COVID-19 poses a threat to social stability globally, which requires efficacious governance and public cooperation. To handle the COVID-19 pandemic, the Chinese national news media have mobilized the public to identify and collaborate with the nation. This paper investigates how war, chess, and examination metaphors, nation personification, and metonymies (i.e. the part representing the whole) are utilized in news reports and editorials to achieve the purpose. We adhere to the theoretical framework of Critical Metaphor Analysis and analyze 156 articles that are sourced from Xinhua News Agency and People’s Daily from January 22, 2020 to February 13, 2020. Besides demonstrating the specific entailments of these metaphors and metonymies, we also involve scenarios such as the ‘Heroic Fight’ scenario and the ‘Harmonious Family’ scenario. Those framing devices function to evoke patriotism and reinforce national identity by activating collective, historical, and cultural memories and evaluating in-group members positively.

Background: The primary manifestations of Alzheimer’s disease (AD) include cognitive decline and brain gray matter volume (GMV) atrophy. Recent studies have found that plasma phosphorylated-tau (p-tau) concentrations perform better in diagnosing, differentiating, and monitoring the progression of AD. However, the correlation between plasma p-tau, GMV, and cognition remains unclear. Objective: To investigate whether GMV plays a mediating role in the association between plasma p-tau concentrations and cognition. Methods: In total, 99 participants (47 patients with AD and 52 cognitively unimpaired [CU] individuals) were included. All participants underwent neuropsychological assessments, laboratory examinations, and magnetic resonance imaging scans. Plasma p-tau217 and p-tau181 concentrations were measured using an enzyme-linked immunosorbent assay kit. Voxel-based morphometry was performed to assess participants’ brain GMV. Partial correlation and mediation analyses were conducted in AD group. Results: Plasma p-tau concentrations were significantly higher in the AD group than in the CU group. Patients with AD had significant brain GMV atrophy in the right hippocampus, bilateral middle temporal gyrus, and right inferior temporal gyrus. In the AD group, there were significant correlations between plasma p-tau217 concentrations, GMV, and Mini-Mental State Examination (MMSE) scores. Brain GMV of the right hippocampus mediated the association between plasma p-tau217 concentrations and MMSE scores. A significant correlation between plasma p-tau181 and MMSE scores was not identified. Conclusion: The findings indicate that p-tau217 is a promising biomarker for central processes affecting brain GMV and cognitive function. This may provide potential targets for future intervention and treatment of tau-targeting therapies in the early stages of AD.

Since the pandemic broke out in 2020, China has widely presented the covid crisis in its mass media and actively constructed collective identity thereof to mobilize medical workers, unify political stances, boost domestic solidarity, and promote international support. This paper combines the Discourse-Historical Approach and a multimodal perspective to investigate how the Chinese state-run news agency People’s Daily discursively achieves these purposes on TikTok. A combination of qualitative and quantitative methods is used to present the high-frequency topoi of justifying the crisis and referential and predicational strategies of shaping collective identity within, which can fall into four dimensions: positive Self, negative Self, negative Others, and positive Others. The linguistic resources can be intensified/mitigated by visual-aural ensembles, which can draw the audience’s attention and arouse their emotional attachments. This study also summarizes the embedded values in the discourses and situates them in socio-political contexts.

Type 2 diabetes mellitus (T2DM) is associated with brain damage and cognitive decline. Despite the fact that the thalamus involves aspects of cognition and is typically affected in T2DM, existing knowledge of subregion-level thalamic damage and its associations with cognitive performance in T2DM patients is limited. The thalamus was subdivided into 8 subregions in each hemisphere. Resting-state functional and structural MRI data were collected to calculate resting-state functional connectivity (rsFC) and gray matter volume (GMV) of each thalamic subregion in 62 T2DM patients and 50 healthy controls. Compared with controls, T2DM patients showed increased rsFC of the medial pre-frontal thalamus, posterior parietal thalamus, and occipital thalamus with multiple cortical regions. Moreover, these thalamic functional hyperconnectivity were associated with better cognitive performance and lower glucose variability in T2DM patients. However, there were no group differences in GMV for any thalamic subregions. These findings suggest a possible neural compensation mechanism whereby selective thalamocortical functional hyperconnectivity facilitated by better glycemic control help to preserve cognitive ability in T2DM patients, which may ultimately inform intervention and prevention of T2DM-related cognitive decline in real-world clinical settings.

The macroscopic failure of concrete used to form an artificial dam in an underground coal mine reservoir is the final manifestation of crack propagation and penetration: the mode of crack propagation is key to the safety evaluation of concrete structures. To improve the performance of concrete in such an artificial dam forming an underground coal mine reservoir, many scholars have investigated nano-modified concrete, but the effects of nanomaterials on the crack propagation mode and failure mode of saturated concrete are not yet fully understood. In the present research, uniaxial compression and acoustic emission (AE) experiments were conducted on saturated concrete specimens containing different types and amounts of nanomaterials, and the crack propagation mode, failure mode, and bearing capacity of different saturated concrete specimens were compared and studied. The results show that: the failure mode, crack propagation mode, and bearing capacity of saturated concrete can be modified by adding nanomaterials, and the type and content thereof can affect the modification effect provided. Considering the strength, bearing capacity, and crack-propagation mode, saturated concrete specimens containing 3 wt% nano-Al2O3 exhibit the best performance, with characteristics of high strength, high bearing capacity, steady crack propagation, and small-scale cracking. The results have guiding significance for damage-warning and safety assessment of hydraulic structures.

The fusiform gyrus (FG) subserves a range of visual cognitive functions likely arising from its distinct subregions that present different connectivity profiles. Nevertheless, the molecular basis underlying such connectivity variability across FG subregions is still an open question. Resting-state functional magnetic resonance imaging (fMRI) data were collected from a discovery dataset (361 healthy subjects) and two independent cross-race, cross-scanner validation datasets (103 and 329 healthy subjects). We adopted a newly developed standardized pipeline to process gene expression data from the Allen Human Brain Atlas. Fine-grained FG subregions derived from the Human Brainnetome Atlas were utilized to measure seed-based resting-state functional connectivity (rsFC). Then, transcriptome-neuroimaging spatial association analyses were conducted to identify genes related to rsFC of each FG subregion. Results showed that rsFC of the left A37mv was associated with expression measures of 1063 genes, while there were no expression-rsFC correlations for the other subregions. The 1063 genes were mainly enriched for biological functions and pathways related to synaptic transmission, neurons, and neurotransmitter systems as well as for autism spectrum disorder. Specific expression analyses revealed that these rsFC-related genes were specifically expressed in brain tissue, in cortical neurons and immune cells, and during nearly all developmental periods. In addition, these genes were associated with multiple behavioral domains such as vision, language, and sensation. Finally, protein-protein interaction (PPI) analysis demonstrated that the genes could construct a PPI network with 37 hub genes. These findings may offer unique insights into the molecular basis underlying the functional heterogeneity of the FG.

Grain boundaries and pores commonly manifest in graphene sheets during experimental preparation. Additionally, pores have been intentionally incorporated into graphene to fulfill specific functions for various applications. However, how does the simultaneous presence of pores and grain boundaries impact the mechanical properties of graphene? This paper establishes uniaxial tension models of single-layer graphene-containing pores and three types of experimentally observed. The effect of interaction between pores and grain boundaries on the fracture strength of graphene was studied respectively for three types of grain boundaries by employing molecular dynamics simulations and considering factors such as pore size, the distance between pores and grain boundaries, and loading angle. A competitive mechanism between the intrinsic strength of pristine graphene with grain boundaries (referred to as pristine GGBs), which varies with the loading angle and the fracture strength of graphene sheets with pores that changes with the size of the pores, governs the fracture strength and failure modes of GGBs with pores. When the former exceeds the latter, the fracture strength of GGBs with pores primarily depends on the size of the pores, and fractures occur at the edges of the pores. Conversely, when the former is lower, the fracture strength of GGBs with pores relies on the loading angle and the distance between pores and grain boundaries, leading to grain boundary rupture. If the two strengths are comparable, the failure modes are influenced by the distance between pores and grain boundaries as well as the loading angle. The findings further elucidate the impact of coexisting grain boundaries and pores on the fracture behavior of graphene, providing valuable guidance for the precise design of graphene-based devices in the future.

For deep excavation projects with weak strata such as filled stones and mud, not only is excavation difficult, but instability and deformation problems are also prone to occur. In order to explore the sensitivity of weak strata parameters to the deformation of counterweight double-row piles, this paper takes a deep foundation pit project in Shenzhen as the research background, uses the finite element analysis software MIDAS GTS NX to conduct numerical simulation analysis of the excavation process of the foundation pit project, and compares the numerical simulation analysis results with on-site measured data. Finally, orthogonal test method and theoretical analysis method are used to analyze the sensitivity of three weak strata parameters on the deformation of counterweight double-row piles. The results show that the cohesive force C is the key influencing factor, the internal friction angle φ is an important influencing factor, and the elastic modulus E is a general influencing factor. The research results have certain engineering guidance significance for the design and construction of deep foundation pit projects in weak strata.

SUMMARY Numerical simulation of non-linear elastic wave propagation in rocks is indispensable for understanding stress/damage dependence of wave velocity changes and the associated micromechanisms. A numerical microstructural model is presented here to investigate seismic velocity changes due to stress and damage. By introducing pre-existing cracks and considering the valid microstructures in the bonded particle model, the proposed method successfully reproduces velocity changes of experiments on dry Lac du Bonnet granite and dry Darley Dale sandstone in both loading and unloading processes. Velocity increasing results from the closure of pre-existing cracks during loading stages, while the reopen of cracks during the unloading process causes velocity decreasing. Particle velocity vectors are used to illustrate wave propagation in a micromechanical way. P wave wave fronts are observed from the source to travel through the model, and wave intersections are clearly shown in the medium when the tensile wave front meets the compressive wave. The microstructure of the model shows a significant effect on rock mechanical behaviour and velocities and lends credibility to the velocity simulation. The valid microstructure produces realistic mechanical behaviour and velocity changes. Also, it replicates the initial hardening in the axial stress versus the axial strain curve, while invalid microstructures (e.g. cement overlap) underestimate the elastic modulus. The simulations also show that the wave velocities scale with the square root of the corresponding component of the coordination number, which can be used to quantify the mechanisms behind the velocity changes. Direct relations were established between velocity changes and opened crack density, which displays a similar tendency compared with predictions of the effective elastic theory. The microstructural model provides the ability to simulate the macro behaviour of rock under loadings in a more realistic manner and to directly examine the microprocesses underlying velocity changes.

Brain structural damage is a typical feature of schizophrenia. Investigating such disease phenotype in patients with drug-naive first-episode schizophrenia (DFSZ) may exclude the confounds of antipsychotics and illness chronicity. However, small sample sizes and marked clinical heterogeneity have precluded definitive identification of gray matter volume (GMV) changes in DFSZ as well as their underlying genetic mechanisms. Here, GMV changes in DFSZ were assessed using a neuroimaging meta-analysis of 19 original studies, including 605 patients and 637 controls. Gene expression data were derived from the Allen Human Brain Atlas and processed with a newly proposed standardized pipeline. Then, we used transcriptome–neuroimaging spatial correlations to identify genes associated with GMV changes in DFSZ, followed by a set of gene functional feature analyses. Meta-analysis revealed consistent GMV reduction in the right superior temporal gyrus, right insula and left inferior temporal gyrus in DFSZ. Moreover, we found that these GMV changes were spatially correlated with expression levels of 1,201 genes, which exhibited a wide range of functional features. Our findings may provide important insights into the genetic mechanisms underlying brain morphological abnormality in schizophrenia.

Understanding how microbial communities adapt to environmental stresses is critical for interpreting ecological patterns and microbial diversity. In the case of the Gobi Desert, little is known on the environmental factors that explain hypolithic colonization under quartz stones. By analyzing nine hypolithic communities across an arid gradient and the effects of the season of the year in the Hexi Corridor of this desert, we found a significant decrease in hypolithic colonization rates (from 47.24 to 15.73%) with the increasing drought gradient and found two distinct communities in Hot and Cold samples, which survived or proliferated after a hot or a cold period. While Cold communities showed a greater species diversity and a predominance of Cyanobacteria, Hot communities showed a predominance of members of the Proteobacteria and the Firmicutes. In comparison, Cold communities also possessed stronger functions in the photosynthesis and carbon metabolism. Based on the findings of this study, we proposed that the hypolithic communities of the Hexi Corridor of the Gobi Desert might follow a seasonal developmental cycle in which temperature play an important role. Thus after a critical thermal threshold is crossed, heterotrophic microorganisms predominate in the hot period, while Cyanobacteria predominate in the cold period.

Background Vitamin D is engaged in various neural processes, with low vitamin D linked to depression and cognitive dysfunction. There are gender differences in depression and vitamin D level. However, the relationship between depression, gender, vitamin D, cognition, and brain function has yet to be determined. Methods One hundred and twenty-two patients with major depressive disorder (MDD) and 119 healthy controls underwent resting-state functional MRI and fractional amplitude of low-frequency fluctuations (fALFF) was calculated to assess brain function. Serum concentration of vitamin D (SCVD) and cognition (i.e. prospective memory and sustained attention) were also measured. Results We found a significant group-by-gender interaction effect on SCVD whereby MDD patients showed a reduction in SCVD relative to controls in females but not males. Concurrently, there was a female-specific association of SCVD with cognition and MDD-related fALFF alterations in widespread brain regions. Remarkably, MDD- and SCVD-related fALFF changes mediated the relation between SCVD and cognition in females. Conclusion Apart from providing insights into the neural mechanisms by which low vitamin D contributes to cognitive impairment in MDD in a gender-dependent manner, these findings might have clinical implications for assignment of female patients with MDD and cognitive dysfunction to adjuvant vitamin D supplementation therapy, which may ultimately advance a precision approach to personalized antidepressant choice.

Dapagliflozin has been used to treat patients with type 1 diabetes mellitus; however, the actual drug efficacy of dapagliflozin on glycated hemoglobin (HbA1c) and whether there is a rebound from dapagliflozin efficacy on HbA1c remain unknown. The present study aimed to explore the actual therapeutic effect and rebound situation of dapagliflozin on HbA1c in type 1 diabetes mellitus patients. A total of 1,594 type 1 diabetes mellitus patients were enrolled for analysis using a non-linear mixed effect model from randomized controlled trials from published literature works including two 5 mg/day dapagliflozin dosage groups and three 10 mg/day dapagliflozin dosage groups. The change rate of HbA1c from a baseline value was chosen as a dapagliflozin pharmacodynamic evaluation index. After deducting control group effects, the therapeutic effect of 5 and 10 mg/day dapagliflozin on HbA1c in type 1 diabetes mellitus patients had no significant difference. In addition, the actual maximal efficacy (AEmax) of dapagliflozin on HbA1c was -6.24% at week 9. When it reached the AEmax, the dapagliflozin pharmacodynamic rebound on HbA1c occurred, and when the treatment was continued for 0.5 and 1 year, the actual efficacies were -4.70% (75% AEmax) and -3.27% (52% AEmax), respectively. This was the first time to clarify the actual therapeutic effect and rebound situation of dapagliflozin on HbA1c in type 1 diabetes mellitus patients, providing a reference value for clinical practices.

Neuronal oscillations within certain frequency bands are assumed to associate with specific neural processes and cognitive functions. To examine this hypothesis, transcriptome-neuroimaging spatial correlation analysis was applied to resting-state functional magnetic resonance imaging data from 793 healthy individuals and gene expression data from the Allen Human Brain Atlas. We found that expression measures of 336 genes were correlated with fractional amplitude of low-frequency fluctuations (fALFF) in the slow-4 band (0.027–0.073 Hz), whereas there were no expression-fALFF correlations for the other frequency bands. Furthermore, functional enrichment analyses showed that these slow-4 fALFF-related genes were mainly enriched for ion channel, synaptic function, and neuronal system as well as many neuropsychiatric disorders. Specific expression analyses demonstrated that these genes were specifically expressed in brain tissue, in neurons, and during the late stage of cortical development. Concurrently, the fALFF-related genes were linked to multiple behavioral domains, including dementia, attention, and emotion. In addition, these genes could construct a protein–protein interaction network supported by 30 hub genes. Our findings of a frequency-dependent genetic modulation of spontaneous neuronal activity may support the concept that neuronal oscillations within different frequency bands capture distinct neurobiological processes from the perspective of underlying molecular mechanisms.

Abnormal cell proliferation caused by abnormal transcription regulation mechanism seems to be one of the reasons for the progression of breast cancer and also the pathological basis. MicroRNA-142-5p (miR-142-5p) is a low-expressed miRNA in breast cancer. The role of MKL-1s regulation of DNMT1 in breast cancer cell proliferation and migration is still unclear. MKL-1 (myocardin related transcription factor A) can bind to the conserved cis-regulatory element CC (A/T) 6GG (called CarG box) in the promoter to regulate the transcription of miR-142-5p. The expressions of miR-142-5p and MKL-1 are positively correlated. In addition, it has been proved that DNMT1 is the target of miR-142-5p, which inhibits the expression of DNMT1 by targeting the 3-UTR of DNMT1, thereby forming a feedback loop and inhibiting the migration and proliferation of breast cancer. Our data provide important and novel insights into the MKL-1/miR-142-5p/DNMT1/maspin signaling pathway and may become a new idea for breast cancer diagnosis, treatment, and prognosis.

The outer dimensions and 3D model of ring forgings in industrial production are the important parameters to ensure the quality of the products. In order to realize the measurement of the outer dimensions and 3D model of the ring forgings, a measuring method for the ring forgings combining with a CCD camera and a 2D laser scanner is proposed in this paper. Firstly, the image information of the ring forgings is obtained by the CCD camera. The radiuses of different sections for ring forgings are acquired by pixel calibration and edge detection. Secondly, the point cloud data of different sections are obtained by laser scanner with the movement of the transmission mechanism. Then, the position discrimination method of straight line fitting is used to reduce the error data. And the radius matching method is used in the further processing of the point cloud data based on image detection, and the point cloud data with high accuracy is further extracted. Finally, the 3D point cloud data and 3D model of the ring forgings are obtained by rotating the 2D point cloud data. The outer dimensions of forgings are derived from the 3D model. The experiments were carried out with two different ring forgings. According to the experimental results, the feasibility of the method proposed in this paper is verified.

Tissue inhibitor matrix metalloproteinase 1 (TIMP1) has been reported to act as a tumor oncogene in colon cancer. However, little is known about the biological role of TIMP1 in gastric cancer. In this study, we found that the expression of TIMP1 in GC tissues was upregulated compared with the normal gastric tissues. TIMP1 was confirmed as a direct target of miR-6745 and silencing TIMP1 mimicked the effects of miR-6745 in GC cells. Further mechanism studies have shown that miR-6745 inhibits the Wnt/β-catenin pathway by targeting TIMP1, thereby inhibiting cell proliferation, migration and invasion. In addition, through the analysis of GC tissues, a negative correlation between miR-6745 and TIMP1 was found in 42 GC tissues. Our findings indicate that the miR-6745-TIMP1 axis regulates Wnt/βcatenin signaling and participates in GC tumorigenesis and provide a potential therapeutic target for preventing GC progression.

An accurate understanding of the breakage mechanism of broken coal and rock mass and its coupling relationship with stress and porosity is important for achieving efficient and safe production in coal mines, storage and utilisation of gas and water resources in goafs, and environmental ecological protection. In this study, a novel 3D simulation method is proposed for broken rock and coal granule compaction and breakage. This method can simulate the re-breakage characteristics of broken rock and coal granules during laterally confined compression (LCS). On this basis, numerical simulations combined with laboratory tests are conducted to quantitatively analyse the stress, porosity, and breakage rate evolution characteristics of broken rock granules during LCS. The entire loading process of broken rock granules is divided into three stages: self-adjustment, broken, and elastic. The stress evolution and breakage evolution characteristics of the broken rock granules during each loading stage are delineated. The breakage characteristics of broken rock granules are the main reasons for the evolution of stress, porosity, and breaking rate. In the elastic stage, only uniform compressive stress acts on the broken rock granule, inhibiting further breakage of the sample. When the loading stress reaches the tensile strength of the broken rock granules, the breakage rate of the models increases the fastest. The effects of the broken sample strength and sample size on the breakage characteristics and stress evolution law of the broken models during loading are further discussed. The secant modulus of the broken models in the elastic stages is approximately equal to the elastic modulus of the coal and rock samples. The coordination number evolution law of the broken granules during loading is the main factor affecting its breakage.

Microbiota of lower female reproductive tract is special in its microorganism composition with Lactobacillus as the predominant bacteria. A few of Lactobacillus species have been identified to benefit the inhibition of inflammatory and malignant diseases. Lacticaseibacillus casei LH23 is a strain isolated from traditional fermented food and had been demonstrate to ameliorate DSS-induced colitis in mice. In the present study, effects of Lacticaseibacillus casei LH23 on cervical cancer cells were investigated. Supernatants of lysates and heat-inactivated Lacticaseibacillus casei LH23 were found to inhibit the expression of human papillomavirus genes E6/E7 which is the main causative factor of cervical cancer. With MTT, EdU staining, and TUNEL staining assays, Lacticaseibacillus casei LH23 was shown to suppress the proliferation and induced the apoptosis of cervical cancer cells. Additionally, with wound-healing and Western-blot assays, Lacticaseibacillus casei LH23 was shown to slowdown the migration of cervical cancer cells and altered the expression of metastasis-related genes. These results demonstrated the anti-cervical cancer potential of Lacticaseibacillus casei LH23.

The electric vehicles (EVs) have played an important role in people's traveling due to the clean energy and friendly environment. Considering the endurance issue of EVs, wireless charging is regarded as a convenient and effective solution. However, the charging power and the efficiency would be greatly affected when the transmitting coil (TC) and the receiving coils are nonaligned. In this article, we present a flexible and high-accuracy positioning device based on electromagnetic balance, which could automatically achieve wireless charging and two-level positioning without communication between TC and EVs during the location. Besides, a six-coil cross-coupled circuit model has been built to optimize the key parameters that affect the positioning accuracy. The theoretical analysis and experimental results show that the proposed structures and location method have certain advantages in positioning accuracy, operation complexity, and practicability. Within the range of first-level positioning, the accuracy could achieve 1 cm. There is a high fitting performance between the simulation and experimental results, which verify the correctness of the theoretical analysis and the validity of the structures. Moreover, an additional comparative experiment has been designed to verify the correctness of the proposed method used in the wireless charging positioning system with ferrite cores and a magnetic shielding layer.

The human visual cortex is a heterogeneous entity that has multiple subregions showing substantial variability in their functions and connections. We aimed to identify genes associated with resting-state functional connectivity (rsFC) of visual subregions using transcriptome-neuroimaging spatial correlations in discovery and validation datasets. Results showed that rsFC of eight visual subregions were associated with expression measures of eight gene sets, which were specifically expressed in brain tissue and showed the strongest correlations with visual behavioral processes. Moreover, there was a significant divergence in these gene sets and their functional features between medial and lateral visual subregions. Relative to those associated with lateral subregions, more genes associated with medial subregions were found to be enriched for neuropsychiatric diseases and more diverse biological functions and pathways, and to be specifically expressed in multiple types of neurons and immune cells and during the middle and late stages of cortical development. In addition to shared behavioral processes, lateral subregion associated genes were uniquely correlated with high-order cognition. These findings of commonalities and differences in the identified rsFC-related genes and their functional features across visual subregions may improve our understanding of the functional heterogeneity of the visual cortex from the perspective of underlying genetic architecture.

The identification and treatment of mining-induced ground fissures are of great significance to mine safety and ecological and environmental protection. In this study, a novel method for ground fissure identification and exploration by infrared remote sensing onboard an unmanned aerial vehicle (UAV) was proposed. Using this method, a region of interest (ROI) that includes ground fissures directly above the middle of a long wall face, No. 12401 in the Shangwan coal mine, was monitored continuously during the day and night. Direct field measurements of ground fissure properties were also conducted to provide a calibration dataset for UAV measurements. Using the direct visible image at 5:00 pm as a reference, the average errors of the length and maximum width of Fissure I obtained from infrared images from 9:00 pm to 5:00 am on the next day, were estimated to be 1.8% and 6.5%, respectively. The diurnal variation of the fissure temperature is sinusoidal, and the range of temperature variation in the fissure decreases with the increase in depth. There is an apparent difference between the two common types of fissures depending on whether the fissure has a direct connection to an aquifer or a goaf. In this study, UAV, infrared thermal imager, and visible light camera data were successfully employed to effectively identify mining-induced ground fissures. In addition, the fissure detection error was validated, and the appropriate time for utilizing this method was obtained. Our results show that to identify the two aforementioned types of fissures, monitoring should be conducted between 3:00 am and 5:00 am. This study lays a foundation for the study and application of UAV and infrared thermal imagers for the identification of ground fissures induced by underground mining in large areas.

The novel duck reovirus (NDRV) is an emerging pathogen that causes disease in various waterfowl species. Since the outbreak, it has caused huge economic losses to the duck industry in China. A rapid, reliable, and high-throughput method is required for epidemiological investigation and evaluation of vaccine immunogenicity. A good first step would be establishing an enzyme-linked immunosorbent assay (ELISA) that could detect NDRV antibodies in different breeds of ducks and geese from the serum and egg yolk. This study used a recombinant NDRV σB protein and a corresponding horseradish peroxidase (HRP)-labeled monoclonal antibody to develop a blocking ELISA (B-ELISA). The cutoff value of the B-ELISA was 37.01%. A total of 212 serum samples were tested by the B-ELISA, and the virus neutralization test (VNT) was the gold standard test. The sensitivity and specificity of the B-ELISA were 92.17% (106/115) and 97.94% (95/97), respectively. The agreement rates between the B-ELISA and VNT were 94.81% (kappa value, 0.896). The B-ELISA could specifically recognize anti-NDRV sera without cross-reacting with other positive serums for other major diseases in ducks and geese. The inter- and intra-assay coefficients of variation (CVs) of the B-ELISA and VNT assays were acceptable. In conclusion, the novel B-ELISA could be a rapid, simple, safe, and economically attractive alternative to the VNT in assessing duck flocks' immunity status and in epidemiological surveillance in multiple waterfowl species.

Lonicerae Japonicae Flos(LJF), a bulk medicinal material, has long been used in clinical settings. The main/Dao-di production areas are Shandong, Henan, and Hebei. However, no systematic study on the difference in volatile components of LJF from different areas is available at the moment. In this study, gas chromatography-mass spectrometry(GC-MS) was used to detect the volatile components in 30 batches of LJF from 3 main production areas. Based on the relative odor activity value(ROAV), the key aroma components were analyzed. Multivariate statistical analysis was performed to analyze the differential components and characteristic aroma components in the samples from the 3 areas. Finally, 113 volatiles were identified from the samples, which were mainly alcohols, esters, acids, aldehydes, ketones, and alkenes. Among the common components of the three areas, linalool, myristic acid, and α-linolenic acid methyl ester had high content. A total of 15 key and 9 modifying aroma components in LJF were determined based on ROAV. The 15 differential components can be used for origin identification. Among them,(E, E)-2,4-decadienal and hexanal contributed a lot to the aroma of LJF from Henan and α-nerol was a characteristic aroma component of LJF in Hebei. In addition, lauryl aldehyde was a biomarker of LJF from Shandong. This study can provide a reference for the origin identification and quality evaluation of LJF.

It is always big challenges for hyaluronic acid (HA) in transmembrane absorbing and efficient delivering to the skin. Pep-1, as one of the cell-penetrating peptides, has been documented to permeate various substances across cellular membranes without covalent binding. Here, a novel hyaluronic acid binding peptide (named HaBP) is designed, and then combine with Pep-1 to enhance the cell-penetrating efficiency of HA. The results of ELISA and immunofluorescence assay show that HaBP could bind with HA very well, and a combination of Pep-1 and HaBP could efficiently improve the transmembrane ability of HA. Furthermore, HA gradually enter the dermis from the surface of the skin in mice when it is administrated with both HaBP and Pep-1, while there are no obvious allergies or other adverse reactions during this process. This study find a new method to promote the efficient transmembrane and transdermal absorption of HA, and throw some light on further research on the development of hyaluronic acid and its related cosmetics or drugs. Underground water reservoir (UWR) technology has been used to protect mine water resources. The main underground space for UWRs is the caving zone formed by longwall mining. Broken rock mass movement in the caving zone affects the porosity distribution and seepage characteristics during the UWR’s circulation, storage, and discharge. We used a fluid-solid coupled model, implemented in PFC3D and combined with Python, to simulate water seepage in the caving zone. The influence of particle size and fluid element size on the error of the simulation results was analysed. Then, we investigated the effect of seepage on broken rock mass porosity under different stress states. Simulations with various particle sizes and compaction stresses show that the average displacement of broken rock decreases exponentially with increased stress. Because large broken particles form the primary bearing structure in the model, it is mostly small particles that move under high-stress levels, so porosity is less affected. Larger particles also migrate if the compaction stress is low; this significantly changes the porosity and leads to roof uplift. Finally, we designed an engineering scale model based on our simulation method using the actual UWR situation in a coal mine.

This study investigated the development and formation of ground fissures at panel 12,401 of the Shangwan Colliery in the Shendong mining area, characterized by the current largest mining height and an aeolian sand surface in China. An integrated monitoring method was employed to examine the ground subsidence, distribution and development pattern of ground fissures, and the relationship between fissure width and depth, including an unmanned aerial vehicle, ground penetrating radar, and real-time kinematic and manual measurements. Then, the development and formation mechanisms of ground fissures were elucidated through theoretical analysis and physical simulation. The results indicate that ultra large mining height longwall panel extraction causes a greater ground subsidence and horizontal deformation than those of the conventions, resulting in more severe damage in the permanent margin area. The ratio of burial depth to mining height (H/M) decreases to less than 30 due to the ultra large mining height, and deep-burial panel displays the characteristics of shallow coal seam mining, such as “step subsidence”. The sliding instability of the primary key stratum causes the simultaneous cutting-off of the aeolian sand layer to form step-like ground fissures. The cause of the tensile fissures is the horizontal surface deformation exceeding the deformation threshold of the aeolian sand layer, and the dynamic tensile fissures were self-restored in 2 weeks as the panel advanced. Step-like fissures connected the working face with the ground, but the depth of the tensile fissures was less than 5.0 m and increased linearly with the fissure width.

As coal mining is extended from shallow to deep areas along the western coalfield, it is of great significance to study weakly cemented sandstone at different depths for underground mining engineering. Sandstones from depths of 101.5, 203.2, 317.3, 406.9, 509.9 and 589.8 m at the Buertai Coal Mine were collected. The characteristic strength, acoustic emission (AE), and energy evolution of sandstone during uniaxial compression tests were analyzed. The results show that the intermediate frequency (125–275 kHz) of shallow rock mainly occurs in the postpeak stage, while deep rock occurs in the prepeak stage. The initiation strength and damage strength of the sandstone at different depths range from 0.23 to 0.50 and 0.63 to 0.84 of peak strength (σc), respectively, decrease exponentially and are a power function with depth. The precursor strength ranges from 0.88σc to 0.99σc, increases with depth before reaching a depth of 300 m, and tends to stabilize after 300 m. The ratio of the initiation strength to the damage strength (k) ranges from 0.25 to 0.62 and decreases exponentially with depth. The failure modes of sandstone at different depths are tension-dominated mixed tensile-shear failure. Shear failure mainly occurs at the unstable crack propagation stage. The count of the shear failure bands before the peak strength increases gradually, and increases first and then decreases after the peak strength with burial depth. The cumulative input energy, released elastic energy and dissipated energy increase with depth. The elastic release rate ranges from 0.46 × 10–3 to 198.57 × 10–3 J/(cm3 s) and increases exponentially with depth.
