Abstract: We report the properties of primary cosmic-ray sulfur (S) in the rigidity range 2.15 GV to 3.0 TV based on 0.38 × 106 sulfur nuclei collected by the Alpha Magnetic Spectrometer experiment (AMS). We observed that above 90 GV the rigidity dependence of the S flux is identical to the rigidity dependence of Ne-Mg-Si fluxes, which is different from the rigidity dependence of the He-C-O-Fe fluxes. We found that, similar to N, Na, and Al cosmic rays, over the entire rigidity range, the traditional primary cosmic rays S, Ne, Mg, and C all have sizeable secondary components, and the S, Ne, and Mg fluxes are well described by the weighted sum of the primary silicon flux and the secondary fluorine flux, and the C flux is well described by the weighted sum of the primary oxygen flux and the secondary boron flux. The primary and secondary contributions of the traditional primary cosmic-ray fluxes of C, Ne, Mg, and S (even Z elements) are distinctly different from the primary and secondary contributions of the N, Na, and Al (odd Z elements) fluxes. The abundance ratio at the source for S/Si is 0.167±0.006, for Ne/Si is 0.833±0.025, for Mg/Si is 0.994±0.029, and for C/O is 0.836±0.025. These values are determined independent of cosmic-ray propagation.

2. Searching for exotic spin-dependent interactions using rotationally modulated source masses and an atomic magnetometer array

Abstract: We describe a proposed experimental search for exotic spin-dependent interactions using rotationally modulated source masses and an atomic magnetometer array. Rather than further improving the magnetometer sensitivity, noise reduction can be another way to reach higher measurement precision. In this work, we propose to use modulating techniques of the source masses to reduce the noise of the experiment. Better precision can be achieved if the fundamental frequency and harmonics of the rotating source masses are used to detect the new interactions. Furthermore, if an array of magnetometers are applied, the statistical precision can be improved, and some common-mode noises can be canceled. Our analysis and simulations indicate that the proposed experiment scheme can improve the detection precisions of three types of spin-dependent interactions by as much as ∼5 orders of magnitude in the force range of ∼cm to ∼10 m.

Abstract: International audience; This paper presents a sensorless control strategy for seven-phase non-sinusoidal permanent magnet synchronous machine (NSPMSM) with high frequency signal injection (HFSI) method in 5th harmonic subspace. Seven-phase nonsinusoidal machine benefits high torque density due to the use of third harmonic components. Firstly, two torque generation strategy in different harmonic subspaces with the minimum injected rms value and amplitude of current is deduced in this paper. Secondly, in order to reinforce the reliabilities of multiphase machine, the HFSI sensorless control is employed. Comparing with traditional HFSI method, the proposed control strategy is achieved in 5th harmonic subspace, which can avoid the torque ripple caused by the injected signal. Finally, according to simulations results the two torque distribution method is validated and compared and the effectiveness of the HFSI method is verified.

5. Blood vessels regulate primary motor neuronal pathfinding in zebrafish via exosome contained microRNA-22

Abstract: A precise neuro-vascular communication is crucial to orchestrate directional migration and patterning of the complex vascular network and neural system. However, how blood vessels are involved in shaping the proper neuronal formation has not been fully understood. So far, limited studies have reported the discovery and functions of microRNAs (miRNAs) in guiding vascular and neural pathfinding. Currently, we showed that the deficiency of miRNA-22a, an endothelial-enriched miRNA, caused dramatic pathfinding defects both in intersegmental vessels (ISVs) and primary motor neurons (PMNs) in zebrafish embryos. Furthermore, we found the specific inhibition of miR-22a in ECs resulted in the patterning defects of both ISVs and PMNs. However, neuronal block of miR-22a mainly led to the axonal defects of PMN. Then we demonstrated that endothelial miR-22a regulates PMNs axonal navigation via exosome pathway. Sema4c was identified as a potential target of miR-22a through transcriptomic analysis and in silico analysis. Furthermore, luciferase assay and EGFP sensor assay in vivo confirmed the binding of miR-22a with 3’-UTR of sema4c. In addition, Down-regulation of sema4c in the miR-22a morphants significantly neutralized the aberrant patterning of vascular and neural networks. Our study revealed that miR-22a acted as a dual guidance cue coordinating vascular and neuronal patterning and expanded the repertoire of guidance molecules, which might be of use therapeutically to guide vessels and nerves in the relevant diseases that affect both systems.

6. Genome-wide evolutionary signatures of climate adaptation in spotted sea bass inhabiting different latitudinal regions

Abstract: Temperature is an important climatic factor that shapes the distribution of eurythermal species. Thermal adaptation of species is important to both evolutionary biology and climate-change biology because it frequently leads to latitudinal gradients of various phenotypes among populations. Spotted sea bass (Lateolabrax maculatus) has a broad latitudinal distribution range along the marginal seas of the Northwest Pacific, providing an excellent teleost model for climate adaptation studies. We generated over 8.57 million SNP loci using whole genome re-sequencing from 100 samples collected at 14 geographic loci. We built the phylogeographic structure and demographic history of L. maculatus and determined sea surface temperature as the key environmental factor and major driving force for genetic divergence and local adaptation. We also identified distinct selective signatures and functional genes underlying adaptive mechanisms and ecological tradeoffs in the southernmost and northernmost populations inhabiting distinct climatic and latitudinal zones. The results offer an opportunity to better understand the genetic basis of the phenotypic variation in eurythermal fishes inhabiting different climatic regions.

Abstract: Sacubitril-Valsartan in Acute Myocardial Infarction In a randomized trial, 5661 patients with acute myocardial infarction and a reduced left ventricular ejection fraction, pulmonary congestion, or both were assigned to receive either sacubitril-valsartan or ramipril. At a median of 22 months, there was no significant difference between the two groups in the incidence of death from cardiovascular causes or incident heart failure. Background In patients with symptomatic heart failure, sacubitril-valsartan has been found to reduce the risk of hospitalization and death from cardiovascular causes more effectively than an angiotensin-converting-enzyme inhibitor. Trials comparing the effects of these drugs in patients with acute myocardial infarction have been lacking. Methods We randomly assigned patients with myocardial infarction complicated by a reduced left ventricular ejection fraction, pulmonary congestion, or both to receive either sacubitril-valsartan (97 mg of sacubitril and 103 mg of valsartan twice daily) or ramipril (5 mg twice daily) in addition to recommended therapy. The primary outcome was death from cardiovascular causes or incident heart failure (outpatient symptomatic heart failure or heart failure leading to hospitalization), whichever occurred first. Results A total of 5661 patients underwent randomization; 2830 were assigned to receive sacubitril-valsartan and 2831 to receive ramipril. Over a median of 22 months, a primary-outcome event occurred in 338 patients (11.9%) in the sacubitril-valsartan group and in 373 patients (13.2%) in the ramipril group (hazard ratio, 0.90; 95% confidence interval [CI], 0.78 to 1.04; P=0.17). Death from cardiovascular causes or hospitalization for heart failure occurred in 308 patients (10.9%) in the sacubitril-valsartan group and in 335 patients (11.8%) in the ramipril group (hazard ratio, 0.91; 95% CI, 0.78 to 1.07); death from cardiovascular causes in 168 (5.9%) and 191 (6.7%), respectively (hazard ratio, 0.87; 95% CI, 0.71 to 1.08); and death from any cause in 213 (7.5%) and 242 (8.5%), respectively (hazard ratio, 0.88; 95% CI, 0.73 to 1.05). Treatment was discontinued because of an adverse event in 357 patients (12.6%) in the sacubitril-valsartan group and 379 patients (13.4%) in the ramipril group. Conclusions Sacubitril-valsartan was not associated with a significantly lower incidence of death from cardiovascular causes or incident heart failure than ramipril among patients with acute myocardial infarction. 10. Exosome-mediated miR-1290 regulation of SLU7 affects hepatocellular carcinoma process

Abstract: Objective: Liver cancer, as a very common malignant tumor in clinic, has a high incidence rate and a poor prognosis. Understanding the relationship of exosomes miR-1290 and SLU7 with liver cancer may be a breakthrough in the diagnosis and treatment of liver cancer in the future.Methods: Altogether 64 patients with liver cancer and 58 healthy people in our hospital were collected, and liver cancer cells were purchased. qPCR was used to detect the mRNA levels of miR-1290 and SLU7. Western blot was used to detect the relative expression of SLU7 protein, and the effect of exosomes miR-1290 and SLU7 on the biological behavior of liver cancer cells was analyzed. Double fluorescein reporter was used to verify the targeted relationship between miR-1290 and SLU7.Results: miR-1290 was highly expressed in liver cancer (P< 0.050) and SLU7 was poorly expressed in liver cancer (P< 0.050). Exosomes miR-1290 could inhibit the relative expression of SLU7 protein (p < 0.050), promote the proliferation, invasion, migration and epithelial-mesenchymal transformation of liver cancer cells, and reduce the ability of apoptosis (P< 0.050). Double luciferase report and rescue experiment confirmed that miR-1290 and SLU7 had targeted relationship (P< 0.050).Conclusion: Exosomes miR-1290 may be a potential target for the diagnosis and treatment of liver cancer in the future through targeting inhibition of SLU7 in the occurrence and development of liver cancer.

11. Efficient Regioselective Photogeneration of a Five-Membered Azaheterocyclic AIEgen for Cancer Cell Differentiation with Unusual Photoamplification Effect

Abstract: Five-membered azaheterocycles, mostly suffered from various synthetic problems, hold a special status in pharmaceutical chemistry and life science. Here, based on green chemistry, a highly efficient and remarkably regioselective photoreaction was developed under mild conditions for constructing fused five-membered azaheterocycles with aggregation-induced emission (AIE) characteristics. After the photoreaction, one of the most noteworthy optical features is the abruptly increased emission induced by UV irradiation. Making use of this, fluorescent photopatterns were readily fabricated in solution and solid support with improved signal-to-background ratio. Due to the excellent biocompatibility and unique chemical structure of the azaheterocycles, selective mitochondria-targeted cancer cell recognition was realized at an ultralow dye concentration. Notably, an unusual photoamplification effect and an exceptional anti-photobleaching phenomenon were observed thanks to the occurrence of the photoreaction. It is not only the first time that fused five-membered azaheterocyclic AIE luminogen was synthesized quantitatively via an unconventionally yet highly regioselective photoreaction, but also the first attempt to combat photobleaching problem by the photoamplification effect.

12. Relationship between renal biopsy and renal survival in elderly patients with chronic kidney disease: A propensity score matching approach

Abstract: Background The effect of renal biopsy on the prognosis of elderly patients with chronic kidney disease remains unclear. Thus, in this study, we aimed to evaluate the relationship between renal biopsy and renal survival in this population.Methods In this multi-centre retrospective study, the baseline characteristics among three groups were balanced by propensity matching. All patients were divided into three groups according to age and renal biopsy. The clinicopathological features at biopsy and renal outcomes during the follow-up were collected and analysed. Renal outcomes were defined as estimated glomerular filtration rate < 15 mL/min/1.73 m2, dialysis, renal transplantation, or death. The prognostic effects of renal biopsy were evaluated using Cox regression models. Results A total of 1313 patients were identified. After propensity matching, 390 patients were selected and divided into three groups. After a total follow-up period of 55 months, 20 (13.3%) patients (47.6% group 1 vs 7.41% group 2 vs 39.1% group 3) reached renal outcomes. No significant differences were found in renal outcomes among aged patients whether they underwent renal biopsy or not. Cox regression analysis revealed risk factors in aged patients including low albumin and high levels of proteinuria and serum creatinine (P < 0.05). Platelet count was significant only in aged patients who underwent renal biopsy (hazard ratio: 0.642, P < 0.05). Conclusion In conclusion, renal biopsy in the elderly has not shown benefits in terms of renal survival, conservative treatment appears to be a viable therapeutic option in the management of those people.

15. Excitation delocalization in porphyrin nanorings

Abstract: This thesis is concerned with the photophysical properties of porphyrin nanorings. In particular, the impact of symmetry, conformation and structure on electronic delocalization, radiative rate and emission polarization memory loss are investigated and exciton migration is probed on a number of porphyrin nanorings. Ultrafast time-resolved spectroscopy techniques are used to study the fluorescence dynamics of these newly synthesized compounds. First, photophysical implications of lifting the rotational symmetry are investigated using a number of small porphyrin rings specially modified for this purpose. For more severe symmetry distortions, increased optical transitions to the lowest excited state have been observed, which is dipole-forbidden due to symmetry reason. Also, the degeneracy in the allowed first excited state is lifted for broken symmetry, leading to a polarization switching effect. The effect of conformation on emission depolarization memory loss is studied using a series of similarly sized porphyrin rings with rigid structures exhibiting various degrees of out-of-plane distortion. Excitations can access any segment on the nanoring and this is not affected by the conformation. However, severe out-of-plane distortions lead to lowered emission anisotropy; this conclusion is further supported by molecular dynamics simulation. Finally, exciton migration within porphyrin ring complex is investigated. Russian doll complex which consists of two concentric porphyrin rings acts as a single emitter, even though spectral features of both ring components are found in the absorption spectrum. Lifetimes and radiative rates of the complex and the individual rings components further confirmed that excitons migrate from the outer 12-ring to the inner 6-ring within 40 ps. A nanotube consisting of two conjugated 6 porphyrin rings linked by dimers acting as staves also exhibits energy migration. Emission anisotropy measurements suggest that excitation is transferred from the staves of the nanotube to the ring plane effectively within 280 fs.

Abstract: Ca$_3$Ti$_2$O$_7$ is an experimentally confirmed hybrid improper ferroelectric material, in which the electric polarization is induced by a combination of the coherent TiO$_6$ octahedral rotation and tilting. In this work, we investigate the tuning of ferroelectricity of Ca$_3$Ti$_2$O$_7$ using iso-valent substitutions on Ca-sites. Due to the size mismatch, larger/smaller alkaline earths prefer $A$'/$A$ sites respectively, allowing the possibility for site-selective substitutions. Without extra carriers, such site-selected iso-valent substitutions can significantly tune the TiO$_6$ octahedral rotation and tilting, and thus change the structure and polarization. Using the first-principles calculations, our study reveals that three substituted cases (Sr, Mg, Sr+Mg) show divergent physical behaviors. In particular, (CaTiO$_3$)$_2$SrO becomes non-polar, which can reasonably explain the suppression of polarization upon Sr substitution observed in experiment. In contrast, the polarization in (MgTiO$_3$)$_2$CaO is almost doubled upon substitutions, while the estimated coercivity for ferroelectric switching does not change. The (MgTiO$_3$)$_2$SrO remains polar but its structural space group changes, with moderate increased polarization and possible different ferroelectric switching paths. Our study reveals the subtle ferroelectricity in the $A_3$Ti$_2$O$_7$ family and suggests one more practical route to tune hybrid improper ferroelectricity, in addition to the strain effect.

For colorectal cancer (CRC), it is unclear whether the differential association of height by sex is real or is due to confounding or bias inherent in observational studies. We performed a Mendelian randomization study to examine the association between height and CRC risk. Methods: To minimize confounding and bias, we derived a weighted genetic risk score predicting height (using 696 genetic variants associated with height) in 10 226 CRC cases and 10 286 controls. Logistic regression was used to estimate odds ratios (OR) and 95% confidence intervals (95% CI) for associations between height, genetically predicted height and CRC. Results: Using conventional methods, increased height (per 10-cm increment) was associated with increased CRC risk (OR = 1.08, 95% CI = 1.02–1.15). In sex-specific analyses, height was associated with CRC risk for women (OR = 1.15, 95% CI = 1.05–1.26), but not men (OR = 0.98, 95% CI = 0.92–1.05). Consistent with these results, carrying greater numbers of (weighted) height-increasing alleles (per 1-unit increase) was associated with higher CRC risk for women and men combined (OR = 1.07, 95% CI = 1.01–1.14) and for women (OR = 1.09, 95% CI = 1.01–1.19). There was weaker evidence of an association for men (OR = 1.05, 95% CI = 0.96–1.15). Conclusion: We provide evidence for a causal association between height and CRC for women. The CRC-height association for men remains unclear and warrants further investigation in other large studies.

Abstract: Repair mortar is commonly used to rehabilitate reinforced concrete structures or components that exhibit a relatively high level of distresses. Yet, this repair mortar can be contaminated by salt from its service environment. This work employs a two-dimensional finite element model to investigate the transport behavior of ionic species in salt-contaminated and water-saturated repair mortar under an externally applied electric field. The model was experimentally validated and then utilized to evaluate the effectiveness of electrochemical chloride extraction (ECE) with or without electrical injection of corrosion inhibitor (EICI). In the case study, both the ECE alone and the ECE+EICI treatment was found effective in decontaminating the zone in front of the steel rebar. In both techniques, the magnitude of current density has a significant effect on removing chloride out of the mortar and increasing the pH of the pore solution near the rebar, whereas the treatment time any not have a significant effect under some scenarios. The injection of the organic corrosion inhibitor significantly slowed down the removal of chloride. Changes in the ionic distribution in the mortar were generally beneficial in reducing the corrosion risk of the steel rebar and thus extending the service life of the repair mortar.

Abstract: A large number of oil and gas reservoirs across the world are naturally fractured, f r om which a significant amount of hydrocarbons are produced. Naturally fractured reservoirs, like all reservoirs, are exploited in t w o stages: primary recovery and secondary recovery (sometimes followed by tertiary recovery, i.e. enhanced oil recovery (EOR)), with different recovery mechanisms. During primary production, the reservoir is produced by fluid expansion. In secondary production and EOR, since the fractures are much more permeable than the matrix, the injected water or EOR agent flows rapidly through the fracture network and surrounds the matrix blocks. Oil recovery then depends on efficient delivery of water or EOR agent to the matrix through the fracture network.

Abstract: We present a measurement of the cosmic ray (e^+ + e^-) flux in the range 0.5 GeV to 1 TeV based on the analysis of 10.6 million (e^+ + e^-) events collected by AMS. The statistics and the resolution of AMS provide a precision measurement of the flux. The flux is smooth and reveals new and distinct information. Above 30.2 GeV, the flux can be described by a single power law with a spectral index γ = −3.170 +/- 0.008 (stat + syst) +/- 0.008 (energy scale)

24. < a > Prismatic, < a > basal, and < c plus a > slip strengths of commercially pure Zr by micro-cantilever tests

Abstract: Slip strengths of 〈a〉 basal, 〈a〉 prism, and 〈c+a〉 pyramidal systems in commercially pure zirconium have been determined using micro-cantilever testing. A range of single crystal cantilevers 0.5 μm to 10 μm wide, oriented for single slip were prepared using focused ion beam (FIB) machining and subsequently deflected using a nanoindenter. The critical resolved shear stress (τcrss) was found by fitting a crystal plasticity finite element model to the experimental load–displacement data for these micro-bending tests. All the three slip systems in α-Zr show a marked size effect in bending described well by τCRSS(W) = τ0 + AWn, where W is the cantilever width, τ0 is the CRSS at the macro scale and n = ∼−1. The exponent, n, of near −1 is in good accord with hardening caused by the back stress generated by dislocations piling up at a diffuse barrier caused by the reduction of stress near the neutral axis. The macro scale CRSS values were used to successfully simulate deformation of a conventional macroscopic compression test.

25. A discrete dislocation plasticity study of the micro-cantilever size effect

Abstract: Micro-cantilevers are increasingly used to extract elastic and plastic material properties through controlled bending using a nanoindenter. Focused Ion Beam milling can be used to produce small scale single crystal cantilevers with cross-sectional dimensions on the order of microns, and electron backscatter diffraction (EBSD) allows cantilevers to be milled from a grain with a desired crystal orientation. Micro-cantilever bending experiments suggest that sufficiently smaller cantilevers are stronger, which is generally believed to be related to the effect of the neutral axis on the evolution of the dislocation structure. A planar model of discrete dislocation plasticity was used to simulate end-loaded cantilevers to interpret the behaviour observed in experiments. The model allowed correlation of the initial dislocation source density and resulting slip band spacing to the experimental load displacement curve. There are similarities between the predictions of this model and those of earlier discrete dislocation plasticity models of pure bending. However, there are notable differences, including a strong source density dependence of the size effect that cannot be explained by geometrically necessary dislocation (GND) arguments, and the effect of the cantilever stress distribution on the locations of soft pile-ups. The planar model was used to identify zero resolved shear stress isolines, rather than the neutral axis, as controlling the soft pile-up location, and source spacing as limiting the slip band spacing in the observed size effect; strengthening was much greater in the source-limited regime. The effect of sample dimensions and dislocation source density were investigated and compared to small scale mechanical tests conducted on titanium and zirconium. The calculations predict a scaling exponent n≈1 for the dependence of stress on size if size is normalised by the average source spacing and a term representing the size-independent flow stress is included, whereas the simple power-law form ordinarily used to fit experimental data significantly underestimates n.

26. Micro-scale fracture experiments on zirconium hydrides and phase boundaries

Abstract: Fracture properties of micro-scale zirconium hydrides and phase boundaries were studied using microcantilever testing methods. FIB-machined microcantilevers were milled on cross-sectional surfaces of hydrided samples, with the most highly-stressed regions within the δ-hydride film, within the α-Zr or along the Zr-hydride interface. Cantilevers were notched using the FIB and then tested in bending using a nanoindenter. Load-displacement results show that three types of cantilevers have distinct deformation properties. Zr cantilevers deformed plastically. Hydride cantilevers fractured after a small amount of plastic flow; the fracture toughness of the δ-hydride was found to be 3.3 ± 0.4 MPam1/2 and SEM examination showed transgranular cleavage on the fracture surfaces. Cantilevers notched at the Zr-hydride interface developed interfacial voids during loading, at loads considerably lower than that which initiate brittle fracture of hydrides.

27. Grain size effects in hcp polycrystals: from GNDs to blocky alpha

Abstract: Typically a ‘smaller is strong’ size effect is seen when testing objects at the micro and nanoscales. This has significant consequences when using these tests to replicate and understand component level performance, for instance in materials discovery programmes. In this computational plasticity study, we follow the micro-cantilever experimental approach of Gong et al. for determination of size-dependent hcp crystal slip strengths and replicate size sensitivity using length scale dependent crystal plasticity modelling. A fundamental derivation of the back stress term required to harden slip systems according to the mechanism of dislocation pile up is introduced. Model micro-beam single crystals in Ti-6Al under four-point bending were examined which showed that the size-independent slip strength could be correctly determined but that the size-strengthening effect was under predicted. This was attributed to the averaging of discrete dislocation behaviour in to the continuum slip rule required within the crystal plasticity formulation

Abstract: A precision measurement by the Alpha Magnetic Spectrometer on the International Space Station of the positron fraction in primary cosmic rays in the energy range from 0.5 to 350 GeV based on 6.8 x 10(6) positron and electron events is presented. The very accurate data show that the positron fraction is steadily increasing from 10 to similar to 250 GeV, but, from 20 to 250 GeV, the slope decreases by an order of magnitude. The positron fraction spectrum shows no fine structure, and the positron to electron ratio shows no observable anisotropy. Together, these features show the existence of new physical phenomena. Abstract: The Marinesco-Sjogren syndrome (MSS) (MIM 248800) is an autosomal recessive condition characterised by cataracts, ataxia, and growth and mental retardation. Chronic myopathy is a common feature. Peripheral neuropathy and acute rhabdomyolysis have been described occasionally in MSS. To date, no gene for it has been localised. Congenital cataracts-facial dysmorphism-neuropathy syndrome (CCFDN) (MIM 604168) is a recently delineated autosomal recessive condition,1 so far only described in a specific gypsy group originating from Bulgaria. This disorder was localised by linkage analysis to 18qter, telomeric to the marker D18S1141.2 CCFDN alleles showed a highly conserved haplotype in the region D18S1141-D18S70-D18S1268 consistent with genetic homogeneity and a single founder mutation. Since then, the disease locus has been reduced to the interval located between markers D18S1095 and D18S1390.3,4 MSS and CCFDN share common clinical features and are considered to be differential diagnoses. Recently, Merlini et al 4 proposed that the CCFDN syndrome and one subtype of MSS (Marinesco-Sjogren/myoglobinuria), also only described in gypsy patients, are genetically identical. We report here the clinical and linkage analysis of one gypsy family and one Turkish family in which patients presented with congenital or juvenile cataracts and ataxia. Both families were initially diagnosed as MSS. However, our study shows that they are clinically and genetically distinct. We found that the gypsy family had CCFDN features and was linked to 18qter whereas the Turkish family had typical MSS features and was not linked to 18qter. Abstract: Humane use of preclinical large animal cancer models plays a critical role in understanding cancer biology and developing therapeutic treatments. Among the large animal candidates, goats have great potentials as sustainable sources for large animal cancer model development. Goats are easier to handle and cheaper to raise. The genome of the goats has been sequenced recently. It has been known that goats develop skin, adrenal cortex, breast and other types of cancers. Technically, goats are subject to somatic cell nuclear transfer more efficiently and exhibit better viability through the cloning process. Towards the development of a goat cancer model, we created a transgenic goat fetal fibroblast (GFF) cell as the donor cell for SCNT. Human mutated K-ras (hK-rasG12D) was chosen as the transgene, as it is present in 20% of cancers. Both hK-rasG12D and a herpes simplex viral thymidine kinase (HSV1-tk) reporter genes, flanked by a pair of LoxP sites, were knocked in the GFF endogenous K-ras locus through homologous recombination. Following Cre-mediated activation (with a 95% activation efficiency), hK-rasG12D and HSV1-tk were expressed in the transgenic GFF cells, evidently through the presence of corresponding mRNAs, and confirmed by HSV1-tk protein function assay. The hK-rasG12D expressing GFF cells exhibited enhanced proliferation rates and an anchorage-independent growth behavior. They were able to initiate tumor growth in athymic nude mice. In conclusion, after activating hK-rasG12D gene expression, hK-rasG12D transgenic GFF cells were transformed into tumorgenesis cells. Transgenic goats via SCNT using the above-motioned cells as the donor cells have been established.

33. The nature of floating cells in human embryonic stem cell culture

Abstract: A consistent presence of floating cells is a common phenomenon in cultures of human embryonic stem cells (hESCs). However, little attention has been paid to their existence. It is currently believed that unavoidable imperfections in culture conditions lead the cells to undergo senescence and apoptosis resulting in unattached cells floating in the culture medium. Inspired by recent studies on mitotic activities in human embryonic stem cell colonies, we believe the existence of floating cells is not simply the result of unfavorable growth conditions but an intrinsic phenomenon resulted from maintaining the pluripotency of hESCs under the culture conditions. We tested this hypothesis with a set of systematic experiments and discovered: 1) the ratio of floating cells to attached cells was significantly increased with culture time; 2) the number of floating cells could be manipulated. For example, we were able to reduce the number of floating cells by providing the colonies with more horizontal or vertical cultural spaces and maintaining the cells’ pluripotency. The results open a new avenue to increase the stem cell culture efficiencies by rescuing the floating cells. On the other hand, by placing a physical barrier on the top of colonies, the number of floating cells was decreased, at the same time, hESCs also showed signs of differentiation. In addition, when inducing cells to differentiate with retinoic acid, the number of floating cells no longer increased with prolonged culture time. Taken together, these results suggested that continuous cell division across the colonies is responsible for the emergence of floating cells during hESC culture. This is quite different from the bacterial colony growth where the cells in the center of colonies are quiescent. Our results indicated that continuous cell division, even at the cost of floating cells formation, is essential for human embryonic stem cell proliferation.

Abstract: Expanding on our previous findings demonstrating that microparticles (MPs) spread cancer multidrug resistance, we now show that MPs sequester drugs, reducing the free drug concentration available to cells. MPs were isolated from drug-sensitive and drug-resistant sub-clones of a human breast adenocarcinoma cell line and from human acute lymphoblastic leukemia cells. MPs were assessed for size, mitochondria, RNA and phospholipid content, P-glycoprotein (P-gp) expression and orientation and ATPase activity relative to drug sequestration capacity. Of the drug classes examined, MPs sequestered the anthracycline class to a significant degree. The degree of sequestration was likely due to the size of MPs and thus the amount of cargo they contain, to which the anthracyclines bind. Moreover, a proportion of the P-gp present on MPs was inside-out in orientation, enabling it to influx drugs rather than its typical efflux function. This was confirmed by surface immunofluorescence and by assessment of drug-stimulated ATPase activity following MP permeabilization. Thus we determined that breast cancer MPs carried a proportion of their P-gp oriented inside-out, providing active sequestration within the microvesicular compartment. These results demonstrate a capacity for MPs to sequester chemotherapeutic drugs, which has a predominantly active sequestration component for MPs derived from drug-resistant cells and a predominantly passive component for MPs derived from drug-sensitive cells. This reduction in available drug concentration has potential to contribute to a parallel pathway and complements that of the intercellular transfer of P-gp. These findings lend further support to the role of MPs in limiting the successful management of cancer.© 2013 Elsevier B.V. All rights reserved.

Abstract: Multidrug resistance (MDR) is a major impediment to the overall success of chemotherapy in clinical oncology. MDR has been primarily attributed by the ATP-dependent transmembrane proteins, P-glycoprotein (P-gp, ABCB1) and Multidrug Resistance-Associated Protein 1 (MRP1, ABCC1). These proteins maintain sublethal concentrations of intracellular chemotherapeutics by virtue of their drug efflux capacity. In this study, we report the acquisition and dissemination of functional MRP1 via microparticle (MP) mediated intercellular transfer. After we showed the transfer and functionality of P-gp in drug sensitive recipient cells, we report the transfer and time-dependent functionality of MRP1 in drug sensitive leukaemia cells following exposure to MPs shed by MRP1-overexpressing MDR cells. We also demonstrate a remarkable capacity for MPs shed from cells with a P-gp dominant resistance profile to re-template a pre-existing MRP1 dominant profile in recipient cells. These findings have significance in understanding the molecular basis for tumour dominant phenotypes and introduce potential new strategies and targets for the acquisition of MDR and other deleterious traits. © 2013 Elsevier B.V.

36. FDI and environmental regulation: Pollution haven or a race to the top?

Abstract: Increasing foreign direct investment (FDI) flows accompanied with globalization have raised the concern of a "race to the bottom" phenomenon in environmental protection. This is because footloose investors of "dirty" industries tend to relocate to "pollution havens" of the developing world. However when pollutant is transboundary (as in the case of greenhouse gases), the source country's incentive to relocate and the recipient country's willingness to host such industries are not straightforward. This article studies the relationship between FDI and environmental regulation using a North-South market share game model in a two-country setting, when pollution is transboundary. Contrary to the pollution haven hypothesis, our model shows that if market sizes of the two countries are small, FDI will raise the emission standard of the host country, resulting in a "race-to-the-top" phenomenon; but if market sizes are large enough, FDI will not change the emission standard of the South (from its laxest form), a finding that is consistent with the "regulatory chill" argument. Equilibrium FDI is contingent on the fixed cost of FDI, as the traditional proximity-concentration tradeoff theory predicts. © 2011 Springer Science+Business Media, LLC.

Abstract: Drug resistance is a major cause of cancer treatment failure, with multidrug resistance (MDR) being the most serious, whereby cancer cells display cross-resistance to structurally and functionally unrelated drugs. MDR is caused by overexpression of the efflux transporters P-glycoprotein (P-gp) and multidrug resistance-associated protein 1 (MRP1). These transporters act to maintain sublethal intracellular drug concentrations within the cancer cell, making the population treatment unresponsive. Recently, we discovered a novel nongenetic basis to MDR whereby microparticles (MPs) transfer P-gp intercellularly from MDR donor cells to drug-sensitive recipient cells. MPs isolated from MDR leukemia and breast cancer cells were cocultured with their drug-sensitive counterparts. P-gp transfer was assessed by direct immunolabeling, and acquired transcripts and regulatory microRNAs by quantitative real-time PCR. We show that MDR MPs incorporate nucleic acids; MPs change recipient cells'transcriptional environment to reflect donor MDR phenotype, and distinct pathways exist among cancers of different origin that may be dependent on donor cells'ABCB1 overexpression. We demonstrate that this pathway exists for both hematological and nonhematological malignancies. By conferring MDR and "retemplating"the transcriptional landscape of recipient cells, MPs provide a novel pathway, having implications in the dissemination and acquisition of deleterious traits in clinical oncology. © FASEB.

Abstract: The equilibrium susceptibility of uniaxial paramagnets is studied in a unified framework which permits to connect traditional results of the theory of quantum paramagnets, $\Sm=1/2, 1, 3/2$, ..., with molecular magnetic clusters, $\Sm\sim5, 10, 20$, all the way up, $\Sm=30, 50, 100$,... to the theory of classical superparamagnets. This is done using standard tools of quantum statistical mechanics and linear response theory (the Kubo correlator formalism). Several features of the temperature dependence of the susceptibility curves (crossovers, peaks, deviations from Curie law) are studied and their scalings with $\Sm$ identified and characterized. Both the longitudinal and transverse susceptibilities are discussed, as well as the response of the ensemble with anisotropy axes oriented at random. For the latter case a simple approximate formula is derived too, and its range of validity assessed, so it could be used in modelization of experiments., 32 pages, 5 figures. In this paper we investigate the asymptotic behaviour (as n→σ) of the zeros of πn, πn′ and also of the zeroes of certain closely related rational approximants to f. Abstract: To examine relationships between body mass index (BMI) and psychological correlates in Chinese school adolescents during the period of economic transition. Baseline data of 1655 Chinese adolescents aged 11–15 y were retrieved from a longitudinal smoking cessation and health promotion program in Wuhan, China. Assessments of body weight and height, depressive symptoms, perceived peer isolation (PPI) and perceived availability of social support (PASS) were collected. Based on the International Obesity Task Force (IOTF) age-and sex-specific BMI cutoffs, 12.5% of boys and 9.2% of girls were overweight. In girls, high BMI was significantly related to higher self-reported depressive symptoms, and was dominantly mediated by PPI. On the contrary, high BMI boys reported significantly lower levels of PPI although high PPI level aggravated depressive symptoms. For both girls and boys, the observed effect of PPI on the relationship between BMI and depressive symptoms was sustained only in low PASS boys and girls. The present study revealed different effects of PPI on the association of BMI and depressive symptoms between boys and girls, which were buffered by levels of PASS. The findings of this study may contribute to our understanding of the influences of psychological correlates in pediatric overweight in the Eastern cultural environment.

Abstract: Background: The renin-angiotensin-aldosterone system (RAAS) is involved in the pathogenesis of insulin resistance and type 2 diabetes in the general population. The RAAS is activated during pregnancy. However, it is unknown whether the RAAS contributes to glycemia in pregnant women. Methods: Plasma renin activity (PRA) and plasma aldosterone levels were quantified at delivery in 689 Chinese mothers. An oral glucose tolerance test in fasted women was performed in the second trimester of pregnancy. The diagnosis of gestational diabetes mellitus (GDM) and impaired glucose tolerance during pregnancy were made according to the guidelines of the Chinese Society of Obstetrics.
