La enfermedad de Gumboro, causada por el virus de la enfermedad infecciosa de la bolsa (IBDV), es una enfermedad aguda, altamente contagiosa y de distribución mundial que afecta a pollos jóvenes. IBDV produce depleción de los linfocitos B que maduran en la bolsa de Fabricio (BF) por lo cual ocasiona atrofia de dicho órgano e inmunosupresión; y, de acuerdo a la cepa viral, una alta mortalidad en pollos jóvenes. En este trabajo de tesis se implementó la metodología de obtención de virus fowlpox recombinantes (FWPVr) y se obtuvo un inmunógeno basado en FWPVr que expresa como antígeno de interés la proteína VP2 de IBDV, principal antígeno viral inductor de respuestas inmunes protectoras. En primer lugar, se diseñó y construyó un vector de transferencia que permite obtener FWPVr por intercambio alélico con el gen fp9.030, como sitio blanco de inserción. Con este vector se obtuvieron los virus recombinantes FW-VP2 que portan y expresan la secuencia codificante de la proteína VP2. Mediante la evaluación de curvas de crecimiento viral in vitro se demostró que la inserción de genes foráneos interrumpiendo el gen fp9.030 no afecta la capacidad replicativa del virus recombinante. Posteriormente, se evaluó la protección conferida por una única inmunización con FW-VP2 por vía subcutánea o por vía intramuscular en pollos SPF (certificados como libres de patógenos específicos). Se observó que la aplicación de una dosis de FW-VP2 por vía subcutánea fue suficiente para reducir significativamente la atrofia de la BF detectándose sólo daños histopatológicos leves. Por otro lado, una única dosis de FW-VP2 aplicadopor vía intramuscular indujo protección frente al desafío con IBDV ya que se observó una reducción significativa del título del virus de desafío presente en las BF y un daño histopatológico leve a moderado en dicho órgano. Luego, el candidato vacunal FW-VP2 se evaluó en esquemas de inmunización de dosis múltiples. El esquema de vacunación de dos dosis con FW-VP2 redujo significativamente la atrofia de la BF (evaluada por el índice bursal y el daño histopatológico) y el título de IBDV en la BF. La aplicación de una tercera inmunización aumentó significativamente la cantidad de anticuerpos anti-VP2 circulantes pero no mejoró los parámetros de protección inducidos con dos inmunizaciones. Finalmente, se confirmó que las inmunizaciones con FW-VP2 inducen una potente respuesta anti-vector que impiden la formación de las lesiones nodulares características de las vacunas de FWPV (“toma de la vacuna”) y, subsecuentemente, el efecto de aplicación de las dosis de refuerzo (booster). En conjunto, los resultados demuestran que se implementó exitosamente la metodología de obtención de FWPV recombinantes y se confirmó que los virus FW-VP2 inducen protección frente al desafío con una cepa de virulencia intermedia de IBDV. A futuro, se evaluará la protección inducida por el candidato vacunal FW-VP2 frente a aislamientos de campo de IBDV y FWPV, con el propósito de desarrollar una vacuna bivalente para el sector avícola. Gumboro´s disease, caused by infectious bursal disease virus (IBDV), is an acute, highly contagious disease with worldwide distribution that affects young chickens. IBDV produces depletion of B lymphocytes that mature in the bursa of Fabricius (BF), causing atrophy of this organ and immunosuppression; and, according to the viral strain, a high mortality in young chickens. In this thesis, the methodology for obtaining recombinant fowlpox virus (FWPVr) was implemented and an immunogen based on FWPVr expressing VP2 protein of IBDV, the main viral antigen that induces protective immune responses, was obtained. First, a transfer vector to obtain FWPVr by allelic exchange with the fp9.030 gene (target insertion site) was designed and constructed. With this vector, the recombinant FW-VP2 viruses carrying and expressing the coding sequence of VP2 protein were obtained. Through the evaluation of viral growth curves in vitro, it was demonstrated that the insertion of foreign genes interrupting the fp9.030 gene does not affect the replicative capacity of the recombinant virus. Subsequently, the protection conferred by a single immunization with FW-VP2 was evaluated by subcutaneous or intramuscular injection in specific pathogen-free chickens. It was observed that the application of a subcutaneous dose of FW-VP2 was enough to significantly reduce the atrophy of the BF because only mild histopathological lesions were detected. On the other hand, a single intramuscular dose of FW-VP2 induced protection against the challenge with IBDV since ir was observed both a significant reduction of the challenge virus titer in BF and mild to moderate histopathological lesions in this organ. Then, the vaccine candidate FW-VP2 was evaluated in multiple dose immunization schemes. The two-dose vaccination scheme with FW-VP2 significantly reduced BF atrophy (assessed by bursal index and histopathological lesions) and the IBDV titer in BF. The application of a third immunization significantly increased the amount of circulating anti-VP2 antibodies but did not improve the protection parameters induced with two immunizations. Finally, it was confirmed that immunizations with FW-VP2 induce a potent anti-vector response that prevents the formation of the nodular lesions characteristic of the FWPV vaccines ("vaccine takes") and, subsequently, the effect of application of the booster dose. Overall, the results show that the methodology for obtaining recombinant FWPVs was successfully implemented and it was confirmed that FW-VP2 viruses induce protection against challenge with a strain of IBDV with intermediate virulence. In the future, the protection induced by the vaccine candidate FW-VP2 will be evaluated against IBDV and FWPV field strains, with the purpose of developing a bivalent vaccine for the poultry industry.
