Uno de los retos del sector de vacuno de carne es producir carne de calidad lo que implica que presente una cantidad suficiente de grasa intramuscular ya que influye de forma positiva en las propiedades sensoriales de la carne. Los mecanismos implicados en el desarrollo del tejido adiposo intramuscular no se conocen en su totalidad, por lo que el estudio de los factores involucrados en la miogénesis podría ayudar a entender mejor el desarrollo de la grasa intramuscular ya que está constatado que existe una competencia por la determinación celular a partir de las Células Madre Mesenquimales entre miocitos, adipocitos y fibroblastos. En este Trabajo Fin de Máster se ha puesto a punto la metodología, utilizando la PCR a tiempo real, para analizar el nivel de expresión génica de los principales Factores de Regulación Miogénica (MRFs): myogenic factor-5 (MYF5), myogenic differentiation (MYOD) y myogenin (MYOG). De esta forma, el estudio de los niveles de ARN mensajero podría dar lugar a la identificación de los elementos implicados en el desarrollo del tejido muscular. Por otro lado, se ha realizado una primera experiencia en la que mediante PCR a tiempo real se cuantifica el nivel de expresión de los MRFs en cuatro músculos (Longissimus dorsi, Semitendinoso, Masseter y Sternomandibularis) en terneros de la raza Pirenaica. Los resultados muestran la existencia de diferencias significativas en el nivel de expresión de estos factores de transcripción en los músculos, que podrían relacionarse con las características específicas de cada uno en relación con el contenido de grasa, fibras musculares y la actividad metabólica. Los músculos que presentan un menor contenido de grasa, Longissimus dorsi y Semitendinosus, cuentan con un mayor nivel de expresión de los MRFs. Por el contrario, el músculo Masseter cuenta con niveles de expresión menores de estos factores de transcripción ya que al tener un mayor contenido de fibras de tipo oxidativo presenta una mayor deposición de grasa One of the challenges of the beef cattle industry is to produce meats of high eating quality, which means that the meat must contain enough amount of intramuscular fat, as it positively influences sensory properties of the meat. In this work it has been optimized the methodology, using real-time PCR, to analyze the gene expression levels of the main Myogenic Regulatory Factors (MRFs): myogenic factor-5 (MYF5), myogenic differentiation (MYOD) and myogenin (MYOG). Thus, the study of mRNA levels might lead to the identification of factores involved in the muscle tissue development. On the other hand, a first experiment using real-time PCR has been performed and quantified the level of expression of the MRFs in four muscles (Longissimus dorsi, Semitendinosus, Masseter and Sternomandibularis) in Pyrenean breed calves. The results showed significant differences in the expression of these transcription factors in the four muscles. This fact might be related to the specific characteristics of each muscle in relation to the fat content, muscle fibers and metabolic activity. Muscles with a lower fat content, Longissimus dorsi and Semitendinosus, presented a higher level of expression of MRFs. Bartonellosis and rickettsiosis are commonly reported in Peru. In order to detect Bartonella sp. and Rickettsiasp. in fleas, ticks and lice, specimens from five distinct locations in Peru (Marizagua, Cajaruro, Jamalca, Lonya Grande and El Milagro) were collected and screened for the presence of these bacteria using PCR and later confirmation by DNA sequencing. The specimens collected were distributed in 102 pools (76 Ctenocephalides felis, 2 Ctenocephalides canis, 16 Pulex irritans, 5 Pediculus humanus, 2 Rhiphicephalus sanguineus, and 1 Boophilus spp.), where Bartonellawas detected in 17 pools (6 of C. felis, 9 of P. irritans, 1 of C. canis, and 1 P. humanus). Also, Rickettsiawas detected in 76 pools (62 C. felis, 10 P. irritans, 2 P. humanus, and 2 C. canis). Bartonella clarridgeiaewas detected in C. felis, C. canisand P. irritanspools at 5.3%, 50% and 12.5%, respectively.Bartonella rochalimaewas detected in one C. felisand two P. irritanspools at 1.3% and 12.5%, respectively. Furthermore, B. henselaewas detected in one C. felispool and one P. humanuspool corresponding to 1.3% and 20%, respectively; and Bartonella spp.was also found in 5 pools of P. irritansat 31.3%. Additionally, R. feliswas detected in C. felis, C. canisand P. irritanspools at 76.3%, 100% and 37.5%, respectively; and Rickettsia spp. was detected in C. felis, P. irritansand P. humanuspools at 5.3%, 25% and 40%, respectively.
