Abstract

International audience; In this work, we show how optimal control theory can be used to optimize membrane filtration processes. The objective is to determine the optimal switching between filtration and backwash sequences in order to maximize the net water production per membrane area of microfiltration or ultrafiltration systems over a given period of time. The optimal solutions, whether the backwash flux is constant or variable, have been derived for a membrane filtration process operating at constant transmembrane pressure using a class of filtration models defined only by their qualitative behaviors. The optimal solution is applied in a simulation case study by considering a specific model validated on data recovered from the literature. The robustness of the approach has been evaluated for different disturbances.

Abstract

International audience; Second generation anticoagulant rodenticides (SGARs), pesticides used worldwide to control rodent populations, exist in two diastereoisomer chemical species because they own two stereogenic centers. A core-shell LC-MS/MS multi-residue method for comprehensive quantitative analysis of the diastereoisomers of five SGARs as well as three first generation anticoagulant rodenticide molecules has been fully validated in liver of rats according to a bioanalytical guideline. A core-shell column (superficially porous particles) has been chosen for its ability to separate the diastereomers of bromadiolone, difenacoum, brodifacoum, flocoumafen and difethialone and for its robustness to rat liver extracts. The highly selective chromatographic separation of the diastereoisomers contributes to good signal to noise ratios and then enhances the sensitivity of the method compared to the ones of fully porous columns. An elution gradient has been optimized with 10mM ammonium acetate and acetonitrile as aqueous/organic mobile phase respectively. Triple quadrupole mass detector has been used to achieve specifity and LLOQ from 0.92 to 2.2 ng/g for each diastereoisomer, or first generation anticoagulant rodenticides. Then we evidenced diastereoisomeric ratios in liver of rats issued from not controlled exposure of wild rats (Rattus norvegicus) trapped in a French Parisian park through a campaign of rodent eradication. We compared them to diastereoisomeric ratios in SGARs commercial baits that contain both isomers, and showed that one of the two diastereoiomers had nearly disappeared in liver of rats. The proportions of cis-bromadiolone and trans-difenacoum were really lowered compared to the baits: 5/7 and 9/12 rats had only trans-bromadiolone and cis-difenacoum hepatic residues respectively. Liver persistence of the two diastereoisomers of bromadiolone and difenacoum was different due to differences in their pharmacokinetics in wild rats. The new Core-shell LC-MS/MS method is particularly well adapted for further exploration of diastereoisomers ratios in rodent and predatory wildlife biological samples in order to evaluate ecological consequences of actual baits, to explore new formulated baits with a good balance between efficacity (ability to kill rodents) and diastereoisomers persistence, and hopefully to mitigate exposure of non-target species.

Abstract

International audience; Two novel strictly anaerobic bacteria, strains Bs105(T) and Bs107(T), were isolated from a deep aquifer-derived hydrocarbonoclastic community. The cells were rod-shaped, not motile and had terminal spores. Phylogenetic affiliation and physiological properties revealed that these isolates belong to two novel species of the genus Desulfotomaculum. Optimal growth temperatures for strains Bs105(T) and Bs107(T) were 42 and 45 degrees C, respectively. The estimated G+C content of the genomic DNA was 42.9 and 48.7 mol%. For both strains, the major cellular fatty acid was palmitate (C-16 : 0). Specific carbon fatty acid signatures of Gram-positive bacteria (iso-C-17 : 0) and sulfate-reducing bacteria (C(17 : 0)cyc) were also detected. An insertion was revealed in one of the two 16S rRNA gene copies harboured by strain Bs107(T). Similar insertions have previously been highlighted among moderately thermophilic species of the genus Desulfotomaculum. Both strains shared the ability to oxidize aromatic acids (Bs105(T) : hydroquinone, acetophenone, paratoluic acid, 2-phenylethanol, trans-cinnamic acid, 4-hydroxybenzaldehyde, benzyl alcohol, benzoic acid 4-hydroxybutyl ester; Bs107(T) : ortho-toluic acid, benzoic acid 4-hydroxybutyl ester). The names Desulfotomaculum aquiferis sp. nov. and Desulfotomaculum profundi sp. nov. are proposed for the type strains Bs105(T) (= DSM 24088(T) = JCM 31386(T)) and Bs107(T) (= DSM 24093(T) = JCM 31387(T)).

Abstract

In a coastal lagoon with a shallow, 25 m water column off the southwest coast of New Caledonia, large-volume ( ∼ 50 m3) mesocosm experiments were undertaken to track the fate of newly fixed nitrogen (N). The mesocosms were intentionally fertilized with 0.8 µM dissolved inorganic phosphorus to stimulate diazotrophy. N isotopic evidence indicates that the dominant source of N fueling export production shifted from subsurface nitrate (NO3−) assimilated prior to the start of the 23-day experiments to N2 fixation by the end of the experiments. While the δ15N of the sinking particulate N (PNsink) flux changed during the experiments, the δ15N of the suspended PN (PNsusp) and dissolved organic N (DON) pools did not. This is consistent with previous observations that the δ15N of surface ocean N pools is less responsive than that of PNsink to changes in the dominant source of new N to surface waters. In spite of the absence of detectable NO3− in the mesocosms, the δ15N of PNsink indicated that NO3− continued to fuel a significant fraction of export production (20 to 60 %) throughout the 23-day experiments, with N2 fixation dominating export after about 2 weeks. The low rates of organic N export during the first 14 days were largely supported by NO3−, and phytoplankton abundance data suggest that sinking material primarily comprised large diatoms. Concurrent molecular and taxonomic studies indicate that the diazotroph community was dominated by diatom–diazotroph assemblages (DDAs) at this time. However, these DDAs represented a minor fraction (2 fixation; they were thus not important for driving export production, either directly or indirectly. The unicellular cyanobacterial diazotroph, a Cyanothece-like UCYN-C, proliferated during the last phase of the experiments when N2 fixation, primary production, and the flux of PNsink increased significantly, and δ15N budgets reflected a predominantly diazotrophic source of N fueling export. At this time, the export flux itself was likely dominated by the non-diazotrophic diatom, Cylindrotheca closterium, along with lesser contributions from other eukaryotic phytoplankton and aggregated UCYN-C cells, as well as fecal pellets from zooplankton. Despite comprising a small fraction of the total biomass, UCYN-C was largely responsible for driving export production during the last ∼ 10 days of the experiments both directly ( ∼ 5 to 22 % of PNsink) and through the rapid transfer of its newly fixed N to other phytoplankton; we infer that this newly fixed N was transferred rapidly through the dissolved N (including DON) and PNsusp pools. This inference reconciles previous observations of invariant oligotrophic surface ocean DON concentrations and δ15N with incubation studies showing that diazotrophs can release a significant fraction of their newly fixed N as some form of DON.

Abstract

International audience; Dinitrogen (N-2) fixation by prokaryotic microorganisms provides bioavailable nitrogen in oligotrophic marine ecosystems such as coral reefs. The widespread though largely unknown heterotrophic diazotrophs require dissolved organic matter (DOM) for their nutrition. In coral reef ecosystems, DOM-rich mucus released by corals potentially sustains heterotrophic diazotrophic growth and activity. In this study, we investigated the effect of coral mucus on planktonic N-2 fixation in in situ experiments over a seasonal cycle within a New Caledonian lagoon, as well as in in vitro experiments in which the effect of mucus was monitored for 72 h. During the field experiment, N-2 fixation rates ranged between 0.12 and 7.90 nmol N1(-1) d(-1). Despite the highest N-2 fixation being found after mucus release, no significant difference was measured between the seawater surrounding the coral before and after mucus release. Similarly, the addition of mucus during the in vitro experiment enhanced N-2 fixation rates 1.5-fold, but this increase was not significantly different from the control. The abundance of 2 dominant populations of diazotrophs associated with corals and their surrounding seawater environments (unicellular cyanobacteria and rhizobia) found within pure mucus samples was on average 18-fold higher than in the surrounding seawater in the summer period and 400-fold higher in the winter. Our results suggest that although coral mucus does not influence planktonic N-2 fixation, the release of large numbers of diazotrophic cells associated with the mucus likely influences the abundance and diversity of diazotroph populations within the lagoon waters.

Abstract

Instream channel restoration is a common practice in river engineering that presents a challenge for research. One research gap is the development of monitoring techniques that allow for testable predictions of sediment transport and supply. Here we use Radio Frequency Identification (RFID) transponders to compare the short-term (1-year) sediment transport response to flood events in a restored and a control reach. The field site is Wilket Creek, an enlarged creek in a fully urbanized catchment without stormwater management control in Toronto, Ontario. The responses to three flooding periods, each of which are at or above the design bankfull discharge, are described. Key results are that (i) particle mobility is lower in the restored reach for all three periods, (ii) full mobility occurs in the control reach during the first two floods while partial mobility occurs in the restored reach, and (iii) the constructed morphology exerted a controlling influence on particle entrainment, with higher mobility in the pools. Log-transformed travel distances exhibit normal distributions when grouped by particle size class, which allows a statistical comparison with power law and other predictive travel-distance relations. Results show that three bedload transport conditions can occur, with partial mobility associated with a mild relation between particle size and travel distance and full mobility associated with either a flat or steep relation depending on the degree of integration of particles in the bed. Recommendations on seeding strategy and sample sizes are made to improve the precision of the results by minimizing confidence intervals for mobility and travel distances. Even in a short term study, the RFID sediment tracking technique allows a process-based assessment of stream restoration outcomes that can be used to justify the instream intervention and plan future attempts to stabilize and enhance the system.

International audience; 20 This study aims to estimate saturated hydraulic conductivity in a silty loam soil and compare modelled data with 21 experimental ones. The flow characteristics of twelve undisturbed soil cores (5 cm in diameter × 6 cm high) were 22 measured in the laboratory after performing X-ray computed microtomography (microCT) analysis. MicroCT 3D 23 imaging was integrated with an existing pore morphologic model and a numerical simulation based on mesh-24 free smoothed particle hydrodynamics (SPH) to calculate the water flow through the macropore network 25 (pores N 40 μm). Results showed that the proposed SPH method was able to predict hydraulic conductivity of 26 large-sized samples as falling in the range of the experimental ones. By contrast the morphologic model generally 27 underestimated the water flow and was slightly affected by the pore shape. Increasing microCT imaging resolu-28 tion and expanding the variability with other soil types will improve the understanding of the role of micropore 29 size and morphology on water conductivity. 30

Abstract

International audience; On the coast of Northern Dobrogea, south of the Danube delta, the Greek settlement of Orgame was founded in the mid 7th c. BC, probably by Milesian colonists. The ancient city was located on the Cape Dolojman which today overlooks a large lagoon complex. We undertook a chronostratigraphic study to:(i) understand coastal changes around Cape Dolojman since ca. 5000 years BP in connection with the construction of the Danube delta lobes, and (ii) identify potential sediment impacts related to human occupation of the site. Three cores were extracted from the lagoon area. Sedimentological and biological analyses were undertaken to reconstruct the evolution of the coastal palaeoenvironments. The results show a closure of the marine bay around 3500 cal. BP and its transformation into a lagoon environment. The first major environmental change was due to the construction of the lobe St. George I and the formation of the barrier Lupilor. Around 2000 cal. BP, the formation of an intra-lagoonal lobe, the Dunavatz, led to the gradual transformation of the lagoon into a fluvial-dominated system. Paradoxically, lagoon waters today still wash the ancient Greek harbour environment, which has not been totally infilled by alluvial sediments. To understand this paradox, in a context of coastal progradation, we compared and contrasted the geomorphological data with the nearby city of Istros/Histria, which was already landlocked at this time. The location of these two Greek colonies relative to the coastal sediment cell and barriers partly explains their contrasting palaeoenvironmental evolution. Until 2650 cal. BP, the increase in charcoal and organic matter in sedimentary archives is interpreted as an anthropogenic signal for a more extensive use of the vegetation cover following the foundation of the city of Orgame (e.g. for domestic use and funeral rites).

Abstract

International audience; This paper considers the sectoring of a large water distribution network into district metered areas (DMAs) and simultaneously optimizes rehabilitation of the network with new pipes, control valves, and storage tanks. Because available water resources are much lower in the dry season, both the design and operational settings are optimized to satisfy water-demand, water-quality, and pressure constraints, as well as efficiency indices under stringent conditions. Because of the heterogeneity of the multiple decision variables and the complicated way they interact through the multiple objectives (some complimentary and some conflicting), it is not possible to fully automate the simultaneous sectorization, rehabilitation, and operational optimization. Therefore, a multistage approach is used where engineering judgment and network graph simplification and visualization tools are used to find a good feasible solution that serves as a first guess for further optimization of sectors and operational settings, to achieve feasible solutions with better cost of implementation, demand similarity among DMAs, and better pressure uniformity in operations. A multiobjective agent swarm optimization framework is used to iteratively change the sectors at the boundaries. For the final configuration, sequential linear programming is used to find optimal valve and pump settings.

International audience; In this paper, we present and discuss simultaneous records of current and wideband electric field waveforms at 380 km distance from the strike point associated with an upward bipolar flash initiated from the Säntis Tower. The flash contains 23 negative strokes and one positive stroke. The height of the ionospheric reflection for the positive pulse was inferred to be about 94.9 km, a value which is significantly higher than for negative pulses of this same flash, which range from 73 to 81 km. It is also found that the ratio of the peak field to the current peak is about two times smaller for the positive pulse compared to negative pulses. This difference can be attributed to a lower return stroke speed for the positive stroke compared to that for negative strokes, and also to the fact that the enhancement of the electric field due to the presence of the tower and the mountain might be more significant for negative pulses, which are characterized by faster risetimes, than for the positive pulse. International audience; The determination of the chemical nature of the organic matter associated with phytoliths remains a challenge. This difficulty mainly stems from amounts of organic carbon (C) that are often well below the detection limit of traditional spectroscopic tools. Conventional solid-state 13 C Nuclear Magnetic Resonance (NMR) is widely used to examine the nature and structure of organic molecules, but its inherent low sensitivity prohibits the observation of diluted samples. The recent advent of commercial microwave source in the terahertz range triggered a renewed interest in the Dynamic Nuclear Polarization (DNP) technique to improve the signal to noise ratio of solid-state NMR experiments. With this technique, the 13 C spectrum of a phytolith sample containing 0.1% w/w C was obtained overnight with sufficient quality to permit a semi-quantitative analysis of the organic matter, showing the presence of peptides and carbohydrates as predominant compounds. Considering the natural abundance of the 13 C isotope, this experiment demonstrates that DNP NMR is sufficiently sensitive to observe spin systems present in amounts as low as a few tens of ppm. The molecular analysis of natural organic matter in organisms, soils or sediments is a challenging field for our understanding of the processes involved in the carbon cycle. Among the many challenges to be met is the difficulty in obtaining a quantitative or semi-quantitative speciation of the organic carbon, with the least possible preparation steps. Indeed, several chemical steps are often required to purify the molecular compounds to be analyzed , from the minerals they are associated with 1-3. Unfortunately these steps can modify the molecular nature of the compounds to be analyzed, especially when HF is involved 4. In this context, 13 C Nuclear Magnetic Resonance (NMR), is an attractive method since it is element specific and therefore does not necessarily require prior extraction of the organic C, as long as the mineral matrix is free from paramagnetic elements (e.g. Fe(II), Fe(III), Cu(II), etc.) which cause substantial signal loss by broadening the lines beyond detection 5-7. Furthermore, 13 C NMR provides a detailed analysis of the nature of the organic C from its surrounding matrix, and the quantitative exploitation can be easily performed using an internal or external reference, or without calibration when only relative proportions are needed. This technique has been successfully used to investigate plant tissuese.g 8. and transformation of organic matter in soils e.g. refs 9-11. In this context, 13 C NMR analysis of organics associated with biogenic silica is a favorable case. Abstract

Systems biology promises to revolutionize medicine, yet human wellbeing is also inherently linked to healthy societies and environments (sustainability). The IDEA Consortium is a systems ecology open science initiative to conduct the basic scientific research needed to build use-oriented simulations (avatars) of entire social-ecological systems. Islands are the most scientifically tractable places for these studies and we begin with one of the best known: Moorea, French Polynesia. The Moorea IDEA will be a sustainability simulator modeling links and feedbacks between climate, environment, biodiversity, and human activities across a coupled marine–terrestrial landscape. Abstract

International audience; This article reports the design of 13C-dating, the first method to calculate the relative age of molecular substance homologues occurring in fractions from the same soil sample. Soil is a major carbon pool impacting modern climate by CO2 release and uptake. Molecular substances that sequester carbon in soils are poorly known due to the absence of methods to study molecular-level C dynamics over agricultural time scales, e.g. 0-200 years. Here I design a method to calculate the relative age of the plant-derived C31 n-alkane occurring in 6 fractions from a soil sample naturally 13C-labelled by maize cropping during 23 years. Soil fractions are the bulk soil extract, two humin-encapsulated fractions and three particle-size fractions. Results show that C31 n-alkane homologues have relative ages ranging from - 6.7 years for the humin-encapsulated homologue to + 25.1 years for the 200-2000 μm fraction homologue. Such a wide variation of 31.8 years evidences temporal pools of molecular substances in soil. This finding also reveals that physical encapsulation can strikingly change the dynamics of a single molecular substance. 13C-dating thus allows to assess the carbon storage potential of molecular substances from crop soils. Such knowledge will help to identify molecular compounds, associated soil pools and agricultural practices that favour carbon sequestration. 13C-dating is further applicable to any environmental sample containing organic matter subjected to a 13C isotope shift with time. 13C-dating will also help to study the sequestration and delayed release of chemicals in various disciplines, such as pollutants in environmental sciences, pharmaceuticals in medicine, and nutrients in food science.

Cette recherche sur la genèse du projet routier intitulé "voie de desserte et de contournement de Besançon" a été menée avec le soutien du ministère de la Recherche et de la Technologie (département Homme, Travail, Technologies, appel à projets 92 "Modernisation des services publics") et du ministère de l'Equipement, du Logement, des Transports et de la Mer (direction de la Recherche et de l'Innovation). Au cours de l'étude, un grand nombre d'élus et de fonctionnaires de l'Etat et des collectivités locales concernées par le projet, ainsi que des militants de plusieurs associations locales, ont aimablement prêté leur collaboration au chercheur; qu'ils en soient sincèrement remerciés. La majorité des matériaux de recherche utilisés ici ont été recueillis à la mairie de Besançon et à la Direction départementale de l'Equipement du Doubs. Abstract

It is presented in this article, a simplified theory of the active lift turbine which has been the subject of several patent[4, 5, 11]. A simplified theory is proposed to extend the Betz limit of the yield on vertical axis wind turbine. This work can be extended either on wind driven or marine current turbine. Based on kinetic energy calculation , that theory demonstrates that the radial force acting on the blade can be used to extend the maximum recoverable power, mainly by transforming a linear motion into a rotating motion. The geometry of this new type of vertical axis turbine is described. Finally the driving power is calculated through the tangential power coefficients. Consequences on the velocity variation generation and induced vibrations are exposed.
