусных заболеваний малины и создания высокоустойчивых сортов. В настоящее время на малине выявлено более 20 вирусных болезней, из которых наибольшее распространение получили 5 из них: кольцевая пятнистость малины (RpRSV), мозаика резухи (ArMV), латентная кольцевая пятнистость земляники (SLRSV), черная кольцевая пятнистость томата (TBRV) и кустистая карликовость малины (RBDV). Зараженные вирусами растения малины снижают энергию роста, коэффициент размножения, семенную продуктивность, всхожесть семян, качество плодов и урожайность. Использование в скрещиваниях инфицированных исходных родительских форм приводит к получению заведомо больных гибридов, что не позволяет объективно их оценить по основным хозяйственно-ценным признакам и свойствам. Для использования в селекции оздоровленного от вредоносных вирусов гибридного фонда малины в каждом селекционно-питомниководческом центре необходимо разработать программу создания генофонда высшего качества и строго её соблюдать. Основными пунктами этой программы являются использование в скрещиваниях только свободных от вредоносных вирусов родителей; регулярное ретестирование исходных форм; проведение гибридизации в условиях закрытого грунта, исключающего лёт насекомых и неконтролируемое перемещение зараженной пыльцы или размещение здоровых родительских форм на отдельных участках с пространственной изоляцией 1,5-2 км; оздоровление ценных родительских форм; немедленное удаление с участков сильно восприимчивых генотипов. Надёжный способ повышения устойчивости к заражению вредоносными вирусами – генетическая устойчивость. Решить эти задачи можно только при комплексном подходе. Для этого необходимо создание современных селекционно-питомниководческих центров, где будут проводиться совместные исследования селекционеров, вирусологов и питомниководов, The problem to limit spreading of malicious viral diseases of raspberry and create high-resistant varieties is acute in the world. At present, more than 20 viral diseases have been revealed on raspberry 5 of which are the most widespread: ring spot of raspberry (RpRSV), rock-cress mosaic (ArMV), latent ring spot of raspberry (SLRSV), black ring spot of tomato (TBRV) and bushy nanism of raspberry (RBDV). The plants of raspberry infected with viruses reduce growth energy, multiplication coefficient, seed production, seed germination, fruit quality and yields. The use in the crosses of infected original parental forms leads to obtaining the obviously sick hybrids that does not allow to evaluating them objectively for their main economic characteristics and properties. To use in the breeding of hybrid fund raspberry sanitized from malicious viruses it is necessary to develop a program of creation of the highest quality gene pool in each breeding and nursery center and to observe it strictly. The main points of this program are the use in crossings only parents which are free from malicious viruses; regular retesting of the original forms; carrying out of hybridization in the conditions of the closed ground, excluding flight of insects and uncontrolled movement of the contaminated pollen or placement of healthy parental forms on separate plots with spatial isolation 1.5-2km; sanitation of valuable parental forms; immediate removing of highly susceptible genotypes from the sites. A reliable way to increase resistance to infection with malicious viruses is genetic stability. These tasks can be solved only with an integrated approach. For this purpose it is necessary to create contemporary breeding and nursery centers where joint researches of breeders, virologists and nursery workers will be conducted, №2 (2018)

11. Неравномерное окрашивание плодов томата: вероятные причины и пути преодоления

Исследования проводили в 2015–2016 годах в Московской области. Были оценены 125 коммерческих гибридов томатов отечественной и зарубежной селекции. Цель работы – определение причины явления неравномерного окрашивания плодов. Проведенные исследования выявили, что в результате неравномерного окрашивания плодов томата помимо товарных качеств, резко ухудшаются и диетические качества плодов. Установлено, что вне зависимости от этиологии заболевания, приводящего к проявлению симптомов «неравномерного окрашивания плодов», ценность таких плодов резко снижается не только из-за чисто визуальной непривлекательности, но и в связи с уменьшением содержания полезных для здоровья человека веществ. Причем максимальное снижение наблюдается именно по самому ценному в плане пользы для здоровья человеческого организма пигменту – ликопину. Отмечено двукратное снижение содержания антиоксиданта ликопина в плодах с симптомами неравномерного окрашивания. Идентификация вирусов методами ИФА и ПЦР в тканях плодов с симптомами обесцвечивания позволили выделить такие вирусы как TMV, ToMV, TSWV и различное их сочетание. Показана стабильная эффективность препарата Фармайод против синдрома неравномерного окрашивания плодов томата. Можно предположить, что имеется сортовая реакция не только по интенсивности развития симптомов неравномерного окрашивания плодов, но и по чувствительности к лечебному эффекту применения Фармайода. Анализ плодов с нормальной окраской, взятых с растений, на которых ранее были отмечены симптомы неравномерного окрашивания, после двух обработок Фармайодом, показал присутствие в плодах вирусов TMV и ToMV. На дату проведения первой обработки плоды с неравномерной окраской отмечали на I и II кистях на всех трех гибридах. Средняя доля (%) поражения на гибриде F1 Океан составила 22,3%, на F1 Алая Каравелла − 35%, на F1 Тривет – 51,9%. По учетам на I–VI кистях после третьей обработки симптомы обесцвечивания наблюдали лишь на единичных плодах: у F1 Океан − 10%, у F1 Алая Каравелла − 2%., Studies were conducted in 2015-2016 years in the Moscow region. 125 commercial hybrids of tomatoes of domestic and foreign selection were evaluated. The purpose of the work is to determine the cause of the phenomenon of uneven staining in fruits. The carried out researches have revealed that as a result of uneven staining of tomato fruits, in addition to commercial qualities, the dietary quality of fruits also sharply worsens. It is established that regardless of the etiology of the disease, which leads to the manifestation of the symptoms of "uneven staining of fruits," the value of such fruits is sharply reduced not only because of purely visual unattractiveness, but also because of the decrease in the content of substances beneficial to human health. And the maximum reduction is observed for the most valuable in terms of health benefits of the human body pigment - lycopene. There was a twofold decrease in the content of lycopene antioxidant in fruits with symptoms of uneven staining. Identification of viruses using ELISA and PCR in tissues of fetuses with symptoms of discoloration made it possible to isolate such viruses as TMV, ToMV, TSWV and their various combinations. The stable efficacy of Farmayod against the syndrome of uneven staining of tomato fruit is shown. It can be assumed that there is a varietal reaction not only in terms of the intensity of the development of symptoms of uneven staining of fruits, but also in sensitivity to the therapeutic effect of the application of Farmayod. Analysis of fruit with normal color taken from plants that previously had symptoms of uneven staining, after two treatments by Farmayod, showed the presence of TMV and ToMV in the fruit. On the date of the first treatment, fruits with uneven coloring were noted on I and II brushes on all three hybrids. The average percentage (%) of damage on the F1 Ocean was 22.3%, on F1 Alaya Caravel- 35%, on F1 Trivet - 51.9%. According to the accounts on I-VI brushes after the third treatment, the symptoms of discoloration were observed only on single fruits: F1 Ocean - 10%, F1 Alaya Caravel - 2%., №4 (2018)

- Abstract
Antibodies (ABs) that target autoantigens were more abundant in the blood of humans and animals suffering from certain autoimmune and viral diseases than in the blood of healthy donors. The emergence of ABs with diverse types of catalytic activity is among the earliest manifestations of certain autoimmune diseases. The putative mechanisms that underlie the accumulation of autoantibodies and abzymes in different autoimmune diseases are addressed in the present review. The extraordinary diversity of abzymes with various types of catalytic activity is discussed.

- Abstract
The epidemiological HIV infection situation in the Ukraine in the past 4 years is analyzed. In patients with AIDS, the clinical manifestations of tuberculosis are atypical; prolonged intoxication, negative tuberculin tests, no bacterial isolation in half the cases. These patients should be treated with 4 - 5 antituberculous drugs, by taking into account the fact that only long-term continuous therapy may yield a positive effect.

- Abstract
Altogether 155 patients with a newly detected positive reaction to HIV (a human immunodeficiency virus) were investigated in the Republic of Burundi. Chest x-ray was done in 80 of them. Pulmonary tuberculosis was diagnosed in 2 of them, pneumonia (chronic, interstitial and bronchial)--in 15. Enhancement and deformity of lung marking were detected in 45 patients (coincidence with clinical signs of acute bronchitis was found but in 5 of them). A conclusion has been made of interstitial pneumonias being typical of HIV-infected patients and of frequent enhancement of lung marking in the preclinical stage of AIDS.
