An indirect ELISA against capsular polysaccharide of Salmonella typhi was developed to evaluate the immune response to the Polysaccharide Vi vaccine ) vaccine against the typhoid fever. The polystyrene plates were first coated with Poly-I-Lysine (Sigma) and polysaccharide antigen purified in the Finlay Institute. Then the plates were incubated with human serum samples. Antibodies anti polysaccharide Vi were detected by addition of anti-human Polyvalent Immunoglobulins / alkaline phosphatase conjugate (Sigma), and the reaction was developed by the substrate specific degradation (p-Nitrophenyl Phosphate). The detection limit of the assay was 0,045 Units of Haemagglutination per mL of specific immunoglobulins in human serum. Intra and inter assay unprecision were below 10%.
