Focusing on the 2019 European Parliament campaign, we investigate parties’ engagement in negative campaigning on Facebook and the relationship to a parties’ ideology and their status as governing versus opposition party at the national level. Manual coding of 8,153 Facebook posts of parties from twelve European countries shows parties create less negative posts than positive and neutral ones. However, these negative posts attract more shares than positive, neutral, and balanced statements, which increases their prominence on the platform. Hence, users and algorithms create a negative campaign environment on Facebook to a greater extent than parties.

Clinicians have published research and reports on calcium and phosphate solubility within parenteral nutrition (PN) for over 40 years. Foundational empirical laboratory investigation in the 1980s motivated by the prevalence of neonatal rickets and osteomalacia in the Neonatal Intensive Care Unit (NICU) population led to precipitation curves that have guided PN prescribing and compounding. Over subsequent decades, numerous publications have expanded the knowledge of factors influencing calcium and phosphate solubility in formulating optimal and safe PN admixtures. Failure to adhere to known principles has led to serious injury and death. Known solubility curves are derived from empiric analysis of a finite number of conditions and concentrations, whereas custom PN orders vary widely and rarely match the admixture composition in the data set used to derive the published precipitation curves. Various commercial platforms have been developed to aid the pharmacist in assessing the potential for precipitation when evaluating a PN order. Some applications plot the calcium and phosphate concentrations of the prescribed PN against known published graphs most similar to the order, allowing the pharmacist to judge the risk of precipitation. Other approaches use intellectually protected trade secret algorithms to determine calcium and phosphate solubility across a continuum of conditions. This publication reports equations that have been used successfully for over 2 decades in our regional network of NICUs and shared with others to determine safe prescribing limits for calcium and phosphate concentrations using an electronic PN prescribing program.

Existing research has begun to document some of the troubling links between COVID-19 news consumption and emotional and psychological well-being, but precisely which sources of news are more likely to be related to such phenomena is still relatively unknown. Given the greater likelihood of encountering disinformation, rumour and other content of dubious origin via interpersonal networks (e.g., friends and family) on social media, we might assume that this is liable to spark greater confusion, fear, panic, anxiety and other negative emotions in comparison to, say, mainstream media. But through a nationally representative survey of U.K. news users we show that, on the whole, there were no significant statistical or practical differences in audiences’ emotional responses to the news between those who rely on social media as their primary source for COVID-19 news, and those who rely on other sources of news. Rather, we observe similarly high levels of negative and positive emotional responses for all types of news source. Findings are discussed in light of ongoing debates around news sources, emotions and public health.

X-linked retinitis pigmentosa (RP) is a severe form of RP, often with early macular involvement. This study aimed to characterise the natural history of patients with a diagnosis of X-linked RP due to

Acylglycerol kinase (AGK) is a mitochondrial lipid kinase that contributes to protein biogenesis as a subunit of the TIM22 complex at the inner mitochondrial membrane. Mutations in AGK cause Sengers syndrome, an autosomal recessive condition characterized by congenital cataracts, hypertrophic cardiomyopathy, skeletal myopathy, and lactic acidosis. We mapped the proteomic changes in Sengers patient fibroblasts and AGKKO cell lines to understand the effects of AGK dysfunction on mitochondria. This uncovered down-regulation of a number of proteins at the inner mitochondrial membrane, including many SLC25 carrier family proteins, which are predicted substrates of the complex. We also observed down-regulation of SFXN proteins, which contain five transmembrane domains, and show that they represent a novel class of TIM22 complex substrate. Perturbed biogenesis of SFXN proteins in cells lacking AGK reduces the proliferative capabilities of these cells in the absence of exogenous serine, suggesting that dysregulation of one-carbon metabolism is a molecular feature in the biology of Sengers syndrome.

Background Cystic echinococcosis (CE) is a zoonotic neglected tropical disease (zNTD) which imposes considerable financial burden to endemic countries. The 2021–2030 World Health Organization’s roadmap on NTDs has proposed that intensified control be achieved in hyperendemic areas of 17 countries by 2030. Successful interventions for disease control, and the scale-up of programmes applying such interventions, rely on understanding the associated costs and relative return for investment. We conducted a scoping review of existing peer-reviewed literature on economic evaluations of CE control strategies focused on Echinococcus granulosus zoonotic hosts. Methodology/Principal findings Database searches of Scopus, PubMed, Web of Science, CABI Direct and JSTOR were conducted and comprehensively reviewed in March 2022, using predefined search criteria with no date, field or language restrictions. A total of 100 papers were initially identified and assessed for eligibility against strict inclusion and exclusion criteria, following the Preferred Reporting Items for Systematic reviews and Meta-Analyses extension for Scoping Reviews (PRISMA-ScR) guidelines. Bibliography review of included manuscripts was used to identify additional literature. Full review of the final manuscript selection (n = 9) was performed and cost data for control interventions were extracted. Conclusions/Significance There are very little published data pertaining to the cost and cost effectiveness of CE control interventions targeting its zoonotic hosts. Data given for costs are often incomplete, thus we were unable to perform an economic analysis and cost effectiveness study, highlighting a pressing need for this information. There is much scope for future work in this area. More detailed information and disaggregated costings need to be collected and made available. This would increase the accuracy of any cost-effective analyses to be performed and allow for a greater understanding of the opportunity cost of healthcare decisions and resource allocation by stakeholders and policy makers for effective and cost-effective CE control.

European politics is increasingly being contested along two dimensions: the economic left-right dimension and a relatively new dimension focused on European integration and immigration. We test this framework at the party and individual-levels in the European Union. First, we use the Chapel Hill Expert Survey to demonstrate that there is no simple relationship between these dimensions at the party level in many European Union countries, and in fact the two dimensions are increasingly orthogonal. We then use the 2019 European Elections Study to show that the transnational-nationalist dimension significantly improves vote choice models relative to models that ignore this dimension. Even more striking, the transnational-nationalist dimension is not just significant, but actually improves vote choice models as much or more than the economic left-right dimension.

No contexto da Pandemia do Coronavírus 2019-20 os serviços psicológicos mediados pelas Tecnologia da Informação e Comunicação se tornaram essenciais para o equilíbrio da saúde mental da população em distanciamento social, isolamento domiciliar e/ou em quarentena, embora, desafiador ao considerar a ambiguidade da Sociedade da Informação e do Conhecimento. Estes serviços são atividades clínicas realizadas por psicológicos cadastros nos Conselhos Federal e Regionais de Psicologia através do e-Psi. O objetivo deste artigo versa na reflexão crítica sobre a tecnologia visualizando os atendimentos psicológicos mediados pelas Tecnologias da Informação e Comunicação no contexto da Pandemia do Coronavírus 2019-20 mentalizando a reconfiguração e/ou rompimento de paradigmas/obstáculos a partir de seus usos na Psicologia. O artigo caminha pela abordagem qualitativa, método descritivo-compreensivo-interpretativo e inclinação interdisciplinar de autores como possibilidade de edificação de um novo prisma acerca da construção, acesso e uso das Tecnologias da Informação e Comunicação na Psicologia na mediação dos serviços clínicos. O aporte teórico caminha pela área da Ciência da Informação e se apoia nas teorias da Revolução da Tecnologia da Informação e Filosofia da Tecnologia. Quando se considera a relação tecnologia, Psicologia, comportamento humano e serviços clínicos a Psicologia utiliza teorias clássicas na interpretação de novos construtos comportamentais e sociais originados na Sociedade da Informação e do Conhecimento. Entre 1995 a 2012 na perspectiva histórica-epistémico houve avanços significativos acerca dos estudos teóricos, técnicos e epistêmicos sobre os atendimentos psicológicos mediados pelas Tecnologia da Informação e Comunicação. Contudo, no contexto da contexto da Pandemia do Coronavírus 2019-20 emergem as necessidades de repensar as reservas, desconfianças e desafios quanto ao uso das tecnologias na psicologia clínica, visualizando os serviços posológicos como atividade efetiva, eficiente, eficaz, de qualidade e de alcance para os grupos sociais.

In order to summarize recent research on the prevention of allergies—particularly asthma—and stimulate new activities for future initiatives, a virtual workshop sponsored by the EAACI Clemens von Pirquet foundation and EUFOREA was held in October 2021. The determinants of the “allergic march” as well as the key messages from intervention studies were reviewed by an international faculty of experts. Several unmet needs were identified, and a number of priorities for future studies were proposed.

Here, the feasibility of using nonimaging pulse-echo ultrasound as a method of noninvasively detecting a bolus in the proximal esophagus was demonstrated. To accomplish this, patient swallows were recorded on a clinical ultrasound device with research interface that allowed for collection of the pulse-echo data. These ultrasound data of the proximal esophagus were processed with a series of signal processing techniques in both the temporal and spectral domains, which revealed characteristic signatures that were unique for both liquid and food boluses compared to the normal collapsed esophageal state. Since substantial amounts of laryngopharyngeal reflux are gaseous in nature, future work will revolve around expanding the data set to include boluses of gaseous refluxate, a standardized methodology for capturing bolus events; developing automated detection tools for identifying laryngopharyngeal reflux for an extended duration; and assessing technology limitations due to user error.

Titania microspheres have attracted substantial attention for a variety of applications, including ion scavenging, catalysis, and energy generation, though most synthetic techniques are limited to a few basic morphologies and narrow size ranges. Here, an intensified microfluidic strategy for continuous synthesis of anatase titania microspheres is presented. In-flow photo crosslinking, incorporated with a flow reactor and polar aprotic solvent, enables access to precursor compositions up to an order of magnitude higher than those previously reported, with size tunability approaching two orders of magnitude. Morphological and surface area effects associated with precursor composition are explored, resulting in hollow, yolk–shell, macroporous, and dense titania microspheres containing no detectable rutile phase and possessing surface areas exceeding 350 m2 g−1 post calcination. Furthermore, effects of calcination temperature and time on the surface area, crystallinity and phase composition, and morphology of the synthesized titania microspheres are studied in detail. The synthesized microspheres are shown to remain completely in the anatase phase, even at temperatures up to 900 °C, far beyond the expected phase transition temperature. Thus, the breadth of attainable morphologies, specific surface areas, and phase compositions present a variety of intriguing substrate candidates for such applications as heterogeneous (photo) catalysis, adsorption and ion capture, electrochemistry, and photovoltaics.

Based on a longitudinal research design (2006–2017), this article analyses how Guardian journalists engage in “below the line” comment spaces; what factors shape this engagement; and how this has evolved over time. The article combines a large-scale quantitative analysis of the total number of comments made (n = 110,263,661) and a manual content analysis of all comments made by 26 journalists (n = 5448) and their broader writing practices with 18 semi-structured interviews conducted in two phases (13 in 2012 and 5 repeated in 2017–18). The results show that there is considerable interest in comment spaces amongst readers, with exponential growth in user commenting. Furthermore, there has been significant engagement below the line by some Guardian journalists, and this is often in the form of direct and sustained reciprocity. Journalist commenting has waned in recent years due to difficulties coping with the volume of comments; changes in editorial emphasis; concerns over incivility and abuse; and a decrease in perceived journalistic benefits of commenting, alongside the rise in importance of Twitter. When journalists comment, they do so in a variety of ways and their comments are often substantive, significantly adding to the story by, for example, defending and explaining their journalism practice.

© The Author(s) 2019. This introduction unpacks the eight articles that make up this Journalism special issue about election reporting. Taken together, the articles ask: How has election reporting evolved over the last century across different media? Has the relationship between journalists and candidates changed in the digital age of campaigning? How do contemporary news values influence campaign coverage? Which voices – politicians, say or journalists – are most prominent? How far do citizens inform election coverage? How is public opinion articulated in the age of social media? Are sites such as Twitter developing new and distinctive election agendas? In what ways does social media interact with legacy media? How well have scholars researched and theorised election reporting cross-nationally? How can research agendas be enhanced? Overall, we argue this Special Issue demonstrates the continued strength of news media during election campaigns. This is in spite of social media platforms increasingly disrupting and recasting the agenda setting power of legacy media, not least by political parties and candidates who are relying more heavily on sites such as Facebook, Instagram and Twitter to campaign. But while debates in recent years have centred on the technological advances in political communication and the associated role of social media platforms during election campaigns (e.g. microtargeting voters, spreading disinformation/misinformation and allowing candidates to bypass media to campaign), our collection of studies signal the enduring influence professional journalists play in selecting and framing of news. Put more simply, how elections are reported still profoundly matters in spite of political parties’ and candidates’ more sophisticated use of digital campaigning.

Despite the sustained growth in journalism as a choice of degree path for young people, our understanding of students’ aspirations and motivations remains relatively underdeveloped. At the same time, journalism careers appear increasingly uncertain, as the industry responds to digitalization and convergence. In this mixed-methods study - employing 35 interviews and a survey of 837 UK journalism students - we ask what areas of journalism do students aspire towards, how do they feel about their future career prospects, and what is motivating them to study journalism in the first place? We find that intrinsic motivations (calling and talent, dynamic job) prevail over public service ones, with students drawn to soft news beats over hard news. Aspirations are also strikingly gendered, opening up questions of journalism education in this process. We also find that while students articulate an aspirational career in respected media outlets, they are pragmatic about their immediate career prospects. Here, journalism education appears to play a significant role in socialising students towards careers beyond journalism. Findings are discussed in the light of ongoing debates around journalistic socialisation and the future of journalism.

The relationship between media, sport, nations, and nationalism is well established; yet, there is an absence of these discussions at the intersection of communication, Paralympics, and disability studies. This omission is particularly significant considering the rapid commodification of the Paralympic spectacle, exacerbated by the entry of Channel 4 (C4) as the UK Paralympic rights holders, that has seen the games become an important site of disability (re-)presentation. In this article, we focus on the construction of national, normative, disabled bodies in Paralympic representation drawn from an analysis of three integrated data sets from C4’s broadcasting of the Rio 2016 Paralympics: interviews with C4 production and editorial staff, quantitative content analysis, and qualitative moving image analysis. We highlight the strategic approach taken by C4 to focus on successful medal-winning athletes, the implications this has on the sports and disability classifications given media coverage, and the role of affective high-value production practices. We also reveal the commercial tensions and editorial decisions that broadcasters face with respect to which disabilities/bodies are made hypervisible—and thereby those which are marginalized—as national disability sport icons that inculcate preferred notions of disability and the (re-)imagined nation.

X-linked retinitis pigmentosa (RP) is a severe form of RP, often with early macular involvement. This study aimed to characterise the natural history of patients with a diagnosis of X-linked RP due to RP2 mutations. Clinical details, best-corrected visual acuity (BCVA) and multimodal retinal imaging were retrospectively collected from patients with RP2 variants from Moorfields Eye Hospital (London, UK). Measures of the ellipsoid-zone (EZ) width, central retinal thickness (CRT), and thickness of the photoreceptor and retinal pigment epithelium complex (PR+RPE, taken between the external limiting membrane and RPE) were extracted from spectral-domain optical coherence tomography (SD-OCT) scans. A total of 47 affected males (median baseline age: 20 years, IQR: 12.5–36.5) were included, and 41 had two or more visits (median follow-up: 8.0 years, IQR: 3.2–14.5). A total of 24 RP2 variants were identified, 13 of which were novel. BCVA dropped from 0.66 LogMAR at baseline (IQR, 0.35–1.4) to 1.3 LogMAR at the most recent visit (IQR: 0.6–1.4). SD-OCT revealed a prevalent outer retinal atrophy (n = 23/35, 65.7%), and measurable EZ width at baseline in 34.3% of patients (n = 12). Age significantly affected all quantitative measures (p < 0.001) except EZ width (p = 0.58), with exponential decays of 46–49% and 12.6–33.9% per decade for BCVA and SD-OCT measures, respectively. RP2 patients exhibited rapid progression to outer retina atrophy and early macular involvement with substantial vision loss by age 30–40.

Existing research has documented the dynamics of increased news consumption alongside – paradoxically – increased news avoidance during the Covid-19 pandemic, highlighting its adverse effects on mental health and emotional wellbeing. However, for methodological and theoretical reasons, research still lacks specifics on what types of negative psychological responses were directly triggered by pandemic news, how prevalent they were in the population, how they manifested in daily life, and what could be the alternatives to them. Further, the almost exclusive focus on negative effects has led to a relative negligence of the positive sides of pandemic news. This study takes a mixed-method approach to address these gaps, combining 59 interviews and a follow-up survey with a representative sample of 2,015 adults across the UK. We found that pandemic news consumption, driven primarily by the need for personalised surveillance in an uncertain situation, oscillated in parallel with its severity and associated lockdown restrictions. The influx of repetitive bad news triggered many negative feelings besides general pandemic anxiety – namely fear, despair and moral outage (anger and disgust). This led to various alterations of daily routines, including news avoidance. Such adverse effects were offset by the reassurance, happiness and hope that the news did, at least occasionally, brought to audiences during the pandemic. Participants suggested several potential “good news” categories that point to the need for constructive news forms that not only inform but also inspire, motivate and/or empower people in personal or collective ways.

Websites are malleable: users can run code in the browser to customize them. However, this malleability is typically only accessible to programmers with knowledge of HTML and Javascript. Previously, we developed a tool called Wildcard which empowers end-users to customize websites through a spreadsheet-like table interface without doing traditional programming. However, there is a limit to end-user agency with Wildcard, because programmers need to first create site-specific adapters mapping website data to the table interface. This means that end-users can only customize a website if a programmer has written an adapter for it, and cannot extend or repair existing adapters. In this paper, we extend Wildcard with a new system for end-user web scraping for customization. It enables end-users to create, extend and repair adapters, by performing concrete demonstrations of how the website user interface maps to a data table. We describe three design principles that guided our system’s development and are applicable to other end-user web scraping and customization systems: (a) users should be able to scrape data and use it in a single, unified environment, (b) users should be able to extend and repair the programs that scrape data via demonstration and (c) users should receive live feedback during their demonstrations. We have successfully used our system to create, extend and repair adapters by demonstration on a variety of websites and we provide example usage scenarios that showcase each of our design principles. Our ultimate goal is to empower end-users to customize websites in the course of their daily use in an intuitive and flexible way, and thus making the web more malleable for all of its users.

In the context of the UK’s impending withdrawal from the European Union, we characterise the 2019 EU election in the UK as a ‘ghost campaign’, which did not have the attention of the major parties, let alone the voter. Such a backdrop may have invited parties to innovate or experiment with Facebook campaigning strategies, but the campaign was largely what one might expect in terms of the exploitation of the affordances of Facebook: cautious. Similarly, it followed the patterns of previous UK European Parliamentary elections by focusing largely on national and domestic politics with the parties offering persuasive communication intended to deliver them votes on election day. Perhaps unsurprisingly, Brexit dominated the topical focus of party Facebook posts.

COJO for COVID Recovery is a UKRI/AHRC-funded rapid COVID-19 response research project that addresses the role that constructive journalism (COJO), particularly solution-focused journalism, can play in the pandemic recovery. The research set out in this report details the findings of the project’s initial in-depth interviews and survey with UK news audiences. Four key sets of findings regarding COVID-19 news consumption and experience emerge from this analysis. 1. The high interest in COVID-19 news was associated with the need to monitor and navigate the uncertain evolution of the pandemic as it affected audiences. 2. There was a clear fluctuation that followed “an inverse Bell curve” (up-down-up) pattern of COVID-19 news consumption which was in parallel with the severity of the pandemic and its associated lockdown restrictions. 3. Many of our participants reported switching off from the news or avoiding it for reasons linked to mental and emotional health, including negativity, repetitive reporting and information overload. 4. Participants demanded the media provide more positive news that helps to lift the mood of the public and/or offers ways out of the pandemic.

This preprint presents preliminary findings from a major new study that examines UK audience perceptions of solutions-oriented journalism in the context of the COVID-19 pandemic. Using a nationally representative survey (n=2015) and in-depth interviews (n=59), members of the UK public were asked about their evaluations of problems-focussed news and solutions-orientated news. Findings suggest that UK audiences are – on the whole – positive towards the concept of solutions-oriented news, identifying its potential to enhance their interest in the news, feel more positive about the potential for change, and empower them to take actions in their local community. We also reveal some of the demographic factors that are associated with support for solutions journalism.

The mechanisms by which cells respond and adapt to oxidative stress are largely unknown but are key to developing a rationale for cancer therapies that target antioxidant pathways. APR-246 is a mutant-p53 targeted therapeutic currently under clinical investigation in myeloid dysplastic syndrome (MDS) and acute myeloid leukemia1. Whilst the mechanism of action of APR-246 is thought to be reactivation of wild-type p53 activity through covalent modification of cysteine residues in the core domain of mutant-p53 protein2,3, here we report that the anti-neoplastic capacity of APR-246 lies predominantly in the conjugation of free cysteine. Genome-wide CRISPR perturbation screening, metabolite profiling and proteomics in response to APR-246 treatment in mutant-p53 cancer cells highlighted the role of GSH and mitochondrial metabolism in determining APR-246 efficacy. APR-246 sensitivity was increased through loss of key enzymes in mitochondrial one-carbon metabolism,SHMT2andMTHFD1L, due to diminished glycine supply forde novoGSH synthesis. Critically, we show that APR-246 induces iron-dependent, apoptotic machinery-independent cell death, ferroptosis. Whole-cell proteomics analyses indicated an upregulation of proteins involved in iron-sulfur cluster biogenesis (eg. FDX1). GSH, acetyl-CoA and NADH levels were also depleted in APR-246 treated cells. Importantly, we found that APR-246 inhibits iron-sulfur cluster biogenesis in the mitochondria of cancer cells through cysteine conjugation. This work not only details novel determinants of APR-246 activity in cancer cells, but also provides a clinical roadmap for targeting antioxidant pathways in tumours - beyond targeting mutant-p53 tumours.

Customizing software should be as easy as using it. Unfortunately, most customization methods require users to abruptly shift from using a graphical interface to writing scripts in a programming language. We introduce data-driven customization, a new way for end users to extend software by direct manipulation without doing traditional programming. We augment existing user interfaces with a table view showing the structured data inside the application. When users edit the table, their changes are reflected in the original UI. This simple model accommodates a spreadsheet formula language and custom data-editing widgets, providing enough power to implement a variety of useful extensions. We illustrate the approach with Wildcard, a browser extension that implements data-driven customization on the web using web scraping. Through concrete examples, we show that this paradigm can support useful extensions to many real websites, and we share reflections from our experiences using the tool. Finally, we share our broader vision for data-driven customization: a future where end users have more access to the data inside their applications, and can more flexibly repurpose that data as part of everyday software usage.

Programmers are told "depend on interfaces, not implementations." But, given a program, is it possible even to assess objectively whether such advice has been followed? Programmers frequently talk in ways like this about dependence, but the very term, like many used in software engineering, has hitherto eluded precise definition. In this work, we resolve a variety of confusions about dependence, and present a formal definition unifying multiple varieties of software dependence, grounded in Halpern and Pearl's theory of actual causation. This definition is parameterized by the formal system characterizing the property of interest, and by constraints on "reasonable changes" to the program. By picking different choices of formal system, one can specialize our definition to characterize several notions of dependence, including build, correctness, and performance dependences. Overall, our work provides a path to making conversations about software dependence fully objective, and might serve as a basis for future work that automatically checks forms of dependence that were previously too abstract or high-level to be candidates for tools.

This article provides a description of the rationale and processes adopted by the Gulf Coast Health Alliance: Health Risks related to the Macondo Spill consortium to evaluate and communicate the risk of exposure to polycyclic aromatic hydrocarbons (PAHs) in seafood over several years following the Deepwater Horizon disaster and subsequent oil spill. We examined gaps in knowledge associated with PAH toxicity following exposure to petrogenic (oil-derived) PAHs by studying the metabolic fate of PAHs and their potential toxicity using sophisticated analytical methods. Using the data generated, we developed a risk communication strategy designed to meet the needs of the stakeholder communities including a consumption guideline calculator, a web-based tool to reconcile seafood consumption with risk of adverse health effects.
