Routine blood culture is used for the detection of bloodstream infections by aerobic and anaerobic bacteria and by common pathogenic yeasts. A retrospective study was conducted in a public hospital in Maceió-AL, by collecting data of all medical records with positive blood cultures. Out of the 2,107 blood cultures performed, 17% were positive with Staphylococcus coagulase negative (51.14%), followed by Staphylococcus aureus (11.21%) and Klebsiella pneumoniae (6.32%). Gram-positive bacteria predominated among positive blood cultures, highlighting the group of Staphylococcus coagulase-negative. While Gram-negative bacteria had a higher number of species among positive blood cultures. Resumo A cultura sanguínea de rotina é usada para a detecção de infecções na corrente sanguínea por bactérias aeróbias e anaeróbias e por leveduras patogênicas comuns. Estudo retrospectivo realizado em hospital público de Maceió-AL, por meio da coleta de dados de todos os prontuários com culturas sanguíneas positivas. Das 2.107 culturas sanguíneas realizadas, 17% foram positivas com Staphylococcus coagulase negativo (51,14%), seguido por Staphylococcus aureus (11,21%) e Klebsiella pneumoniae (6,32%). As bactérias Gram-positiva predominaram entre as culturas de sangue positivas, destacando-se o grupo das Staphylococcus coagulase-negativo. Enquanto as bactérias Gram-negativas apresentaram um número maior de espécies entre as culturas de sangue positivas.

Background Bacteria play a suspected role in the development of several cancer types, and associations between the presence of particular bacteria and prostate cancer have been reported. Objective To provide improved characterisation of the prostate and urine microbiome and to investigate the prognostic potential of the bacteria present. Design, setting, and participants Microbiome profiles were interrogated in sample collections of patient urine (sediment microscopy: n = 318, 16S ribosomal amplicon sequencing: n = 46; and extracellular vesicle RNA-seq: n = 40) and cancer tissue (n = 204). Outcome measurements and statistical analysis Microbiomes were assessed using anaerobic culture, population-level 16S analysis, RNA-seq, and whole genome DNA sequencing. Results and limitations We demonstrate an association between the presence of bacteria in urine sediments and higher D’Amico risk prostate cancer (discovery, n = 215 patients, p < 0.001; validation, n = 103, p < 0.001, χ2 test for trend). Characterisation of the bacterial community led to the (1) identification of four novel bacteria (Porphyromonas sp. nov., Varibaculum sp. nov., Peptoniphilus sp. nov., and Fenollaria sp. nov.) that were frequently found in patient urine, and (2) definition of a patient subgroup associated with metastasis development (p = 0.015, log-rank test). The presence of five specific anaerobic genera, which includes three of the novel isolates, was associated with cancer risk group, in urine sediment (p = 0.045, log-rank test), urine extracellular vesicles (p = 0.039), and cancer tissue (p = 0.035), with a meta-analysis hazard ratio for disease progression of 2.60 (95% confidence interval: 1.39–4.85; p = 0.003; Cox regression). A limitation is that functional links to cancer development are not yet established. Conclusions This study characterises prostate and urine microbiomes, and indicates that specific anaerobic bacteria genera have prognostic potential. Patient summary In this study, we investigated the presence of bacteria in patient urine and the prostate. We identified four novel bacteria and suggest a potential prognostic utility for the microbiome in prostate cancer. Publisher PDF

Introduction Deep neck infections are a group of diseases with serious complications and mortality, which can occur as a result of common diseases in the community and which have surgical and medical treatment options. Objectives Patients ages, genders, complaints, physical examination findings, hospitalization complaints, history of antibiotic use before the application, additional diseases, radiological tests and analysis of examinations, type of treatment method, antibiotic agents selected in treatment, bacterial culture results, duration of hospitalization, complications, mortality rates were systematically recorded. In the study, anaerobic bacterial factors, which are difficult to produce in routine, were produced by considering special transport conditions and culture media. Methods A total of 74 patients who were hospitalized in the Department of Otorhinolaryngology, University of Mersin, between 01.07.2016 and 01.07.2017 for deep neck infection were evaluated prospectively. The study included 37 female and 37 male patients. The ages of the patients ranged from 1 to 69 and the mean age was 31 years. Results According to the analysis of the obtained data, there was a statistically significant relationship between the patients with additional diseases and the treatment modalities of the patients (p = 0.017). The surgical treatment rate was increased in this group of patients. In patients with a history of antibiotic use, it was found that patients in the pediatric group were in hospital longer in terms of length of stay compared to adults (p = 0.036). In adult patients who underwent surgery, the absorptive long axis was found to be longer in mm than in patients receiving isolated medical treatment (p = 0.008). Conclusions Deep neck infections is a disease group that seriously concerns public health, with significant mortality and morbidity. Ensuring airway safety of patients should be the first intervention. Abscesses located lateral to the tonsil capsule may not drain adequately without concomitant tonsillectomy.

A growing body of evidence has demonstrated the importance of the gut microbiome in human health. In general, fecal microbial samples are used to study the mechanisms of relevant diseases. In this context, it is worth mentioning that an optimized cryopreservation method is urgently needed to successfully perform clinical diagnosis, therapy, and scientific investigations of the gut microbiome without affecting its viability and biological activity. In this study, we aimed to test the relative cryopreservation efficiency of different nontoxic natural cryoprotectants using infant fecal and meconium samples. First, we selected two facultative and two obligate anaerobic bacteria as the experimental gut microbial strain to compare these cryoprotectants' toxicity and concentration-dependent bacteria viabilities after cryopreservation, then the viabilities and bacterial diversity of mixed facultative and obligate anaerobic bacteria. Finally, we explored the effects of optimized cryoprotectants for meconium and infant feces after cryopreservation using 16S rRNA sequencing analyses. In addition, to better understand the effectiveness of these cryoprotectants, we used different freeze-thaw conditions mimicking real-life situations in the process of distribution. We found that the better choice for the infant fecal sample's cryopreservation was 100 mg/mL trehalose, whereas 200 mg/mL trehalose/betaine was the optimum choice for meconium cryopreservation. We hope that our results will shed light on the importance of natural cryoprotectants toward the long-term and stable viability of invaluable human gut microbial specimens.

Objectives Knowledge of the microbiological aetiology of periprosthetic joint infection (PJI) is essential to its management. Contemporary literature from the United States on this topic is lacking. This study aimed to identify the most common microorganisms associated with types of arthroplasty, the timing of infection, and clues to polymicrobial infection. Methods We performed an analytical cross-sectional study of patients 18 years of age or older with hip or knee PJI diagnosed at our institution between 2010 and 2019. PJI was defined using the criteria adapted from those of the Musculoskeletal Infection Society. Cases included PJI associated with primary or revision arthroplasty and arthroplasty performed at our institution or elsewhere. Results A total of 2067 episodes of PJI in 1651 patients were included. Monomicrobial infections represented 70% of episodes (n = 1448), with 25% being polymicrobial (n = 508) and the rest (5%, n = 111) culture-negative. The most common group causing PJI was coagulase-negative Staphylococcus species (other than S. ludgunensis) (37%, n = 761). The distribution of most common organisms was similar regardless of arthroplasty type. The S. aureus complex, Gram-negative bacteria, and anaerobic bacteria (other than Cutibacterium species) were more likely to be isolated than other organisms in the first year following index arthroplasty (OR 1.7, 95%CI 1.4–2.2; OR 1.5, 95%CI 1.1–2.0; and OR 1.5, 95%CI 1.0–2.2, respectively). The proportion of culture-negative PJIs was higher in primary than revision arthroplasty (6.5% versus 3%, p 0.0005). The presence of a sinus tract increased the probability of the isolation of more than one microorganism by almost three-fold (OR 2.6, 95%CI 2.0–3.3). Conclusions Joint age, presence of a sinus tract, and revision arthroplasties influenced PJI microbiology.

La Trobe University Faculty of Science, Technology and Engineering Murray Darling Freshwater Research CentreMDFRC item.The development and demise of substantial populations of bacteriochlorophyll (BChl) a and d containing phototrophs are documented within the bottom waters of a billabong (oxbow lake) in south-eastern Australia. The observation of such populations within a freshwater body is unusual, illustrating an unexpected source of organic carbon within the floodplain ecosystem of one of Australia's principal rivers. Values of up to 1086 µg L−1 BChl a and 490 µg L−1 BChl d were recorded during an 11 month study of physico-chemical parameters, algae and bacteria. An unusual density stratification was observed within the billabong, fostered by an increase in bivalent ions at depth, and leading to persistent anoxia below 0.75 m depth over summer. A marked vertical distribution was observed for both algae and bacteria, that for the bacteria being lost with a mixing event in late March. The presence of up to 74 µg L−1 BChl a at the surface of the billabong during the summer months is ascribed to entrainment of bacteria within bubble plumes rising from the sediments.

Highlights • Periapical lesions promotes higher T-bet expression and lower GATA3 and FOXP3 expression. • Periapical lesion promotes increased GATA3 and decreased FOXP3 expression in the group with 4 periapical lesions (4PL) • The adaptive immune system, with a predominance of the Th1 polarization, may be involved in the development of insulin resistance in rats with Periapical lesions., Objective Apical periodontitis (AP) is a chronic or acute inflammatory disease usually developed from endodontic infections, predominantly due to gram-negative anaerobic bacteria invading the dental pulp. This study aimed to evaluate lymphocyte markers to assess the involvement of adaptive immunity in insulin resistance (IR) in a rat model of AP. Design. Forty-five male Wistar albino rats were divided into 3 groups (control, 1AP and 4AP). AP was induced in the upper right first molar (1AP), and in the first and second upper and lower right molars (4AP). The spleen was collected to evaluate the expression of transcription factors involved in lymphocyte polarization, including T-bet (Th1), GATA3 (Th2), and FOXP3 (Treg). Blood samples were assessed for serum cytokine levels transcribed by the respective lymphocyte polarizations, INF-γ (Th1), IL-4 (Th2) and TGF-β (Treg). In addition, glucose and insulin levels were measured to evaluate IR by the HOMA-IR method. Results The results showed higher T-bet expression on AP groups, along with lower GATA3 and FOXP3 expression in the 1AP, in addition to increased GATA3 and decreased FOXP3 expression in the 4AP group compared to the CN group. There was no difference in the INF-γ levels, while IL-4 was decreased in the AP groups. Taken together, these results suggest that the adaptive immune system, with a predominance of the Th1 polarization, may be involved in the development of IR in rats with AP. Conclusions AP promotes increase in the expression of T-bet (4AP) and decrease of FOXP3 expressions and IL-4 levels (1AP and 4AP). However, depending on the number of lesions (1 or 4 lesions), the expression of GATA3 appears differently. Thus, innate immunity and adaptive immunity may contribute to the IR observed in rats with AP.

The problem of treatment and prevention of infectious-inflammatory post extraction complications in modern surgical dentistry are still challenging, despite the great advances in this field. Many etiological factors are identified as contributing to the development of such complications, but at present scientists are paying much attention to the infectious factor, which is associated with the microflora of the tooth socket after tooth removal. According to the literature, the most common post-extraction complication during an outpatient surgical appointment is alveolitis, which accounts for 1-35% of all cases of tooth extraction. To combat an infectious etiological factor of alveolitis, dental surgeons apply topical antiseptics with a broad spectrum of action such as iodoform and chlorhexidine. As a topical antiseptic drug in the treatment and prevention of oral infectious and inflammatory complications, a domestic antiseptic, Decasan, based on decamethoxine, is becoming more widely applied. Previous studies have shown that this antiseptic has antimicrobial effect against gram-positive and gram-negative, aerobic and anaerobic bacteria; this requires further in-depth studies of its properties, which could be applied in the dental surgical practice to treat and prevent infectious-inflammatory post extraction complications. Chlorhexidine bigluconate is a cationic biguanide. Penetrating into the intracellular membranes of bacteria, it impedes the oxygen consumption and leads to the death of bacterial cells. This antiseptic is widely used in dental practice. Treatment of alveolitis is carried out by using the standard method, namely by using iodoform tamponade of the tooth socket in combination with other medicines. Iodoform (triiodomethane) is a yellow crystalline substance with a strong characteristic odor, practically insoluble in water. In dentistry, this drug is used as an antiseptic in powder form, or combined pastes. The purpose of this in vitro study is to investigate the antimicrobial properties of Decasan, chlorhexidine and iodoform against museum microbial strains. Materials and methods. Museum strains of Staphylococcus aureus ATCC 25923, Staphylococcus epidermidis ATCC 14990, Enterococcus faecalis ATCC 29212, Esherichia coli ATCC 25922, Candida albicans ATCC 10231, obtained from L.V. Gromashevsky Institute of Epidemiology and Infectious Diseases, National Academy of Medical Sciences of Ukraine, were used as studied cultures of microorganisms. The antimicrobial effect of decasan, chlorhexidine and iodoform was assessed by quantitative method of serial dilutions in broth and agar according to the order №167, dated 5.04.2007 On approval of guidelines “Determination of sensitivity of microorganisms to antibacterial drugs". Conclusion. According to the data obtained, decasan and chlorhexidine demonstrate higher bacteriostatic and bactericidal effect on the studied museum microbial strains at a concentration thousands of times lower than the concentration of iodoform.

Volatile methyl siloxane is a trace impurity in biogas produced by anaerobic bacteria. It decreases the energy utilization of biogas and thus needs to be removed. In this study, six reduced graphene-oxide aerogels (rGOAs) were prepared from industrial-grade multilayer graphene oxide by a hydrothermo-chemical reduction method in the presence of various amine additives, and the use of the rGOAs as adsorbents for hexamethyldisiloxane (L2) removal was investigated. The developed rGOAs were characterized via the Brunner-Emmet-Teller technique, Fourier-transform infrared spectroscopy, scanning electron microscopy, transmission electron microscopy, and X-ray diffraction. The results revealed that the different pKa values of the amine molecules led to an increase in the rGOA contact angle, the specific surface area and the total pore volume in the following sequence: 0-rGOA

The aim of this study was to determine the common uses and evaluate the microbiological safety of raw milk sold in southern Benin. To determine the different uses of raw milk, 345 individuals were surveyed in three locations (Allada, Ouidah, and Zongo). Per location, 115 individuals were randomly sampled. For the microbial analysis, milk samples were collected from the three targeted locality in southern Benin (Allada, Ouidah, and Zongo). Our data shows that the mean values (CFU/ml) of the analyzed samples vary according to the research organisms. Thus, it is recorded 1.8 × 108 for total aerobic mesophilic flora (TMC), 4.0 × 107 for fecal coliforms (FC), 3.5 × 107 for Escherichia coli, 2.8 × 107 for total coliforms (TC), 2.1 × 107 for Fecal Streptococci (FS), 1.6 × 107 for yeasts and molds (YM), 1.7 × 107 for sulfur reducing anaerobic bacteria (SRA) and 1.2 × 107 for Staphylococcus spp. None of the milk samples contained Salmonella spp. Globally, milk samples had important bacterial load with the highest values for those collected from Zongo and Ouidah. It can be said that raw milk sold in the Southern Benin’s markets does not comply with good hygienic practice rules in milking, storage, transportation and sale. Thus, raw cow milk sold presents a serious health risk for potential consumers. Key words: Raw milk, microbiological quality, food safety, pathogens, Benin.

AIM: The study was done to assess the antimicrobial effectiveness of Chitosan Nanoparticles (CNPs), Chlorhexidine (CHX), and their combination (CHX/CNPs) versus that of Sodium hypochlorite (NaOCl) in patients with mandibular necrotic premolars and to evaluate their effects on post-operative pain after single-visit endodontic treatment. MATERIALS AND METHODS: Sixty patients with necrotic mandibular premolars were divided randomly to four groups (n = 15) according to the used irrigating solution. Instrumentation was done using rotary ProTaper files. During instrumentation, irrigation was done using 2.5% NaOCl; afterward, canals were flushed with sterile saline. A final flush with the study irrigants was done as follows: 3% CNPs for Group A, 2% CHX for Group B, CHX/CNPs for Group C, and 5.25% NaOCl for Group D. Samples were collected from root canals before and after canal preparation then cultured to assess the number of colony-forming units/ml. All patients were instructed to record their pre- and post-operative pain levels on a numerical rating scale. RESULTS: CNPs and CHX/CNPs were significantly more effective than either CHX or NaOCl; however, there was no significant difference between them against anaerobic bacteria. All tested irrigants were similarly effective against aerobic bacteria. CNPs and CHX/CNPs were associated with significantly lower post-operative pain levels in the first 24 h after treatment. CONCLUSIONS: CNPs and its combination with CHX are significantly more effective than both CHX and NaOCl against anaerobic bacteria isolated from necrotic mandibular premolars. Post-operative pain intensity was significantly lower with CNPs and CNPs/CHX combination than with either NaOCl or CHX.

The present study was conducted to investigate the effect of Aspergillus oryzae inclusion on ruminal fermentation, feed intake and dry matter (DM) apparent digestibility in cows fed Cenchrus purpureus forage and concentrate. Cows were randomly assigned in a replicated 4 × 4 Latin square design, into four treatments: control and three doses of A. oryzae culture (1, 2 and 3 g/d). Experimental periods had 21 days, in which 14 days were allowed for treatment adaptation and 7 days for sampling. Addition of 2 g/d increased (p = 0.0054) total anaerobic bacteria. For cellulolytic bacteria and fungal populations increases (p < 0.0001) with all doses were observed. Highest concentrations of total short chain fatty acids (SCFAs), acetate, isovalerate and valerate were observed with 2 g/d of additive. Propionate concentration and acetate: propionate (Ac:Pr) ratio were unaffected by treatments. A. oryzae addition did not modify ruminal pH. A decrease of ammonia-N was observed with 2 g/d of additive. Dry matter intake and apparent digestibility of DM were increased (p = 0.0171 and p = 0.0023, respectively) with the fungal culture addition. It is concluded that Aspergillus oryzae strain H/6.28.1 stimulates ruminal fermentation and improves feed intake and dry matter apparent digestibility in cows feeding with Cenchrus purpureus forage and concentrate.

Energy consumption places growing demands on modern lifestyles, which have direct impacts on the world’s natural environment. To attain the levels of sustainability required to avoid further consequences of changes in the climate, alternatives for sustainable production not only of energy but also materials and chemicals must be pursued. In this respect, syngas fermentation has recently attracted much attention, particularly from industries responsible for high levels of greenhouse gas emissions. Syngas can be obtained by thermochemical conversion of biomass, animal waste, coal, municipal solid wastes and other carbonaceous materials, and its composition depends on biomass properties and gasification conditions. It is defined as a gaseous mixture of CO and H2 but, depending on those parameters, it can also contain CO2, CH4 and secondary components, such as tar, oxygen and nitrogenous compounds. Even so, raw syngas can be used by anaerobic bacteria to produce biofuels (ethanol, butanol, etc.) and biochemicals (acetic acid, butyric acid, etc.). This review updates recent work on the influence of biomass properties and gasification parameters on syngas composition and details the influence of these secondary components and CO/H2 molar ratio on microbial metabolism and product formation. Moreover, the main challenges, opportunities and current developments in syngas fermentation are highlighted in this review.

A central goal of microbiome research is to understand the factors that balance gut-associated microbial communities, thereby creating longitudinal and cross-sectional heterogeneity in their composition and density. Whereas the diet dictates taxa dominance, microbial communities are linked intimately to host physiology through digestive and absorptive functions that generate longitudinal heterogeneity in nutrient availability. Additionally, the host differentially controls the access to electron acceptors along the longitudinal axis of the intestine to drive the development of microbial communities that are dominated by facultatively anaerobic bacteria in the small intestine or obligately anaerobic bacteria in the large intestine. By secreting mucus and antimicrobials, the host further constructs microhabitats that generate cross-sectional heterogeneity in the colonic microbiota composition. Here we will review how understanding the host factors involved in generating longitudinal and cross-sectional microbiota heterogeneity helps define physiological states that are characteristic of or appropriate to a homeostatic microbiome.

The world's energy supply is very dependent on non-renewable energy in the form of fossil fuels. This causes fossil fuels depletion and the need for alternative energy sources such as biogas. Biogas is produced from the fermentation process of organic matter with the help of anaerobic bacteria in free oxygen absence. This study aims to produce biogas from cabbage and tomato waste separately. Biogas production was carried out by varying feed concentrations of 100 g/L, 200 g/L, and 300 g/L for cabbage waste and 81.6 g/L; 215 g/L; and 237 g/L for tomato waste. This study consisted of 10 days seeding and acclimatization process, followed by a start-up stage using insulated anaerobic bioreactors. The largest methane from cabbage and tomato waste was 60% at a feed concentration of 200 g/L and 50% at a feed concentration of 237 g/L, respectively. The maximum growth rates (µm) for biogas from cabbage and tomato waste were 0.122 day-1 and 0.121 day-1, respectively.

At feed-out, aerobic spoilage of silage enables an increase in anaerobic spore-forming bacteria (ANSB) that may enter the total mixed ration (TMR). The aim of our study was to understand whether in hot summers the silage structures and management may affect the level of ANSB in milk for long-ripening cheese production. A survey of silage facilities, management, and their relationships with silage, TMR, feces, and milk ANSB most probable number (MPN) content was conducted in the Po Valley during summer months. Silo type did not affect the mean ANSB, but only the wideness of their value distributions, with a narrow range for bags and a wider range for bunkers. The unloading equipment affected the ANSB count, the front-end loader with cutter was associated with a lower ANSB count—probably as a result of the reduced surface left after daily silage removal. Silo length and daily removed face width were the main factors affecting contamination of silage by spore-forming bacteria during summer, with longer silos and wider surface removal reducing ANSB contamination—probably as a consequence of reduced aerobic spoilage at the silage surface. The silage contamination by spore-forming bacteria within a log10 2 MPN g−1 allowed a low concentration of spore-forming bacteria at the farm bulk milk tank level. Fecal ANSB levels did not factor into the regression that explains the ANSB in farm milk. It has been found that silage facilities’ features and their management are an important first step to reduce the extent of ANSB contamination at the farm level.

Water is essential to the life of every living organism. The water used by the consumer must be safe for the preservation of his health. The objective of this study was to assess the quality of well water used in households in the city of Korhogo. To do this, water samples were taken from the wells of different households. Parameters such as pH, temperature, electrical conductivity and total dissolved solids were measured at the sampling sites. Nitrates and ammonium were determined by spectrophotometric methods using sodium salicylate and indophenol blue, respectively. The membrane filtration technique has been used for the enumeration of total coliforms, Escherichia coli, fecal enterococci, sulfite-reducing anaerobic bacteria, yeasts and moulds. The agar incorporation method was used for the enumeration of mesophilic aerobic germs. The non-conformity relative to pH was 73%. All wells were contaminated with nitrates and ammonium. Non-compliance rates of 80% and 53% were obtained for nitrates and ammonium respectively. Non-compliance with mesophilic aerobic germs, total coliforms, E. coli and fecal enterococci was 100% for each indicator. No spore of sulphite-reducing clostridia has been identified. Contamination rates of 80% (mould) and 67% (yeast) were recorded. The moulds isolated from the various water samples belong to the genus Aspergillus. The quality of well water in households is not suitable for human consumption. These waters require treatment or household residents must switch to other safe sources of drinking water.

Sulfoquinovose is the polar headgroup of plant sulfolipids and is a globally abundant organosulfur compound, and its degradation by bacteria is an important component of the sulfur cycle. Sulfoquinovose degradation by certain bacteria, including Escherichia coli, produces dihydroxypropanesulfonate (DHPS), which is further converted by anaerobic bacteria into 3-hydroxypropanesulfonate (3-HPS), through the catalytic action of DHPS dehydratase (a member of the glycyl radical enzyme family), and sulfopropionaldehyde reductase HpfD (a member of the metal-dependent alcohol dehydrogenase family). Here we report biochemical investigation of Hungatella hathewayi HpfD. In addition to 3-HPS, HpfD also displayed high catalytic activities for NAD+ -dependent oxidation of 4-hydroxybutanesulfonate (4-HBS) and γ-hydroxybutyrate (GHB). The highest activity was obtained with Fe2+ or Mn2+ as the divalent metal cofactor. Indigo is a fabric dye that requires reduction by microbial activity or chemical reagents to render it soluble in water. Sources of indigo for fermentation are primarily divided into composted indigo-containing plants and indigo extracted from plants. To elucidate the factors responsible for bacterial diversity, and for sustaining reduced state of indigo in different preparations, this study assessed fermentation-derived fluids using composted plant leaves, sukumo, and extracted indigo (Ryukyu-ai paste, and Indian indigo cake) prepared using different procedures. Regardless of the indigo source, obligate anaerobic bacteria, including the families Proteinivoraceae and Tissierellaceae, predominate (16.9-46.1%), suggesting their high affinity for this fermentation ecosystem (hyperalkaline and low redox potential). Moreover, bacterial communities in sukumo fermentations are more diverse than those from indigo extracts with the diversity tending to increase based on the fermentation period. Our results further suggest that the microbiota composition in sukumo fermentation is associated with the various bacterial nutrients derived from sukumo, including seed microorganisms. In addition, the debris derived from sukumo can reduce the pH stress experienced by the microorganisms. Further, regardless of 5.4 years difference in the fermentation age, the bacterial flora in two Ryukyu-ai batches exhibit similar features with low microbial diversities. The uniformity of the nutrient, along with the simple, yet strong, bacterial network in Ryukyu-ai fluids may be responsible for the stable bacterial flora composition. Taken together, these results indicate that the microbiota in indigo fermentation is highly influenced by the seed culture, the nutrient derived from raw materials, and the fermentation conditions.

Arsenic is responsible for water pollution in many places around the world and presents a serious health risk for people. Lake Van is the world's largest soda lake, and there are no studies on seasonal arsenic pollution and arsenic-resistant bacteria. We aimed to determine the amount of arsenic in the lake water and sediment, to isolate arsenic-metabolizing anaerobic bacteria and their identification, and determination of arsenic metabolism. Sampling was done from 7.5 m to represent the four seasons. Metal contents were determined by using ICP-MS. Pure cultures were obtained using the Hungate technique. Growth characteristics of the strains were determined at different conditions as well as at arsenate and arsenite concentrations. Molecular studies were also carried out for various resistance genes. Our results showed that Lake Van's total arsenic amount changes seasonally. As a result of 16S rRNA sequencing, it was determined that the isolates were members of 8 genera with arsC resistance genes. In conclusion, to sustain water resources, it is necessary to prevent chemical and microorganism-based pollution. It is thought that the arsenic-resistant bacteria obtained as a result of this study will contribute to the solution of environmental arsenic pollution problems, as they are the first data and provide the necessary basic data for the bioremediation studies of arsenic from contaminated environmental habitats. Green propolis may represent a promising therapeutic alternative against dental anaerobic pathogens because of its antimicrobial action. This study aimed to evaluate the antimicrobial and antibiofilm actions of Brazilian green propolis aqueous extract (BGP-AqExt) against dental anaerobic bacteria. The minimum inhibitory concentration (MIC) and minimum microbicide concentration (MMC) of the extract were determined against the standard strains (ATCC) of Fusobacterium nucleatum, Parvimonas micra, Prevotella intermedia, Porphyromonas gingivalis and Porphyromonas endodontalis. BGP-AqExt was chemically characterized by high-performance liquid chromatography with diode-array detection (HPLC-DAD) analysis. Antibiofilm action was measured by MTT and crystal violet tests. The data were statistically analyzed by ANOVA and Tukey (5%) tests. The extract had antimicrobial action against all tested anaerobic bacteria, with an MIC value of 55 mg/mL for all bacteria, an MMC of 27.5 mg/mL for F. nucleatum and P. micra and 55 mg/mL for P. intermedia. Chemically, BGP-AqExt is composed of quercetin, gallic acid, caffeic and p-coumaric acid, drupani, kaempferol and Artepillin C. Significant reductions in biomass and metabolic action of biofilms were found after BGP-AqExt application. Therefore, BGP-AqExt has an antimicrobial and antibiofilm effect against dental anaerobic bacteria.

Some anaerobic bacteria, particularly Clostridium species, produce extracellular cellulolytic and xylanolytic enzymes as multienzyme complexes (MECs). However, an amylolytic/xylanolytic/cellulolytic multienzyme complex (AXC-MEC) from anaerobic bacteria is rarely found. In this work, the glycoprotein AXC-MEC, composed of subunits of amylolytic, xylanolytic, and cellulolytic enzymes, was isolated from crude extracellular enzyme of the mesophilic anaerobic bacterium Clostridium manihotivorum CT4, grown on cassava pulp, using a milled cassava pulp column and Sephacryl S-500 gel filtration chromatography. The isolated AXC-MEC showed a single band upon native-polyacrylamide gel electrophoresis (native-PAGE). Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) showed at least eight protein bands of the multienzyme complex which predominantly exhibited amylolytic enzyme activity, followed by xylanolytic and cellulolytic enzyme activities. The AXC-MEC is highly capable of degrading starch and non-starch polysaccharides present in cassava pulp into glucose and oligosaccharides, without conventional pretreatment. Base on the genomic analysis of C. manihotivorum CT4, we found no evidence of the known structural components of the well-known multienzyme complexes from Clostridium species, cellulosomes such as scaffoldin, cohesin, and dockerin, indicating that AXC-MEC from strain CT4 exhibit a different manner of assembly from the cellulosomes. These results suggest that AXC-MEC from C. manihotivorum CT4 is a new MEC capable of hydrolyzing cassava pulp into value-added products, which will benefit the starch industry. Chronic periodontal disease and oral bacteria dysbiosis can lead to the accumulation of genetic mutations that eventually stimulate Oral Squamous Cell Cancer (OSCC). The annual incidence of OSCC is increasing significantly, and almost half of the cases are diagnosed in an advanced stage. Worldwide there are more than 380,000 new cases diagnosed every year, and a topic of extensive research in the last few years is the alteration of oral bacteria, their compositional changes and microbiome. This review aims to establish the relationship between bacterial dysbiosis and OSCC. Several bacteria implicated in periodontal disease, includingFusobacterium nucleatum,Porphyromonas gingivalis,Prevotella intermedia, and someStreptococcusspecies, promote angiogenesis, cell proliferation, and alteration in the host defense process; these same bacteria have been present in different stages of OSCC. Our review showed that genes involved in bacterial chemotaxis, the lipopolysaccharide (LPS) of the cell wall membrane of gram negatives bacteria, were significantly increased in patients with OSCC. Additionally, some bacterial diversity, particularly withFirmicutes, andActinobacteriaspecies, has been identified in pre-cancerous stage samples. This review suggests the importance of an early diagnosis and more comprehensive periodontal therapy for patients by the dental care professional.

Biogas merupakan gas mudah terbakar yang dihasilkan dari proses fermentasi bahan-bahan organik oleh bakteri anaerob. Senyawa tersebut didalam digester akan dikonversi menjadi senyawa metan yang dapat dibakar sebagai sumber energi. Bahan tersebut adalah kotoran sapi, tanaman hydrilla verticillata, tanaman eceng gondok, dan tanaman rumput gajah. Pada penelitian kali ini mencoba untuk memanfaatkan bahan-bahan kombinasi antara Kotoran sapi dengan variasi ketiga tanaman dengan perbandingan 2 : 1 dan kombinasi variasi tanaman dengan perbandingan 1 : 1. Untuk membandingkan 3 tanaman tersebut manakah yang optimal menghasilkan biogas. Parameter yang dianalisa terdiri dari kadar air, C/N rasio, suhu, tekanan, lama nyala api dan kadar gas metan. Hasil penelitian menunjukkan bahwa biogas terbaik diperoleh pada waktu fermentasi selama 30 hari pada variasi campuran bahan kotoran sapi dengan hydrilla verticillata pada perbandingan 2 : 1 dengan kadar air sebesar 41,3%, rasio C/N sebesar 21,5%, 50,4% kadar gas metan. Nonalcoholic fatty liver disease is a hepatic disorder with deposition of fat droplets, and has a high risk of progression to steatosis-related hepatitis and irreversible hepatic cancer. Metronidazole (MNZ) is an antiprotozoal and antimicrobial agent widely used to treat patients infected with anaerobic bacteria and intestinal parasites; however, MNZ has also been shown to induce liver tumors in rodents. To investigate the effects of MNZ on steatosis-related early-stage hepatocarcinogenesis, male rats treated with N-nitrosodiethylamine following 2/3 hepatectomy at week 3 were received a control basal diet, high fat diet (HFD), or HFD containing 0.5% MNZ. The HFD induced obesity and steatosis in liver, accompanied by altered expression of Pparg and Fasn, genes related to lipid metabolism. MNZ increased nuclear translocation of lipid metabolism-related transcription factor peroxisome proliferator-activated receptor gamma in hepatocytes, together with altered liver expression of lipid metabolism genes (Srebf1, Srebf2, Pnpla2). Furthermore, MNZ significantly increased the number of preneoplastic liver foci, accompanied by DNA double-strand breaks and late-stage autophagy inhibition, as reflected by increased levels of γ-H2AX, LC3, and p62. Therefore, MNZ could induce steatosis-related hepatocarcinogenesis by inducing DNA double-strand breaks and modulating autophagy in HFD-fed rats.

Fusobacterium necrophorum is a very rare cause of endocarditis. We herein report a case of F. necrophorum endocarditis with liver abscesses in a 51-year-old woman. This is the first reported case of monomicrobial F. necrophorum endocarditis to present in a patient over 50 years old. We also reviewed 10 reported cases, including the present case. Our review indicated that anaerobic bacteria, including Gram-negative anaerobic bacilli such as F. necrophorum, should be considered in the differential diagnosis of infective endocarditis, especially in patients without preexisting organic heart disease.

Background To evaluate the safety and efficacy of arsenic trioxide (ATO) combined with VNP20009 in treating the advanced hepatocellular carcinoma (HCC). Methods The proliferation assay, migration assay and real-time PCR analyses were performed to assess the impact of ATO combined with VNP20009 on the McA-RH7777 cells. Forty Buffalo rats were orthotopically implanted with HCC in the livers and randomly divided into four groups: (A) ATO plus VNP20009; (B) ATO; (C) VNP20009; and (D) control. ATO (2 mg/kg) was administered by peritoneal injection once a day and continued for five days. VNP20009 (about 1×107 CFU) was directly injected into the tail vein. MRI examinations were performed to access the tumor responses one and 2 weeks later, respectively. Micro CT scans of chest were performed to assess the lung metastases. Hematoxylin-eosin (HE) staining and immunohistochemical analyses were performed to analyze the tumor tissues. Results In the in vitro experiments, VNP20009 suppressed the proliferation of McA-RH7777 cells, attenuated their migration ability, and weakened the potential of metastases. MRI examinations showed that the mean residual tumor volumes of ATO plus VNP20009 group on the 7th day and 14th day after the administration of ATO combined with VNP20009 were significantly smaller than those of other groups. Micro CT scans revealed that the lung metastases rates of ATO plus VNP20009 group and VNP20009 group were significantly lower than those of other groups. Immunohistochemical analyses displayed that the levels of VEGF and Vimentin in the tumors of ATO plus VNP20009 group were obviously lower than those of other groups. The median survival of rats in the ATO plus VNP20009 group was longer than those of other groups. Conclusions The strategy of ATO combined with VNP20009 was safe and had a potential to inhibit tumor growth, decrease the lung metastases, and prolong the overall survival in treating the advanced HCC. The two complementary interventions may have synergistic effects.

Hindgut of wild ruminants harbours diversified anaerobic bacteria with promising fiber degrading ability. Fibrolytic enzyme activity is strongly influenced by diet and host species which till date remains unexplored for harnessing their optimum benefits. The present study was conducted to isolate and characterize fiber degrading anaerobic bacteria from faeces of wild blue bull (Boselaphus tragocamelus) inhabiting in semiarid regions. A total of 167 isolates were obtained from 85 faecal samples using M-10 medium, on the basis of clear zones formed on Congo red plates 20 isolates were selected and designated as NLG1-20 for microscopic and biochemical characterization. Further, molecular confirmation was done by PCR analysis with universal 16S rDNA primers. All isolates were obligatory anaerobes except, NLG4, NLG19 and NLG20. Majority of the sugars tested were utilized by most of the isolates except arabinose. Fibrolytic enzyme activities revealed that NLG1 had highest endoglucanase activity, NLG13 had highest exoglucanase activity while NLG8 showed maximum xylanase activity. In case of FPase assay, highest and lowest values were observed in isolate NLG11 (8.96 U/mL) and NLG8 (5.58 U/mL), respectively. Phylogenetic analysis of the isolates revealed a highly diverse group of microbes mainly belonging to the family Paenibacillaceae which have not been previously characterized in ruminants for fiber degradation. Therefore, results obtained in the present study indicated that the screened isolates showed promising fiber degrading potential in terms of filter paper assay and fibrolytic enzyme activity which can be explored further for improving lignocellulose digestibility in ruminants as an additive.

Background: Diabetes mellitus is a chronic disease that is present in almost every country around the world. Diabetics are known to suffer more from gingivitis/periodontitis than the general population. The projected increase in the diabetic population will only lead to an increase in oral diseases, such as gingivitis in this population. Aim: This study evaluates an Ayurvedic toothpaste for the control of gingivitis in a diabetic population. aterials and Methods: In vitro and in vivo evaluations were performed on this Ayurvedic toothpaste. The in vitro study used a well known anaerobic biofilm model. In the in vivo clinical studies, diabetic subjects were evaluated for plaque and gingivitis using classical methods. The subjects were also evaluated using traditional Ayurvedic endpoints, namely Raktasrava (gum bleeding), Shotha (inflammation), and Dantamamsa mriduta (sponginess of gums). Results: The results of these evaluations demonstrate that the Ayurvedic toothpaste has antibacterial efficacy against anaerobic bacteria. Its use by diabetic subjects resulted in a reduction in plaque and gingival indices in two separate studies lasting up to 6 months. In addition, there was an improvement in the Ayurvedic endpoints in these clinical studies as well. Conclusions: This study demonstrates that an Ayurvedic toothpaste has a positive oral health benefit in diabetic subjects who suffer from gingivitis. Key Messages: The research reported within demonstrates that this Ayurvedic toothpaste has a positive oral health benefit in diabetic subjects who suffer from gingivitis. Hot springs harbour diverse and interesting groups of microorganisms adapted to extreme conditions. However, due to limitations in the culture-dependent approach, most of such thermophiles remain uncultured and unexplored. Hence, this study was conducted to gain a comprehensive understanding of the bacterial diversity of Mahapelessa hot spring, Sri Lanka using both culture-dependent and culture-independent approaches. The in situ temperature of the water sample was 44.5 °C and the pH was 8.14. 16S rRNA Sanger sequencing of DNA extracted from the 18 bacterial isolates revealed the presence of eight genera belonging to two phyla: Proteobacteria (84%) and Firmicutes (16%) and the most abundant genus being Klebsiella. A total of 23 bacterial phyla representing 80 classes, 43 orders, 123 families, 205 genera and 83 species were detected by 16S rRNA V3-V4 region by amplicon metagenome sequencing of DNA extracted from water samples, where the most abundant phylum was the Proteobacteria (57.39%), followed by Firmicutes (23.7%) and Chloroflexi (4.14%). The three phyla Actinobacteria, Planctomycetes and Bacteroidetes were also detected less than 3% in abundance while 4.48% of bacteria could not be fit into any known phylum. The most abundant genera were Burkholderia (14.87%), Desulfotomaculum (7.23%) and Stenotrophomonas (6.1%). Four strictly anaerobic bacteria, Anaerosolibacter carboniphilus (0.71%), Bellilinea caldifistulae (0.04%), Salimesophilobacter vulgaris (0.1%), Anaerobacterium chartisolvens (0.12%); two potential plant growth-promoting bacteria, Azospirillum halopraeferens (0.04%) and Bradyrhizobium liaoningense (0.16%) and one potential alkali tolerant and sulphate-reducing bacterium, Desulfovibrio alkalitolerans (0.45%) were recorded. Pigmentiphaga sp. was isolated from Mahapelessa hot spring and to the best of our knowledge, this is the first record of this genus from a hot spring. This study gives insight into the vast bacterial diversity present in the Mahapelessa hot spring from the culture-independent approach which could not be identified using standard culturing techniques.

Periprosthetic joint infection (PJI) represents a major cause of failure following hip replacement surgery, setting an immense financial cost on society. Despite the fact that a variety of bacteria is incriminated for these infections, PJIs caused by anaerobic bacteria are the most devastating. leading to serious complications. Data on diagnosis and treatment of anaerobic PJIs are mainly gained from case reports, along with a few studies. Peptoniphilus asaccharolyticus is a rare Gram-positive anaerobic coccus, rarely, isolated from osseous infections. There is no recent literature data reporting isolation of Peptoniphilus asaccharolyticus from periprosthetic joint infections. The purposes of this case report are to present a rare periprosthetic joint infection caused by an anaerobic bacterium (Peptoniphilus asaccharolyticus), provide details on diagnosis and management of this coccus, and report recent literature data on periprosthetic joint infections by anaerobic bacteria.

BACKGROUND Acne vulgaris is a chronic inflammatory skin disease of the pilosebaceous units which can affect the individual's physiological and psychological health. Abnormal growth of lipophilic anaerobic bacteria such as Propionibacterium acnes is reported to be a major factor in the development of acne. However, the relationship between skin microorganisms and acne has not been fully elucidated. Our study aimed to explore the microbial differences between patients with acne and healthy controls (HCs). METHODS The study involved 16 participants diagnosed with acne vulgaris and 5 HCs. We collected skin microbe samples from the cheeks, brow, forehead, neck, chin, or chest of the participants with sterile cotton swabs depending on the location of the acne lesions. Cutaneous microbe samples from the participants were tested by 16s sequencing. RESULTS Patients with acne showed increased diversity of skin microbiota in their samples. OTU535601 (Lachnospiraceae), OTU4460604 (Clostridiales), OTU3217705 (Moraxellaceae), OTU1066814 (Prevotella), and OTU455671 (Lactococcus garvieae) were the top 5 most abundant species found in patients with acne but were not present in HCs. OTU423327 (Achromobacter), OTU4423360 (Stenotrophomonas), OTU993127 (Porphyromonas), OTU677680 (Prevotella), and OTU269901 (Pseudomonas) were the top 5 most abundant species in HCs but were not found in patients with acne. CONCLUSIONS The present study has analyzed and compared the diversity and abundance of microorganisms and the characteristics of the main pathogenic bacteria in patients with acne and HCs. Our findings indicate the importance of maintaining the skin's commensal microflora balance with the development of acne vulgaris.

Thermophilic anaerobic bacteria have received increasing interest as a promising bioprocessing platform for bioethanol production due to their broad substrate range and higher operating temperature. However, ethanol concentration is still lower than the commercial requirements on account of the high by-product formation and low ethanol tolerance. Our previous transcriptome analysis showed the gene expression level of pyruvate formate lyase-activating protein (PflA) was significantly down-regulated when Thermoanaerobacterium aotearoense SCUT27/Δldh was stimulated by various concentrations of ethanol. Based on this, we constructed the mutant strain T. aotearoense SCUT27/Δldh/ΔpflA, which not only possesses higher ethanol tolerance and production but also has enhanced ethanol/acetate ratio that is important for economical ethanol production. Compared to T. aotearoense SCUT27/Δldh, the mutant strain could resist to 5% (v/v) exogenous ethanol and produce 49 % more ethanol (38.82 g/L vs. 26.05 g/L) in 5-L bioreactor. In addition, the final concentration of ethanol reached 45.01 g/L by immobilized cell fermentation, which is the highest value obtained in T. aotearoense SCUT27. Simultaneously, the ethanol/acetate ratio of the mutant strain achieved 6.82, which was 2.16-fold higher than that of the parental strain.

Objective: No information exists about whether acute histologic chorioamnionitis (acute-HCA) is more advanced and severe, and intra-amniotic inflammation is more frequent and intense according to outside in neutrophil migration within the same chorio-decidua. The objective of current study is to examine this issue. Materials and methods: We included 106 singleton preterm-births (gestational age at delivery: 20–34 weeks) due to either preterm-labor or preterm-PROM in the context of acute chorio-deciduitis. Study-population was divided into 3 groups according to outside-in neutrophil migration within chorio-decidua as follows: 1) group-1: ‘inflammation restricted to the decidua’ (n = 22); 2) group-2: ‘inflammation restricted to the MT of chorion and the decidua’ (n = 31); 3) group-3: ‘inflammation in the CT of chorion’ (n = 53). We examined the frequency of inflammation in each placental compartment beyond chorio-decidua (i.e., amnion, umbilical cord, and chorionic-plate), and total grade (1–8) of acute-HCA. Moreover, the frequency of intra-amniotic infection (defined as positive amniotic-fluid culture for aerobic and anaerobic bacteria and genital mycoplasmas) and intra-amniotic inflammation (defined as amniotic fluid WBC ≥ 19 cells/mm3), and an intra-amniotic inflammatory response gauged by amnioticfluid WBC count (cells/mm3) were examined in 50 amniotic fluid samples within 7 days of birth. We identified a suite of steroid biomarkers in sediments and surface soils around Lake Ximencuo, a typical glacial lake in the Nianbaoyeze region, eastern Qinghai-Tibet Plateau (QTP). Various sterols, stanols, sterones, and sterenes appear in the sediments and surface soil samples. Among these steroids, plant-derived β-sitosterol and the 5α-stigmastanol that is derived from it exhibit high relative abundances. Notably, we found that the relative abundance of 5α-stigmastanol and the 5α-stigmastanol/n-alkanol ratio in a core of Lake Ximencuo sediment increased significantly in the last 200–300 year. These biomarker changes are broadly consistent with the growth of human settlement and the rapid development of animal husbandry in this region, which resulted in the former nomadic pastoral lifestyles gradually being transformed into permanent settlements since the early eighteenth century. The environmental and cultural changes led to greater delivery to soils around the lake of β-sitosterol that was transformed into 5α-stigmastanol by microbial communities in soil horizons, which include aerobic and anaerobic bacteria, and eventually surface runoff transport of the increased abundance of 5α-stigmastanol into Lake Ximencuo. We further postulate that the progressively greater human impacts in the Nianbaoyeze region over the past 200–300 year have modified the kinds of microbes that participate in the biogeochemical cycles of local soils.

Sulfonates include diverse natural products and anthropogenic chemicals and are widespread in the environment. Many bacteria can degrade sulfonates and obtain sulfur, carbon, and energy for growth, playing important roles in the biogeochemical sulfur cycle. Cleavage of the inert sulfonate C–S bond involves a variety of enzymes, cofactors, and oxygen-dependent and oxygen-independent catalytic mechanisms. Sulfonate degradation by strictly anaerobic bacteria was recently found to involve C–S bond cleavage through O2-sensitive free radical chemistry, catalyzed by glycyl radical enzymes (GREs). The associated discoveries of new enzymes and metabolic pathways for sulfonate metabolism in diverse anaerobic bacteria have enriched our understanding of sulfonate chemistry in the anaerobic biosphere. An anaerobic environment of particular interest is the human gut microbiome, where sulfonate degradation by sulfate- and sulfite-reducing bacteria (SSRB) produces H2S, a process linked to certain chronic diseases and conditions.

Background: Flavonifractor plautii is a strictly anaerobic rod shaped bacterium belonging to the family of Clostridiales. It is a commensal of the human intestinal microbiota which was seldom isolated from clinical samples, therefore clinical data are scarce. To date, only four cases of F. plautii infections were described, all occurring in immunosuppressed patients. Case presentation: We report a case where F. plautii was isolated from the blood culture of a severe burn victim and identified by matrix-assisted laser desorption ionization time-of-flight mass spectrometry. Discussion: To the best of our knowledge, this is the first case of F. plautii blood stream infection described in a burn patient.

Microaerobic hydrolysis and acidification (MHA), as a promising pre-treatment method of industrial wastewater, is drawing increasing attention to enhance the hydrolysis-acidification rate and inhibit the production of toxic gas H2S. In the present work, a pilot-scale MHA reactor coupled with anoxic-oxic (A/O) processes for treating the petrochemical wastewater was established and the mechanism and application of the MHA reaction were explored. The results showed that the ratio of VFA/COD was increased by 43-90% and low effluent S2- concentration (less than 0.2 mg/L) was obtained after MHA treatment with 5.5-13.8 L air m-3 h-1 supply. The MHA sludge exhibited a good settleability, a higher protease activity and plentiful community diversity. In addition to the dominant anaerobic bacteria responsible for hydrolysis and acidification such as Clostridiales uncultured, Anaerovorax, Anaerolineaceae uncultured and Fastidiosipila, the sulfate reducing bacteria involving Desulfobacter, Desulfomicrobium and Desulfobulbus, the sulphur oxidizing bacteria involving Thiobacillus, Arcobacter and Limnobacter, the nitrifies such as Nitrosomonadaceae uncultured and Nitrospira, and denitrifies Thauera were also identified. MHA pre-treatment guaranteed the efficacy and stability of the following A/O treatment. The removal efficiency of COD and ammonium of the MHA-A/O system remained at around 78.3% and 80.8%, respectively, although the organic load fluctuated greatly in the influent.

Lemierre's syndrome (LS) is characterized by septic thrombophlebitis of the internal jugular vein with septicemia and metastatic infection following an oropharyngeal infection. LS is rare but can cause infective endocarditis (IE), complicating IE management. We report a case of IE secondary to thrombophlebitis in the left vertebral vein following pharyngitis (LS variant) with distinctively severe manifestations, including metastatic infection and severe neurological impairment with multiple cerebral infarctions. A pedunculated abscess was noted on the left ventricular free wall. Despite the patient's highly impaired consciousness level (i.e., comatose state), we performed early surgery to remove the abscess after excluding LS-related brain complications. Preoperative antibiotics included clindamycin to cover LS-related anaerobic bacteria, and thrombophlebitis required postoperative anticoagulation. By managing LS as well as IE, the infection was controlled, and the neurological status normalized. This report provides insights into the perioperative management of IE secondary to LS.

Intermittent fasting (IF) is the practice of restricting food intake for 12-48 hours per fasting cycle over a prolonged period of time. Previous study shows beneficial health effects such as weight loss and lower risk for cardiometabolic diseases. Although reduced calorie intake may account for some of the observed benefits of IF, exact mechanisms are still unclear. Recent evidence indicates that IF may lead to remodeling and increased taxonomic diversity in the human gut microbiome. In particular, the Lachnospiraceae family of anaerobic bacteria increased during fasting. This family, in the order Clostridiales, promotes butryogenesis in the gut, a process that is associated with healthful metabolic and prolongevity effects. IF-associated alterations to the microbiome may play a key role in the metabolic and potential healthspan-enhancing benefits of IF and dietary restriction.

Methylmercury (MeHg) is a highly toxic form of mercury that can bioaccumulate in fish tissue. Methylmercury is produced by anaerobic bacteria, many of which are also capable of MeHg degradation. In addition, demethylation in surface waters can occur via abiotic sunlight-mediated processes. The goal of the present study was to understand the relative importance of microbial Hg methylation/demethylation and abiotic photodemethylation that govern the mass of MeHg within an aquatic system. The study location was the Hells Canyon complex of 3 reservoirs on the Idaho–Oregon border, USA, that has fish consumption advisories as a result of elevated MeHg concentrations. Our study utilized stable isotope addition experiments to trace MeHg formation and degradation within the water column of the reservoirs to understand the relative importance of these processes on the mass of MeHg using the Water Quality Analysis Simulation Program. The results showed that rates of MeHg production and degradation within the water column were relatively low (0.5 m below the surface. These results can be used to identify the relative importance of MeHg processes that can help guide reservoir management decisions.

Vaginal microbiota provide the first line of defense against urogenital infections primarily through protective actions of Lactobacillus species Perceived stress increases susceptibility to infection through several mechanisms, including suppression of immune function. We investigated whether stress was associated with deleterious changes to vaginal bacterial composition in a subsample of 572 women in the Longitudinal Study of Vaginal Flora, sampled from 1999 through 2002. Using Cox proportional hazards models, both unadjusted and adjusted for sociodemographic factors and sexual behaviors, we found that participants who exhibited a 5-unit-increase in Cohen’s Perceived Stress Scale had greater risk (adjusted hazard ratio (HR) = 1.40, 95% confidence interval (CI): 1.13, 1.74) of developing molecular bacterial vaginosis (BV), a state with low Lactobacillus abundance and diverse anaerobic bacteria. A 5-unit increase in stress score was also associated with greater risks of transitioning from the L. iners-dominated community state type (26% higher) to molecular-BV (adjusted HR = 1.26, 95% CI: 1.01, 1.56) or maintaining molecular-BV from baseline (adjusted HR = 1.23, 95% CI: 1.01, 1.47). Inversely, women with baseline molecular-BV reporting a 5-unit stress increase were less likely to transition to microbiota dominated by L. crispatus, L. gasseri, or L. jensenii (adjusted HR = 0.81, 95% CI: 0.68, 0.99). These findings suggest that psychosocial stress is associated with vaginal microbiota composition, inviting a more mechanistic exploration of the relationship between psychosocial stress and molecular-BV.

HPVs representing the most common sexually transmitted disease are a group of carcinogenic viruses with different oncogenic potential. The immune system and the vaginal microbiome represent the modifiable and important risk factors in HPV-induced carcinogenesis. HPV infection significantly increases vaginal microbiome diversity, leading to gradual increases in the abundance of anaerobic bacteria and consequently the severity of cervical dysplasia. Delineation of the exact composition of the vaginal microbiome and immune environment before HPV acquisition, during persistent/progressive infections and after clearance, provides insights into the complex mechanisms of cervical carcinogenesis. It gives hints regarding the prediction of malignant potential. Relative high HPV prevalence in the general population is a challenge for modern and personalized diagnostics and therapeutic guidelines. Identifying the dominant microbial biomarkers of high-grade and low-grade dysplasia could help us to triage the patients with marked chances of lesion regression or progression. Any unnecessary surgical treatment of cervical dysplasia could negatively affect obstetrical outcomes and sexual life. Therefore, understanding the effect and role of microbiome-based therapies is a breaking point in the conservative management of HPV-associated precanceroses. The detailed evaluation of HPV capabilities to evade immune mechanisms from various biofluids (vaginal swabs, cervicovaginal lavage/secretions, or blood) could promote the identification of new immunological targets for novel individualized diagnostics and therapy. Qualitative and quantitative assessment of local immune and microbial environment and associated risk factors constitutes the critical background for preventive, predictive, and personalized medicine that is essential for improving state-of-the-art medical care in patients with cervical precanceroses and cervical cancer. The review article focuses on the influence and potential diagnostic and therapeutic applications of the local innate immune system and the microbial markers in HPV-related cancers in the context of 3P medicine.

In an attempt to find versatile and readily accessible coumarin scaffolds with a broad spectrum of biomedical properties, seven bisimidazole–coumarin conjugates were synthesized through several steps starting from metacetamol and malonic acid. Spectroscopical techniques including FTIR, 1H-NMR, and 13C-NMR were employed to confirm the chemical backbone of the synthesized conjugates. Their biomedical potentials to behave as anti-tumorous, anti-aerobic gram-negative bacterial, anti-anaerobic bacterial, antifungal, free-radical quencher, and anticoagulant agents were evaluated. The anti-tumorous potential was investigated via an MTT-based assay against eight neoplastic-cell lines, while the anti-aerobic gram-negative bacterial potential was inspected using a broth-dilution assay against six standard bacterial strains. The anti-anaerobic bacterial potential was detected utilizing a Brucella-agar dilution assay against four standard bacterial strains, whereas the antifungal potential was estimated via a Sabouraud-dextrose broth dilution method against two standard fungal strains. The antioxidant potential was determined by following the capacity of the conjugates to quench the hydroxyl and DPPH radical phenotypes. Finally, the anticoagulant potential was detected in vivo by monitoring prothrombin-time following oral administration in rabbits. From the gathered outcomes, the author reported three main results. The conjugates have promising and broad biomedical effects. Conjugate Y7 has a potent and broad-range antineoplastic effect with a high capacity to quench the damaging free radicals, while being free from anticoagulant activity. Conjugate Y1 exhibited a potent antimicrobial activity with a spectrum involving pathogenic aerobic and anaerobic bacteria, and fungi. It is concluded that conjugate Y7 may represent a potential applicant as an antineoplastic agent, while conjugate Y1 may afford an agent that is free from anticoagulant activity with high potency as a broad spectrum antimicrobial compound.

Leg ulcers are estimated to occur in 1%–10% of North American patients with sickle cell disease (SCD). Their pathophysiology remains poorly defined, but as with other chronic wounds, it is hypothesised that the microbial milieu, or microbiome, contributes to their healing and clinical outcomes. This study utilises 16S ribosomal RNA (rRNA) gene sequencing to describe, for the first time, the microbiome of the SCD leg ulcer and its association with clinical factors. In a cross‐sectional analysis of 42 ulcers, we recovered microbial profiles similar to other chronic wounds in the predominance of anaerobic bacteria and opportunistic pathogens including Staphylococcus, Corynebacterium, and Finegoldia. Ulcers separated into two clusters: one defined by predominance of Staphylococcus and smaller surface area, and the other displaying a greater diversity of taxa and larger surface area. We also find that the relative abundance of Porphyromonas is negatively associated with haemoglobin levels, a key clinical severity indicator for SCD, and that Finegoldia relative abundance is negatively associated with CD19+ B cell count. Finally, ratios of Corynebacterium:Lactobacillus and Staphylococcus:Lactobacillus are elevated in the intact skin of individuals with a history of SCD leg ulcers, while the ratio of Lactobacillus:Bacillus is elevated in that of individuals without a history of ulcers. Investigations of the skin microbiome in relation to SCD ulcer pathophysiology can inform clinical guidelines for this poorly understood chronic wound, as well as enhance broader understanding about the role of the skin microbiome in delayed wound healing.

The aim of the study was to evaluate the pathogenic potential of Bacteroides pyogenes, rarely identified in clinical laboratories anaerobic bacteria. To increase the knowledge about this poorly understood anaerobic microorganism, the study also includes cases of infections described so far in the literature. Only the use of 16S rRNA sequencing and mass spectrometry technique allowed the identification of B. pyogenes from clinical specimens. We reported 13 severe human infections caused by B. pyogenes. Bacteria were cultured from the wound after biting by animals, chronic infections within the oral cavity, from patients with histologically or radiological proven osteomyelitis, surgical site infection, and from urine sample collected after a urological procedure. Most (9/13) of the patients required hospitalization. Almost 70% of them needed urgent admission via the emergency room. Two inpatients due to a life-threatening condition were admitted to the intensive care unit. Almost 50% of isolates were resistant to penicillin. All resistant to penicillin strains were isolated from skin and mucous membrane infections.

Volatile organic compounds (VOCs) become concerned pollutants in landfill gases, and their composition and concentration varied significantly during waste decomposition. Many environmental factors are known to affect VOC emissions, while the effect of indigenous bacteria in wastes on VOC production remains elusive. In this study, a simplified anaerobic degradation experiment, with the single substrate and the purified bacteria from a landfill, was set up to measure the degradation process and the dynamic changes of VOCs. The experiment excluded the abiotic factors for VOC variation. The two isolated bacteria, identified as Sporanaerobacter acetigenes and Clostridium sporogenes, could anaerobically ferment amino acids by Stickland reaction. They produced 51 and 57 species of VOCs in the experiment, respectively. The concentration changes of VOCs over bacterial growth and fermentation were clustered into four types by principal component analysis: three profiles were regular, similar to the variation of nitrate, hydrogen sulfide, and the major fermentation products (carbon dioxide, ammonium, and volatile organic acids), respectively; while one profile was unique to any degradation indicator. The various concentration profiles indicated different origins for VOCs, possibly from the extracellular environment, fermentation, and secondary reactions. The findings provide insights into the understanding of VOC diversity and variability during waste decomposition.
