La reforma laboral aprobada el 28 de diciembre de 2021 ha consolidado, de manera definitiva, como instrumento clave de flexibilidad interna, una figura que ha demostrado, durante la crisis sanitaria por COVID19, que es una herramienta clave para empresas y trabajadores ante situaciones complejas. Una herramienta que facilita el tránsito hacia la recuperación empresarial mientras que preserva el empleo, siendo, por tanto, un elemento de equilibrio entre los intereses de los antagonistas sociales en el marco de las relaciones de trabajo. El RDL 32/2021 ha configurado una ordenación normativa fruto del equilibrio entre el respeto a lo ya existente, la incorporación de aquellos aspectos que han sido probados con éxito durante la pandemia y aquellos otros elementos que han pretendido mejorar su diseño anterior. La eficacia de la medida ya quedó acreditada en momentos de máxima dificultad, queda ver cómo responderá en el período postpandémico, pero, sea como sea, el éxito de la misma ya se concreta por el hecho de venir respaldada por un acuerdo en el marco del Diálogo Social. The labour law reform approved on 28 December 2021 in Spain has definitively consolidated, as a key instrument of internal flexibility in companies, a figure that has been shown, during the health crisis by COVID-19, to be a key tool for companies and workers in complex situations. A tool that facilitates the transition towards business recovery while preserving employment, and is therefore an element of balance between the interests of the social antagonists in the framework of labour relations. Royal Decree-law 32/2021 has configured a regulatory system that is the result of a balance between respect for what already exists, the incorporation of those aspects that have been successfully tested during the pandemic and those other elements that have sought to improve on its previous design. The effectiveness of the measure has already been accredited in times of maximum difficulty; it remains to be seen how it will respond in the post-pandemic period, but, be that as it may, its success has already been confirmed by the fact that it is backed by an agreement within the framework of the Social Dialogue. [Background] During the last three decades systemic fungal infections associated to immunosuppressive therapies have become a serious healthcare problem. Clinical development of new antifungals is an urgent requirement. Since fungal but not mammalian cells are encased in a carbohydrate-containing cell wall, which is required for the growth and viability of fungi, the inhibition of cell wall synthesizing machinery, such as β(1,3)-D-glucan synthases (GS) and chitin synthases (CS) that catalyze the synthesis of β(1-3)-D-glucan and chitin, respectively, represent an ideal mode of action of antifungal agents. Although the echinocandins anidulafungin, caspofungin and micafungin are clinically well-established GS inhibitors for the treatment of invasive fungal infections, much effort must still be made to identify inhibitors of other enzymes and processes involved in the synthesis of the fungal cell wall. [Purpose] Since natural products (NPs) have been the source of several antifungals in clinical use and also have provided important scaffolds for the development of semisynthetic analogues, this review was devoted to investigate the advances made to date in the discovery of NPs from plants that showed capacity of inhibiting cell wall synthesis targets. The chemical characterization, specific target, discovery process, along with the stage of development are provided here. [Methods] An extensive systematic search for NPs against the cell wall was performed considering all the articles published until the end of 2020 through the following scientific databases: NCBI PubMed, Scopus and Google Scholar and using the combination of the terms “natural antifungals” and “plant extracts” with “fungal cell wall”. [Results] The first part of this review introduces the state of the art of the structure and biosynthesis of the fungal cell wall and considers exclusively those naturally produced GS antifungals that have given rise to both existing semisynthetic approved drugs and those derivatives currently in clinical trials. According to their chemical structure, natural GS inhibitors can be classified as 1) cyclic lipopeptides, 2) glycolipids and 3) acidic terpenoids. We also included nikkomycins and polyoxins, NPs that inhibit the CS, which have traditionally been considered good candidates for antifungal drug development but have finally been discarded after enduring unsuccessful clinical trials. Finally, the review focuses in the most recent findings about the growing field of plant-derived molecules and extracts that exhibit activity against the fungal cell wall. Thus, this search yielded sixteen articles, nine of which deal with pure compounds and seven with plant extracts or fractions with proven activity against the fungal cell wall. Regarding the mechanism of action, seven (44%) produced GS inhibition while five (31%) inhibited CS. Some of them (56%) interfered with other components of the cell wall. Most of the analyzed articles refer to tests carried out in vitro and therefore are in early stages of development. [Conclusion] This report delivers an overview about both existing natural antifungals targeting GS and CS activities and their mechanisms of action. It also presents recent discoveries on natural products that may be used as starting points for the development of potential selective and non-toxic antifungal drugs. The goalkeeper and defensive player’s degree of opposition could influence throwing velocity and thereby success of scoring goals. Therefore, the aim of this study was to investigate the influence of opposition and contact on throwing velocity and effectiveness in elite team handball players from first and second offensive lines. Throwing velocity from 2893 throws carried out in 47 matches during the Men’s World Handball Championship were analysed. A higher throwing velocity was obtained with opposition and no contact situations. In the comparison between the first and second offensive line players, the same results were obtained in relation to the higher throw speed. Significant relationships were found between the attack effectiveness and the type of opposition and contact. Throws carried out with no opposition either with or without contact are those that more likely end in a goal, while opposition without and with contact decreases the effectiveness in scoring goals. Concerning playing positions, both first and second line showed this significant relationship between attack effectiveness and the type of opposition and contact. It was concluded that opposition and defensive contact have a negative and significant effect on the scoring efficacy. Defensive contact is a determining factor causing a significant decrease on throwing velocity.

Trabajo presentado en el XIV Congreso Nacional de Micología EFI3, celebrado en Tarragona (España), del 19 al 21 de septiembre de 2018, La pared celular es una estructura esencial en las células fúngicas que se encuentra ausente en las células de animales. Es una estructura rígida, externa a la membrana plasmática, que proporciona a la célula protección mecánica y resistencia a la presión osmótica interna. A pesar de su rigidez, la pared celular es capaz de adaptarse a los cambios morfológicos que se producen durante el ciclo de vida de la célula fúngica. Así, durante la citocinesis se produce la biosíntesis de un septo de división simultánea a la contracción de un anillo de actomiosina. La levadura de fisión Schizosaccharomyces pombe es un hongo unicelular con forma bacilar que crece por extensión de los extremos y se divide por fisión en la zona media, generando dos células hijas del mismo tamaño. S. pombe es un organismo modelo para el estudio de la citocinesis y el crecimiento polarizado porque su morfología, tamaño y división son extremadamente reproducibles. La pared celular de S. pombe posee dos tipos de glucanos como componentes estructurales mayoritarios y esenciales: ß-(1,3)-D-glucano con algunas ramas ß-(1,6) y ¿-(1,3)-D-glucano. Bgs4 y Ags1 son las enzimas responsables de la síntesis de estos polímeros. También hay una pequeña cantidad de ß-(1,3)-D-glucano lineal localizado en el septo primario y en las áreas de crecimiento. Bgs1 es la enzima responsable de la síntesis de este ß-(1,3)-glucano lineal que es esencial para la formación del septo y para la viabilidad celular. En esta ponencia se presentan resultados recientes que profundizan en el conocimiento del proceso de septación de la levadura de fisión describiendo la colaboración de Bgs1 con proteínas del anillo de actomiosina, como la paxilina y la fosfatasa calcineurina, en la formación del surco de división. Además, presentaremos resultados preliminares sobre la función de Bgs1 y el posible papel del glucano lineal en el establecimiento del crecimiento polarizado.

Trabajo presentado en el Fungal Cell Wall, celebrado en el Ensenada (Baja California, México), del 9 al 12 de octubre de 2017, Cytokinesis is the final process of the cell cycle, which gives rise to two independent daughter cells once mitosis is completed. In fungi, as in animal cells, cytokinesis requires the formation and closure of a cleavage furrow that is achieved by constriction of an actomyosin ring and invagination of the plasma membrane. Additionally, since fungal cells are surrounded by a rigid cell wall, cytokinesis requires the formation of a septum coincident with the ring constriction and membrane ingression. Fission yeast Schizosaccharomyces pombe is a unicellular, rod-shaped fungus that divides by medial fission, generating two daughter cells of equal size. Two types of glucans are the major structural components of S. pombe wall: ß(1,3)-D-glucan with some ß(1,6) branches, and ¿(1,3)-D-glucan. Bgs4 and Ags1 are the enzymes responsible for the synthesis of these polymers. Additionally, there is a small amount of linear ß(1,3)-D-glucan located at the growing areas and the primary septum. Bgs1 is the enzyme responsible for linear B(1,3)glucan synthesis. We will present data about the reciprocal collaboration between the enzymes responsible for cell wall synthesis and the actomyosin ring during cytokinesis, and will discuss the importance of the cell wall and the glucan synthases in the formation of the cleavage furrow, the contraction of the actomyosin ring, and the construction of a septum able to support the high turgor pressure of the cell. In addition, we will present preliminary results on the possible role of Bgs1 and the linear glucan in the establishment of polarized growth. Bgs1 depletion causes delocalization of the actin patches and, as a consequence, diffusion of the sterol-rich membrane domains. Moreover, in the absence of the main polarity markers, Tea1 or Tea4, Bgs1 depletion induces complete loss of polarity and causes multinucleated rounded cells even in the presence of osmotic stabilizer. Our results suggest that Bgs1 is a redundant polarity cue that collaborates with Tea1-Tea4 in the maintenance of the fission yeast morphology.

In fungi, as in animal cells, cytokinesis requires the formation of a cleavage furrow that is achieved by constriction of an actomyosin ring and invagination of the plasma membrane. Additionally, since fungal cells have a polysaccharide cell wall outside the plasma membrane, cytokinesis requires the formation of a septum coincident with the membrane ingression. The septum biosynthesis must be strictly coordinated with the deposition of new plasma membrane material and closure of the actomyosin ring, in such a way that no cell lysis occurs during the whole process. Fission yeast Schizosaccharomyces pombe is a unicellular, rod-shaped fungus that has become a popular model organism for the study of actomyosin ring formation and constriction during cell division. We will present recent advances about the reciprocal collaboration between the enzymes responsible for the cell wall synthesis and some components of the actomyosin ring. Our recent studies unveiled the importance of this collaboration in the formation of the cleavage furrow and the construction of a cross-walled septum able to support the high turgor pressure of the cell. In addition, we will discuss the importance of the cell wall in the steady progression of the cleavage furrow.

Resumen del póster presentado al American Society for Cell Biolog (ASCB) Annual Meeting, celebrado en San Francisco, California (US) del 3 al 7 de diciembre de 2016., Fission yeast cells are rod‐shaped, grow by tip extension, and divide by medial fission, generating two daughter cells of equal size. Like other fungi, fission yeast possesses an extracellular polysaccharidebased structure called the cell wall. Microtubule‐mediated delivery to the cell ends of the Kelch‐repeat containing Tea1 protein and the SH3 domain containing Tea4 protein, is required to establish the cell grow at the tips. The Tea1‐Tea4 complex is able to mediate the polarization of the sterol‐rich membrane domains that recruit other factors, such as the GTPase Cdc42 which is required to maintain the shape and dimensions of the cell. Bgs1 is the enzyme responsible for linear B(1,3)glucan synthesis. This cell wall polymer is mainly present in the primary septum and, in lower amount, in the cell wall including the cell tips. Bgs1 depletion generates multiseptated cells with defective septa. We show here that Bgs1 depletion also causes delocalization of the actin patches and, as a consequence, diffusion of the sterol‐rich membrane domains. Additionally, Bgs1 depletion causes other glucan synthases, normally concentrated at the growing poles, to spread throughout the membrane. Moreover, in the absence of Tea1 or Tea4 polarity markers, Bgs1 depletion induces complete loss of polarity and causes multinucleated rounded cells even in the presence of osmotic stabilizer. Bgs1 loss of function causes the same phenotypes as the Bgs1 depletion. Our results suggest that functional Bgs1, through the control of actin patches, is a redundant polarity cue that collaborates with Tea1‐Tea4 in the maintenance of the fission yeast morphology.

Additionally, because fungal cells have a polysaccharide cell wall outside the plasma membrane, cytokinesis requires the formation of a septum coincident with the membrane ingression. Fission yeast Schizosaccharomyces pombe is a unicellular, rod-shaped fungus that has become a popular model organism for the study of actomyosin ring formation and constriction during cell division. Resumen del trabajo presentado a la 10ª Reunión de la Red Española de Levaduras, celebrada en El Escorial (Madrid) del 16 al 18 de diciembre de 2015., Cytokinesis is the final stage of the eukaryotic cell cycle where, after chromosome segregation, the formation of a cleavage furrow separates the cell into two new cells. Furrow formation is achieved by closure of a contractile actomyosin ring (CR) that invaginates the plasma membrane. In fungal cells, it also requires simultaneous synthesis of a division septum structure that will constitute the new cell wall upon cell separation. Thus, the septum biosynthesis must be strictly coordinated with the deposition of new plasma membrane material and CR closure. The cell wall is an external structure, essential for the maintenance of fungal cell integrity. The α(1,3) and β(1,3)glucans are the two main structural polymers of fission yeast cell wall. Schizosaccharomyces pombe contains four β(1,3)glucan synthase subunits, Bgs1 to Bgs4, that share high identity with fungal and plant homologues. Interestingly, all of them and the α(1,3)glucan synthase Ags1 are essential. Our group is interested in the study of Ags1, Bgs1 and Bgs4, the main subunits involved in the synthesis of the different cell wall and septum glucans. The septum is a three-layered structure of a middle primary septum (PS) flanked by a secondary septum (SS) on each side. Bgs1 is responsible for the linear β(1,3)glucan synthesis of the PS which interacts specifically and with high affinity with the fluorochrome Calcofluor white (CW). Bgs1 shares an essential function with CR paxillin, collaborating for septum formation. Bgs4 is responsible for the branched β(1,3)glucan which is required for connecting extracellular cell wall and CR during cytokinesis; and Ags1 for the α(1,3)glucan which is essential for the PS adhesion strength needed to withstand the turgor pressure during cell abscission. Both Bgs4 and Ags1 are responsible for the SS formation and both are essential for cell integrity. Our group is interested in studying the cytokinesis process. It is generally believed in all eukaryote cells that furrow ingression and CR constriction initiate after nuclear mitosis completion at the end of anaphase or the onset of telophase. Thus, it is assumed a sequential process in which furrow ingression will not start until mitosis completion, when the chromosomes are segregated and the spindle disassembled. However, we have found that septum synthesis and therefore CR constriction, initiate in early-anaphase overlapping nuclear mitosis. The regulation of septation onset with regards to the CR proteins, activation of the septation initiation network (SIN), cell cycle mitotic kinase and cyclin, cell size, spindle pole body asymmetry, etc. was studied. Our data show that mitosis and septation are overlapped processes, but still ensuring a safe mitosis in which septum synthesis only initiates after successful chromosome segregation.

Resumen del trabajo presentado a la EMBO Conference on Fission Yeast: Pombe 2013; 7th International Fission Yeast Meeting celebrada en Londres (UK) del 24 al 29 de junio de 2013., The cell wall is a structure external to the plasma membrane. Its integrity is vital and it constitutes the exoskeleton that confers mechanical strength and osmotic resistance to the fungal cell. The main structural component of the Schizosaccharomyces pombe cell wall is the β(1,3)glucan. Fission yeast contains four essential putative β(1,3)glucan synthase (GS) subunits, Bgs1 to Bgs4. Bgs1 localizes to the contractile ring and septum, and it is responsible for the synthesis of a special linear β(1,3)glucan and the primary septum that this glucan generates. Besides, Bgs1 is also localized to the growing poles. The establishment of growth polarity is controlled by a complex termed "polarisome", which is at least comprised by the marker proteins Tea1 and Tea4, the membrane protein Mod5, Bud6, the formin For3 and the GTPase Cdc42. Bgs1 depletion produces cells with bifurcated ends. We are studying the role of Bgs1 in growth polarity and we found that bgs1+ shut-off in tea1Δ or tea4Δ mutants originates spherical cells. However, this effect is not produced by bgs1+ shut-off in mod5Δ, bud6Δ or for3Δ single, double or triple mutants. This suggests that both Bgs1 and Tea proteins cooperate in an essential function for polarity establishment and maintenance. We also found that Bgs1 absence produces Mod5-GFP, actin patches and lipid-rafts delocalization all around the plasma membrane. We were able to cause actin patches delocalization by using low concentrations of the actin depolymerizing drug Latrunculin B, and observed that actin patches delocalization in tea1Δ and tea4Δ cells produces the loss of polarity. It has been described that some cdc42 thermosensitive mutants and the pob1Δ or wsp1Δ deletion mutants present delocalized actin patches around the cell periphery. Combination of these mutations with the tea1Δ and tea4Δ mutations resulted in spherical cells. These results show that Bgs1 has a role in cell polarity by controlling the actin patches localization.

Trabajo fin de grado presentado en la Universidad de Salamanca, Grado en Biología/Biotecnología

La situación demográfica actual en España está marcada por una tasa de natalidad reducida y una esperanza de vida cada vez mayor, convirtiendo nuestro país en uno de los más envejecidos del mundo. Esta situación se refleja en nuestro mercado de trabajo que ve afectada su fuerza laboral por el envejecimiento poblacional y sus circunstancias. Ante dichos acontecimientos, este Trabajo Final de Grado analiza en sus diferentes apartados los factores principales que están presentes en la configuración del mercado de trabajo de las personas mayores de 45 años, dedicando un amplio apartado dentro de dentro del marco teórico al análisis de desempleo de larga duración y su especial incidencia sobre este colectivo. El trabajo finaliza exponiendo la protección social destinada a este colectivo por parte de las administraciones y la regulación en la que esta se sustenta. Se concluye estableciendo que es necesario que los gobernantes tengan en cuanta toda esta situación e implanten medidas en aras a alcanzar soluciones realmente eficaces para evitar la precariedad laboral del colectivo. The current demographic situation in Spain is marked by a reduced birth rate and an increasing life expectancy, making our country one of the oldest in the world. This situation is reflected in our labor market, affected by the aging population and its circumstances. Given these events, throughout its different sections this project analyzes the main factors that are present in the configuration of the labor market of people over 45 years of age, dedicating a wide section within the theoretical framework to the analysis of long-term unemployment and its special impact on this group. The work study ends by exposing the social protection for this group by the administrations and the regulation on which it is based. It concludes by establishing that it is necessary for the rulers to take the situation into account and implement measures in order to reach really effective solutions to avoid the precariousness of the collective's work.

La “Horchata de chufa" o más comúnmente conocida como “Horchata” es una bebida tradicional, no alcohólica, dulce y de aspecto blanquecino obtenida a partir de tubérculos de chufa (Cyperus esculentus L. var. Sativus Boeck) consumida fundamentalmente en la Comunidad Valenciana durante la época de verano, aunque en los últimos años ha experimentado un crecimiento en su consumo tanto a nivel español como en el resto del mundo. Durante el proceso de elaboración de la “Horchata” se producen una gran cantidad de residuos y subproductos tanto sólidos como líquidos, donde los sólidos pueden representar hasta el 60% del material vegetal. Estos subproductos se han considerado tradicionalmente como desechos industriales sin valor comercial y se han utilizado fundamentalmente como pienso para animales. Los subproductos líquidos destacan por su alto contenido en prebióticos, antioxidantes y como sustituto del agua en productos cárnicos. Por otra parte, los subproductos sólidos pueden ser utilizados para diferentes usos, debido a su alto contenido en fibra dietética, compuestos antioxidantes bioactivos, además de tener buenas propiedades tecnológicas (capacidad de retención de agua, capacidad de emulsión y estabilidad de emulsión). El objetivo de la presente Tesis Doctoral es la recuperación caracterización de diversos nutrientes y compuestos bioactivos obtenidos a partir de subproductos sólidos del proceso de elaboración de la “horchata”. Fundamentalmente, el estudio se ha centrado en la extracción de aceite y evaluación del contenido graso y perfil de ácidos así como vitamina E del mismo. También se ha evaluado el contenido de compuestos antioxidantes bioactivos, capacidad antioxidante total y parámetros fisicoquímicos de los extractos obtenidos. Para obtener los extractos, se utilizaron diferentes técnicas de extracción, tanto métodos convencionales (ej. extracción convencional sólido/líquido, Soxhlet, etc.) como procesos innovadores (ej. fluidos supercríticos (SC-CO₂)), también conocidos como “extracción verde”. Posteriormente, los extractos se analizaron mediante la utilización de diferentes técnicas analíticas, como espectrofotometría y métodos cromatográficos como por ejemplo cromatografía de gases con detector de ionización de llama, cromatografía líquida acoplada a espectrometría de masas en tándem, etc. También se revisó en la literatura disponible el potencial uso de otros procesos innovadores como ultrasonidos, pulsos eléctricos, altas presiones, solas y/o en combinación con enzimas, expresión mecánica asistida por gas, microondas, los cuales también se han mostrado interesantes de cara a la valorización de los subproductos obtenidos durante la elaboración de la “Horchata”. Tras evaluar los resultados obtenidos, se observó cómo los subproductos de la "horchata" presentan un contenido importante de aceite, y compuestos antioxidantes bioactivos, aunque es necesario seleccionar el método de extracción apropiado para conseguir recuperar estos compuestos de la manera adecuada. Se observó que el contenido de nutrientes y compuestos antioxidantes bioactivos difiere de acuerdo al método de extracción utilizado. Por ejemplo, tras utilizar la extracción con fluidos supercríticos, se observó un aumento en la extracción de aceite tras aumentar la presión utilizada, aunque el rendimiento fue inferior al obtenido tras utilizar extracción convencional mediante el método Folch o Soxhlet. Eso sí, en el caso de los fluidos supercríticos se evitó la utilización de disolventes tóxicos. Asimismo, el aceite extraído presentó una mayor estabilidad oxidativa tras aplicar SC-CO2 a 30 y 40 MPa, en comparación con las extracciones realizadas a presiones más bajas o al procedimiento de extracción convencional. La extracción con SC-CO2 fue más eficiente para recuperar compuestos fenólicos lipofílicos con alta capacidad antioxidante en comparación con el procedimiento convencional de extracción (método Folch). Se observó una mayor extracción de los diferentes compuestos individuales, identificados y cuantificados por cromatografía líquida acoplada a espectrometría de masas en tándem cuando se aumentó la presión de tratamiento obteniendo los valores más altos cuando la presión fue de 40 MPa. Por otro lado, también se observó que tras utilizar la extracción convencional sólido/líquido, modificando el tiempo y la temperatura de extracción así como el disolvente utilizado, la eficiencia de extracción de los compuestos fenólicos totales fue mayor al aumentar la temperatura. Asimismo, concentraciones más altas de etanol y tiempos de extracción prolongados también promovieron una mejora en la extracción de estos compuestos. La capacidad antioxidante también aumentó cuando se utilizaron mayores concentraciones de etanol, con un mayor tiempo de extracción y a mayor temperatura. Estos resultados se podrían traducir en beneficios económicos derivados de la valorización de los subproductos frente a su eliminación con el fin de preservar el medio ambiente de residuos procedentes de la industria horchatera de una manera sostenible, reduciendo la huella de dióxido de carbono y proteger la salud del consumidor mediante la prevención o reducción de los impactos globales del uso de los recursos y la mejora de la eficacia de dicho uso. "Horchata de chufa" or more commonly known as "Horchata" is a traditional, non-alcoholic, sweet and whitish-looking drink obtained from tubers of tiger nut (Cyperus esculentus L. Sativus Boeck), consumed mainly in the Valencian Community during the summer season, although over the last years it has experienced a growth in its consumption both in Spain and in the rest of the world. During "Horchata" production process, a large amount of wastes and by-products are produced, both solid and liquids, where solid ones can represent up to 60% of the plant material. These by-products have been traditionally considered as industrial waste without commercial value and have been used mainly as animal feed. The liquid by-products stand out for their high content in prebiotics, antioxidants and as a substitute for water in meat products. On the other hand, solid by-products an be used for different applications, due to their high content of dietary fiber, antioxidant bioactive compounds, in addition to having good technological properties (water retention capacity, emulsion capacity and emulsion stability, among others). The aim of the present PhD Thesis is the recovery and characterization of nutrients and antioxidant bioactive compounds obtained from "Horchata" solid by-products. The study has been mainly focused on the extraction of oil and the evaluation of its fatty acid content and profile as well as vitamin E amount. The content of antioxidant bioactive compounds, total antioxidant capacity and physicochemical parameters of the obtained extracts have been also evaluated. To obtain the extracts, different extraction techniques were used, both conventional methods (eg conventional solid/liquid extraction, Soxhlet, etc.) and innovative processes (eg. supercritical fluids (SC-CO₂)), also known as "green extraction". Subsequently, the extracts were analyzed using different analytical techniques, such as spectrophotometry and chromatographic methods such as gas chromatography with flame ionization detector, liquid chromatography coupled to tandem mass spectrometry, etc. The potential use of other innovative processes such as ultrasounds, electric pulses, high pressures, alone and/or in combination with enzymes, gas-assisted mechanical expression, microwaves, which are also promising tools for the valorization of the by-products obtained during the elaboration of the "Horchata". After evaluating the results obtained, it was observed how "Horchata" by-products present an important content of oil, and antioxidant bioactive compounds, although it is necessary to select the appropriate extraction method in order to recover these compounds in the proper way. It was observed that the content of nutrients and antioxidant bioactive compounds differs according to the extraction method used. For example, after using extraction with supercritical fluids, an increase in the oil extraction was observed after increasing the pressure used, although the yield was lower than that obtained after using conventional extraction using the Folch or Soxhlet method. Of course, in the case of supercritical fluids the use of toxic solvents was avoided. Likewise, the extracted oil presented a greater oxidative stability after applying SC-CO2 at 30 and 40 MPa, in comparison with the extractions made at lower pressures or the conventional extraction procedure. Extraction with SC-CO2 was more efficient to recover lipophilic phenolic compounds with high antioxidant capacity compared to the conventional extraction method (Folch method). A greater extraction of the different individual compounds was observed, identified and quantified by liquid chromatography coupled to tandem mass spectrometry, when the treatment pressure was increased, obtaining the highest values when the pressure was 40 MPa. On the other hand, it was also observed that after using the conventional solid/liquid extraction, modifying the extraction time and temperature as well as the solvent used, the extraction efficiency of the total phenolic compounds was higher with increasing temperature. Moreover, higher concentrations of ethanol and longer extraction times also promoted an improvement in the extraction of these compounds. Antioxidant capacity also increased when higher ethanol concentrations were used, with a longer extraction time and higher temperature. These results could translate into economic benefits derived from the valuation of by-products as opposed to their elimination in order to preserve the environment.

Memoria presentada por José Ángel Clemente Ramos para optar al Grado de Doctor en Biología que ha sido realizada en el Instituto de Biología Funcional y Genómica, centro mixto de la Universidad de Salamanca (Departamento de Microbiología y Genética) y el Consejo Superior de Investigaciones Científicas., La pared celular es una estructura vital que confiere resistencia mecánica y soporte estructural a la célula fúngica que está ausente en las células animales. El gen bgs1+ de S. pombe codifica una hipotética subunidad catalítica responsable de la síntesis del beta(1,3) glucano lineal implicado en la biosíntesis del septo primario de la pared celular. La ausencia de Bgs1p produce células con defectos en separación y la selección del sitio de crecimiento polarizado originando células ramificadas con múltiples septos. El objetivo durante el desarrollo de este trabajo ha sido el estudio de la función de la proteína beta(1,3)glucán sintasa (GS) Bgs1 más allá de su papel como enzima biosintética sino como elemento de control esencial entre la Ruta de Activación de la Septación (SIN) y el establecimiento de la polaridad del crecimiento donde mediarían de manera antagónica con la Ruta de Morfogénesis (MOR). Hemos observado y demostrado que la ausencia de Bgs1p provoca un fenotipo multipolarizado en células con una baja actividad de la ruta SIN. Este defecto podía identificarse con cualquier elemento de la ruta. Además es específico, ya que no se reproduce en otros mutantes hiperpolarizados ni está asociado a la progresión mitótica de las células. En consonancia con estos resultados un aumento de la actividad de la ruta SIN durante la represión de bgs1+ provocaba el fenotipo opuesto en las células originando una pérdida de la polaridad y morfologías redondeadas. Dada la comunicación recíproca que se produce entre las rutas de señalización celular SIN y MOR decidimos estudiar si una reducción de la actividad MOR suprimía el fenotipo multipolarizado generado por la represión de bgs1+ en células con una baja actividad de la ruta SIN. Se comprobó que la supresión del fenotipo multipolarizado por la ruta MOR era específica y no estaba provocado por un bloqueo en el crecimiento o parada de la progresión mitótica de los mutantes. La explicación molecular más plausible al fenotipo multipolarizado de los mutantes se encontraría en la actividad descontrolada de la ruta MOR. El mecanismo de señalización implicado en la generación del fenotipo multipolarizado está controlado por la GTPasa Cdcp42p a través de la quinasa efectora Shk1p y la formina For3p. Gracias a un detallado estudio de la maquinaria de polaridad se comprobó que el complejo de establecimiento de la polaridad Tea1p-Tea4p y la quinasa Pom1p colaboran con Bgs1p en el mantenimiento del crecimiento polarizado, pero no son necesarias para la generación del fenotipo de multipolaridad en ausencia de Bgs1p y una ruta SIN defectuosa. La acumulación de parches de actina en los nuevos polos de crecimiento y la desorganización del citoesqueleto de tubulina son resulta crritica para el desarrollo del fenotipo multipolarizado. Las micotoxinas son sustancias producidas por determinados hongos presentes en alimentos destinados para alimentación humana o animal, y por su carácter tóxico pueden dar lugar a las enfermedades denominadas micotoxicosis.De ellas son las aflatoxinas (AFs) y la ocratoxina A (OTA) las que mayor preocupación han generado en el Comité Científico de Alimentación Humana de la Unión Europea, debido a los riesgos que el consumo continuado de estas micotoxinas a través de los alimentos comporta para la salud humana, por su poder mutagénico y cancerígeno, que se ha visto demostrado en animales expuestos a alimentos contaminados por ellas. La presencia natural de AFs y OTA está asociada a diferentes alimentos, por el crecimiento de especies fúngicas. Así las AFs se pueden encontrar en maíz, cacahuetes, arroz, vino y semillas de soja, debido al crecimiento en ellos de las especies fúngicas Aspergillus flavus, A. parasiticus y A. nomius. Mientras que la OTA se ha encontrado en cereales y derivados, legumbres, café, especias, cacao, frutos secos, malta y bebidas como la cerveza, por el crecimiento de los hongos Penicillium verrucosum y A. ochraceus. Las medidas legislativas sobre micotoxinas son recientes y en la actualidad cuentan con regulación específica a nivel comunitario las AFs y la OTA, entre otras. Y sólo para algunos alimentos se han establecido contenidos máximos admisibles, ingestas diarias y semanales tolerables y protocolos para su control, en el análisis y toma de muestras. Por este motivo los objetivos de esta tesis han sido:- Estudiar la extracción de AFs y OTA.- Estudiar la determinación de AFs y OTA por cromatografía líquida con diferentes técnicas de detección.- Aplicar los métodos seleccionados al análisis de materias primas y alimentos elaborados.- Calcular la ingesta diaria de AFs y OTA y su comparación con las ingestas diarias admisibles establecidas por el comité mixto de expertos de la FAO/OMS sobre aditivos alimentarios.Para ello se han empleado la técnica de extracción clásica con disolventes orgánicos, la extracción acelerada, dispersión de matriz en fase sólida, extracción con fluidos supercríticos y extracción con columnas de inmunoafinidad. Y se ha realizado estudio de la determinación de AFs y OTA por cromatografía líquida con las técnicas de detección fluorescencia, espectroscopia de masas y masas-masas. Estos métodos se han aplicado para el análisis de cereales y derivados, frutas, frutos secos y piensos procedentes de España, Portugal y Marruecos.Con los datos obtenidos se ha realizado el cálculo de la ingesta diaria de AFs y OTA utilizando los datos de consumo de cada país. El riesgo que implica el consumo de estas micotoxinas se ha evaluado comparando la ingesta diaria real con las ingestas diarias admisibles establecidas por el comité mixto de expertos de la FAO/OMS sobre aditivos alimentarios. Con el trabajo realizado, se puede concluir que: La extracción líquida presurizada de OTA ha proporcionado los resultados más exactos, precisos y reproducibles.La confirmación de la OTA por espectrometría de masas de triple cuadrupolo proporciona resultados más selectivos aunque la sensibilidad es ligeramente superior con la detección de fluorescencia.La incidencia de OTA en arroz ha sido del 13 % para las muestras procedentes de Portugal, 14 % para las de España y 39 % para las de Marruecos. En las muestras de pan analizadas esta incidencia ha sido del 39 % para las procedentes de Portugal y un 48 % para las de Marruecos. En los frutos secos procedentes de Marruecos, se ha obtenido una incidencia del 31 %.La incidencia de AFs en cereales, frutos secos y piensos comercializados en Marruecos es del 51, 26 y 67 %, respectivamente. De ellas, la AFB1 es la más frecuente y de mayor concentración.Las ingestas diarias estimadas de OTA por el consumo de los alimentos analizados son: 1 ng/kg pc/día para España, 4.27 ng/kg pc/día para Portugal y 125.5 ng/kg pc/día para Marruecos. La ingesta diaria estimada de AFs para la población de Marruecos es de 15.22 ng/kg pc/día., Mycotoxins are receiving increasing attention for its toxic effects and high incidence in a wide range of food commodities. Sufficient experimental evidence for carcinogenicity in animal studies has led to the classification of ochratoxin A as a possible human carcinogen (group 2B) and aflatoxin B1 as carcinogen (group 1) by the International Agency for Research on Cancer (IARC 1993). The Joint Committee FAO/WHO of Experts on Food Additives (JECFA) has established a provisional tolerable weekly intake (PTWI) of OTA at 100 ng kg/ of body weight corresponding to approximately 14 ng /kg body weight/day (JECFA 2001) and for aflatoxins as low are possible (ALARA).In Mediterranean and North African (Morocco) Area the climatic conditions characterized by high humidity and temperature and inadequate storage contribute to the potential for a significant mycotoxins exposure of population. Due to the significant health risks, it is important to establish a data collection on the occurrence of aflatoxins and ochratoxin A in foods. Much attention has been paid recently in the world to monitor the presence of aflatoxins and ochratoxin A in agricultural products intended for consumption and/or exportation.So that the objectives of this thesis were: study different OTA extraction methods in several matrices; analyze AF and OTA by liquid chromatography using different detection techniques; study different confirmatory processes of AF and OTA; evaluate the presence of AF and OTA in cereals, cereal products, dry fruits and poultry feed, purchased from supermarkets and markets from Spain, Portugal and Morocco; estimate daily intake of AF and OTA for Spanish, Portuguese and Moroccan population through analyzed samples; and compare AF and OTA estimated daily intake with tolerable daily intake proposed by official organisms, in order to evaluate the risk of exposing to both mycotoxins.
