Occupational therapists usually use assessment and screening forms that can identify the problems faced by the person. In the field of return to work, the initial assessment is usually used to get an overview of a problem faced by the employee, both physically and mentally. This study explores the potential screening and reporting used using the Likelihood Scale of Environment & Occupation (LEO) or is also known as Skala Kemungkinan Persekitaran & Pekerjaan (SKiPP), related to local industry in Malaysia. The re-evaluation of the LEO/SKiPP screening is done to report the latest developments in the development of the instrument. Reliable energy-storage materials that can provide enhanced capacitance with good cycling stability along with improved energy and power densities are exceedingly relevant in the commercial arena to attenuate the prevalent energy shortage. Polymers with structural flexibility and good mechanical and thermal stability can be coupled with carbon-based additives to obtain an electrochemical platform for energy storage without compromising on the required thermal and mechanical attributes. Here, a styrene–maleic anhydride copolymer was modified with a diamine to form a polyimide. The samples were characterized using ultraviolet–visible spectroscopy, carbon-13 (13C) nuclear magnetic resonance spectroscopy, Fourier transform infrared spectroscopy, Fourier transform Raman spectroscopy, thermogravimetric analysis, differential scanning calorimetry, Brunauer–Emmett–Teller analysis, scanning electron microscopy, transmission electron microscopy and X-ray diffraction to understand and investigate the spectral, morphological and electrochemical characteristics of the polymer nanocomposites. Efficacious intercalation of graphene oxide resulted in the formation of polymer nanocomposites that can act as excellent supercapacitor electrodes with a high specific capacitance of 700 F/g at 0.5 A/g and an energy and a power density of 129.23 Wh/kg and 5572 W/kg, respectively.

Pancreatic β-cells are susceptible to oxidative stress, leading to β-cell death and dysfunction due to enhanced ROS levels and type 2 diabetes. To inhibit the β-cells damages induced by the oxidative stress, the present study investigates the beneficial effect of various peptides (WL15, RF13, RW20, IW13 and MF18) of immune related proteins (cysteine and glycine-rich protein 2, histone acetyltransferase, vacuolar protein sorting associated protein 26B, serine threonine-protein kinase and CxxC zinc finger protein, respectively). Also, the molecular mechanism of WL15 from cysteine and glycine-rich protein 2 on β-cell regeneration was identified through PEPCK and insulin pathway.In this study, a total of five peptides including WL15, RF13, RW20, IW13, and MF18 were derived from immune-related proteins such as cysteine and glycine-rich protein 2, histone acetyltransferase, vacuolar protein sorting associated protein 26B, serine threonine-protein kinase and CxxC zinc finger protein, respectively. These protein sequences were obtained from an earlier constructed transcriptome database of a teleost Channa striatus. The identified peptides were evaluated for their antioxidant as well as antidiabetic activity. Based on the in silico analysis and in-vitro screening experiments, WL15 was predicted to have better antioxidant and antidiabetic activity among the five different peptides. Therefore, WL15 alone was further analyzed for apoptosis, antioxidant capacity, glucose metabolism, and gene expression performance, which was investigated on the alloxan (500 µM) induced zebrafish in vivo larval model.The results showed alloxan exposure to zebrafish larvae for a day, the ROS was generated in the β-cells. Interestingly, WL15 treatment showed a protective effect by reducing the toxicity of alloxan exposed zebrafish larvae by increasing their survival and heart rate. Moreover, WL15 reduced the intracellular ROS level and apoptosis in alloxan-induced larvae. The superoxide anion and lipid peroxidation levels are also reduced by improving the glutathione content after the WL15 treatment. Besides, WL15 treatment increased the proliferation rate of β-cells and decreased the glucose level. Further, the gene expression studies revealed that WL15 treatment normalized the PEPCK expression while upregulating the insulin expression in alloxan exposed larvae.Overall, the findings indicate that WL15 of cysteine and glycine-rich protein 2 can act as a potential antioxidant for type 2 diabetes patients in respect of improving β-cell regeneration.

Background: Cryptogenic strokes are common in young adults. Patent foramen ovale (PFO) is an important cause of cryptogenic ischemic strokes. Transcranial Doppler (TCD) with bubble contrast is a noninvasive bedside tool in screening for PFO and other right to left shunt (R-L shunt). Percutaneous PFO closure in selected patients with a high risk for paradoxical emboli is beneficial. Data on PFO in young cryptogenic strokes from India are limited. Aims: To determine the utility of screening for R-L shunt using TCD in young patients with cryptogenic strokes and to identify clinical predictors of an R-L shunt. Materials and methods: This was a hospital-based prospective study conducted between January 2013 and December 2019 in a tertiary hospital in South India. All consecutive patients with ischemic stroke and ages between 18 and 45 years were included. TCD with bubble contrast study was performed on all patients. Those who were TCD bubble contrast study positive and had features of an embolic stroke of undetermined source (ESUS) underwent transesophageal echocardiography (TEE) to confirm a PFO and to look for its high-risk features. Selected ESUS patients with PFO and associated high-risk features as identified on TEE underwent percutaneous PFO device closure. All patients were followed up in the stroke and cardiology clinics. Results: During the study period, 6,197 patients with ischemic strokes were screened for eligibility of which 304 (4.9%) were between the age of 18 and 45 years. Of these, 300 patients with ischemic stroke in young underwent the TCD bubble contrast study. R-L shunt was found in 121 (40.3%) patients. Based on an extensive etiological evaluation, 72 patients were identified to have an ESUS and underwent TEE for confirming PFO. Of these, 65 patients had PFO, four were negative, and three were found to have extracardiac shunts. Based on clinical findings, imaging features, and high-risk features on TEE, 29 patients underwent PFO closure. Activity at the time of stroke onset equivalent to a Valsalva maneuver (p ≤ 0.01), isolated cortical infarction (p = 0.027), and posterior circulation involvement (p = 0.0135) were significantly associated with the presence of an R-L shunt. The patients who had a higher modified anatomical-functional risk of paradoxical embolism (AF-RoPE) score, a high-grade shunt on the TCD bubble contrast study had a longer length of the tunnel and had the presence of an interatrial septal aneurysm (p = 0.012) were referred for PFO device closure. Conclusions: R-L shunt is common in cryptogenic ischemic strokes in young. TCD with bubble contrast study is a noninvasive and feasible bedside tool to detect them. Applying the ESUS criteria in these cryptogenic strokes with a positive TCD bubble contrast study can be then used for selecting patients for more invasive tests like TEE. High-risk PFOs picked up with TEE can be then considered for PFO closure for secondary stroke prevention. The history of Valsalva maneuver-like activity (such as lifting heavy weights or straining) at the time of stroke onset can be a clinical predictor for the presence of an R-L shunt. In addition to isolated cortical infarction, the presence of posterior circulation infarct in ESUS can predict the presence of an R-L shunt.

The management of employee mental health in the workplace is essential. Good leadership by employers can increase employees' productivity and directly improve their quality of life. This study explores the use of more objective evaluators in formulating employers' action plans on workplace psychosocial risks. This survey was conducted using an employer practice evaluation form (Amalan Majikan: AM). Seventy-seven employers have filled out this survey voluntarily. Seven employers (9.1%) received an excellent result equivalent to five stars rating. This rating can be used as a reference for employers' practice managing psychosocial risk in the workplace. Boswellia serrate is an ancient and highly valued ayurvedic herb. Its extracts have been used in medicine for centuries to treat a wide variety of chronic inflammatory diseases. However, the mechanism by which B. serrata hydro alcoholic extract inhibited pro-inflammatory cytokines in zebrafish (Danio rerio) larvae with LPS-induced inflammation remained unknown.LC-MS analysis was used to investigate the extract's phytochemical components. To determine the toxicity of B. serrata extract, cytotoxicity and embryo toxicity tests were performed. The in-vivo zebrafish larvae model was used to evaluate the antioxidant and anti-inflammatory activity of B. serrata extract.According to an in silico study using molecular docking and ADMET, the compounds acetyl-11-keto-boswellic and 11-keto-beta-boswellic acid present in the extract had higher binding affinity for the inflammatory specific receptor, and it is predicted to be an orally active molecule. In both in-vitro L6 cells and in-vivo zebrafish larvae, 160 µg/mL concentration of extract caused a high rate of lethality. The extract was found to have a protective effect against LPS-induced inflammation at concentrations ranged between 10 and 80 µg/mL. In zebrafish larvae, 80 µg/mL of treatment significantly lowered the level of intracellular ROS, apoptosis, lipid peroxidation, and nitric oxide. Similarly, zebrafish larvae treated with B. serrata extract (80 µg/mL) showed an increased anti-inflammatory activity by lowering inflammatory specific gene expression (iNOS, TNF-α, COX-2, and IL-1).Overall, our findings suggest that B. serrata can act as a potent redox scavenger against LPS-induced inflammation in zebrafish larvae and an inhibitor of specific inflammatory genes.

Parabens are synthetic antimicrobial compounds used as a preservative for extending the shelf life of food, pharmaceutical and cosmetic products. The alkyl chain length of the paraben esters positively correlates with their antimicrobial property. Hence, long-chain paraben esters, namely butylparaben and propylparaben, are used in combination as they have better solubility and antimicrobial efficacy. Extensive use of parabens has now resulted in the ubiquitous presence of these compounds in various human and environmental matrices. During early life, exposure to environmental contaminants is known to cause oxidative-stress mediated apoptosis in developing organs. The brain being one of the high oxygen-consuming, metabolically active and lipid-rich organ, it is primarily susceptible to reactive oxygen species (ROS) and lipid peroxidation (LP) induced neuronal cell death. The primary cause for the impairment in cognitive and emotional neurobehvioural outcomes in neurodegenerative disease was found to be associated with neuronal apoptosis. The present study aimed to study butylparaben and propylparaben's effect on zebrafish during early embryonic stages. Besides this, the association between alteration in anxiety-like neurobehavioral response with oxidative stress and antioxidant status in head region was also studied. The study results showed variation in the toxic signature left by butylparaben and propylparaben on developmental parameters such as hatching rate, survival and non-lethal malformations in a time-dependent manner. Data from the light-dark preference test showed embryonic exposure to butylparaben and propylparaben to trigger anxiety-like behavior in zebrafish larvae. In addition, a significant increase in intracellular ROS and LP levels correlated with suppressed antioxidant enzymes: superoxide dismutases (SOD), catalases (CAT), Glutathione peroxidase (GPx), glutathione S-transferase (GST), and Glutathione (GSH) activity in the head region of the zebrafish larvae. Acetylcholinesterase (AChE) activity was also suppressed in the exposed groups, along with increased nitric oxide production. The overall observations show increased oxidative stress indices correlating with upregulated expression of apoptotic cells in a dose-dependent manner. Collectively, our findings reveal butylparaben and propylparaben as an anxiogenic neuroactive compound capable of inducing anxiety-like behavior through a mechanism involving oxidative-stress-induced apoptosis in the head of zebrafish larvae, which suggests a potential hazard to the early life of zebrafish and this can be extrapolated to human health as well.

Peptide-based drug development is an emerging and promising approach in cancer therapeutics. The present study focuses on understanding the mechanism of MP12 peptide (MDNHVCIPLCPP) derived from cysteine-rich trypsin inhibitor protein of virulence factor of pathogenic fungus Aphanomyces invadans. MP12 is involved in antiproliferative activity against the human laryngeal epithelial cell (Hep-2), demonstrated in this study. MP12 sequence showed a significant binding score and has multiple hydrogen bond interactions with the proteins that play a vital role in apoptotic pathways such as Bcl-2, caspase-3, caspase-7, and XIAP. Based on the bioinformatics characterization and molecular docking result, further study was focused on MP12 antiproliferative activity. Cell-to-cell communication is essentially required in bacteria for the production of multiple virulence factors and successful colonization in the host. Targeting the virulence factors production without hampering the growth of the pathogens is a potential strategy to control pathogenesis. To accomplish this, a total of 43 mangrove isolates were screened for quorum quenching (QQ) activity against Pseudomonas aeruginosa (PA), in which eight bacteria have shown antibiofilm activity without hampering the growth of the PA. Prominent QQ activity was observed in Bacillus subtilis BR4. Wireless sensor networks attract so much attention in current IoT-enabled industrial and domestic applications having either homogeneous or heterogeneous sensors deployed to acquire information of intent. WSNs are designed to operate using self-powered sensor nodes as their choice of application is geographic critical. Such nodes must support energy efficiency so that network longevity becomes high. Cluster head selection plays a crucial stage in a WSN architecture which mainly focuses on the minimization of network energy consumption. It groups sensor nodes in such a way that a sophisticated network cluster is formed to have enhanced life time besides a low power consumption. A popular clustering technique, known as LEACH and its variants, is found to be energy efficient compared to its counterparts. The authors propose a novel fully connected energy efficient clustering (FCEEC) mechanism using the electrostatic discharge algorithm to establish a fully connected network with shortest path routing from sensor nodes (SNs) to cluster head (CH) in a multihop environment. The proposed electrostatic discharge algorithm (ESDA) enhances network life time while attaining energy efficient full connectivity between sensor nodes. As a result of ESD, the dead node count is reduced significantly so that the network longevity is increased. In the end, simulation results exhibited improved performance metrics such as energy efficiency, dead node count, packet delivery, and network latency compared to certain conventional CH selection approach.

In this work, platinum nanoparticles (Pt) decorated on a spinel group, zinc cobalt oxide (ZnCo2O4) 3-D flower was intended to detect caffeine electrochemical sensor. Pt particles were used to enhance the performance of hydrothermally prepared 3-D ZnCo2O4 flower. So Pt nanoparticles were decorated on each thin nanosheet of self-assembled 3-D flower-like ZnCo2O4. The thin transparent nanosheet of the ZnCo2O4 flower will benefit electrochemical sensors by simplifying its ion exchange. Therefore, Pt@ZnCo2O4 shows an excellent electrochemical conductivity, which promotes the caffeine-based sensor's rapid electrochemical oxidation. Using amperometric i-t at an applied potential of 1.45 V, Pt@ZnCo2O4 modified electrode shows widespread of two linear ranges (0.05 to 265.55 μM and 295.55 to 757.55 μM) with a low detection limit (0.0114 and 0.01657 μM) and high sensitivity (3.419 and 1.862 μA μM−1 cm−2). Moreover, good results were obtained for the real-time detection of caffeine in sugar-free cola beverage and energy drink samples. Henceforth, the developed method could be an ideal way to detect caffeine in real samples.

A novel antibacterial immunostimulant using Platinum nanoparticles (PtNPs) and lectin from Metapenaeus dobsoni (Md-Lec) was developed. The Md-Lec and PtNPs (Pt-lec) hybrid formed through non-covalent interaction exhibits antimicrobial activity against fish specific pathogens by affecting membrane integrity and producing excess reactive oxygen species. The therapeutic efficacy of Pt-lec was demonstrated through rescuing Aeromonas hydrophila infected Nile Tilapia. Pt-lec prevents the infection spreading and reduces the bacterial bioburden in less than 12 h, and as a result of this the fish were restored to normalcy. To assess immunostimulation, we studied the expression of three different immune related genes, namely LEC, Myd88 and COX-2 in the gills, liver, spleen and kidney of fish under various experimental conditions. Our results showed that Pt-lec treatment appeared to be better when compared to lectin alone in enhancing the expression of Myd88 and COX-2, but LEC was not as expected. These results suggest that Pt-lec has the ability to protect Nile Tilapia against bacterial infection by restricting bacterial bioburden through their direct effects on the bacterial membrane and indirectly through their effects on host immune-related gene expression. This hybrid could have potential “green” application in fish farming in rescuing infected animals when compared to widely and unregulated antibiotics.

To use machine learning to discover web application security issues (ML). Web applications are notoriously difficult to analyses due to their variety and bespoke construction. Machine learning protects websites and other digital assets. It uses human-labeled data to deliver automated analytical tools with web-programming semantics. We introduce Mitch as the black box ML solution for detecting Cross-Site Request Forgery (CSRF). With Mitch’s help, we discovered 35 new CSRFs across 20 major websites and 3 new CSRFs in live production applications. In the end, working code will be utilized to prove Mitch’s worth.

A semisynthetic strategy has been developed for the synthesis of novel 6-gingerol based analogs using simple and robust chemistries.

INTRODUCTION: Rifampicin conjugated (R-CP), and rifampicin -isoniazid dual conjugated (RI-CP) norbornene-derived nanocarriers are newly designed for pH stimuli-responsive delivery of tuberculosis (TB) drugs. Its biosafety level is yet to be well established. OBJECTIVES: To assess the impacts of the nanocarriers on liver cells using zebrafish animal model and human liver cell line model (HepG2). METHODS: Initially, lethal dose concentration for the norbornene-derived nanocarrier systems in zebrafish was determined. The toxic effects were analysed at the sub-lethal drug concentration by histopathological study, total GSH level, gene expression and DNA damage in zebrafish liver cells. Fish erythrocyte nuclear abnormalities were also evaluated. Cell viability and oxidative stress level (ROS generation) after exposure to the nanoconjugates was determined using HepG2 cell in the in vitro study. RESULTS: In vivo studies of both R-CP and RI-CP showed 100% mortality at 96 hours for exposure concentration >100mg/l and showed toxic changes in zebrafish liver histology, GSH, and DNA damage levels. A noticeable upregulated PXR, CYP3A and cyp2p6 genes was observed in RI-CP exposure than in RIF or R-CP molecules. The in vitro study revealed a dose-dependent effect on cell viability and ROS generation for RIF, R-CP and RI-CP exposures in HepG2 cells. CONCLUSION: The current study reports that the rifampicin conjugated (R-CP) and rifampicin-isoniazid conjugated (RI-CP) norbornene derived nanocarriers exhibit enhanced toxic responses in both adult zebrafish and HepG2 cells. The pH-sensitive norbornene derived nanocarriers on conjugation with different drugs exhibited varied impacts on hepatic cells. Hence the present investigation recommends a complete metabolomics analysis and norbornene carrier-drug interaction study to be performed for each drug conjugated norbornene nanocarrier to ensure its biosafety.

GR15 is a short molecule or peptide composed of aliphatic amino acids and possesses to have antioxidant properties. The GR15, 1GGGAFSGKDPTKVDR15 was identified from the protein S-adenosylmethionine synthase (SAMe) expressed during the sulfur departed state of Arthrospira platensis (spirulina or cyanobacteria). The in-silico assessment and the structural features of GR15 showed its antioxidant potency. Real-time PCR analysis found the up-regulation of ApSAMe expression on day 15 against oxidative stress due to 10 mM H2O2 treatment in A. platensis (Ap). The antioxidant activity of GR15 was accessed by the cell-free antioxidant assays such as ABTS, SARS, HRAS and NO; the results showed dose-dependent antioxidant activity. The toxicity assay was performed in both in vitro and in vivo models, in which peptide does not exhibit any toxicity in MDCK cell and zebrafish embryos. The intercellular ROS reduction potential of GR15 peptide was also investigated in both in vitro and in vivo models including LDH assay, antioxidant enzymes (SOD and CAT), and fluorescent staining assay (DCFDA, Hochest and Acridine orange sting) was performed; the results showed that the GR15 peptide was effectively reduced the ROS level. Further, RT-PCR demonstrated that GR15 enhanced the antioxidant property and also up-regulated the antioxidant gene, thus reduced the ROS level in both in vitro and in vivo models. Based on the results obtained from this study, we propose that GR15 has the potential antioxidant ability; hence further research can be directed towards the therapeutic product or drug development against disease caused by oxidative stress.

Effect of bamboo vinegar powder (BVP) on growth, immunity, disease resistance, and immune-related gene expressions in juvenile Asian sea bass (barramundi), Lates calcarifer against Vibrio anguillarum was investigated. V. anguillarum infected fish fed by 2g BVP kg-1 diet exhibited significant growth, albumin (AB), serum total protein (TP), and globulin (GB) levels. Similarly, enhanced lysozyme (LZM), phagocytic (PC), anti-protease (AP), respiratory burst (RB), bactericidal (BC) activities and total immunoglobulin (Ig) levels were observed in fish fed 2g BVP kg-1. In addition, use of 2g BVP kg-1 in diet modulated several immune related gene expressions against Vibrio disease. This study exhibited a positive effect of dietary 2g BVP kg-1 diet on hemato-biochemical and immunological responses, which enhance the immunocompetence and health status of L. calcarifer against V. anguillarum infection.

Social and emotional intelligence has understood their own and other's feelings and reactions according to their needs. The therapeutic relationship, which considered as a foundational concept among occupational therapy practitioners, which refers to establish rapport with the patient and the therapist to understand the patient's needs and set the goal respectively. To determine the relationship between social intelligence (SI), emotional intelligence (EI) and therapeutic relationship (TR) among clinical occupational therapists. The study used quantitative non-experimental correlational design. Eighty participants recruited from eight hospitals in Riyadh. Data conducted using a closed-ended questionnaire. It contains several questions in social intelligence (MESI), emotional intelligence (Assessing emotional scale), and therapeutic relationship between them (STAR). The questionnaire distributed to clinical occupational therapists in the form of an electronic questionnaire to facilitate the data collection process. The results releveled that there was a significant relationship in social intelligence, emotional intelligence and therapeutic relationship among clinical occupational therapists (p = < 0.05). Future study needs to provide the awareness of social intelligence, emotional intelligence and therapeutic relationship among clinical occupational therapist. Supramolecular compounds are capable of undergoing various non-covalent interactions with other molecules like host-guest chemistry, controlled encapsulation, hydrophobic interactions, a variety of delivering strategies, electron affinity induced interactions, etc. These properties of supramolecular compounds make them attractive to be effective drug carriers. This review will describe in detail the recent interesting reports on the structure-activity relationship of supramolecular compounds in drug delivery. The supramolecular compounds considered are cyclodextrins, cucurbiturils, avidin, biotin, rotaxanes, viologens, and dendrimers. Studying the opportunities given by these supramolecular compounds in drug delivery will pave the way for novel efficient drug delivery systems for the future.

Background Glioblastoma (GBM) remains a largely incurable disease as current therapy fails to target the invasive nature of glioma growth in disease progression and recurrence. Here, we use the FDA-approved drug and small molecule Hippo inhibitor Verteporfin (VP) to target YAP-TEAD activity, known to mediate convergent aspects of tumor invasion/metastasis, and assess the drug’s efficacy and survival benefit in GBM models. Methods Up to 8 low-passage patient-derived GBM cell lines with distinct genomic drivers, including 3 primary/recurrent pairs, were treated with VP or vehicle (VEH) to assess in vitro effects on proliferation, migration, invasion, YAP-TEAD activity, and transcriptomics. Patient-derived orthotopic xenograft (PDX) models were used to assess VP’s brain penetrance and effects on tumor burden and survival. Results VP treatment disturbed YAP/TAZ-TEAD activity; disrupted transcriptome signatures related to invasion, epithelial-to-mesenchymal, and proneural-to-mesenchymal transition, phenocopying TEAD1-knockout effects; and impaired tumor migration/invasion dynamics across primary and recurrent GBM lines. In an aggressive orthotopic PDX GBM model, short-term VP treatment consistently diminished core and infiltrative tumor burden, which was associated with decreased tumor expression of Ki67, nuclear YAP, TEAD1, and TEAD-associated targets EGFR, CDH2, and ITGB1. Finally, long-term VP treatment appeared nontoxic and conferred survival benefit compared to VEH in 2 PDX models: as monotherapy in primary (de novo) GBM and in combination with Temozolomide chemoradiation in recurrent GBM, where VP treatment associated with increased MGMT methylation. Conclusions We demonstrate combined anti-invasive and anti-proliferative efficacy for VP with survival benefit in preclinical GBM models, indicating potential therapeutic value of this already FDA-approved drug if repurposed for GBM patients.

Existing methods use static path identifiers, making it easy for attackers to conduct DDoS flooding attacks. Create a system using Dynamic Secure aware Routing by Machine Learning (DAR-ML) to solve healthcare data. A DoS detection system by ML algorithm is proposed in this paper. First, to access the user to see the authorized process. Next, after the user registration, users can compare path information through correlation factors between nodes. Then, choose the device that will automatically activate and decrypt the data key. The DAR-ML is traced back to all healthcare data in the end module. In the next module, the users and admin can describe the results. These are the outcomes of using the network to make it easy. Through a time interval of 21.19% of data traffic, the findings demonstrate an attack detection accuracy of over 98.19%, with high precision and a probability of false alarm.

Antioxidant natural products and their analogs especially phenolic compounds, exhibit diverse biological properties, including anti-inflammatory, antioxidant, and anticancer activities. Ginger which is widely used worldwide for various beneficial effects also contains several phenolic antioxidants, and 6-gingerol is one of the natural products studied extensively. However, the molecular mechanism of synthetically synthesized 6-gingerdione (compound 1) from 6-gingerol was not known. In this study, compound 1 and methylated 6-gingerdione (compound 2) were obtained semi synthetically from 6-gingerol. Compound 1 and 2 are subjected to SwissADME prediction. Then the protective effect of compound 1 was analyzed in 2 % EtOH induced HepG2 cells and zebrafish larvae. Hydroxyl and nitric oxide scavenging assays reveal that compound 1 showed more antioxidant activity than compound 2 at 50 μM. Moreover, compound 1 exhibited good anti-inflammatory activity via lipoxygenase inhibition and proteinase inhibition. Apoptosis and oxidative stress in HepG2 cells were induced by 2 % EtOH and treated with compound 1. Compound 1 significantly inhibited the EtOH induced nitric oxide production, apoptosis, and ROS generation in HepG2 cells. Encouraged by the in-vitro antioxidant and anti-inflammatory activities, compound 1 was then investigated for its protective effect in 2 % EtOH induced ALD zebrafish larva. Compound 1 protected the zebrafish larvae from liver injury by suppressing inflammatory (COX-2, TNF-α, and IL-1β) and lipogenic genes (C/EBP-α, SREBP1, and IL-1β) while upregulating the antioxidant gene. Our findings indicate that compound 1 synthesized from 6-gingerol ameliorated liver injury that likely, contributes to its potential antioxidant and anti-inflammatory properties.
