Steroidlerin tayini genel olarak immunoassay yöntemler ve kromatografik yöntemlerle yapılmaktadır. Söz konusu yöntemler zaman alıcı ön işlemler ve pahalı ekipmanlar içerir. Ancak mikrobiyal biyosensörler ve moleküler damgalanmış polimer (MIP) tabanlı elektrokimyasal sensörlerin kullanılmasıyla hızlı, duyarlı, ucuz ve seçimli steroid tayini yapılabilir. Bu tez çalışmasında kortizol-kortizon ve prednisolona yönelik MIP sensörleri, kolesterole yönelik ise mikrobiyal biyosensör sistemi geliştirilmiştir. MIP sensörleri altın nanopartikül kaplanmış grafit elektrot yüzeyinde pirolün kalıp molekül varlığında elektropolimerizasyonu yöntemi ile hazırlanmıştır. Sensör cevapları Diferansiyel Puls (DP) yöntemi ile Ferri/Ferro çözeltisinden izlenmiştir. Kortizon-kortizol ve prednisolona yönelik geliştirilen MIP sensörlerinde kalıp molekül/monomer oranı, inkübasyon ve ekstraksiyon süresi optimizasyonları yapılmıştır. Kortizon-MIP sensörünün doğrusal tayin aralığı 1-100 μM, tayin limiti 0,25 μM olarak bulunurken Prednisolon-MIP sensörünün doğrusal tayin aralığı 1-75 μM, tayin limiti ise 0,35 μM olarak bulunmuştur. Tekrarlanabilirlik denemesinde Kortizon-MIP sensörünün 50 μM kortizon için (n=10) ortalama (¯x), standart sapma (±S.S.) ve % varyasyon katsayısı (%VK) değerleri sırasıyla 48,10 μM, ±2,42μM ve %5,04 olarak bulunurken, 50 μM prednisolon için (n=10) ortalama (¯x), standart sapma ve % Varyasyon katsayısı değerleri sırasıyla 48,69 μM, ±1,06 μM ve %2,19 olarak bulunmuştur. MIP sensörleri ile substrat spesifikliği, girişim etkisi ve depo kararlılığı denemeleri yapılmıştır. Ayrıca geliştirilen MIP sensörleri ile liyofilize idrar ve serum örneklerinde kortizol ve prednisolon tayini yapılmıştır. Mikrobiyal biyosensör geliştirilmesi aşaması kapsamında biyoalgılayıcı olarak kolesterol metabolizması indüklenmiş ve organik çözgene dayanıklı Pseudomonas sp. türü bakteriler ve katalaz enzimi kullanılmıştır. Pseudomonas sp. ve katalaz enzimi çok duvarlı karbon nanotüp (MWCNT)/iyonik sıvı (IL)/karbon pasta elektrot (CPE) içerisine immobilize edilmiştir. Biyosensörün immobilizasyon aşamaları CV ve impedans spektroskopisi ile karakterize edilmiştir. Ölçümlerde mediyatör sistemi olarak hidrokinon kullanılmıştır. Amperometrik çalışma potansiyeli -0,2 V olarak bulunmuştur. Optimizasyon ve karakterizasyon çalışmaları amperometrik yöntem ile yapılmıştır. Mikrobiyal biyosensörün çalışma koşullarının optimizasyonu denemeleri yapılmış ve optimum sıcaklık 35 °C, optimum pH : 7,0 (Fosfat tamponu, 50 mM) olarak bulunmuştur. Bakteri miktarının biyosensör cevabı üzerine etkisi incelenmiştir. Kolesterol tayinine yönelik olarak geliştirilen bu mikrobiyal biyosensörün doğrusal tayin aralığı 5-600 μM tayin limiti ise 1,52 μM olarak bulunmuştur. Substrat spesifikliği ve girişim etkisi denemelerinde kortizon, kortizol, testosteron, β-sitosterol ve östradiol gibi steroid bileşiklerle glukoz, askorbik asit gibi endojen analitlerin varlığının biyosensör cevabına etkisi incelenmiştir. Optimizasyon ve karakterizasyon çalışmaları tamamlanan mikrobiyal biyosensörün doygun siklohegzandaki kolesterol cevabı incelenmiş ve sulu ortamla karşılaştırılmıştır. Geliştirilen mikrobiyal biyosensörle liyofilize serum örneklerinde hem sulu fazda hem de siklohegzan fazında kolesterol tayini yapılmıştır., The determination of steroids is generally performed by immunoassay methods and chromatographic methods. Such methods include time consuming preprocessing and expensive equipment. However, using microbial biosensors and MIP based electrochemical sensors, rapid, sensitive, inexpensive and selective steroid detection can be performed. In this thesis, MIP sensors for cortisol-cortisone and prednisolone and microbial biosensor system for cholesterol have been developed. MIP sensors were prepared by electropolymerization method in the presence of pyrole and template molecule on gold nanoparticle coated graphite electrode surface. Sensor responses were monitored by Ferri / Ferro solution with Differential Pulse (DP) method. Template molecule / monomer ratio, incubation and extraction duration optimizations were performed in the MIP sensors that developed for cortisone-cortisol and prednisolone. The linear detection range of the Cortisone-MIP sensor was found to be 1-100 μM and the detection limit 0.25 μM, while the linear detection range of the Prednisolon-MIP sensor was found to be 1-75 μM and the detection limit 0.35 μM. In the reproducubility experiments average value (x), standard deviation (±) and variation coefficient (CV %) values for the 50 M cortisone (n = 10) by the Cortisone-MIP sensor were calculated to be 48.10 μM, ± 2.42 M and 5.04%, respectively, and for the prednisolone (n = 10) were found as 48.69 μM, ± 1.06 μM and 2.189% respectively. MIP sensors have been used to test substrate specificity, interferenceeffect, and storage stability. In addition, the developed MIP sensors were tested for cortisol-cortisone and prednisolone in lyophilized urine and serum samples. Within the microbial biosensor development step, cholesterol metabolism induced and organic phase tolerant Pseudomonas sp. and catalase enzyme were used as a biocomponent of biosensor. Pseudomonas sp. and the catalase enzyme were immobilized into multi-wall carbon nanotube (MWCNT) / ionic liquid (IL) / carbon paste electrode (CPE). Immobilization steps of the biosensor were characterized by CV and impedance spectroscopy. Hydroquinone was used as the mediator system in the measurements. The amperometric working potential was found to be -0.2 V. Optimization and characterization studies were also carried out by amperometric method. The optimization of working conditions of microbial biosensor were carried out and optimum temperature and pH were found to be 35 °C and 7.0 (Phosphate buffer, 50 mM). The effect of amounts of bacteria on the biosensor responses was investigated. The linear range of this microbial biosensor that developed for the determination of cholesterol was found to be 5-600 μM and the detection limit was found as 1.52 μM. The effect of steroid compounds such as cortisone, cortisol, testosterone, β-sitosterol and estradiol, and the presence of endogenous analytes such as glucose and ascorbic acid on the biosensor response was investigated in substrate specificity and interference effect experiments. After optimization and characterization studies of the microbial biosensor, the biosensor response to cholesterol in saturated cyclohegzan was investigated and also responses obtained were compared with response obtained by water phase. Cholesterol determination was carried out both in cyclohegzan and in water phase by developed microbial biosensor.

Çalışmamız kalıp molekül kloramfenikol, momomer metakrilik asit ve çapraz bağlayıcı etilenglikoldimetakrilat temelli moleküler baskılanmş polimer hazırlanmasını içerir. Sentezlenen MIP süt numunelerinden kloramfenikol (CAP), florfenikol (FF) ve tiyamfenikol (TAP)’ün LC-MS/MS analizini takip eden katı faz ekstraksiyonu için kullanılmıştır. Moleküler baskılanmış polimerler (MIP’ler) sulu süspansiyon polimerizasyonu ve çöktürme polimerizasyonu metotları ile sentezlendi. Elde edilen moleküler baskılanmış polimer mikro küreleri taramalı elektron mikroskobu (SEM), fourier dönüşümlü kızıl ötesi spektroskopi (FTIR), termogravimetrik analiz (TGA) ile karakterize edildi. CAP bileşiğinin sentezlenen polimer üzerindeki adsorpsiyon davranışı analit konsantrasyonu, polimer-analit etkileşim süresi ve çözelti pH’ı parametrelerini içerecek şekilde optimize edildi. MIP ve kontrol polimeri NIP için bağlanma izotermleri çizildi. Scatchard analizlerinde çöktürme polimerizasyonu ile sentezlenen MIP için spesifik ve spesifik olmayan bağlanma bölgeleri tayin edildi, buna karşın süspansiyon polimerizasyonu ile elde edilen MIP’ler kontrol polimerine göre zayıf adsorpsiyon özellikleri gösterdi. Çöktürme polimerizasyonu ile sentezlenen MIP ve kontrol polimeri NIP için adsorpsiyon özellikleri batch modu deneyleri ile karakterize edildi. Scatchard analizi çıktılarından MIP, yüksek seçici bölgeleri için Qmax = 2,238 mg g-1 ve Kd = 4,156 mg mL-1 değerleri verirken düşük seçici bölgeler Qmax = 3,68 mg g-1 ve Kd = 31,88 mg mL-1 değerlerini gösterdi. Kontrol polimeri NIP, Qmax = 1,238 mg g-1 ve Kd = 20,38 mg mL-1 değerleri ile MIP’e kıyasla daha düşük bağlanma karakteristikleri göstermektedir. CAP bileşiği için baskılama faktörü 3,44 (QMIP/QNIP) olarak tayin edildi ve göreceli baskılama faktörleri QCAP/QFF ve QCAP/QTAP sırasıyla 1,22 ve 1,24 olarak belirlendi. Sentezlenen MIP, süt numunelerinden fenikol bileşiklerinin katı faz ekstraksiyonunda başarı ile uygulandı. CAP, FF ve TAP fenikolleri ile spike edilen (0,01 – 10 mg L-1) süt numunelerine MIP ile hazırlanmış SPE kartuşlarında katı faz ekstraksiyonu uygulandı. Önceden belirlenmiş SPE şartlarının süt numunelerine uygulanması ile CAP için 82,5±5,6 – 91,8±3,7; FF için 73±5,3 – 83,6±4,8; ve TAP için 71,2±4,1 – 72,6±3,6 aralığında geri kazanım değerleri elde edildi. Kloramfenikol, florfenikol ve tiyamfenikolün süt numunelerinden çöktürme polimerizasyonu ile sentezlenen MIP’e dayalı katıfaz ekstraksiyonunun takip eden LCMS/ MS analizleri başarı ile gerçek numunelere uygulanabilir. abstract Our study involves preparation of molecularly imprinted polymer based on template molecule chloramphenicol, monomer metacrylic acid and crosslinker ethyleneglycoldimethacrylate, synthesized MIP is used for solid phase extraction of chloramphenicol (CAP), florfenicol (FF) and thiamphenicol (TAP) from milk samples followed by LC-MS/MS analysis. Molecularly imprinted polymers (MIP’s) are synthesized by aqueous suspension polymerization and precipitation polymerization methods. Resulting molecularly imprinted polymer microspheres (MIPM’s) were characterized with scanning electron microscope (SEM), fourier transformed infrared spectroscopy (FTIR) and thermogravimetric analysis (TGA). Adsorption behavior of CAP on synthesized polymers were optimzed including parameters concentration of analyte, polymer-analyte contact time and pH of solution. Binding isotherms for MIP and control polymer NIP are plotted. According to the Scatchard plot analysis specific and non-specific binding sites were determined for precipitation polymerization synthesized MIP while suspenson polymerization resulted MIP showed weak adsorption properties compared with control polymer. MIP synthesized by precipitation polimerization and its control polymer NIP’s adsorption properties were characterized with batch mode experiments. Outcomes from Scatchard plot analysis of MIP gives Qmax = 2,238 mg g-1 and Kd = 4,156 mg mL-1 values for high affinity sites whereas low affinity sites showed Qmax = 3,68 mg g-1 and Kd = 31,88 mg mL-1. Control polymer NIP gives Qmax = 1,238 mg g-1 and Kd = 20,38 mg mL-1 values showing low binding characteristics with respect to the MIP. Imprinting factor (IF) for template compound CAP was defined as 3,44 (QMIP/QNIP) and relative imprinting factors in optimized batch mode experiments QCAP/QFF and QCAP/QTAP were 1,22 and 1,24 respectively. Synthesized MIP was applied succesfuly to the solid phase extraction (SPE) of fenicol compounds from milk samples. Milk samples spiked with phenicols CAP, FF and TAP (0,01-10 mg L-1) were subjected to the solid phase extraction with MIP prepared SPE cartridges. Under pre-defined SPE conditions applied to the milk samples, recoveries were between 82,5±5,6 – 91,8±3,7 for CAP, 73±5,3 – 83,6±4,8 for FF and 71,2±4,1 – 72,6±3,6 for TAP compounds. Precipitation polymerization synthesized MIP based SPE extraction of chloramphenicol, florfenicol and thiamphenicol from milk samples followed by LCMS/ MS analysis can be succesfully applied to the real samples.

İnsülin canlı sistemde glukoz düzeyinin kontrolünü sağlayan ve metabolizma üzerinde önemli etkileri olan bir hormondur.İnsülin üretimi veya insülin kullanımı azaldığında diabetik hastalıklar oluşmaktadır. İnsülinin ayrılması, saflaştırılması veya saf proteinler, diabetik ilaçların saf materyallerini oluşturdukları için büyük bir öneme sahiptirler. Bu noktada adsorpsiyon yoluyla proteinlerin saflaştırılması biyomedikal ve biyoteknolojik uygulamalar için oldukça önemlidir. Çalışmada insülin adsorpsiyonu için afinite adsorpsiyon tekniği çalışıldı. Bu amaçla insülin adsorpsiyonu için insülin-baskılı süpermakrogözenekli kriyojel hazırlandı. N-metakriloil-L-histidin metilester (MAH) monomer olarak seçildi. MAH, Hacettepe Ünv. Fen Fak. Kimya Bölümü, Biyokimya Ana Bilim Dalı' nda Prof. Dr. Adil Denizli ve çalışma grubu tarafından metakriloil klorür ve L-histidin metil esterden sentezlendi. İlk aşamada insülin MAH ile komplekleştirilerek, insülin baskılı p(HEMA-MAH) kriyojel serbest polimerizasyon reaksiyonu ile polihidroksimetakrilat p(HEMA), N,N,N',N'- tetrametilendiamit (TEMED) ve amonyum persülfat (APS) ile buz banyosunda hazırlandı. Daha sonra 0.1 M Glisin- HCl tamponu (pH: 3.5) ile insülin kriyojelden uzaklaştırıldı. Kriyojel taramalı elektron mikroskobu (SEM), Fourier infrared spektroskopisi (FT-IR) ve şişme testi ile karakterize edildi. P(HEMA) için kriyojelin denge şişme oranı 8.56 g H2O / g polimer ve insülin baskılanmış p(HEMA-MAH) için 7.2 g H2O / g polimer olarak bulundu. İnsülin adsorpsiyon çalışmaları sürekli sistemlerde farklı şartlar altında (akış hızı, ortamın pH'sı, insülin konsantrasyonu, sıcaklık ve iyonik şiddet gibi) araştırıldı. Sulu çözeltide p(HEMA-MAH) kriyojelin, insülin adsorpsiyon kapasitesi 8.13 g H2O / g polimer olarak belirlendi. Adsorpsiyon kapasitesinde anlamlı bir azalma olmaksızın, p(HEMA-MAH) kriyojeli ile insülinin tekrarlanabilir bir şekilde adsorplandığı-desorplandığı gözlendi. Insulin is one of the most important hormone and it has profound effects on metabolism by controlling of glucose homeostasis. When insulin production or the use of insulin ability decrease these disorders cause diabetic illness. Separation and purification of insulin or purified proteins are very important because of to obtain as a purified raw materials of diabetics drugs. Protein separation techniques gain more importance at this point. Protein purification via adsorption is of significant importance in biomedical and biotechnological purposes. In present study, affinity adsorption technique is studied for insulin adsorption. For this aim insulin-imprinted supermacroporous cryogel was prepared for the insulin adsorption. N- methacryloyl- L- histidin methylester (MAH) was choosen as the monomer. MAH was synthesized using methacryloyl chloride and L- histidinemethyl ester by Hacettepe University Faculty of Science Department of Biochemistry Prof. Dr. Adil Denizli?s group. In the first step, insulin was complexed with MAH and insulin-imprinted p(HEMA-MAH) cryogel was prepared by free radical polymerization by polyhydroxymethacrylate p(HEMA), N,N,N?,N?? tetrametiletilendiamin (TEMED) and ammmonium persulphate (APS) in an ice bath. Then, insulin was removed from cryogel by using 0.1 M glycine-HCl buffer (pH: 3.5). Cryogel was characterized by using scanning electron microscopy (SEM), Fourier transfer infrared spectroscopy and swelling test. The equilibrium swelling ratios of the cryogels were 8.56 g H2O/g polymer for p(HEMA) and 7.20 g H2O/g polymer for insulin-imprinted p(HEMA-MAH). Insulin adsorption experiments were carried out under different conditions in continuous system (i.e., flow rate, medium pH, insulin concentration, temperature and ionic strength). Insulin adsorption capacity of p(HEMA-MAH) cryogel from aqueous solution was found to be 8.13 mg/g polymer. It was observed that insulin could be repeatedly adsorbed and desorbed with p(HEMA-MAH) cryogel without any significant loss in the adsorption capacity.
