Sociologists and public health scholars have called attention to the rise of social isolation and loneliness in the US. Considering these developments, it is vital to extend the sociological imagination to better understand the forms of meaningful connection and social relationship characteristics sought by individuals. The growing Authentic Movement represents a series of decentralised social groups in the US and abroad that focus on Authentic Relating and Circling Practices. This ethnographic research combines semi-structured interviews and participant observation techniques to examine how these groups promote and allow for participants to explore concepts of emotion intimacy and alternative ways to form authentic connections with others in psychologically safe and (semi-) structured environments. In addition, the research aims to explore how these communities specifically address the rising trends and social problem of social isolation and limited meaningful (emotional) connection with others.

There is a well-established, steady trend of religious disaffiliates in the USA. However, little qualitative research has been devoted to documenting the experience of religious disaffiliation or “exiting,” specifically in the context of identity reconstruction and well-being. This study investigates an understudied subgroup of exiters—individuals who have left Christian fundamentalist religions. Drawing on 24 in-depth interviews, this research reveals the social psychological processes through which former religious participants reconstruct their identity and self-concepts related to their well-being. The results demonstrate that while it is challenging in the beginning stages of the exiting process to develop a new identity and improve self-concepts, over time the construction of a nonreligious identity eventually contributed to participants’ greater well-being.

Fevipiprant , a prostaglandin D 2 receptor 2 antagonist, is in clinical development as a treatment for asthma. The goal of this study was to assess the potential of fevipiprant to cause drug-drug interactions (DDI) as a perpetrator, that is, by altering the pharmacokinetics (PK) of co-medications. In vitro drug interaction studies of clinically relevant drug metabolizing enzymes and transporters were conducted for fevipiprant and its acyl glucuronide (AG) metabolite. Comparison of K i values with unbound systemic or portal vein steady-state plasma exposure of fevipiprant and its AG metabolite revealed the potential for inhibition of organic anion transporting polypeptide 1B1 (OATP1B1) transporters (R-value of 5.99), while other targets including cytochrome P450 enzymes were not, or only marginally, inhibited. Consequently, an open-label, two-part, two-period, single-sequence clinical study assessed the effect of fevipiprant 450 mg QD on the pharmacokinetics of simvastatin 20 mg and rosuvastatin 20 mg, two statins with different dependency in OATP1B1-mediated hepatic uptake, in healthy adult volunteers. The study also assessed the pharmacogenetics of the SLCO1B1 gene, which encodes OATP1B1. Clinically, fevipiprant 450 mg QD showed a low potential for interaction and increased the peak concentrations of simvastatin acid and rosuvastatin by 2.23- and 1.87-fold, respectively, with little or no impact on total exposure. Genotype analysis confirmed that SLCO1B1 genotype influences statin pharmacokinetics to a similar extent either with or without fevipiprant co-administration. In summary, fevipiprant at 450 mg QD has only minor liabilities as a perpetrator for DDI.

Because themes play such a central role in the presentation of qualitative research results, we propose a new method, Iterative Thematic Inquiry (ITI), that is guided by the development of themes. We begin by describing how ITI uses pragmatism as a theoretical basis for linking beliefs, in the form of preconceptions, to actions, in the form of data collection and analysis. Next, we present the four basic phases that ITI relies on: assessing beliefs; building new beliefs through encounters with data; listing tentative themes; and, evaluating themes through coding. We also review several notable differences between ITI and existing methods for qualitative data analysis, such as thematic analysis, grounded theory, and qualitative content analysis. The use of ITI is then illustrated through its application in a study of exiters from fundamentalist religions. Overall, the two most notable features of ITI are that it begins the development of themes as early as possible, through an assessment of initial preconceptions, and that it relies on writing rather than coding, by using a continual revision of tentative results as the primary procedure for generating a final set of themes.

Rationale Fevipiprant, an oral CRTh2 antagonist, inhibited OATP1B1, OATP1B3 and OATP2B1 in vitro. The [I]/Ki ratios indicated a potential for inhibition of OATP1B1 ([I]/Ki=0.22), while interaction with OATP1B3 and OATP2B3 appeared unlikely ([I]/Ki=0.01 and 0.0028, respectively). We assessed clinical effect of orally co-administered fevipiprant on the PK of OATP1B1 substrates simvastatin and rosuvastatin. Methods This was an open-label, two-part, two-period, single-sequence study in healthy volunteers to study effect of fevipiprant 450mg o.d. on the PK of simvastatin 20mg (Part 1) and rosuvastatin 20mg (Part 2). Subjects were genotyped as per known SNPs of the SLCO1B1 gene. Results Part 1 and Part 2 included 17 and 16 subjects, respectively. Co-administration of fevipiprant increased simvastatin acid Cmax by 2.23-fold with little effect on AUC. Simvastatin Cmax increased 20% and AUCinf and AUClast 9% and 11%, respectively. Co-administration of fevipiprant increased rosuvastatin Cmax 1.87-fold with no effect on AUC (Table). Cmax of statins increased in subjects who carried one of the SNPs. Increase in statin exposure upon co-administration of fevipiprant was comparable between SNP carriers and wild-type subjects. Conclusion Co-administration of fevipiprant increased Cmax of both statins with little effect on AUC. Increase in statin exposure was comparable between SNP carriers and wild-type subjects.
