Background While adult outcome in autism spectrum disorder (ASD) is generally measured using socially valued roles, it could also be understood in terms of aspects related to health status – an approach that could inform on potential gender differences. Methods We investigated gender differences in two aspects of outcome related to health-status, i.e. general functioning and self-perceived health status, and co-occurring health conditions in a large multi-center sample of autistic adults. Three hundred and eighty-three participants were consecutively recruited from the FondaMental Advanced Centers of Expertise for ASD cohort (a French network of seven expert centers) between 2013 and 2020. Evaluation included a medical interview, standardized scales for autism diagnosis, clinical and functional outcomes, self-perceived health status and verbal ability. Psychosocial function was measured using the Global Assessment of Functioning scale. Results While autistic women in this study were more likely than men to have socially valued roles, female gender was associated with poorer physical and mental health (e.g. a 7-fold risk for having three or more co-occurring physical health conditions) and a poorer self-perceived health status. Psychosocial function was negatively associated with depression and impairment in social communication. Half of the sample had multiple co-occurring health conditions but more than 70% reported that their visit at the Expert Center was their first contact with mental health services. Conclusions To improve objective and subjective aspects of health outcome, gender differences and a wide range of co-occurring health conditions should be taken into account when designing healthcare provision for autistic adults.

Although reported in 15% of affected individuals, little is known about the specificity of their clinical profiles. Adaptive skills represent a holistic approach to a person's competencies and reflect specifically in autism, their strengths and difficulties. Methods In this study, we hypothesised that individual with ASD with a history of MIA (MIA+) could be more severely socio-adaptively impaired than those without MIA during pregnancy (MIA−). To answer this question, we considered two independent cohorts of individuals with ASD (PARIS study and FACE ASD) screened for pregnancy history, and used a supervised and unsupervised statistical approach. Results We included 295 mother-child dyads with 14% of them with MIA+. We found that ASD-MIA+ individuals displayed more severe maladaptive behaviors, specifically in their socialization abilities. MIA+ directly influenced individual's socio-adaptive skills, independent of other covariates, including ASD severity. Interestingly, MIA+ may affected persistently the socio-adaptive behavioral trajectories of individuals with ASD. Limitations The current study has a retrospective design with possible recall bias regarding the MIA event and, even if pooled from two cohorts, has a relatively small population. In addition, we were limited by the number of covariables available potentially impacted socio-adaptive behaviors. Larger prospective study with additional dimensions related to ASD is needed to confirm our results Conclusions Specific pathophysiological pathways may explain these clinical peculiarities of ASD- MIA+ individuals, and may open the way to new perspectives in deciphering the phenotypic complexity of autism and for the development of specific immunomodulatory strategies.

This article describes the setup and performance of the near and far detectors in the Double Chooz experiment. The electron antineutrinos of the Chooz nuclear power plant were measured in two identically designed detectors with different average baselines of about 400 m and 1050 m from the two reactor cores. Over many years of data taking the neutrino signals were extracted from interactions in the detectors with the goal of measuring a fundamental parameter in the context of neutrino oscillation, the mixing angle {\theta}13. The central part of the Double Chooz detectors was a main detector comprising four cylindrical volumes filled with organic liquids. From the inside towards the outside there were volumes containing gadolinium-loaded scintillator, gadolinium-free scintillator, a buffer oil and, optically separated, another liquid scintillator acting as veto system. Above this main detector an additional outer veto system using plastic scintillator strips was installed. The technologies developed in Double Chooz were inspiration for several other antineutrino detectors in the field. The detector design allowed implementation of efficient background rejection techniques including use of pulse shape information provided by the data acquisition system. International audience; Chloroazaphosphatranes, the corresponding halogenophosphonium cations of the Verkade superbases, were evaluated as a new motif for halogen bonding (XB). Their modulable synthesis allowed for synthetizing chloroazaphosphatranes with various substituents on the nitrogen atoms. The binding constants determined from NMR titration experiments for Cl − , Br − , I − , AcO − , and CN − anions are comparable to those obtained with conventional iodine-based monodentate XB receptors. Remarkably, the protonated azaphosphatrane counterparts display no affinity for anions under the same conditions. The strength of the XB interaction is, to some extent, related to the basicity of the corresponding Verkade superbase. The halogen bonding abilities of this new class of halogen donor motif were also revealed by the Δδ(31 P) NMR shift observed in CD 2 Cl 2 solution in the presence of triethylphosphine oxide (TEPO). Thus, chloroazaphosphatranes constitute a new class of halogen bond donors, expanding the repertory of XB motifs mainly based on C Ar −I bonds.

International audience; The resistance-nodulation-division (RND)-type efflux pumps AcrAB and MdtABC contribute to multidrug-resistance (MDR) in Gram-negative bacteria. Photorhabdus is a symbiotic bacterium of soil nematodes that also produces virulence factors killing insects by septicaemia. We previously showed that mdtA deletion in Photorhabdus laumondii TT01 resulted in no detrimental phenotypes. Here, we investigated the roles of the last two putative RND transporters in TT01 genome, AcrAB and AcrAB-like (Plu0759-Plu0758). Only ΔacrA and ΔmdtAΔacrA mutants were multidrug sensitive, even to triphenyltetrazolium chloride and bromothymol blue used for Photorhabdus isolation from nematodes on the nutrient bromothymol blue-triphenyltetrazolium chloride agar (NBTA) medium. Both mutants also displayed slightly attenuated virulence after injection into Spodoptera littoralis. Transcriptional analysis revealed intermediate levels of acrAB expression in vitro, in vivo and post-mortem, whereas its putative transcriptional repressor acrR was weakly expressed. Yet, plasmid-mediated acrR overexpression did not decrease acrAB transcript levels neither MDR in TT01 WT. While no pertinent mutations were detected in acrR of the same P. laumondii strain grown either on NBTA or nutrient agar, we suggest that AcrR-mediated repression of acrAB is not physiologically required under conditions tested. Finally, we propose that AcrAB is the primary RND-efflux pump, which is essential for MDR in Photorhabdus and may confer adaptive advantages during insect infection.

Antibiotic resistance is an increasing threat to human health. A direct link has been established between antimicrobial self-resistance determinants of antibiotic producers, environmental bacteria, and clinical pathogens. Natural odilorhabdins (ODLs) constitute a new family of 10-mer linear cationic peptide antibiotics inhibiting bacterial translation by binding to the 30S subunit of the ribosome. These bioactive secondary metabolites are produced by entomopathogenic bacterial symbiont Xenorhabdus (Morganellaceae), vectored by the soil-dwelling nematodes. ODL-producing Xenorhabdus nematophila symbionts have mechanisms of self-protection. In this study, we cloned the 44.5-kb odl biosynthetic gene cluster (odl-BGC) of the symbiont by recombineering and showed that the N-acetyltransferase-encoding gene, oatA, is responsible for ODL resistance. In vitro acetylation and liquid chromatography-tandem mass spectrometry (LC-MS/MS) analyses showed that OatA targeted the side chain amino group of ODL rare amino acids, leading to a loss of translation inhibition and antibacterial properties. Functional, genomic, and phylogenetic analyses of oatA revealed an exclusive cis-link to the odilorhabdin BGC, found only in X. nematophila and a specific phylogenetic clade of Photorhabdus. This work highlights the coevolution of antibiotic production and self-resistance as ancient features of this unique tripartite complex of host-vector-symbiont interactions without odl-BGC dissemination by lateral gene transfer.

A θ oscillation analysis based on the observed antineutrino rates at the Double Chooz far and near detectors for different reactor power conditions is presented. This approach provides a so far unique simultaneous determination of θ and the total background rates without relying on any assumptions on the specific background contributions. The analysis comprises 865 days of data collected in both detectors with at least one reactor in operation. The oscillation results are enhanced by the use of 24.06 days (12.74 days) of reactor-off data in the far (near) detector. The analysis considers the ν¯ interactions up to a visible energy of 8.5 MeV, using the events at higher energies to build a cosmogenic background model considering fast-neutrons interactions and Li decays. The background-model-independent determination of the mixing angle yields sin(2θ) = 0.094 ± 0.017, being the best-fit total background rates fully consistent with the cosmogenic background model. A second oscillation analysis is also performed constraining the total background rates to the cosmogenic background estimates. While the central value is not significantly modified due to the consistency between the reactor-off data and the background estimates, the addition of the background model reduces the uncertainty on θ to 0.015. Along with the oscillation results, the normalization of the anti-neutrino rate is measured with a precision of 0.86%, reducing the 1.43% uncertainty associated to the expectation. This randomized controlled trial aimed to evaluate the effectiveness of the Eye Movement Desensitization and Reprocessing Protocol for Recent Critical Incidents and Ongoing Traumatic Stress (EMDR-PRECI) in reducing posttraumatic stress disorder (PTSD), anxiety, and depression symptoms related to the work of first responders on active duty. Participants were randomly assigned to two 60-minute individual treatment sessions (N = 30) or to a no-treatment control condition (N = 30). They completed pre-, post-, and follow-up measurements using the Posttraumatic Stress Disorder Checklist for Diagnostic and Statistical Manual of Mental Disorders, fifth edition (DSM-5) (PCL-5) and the Hospital Anxiety and Depression Scale (HADS). Data analysis by repeated measures analysis of variance (ANOVA) showed clear effects of the EMDR-PRECI in reducing PTSD work-related symptoms in the treatment group with symptom reduction maintained at 90-day follow-up with a large effect size (d = 3.99), while participants continued to experience direct exposure to potentially traumatic work-related events during the follow-up period. Data analysis by repeated measures ANOVA revealed a significant interaction between time and group, F (2,116) = 153.83, p < .001, ηP2 = .726 for PTSD, and for anxiety F (1,58) = 37.40, p < .005, ηP2 = .090, but not for depression. A t-test showed a clear decrease for depression symptoms for the treatment group with statistically significant results. The study results suggest that the EMDR-PRECI could be an efficient and effective way to address first responders' work-related PTSD, anxiety and depression symptoms. Future research is recommended to replicate these results and to investigate if symptom improvement also results in the reduction of physical health symptoms and early retirement for PTSD-related reasons among first responders.

NOSO-502 is a preclinical antibiotic candidate of the odilorhabdin class. This compound exhibits activity against Enterobacteriaceae pathogens, including carbapenemase-producing bacteria and most of the colistin (CST)-resistant strains. Among a collection of CST-resistant Klebsiella pneumoniae strains harboring mutations in the genes pmrAB, mgrB, phoPQ, and crrB, only those bearing mutations in the gene crrB were found to be resistant to NOSO-502. CrrB is a histidine kinase which acts with the response regulator CrrA to modulate the PmrAB system, which induces the restructuring of lipopolysaccharide on the outer membrane and thus leads to CST resistance. Moreover, crrB mutations also enhance the transcription of neighboring genes, such as H239_3063, encoding an ABC transporter transmembrane region, H239_3064, encoding a putative efflux pump also known as KexD, and H239_3065, encoding an N-acetyltransferase. To elucidate the mechanism of resistance to NOSO-502 induced by CrrB missense mutations in K. pneumoniae, mutants of NCTC 13442 and ATCC BAA-2146 strains resistant to NOSO-502 and CST with single amino acid substitutions in CrrB (S8N, F33Y, Y34N, W140R, N141I, P151A, P151L, P151S, P151T, or F303Y) were selected. Full susceptibility to NOSO-502 was restored in crrA- or crrB-deleted K. pneumoniae NCTC 13442 CrrB (P151L) mutants, confirming the role of CrrAB in controlling this resistance pathway. Deletion of kexD (but not other neighboring genes) in the same mutant also restored NOSO-502-susceptibility. Upregulation of the kexD gene expression was observed for all CrrB mutants. Finally, plasmid expression of kexD in a K. pneumoniae strain missing the locus crrABC and kexD significantly increased resistance to NOSO-502.

This randomized controlled trial extended the investigation previously conducted by Jarero et al. (2015) which found that the eye movement desensitization and reprocessing Integrative Group Treatment Protocol adapted for ongoing traumatic stress (EMDR-IGTP-OTS) was effective in reducing posttraumatic stress disorder (PTSD) symptoms related to the diagnosis and treatment of different types of cancer in adult women. The current study sought to determine if the results could be replicated and if the treatment would also be effective in reducing symptoms of anxiety and depression. Participants in treatment (N = 35) and no-treatment control (N = 30) groups completed pre, post, and follow-up measurements using the Posttraumatic Stress Disorder Checklist for the Diagnostic and Statistical Manual of Mental Disorders (5th ed.; DSM-5) (PCL-5) and the Hospital Anxiety and Depression Scale (HADS). Data analysis by repeated measures analysis of variance (ANOVA) showed that the EMDR-IGTP-OTS was effective in significantly reducing symptoms of PTSD, anxiety, and depression, with symptoms maintained at 90-day follow-up and with large effect sizes (e.g., d = 1.80). A comparison of the treatment and no-treatment control groups showed significantly greater decreases for the treatment group on symptoms of PTSD, anxiety, and depression. No significant correlation was found when exploring the relationship between scores on the Adverse Life Experiences scale and scores indicating pretreatment severity of PTSD, anxiety, and depression. This study suggests that EMDR-IGTP-OTS may be an efficient and effective way to address cancer-related posttraumatic, depressive, and anxious symptoms.

The aim of this preliminary study was to evaluate the effectiveness of specially trained and supervised paraprofessionals in administering the eye movement desensitization and reprocessing Integrative Group Treatment Protocol (EMDR-IGTP) to reduce work-related posttraumatic stress disorder (PTSD) symptoms. The 2 paraprofessionals in this study were specially selected and trained in the application of the EMDR-IGTP and then provided treatment in an uncontrolled clinical trial to 37 clients from 3 non-governmental organizations in Bolivia. The participants were adult staff members (protective services workers, caregivers, psychologist, lawyers, and social workers) who provided care to children and adolescents with severe interpersonal trauma. Four EMDR-IGTP sessions within a parallel 2-week period were administered for each randomly assigned group. The Posttraumatic Stress Disorder Checklist for DSM-5 (PCL-5) was administered at pretreatment and 30 and 90 days’ posttreatment. A repeated measures analysis of variance (ANOVA) determined that PCL-5 score means differed statistically significantly between time points F(2, 72) = 574.53, p < .001, ηP2 = .94. The study presents preliminary evidence scaling up EMDR therapy in a low- and middle-income country, making it possible to reach larger numbers of people in a shorter time, thereby offering an operational advantage. The study has limitations specially related to the size of the sample, the use of only one measure, and the lack of comparison with a control group or treatment. Further studies are required to present large samples with more measures and comparison of results with another therapy or control group.

Purpose The purpose of this paper is to present a sustainable, community-based action model to improve the care and protection of children between the ages of zero and eight years who have been left behind due to migration. One facet of the program is to train community leaders to teach an experiential program to facilitate and increase the likelihood of the development of life habits in children, caretakers, teachers, mothers, and fathers that will improve the caretaker-infant relationship. Design/methodology/approach Community leaders were evaluated via a pre-training questionnaire and a post-training questionnaire. Findings The evaluation of the training of the community leaders revealed the following statistically significant results: an increase in the level of a number of life habits, which include self-awareness, management of emotions, empathy, assertive communication, decision-making, critical thinking, conflict negotiation and resolution, and teamwork; a reduction in the degree to which psychosocial barriers hinder personal development; an increase in the level of autonomy and self-efficacy; an increase in the level of knowledge that participants have about the topics of the workshops: child development, child-rearing and academic potential, as well as migration; and positive changes in the attitudes that they have regarding certain topics related to child development. Originality/value The program was conducted in Hidalgo, Mexico, a state whose high rates of migration put the children at greater risk for developmental delays if they lack adult support.

ResumenObjetivosEstudiar los cambios endoteliales y el grosor central de la córnea (CCE) que presentan los pacientes operados de catarata con el método convencional de facoemulsificación y el asistido con láser de femtosegundos.MétodoEstudio prospectivo, comparativo, experimental y longitudinal de 65 pacientes con catarata. Se realizó microscopia especular y paquimetría ultrasónica prequirúrgica al primer día, a la semana y al mes postoperatorio. Se incluyeron pacientes con catarata candidatos a cirugía por facoemulsificación, mayores a 45 años, sin enfermedad corneal y con buena dilatación pupilar.ResultadosSe obtuvo una muestra de 65 pacientes, 35 del grupo de femtosegundos y 30 en el grupo de facoemulsificación. Todas las características demográficas fueron similares en ambos grupos, al igual que la dureza de las cataratas, CCE y paquimetría central preoperatoria.El tiempo efectivo de facoemulsificación fue de 4.17 en el grupo de femtosegundos, en comparación con 8.21 en el grupo de facoemulsificación, con una diferencia estadísticamente significativa. En el conteo de células endoteliales y la paquimetría ultrasónica no se encontró diferencia estadísiticamente significativa entre los 2 grupos.ConclusionesAunque se observó una diferencia estadísticamente significativa en cuanto al tiempo efectivo de faco, siendo menor en el grupo de femtosegundos, no se encontraron diferencias estadísticamente significativas en el CCE y grosor corneal central entre el grupo de facoemulsificación convencional y el asistido por láser de femtosegundos.AbstractObjectivesTo compare the endothelial cell count and central corneal volume between the group of conventional phacoemulsification and the femtosecond laser assisted surgery.MethodProspective, comparative, experimental and randomized study of 64 cataract patients who had surgery between May and August of 2013. Patients where randomly choose to have either femtosecond laser assisted surgery (Femto group) or the conventional phacoemulsification (Phaco group). The endothelial cell count was determined by specular microscopy preoperatively, 1 day, 1 week, and 1 month postoperatively. The same process was applied to assess the central corneal volume by measuring it with ultrasonic paquimetry. We included patients with similar cataract density, older than 40 years old and with no corneal pathology.ResultsThe mean age was 66.68±11.74 years in the femto group and 72.2±8.82 on the phaco group. In the multivariable modeling, there were no significant differences in the mean central corneal volume (P=.44) and the endothelial cell count (P=.45). There was not statistical significance difference between the central corneal volume and the endothelial cell count of the conventional phaco and the femto group.ConclusionsThe femtosecond laser-assisted surgery has the same impact in corneal swelling and endothelial trauma that the conventional phaco surgery.

Lipid A is a potent endotoxin, and its fatty acids (lauric, myristic, and sometimes palmitic acid) anchors lipopolysaccharide (LPS) into the outer leaflet of the outer membrane of most Gram-negative bacteria. The highly anionic charge of the glucosamine lipid A moiety makes the LPS a powerful attractant for cationic antimicrobial peptides (AMPs). AMPs are major component of innate immunity that kill bacteria by permeabilization of lipid bilayers. Secondary lipid A acylation of Klebsiella pneumoniae, involving the acyltransferase LpxM (formally, msbB or WaaN) that acylates (KDO)2-(lauroyl)-lipid IV-A with myristate during lipid A biosynthesis, has been associated with bacterial resistance to AMPs contributing to virulence in animal models. We investigated here the role of the msbB gene of the entomopathogenic bacterium Photorhabdus luminescens in AMP resistance, by functional complementation of the AMP susceptible K. pneumoniae lpxM mutant with the P. luminescens msbB gene. We showed that msbB (lpxM) gene of P. luminescens is able to enhance polymyxin B, colistin and cecropin A resistance of K. pneumoniae lpxM mutant, compared to the non-complemented mutant. However, we could not obtain any msbB mutant of Photorhabdus by performing allelic exchange experiments based on positive selection of sucrose highly resistant mutants. We thus suggest that msbB-mediated Photorhabdus lipid A acylation is essential for outer membrane low-permeability and that modification of lipid A composition, fluidity and osmosis-resistance have an important role in the ability of Photorhabdus to grow in sucrose at high concentrations.

L'objectif de cette recherche est d'évaluer l'efficacité du protocole de traitement intégratif de groupe par la désensibilisation et le retraitement par les mouvements oculaires (PTIG-EMDR) dans la réduction des symptômes d'état de stress post-traumatique (ESPT) liés au diagnostic et au traitement de différents types de cancer chez des femmes adultes. La thérapie intensive PTIG-EMDR a été administrée pendant trois jours consécutifs, deux fois par jour, à 24 femmes adultes diagnostiquées avec différents types de cancer (cancer du col de l'utérus, du sein, du côlon, de la vessie et de la peau) et présentant des symptômes d'ESPT liés à leur diagnostic et à leur traitement. Les données ont été analysées à l'aide d'une ANOVA factorielle sur les effets du PTIG-EMDR, évalués avec le Short PTSD Rating Interview (entretien court d'évaluation de l'ESPT) comme variable dépendante, et le groupe (deux groupes de patientes : phase active et phase de suivi du traitement du cancer) et le temps (quatre intervalles de temps) comme variables indépendantes. Des analyses post hoc ont été effectuées. Les résultats ont montré des effets principaux significatifs pour le temps et le groupe. Aucune interaction importante n'a été observée. Les résultats ont également montré une amélioration subjective globale chez les participantes. Cette étude pilote suggère qu'une administration intensive du PTIG-EMDR peut être un apport valable pour des patients atteints du cancer et présentant des symptômes d'ESPT liés au diagnostic et au traitement. D'autres recherches comportant des études contrôlées randomisées seront nécessaires pour démontrer l'efficacité du PTIG-EMDR sur cette population.

Childhood obesity is a critical and growing public health crisis across the world, with implications ranging from the development of noncommunicable diseases such as diabetes, which has been especially revelant in recent months due to the associated risk with COVID-19 recovery, to psychosocial consequences such as low self-esteem. Habits pertaining to diet and exercise may affect a child’s risk of becoming obese, with consumption of processed foods or a lack of physical exercise as risk factors. The objective of this study was to implement a school-based progam in two cities in the State of Chihuahua, Mexico, with the goal of preventing the development of childhood obesity through building life skills, gaining knowledge, and reducing psychosocial barriers to adopting a healthier lifestyle. Teachers were trained in workshops which prepared them to facilitate life skills, reduce psychosocial barriers and disseminate information through interactive activities with their fourth and fifth grade students, as well as engage in conferences with parents to review similar content regarding healthy practices. Results showed a statistically significant increase in the teacher’s role as a health promoter in the classroom, compared with a no treatment group (F1,74=4,302, p=0.042, η2=0.57), an increase in knowledge about diabetes and health care, as well as increased capability in taking care of their own diet (F1,74=6.103, p=0.006, η2=0.937). Students’ results in the treatment group showed a significant effect in knowledge about the healthy portion size of various types of food including the importance of daily consumption of vegetables (F1,1596=4.033, p=0.045), a significant decrease in consumption of junk food and sodas (F1, 1593=7.074, p=0.008), and a significant decrease in drinking soda (F1,1593=6.618, p=0.010), compared with a no treatment group. Parents increased their knowledge of maintaining healthy eating habits and the importance of exercise as well as their self confidence to promote healthy habits in the family. These results demonstrate the success of the implementation of this comprehensive model based on life skills and intrinsic empowerment in encouraging behaviors as well as emotional and physical habits that reduce risk of childhood obesity, although long-term tracking of indicators such as body mass index (BMI) are needed to determine the efficacy of the program over an extended period of time.

This longitudinal multicenter randomized controlled trial has three objectives 1) to evaluate the EMDR-PRECI in the remission of the PTSD Diagnosis on female minors who were victims of sexual and/or physical violence compared to treatment as usual (TAU), 2) to evaluate the effectiveness of the EMDR-PRECI in the reduction of the PTSD, anxiety, and depression symptoms of the participants compared with TAU, and 3) to expand the knowledge on the existing correlation between the PCL-5 core criteria for PTSD provisional diagnosis and the CAPS-5 PTSD clinical diagnosis on this population. The study was conducted in 2019 in the cities of Puebla, Mexico, and Mexico City, in three sites of a non-profit organization who provide services to women in vulnerable situations. A total of 32 minors met the inclusion criteria. Participants’ ages ranged from 12 to 17 years old (M =15.35 years old). Participation was voluntary with the participant’s and their legal guardian's consent. It is relevant to mention that 31 of the 32 participants were pregnant during treatment with an average of 24.63 weeks (6.15 months) of pregnancy. All patients in the EMDR treatment group did not meet PTSD diagnostic criteria after treatment and at 90-days-follow-up assessments, while all patients in the TAU group maintained their PTSD baseline clinical diagnosis after treatment and at 90-days-follow-up assessments. Le protocole EMDR (désensibilisation et retraitement par les mouvements oculaires) individuel pour une utilisation paraprofessionnelle dans les situations de traumatisme aigu (EMDR-PROPARA) fait partie d'un projet développé à l'initiative du Dr Francine Shapiro. Cet essai clinique randomisé a examiné l'efficacité de ce protocole, administré par des thérapeutes EMDR expérimentés. Trente-neuf premiers intervenants en service actif ayant subi des traumatismes ont été répartis de manière aléatoire en deux groupes et ont reçu des séances de quatre-vingt-dix minutes, soit d'EMDR-PROPARA, soit de thérapie de soutien. Les participants du groupe EMDR-PROPARA ont montré une amélioration immédiatement après le traitement, et leurs scores au Short PTSD Rating Interview (SPRINT), lors d'un suivi à trois mois, avaient encore baissé. Comparativement, les participants du groupe ayant bénéficié de la thérapie de soutien présentaient une amélioration non significative après le traitement et une augmentation des scores au SPRINT lors du second suivi. La différence significative entre les deux traitements donne un support préliminaire à l'hypothèse de l'efficacité de l'EMDR-PROPARA pour réduire la gravité des symptômes post-traumatiques et favoriser une amélioration subjective globale. Les auteurs recommandent que des études mieux contrôlées soient menées afin d'évaluer plus précisément l'efficacité de cette intervention.

This research evaluated the effectiveness of the Eye Movement Desensitization and Reprocessing (EMDR) Protocol for Recent Critical Incidents (EMDR-PRECI) in reducing posttraumatic stress symptoms related to the explosion in an explosives manufacturing factory north of Mexico City that killed 7 employees. The EMDR-PRECI was administered on 2 consecutive days to 25 survivors who had posttraumatic stress symptoms related to the critical incident. Participants’ mean score on the Short PTSD Rating Interview (SPRINT) was 22, well above the clinical cutoff of 14. They were randomly assigned to immediate and waitlist/delayed treatment conditions and therapy was provided within 15 days of the explosion. Results showed significant main effects for the condition factor, F(1, 80) = 67.04, p < .000. SPRINT scores were significantly different across time showing the effects of the EMDR therapy through time, F(3, 80) = 150.69, p < .000. There was also a significant interaction effect, condition by time, F(2, 80) = 55.45, p < .001. There were significant differences between the two treatment conditions at Time 2 (post-immediate treatment vs. post-waitlist/delayed), t(11) = −10.08, p < .000. Treatment effects were maintained at 90-day follow-up. Results also showed an overall subjective improvement in the participants. This randomized controlled trial provides evidence for the efficacy of EMDR-PRECI in reducing posttraumatic stress symptoms after a technological disaster.

The purpose of this research is to evaluate the effectiveness of the eye movement desensitization and reprocessing (EMDR) Integrative Group Treatment Protocol (EMDR-IGTP) in reducing posttraumatic stress disorder (PTSD) symptoms related to the diagnosis and treatment of different types of cancer in adult women. EMDR-IGTP intensive therapy was administered for 3 consecutive days, twice daily, to 24 adult women diagnosed with different types of cancer (cervical, breast, colon, bladder, and skin) who had PTSD symptoms related to their diagnosis and treatment. The data was analyzed using factorial ANOVA with the effects of the EMDR-IGTP evaluated with the Short PTSD Rating Interview as dependent variable and group (two groups of patients: active phase and follow-up phase of cancer treatment) and time (four time points) as independent variables. Post hoc analyses were carried out. Results showed significant main effects for time and group. No significant interaction was found. Results also showed an overall subjective improvement in the participants. This pilot study suggests that intensive administration of the EMDR-IGTP can be a valuable support for cancer patients with PTSD symptoms related to their diagnosis and treatment. Further research with randomized controlled studies is needed to demonstrate the effectiveness of EMDR-IGTP in this population.

There is a complex interplay between the regulation of flagellar motility and the expression of virulence factors in many bacterial pathogens. Here, we review the literature on the direct and indirect roles of flagellar motility in mediating the tripartite interaction between entomopathogenic bacteria (Photorhabdus and Xenorhabdus), their nematode hosts, and their insect targets. First, we describe the swimming and swarming motility of insect pathogenic bacteria and its impact on insect colonization. Then, we describe the coupling between the expression of flagellar and virulence genes and the dynamic of expression of the flagellar regulon during invertebrate infection. We show that the flagellar type 3 secretion system (T3SS) is also an export apparatus for virulence proteins in X. nematophila. Finally, we demonstrate that phenotypic variation, a common property of the bacterial symbionts of nematodes, also alters flagellar motility in Photorhabdus and Xenorhabdus. Finally, the so-called phenotypic heterogeneity phenomenon in the flagellar gene expression network will be also discussed. As the main molecular studies were performed in X. nematophila, future perspectives for the study of the interplay between flagellum and invertebrate interactions in Photorhabdus will be discussed.

Determine the frequency of granulomatosis with polyangiitis (GPA) associated with non-identified ANCA (non-MPO, non-PR3 ANCA) and secondarily compare their clinic with GPA associated with MPO-positive or PR3-positive ANCA.In a monocentric retrospective observational study, clinical data of 398 patients with non-identified ANCA (titer of ANCA at least 1/80 by immunofluorescence on ethanol fixed PMN) was gathered over a period of 6 years. GPA patients from this population were compared with GPA patients with identified ANCA on the basis of clinical, biological, immunological and histological features.The most common diseases associated with non-identified ANCA were inflammatory bowel diseases accounting for 17% of diseases. GPA accounted for only 1.8% of cases. There were no significant differences in terms of clinical and histological characteristics between GPA with non-identified ANCA and GPA with identified ANCA, but significantly higher CRP levels were observed in GPA patients with identified ANCA (p = 0.005). Localized disease (ear, nose and throat and/or lung involvement without any other systemic involvement) was more frequent in the group of GPA with nonidentified ANCA (p = 0.047) as compared to GPA with identified ANCA. This explains that the former group of patients was less frequently treated by cyclophosphamide than the latter (p = 0.016).GPA with non-MPO, non-PR3 ANCAs is relatively rare. Our study suggests that GPA with nonidentified ANCA differs from GPA with identified ANCA by the frequency of localized forms.

Our chapter looks at the situation of women in Mexico through the eyes of the expansion of their human capabilities, so that they may reach their potential. We start by looking at the current state of affairs of women in Mexico. We explain what the pressure to conform, family expectations and values, and gender roles and expectations entail and how they affect different areas of women’s lives. We then go on to explore women’s sexuality, health, legal issues and their role in politics. In the next section of the chapter, we present the Framework for Enabling Empowerment (FrEE) and some of its applications through I want to I can programs, as a base for successful program development for women in Mexico. Our conclusions point to the fact that the situation of women in Mexico has improved in the last 100 years and we summarize the need to further expand their freedoms and capabilities.

El Protocolo Individual de Terapia de Reprocesamiento y Desensibilización a través del Movimiento Ocular para uso de Paraprofesionales en situaciones de trauma agudo (EMDR-PROPARA), es parte de un proyecto desarrollado bajo la iniciativa de la Dra. Francine Shapiro. Este estudio aleatorio controlado investigó la efectividad del protocolo administrado por Terapeutas EMDR con experiencia. En el estudio participaron 39 auxiliadores traumatizados y en servicio activo, que fueron asignados al azar para recibir una sesión de 90 minutos, ya fuera del EMDR-PROPARA o de Consejería de Apoyo. Los participantes tratados con el EMDR-PROPARA mostraron beneficios inmediatos después del tratamiento; con un decremento de los puntajes de la Short PTSD Rating Interview (SPRINT), que continuó observándose hasta el segundo seguimiento a los tres meses. En comparación, los participantes que fueron tratados con Consejería de Apoyo, tuvieron un decremento no significativo después del tratamiento y un aumento en los puntajes del SPRINT en el segundo seguimiento. Las diferencias significativas entre los dos tratamientos proporcionan evidencia preliminar, que apoya la eficacia del EMDR-PROPARA para reducir la severidad de los síntomas postraumáticos y en la mejoría subjetiva global. Se recomienda realizar mayor investigación controlada para evaluar de manera más extensa la eficacia de esta intervención.

Bacteria of the genus Xenorhabdus are symbionts of soil entomopathogenic nematodes of the genus Steinernema. This symbiotic association constitutes an insecticidal complex active against a wide range of insect pests. Unlike other Xenorhabdus species, Xenorhabdus poinarii is avirulent when injected into insects in the absence of its nematode host. We sequenced the genome of the X. poinarii strain G6 and the closely related but virulent Xenorhabdus doucetiae strain FRM16. G6 had a smaller genome (500-700 kb smaller) than virulent Xenorhabdus strains and lacked genes encoding potential virulence factors (haemolysins, type 5 secretion systems, enzymes involved in the synthesis of secondary metabolites, toxin-antitoxin systems). The genomes of all the X. poinarii strains analysed here had a similar small size. We did not observe the accumulation of pseudogenes, insertion sequences or decrease in coding density usually seen as a sign of genomic erosion driven by genetic drift in host-adapted bacteria. Instead, genome reduction of X. poinarii seems to have been mediated by the excision of genomic blocks from the flexible genome, as reported for the genomes of attenuated free pathogenic bacteria and some facultative mutualistic bacteria growing exclusively within hosts. This evolutionary pathway probably reflects the adaptation of X. poinarii to specific host.

The aim of this longitudinal multisite randomized controlled trial (RCT) was to evaluate the effectiveness of the EMDR-Integrative Group Treatment Protocol for Ongoing Traumatic Stress (EMDR-IGTP-OTS) in reducing severe posttraumatic stress disorder, depression, and anxiety symptoms in refugee minors living in Valencia, Spain. The study was conducted in 2019 in the city of Valencia, Spain, in six sites for refugees (homes or residences). A total of 184 minors (all males) met the inclusion criteria. Participants’ ages ranged from 13 to 17 years old (M =16.36 years). Participants’ time since the arrival to the sites in Valencia varied from January 2018 to March 2019. Participation was voluntary with the participants and their legal guardian's consent. In this study, intensive EMDR therapy was provided. This intensive format allowed the participants to complete the full course of treatment in a short period. Treatment was provided simultaneously by eighteen licensed EMDR clinicians formally trained in the protocol administration divided into six teams. Each of the treatment group participants received an average of eight hours of treatment, provided during nine group-treatment sessions, three times daily during three consecutive days in a setting inside the six refugee sites. EMDR-IGTP-OTS treatment focused only on the distressing memories related to their life as refugees and did not address any other memories. No adverse effects were reported during treatment or at three-month post-treatment assessment. Data analysis by repeated measures ANOVA showed that the EMDR-IGTP-OTS was effective in reducing PTSD symptoms in the treatment group (F (2, 122) = 43.17p

Spodoptera littoralis, one of the major pests of many important crop plants, is more susceptible to Bacillus thuringiensis aizawai delta-endotoxins than to those of Bacillus thuringiensis kurstaki. Within the framework of the development of efficient bioinsecticides and the prevention against insect resistance, we tested the effect of mixing B. thuringiensis kurstaki delta-endotoxins and Photorhabdus luminescens cells on S. littoralis growth. The obtained results showed that the growth inhibition of this insect was more effective when B. thuringiensis kurstaki spore-crystal mixture and Photorhabdus luminescens cells were used in combination. Furthermore, this synergism is mainly due to the presence of Cry1Ac, which is one of the three delta-endotoxins that form the crystal of B. thuringiensis kurstaki strain BNS3 in addition to Cry1Aa and Cry2Aa. This work shows a possibility to use B. thuringiensis as a delivery means for Photorhabdus bacteria in order to infect the insect hemocoel and to reduce the risk of developing resistance in the target organism.

The eye movement desensitization and reprocessing (EMDR) individual protocol for paraprofessional use in acute trauma situations (EMDR-PROPARA) is part of a project developed at the initiative of Dr. Francine Shapiro. This randomized clinical trial examined the effectiveness of the protocol administered by experienced EMDR therapists. There were 39 traumatized first responders on active duty randomly assigned to receive two 90-min sessions of either EMDR-PROPARA or of supportive counseling. Participants in the EMDR-PROPARA group showed benefits immediately after treatment, with their scores on the Short PTSD Rating Interview (SPRINT) showing further decreases at 3-month follow-up. In comparison, supportive counseling participants experienced a nonsignificant decrease after treatment and an increase in the SPRINT scores at the second follow-up. The significant difference between the two treatments provides preliminary support for EMDR-PROPARA’s effectiveness in reducing severity of posttraumatic symptoms and subjective global improvement. More controlled research is recommended to evaluate further the efficacy of this intervention.

Bacteria of the genus Xenorhabdus are symbionts of soil entomopathogenic nematodes of the genus Steinernema. This symbiotic association constitutes an insecticidal complex active against a wide range of insect pests. Within Xenorhabdus bovienii species, the X. bovienii CS03 strain (Xb CS03) is nonvirulent when directly injected into lepidopteran insects, and displays a low virulence when associated with its Steinernema symbiont. The genome of Xb CS03 was sequenced and compared with the genome of a virulent strain, X. bovienii SS-2004 (Xb SS-2004). The genome size and content widely differed between the two strains. Indeed, Xb CS03 had a large genome containing several specific loci involved in the inhibition of competitors, including a few NRPS-PKS loci (nonribosomal peptide synthetases and polyketide synthases) producing antimicrobial molecules. Consistently, Xb CS03 had a greater antimicrobial activity than Xb SS-2004. The Xb CS03 strain contained more pseudogenes than Xb SS-2004. Decay of genes involved in the host invasion and exploitation (toxins, invasins, or extracellular enzymes) was particularly important in Xb CS03. This may provide an explanation for the nonvirulence of the strain when injected into an insect host. We suggest that Xb CS03 and Xb SS-2004 followed divergent evolutionary scenarios to cope with their peculiar life cycle. The fitness strategy of Xb CS03 would involve competitor inhibition, whereas Xb SS-2004 would quickly and efficiently kill the insect host. Hence, Xenorhabdus strains would have widely divergent host exploitation strategies, which impact their genome structure.

Resume L’hypothese neurodeveloppementale est aujourd’hui l’un des modeles phares pour expliquer la schizophrenie. Selon ce modele, la schizophrenie est la consequence d’anomalies cerebrales precoces engendrees par l’interaction de facteurs genetiques et de facteurs environnementaux. Les donnees en faveur de ce modele sont nombreuses. Plusieurs etudes retrouvent des anomalies perinatales beaucoup plus nombreuses que chez les sujets controles. Sur le plan clinique, les sujets schizophrenes presentent de multiples deficits cognitifs bien avant l’eclosion de la maladie. Ces anomalies parfois subtiles sont selon le modele neurodeveloppemental l’expression des anomalies precoces du systeme nerveux central. Ces donnees sont concordantes avec les donnees en imagerie cerebrale qui montrent des anomalies precoces mais egalement des changements progressifs du systeme nerveux central. Enfin, les donnees genetiques et histologiques confirment que les genes impliques dans la schizophrenie sont les genes codant les differentes etapes du developpement du systeme nerveux central.

Xenorhabdus nematophila engages in complex interactions with invertebrates, through its symbiosis with soil nematodes and its pathogenicity to a broad range of insect larvae. Among the regulatory proteins of Xenorhabdus involved in host interactions, the sigma factor FliA and the regulator FliZ, expressed from the fliAZ operon, play a key role in mediating the production of exoenzymes, motility and full virulence in insects (Lanois et al., 2008). In this study, we investigated the dynamics of the FliA-dependent flagellin gene fliC and FliZ-dependent haemolysin genes xaxAB during insect infection and nematode association by carrying out real-time expression analysis using an unstable GFP monitoring system. We showed that expression of the FliAZ-dependent genes in infected insects is not restricted to a specific tissue but increases significantly just prior to host death and reaches a maximal level in larvae cadaver. Using an iron availability reporter construct, we also showed that iron starvation conditions inhibit expression of FliAZ-dependent genes in vitro, as well as during the first steps of the infectious process. These findings shed further light on the role of the FliAZ regulon in the Xenorhabdus life cycle and suggest that iron may constitute a signal governing Xenorhabdus adaptation to shifting host environments.
