ABSTRACT The objective of this study was to identify differentially expressed genes (DEG) between females and males in early bovine embryos. Superovulated cattle (n = 4) in the age span of 16–18 months were artificially inseminated with semen from the same bull that has been proven to be fertile. Blastocysts were collected by routine non-surgical uterine flushing on day 7 after insemination. This study determines the sex of embryos using a micro-injection pipette to collect a few blastomeres through the Zona pellucida. The remaining blastomeres were used for single-cell RNA sequencing (scRNA-seq) analysis on the Illumina platform, followed by differential expression analysis, Gene Ontology (GO) function analysis, and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis. Modeling and simulation of pick cutting hard concretion coal process are taken as a key link to analyze the stress variation law. To this aim, the properties of hard concretion coal in the target mining area were tested. Related model with high-quality common grid was generated using the mapping method through Hypermesh. the coal breaking process was performed using LS-DYNA, and the stress variation law of coal and hard concretion was analyzed. The results showed that the closer the action position to the middle point of long axis of hard concretion, the greater stress peak would be generated, stripping of the pick occurred when it acted on the 2/5 and 1/2 positions of the long axis. Using similarity theory, the model of hard concretion coal was made and the pick cutting experiment was performed. Compared with simulation torque result, the correctness of modeling and simulation has been verified.

Bioactive peptides have been extensively applied for developing health-promoting foods. To improve the value of Laminaria japonica proteins (LJPs), anti-hypertensive peptides were prepared by using controlled enzymatic hydrolysis along with response surface methodology to improve their yields. Eight anti-hypertensive peptides with tyrosine (Tyr) at the C-terminal were determined through an efficient RP-HPLC method and molecular docking was performed to reveal the underlying mechanisms behind their effectiveness of anti-hypertension. According to the results, the strongest activity of hydrolysates was achieved under enzyme ratio (papain: alcalase: trypsin) 1:2:1, pH 8.0, temperature 60°C, substrate concentration 1.7%, enzyme addition 4.4%, and hydrolysis time 5.1 h. Furthermore, molecular docking results displayed that anti-hypertensive peptides could bind with ACE active sites through forming a stable composite structure, thus causing a significant reduction to activities. These results demonstrated the potential applications of LJPs as a source of anti-hypertensive peptides and provided a reference for the industrialized anti-hypertensive productions.
