- Abstract
Objective: Stem cell transplantation is considered to be one of the available treatments for malign or hereditary blood diseases and bone marrow failure. Peripheral blood progenitor cells (PBPC) are widely used for this technique. Recently a new type of stem cell with a pluripotent potential has been identified. These cells, called very small embryonic-like (VSEL) stem cells, are thought to be found in peripheral blood (PB) in adult individuals. The aim of this study was to obtain and identify VSEL stem cells using a peripheral blood source. Material and Method: VSEL cells were isolated from mononuclear and erythrocyte layers obtained by using lysis and ficoll gradient methods from the materials taken from donors. The presence of NANOG, OCT4, SSEA-4 and CXCR-4 embryonal stem cell markers by flow cytometry and immunofluorescence staining was investigated. The presence of NANOG and OCT4 proteins was investigated using the Western blot method. Results: According to the flow cytometry results, the number of cells carrying the VSEL marker was higher in the debris layer than in the CD45-population. Western and immunofluorescence results showed high levels of OCT4 and NANOG expression. It was also determined that these proteins are present in both the cytoplasm and the nucleus. Conclusions: High expression of these proteins, known as pluripotent stem cell markers, in adult peripheral blood brings to mind the question of how they work in differentiated tissues. These findings strengthen the thesis of a new stem cell population that have pluripotent markers in peripheral blood and provide the basis for future clinical studies about this point. Son yıllarda, zeytinyağının faydalı etkilerinin sadece yüksek oleik asit içeriğiyle ilişkili değil, aynı zamanda içerdiği fenolik bileşenlerin antioksidan aktivitesiyle de ilişkili olduğu kanıtlanmıştır. Bu çalışmada, sızma zeytinyağı fenolik ekstraktının (ZFE), insan periferik kan mononükleer hücrelerinin (PKMH) nükleer ve mitokondriyal DNA'sındaki bazal DNA hasarı ve oksidatif stres kaynaklı DNA hasarı üzerindeki etkisi incelendi. Hücreler, 35 yaşındaki sağlıklı iki erkek ve iki kadın gönüllünün kan örneklerinden izole edildi. Hücrelere diyetle alınabilecek ve sitotoksik etkisi olmayan ZFE konsantrasyonları ile ön-uygulama yapıldı. Hücreler üzerinde oksidatif stres oluş-turmak için 30 dakika hidrojen peroksit (H2O2, 200 µM) ile uygulama yapıldı. DNA hasarları gene-özgü QPCR yöntemi ile ölçüldü. Hücrelere ZFE ile ön uygulama yapılması her iki genomdaki bazal DNA hasar seviyesi üzerin-de olumsuz bir etkiye neden olmadı. H2O2 ile oluşturulan oksidatif stres, nDNA (nükleer DNA) (1.43 hasar/10kb) ile karşılaştırıldığında mtDNA'da (mitokondriyal DNA) (2.76 hasar/10kb) iki kat daha fazla hasara neden oldu. Bununla birlikte, oksidatif stres öncesi ZFE ile ön-uygulama yapılması hem mtDNA hem de nDNA üzerinde koru-yucu etki göstermedi. Sonuçta, sızma zeytinyağı tüketimi sonrası doku ve vasküler sistemde bulunabilecek iki farklı fenolik ekstrakt konsantrasyonunun, insan PKMH’lerde oksidatif DNA hasarlarını önleyici bir etkiye sahip olmadığı belirlendi. Bu sonuçlar, zeytinyağının sindirim sistemi üzerinde daha fazla sahip olduğu bilinen faydalı etkisinin tüketiminden hemen sonraki daha yüksek konsantrasyonlarından kaynaklanabileceğini işaret etmektedir

Amaç: Lizozomal bir enzim olan alfa-naftil asetat esteraz (ANAE) enzimi pek çok türde periferik kan yaymalarında T-lenfosit, B-lenfosit ve monositlerin birbirlerinden ayırt edilmelerinde kullanılmaktadır. Bu araştırmada, Topoz olarak da bilinen Kırgızistan dağ yaklarında (Bos grinniens) periferik kan lökositlerinin ANAE enzimi pozitivitelerinin belirlenmesi amaçlandı. Gereç ve Yöntem: Bu çalışmada materyal olarak 13 adet 2–4 yaşlı sağlıklı Kırgız Dağ Yak'ından alınan periferik kan örnekleri kullanıldı. Örneklerden hazırlanan frotilerde ANAE demonstrasyonları gerçekleştirildi. Bulgular: ANAE pozitivitesi tüm monositlerde ve sadece bazı lenfositlerde gözlendi. Lenfositlerdeki pozitivite 1-4 adet kırmızı-kahverengi sitoplazmik granüllerle karakterize nokta tarzındayken monositlerin tamamı tüm sitoplâzmaları boyunca yaygın, ince taneli bir enzimatik reaksiyona sahipti. Nötrofil ve eozinofil lökositlerde ise enzimatik reaksiyon gözlenmedi. Kırgız dağ yaklarında ortalama ANAE pozitif lenfosit oranının %58,85±1,38 olduğu tespit edildi. Öneri: Kırgızistan dağ yakları hakkında yapılan çalışmalar söz konusu hayvanların yaşam koşulları ve sert mizaçları nedeniyle son derece sınırlı kalmıştır. Dolayısıyla bu hayvanlar üzerinde yapılacak olan her türlü yapısal, histolojik, enzim histokimyasal, fizyolojik, biyokimyasal ve anatomik çalışmaların yanı sıra ülkede görev yapan veteriner hekim klinisyen ya da akademisyenlerin karşı karşıya kalabilecekleri çeşitli hastalıkların olgu sunumlarının, ilerleyen yıllarda bu hayvanlar üzerinde yapılacak olan çalışmalara yön vereceği düşünülmektedir., Aim: A lysosomal enzyme, alpha naphthyl acetate esterase (ANAE), has been used to differentiate T and B lymphocytes, and monocytes in various species in peripheral blood smears. In this study, it was aimed to determine the positivity of the ANAE in peripheral blood leukocytes of Kyrgyzstan's mountain yaks, as also known Topoz,(Bos grinniens). Materials and Methods: As a material, the peripheral blood samples collected from healthy thirteen Kyrgyzstan's mountain yaks, 2–4 year old aged, were used. ANAE was demonstrated on smears prepared from blood samples. Results: ANAE-positivity was observed in all monocytes and only some lymphocytes. ANAE-positivity pattern in lymphocytes was dot like characterized with 1-4 reddish-brown cytoplasmic granules while all monocytes have a diffuse, fine granular enzymatic reaction along whole cytoplasm. No enzymatic reaction was observed in neutrophil and eosinophil leukocytes. The ANAE positive lymphocyte rate was found to be 58,85 % in Kyrgyzstan's mountain yaks. Conclusion: Studies on Kyrgyzstan mountain yaks have been extremely limited due to their living conditions and aggressive temperaments of these animals. Therefore, any kind of structural, histological, enzyme histochemical, physiological, biochemical and anatomical studies on these animals, as well as case reports of various diseases that may be faced by veterinary clinicians or academicians working in the country, will guide the studies to be carried out on these animals in future.

In flow cytometry; FS, SS and through the gate of CD45, it is possible to obtain a good knowledge about the characteristic of bone marrow cellular components. it is very important to understand the structure of normal bone marrow for the identification of pathological conditions. As a result, to recognize healthy bone marrow and blood samples correctly by flow cytometry enables us to make better immunophenotyping., Özet Akım sitometrik analiz ile daha doğru sonuçlara ulaşabilmek için kemik iliği ve periferik kan hücrelerinin fiziksel morfolojisinin iyi bilinmesi gereklidir. Akım sitometride; FS, SS ve CD45 kapısı ile kemik iliği hücresel bileşenleri ve onların karakterleri hakkında iyi bir bilgi edinmek mümkündür. Normal kemik iliği yapısını anlamak, patolojik durumların tanımlanmasında oldukça önemlidir. 6. The in vitro determination of genotoxicity in peripheral lymphocytes of welders exposed to fumes from metal arc welding

Gas metal arc welding is a widely used method in many industrial areas. In this study, the peripheral blood lymphocytes of 23 welders and 25 nonexposed subjects were monitored for genotoxicity. Materials and methods: The blood samples obtained from the subjects were incubated with a culture medium at 37 °C for 72 h. Cytochalasin-B and BrdU were added for micronucleus and replication index. After incubation, the cultures were harvested, stained, and examined. Results: The micronucleus rate (2.165 ± 1.645) of the welders was quite higher than that of those not exposed (1.352 ± 0.978) (P < 0.01). There was a significant difference between micronucleus rates of smoking and nonsmoking subjects. It was found that as the time of exposure to welding fumes increased, so did the micronucleus rates (2.938 ± 1.723) (P < 0.05). A positive correlation was observed between micronucleus frequency and age. The replication index rate (1.026 ± 0.020) of the welders was higher than that of those not exposed (1.019 ± 0.016) (P = 0.01). Conclusion: It is advised that welders who work with gas metal arc welding be instructed and that necessary protective measures be taken because of the risk caused by metal arc welding fumes.

WOS: 000328083200020, This study was performed to determine the effects of pregnancy on the proportion of T-lymphocytes, null lymphocytes and acid phosphatase (ACP-ase) positive peripheral blood lymphocytes and the distribution of the mentioned cells in the decidua basalis region of endometrium in the pregnant mice. For this purpose, mice at 12-14 weeks of age were divided into four groups as non-pregnant control, and at the middle of the first, second, and the third week of the pregnancy, corresponding to early, middle, and late (3rd, 10th, and 17th days of pregnancy) gestational stages respectively (n = 6 for each group). The lowest T-lymphocytes percentage was determined at early and middle pregnancy in the peripheral blood (43.83%) and decidua basalis (10.83 number/0.1 mm(2)), respectively. There was a statistically significant increase in the proportions of the peripheral blood ACP-ase (+) lymphocytes (44.33%) at the mid-gestational period while the lowest ACP-ase positive lymphocyte numbers (5.50 number/0.1 mm(2)) in the decidua basalis was observed at early pregnancy. The highest null cell rates were found at early gestation either in the peripheral blood (11.50%) or in the decidua basalis (7.83 umber/0.1 mm(2)). The lowest percentage of peripheral blood lymphocyte (56.00%) was recorded at the early pregnancy. It was concluded that the number and the distribution of the lymphocyte, T-lymphocyte, null lymphocyte, and ACP-ase positive lymphocyte were affected by pregnancy although there were some differences among the gestational periods.

Memelilerde perifer kanda, vücut sıvılarında ve meme bezi dokusunda yabancı mikroorganizmalara ve cisimlere karşı organizmanın Celluler Savunma görevini üstlenen hücreler Polymorphonuclear Leukocyte 'ler, Macrophage 'lar ve Lenfocyt nelerdir. Normal inek sütlerinde sayısal değer açısından Macrophage 'lar ilk sırayı alırlar. Acut mastitisli inek sütlerinden ilk değişme PMNL 'lerin infiltrasyonu dahi artış tır. ilerlemiş mastitis vakalarında ise sütte her üç hücre sayısında da büyük artışlar gözlenir. Mastitisli sütlerde özellikle PMNL'lerin fagositoz aktivitelerini baskılayan faktörler vardır. Bu faktörler sütteki Lipid-Casein seviyesinin yüksekliği immunoglobulin eksikliği ile ortaya çıkan Opsonization yetersizliği şeklinde karakterize edilebilir. The cells being charged with immunocellular defense against all kinds of Foreign microorganisms and foreign bodies in the blood and body fluids of mammalian animals are mainly macrophages, polymorphonuclear leukocytes (PMNL) and lymphocytes. The macrophages are the most common cells seenin the normal cow milk. The earliest sign of increase in the number of inflammatory cells is noted in the amount of PMNL, in the milk obtained from acutely infected cows. However the number of all the three types of cells are increasing with the time elapsed from infection. There are several factors inhibiting the phagocytic activity of PMNL. These factors characterized the increase in the level of Lipid-Casein in the mi/k, and decreased opsonization potential due to immunoglobulin deficiency.
