This study aims at the synthesis and initial biological evaluation of novel rhenium–tricarbonyl complexes of 3,3′,4′,5,7-pentahydroxyflavone (quercetin), 3,7,4΄-trihydroxyflavone (resokaempferol), 5,7-dihydroxyflavone (chrysin) and 4΄,5,7-trihydroxyflavonone (naringenin) as neuroprotective and anti-PrP agents. Resokaempferol was synthesized from 2,2΄,4-trihydroxychalcone by H2O2/NaOH. The rhenium–tricarbonyl complexes of the type fac-[Re(CO)3(Fl)(sol)] were synthesized by reacting the precursor fac-[Re(CO)3(sol)3]+ with an equimolar amount of the flavonoids (Fl) quercetin, resokaempferol, chrysin and naringenin and the solvent (sol) was methanol or water. The respective Re–flavonoid complexes were purified by semi-preparative HPLC and characterized by spectroscopic methods. Furthermore, the structure of Re–chrysin was elucidated by X-ray crystallography. Initial screening of the neuroprotective properties of these compounds included the in vitro assessment of the antioxidant properties by the DPPH assay as well as the anti-lipid peroxidation of linoleic acid in the presence of AAPH and their ability to inhibit soybean lipoxygenase. From the above studies, it was concluded that the complexes’ properties are mainly correlated with the structural characteristics and the presence of the flavonoids. The flavonoids and their respective Re-complexes were also tested in vitro for their ability to inhibit the formation and aggregation of the amyloid-like abnormal prion protein, PrPSc, by employing the real-time quaking-induced conversion assay with recombinant PrP seeded with cerebrospinal fluid from patients with Creutzfeldt–Jakob disease. In the present study, we developed new functional food materials from chickpea (Cicer arietinum) milk (CM) with desirable nutritional value, health functions, and rheology to mitigate lifestyle and age-related diseases.The anti-oxidant, anti-glycation, and bile acid-lowering properties of CM fermented with the lactic acid bacteria Lactiplantibacillus pentosus Himuka-SU5 (LpFCM) and Lactococcus lactis subsp. lactis Amami-SU1 (LcFCM) were determined in vitro. L. pentosus Himuka-SU5 and L. lactis Amami-SU1 were found to lower the pH from 6.4 to 4.7 at 12 and 48 h, respectively.The lactic acid concentration in LpFCM and LcFCM was 6.9 and 4.4 mg/mL, respectively. Both starters were found to degrade the dissolved proteins in CM, and L. lactis Amami-SU1 produced ammonia.Though the total phenolic content was slightly lower in LpFCM and LcFCM than in unfermented CM, O2− radical-scavenging capacity, 1,1-diphenyl-2-picrylhydrazyl radical-scavenging capacity, and Fe-reducing power were high in both LpFCM and LcFCM. Anti-glycation in a bovine serum albumin-fructose model and the bile acid-lowering capacities of CM were distinctly increased following fermentation.Based on the results of the present study, we inferred that fermented CM can be considered a desirable food material to prevent and ameliorate chronic lifestyle diseases, particularly in the elderly., 公開日: 2023-06-06

This study was aimed to determine the total phenolics contents and antioxidant capacity of white and brown teff grain samples. The free total polyphenolic content (TPPC), bound TPPC and total flavonoids content ranged from 62.1–129.9, 84.6–189.6 mg GAE/100g and 84.4–195.1 mg QE/100g, respectively, in white teff samples, and, 118.6–196.7, 141.1–195.1mgGAE/100g and 97.8–202.5 mg QE/100g, respectively, in brown teff samples. Besides, the free 2,2-diphenyl-1-picrylhydrazyl (DPPH) and bound DPPH ranged from 74.8–98.3, 77.1–99.9 mg AAE/100g, respectively, for white teff samples, and, 68.7–93.1 and 71.2–99.4 mg AAE/100g, respectively, for brown teff samples. This study revealed that total phenolics content was higher in brown teff samples than white teff samples and is in agreement with other reports. However, their DPPH scavenging activities were nearly equal, indicating that both varieties of teff are relevant for human nutrition and health. Furthermore, the dendrogram has shown sharp separation of the samples based on their origin and variety related to the total phenolics parameters. Hence, findings of this study can help consumers appreciate the nutritional value of white and brown teff grains; provide them guidance for teff purchase and production. Oxidative stress and cellular senescence in dermal papilla cells (DPCs) are major etiological factors causing hair loss. In this study, the effect of the Allium hookeri extract (AHE) on hair-inductive and anti-oxidative properties was investigated in human DPCs. As a result, it was found that a non-cytotoxic concentration of the extracts increased the viability and size of the human DPC spheroid, which was associated with the increased expression of hair-growth-related genes in cells. To determine whether or not these effects could be attributed to intracellular anti-oxidative effects, liquid chromatography-mass spectrometry alongside various biochemical analyses are conducted herein. An ingredient called alliin was identified as one of the main components. Furthermore, AHE treatment induced a significant decrease in H2O2-mediated cytotoxicities, cell death, and cellular senescence in human DPCs. Upon analyzing these results with a molecular mechanism approach, it was shown that AHE treatment increased β-Catenin and NRF2 translocation into the nucleus while inhibiting the translocation of NF-κB (p50) through p38 and PKA-mediated phosphorylations of GSK3β, an upstream regulator of those proteins. These results overall indicate the possibility that AHE can regulate GSK3β-mediated β-Catenin, NRF2, and NF-κB signaling to enhance hair-inductive properties and ultimately protect against oxidative stress-induced cellular damage in human DPCs.

Ototoxic hearing loss due to antibiotic medication including aminoglycosides and excess free radical production causes irreversible hair cell injury. Cichoric acid, a naturally occurring phenolic acid, has recently been found to exert anti-oxidative and anti-inflammatory properties through its free radical scavenging capacity. The present study aimed to investigate the protective effects of cichoric acid against neomycin-induced ototoxicity using transgenic zebrafish (pvalb3b: TagGFP). Our results indicated that cichoric acid in concentrations up to 5 μM did not affect zebrafish viability during the 2 h treatment period. Therefore, the otoprotective concentration of cichoric acid was identified as 5 μM under 2 h treatment by counting viable hair cells within the neuromasts of the anterior- and posterior-lateral lines in the study. Pretreatment of transgenic zebrafish with 5 μM of cichoric acid for 2 h significantly protected against neomycin-induced hair cell death. Protection mediated by cichoric acid was, however, lost over time. A terminal deoxynucleotidyl transferase dUTP nick-end labeling (TUNEL) assay and FM4-64 staining, respectively, provided in situ evidence that cichoric acid ameliorated apoptotic signals and mechanotransduction machinery impairment caused by neomycin. A fish locomotor test (distance move, velocity, and rotation frequency) assessing behavioral alteration after ototoxic damage revealed rescue due to cichoric acid pretreatment before neomycin exposure. These findings suggest that cichoric acid in 5 μM under 2 h treatment has antioxidant effects and can attenuate neomycin-induced hair cell death in neuromasts. Although cichoric acid offered otoprotection, there is only a small difference between pharmacological and toxic concentrations, and hence cichoric acid can be considered a rather prototypical compound for the development of safer otoprotective compounds.

The aim of this study was to determine the effect of both the Solid State Fermentation (SSF) technique and the use of Rhizopus oligosporus on the physicochemical changes of fermented de-husked rice flour. Three varieties of de-husked rice, i.e., Mentik Wangi Susu (non-pigmented), red Cempo Merah, and black Jowo Melik (pigmented) were fermented using Rhizopus oligosporus. Fermentation was performed at room temperature with a fermentation time of 0, 24, 48, and 72 hours. The analyzed parameters were proximate composition, bio-active compounds, and pasting profile. The results showed an increase in flour pasting profile, ash, protein, and fat content after the fermentation. The total availability of the total phenolic content (TPC) and antioxidant capacity were also increased. The highest TPC (0.37 mg GAE/g) and antioxidant capacity (1.43 mg TEAC/g) were obtained in the Jowo Melik variety at 72 hours of fermentation. In contrast, anthocyanin and carbohydrate contents decreased as fermentation time increased. The highest anthocyanin content of 0.53 mg/g (after 24-hour fermentation) was obtained in the Jowo Melik variety. In conclusion, 72-hour-fermented black rice flour (Jowo Melik) has a higher potential to be developed as a functional food.

Exposure to arsenic leads to alterations of the cellular machinery and eventually cell death, depending on the dose and duration of exposure. Oxidative stress induced by arsenic is the major mechanism by which it inflicts cellular toxicity, challenging the survival-support - autophagy and culminating in apoptosis in the thymus and spleen of mice. Curcumin, a potent dietary anti-oxidant with known anti-apoptotic and anti-inflammatory properties, was assessed for therapeutic benefits. However, the major caveat of this polyphenol is its low water solubility and limited bioavailability. Therefore, Self Nano-Emulsifying Curcumin (SNEC30) was used to treat mice exposed to arsenic. When administered, SNEC30 effectively ameliorated the adverse effects of arsenic in mice, by restoring structural alterations and reducing ROS-mediated cell death, thereby endorsing the importance of nutraceuticals in counteracting heavy metal-induced cellular toxicity.

Background Chronic hyperglycemia induced oxidative stress and dyslipidemia in diabetic nephropathy may lead to chronic renal damage. Thus, counteracting oxidative stress might represent an interesting approach in alleviating hyperglycemia-induced renal damage. Objective The present experimental work was undertaken to explore nephroprotective efficacy of Curculigo orchiodies in streptozotocin-nicotinamide induced diabetic nephropathy in laboratory animals. Materials and methods Single intraperitoneal introduction of freshly prepared STZ (65 mg/kg) was used for induction of diabetic nephropathy in rats, 15 min after NAD administration (230 mg/kg; i.p.). The evaluation of nephropathy was done by assessment of serum glucose level, insulin level and renal function test (albumin, urea and creatinine). In addition to this, lipid profile as well as oxidative stress (TBARS, superoxide dismutase, catalase and reduced glutathione) was evaluated. Augmented levels of blood glucose, albumin, urea and creatinine confirmed the development of nephropathic symptoms in rats. After 30 days of STZ administration, different doses (150, 300 mg/kg and 600 mg/kg; p.o.) of hydroalcoholic and ethanolic extracts of C. orchiodies were administered to rats for 45 days. Conclusion Curculigo orchiodes significantly attenuated hyperglycemia induced increase in lipid profile, oxidative stress and normalized the renal functions (albumin, urea and creatinine); attributing to the efficacy of C. orchiodies in diabetic nephropathy. These findings suggest that hydroalcholic and ethanolic extract of Curculigo Orchiodes ameliorated the progression of diabetic nephropathy. The observed nephroprotective effect of C. orchiodes is attributed to its hypoglycemic, antioxidant and anti-hyperlipidemic activity.

Couple of ethnopharmacological surveys in the Indian Ladakh and Pakistani Shigar valleys has reported the medicinal use of Acantholimon lycopodioides against cardiac and gastric disorders that however, remains without scientific rationale or experimental validations. Here, we assess the in vitro bio/therapeutic activities of A. lycopodioides extracts as well as chloroform, ethyl acetate, n-butanol and aqueous fractions. The in vitro β-carotene-linoleic acid bleaching and DPPH radical scavenging methods demonstrated a very high anti-oxidative property of chloroform and ethyl acetate fractions compared to others. Cell viability assay (MTT) on human cervical (HeLa), breast (MDA-MB321) and liver (HepG2) cancer cells revealed their differential cytotoxicity, except the chloroform fraction. Of these, the precipitate exerted highest cytotoxicity on HepG2 cells followed by aqueous fraction on MDA-MB321 cells. Notably, the non-cytotoxicity of chloroform fraction coincided with its highest anti-oxidative activity. Further, the chloroform fraction showed marked hepatoprotection (up to 84%) against 3’7’dichlorofluorescin triggered free radicals induced oxidative damage. Also, the hepatoprotective chloroform fraction mildly activated CYP3A4 in HepG2 cells (dual-luciferase assay). Moreover, the A. lycopodioides extracts and fractions showed differential anti-bacterial and anti-fungal activities. Of these, while S. aureus was more sensitive to the water-insoluble extract, ethyl acetate fraction showed moderate activity against E. coli and C. albicans. On the other hand, the chloroform fraction showed promising activity against S. Aureus, C. albicans, P. vulgaris and E. faecalis. In conclusion, our data for the first time, demonstrated promising anti-oxidative, hepatoprotective, anti-cancer, anti-microbial and CYP3A4 activating salutations of A. lycopodioides. This warrants further studies towards isolation and identification of its therapeutically active principles.

Introduction:Equisetum arvense (E. arvense) is an herbal medicinal plant that is mostly found in Limpopo and the North West Provinces of South Africa. Traditionally, the infusions and decoction of this plant are used in the management of type 2 diabetes, compromised immune system and oxidative stress related disorders. We investigated E. arvense in an attempt to authenticate its traditional use by cost-effective thin layer chromatography. Methods:The anti-diabetic activity was assayed by spraying a developed TLC plate with a buffered β-glucosidase enzyme and bovin solution and incubating at 36.9°C for 20 mins. A freshly prepared solution of 2-naphthyl-β-D-glucopyranoside and fast blue salt (1:4 v/v) was used for detecting inhibitory activity of the enzyme by the plant extract. Immune boosting potentials were evaluated by analysing for the presence or absence of β-sitosterol, a known immune booster in the 3 extracts (chloroform, ethanol, and water) and enriched fractions E and F. The antioxidant activity was investigated using 2,2-Diphenyl-1-Picryl-Hydrazyl (DPPH), H2O2 solution free radical scavenging as well as the reducing power of iron (+3) to (+2) by the extracts and enriched fractions. Standards and positive controls were used for the respective assays conducted. Results:Results obtained indicated the potentials of E. arvense extracts and enriched fractions as antidiabetic (inhibits β-glucosidase implicated in type 2 diabetes), immune boosting (presence of β-sitosterol) and antioxidant (IC50 values of 5.48 ± 0.13 - 12.38 ± 0.09 for FRAP, 12.00 ± 0.02 – 14.01 ± 0.23 for DPPH and 18.07 ± 0.06 - 62.01 ± 0.18 for H2O2) assays. The quantified amount of β-sitosterol and the antidiabetic compound in the plant ethanol extract were determined as 323.85 ng/mg and 130.04 mg/ml or 1.3 x 1011 ng/ml respectively. Conclusions:This study has preliminarily authenticated the traditional uses of E. arvense by rural South Africans.

Background: Anethole is the principle essential oil component of anise and fennel. Renal ischemia/reperfusion (RIR) is one of the utmost imperative reasons for acute kidney injury and often associated with high mortality rate. The aim of this study is to investigate the protective effect of anethole on RIR status, exploring the involved mechanisms. Methods: RIR was accomplished by bilateral renal pedicle clamping for 45 min, after which the clamps were removed to achieve the reperfusion phase. Rats were randomized into five groups; Sham, Sham + anethole, RIR, and finally RIR + anethole (125 mg/kg or 250 mg/kg) groups. Animals were given anethole (in specified groups in doses) for 14 days before RIR. Results: RIR-experienced animals developed renal injury evidenced by diminished renal function and histopathological alteration. RIR induced severe oxidative, inflammatory, and apoptotic status within renal tissue. Pre-RIR management with anethole enhanced renal morphology and improved renal function. Anethole amplified GSH content and SOD, CAT, and GPx activities and lowered MDA. Anethole reduced gene and protein expression levels of HMGB1, TLR2, TLR4, MYD88, and NFκB. Anethole distinctly dropped TNF-α, IFN-γ, and MCP-1 levels, increased IL-10, and diminished caspase 3 and 9, reflecting its anti-inflammatory and anti-apoptotic actions. Conclusion: Anethole displayed anti-inflammatory, anti-oxidant, and anti-apoptotic actions against RIR-induced injury. Anethole exhibited renal protective actions, which could be through inhibiting the HMGB1/TLR2, 4/MYD88/NFκB pathway. These results could suggest anethole as a protective agent against RIR.

Background The genus Lycium is commercially known for its nutrient dense goji-berries, among these berries, black goji-berries obtained from Lycium ruthenicum Murr are highly valued and widely used as traditional medicine in trans-himalayan cold desert Ladakh and as functional food in several countries. Methods The current collection of data and literature was done by exploring different scientific portals like SciFinder, Google scholar, PubMed, Dictonary of Natural Products, Institute for Scientific Information, Web of Science and Scopus by searching keywords like black goji berry, crystal pearl, and trans-Himalayan plant. Results Fruits of L. ruthenicum Murr, are overwhelmingly enriched in anthocyanins, proanthocyanidins, polysaccharides, spermine and spermidine alkaloids. The presence of these bioactive phyto-chemicals has been linked with reported anti-diabetic, anti-inflammatory, anti-fatigue, anti-atherosclerosis and neuro-protective properties of black goji berries. A unique color of these berries makes them exceptional as compared to other berries. Conclusions In this article, we have reviewed the variety of high value phytochemicals of Lycium ruthenicum Murr, with a special focus on health promoting anthocyanins which will provide an insight to the readers for exploring novel applications of L. ruthenicum Murr in field of medicine and food industries.

The neoagarooligosaccharides have received growing attention owing to their physiological activities. The aim of this study was the isolation of agarase-producing bacteria for production of agar hydrolysates with special emphasis on their anti-oxidant potential. An agarolytic strain NI125 was isolated from Nelson's Island, Alexandria, Egypt. Based on 16S rRNA analysis and extensive phenotypic characterization, it was identified as Aquimarina agarilytica. Maximum enzyme production was achieved after 24 h incubation at 20°C, and tryptone was recorded to be the best nitrogen source for agarase production. Extracellular agarase was partially purified by ammonium sulfate precipitation. The substrate specificity assay using p-nitrophenyl-α/β-D-galactopyranoside revealed the cleavage of the β-linkage rather than the α-linkage. Neoagarooligosaccharides produced by the partially purified β-agarase expressed promising anti-oxidant properties, with 23% free radical scavenging potential. Notable enhancement of the anti-oxidant potency of the oligosaccharides was achieved (up to 87% scavenging ability) by sulfation of the agar prior to hydrolysis for 12 h with β-agarase. Results obtained suggest the potential application of the produced neoagarooligosaccharides anti-oxidants as promising additives in food and feed products.

Background Inflammation has continued to raise global challenges and Jatropha tanrogenesis (JT) is used traditionally for its management. In this study, the in silico and in vitro anti-inflammatory potential of bioactive sterols were investigated. The active compounds of ethanol extract of JT leaves were identified using Gas chromatography-mass spectrometry (GC.MS) followed by molecular docking against COX-1 and COX-2 using maestro Schrödinger and pharmacokinetic profile prediction using webserver tools. The in vitro anti-inflammatory and anti-oxidantive potentials were investigated using standard protocols. Results GC–MS analysis of ethanol extract of JT leaves revealed the presence of eight (8) compounds, the molecular docking analysis of these compounds demonstrated varying degrees of binding affinities against the target proteins. The extract exhibit concentration dependent anti-oxidant activity with IC50 of 106.383 and 6.00 Fe2+E/g for DPPH and FRAP respectively. The extract showed significant (P P > 0.05) increase was observed at 600 and 1000 µg/ml compared to 200 µg/ml of diclofenac sodium. Ulcerative colitis (UC) is an inflammatory bowel disease that affects the patients’ colorectal area culminating in an inflamed ‘leaky gut.’ The majority of UC treatments only provide temporary respite leading to its relapse. Therefore, this study investigated the efficacy of the enteric-coated ‘Cologrit’ (EC) tablet in alleviating UC-like inflammation. Cologrit is formulated using polyherbal extracts that have anti-inflammatory qualities according to ancient Ayurveda scriptures. Phytochemical profiling revealed the presence of gallic acid, rutin, ellagic acid, and imperatorin in Cologrit formulation. Cologrit treatment decreased inflammation in LPS-induced transformed THP-1 macrophages, and TNF-α-stimulated human colorectal (HT-29) cells through the modulation of NFκB activity, IL-6 production, and NFκB, IL-1β, IL-8, and CXCL5 mRNA expression levels. Cologrit also lessened human monocytic (U937) cell adhesion to HT29 cells. Methacrylic acid-ethylacrylate copolymer-coating of the enteric Cologrit tablets (EC) supported their dissolution, and the release of phytochemicals in the small intestine pH 7.0 environment in a simulated gastrointestinal digestion model. Small intestine EC digestae effectively abridged dextran sodium sulfate (2.5% w/v)-induced cell viability loss and oxidative stress in human colon epithelial Caco-2 cells. In conclusion, the enteric-coated Cologrit tablets demonstrated good small intestine-specific phytochemical delivery capability, and decreased UC-like inflammation, and oxidative stress through the regulation of TNF-α/NFκB/IL6 signaling axis.

Suaeda aegyptiaca is a halophytic plant widely growing in northeast Africa and Asia. The current study reports on the GC-MS analysis of S. aegyptiaca essential oil. The essential oil was prepared using three different methods: cold n-hexane extraction (CHE), hot n-hexane extraction (HHE), and hydro-distillation extraction (HDE). The GC-MS analysis detected twenty-eight compounds in both CHE (97.28%) and HHE (97.35%) and twenty compounds in HDE (98.65%). 2-methyloctacosane (48.72%); 11-decyldocosane (29.20%); and 1, 2-benzenedicarboxylic acid diisooctyl ester (57.87%) were the main constituents in CHE, HHE, and HDE, respectively. Free radical scavenging activity testing using 2,2-diphenyl-1-picrylhydrazyl (DPPH) revealed the notable anti-oxidant potential of HDE (IC50 0.358 mg/mL) compared to ascorbic acid (IC50 0.264 mg/mL). Moreover, in vitro anti-inflammatory activity testing using COX-1 and COX-2 showed the notable activity of HDE (IC50 5.50 µg/mL and 2.59 µg/mL, respectively). The observed anti-inflammatory activity of HDE was further confirmed by the characteristic decrease in TNF-α levels in RAW264.7 to 572.20 Pg/mL compared to the decrease of 442.80 Pg/mL caused by the positive control (Celecoxib®). On the other hand, a cytotoxic activity investigation indicated that CHE was the most potent against the Caco-2 and HCT-116 cell lines (IC50 8.11 and 11.18 µg/mL, respectively), and it was closely followed by HHE (IC50 12.42 µg/mL) against the Caco-2 cell line. Collectively, S. aegyptiaca essential oil prepared by the hydro-distillation method exhibited notable anti-oxidant and anti-inflammatory activities, while the same essential oil prepared by solvent extraction methods displayed a characteristic cytotoxic activity against the Caco-2 and HCT-116 cell lines. These results confirmed that different extraction methods greatly influence the biological potential of an essential oil, which, in turn, is attributable to the different constituents in each extract Moreover, S. aegyptiaca was noted as a promising halophytic plant for more phytochemical and biological investigations.

Urease is an amidohydrolase enzyme that is responsible for fatal morbidities in the human body, such as catheter encrustation, encephalopathy, peptic ulcers, hepatic coma, kidney stone formation, and many others. In recent years, scientists have devoted considerable efforts to the quest for efficient urease inhibitors. In the pharmaceutical chemistry, the thiourea skeleton plays a vital role. Thus, the present work focused on the development and discovery of novel urease inhibitors and reported the synthesis of a set of 1-aroyl-3-[3-chloro-2-methylphenyl] thiourea hybrids with aliphatic and aromatic side chains 4a–j. The compounds were characterized by different analytical techniques including FT-IR, 1H-NMR, and 13C-NMR, and were evaluated for in-vitro enzyme inhibitory activity against jack bean urease (JBU), where they were found to be potent anti-urease inhibitors and the inhibitory activity IC50 was found in the range of 0.0019 ± 0.0011 to 0.0532 ± 0.9951 μM as compared to the standard thiourea (IC50 = 4.7455 ± 0.0545 μM). Other studies included density functional theory (DFT), antioxidant radical scavenging assay, physicochemical properties (ADMET properties), molecular docking and molecular dynamics simulations. All compounds were found to be more active than the standard, with compound 4i exhibiting the greatest JBU enzyme inhibition (IC50 value of 0.0019 ± 0.0011 µM). The kinetics of enzyme inhibition revealed that compound 4i exhibited non-competitive inhibition with a Ki value of 0.0003 µM. The correlation between DFT experiments with a modest HOMO-LUMO energy gap and biological data was optimal. These recently identified urease enzyme inhibitors may serve as a starting point for future research and development.

Micronutrients extracted from natural plants or made by biological synthesis are widely used in anti-aging research and applications. Among more than 30 effective anti-aging substances, employing polyphenol organic compounds for modification or delaying of the aging process attracts great interest because of their distinct contribution in the prevention of degenerative diseases, such as cardiovascular disease and cancer. There is a profound potential for polyphenol extracts in the research of aging and the related diseases of the elderly. Previous studies have mainly focused on the properties of polyphenols implicated in free radical scavenging; however, the anti-oxidant effect cannot fully elaborate its biological functions, such as neuroprotection, Aβ protein production, ion channel coupling, and signal transduction pathways. Caenorhabditis elegans (C. elegans) has been considered as an ideal model organism for exploring the mechanism of anti-aging research and is broadly utilized in screening for natural bioactive substances. In this review, we have described the molecular mechanisms and pathways responsible for the slowdown of aging processes exerted by polyphenols. We also have discussed the possible mechanisms for their anti-oxidant and anti-aging properties in C. elegans from the perspective of different classifications of the specific polyphenols, such as flavonols, anthocyanins, flavan-3-ols, hydroxybenzoic acid, hydroxycinnamic acid, and stilbenes.

The aims of this study were to induce calli from the seeds of three rice varieties (Hommali 105, Munpu, and Niawdum) and investigate their anti-aging potential. First, rice seeds were cultured on a Murashige and Skoog medium (MS medium) supplemented with 2 mg/L of 2,4-Dichlorophenoxyacetic acid (2,4-D), 1 mg/L of 1-Naphthalene acetic acid (NAA), and 1 mg/L of 6-Benzylaminopurine (BAP). After three weeks, the calli were extracted with ethanol. Then, their phenolic contents were determined by spectrophotometer and the amino acids were identified by ultra-performance liquid chromatography (UPLC). Their cytotoxicity, anti-oxidant (potassium ferricyanide reducing power assay (PFRAP), DPPH radical scavenging assay (DPPH), lipid peroxidation inhibition (LPO), and superoxide dismutase activity (SOD)), and anti-aging (keratinocyte proliferation, anti-collagenase, anti-inflammation, and anti-tyrosinase) activities were also investigated. Munpu callus (385%) was obtained with a higher yield than Hommali (322%) and Niawdum (297%) calli. The results revealed that the phenolic and amino acid contents were enhanced in the calli. Moreover, the calli were rich in glutamic acid, alanine, and gamma aminobutyric acid (GABA). The callus extracts showed no cytotoxic effects at a concentration of equal to or lower than 0.25 mg/mL. The highest anti-oxidant activities (PFRAP (0.81 mg AAE/mL), DPPH (68.22%), LPO (52.21%), and SOD (67.16%)) was found in Munpu callus extract. This extract also had the highest keratinocyte proliferation (43.32%), anti-collagenase (53.83%), anti-inflammation (85.40%), and anti-tyrosinase (64.77%) activities. The experimental results suggest that the amounts of bioactive compounds and anti-aging activities of rice seeds can be enhanced by the induction of callus formation.

Celastrol (also called tripterine) is a quinone methide triterpene isolated from the root extract of Tripterygium wilfordii (thunder god vine in traditional Chinese medicine). Over the past two decades, celastrol has gained wide attention as a potent anti-inflammatory, anti-autoimmune, anti-cancer, anti-oxidant, and neuroprotective agent. However, its clinical translation is very challenging due to its lower aqueous solubility, poor oral bioavailability, and high organ toxicity. To deal with these issues, various formulation strategies have been investigated to augment the overall celastrol efficacy in vivo by attempting to increase the bioavailability and/or reduce the toxicity. Among these, nanotechnology-based celastrol formulations are most widely explored by pharmaceutical scientists worldwide. Based on the survey of literature over the past 15 years, this mini-review is aimed at summarizing a multitude of celastrol nanoformulations that have been developed and tested for various therapeutic applications. In addition, the review highlights the unmet need in the clinical translation of celastrol nanoformulations and the path forward.

The objective of this study was to investigate the effects of replacing antibiotics with Kudzu-leaf flavonoids (KLF) on the growth performances, gut epithelial development, and gastrointestinal bacteria diversities of Yellow-feathered broilers. For this purpose, total of 216 1-day-old male Yellow-feathered chickens with the similar birth weight (31.0 ± 1.0 g) were randomly divided into 3 treatments: the control treatment (CON), the kudzu-leaf flavonoids supplement treatment (KLF), and the antibiotics supplement treatment (AGP). All birds were provided with a 56 d-feeding procedure, followed by the measurement of production performances, immune organs, blood anti-oxidant parameters, intestine epithelium development, and cecal microbiota. Results showed the feed conversion ratio significantly decreased after KLF supplement compared with CON (P < 0.05). KLF supplement partly promoted the anti-oxidant capacity on account of the increased activity of Superoxide dismutase (SOD) and the decrease content of malondialdehyde (MDA). Further, as referred to the gastrointestinal development and bacteria, ratio of villus/crypt significantly increased of ileum in KLF treatment (P P < 0.05) after KLF supplementation. Moreover, correlation analysis indicated that probitics including Bifidobacterium, Butyricimonas, Lactobacillus and Streptococcus positively correlated with production performances. In conclusion, KLF supplement may promote feed efficiency and benefit the gastrointestinal health through improving gut bacterial diversity and probiotic bacteria. The KLF might be applied as a proper antibiotic alternative.

Background Curcumin, a derivative of the spice turmeric, has been adopted by Eastern medicine for centuries as an adjunct to treat several medical conditions (e.g., anorexia and arthritis) because of its well-established anti-inflammatory properties. Studies have shown that the use of curcumin in mice models has led to reduction in several inflammatory markers as well as key inflammatory pathway enzymes. As a result, studies in Western medicine have developed to determine if this recognized benefit can be utilized for patients with inflammatory lung diseases, such as asthma. This study will seek to better understand if curcumin can be used as an adjunctive therapy for improving asthma control of patients with moderate to severe asthma; a finding we hope will allow for a more affordable treatment. Methods This study will utilize a randomized, placebo controlled, double blinded pilot superiority phase 2 trial at an outpatient pulmonary clinic in Southern California, USA. Subjects will be receiving Curcumin 1500 mg or matching placebo by mouth twice daily for the study period of 12 weeks. Subjects will be randomized to either a placebo or intervention Curcumin. Subjects will have 6 clinic visits: screening visit, a baseline visit, monthly clinic visits (weeks 4, 8, and 12), at weeks 4, 8, and a follow-up clinic visit or phone-call (week 16). Changes in asthma control test scores, number of days missed from school/work, FEV1 (% predicted), FEV1/FVC ratio, FVC (% predicted), blood eosinophil count, blood total IgE, and FeNO levels will be compared by group over time. Discussion The therapeutic effects of curcumin have been studied on a limited basis in asthmatics and has shown mixed results thus far. Our study hopes to further establish the benefits of curcumin, however, there are potential issues that may arise from our study design that we will address within this paper. Moreover, the onset of the COVID-19 pandemic has resulted in safety concerns that have delayed initiation of our study. This study will contribute to existing literature on curcumin’s role in reducing lung inflammation as it presents in asthmatics as well as patients suffering from COVID-19. There is a dearth of natural remedies available for the treatment of an increasing number of diseases facing mankind. Natural products may provide an opportunity to produce formulations and therapeutic solutions to address this shortage. Curcumin (CUR), diferuloylmethane; I,7-bis-(4-hydroxy-3-methoxyphenyl)-1,6-heptadiene-3,5-dione is the major pigment in turmeric powder which has been reported to exhibit a number of health benefits including, antibacterial, antiviral, anti-cancer, anti-inflammatory and anti-oxidant properties. In this review, the authors attempt to highlight the biological and pharmacological properties of CUR in addition to emphasizing aspects relating to the biosynthesis, encapsulation and therapeutic effects of the compound. The information contained in this review was generated by considering published information in which evidence of enhanced biological and pharmacological properties of nano-encapsulated CUR was reported. CUR has contributed to a significant improvement in melanoma, breast, lung, gastro-intestinal, and genito-urinary cancer therapy. We highlight the impact of nano-encapsulated CUR for efficient inhibition of cell proliferation, even at low concentrations compared to the free CUR when considering anti-proliferation. Furthermore nano-encapsulated CUR exhibited bioactive properties, exerted cytotoxic and anti-oxidant effects by acting on endogenous and cholinergic anti-oxidant systems. CUR was reported to block Hepatitis C virus (HCV) entry into hepatic cells, inhibit MRSA proliferation, enhance wound healing and reduce bacterial load. Nano-encapsulated CUR has also shown bioactive properties when acting on antioxidant systems (endogenous and cholinergic). Future research is necessary and must focus on investigation of encapsulated CUR nano-particles in different models of human pathology.

Introduction: All parts of the mango plant contain secondary metabolites that possess several beneficial properties. In the present study, evaluation of in vitro anti-oxidant, anti-microbial and anti-inflammatory activities of five Indian cultivars of mango fruit peel extracts were carried out.Methods: Different solvent (hexane, ethyl acetate, and methanol) extracts of five Indian cultivars of mango fruit peels were prepared by using soxhalate, and the aqueous extract was prepared by maceration. In vitro anti-oxidant activities of these extracts were determined by using 2,2-Diphenyl-1-picrylhydrazyl (DPPH), 2,2′-azinobis 3-ethylbenzothiazoline-6-sulfonic acid (ABTS), hydrogen peroxide (H2O2) and nitric oxide (NO) radical scavenging methods. Their anti-microbial and anti-inflammatory activities were determined by agar-well diffusion and HRBC (human red blood cell) methods, respectively.Results: All the five mango peel extracts showed good anti-oxidant activities, especially the methanolic extract of peel of Sindhura cultivar showed better IC50 values of DPPH (21.62±1.82 μg/mL), ABTS (21.33±1.94 μg/mL), H2O2 (19.87±2.61 μg/mL) and NO (57.29±2.17 μg/mL) radical scavenging activities than other extracts. It also possessed higher contents of phenolics (169.18±3.28 mg of GAE/g) and flavonoids (26.18±1.84 mg of QE/g) than the other extracts. All peel extracts of five cultivars of mango fruit revealed good anti-microbial activities against bacterial and fungal cultures and also possessed significant anti-inflammatory activity.Conclusion: The present study revealed that all the mango peel extracts have potential antioxidant activity, as well as better anti-microbial and anti-inflammatory activities.

Background: The present study was performed to explore whether the aqueous extract of Senecio biafrae (S. biafrae) roots provide any in vivo protective activity against carbon tetrachloride (CCl4)-induced hepatotoxicity in male albino rats. Methods: Rats (150-200 grams) were grouped into five groups (A-E) of six rats each and were treated orally for twelve days with 72 hourly administration of CCl4 (1 mL/kg) as follows: Group A received distilled water only (negative control), Group B was administered distilled water plus CCl4 (positive control), Group C was administered 400 mg/kg extract and CCl4, Group D received 200 mg/extract and CCl4, while Group E was administered standard drug (Silymarin 25mg/kg, PO). Results: Pre-treatment with the extract of S. biafrae (200 or 400mg/kg) or Silymarin (25mg/kg) caused significant restoration in the biomarkers as evaluated by reducing the levels of malondialdehyde, transaminases and elevating the levels of superoxide dismutase, catalase and glutathione peroxidase activities, which were altered by CCl4 toxicity. The extract at a dose of 400mg/kg demonstrated similar activities comparable to the standard drug (Silymarin). Conclusion: The results of this study indicate that the root extract of S. biafrae possesses hepatoprotective and anti-oxidant properties which may be due to the presence of phytochemicals in it.

The objective of this study was to evaluate the in vitro anti-oxidant, hypo-tensive and the diuretic activities of Origanum glandulosum leaves in rat. Chemical analysis revealed the isolation of katuranin and 5-isopropyl-3-methylphenol. Ethyl acetate extract possessed highest scavenging activity against DPPH and hydroxyl radicals. Intravenous administration of extracts (0.04 to 12 mg/kg body weight) dose-dependently decreased the blood pressure (systolic, diastolic and mean) of the anesthetized rat. Methanol extract showed dose-dependent diuretic activity by increasing the urine output (77%) and the urinary excretion of sodium and potassium. In conclusion, this study supports the use of O. glansulosum in traditional medicine as hypotensive and diuretic agents.

The steady rise in life expectancy, modern life style and changing environmental conditions are responsible for increasing incidence of cancer. A number of chemical drugs have been used for cancer treatment; however the induction of genotoxic, carcinogenic and teratogenic effects limits their use. Alternatively, plant phytochemicals have been proven effective chemopreventive agents. This review illustrates the use of “picrosides” derived from Picrorhiza kurroa for the treatment of cancer. We have detailed the anti-oxidant and anti-inflammatory action of picrosides as the key mechanism in reducing oncogenesis. Action of picrosides on detoxifying enzymes, cell cyle regulation and induction of signal transducers inhibiting apoptosis has also been reviewed. The present review highlights the use of picrosides as an important therapeutic agent against different types of cancer.

RhoA/Rho-associated coiled-coil forming protein serine/threonine kinase (ROCK) has appeared as a potential therapeutic target in numerous diseases, because of its preventing action on various enzymes providing antioxidant and cytoprotective action. Progression and pathophysiology of diabetic nephropathy have also shown potential involvement of oxidative stress and inflammatory pathways. In the present study, we investigated the effect of kaempferol on hyperglycemia-induced activation of RhoA kinase and associated inflammatory signaling cascade. Currently there is only small literature available on the mechanism of anti-diabetic and nephroprotective action of this compound, which creates a void. Therefore, we focused here on the investigation of molecular mechanisms for kaempferol by means of in vitro testing, using rat (NRK-52E) and human renal tubular epithelial cells (RPTEC). Our findings suggest that kaempferol inhibits hyperglycemia-induced activation of RhoA and decreased oxidative stress, pro-inflammatory cytokines (TNF-α and IL-1β) and fibrosis (TGF-β1 expression, extracellular matrix protein expression) in NRK-52E and RPTEC cells. Therefore, kaempferol can be used as a potential therapeutic for the treatment of diabetic nephropathy.

Background Excessive and continuous inflammation may be the main cause of various immune system diseases. Reflux esophagitis (RE) is a common gastroesophageal reflux disease (GERD). Camellia japonica has high medicinal value and has long been used as a traditional herbal hemostatic medicine in China and Korea. The purpose of this study is to explore the antioxidant and anti-inflammatory activities of CJE and its protective effect on RE. Materials and methods Buds from C. japonica plants were collected in the mountain area of Jeju, South Korea. Dried C. japonica buds were extracted with 75% ethanol. DPPH and ABTS radical scavenging assay were evaluated according to previous method. The ROS production and anti-inflammatory effects of C. japonica buds ethanol extract (CJE) were evaluated on LPS-induced RAW 264.7 cell inflammation. The protective effects of CJE on RE were conducted in a RE rat model. Results CJE eliminated over 50% of DPPH and ABTS radical at concentration of 100 and 200 µg/mL, respectively. CJE alleviated changes in cell morphology, reduced production of ROS, NO and IL-1β. Also, down-regulated expression levels of iNOS, TNF-α, phosphorylated NF-κB, IκBα, and JNK/p38/MAPK. CJE reduced esophageal tissue damage ratio (40.3%) and attenuation of histological changes. In addition, CJE down-regulated the expression levels of TNF-α, IL-1β, COX-2 and phosphorylation levels of NF-κB and IκBα in esophageal tissue. Conclusions CJE possesses good anti-oxidation and anti-inflammatory activity, and can improve RE in rats caused by gastric acid reflux. Therefore, CJE is a natural material with good anti-oxidant and anti-inflammatory activity and has the possibility of being a candidate phytomedicine source for the treatment of RE.

In this study, the methanolic extract from seeds of Gardenia jasminoides exhibited strong antioxidant and enzyme inhibition activities with less toxicity to NIH3T3 and HepG2 cells at the concentration of 100 µg/mL. The antioxidant activities (DPPH and ABTS), α-amylase, and α-glucosidase inhibition activities were found higher in methanolic extract (MeOH-E) than H2O extract. Besides, 9.82 ± 0.62 µg and 6.42 ± 0.26 µg of MeOH-E were equivalent to 1 µg ascorbic acid for ABTS and DPPH scavenging, respectively while 9.02 ± 0.25 µg and 6.52 ± 0.15 µg of MeOH-E were equivalent to 1 µg of acarbose for inhibition of α-amylase and α-glucosidase respectively. Moreover, the cell assay revealed that the addition of MeOH-E (12.5 µg/mL) increased about 37% of glucose uptake in insulin resistant (IR) HepG2 as compared to untreated IR HepG2 cells. The LC- MS/MS and GC-MS analysis of MeOH-E revealed a total of 54 compounds including terpenoids, glycosides, fatty acid, phenolic acid derivatives. Among the identified compounds, chlorogenic acid and jasminoside A were found promising for anti-diabetic activity revealed by molecular docking study and these molecules are deserving further purification and molecular analysis.

Objective(s): This study aimed to determine the impact of cannabinoid agonists and antagonists on the mucosal lesion progress in the stomach induced by water-immersion restraint stress (WIRS). Materials and Methods: Rats subjected to WIRS for 4 hr were treated with Dimethyl sulfoxide (DMSO), CBR1 agonist (NADA, 1 mg/kg), CBR1 antagonist (Rimonabant, 1 mg/kg), CBR2 agonist (GW405833 1 mg/kg) or CBR2 antagonist (AM630, 1 mg/kg SC) 30 min before WIRS. Microscopic lesions, oxidative stress, inflammatory cytokines biomarkers, and (Myeloperoxidase) MPO in gastric tissues were determined. Results: Results indicated development of severe gastric lesions with a substantial increase in the contents of (nitric oxide) NO, (malondialdehyde) MDA, (interleukin-1 beta) IL-1β, MPO, (tumor necrosis factor-alpha) TNF-α, and a significant fall in the content of GSH and the activity of PON-1 after WIRS. Conclusion: Treatment with NADA and AM630 protected gastric tissues against ulcers as demonstrated by a decrease in the contents of MDA, TNF-α, MPO, and IL-1β along with an increase in the content of PON-1 activity and GSH in the stomach tissues. On the other hand, treatment with SR141716A or GW405833 showed no protective effects on ulcers development. It seems that cannabinoids exert their antioxidant potential and anti-inflammatory effects against WIRS-induced gastric ulcers by activation of CB1R.

Aster scaber is a wild vegetable cultivated in Korea and is known to contain phytochemicals with various biological activities. The potential antioxidant and anti-obesity effects of A. scaber and their mechanism are yet to be reported. We evaluated the total phenolic, flavonoid, and proanthocyanidin contents and oxygen radical absorbance capacity of A. scaber ethanolic extract (ASE), and analyzed the major phenolic compounds of ASE. Antioxidant activity was measured at the chemical level through 2,2-diphenyl-1-picrylhydrazyl (DPPH), reducing power assay, 2,2′-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS), and fluorescence recovery after photobleaching (FRAP) assay. In addition, it was measured in vitro through inhibition of Reactive oxygen species (ROS) production in 3T3-L1 adipocyte, and inhibition of lipid accumulation was also evaluated. ASE reduced the expression of enzymes involved in the production of ROS and increased the expression of antioxidant enzymes that reduce increased ROS levels. They also reduced the expression of adipogenesis transcription factors that regulate adipocyte differentiation in relation to ROS production, inhibited the expression of lipogenesis-related genes related to fat accumulation through AMP-activated protein kinase (AMPK) activity, and increased expression of lipolysis-related genes. Thus, ASE containing CGA (chlorogenic acid) inhibits ROS production in 3T3-L1 adipocytes, owing to its strong antioxidant activity, and inhibits lipid accumulation caused by oxidative stress. The extract can be used as a potential functional food material for reducing oxidative stress and obesity.

High-fat diet (HFD) usually induces oxidative stress and astaxanthin is regarded as an excellent anti-oxidant. An 8-week feeding trial was conducted to investigate the effects of dietary astaxanthin supplementation on growth performance, lipid metabolism, antioxidant ability, and immune response of juvenile largemouth bass (Micropterus salmoides) fed HFD. Four diets were formulated: the control diet (10.87% lipid, C), high-fat diet (18.08% lipid, HF), and HF diet supplemented with 75 and 150 mg kg−1 astaxanthin (HFA1 and HFA2, respectively). Dietary supplementation of astaxanthin improved the growth of fish fed HFD, also decreased hepatosomatic index and intraperitoneal fat ratio of fish fed HFD, while having no effect on body fat. Malondialdehyde content and superoxide dismutase activity were increased in fish fed HFD, astaxanthin supplementation in HFD decreased the oxidative stress of fish. The supplementation of astaxanthin in HFD also reduced the mRNA levels of Caspase 3, Caspase 9, BAD, and IL15. These results suggested that dietary astaxanthin supplementation in HFD improved the growth performance, antioxidant ability and immune response of largemouth bass.

Quercetin is a natural flavonol antioxidant found in various plant sources and food samples. It is well known for its notable curative effects on the treatment of ophthalmic diseases due to various biological activities, such as antioxidant, anti-inflammatory, and anti-fibrosis activities. This review will discuss the latest developments in therapeutic quercetin for the treatment of keratoconus, Graves' orbitopathy, ocular surface, cataracts, glaucoma, retinoblastoma, and other retinal diseases.

Microalgae are believed to be a promising source of different high value products (HVP). HVP are metabolites which are produced during the growth of microalgae and their production is triggered by the physiochemical growth conditions of the microalgae. The present project is aimed at exploring the presence of bioactive compounds in desert microalgae Desmodesmus sp. and to assess its ability to support and improve health care in Qatar. The isolate was cultivated under conventional conditions as a control, and under 3 different stress conditions to enhance the production of high value products. Then crude algal extracts were used to assess the biological activity. Results of the study suggest that Desmodesmus sp. produces high value products that exhibit great anti-oxidant, anti-proliferative and anti-bacterial activity when cultivated under conventional conditions and under continuous light stress.

The present investigation aimed to evaluate the protective effect of Zingerone (ZIN) against lipopolysaccharide-induced oxidative stress, DNA damage, and cytokine storm in rats. For survival study the rats were divided into four groups (n = 10). The control group was treated with normal saline, Group II received an intraperitoneal (i.p) injection (10 mg/kg) of LPS as disease control. Rats in Group III were treated with ZIN 150 mg/kg (p.o) 2 h before LPS challenge and rats in Group IV were given ZIN only. Survival of the rats was monitored up to 96 h post LPS treatment. In another set, the animals were divided into four groups of six rats. Animals in Group I served as normal control and were treated with normal saline. Animals in Group II were treated with lipopolysaccharide (LPS) and served as disease control. Group III animals were treated with ZIN 2 h before LPS challenge. Group IV served as positive control and were treated with ZIN (150 mg/kg orally). The blood samples were collected and used for the analysis of biochemical parameters like alanine transaminase (ALT), alkaline phosphatase (ALP), aspartate transaminase (AST), blood urea nitrogen (BUN), Cr, Urea, lactate dehydrogenase (LDH), albumin, bilirubin (BIL), and total protein. Oxidative stress markers malondialdehyde (MDA), glutathione peroxidase (GSH), myeloperoxidase (MPO), and (DNA damage marker) 8-OHdG levels were measured in different organs. Level of nitric oxide (NO) and inflammatory markers like TNF-&alpha, IL-1&szlig, IL-1&alpha, IL-2, IL-6, and IL-10 were also quantified in plasma. Procalcitonin (PCT), a sepsis biomarker, was also measured. ZIN treatment had shown significant (p &lt, 0.5) restoration of plasma enzymes, antioxidant markers and attenuated plasma proinflammatory cytokines and sepsis biomarker (PCT), thereby preventing the multi-organ and tissue damage in LPS-induced rats also confirmed by histopathological studies of different organs. The protective effect of ZIN may be due to its potent antioxidant potential. Thus ZIN can prevent LPS-induced oxidative stress as well as inflammatory and multi-organ damage in rats when administered to the LPS treated animals.

Lactoferrin (LAT), a multifunctional protein involved in numerous physiological functions, and the medicinal plant Pelargonium sidoides DC (PEL) have been described for their anti-inflammatory properties. Because the main advantage of natural products consists in administering them in combination rather than as single compound, we aimed to understand whether the combination of PEL and LAT, herein PELIRGOSTIM, could still prove beneficial or additive/synergistic activities during inflammatory conditions. To pursue this goal, we used macrophagic cells (J774.1) and treated them with PEL and LAT in a concentration-dependent manner. We found that PELIRGOSTIM was able to reduce the levels of reactive oxygen species (ROS) and nitrite, effects that were correlated to the release of lower levels of IL-1β after LPS treatment. In addition, the combination of PEL and LAT showed bacteriostatic activities against Staphylococcus aureus and Escherichia coli which had limited growth starting from 5 hours up to 20 hours. This effect was stronger than that observed for penicillin/streptomycin. Our results provide PELIRGOSTIM as an innovative combination of natural products capable to prevent inflammation-, oxidative stress- and microbial-related disorders.

Introduction: Evidence suggests increased oxidative stress and improved total antioxidant status resulting from regular exercise in the heart tissue of diabetic patients. The aim of this study was to investigate the effect of 8 weeks of increasing endurance training on the levels of antioxidant enzymes and the total antioxidant status of the cardiac tissue of diabetic mice. Methods: In this experimental study, 24 Wistar male rats (aged 10 weeks and weighing 256±11.8 g) were divided into 4 groups of 6.The training program lasted for 8 weeks with increasing endurance training. It was measured 48 hours after the completion of the protocol levels of antioxidant enzymes (catalase, glutathione, and glutathione peroxidase) and the total antioxidant status of rat heart tissue. One-way analysis of variance test was used for intergroup comparisons and Pearson test was used to investigate the relationship between indicators. Results: The results showed a significant difference in total antioxidant status, catalase and glutathione (P = 0.001) and were not significant in glutathione peroxidase (P = 0.240). Moreover, a follow-up test showed a significant increase in total antioxidant status and catalase (healthy exercise, healthy control, and diabetic exercise compared to diabetic control), Glutathione (healthy exercise (P = 0.001) and healthy control (P = 0.049) compared to diabetic control); (Healthy exercise (P = 0.003) compared to diabetic exercise. Finally, some correlations between antioxidant enzymes and total antioxidant status were significant. Conclusion: According to research results, increasing endurance training appears to increase levels of antioxidant enzymes and improve total antioxidant status, thereby it reduces oxidative stress in the heart tissue of diabetic mice.

The p53 protein plays a central role in mediating immune functioning and determines the fate of the cells. Its role as a tumor suppressor, and in transcriptional regulation and cytokine activity under stress conditions, is well defined. The wild type (WT) p53 functions as a guardian for the genome, while the mutant p53 has oncogenic roles. One of the ways that p53 combats carcinogenesis is by reducing inflammation. WT p53 functions as an anti-inflammatory molecule via cross-talk activity with multiple immunological pathways, such as the major histocompatibility complex I (MHCI) associated pathway, toll-like receptors (TLRs), and immune checkpoints. Due to the multifarious roles of p53 in cancer, it is a potent target for cancer immunotherapy. Plant flavonoids have been gaining recognition over the last two decades to use as a potential therapeutic regimen in ameliorating diseases. Recent studies have shown the ability of flavonoids to suppress chronic inflammation, specifically by modulating p53 responses. Further, the anti-oxidant Keap1/Nrf2/ARE pathway could play a crucial role in mitigating oxidative stress, leading to a reduction of chronic inflammation linked to the prevention of cancer. This review aims to discuss the pharmacological properties of plant flavonoids in response to various oxidative stresses and immune dysfunctions and analyzes the cross-talk between flavonoid-rich dietary intake for potential disease prevention.

Flaxseed is a plant that grows and is cultivated in more than 50 countries; the main flax producer countries are Canada, China, the United States, and India. The purpose of the present study was to overview the source, chemical compounds, and mechanisms of the therapeutic effects of this valuable plant. For writing this manuscript, we made a list of relevant keywords and phrases, and then we started searching for studies in PubMed, Scopus, and Web of Science databases. The main constituents of flaxseed include lipids, proteins, lignans, fibers, and minerals. Flaxseed is full of antioxidants such as tocopherols, betacarotene, cysteine, and methionine which result in a decrease in blood pressure, heart disease, hepatic and neurological disorders, and increased insulin sensitivity. Flaxseed is commonly used for its antidiabetic and anticancer activities and also it is beneficial for cardiovascular, gastrointestinal, hepatic, urological, and reproductive disorders, and because of these beneficial effects, it is recognized as a medical plant.

Background Catechins, which are polyphenol compounds found in many plants and are an important component of tea leaves, are strong anti-oxidants. Research Many studies seek to enhance the effects of catechins on the human body and boost their protective power against UV radiation. There are many examples of the positive anti-microbial, anti-viral, anti-inflammatory, anti-allergenic, and anti-cancer effects of catechins. Catechins increase the penetration and absorption of healthy functional foods and bio cosmetics into the body and the skin, thus improving their utility. High value-added anti-oxidant substances have been extracted from food and plant sludge, and experiments have shown that catechins are safe when applied to the human body. The stability of catechins is very important for their absorption into the human body and the effectiveness of their anti-oxidant properties. Conclusion Continued research on the strong anti-oxidant effects of catechins is expected to result in many advances in the food, cosmetics, and pharmaceutical industries.

Couple of ethnopharmacological surveys in the Indian Ladakh and Pakistani Shigar valleys has reported the medicinal use of Acantholimon lycopodioides against cardiac and gastric disorders that however, remains without scientific rationale or experimental validations. Here, we assess the in vitro bio/therapeutic activities of A. lycopodioides extracts as well as chloroform, ethyl acetate, n-butanol and aqueous fractions. The in vitro β-carotene-linoleic acid bleaching and DPPH radical scavenging methods demonstrated a very high anti-oxidative property of chloroform and ethyl acetate fractions compared to others. Cell viability assay (MTT) on human cervical (HeLa), breast (MDA-MB321) and liver (HepG2) cancer cells revealed their differential cytotoxicity, except the chloroform fraction. Of these, the precipitate exerted highest cytotoxicity on HepG2 cells followed by aqueous fraction on MDA-MB321 cells. Notably, the non-cytotoxicity of chloroform fraction coincided with its highest anti-oxidative activity. Further, the chloroform fraction showed marked hepatoprotection (up to 84%) against 3′7′dichlorofluorescin triggered free radicals induced oxidative damage. Also, the hepatoprotective chloroform fraction mildly activated CYP3A4 in HepG2 cells (dual-luciferase assay). Moreover, the A. lycopodioides extracts and fractions showed differential anti-bacterial and anti-fungal activities. Of these, while S. aureus was more sensitive to the water-insoluble extract, ethyl acetate fraction showed moderate activity against E. coli and C. albicans. On the other hand, the chloroform fraction showed promising activity against S. Aureus, C. albicans, P. vulgaris and E. faecalis. In conclusion, our data for the first time, demonstrated promising anti-oxidative, hepatoprotective, anti-cancer, anti-microbial and CYP3A4 activating salutations of A. lycopodioides. This warrants further studies towards isolation and identification of its therapeutically active principles.

Surface active agents are characterized for their capacity to adsorb to fluid and solid-water interfaces. They can be classified as surfactants and emulsifiers based on their molecular weight (MW) and properties. Over the years, the chemical surfactant industry has been rapidly increasing to meet consumer demands. Consequently, such a boost has led to the search for more sustainable and biodegradable alternatives, as chemical surfactants are non-biodegradable, thus causing an adverse effect on the environment. To these ends, many microbial and/or marine-derived molecules have been shown to possess various biological properties that could allow manufacturers to make additional health-promoting claims for their products. Our aim, in this review article, is to provide up to date information of critical health-promoting properties of these molecules and their use in blue-based biotechnology (i.e., biotechnology using aquatic organisms) with a focus on food, cosmetic and pharmaceutical/biomedical applications.

Thai Traditional Medicine believes that human body is composed of four elements. It also mentions that skin problems related to each element can be alleviated by using suitable plants. This study aimed to determine the bioactivities related to skin effects of Aloe vera, Cucumis sativus, Alpinia galanga and Phyllanthus emblica. The plants were extracted with 50% ethanol prior to bioactivity evaluation. The highest anti-oxidant activity (IC50, 14.01 µg/ml) was found for P. emblica. Tyrosinase inhibition was found with P. emblica (41.92% inhibition) and A. vera (26.67% inhibition) at 500 µg/ml concentrations. The cells treated with 500 µg/ml A. vera and 15.62 µg/ml P. emblica had after 48 h increased collagen type-1 production by around 14 and 4 -fold from those of untreated cells. Anti-glycation was found with P. emblica. P. emblica, A. galanga and C. sativus at 62.5 µg/ml exhibited anti-inflammatory activity. This information supports evidence based use of these plants as food and cosmetic ingredients.

There exist overwhelming evidence that plant-derived bioactive flavonoids possess strong therapeutic activities against cancer, cardiovascular diseases, and age-related neurodegenerative disorders. Several drug discovery studies have suggested the therapeutic potential of apigenin (APG) are due to its strong anti-inflammatory, anti-oxidant, and anti-tumor properties. APG is found in citrus fruits, vegetables, spices, and herbs. It is well documented that APG can modulate a wide variety of intra- and extra-cellular signaling pathways in in vitro cancer cell lines and in vivo neoplasm systems, and consequently retard or inhibit abnormal growth of tissues. Co-administration of APG with anti-cancer drugs has shown to increase the therapeutic potency of the later. The overall aims and objectives of this review are to highlight the potential role of APG in the prevention and treatment of different types of malignant human cancers. We will also describe different purported biochemical mechanisms associated with the inhibition of cancer cell proliferation and tumor growth.

Aim: Identification of the important processes and the related genes that are dis-regulated in the celiac disease (CD) was the aim of this study. Background: Celiac disease is an autoimmune disorder which is characterized by immune reaction response mostly to wheat gluten. The gluten-free diet is the best-known treatment of the patients. Methods: Significant differentially expressed proteins (DEPs) related to the CD are extracted from a published proteomics study and are included in protein-protein interaction PPI) network analysis by Cytoscape software and its applications. The central proteins and related processes are identified and discussed. Results: Among 53 queried genes, 51 individuals were recognized by the database, and after network construction, 48 ones included in the network, and three genes remained as isolated nodes. Following 50 neighbors, the network was analyzed, and eight central genes were identified as dis-regulated elements. Related processes and the role of the central genes in celiac are discussed in detail. Conclusion: CAT, ENO1, PCK2, ACO2, ALDOOB, GALM, ADA, and ACTBADA as critical genes and Antioxidant activity, carbohydrate metabolism, inflammation, cell growth processes are highlighted as the dis-regulated individuals in CD.

Parkinsonian-like motor deficits in Huntington's Disease (HD) patients are associated with abnormal dopamine neurotransmission in the striatum. Dopamine metabolism leads to the formation of oxidized dopamine quinones that exacerbates mitochondrial dysfunction with production of reactive oxygen species (ROS) that eventually lead to neuronal cell death. We have previously shown that dopamine-induced oxidative stress triggers apoptotic cell death in dopaminergic neuroblastoma SH-SY5Y cells hyper-expressing the mutant polyQ Huntingtin (polyQ-Htt) protein. Dopamine toxicity was paralleled by impaired autophagy clearance of the polyQ-Htt aggregates. In this study, we found that Dopamine affects the stability and function of ATG4, a redox-sensitive cysteine-protein involved in the processing of LC3, a key step in the formation of autophagosomes. Resveratrol, a dietary polyphenol with anti-oxidant and pro-autophagic properties, has shown neuroprotective potential in HD. Yet the molecular mechanism through which Resveratrol can protect HD cells against DA is not known. Here, we show that Resveratrol prevents the generation of ROS, restores the level of ATG4, allows the lipidation of LC3, facilitates the degradation of polyQ-Htt aggregates and protects the cells from Dopamine toxicity. The present findings provide a mechanistic explanation of the neuroprotective activity of Resveratrol and support its inclusion in a therapeutic regimen to slow down HD progression.

BACKGROUND Severe burn damage and its consequences are life threatening which can complicate patients’ health. Medicinal and traditional plants are considered as safe, natural and inexpensive source of treatment for wide variety of diseases. This study assessed beneficial effect of Pistacia atlantica oil on rats burn wound healing and its potential effects on malondialdehyde (MDA), vasculoendothelial growth factor (VEGF), hydroxyprolin and antioxidant status in wound area. METHODS Thirty male rats weighing 200±10 g were randomly divided into three groups (n=10) as follows. Group 1 underwent just burn injury, Group 2 underwent burn injury and received 150 mg/kg/day P. atlantica oil topically, and Group 3 underwent burn injury and received 150 mg/kg/day sulfadiazine cream topically. At the end of the study (day 14), wounded areas were measured and then skin in the burn damage were dissected and anti-oxidative parameter, MDA, VEGF and hydroxyprolin were evaluated. RESULTS P. Atlantica oil significantly increased antioxidant defense, VEGF, hydroxyprolin and reduced MDA levels. It could remarkably reduce wound size compared to burn control group. P. Atlantica oil showed more beneficial effects than sulfadiazine. CONCLUSION P. atlantica resin oil could be considered as a new therapeutic agent for treatment of injuries.
