1. Nitrogen uptake and macronutrients distribution in mango trees (cv. Keitt) under three N fertigation treatments

Abstract

We assessed the effects of N concentration in the irrigation water on nutrient uptake and distribution in leaves and fruit of mango cv. Keitt grown in a lysimeter for four years. We applied three treatments: N1 – no N fertilization (less than 2 mg/L in the tap water); N2 – 10 mg/L N; N3 – 20 mg/L N.Deficient N conditions (N1) generated low vegetative yield, high fruit:leaf ratio, high photosynthetic activity, high leaf P and K concentrations, and high sugar content along with low acidity in the fruit. Excess N concentration (N3) induced vegetative growth, and reduced fruit yield and gas-exchange characteristics. The calculated annual nitrogen uptake heavily depended on the nitrogen supply (N1-26 g/tree; N2-196 g/tree; N3- 185 g/tree).Fruits were the major N sink being 0.82, 0.26 and 0.05 from the total annual N supplied. The N quantities accumulated in N1 fruits during the reproductive season (May-August), were above the N quantities supplied via fertigation, suggesting that N reserve in the vegetative tissues supplied the fruit’s high N demand.The finding shows the importance of adequate nitrogen supply to mango trees and the dangers of excessive fertilization.

Abstract

Using the Cap Analysis of Gene Expression (CAGE) technology, the FANTOM5 consortium provided one of the most comprehensive maps of transcription start sites (TSSs) in several species. Strikingly, ~72% of them could not be assigned to a specific gene and initiate at unconventional regions, outside promoters or enhancers. Here, we probe these unassigned TSSs and show that, in all species studied, a significant fraction of CAGE peaks initiate at microsatellites, also called short tandem repeats (STRs). To confirm this transcription, we develop Cap Trap RNA-seq, a technology which combines cap trapping and long read MinION sequencing. We train sequence-based deep learning models able to predict CAGE signal at STRs with high accuracy. These models unveil the importance of STR surrounding sequences not only to distinguish STR classes, but also to predict the level of transcription initiation. Importantly, genetic variants linked to human diseases are preferentially found at STRs with high transcription initiation level, supporting the biological and clinical relevance of transcription initiation at STRs. Abstract

The current global consumer aerosol products such as deodorants, hairsprays, air-fresheners, polish, insecticide, disinfectant\ud are primarily utilised unfriendly environmental propellant of liquefied petroleum gas (LPG) for over three decades.\ud The advantages of the new innovative technology described in this paper are: (i) no butane or other liquefied\ud hydrocarbon gas; (ii) compressed air, nitrogen or other safe gas propellant; (iii) customer acceptable spray quality and\ud consistency during can lifetime; (iv) conventional cans and filling technology. Volatile organic compounds and greenhouse\ud gases must be avoided but there are no flashing propellants replacements that would provide the good atomisation and\ud spray reach. On the basis of the energy source for atomising, the only feasible source is inert gas (i.e. compressed air),\ud which improves atomisation by gas bubbles and turbulence inside the atomiser insert of the actuator. This research\ud concentrates on using ‘bubbly flow’ in the valve stem, with injection of compressed gas into the passing flow, thus also\ud generating turbulence. Using a vapour phase tap in conventional aerosol valves allows the propellant gas into the liquid\ud flow upstream of the valve. However, forcing bubbly flow through a valve is not ideal. The novel valves designed here,\ud using compressed gas, thus achieved the following objectives when the correct combination of gas and liquid inlets to the\ud valve, and the type and size of atomiser ‘insert’ were derived:\ud 1. Produced a consistent flow rate and drop size of spray throughout the life of the can, compatible with the current\ud conventional aerosols that use LPG: a new ‘constancy’ parameter is defined and used to this end.\ud 2. Obtained a discharge flow rate suited to the product to be sprayed; typically between 0.4 g/s and 2.5 g/s.\ud 3. Attained the spray droplets size suited to the product to be sprayed; typically between 40 mm and 120 mm.

4. Nucleosynthetic molybdenum isotope anomalies in iron meteorites – new evidence for thermal processing of solar nebula material

Abstract

We have investigated nucleosynthetic Mo isotope anomalies in 38 different bulk iron meteorites from 11 groups, to produce by far the largest and most precise dataset available to date for such samples. All magmatic iron groups were found to display deficits in s-process Mo isotopes, with essentially constant anomalies within but significant variations between groups. Only meteorites of the non-magmatic IAB/IIICD complex revealed terrestrial Mo isotopic compositions. The improved analytical precision achieved in this study enables two isotopically distinct suites of iron meteorites to be identified. Of these, the r=p suite encompasses the IC, IIAB, IIE, IIIAB, IIIE and IVA groups and exhibits relatively modest but ‘pure’ s-process deficits, relative to Earth. The second r>p suite includes groups IIC, IIIF and IVB. These iron meteorites show larger s-process deficits than the r=p suite, coupled with an excess of r-process relative to p-process components. Comparison of the results with data for other elements (e.g., Cr, Ni, Ru, Ti, Zr) suggests that the Mo isotope variability is most likely produced by thermal processing and selective destruction of unstable presolar phases. An updated model is proposed, which relates the iron meteorite suites to different extents of thermal processing in the solar nebula, as governed by heliocentric distance. In detail, the r=p suite of iron meteorite parent bodies is inferred to have formed closer to the Sun, where the extent of thermal processing was similar to that experienced by terrestrial material, so that the meteorites exhibit only small s-process deficits relative to Earth. In contrast, the r>p suite formed at greater heliocentric distance, where more subtle thermal processing removed a smaller proportion of r- and p-process host phases, thereby generating larger s-process deficits relative to the terrestrial composition. In addition, the thermal conditions enabled selective destruction of p- versus r-isotope carrier phases, to produce the observed divergence of r- and p-process Mo isotope abundances.

Queried with a protein sequence it returns: multiple sequence alignments, predicted aspects of structure (secondary structure, solvent accessibility, transmembrane helices (TMSEG) and strands, coiled-coil regions, disulfide bonds and disordered regions) and function. The service incorporates analysis methods for the identification of functional regions (ConSurf), homology-based inference of Gene Ontology terms (metastudent), comprehensive subcellular localization prediction (LocTree3), protein-protein binding sites (ISIS2), protein-polynucleotide binding sites (SomeNA) and predictions of the effect of point mutations (non-synonymous SNPs) on protein function (SNAP2). Our goal has always been to develop a system optimized to meet the demands of experimentalists not highly experienced in bioinformatics. To this end, the PredictProtein results are presented as both text and a series of intuitive, interactive and visually appealing figures. Abstract

Drug product selection legislation is intended to achieve savings in the cost of prescription drugs without adversely affecting the quality of care by allowing pharmacists the opportunity to dispense less costly generically equivalent drug products in place of the product which had been prescribed by the physician. Various conditions under which the pharmacists are authorized by state laws to exercise theoption to substitute are discussed. The study then identifies and examines the conditions under which savings may and do occur, using the legislation in Michigan as the model. Over 60,000 acutal prescriptions were examined for the three-year period, April 1, 1974 through March 31, 1977--which covers the period of the year immediately before the legislation became effective and the two subsequent years-to determine: 1) the extent to which substitution is possible; 2) potential savings from generic substitution; and 3) very substantial potential cost savings from drug product selection, the actual savings represent only an extremely small proportion of the potential. This principally is due to the fact of the low rate of substitution among eligible prescriptions. Thus, if the gap between actual and potential savings is to be reduced, more attention must be given to affecting the pattern of drug selection among pharmacists.
