5. The effect of CD39+CD8+ T cells and CD39 and CD73 on tumor cells on the treatment response of epithelial ovarian cancer and patient outcomes

- Abstract
Objective. In this study, we evaluated whether CD39+CD8+ tumor-infiltrating lymphocytes (TILs) and CD39 and CD73 on tumor cells had an effect on survival in epithelial ovarian cancer (EOC) patients treated with platinum-based chemotherapy. Methods. A total of 129 EOC patients between 2009 and 2015 were retrospectively reviewed. The expression of CD39+CD8+ TILs in the TME and CD39 and CD73 on tumor cells was measured by multiplexed quantitative fluorescence (QIF) on tissue microarrays (TMAs). Results. The expression of CD39+CD8+ TILs in the TME and CD39 and CD73 on tumor cells in EOC patients was negatively associated with the response to platinum-based treatment. The area under the ROC curve of CD39+CD8+ TILs and CD39 and CD73 on tumor cells IHC image quantification for predicting the platinum resistance in EOC patients was 0.679, 0.718, 0.682; respectively. Furthermore, the protein expression of CD39+CD8+ TILs and CD39 and CD73 on tumor cells were higher in the platinum Resistant group than in the Sensitive group (PConclusion. Our results show that the expression of CD39+CD8+ TILs and CD39 and CD73 on tumor cells could be useful markers of treatment response and prognosis in EOC patients treated with platinum-based chemotherapy in Chinese women.

The need to effectively and selectively remove radioactive 137Cs and 90Sr from nuclear waste solutions persists to mitigate their environmental mobility and high radiotoxicity. Because it is difficult to effectively remove them from acidic environments that degrade most sorbents, new sorbent materials are highly desirable. Here, efficient removal of Cs+ and Sr2+ is achieved by the composite of layered tin sulfide (Me2NH2)4/3(Me3NH)2/3Sn3S7·1.25H2O (FJSM-SnS) and polyacrylonitrile (PAN) (FJSM-SnS/PAN). The granulous composite possesses regular particle morphology and good mechanical strength as an engineered form. It shows excellent acid-base and γ-irradiation resistance, high maximum adsorption capacities (qm) of 296.12 and 62.88 mg/g for Cs+ and Sr2+ ions, respectively, and high selectivity even in the presence of excess Na+ ions or using lake water. Impressively, qmCs of FJSM-SnS/PAN reaches 89.29 mg/g under even acidic conditions (pH = 2.5). The column loaded with FJSM-SnS/PAN granules exhibits high removal rates (R) toward low-concentration Cs+ and Sr2+ ions under both neutral and acidic conditions. Moreover, the composite can be recycled and reused with high RCs and RSr. This work highlights the great potential of metal sulfide ion-exchangers in engineered form for the efficient removal of Cs+ or Sr2+ ions, especially under acidic conditions, for radionuclide remediation.

The selective capture of radioactive cesium, strontium, and lanthanides from liquid nuclear waste is of great significance to environmental remediation and human health. Herein, the rapid and selective removal of Cs+, Sr2+, and Eu3+ ions is achieved by two metal sulfides (FJSM-SnS-2 and FJSM-SnS-3). Both structures feature [Sn3S7]n2n- layers with the mixed cations of [CH3NH3]+ and [Bmmim]+ (1-butyl-2,3-dimethylimidazolium) as templates. However, the ratios and arrangements of mixed cations in the interlayered spaces are distinct. It is unprecedented that [CH3NH3]+ and [Bmmim]+ in FJSM-SnS-2 are alternatingly arranged in different interlayered spaces, whereas they in FJSM-SnS-3 are located in the same interlayered spaces. It is the first time that the ionic liquid cation and protonated organic amine have been simultaneously incorporated into metal sulfides. Both compounds show high capacities, rapid kinetics, and a wide pH active range for Cs+, Sr2+, and Eu3+. Even under excess Na+ ions, both show excellent selectivity in capturing trace Sr2+ and Eu3+ ions. FJSM-SnS-3 presents the highest KdEu to date. They still retain high removal efficiency even after intense β and γ radiation. Moreover, it is first confirmed by the in situ tracking method of mass spectrometry that the large-sized [Bmmim]+ ions are exchangeable. It is found that the arrangement of cations between interlayered spaces is a crucial factor affecting ion exchange performance. This work will likely change the consensus that large-sized organic cations are difficult to be exchanged and thus further highlight the great potential of metal sulfide ion exchangers for radionuclide remediation.

The control group was given conventional nursing care, and the experimental group received nursing intervention based on the G-Caprini scale. The rate of postoperative lower-limb deep-vein thrombosis in the two groups was compared, and the incidence of VTE and the level of nursing satisfaction in the two groups were statistically analyzed.Results: The incidence of postoperative VTE in each risk class of the G-Caprini scale was lower in the experimental group than in the control group, and the difference was statistically significant (P < 0.01). In the experimental group, 47 patients were very satisfied with the nursing care, 1 was satisfied, and 1 was dissatisfied, which meant the nursing satisfaction rate in the experimental group was 97.96 (48/49). In the control group, 40 patients were very satisfied with the nursing care, 2 were satisfied, 1 was basically satisfied, and 6 were dissatisfied; thus, the satisfaction rate for nursing care in the control group was 87.75%. The difference between the two groups was statistically significant (Ï2 = 19.657, p < 0.05).Conclusion: Nursing interventions based on the G-Caprini rating scale were significantly effective in preventing VTE in patients after gynecological surgery and resulted in higher levels of patient satisfaction in terms of nursing care.Keywords: G-Caprini scale, nursing intervention, gynecological surgery, venous thromboembolism

Two new cesium containing manganese vanadates are synthesized by a facile method. The known compound K2Mn(VO3)4 (1), synthesized via a milder method than the previous hydrothermal reaction, was firstly used as a precursor to prepare two new manganese vanadates, namely CsMn(VO3)3·H2O (2) and Cs4.34Mn1.83(VO3)8·2.965H2O (3) in concentrated Cs+ aqueous solutions at 80 °C. Compound 2 features a three-dimensional (3D) microporous framework constructed from serrated (VO3)n chains interconnected by Mn2+ ions. The Cs+ ions and the lattice water molecules are located in the channels of the structure along the a-axis. In compound 3, the sinusoidal (VO3)n chains are linked by Mn2+ ions into V–O–Mn double chains, which are further weaved by {Mn4O8} units to form a 3D microporous network with channels occupied by disordered Cs+ ions. Compounds 2 and 3 are the first Cs–Mn–V–O examples with a 3D microporous framework. Thermal stabilities of compounds 2 and 3 were investigated by thermogravimetric (TG) analysis. This work provides an effective synthetic method for preparing alkali-metal containing heterometallic vanadates with new composition and structures.

19. The Impact of Non-Invasive Ventilation Combined with Montelukast on Clinical Efficiency and Pulmonary Function in Patients with Bronchial Asthma Complicated by Obstructive Sleep Apnea Hypopnea Syndrome

Our study aims to explore the impact of non-invasive ventilation (NIV) alone or combined with montelukast on clinical efficiency and pulmonary function (PF) in treating patients with bronchial asthma complicated by obstructive sleep apnea hypopnea syndrome (OSAHS).A total of 386 patients with bronchial asthma underwent sleep monitoring. Patients were divided into 3 groups according to the different treatment methods. The changes in PF, apnea hypopnea index (AHI) score and the level of inflammatory factors in all patients before and after treatment were recorded, and the clinical effect following treatment was noted.Following treatment, the clinical efficiency of Group 2 was significantly better than that of both Group 1and the control group, and the therapeutic effect in Group 1 was better than in the control group (P.05). Before treatment, vital capacity (VC), peak expiratory flow (PEF), forced expiratory volume in 1 second/forced vital capacity (FEV1/FVC) and asthma control test (ACT) scores, AHI scores, C-reactive protein (CRP), tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) index were compared between the 3 groups (P.05). In contrast, after treatment the VC, PEF, FEV1/FVC and ACT, AHI, CRP and TNF-α scores and the IL-6 index in the 3 groups were improved compared with before treatment. The indices in Groups 1 and 2 were better than in the control group, and the VC, PEF, FEV1/FVC and ACT, AHI, CRP, and TNF-α scores and IL-6 index in Group 2 reported greater beneficial effect than in Group 1.The combination of NIV and montelukast exerts a beneficial effect in treating patients with bronchial asthma complicated with OSAHS, which holds the potential of effectively improving clinical symptoms and PF, reducing ACT and AHI scores and alleviating inflammatory reactions. Hence, the combination is valid and appropriate for clinical application.

20. Activation of the Nrf2 Antioxidant Pathway by Longjing Green Tea Polyphenols in Mice Livers

Previous studies have revealed that green tea polyphenol (GTP) could protect against liver injury due to oxidative stress. However, the mechanism underlying the bioactive actions of GTP in the liver has not been systematically evaluated. This study aimed to investigate the effect of GTP on the activation of the nuclear factor erythroid-2-related factor 2 (Nrf2)-Kelch-like ECH-associated protein 1 (keap1) pathway, using in silico and in vivo methods. Furthermore, the regulation of Nrf2 downstream target antioxidant response element (ARE) was also evaluated. The high-performance liquid chromatography analysis indicated that GTP includes 9 major compounds, and molecule docking analysis demonstrated that most of these polyphenols have a strong binding affinity with the keap1 Kelch domain, where keap1 binds to the Neh2 domain of Nrf2. Remarkably, the predominant compound of GTP, that is, epigallocatechin gallate, displayed the best binding affinity score, which can fully occupy all 3 polar subpockets of the keap1 Kelch domain. The Nrf2, keap1, and Nrf2 downstream target gene expression levels were changed in the livers compared to the control group. It showed that the Nrf2 expression level was significantly upregulated in GTP-induced mice liver across most treatments, while the keap1 expression level remained unchanged. Subsequently, we observed a significant increasing trend in the expression of the downstream ARE, including antioxidative enzymes, liver phase II enzymes, and liver efflux transporters in mice livers. 21. [Resveratrol suppressed the expression of MIP-2 induced by lipopolysaccharide in osteoblasts via up-regulating SOCS-3]

To explore whether resveratrol dependents on the production of suppressor of cytokine signaling suppressor 3 (SOCS-3) in inhibiting mRNA production of macrophage inflammatory protein-2 (MIP-2) in osteoblasts induced by lipopolysaccharides(LPS) extracted from Porphyromonas endodontalis(P.e).MC3T3-E1 cells were treated with different concentrations of resveratrol (0, 5, 10 and 20 μmol/L) and 20 μmol/L resveratrol for different time( 0, 10, 30, 60, 120 and 180 min). The expression of SOCS-3 protein was detected by Western blot. MC3T3-E1 cells were transfected with mouse SOCS3 siRNA (si-SOCS-3) and control siRNA(si-control). Reverse transcription real-time PCR(real-time RT-PCR) and Western blot was used to detect the silencing efficiency of SOCS-3. Cells were stimulated by 20 μg/mL P.e-LPS for 24 h after transfection, in the absence or presence of 20 μmol/L resveratrol for 1 h , and the changes of MIP-2 mRNA were determined by real-time RT-PCR. Statistical analysis was performed using one-way ANOVA and Dunnett t test with SPSS 13.0 software package.Treatment of MC3T3-El cells with different concentrations of resveratrol caused a significant increase in SOCS-3 protein expression in a dose-dependent manner. During the observation time of 180 min, SOCS-3 protein expression was the highest at 20 μmol/L resveratrol-treated osteoblasts for 60 min. The silencing efficiency of SOCS-3 mRNA was 63.7%. Transfection with SOCS-3 siRNA increased MIP-2 mRNA expression in LPS-stimulated MC3T3-E1 cells and negated the inhibitory effects of resveratrol on LPS-induced MIP-2 mRNA expression(P0.05).Resveratrol inhibits the expression of MIP-2 mRNA in osteoblasts induced by P.e-LPS by up-regulating the expression of SOCS-3 protein.

With the in situ-generated [Pb(MCP)4]2+ (HMCP+ = 1-methyl-4-(carboxyl)pyridinium) or [M(phen)3]2+ (M = Co, Fe and Ni; phen = 1,10-phenanthroline) complexes as structural directing agents and charge-balancing ions, we solvothermally synthesized and structurally characterized four new organic-inorganic hybrid iodoplumbates. Compound K2[Pb(MCP)4]Pb3I10 (1) represents the first K+ and [Pb(MCP)4]2+ co-templated hybrid haloplumbate, and exhibits a curve-like anionic layer of [Pb3I10]n4n-. Compounds [M(phen)3]Pb2I6·CH3CN (M = Co (2), Fe (3) and Ni (4)) have isostructural phases, and feature a one-dimensional (1D) [Pb2I6]n2n- anionic chain characteristic of pyramid-like [PbI5] units. The optical property studies show that compounds 1-4 exhibit semiconductor behaviors with the band gaps of 1.98-2.68 eV. In addition, the title compounds exhibited interesting photoelectrical responsive properties, with the photocurrent density in the order of 1 > 3 > 2 > 4. The thermal stabilities of the title compounds 1-4, as well as the theoretical band structure and density of states (DOS) of compounds 1 and 2 have also been studied.

ObjectiveDNA mismatch repair deficiency is not only thought to promote tumorigenesis but is also suggested to be associated with platinum-based chemotherapy treatment. In this study, we investigated the effects of two genetic polymorphisms in the hMSH2 and hMLH1 genes on the risk of epithelial ovarian cancer and the clinical outcome of patients treated with platinum-based chemotherapy.MethodsA case-control study was performed in 536 epithelial ovarian cancer patients and 532 control women. Genotypes of two polymorphisms were determined by the polymerase chain reaction/ligase detection reaction method. Pearson Chi-square test was used to evaluate genotype distributions and allele frequencies in the patients and controls. Kaplan-Meier survival curves, and univariate and multivariate Cox regression models were used to analyze the effect of polymorphisms on patients’ prognoses.ResultsThe genotype and allele frequencies of the rs2303428 and rs1800734 polymorphisms were not significantly different between the case and control groups. Compared with wild homozygous genotype, the presence of variant alleles (heterozygous and variant homozygous genotypes) did not affect the risk of developing epithelial ovarian cancer. However, survival analysis showed that the rs2303428 polymorphism was related to the prognosis of epithelial ovarian cancer patients. Compared with the TT genotype, patients carrying the C allele had a shorter progression-free survival during the 3- and 5-year follow-up (HR 1.41, 95% CI 1.07 to 1.87 and HR 1.56, 95% CI 1.12 to 2.16, respectively). For the rs1800734 polymorphism, the A allele may significantly increase patients’ progression-free survival compared with the GG genotype in the 5-year follow-up (HR 0.66, 95% CI 0.44 to 0.98).ConclusionOur research suggests that genetic polymorphisms in hMSH2 and hMLH1 may indicate the clinical progression of epithelial ovarian cancer patients treated with platinum-based chemotherapy.

To compare clinical effects of different postoperative rehabilitation modes on lumbar degenerative diseases, and explore influence of rehabilitation mode and other factors on postoperative effect.From June 2013 to July 2016, totally 900 patients were admitted from nine tertiary hospitals in Beijing to perform single segment bone grafting and internal fixation due to lumbar degenerative diseases were prospectively analyzed. There were 428 males and 472 females, the age of patient over 18 years old, with an average of (51.42±12.41) years old;according to patients' subjective wishes and actual residence conditions, all patients were divided into three groups, named as observation group 1 (performed integrated rehabilitation approach and orthopedic treatment model intervention), observation group 2 (performed integrated rehabilitation approach and orthopedic treatment, classified rehabilitation model intervention), and control group(performed routine rehabilitation model intervention). Visual analogue scale(VAS), Oswestry Disability Index(ODI) and Japanese Orthopaedic Association (JOA) were used to evaluate postoperative efficacy among three groups at 24 weeks. Possible factors affecting the postoperative efficacy including age, age grouping, gender, body mass index (BMI), BMI grouping, education level, visiting hospital, payment method of medical expenses, preoperative complications, preoperative JOA score, clinical diagnosis, surgery section, operative method, intraoperative bleeding volume, postoperative complications and rehabilitation mode were listed as independent variables, and postoperative ODI score at 24 weeks as dependent variables. Univariate analysis was used to analyze relationship between influencing factors and postoperative efficacy. Multiple linear regression was used to analyze relationship between influencing factors, rehabilitation mode and postoperative ODI score at 24 weeks, in further to find out the main reasons which affect postoperative efficacy, and to analyze impact of rehabilitation mode on postoperative efficacy.All patients were followed up for 24 weeks after operation. All incisions healed at stage I with stable internal fixation. (1)Evaluation of postoperative efficacy:① There were no statistical differences in preoperative VAS and ODI among three groups(Preoperative JOA score, gender, age could predict postoperative clinical effects of lumbar degenerative diseases in varying degrees treated with single level bone graft fusion and internal fixation. Different rehabilitation modes could improve clinical effects. Intergrated rehabilitation orthopedic treatment model and integrated rehabilitation approach and orthopedic treatment with classifiedrehabilitation model are superior to conventional rehabilitation model in improving patients' postoperative function and relieving pain, which is worthy of promoting in clinical.

Aberrant DNA methylation is considered to play a critical role in the chemoresistance of epithelial ovarian cancer (EOC). In this study, we explored the relationship between hypermethylation of the Mahogunin Ring Finger 1 (MGRN1) gene promoter and primary chemoresistance and clinical outcomes in high-grade serous ovarian cancer (HGSOC) patients. The MALDI-TOF mass spectrometry assays revealed a strong association between hypermethylation of the MGRN1 upstream region and platinum resistance in HGSOC patients. Spearman’s correlation analysis revealed a significantly negative connection between the methylation level of MGRN1 and its expression in HGSOC. In vitro analysis demonstrated that knockdown of MGRN1 reduced the sensitivity of cells to cisplatin and that expression of EGR1 was significantly decreased in SKOV3 cells with low levels of MGRN1 expression. Similarly, EGR1 mRNA expression was lower in platinum-resistant HGSOC patients and was positively correlated with MGRN1 mRNA expression. Kaplan-Meier analyses showed that high methylation of the MGRN1 promoter region and low expression of MGRN1 were associated with worse survival of HGSOC patients. In multivariable models, low MGRN1 expression was an independent factor predicting poor outcome. Furthermore, low expression of EGR1 was also been confirmed to be significantly related to the poor prognosis of HGSOC patients by Kaplan-Meier. The hypermethylation of the MGRN1 promoter region and low expression of MGRN1 were associated with platinum resistance and poor outcomes in HGSOC patients, probably by altering EGR1 expression.

Ethnopharmacological relevance Ginseng (Panax ginseng C. A. Meyer), a traditional Chinese medicine, is widely used in the adjunctive therapy of the liver diseases. Aim of the study Ginsenosides are one kind of the main active ingredients in ginseng. Although hepatoprotective mechanisms of ginsenosides, such as anti-oxidation, anti-inflammation and anti-apoptosis, have been well studies, little is known about the effect of ginsenosides on drug metabolism in liver. Since CYP3A11/3A4 is a major enzyme catalyzing the drug metabolism in liver, an investigation of the enzyme's expression during the progression of a liver disease will gain valuable information about the hepatic drug metabolism. The purpose of this study was to determine the effect of ginsenosides on the expression of hepatic CYP3A11/3A4 in the lipopolysaccharides (LPS) injured human HepG2 cells and mice. We hypothesize that ginsenosides are important to stabilize CYP3A11/3A4 expression in an injured liver. Materials and methods In this study, LPS was intraperitoneally intermittently injected to induce the liver injury in mice. Ginsenosides were intragastrically administered to mice for 7 days to treat the liver injury. Serum biochemical analysis and histopathological study were taken to evaluate the hepatoprotective effect of ginsenosides. The effect of ginsenosides was also evaluated in human HepG2 cells in the presence and absence of LPS. Real-time PCR and western blotting method were used to detect the mRNA and protein levels of CYP3A11/3A4 in mouse liver tissue and human HepG2 cells. The reporter gene-transfected cells were used to identify upstream targets in HepG2 cells. Results LPS injection in mice resulted in the up-regulation of pro-inflammatory cytokines such as IL-1β, IL-6 and TNF-α in liver, up-regulation of hepatic enzymes such as Tbil, ALT, AST and ALP in serum, and down-regulation of CYP3A11/3A4 expression in liver. Ginsenosides treatment reversed the up-regulation of pro-inflammatory cytokines and serum hepatic enzymes elicited by LPS. Pathological results suggest that ginsenosides reduced liver damage. Moreover, ginsenosides reversed the decrease of CYP3A11/3A4 expression in the liver of LPS-injured mouse and in LPS-treated HepG2 cells. To further investigate the regulatory mechanisms, we found that ginsenosides enhanced the rifampicin-induced pregnane X receptor (PXR) transactivation of the CYP3A4 promoter. Treatment of hPXR–over-expressed cells with ginsenosides increased the rifampicin-inducible expression of CYP3A4 in a concentration-dependent manner. Conclusion Ginsenosides reverse the effects of LPS-induced hepatic CYP3A11/3A4 dysfunction, suggesting that the stabilization of the CYP3A11/3A4 expression in an injured liver appears a novel hepatoprotective mechanism of ginsenosides.

The current study was conducted to explore the clinical features and risk factors of patients with asthma complicated by obstructive sleep apnea-hypopnea syndrome (OSAHS). Patients with asthma who underwent polysomnography in our hospital from August 2017 to December 2019 were enrolled in the study. Data on demographics, pulmonary function testing, polysomnography, blood gases, mean pulmonary artery pressure, and vascular endothelial growth factor (VEGF) were compared between the two groups. Of 238 patients with asthma, 93 who also had OSAHS formed the observation group and were subclassified into mild (n = 33), moderate (n = 41), and severe (n = 19) categories, while 145 patients with asthma alone were assigned to the control group. Hai-Yan Sun, Qiu Tang, Jian-Hong Chen, Xiao-Juan Lv, Ye-Qiang Tu, Ding-Ding Yan Department of Gynecologic Oncology, Zhejiang Cancer Hospital, Hangzhou, China Purpose: To determine if postoperative cisplatin concurrent chemoradiotherapy (CCRT) improves the outcome in stage IA/IIB cervical cancer patients with intermediate risk factors, when compared with radiation therapy (RT) alone, and identify the potential eligible populations for this treatment.Patients and methods: We reviewed medical records of 1,240 patients with stage IA/IIB cervical cancer who underwent radical hysterectomy and pelvic lymphadenectomy in our hospital between January 2008 and December 2011. Of the 1,240 patients, 436 displayed 1 or more intermediate risk factors. Of these, we screened 306 patients who underwent RT only or CCRT. We analyzed the effects of CCRT on survival and prognosis.Results: The 5-year progress-free survival (PFS) in the CCRT group was superior to that in the RT-only group (96.0% vs 89.0%, respectively; P=0.031). The 5-year overall survivals (OSs) were not different between the 2 groups (P=0.141). Compared with RT-only group, CCRT did not improve PFS or OS in patients with 1 risk factor, large tumor size, or deep stromal invasion (P>0.05). Compared with RT-only group, CCRT improved PFS (97.9% vs 82.8%; P=0.017) but did not increase OS (97.9% vs 89.7%; P=0.109) in patients with lymphovascular space invasion plus deep stromal invasion/large tumor size. OS (92.3% vs 70.6%; P=0.048) and PFS (92.3% vs 64.7%; P=0.020) in the CCRT group were superior to those in the RT-only group with 3 risk factors. Compared with RT-only group, CCRT was an independent prognostic factor for favorable PFS (hazard ratio [HR] =0.238; 95% CI =0.0827–0.697, P=0.009) and OS (HR =0.192; 95% CI =0.069–0.533, P=0.002).Conclusion: Postoperative CCRT improved survival in stage IA/IIB cervical cancer patients with intermediate risk factors. It is of vital importance to capture lanthanides (nuclear fission products) from waste solutions for radionuclide remediation owing to their hazards. The effective separation of lanthanides are achieved by an acid/base-stable and radiation-resistant vanadate, namely, [Me2 NH2 ]V3 O7 (1). It exhibits high adsorption capacities for lanthanides (qmEu =161.4 mg g-1 ; qmSm =139.2 mg g-1 ). And high adsorption capacities are maintained over a pH range of 2.0-6.9 (qmEu =75.1 mg g-1 at low pH of 2.5). It displays high selectivity for Eu3+ (simulant of An3+ ) against a large excess of interfering ions. It can efficiently separate Eu3+ and Cs+ (or Sr2+ ) with the highest separation factor SFEu/Cs of 156 (SFEu/Sr of 134) to date. The adsorption mechanism is revealed by calculations and XPS, EXAFS, Raman, and elemental analyses. These merits combined with facile synthesis and convenient elution makes the title vanadate a promising lanthanide scavenger for environmental remediation.

Purpose: Regulation of gene expression is fine-tuned by a dynamic equilibrium between repressive modifications and transcriptional activation of histone tails. Jumonji domain-containing 3 (Jmjd3), also known as KDM6B, is a specific histone demethylase for trimethylation on histone H3 lysine 27 (H3K27me3) that specifically removes the methylation of H3K27me3 and promotes gene expression. Our previous study showed that Jmjd3 inhibits serum deprivation-induced osteoblast apoptosis. In this study, we clarified the role of Jmjd3 in tumor necrosis factor-alpha (TNF-α)-induced osteoblast apoptosis. Materials and Methods: Jmjd3 activity was inhibited by GSK-J4. Transfection of osteoblastic murine MC3T3-E1 cells with short hairpin RNA (shRNA) was used to establish stable Jmjd3 knockdown cells. Osteoblast apoptosis was detected using Annexin V-APC/PI staining, cysteinyl aspartate specific protease-3 (caspase-3) activity assays, and Western blot. Real-time polymerase chain reaction (PCR) and chromatin immunoprecipitation (ChIP) assays were performed to clarify the mechanism responsible for Jmjd3-regulated osteoblast apoptosis induced by TNF-α. Results: Based on Annexin V-APC/PI staining, caspase-3 activation, and poly ADP-ribose polymerase (PARP) cleavage, pretreatment with GSK-J4 and knockdown of Jmjd3 by shRNA transfection each inhibited osteoblast apoptosis. Furthermore, knockdown of Jmjd3 decreased the expression of Ras association domain family 5 (RASSF5), which is a pro-apoptotic gene of the Ras associated domain family. H3K27me3 levels in the promoter region of RASSF5 were up-regulated in the Jmjd3 knockdown cells. Conclusions: Jmjd3 regulated TNF-α-induced osteoblast apoptosis by targeting RASSF5.

To apply Flipped classroom combined with case-based learning(CBL) in resident training of endodontics, in order to improve training efficiency.Fifty-one residents from China Medical University, School of Stomatology were randomly divided into 2 groups, twenty-six students in the experimental group were trained with Flipped classroom combined with CBL, the other twenty-five students in the control group were trained with traditional teaching method. At the end of the course, exams and questionnaires were conducted to evaluate the training quality of two different methods. The exams consisted of didactic and operational assessment. The data were analyzed with SPSS 13.0 software package.The results of didactic exam and comprehensive evaluation indicated that experimental group was significantly higher than the control group(P0.05). The results of questionnaires indicated that residents showed much more satisfied with Flipped classroom combined with CBL (P0.05).However, there was no significant difference in operational assessment(P0.05).Compared with traditional teaching method, Flipped classroom combined with CBL can achieve better training effect, which is worthy of further application in dental resident training.

To evaluate the clinical effect of different kinds of laser on 60 molars with abrasion.Thirty patients with 60 abrasive molars were selected according to the inclusion criteria. Molars and premolars were divided into 2 groups randomly. Teeth in the experimental group were treated with Er,Cr:YSGG laser combined with AdperTM Easy One, while teeth in the control group were treated with Nd:YAG laser combined with AdperTM Easy One, composite resin Z350 was selected to restore the defect. The modified USPHS criteria was used to evaluate the treatment effects at recall periods.The data were analyzed using SPSS 13.0 software package.On retention, the B level rate of the experimental group was significantly lower than that of the control group(P0.05) 12 months later. For success rate at 18 months,the difference between the experimental group and the control group was significant(P0.05). At the same time, sensitivity of tooth and overall response in the two groups had no significant difference (P0.05).Although the overall response between the two groups had no significant difference,Er,Cr:YSGG laser shows better effect of retention, which is the preferred option for treatment of abrasive molars.

The focus of the study was to establish an UPLC-ESI-MS/MS method for the qualitative and quantitative determination of VLPVPR in rat plasma. The method was as follows: Protein impurities in the rat plasma samples were precipitated with methanol; A Waters Acquity BEH C18 column (100 mm×2.1 mm, 1.7 μm) was adopted; the column temperature was 25°C; the mobile phase consisted of methanol-water with gradient elution at the steady flow rate of 0.3 mL·min-1; the detection wavelength was 202 nm. Mass spectrometry applied selected ion monitoring mode with m/z 255 (quantitative ion) and m/z 169 (qualitative ion). It was found that excellent linear relationship was obtained from the range of 10~200 ng·mL-1 (r=0.9991), the limit determination (LOD) of VLPVPR was 1.8 ng·mL-1, the recovery rate was 96.33%~100.76%, The inter and intre-day RSD were less than 7%. We thus conclude that a rapid and sensitive UPLC-ESI-MS/MS method was developed and validated for the determination of recombinant antihypertensive peptide VLPVPR in rat plasma. It can be applied to evaluate the pharmacokinetics of VLPVPR.

The purpose of this study is to observe whether Huangshan maofeng tea polyphenols extracts have protection effect for mice liver injuries caused by carbon tetrachloride (CCl4) and its mechanism. Method: High (300 mg/kg), middle (150 mg/kg) and low (75 mg/kg) doses of Huangshan maofeng tea polyphenols were administered for 7 days. One hour after the last gavage, all groups (except the control group) will inject in peritoneal 0.10% CCl4 olive oil solution. After 24 hours, blood from eyeball and liver tissue will be extracted. Changes in content of Alanine aminotransferase (ALT) and Aspartate aminotransferase (AST) will be detected; The expression of the mRNAs and proteins of the four liver enzymes were detected using Real-Time PCR and Western Blot. Result: Huangshan maofeng tea polyphenols low, medium, high dose groups can all decrease the activity of ALT and AST in serum of mice with acute liver injury; Huangshan maofeng tea polyphenols low, medium, high dose groups can all induce mice liver Cyp3a11 mRNA and protein expression. Conclusion: Huangshan maofeng tea polyphenols extracts can protect liver by inducing the expression of Cyp3a11.

Multidrug resistance is the main obstacle in cancer chemotherapy. Emerging evidence demonstrates the important role of autophagy in cancer cell resistance to chemotherapy. Therefore, autophagy inhibition by natural compounds may be a promising strategy for overcoming drug resistance in liver cancer cells. Here, we found that ADCX, a natural cycloartane triterpenoid extracted from the traditional Chinese medicine (TCM) source Cimicifugae rhizoma (Shengma), impaired autophagic degradation by suppressing lysosomal cathepsin B (CTSB) expression in multidrug-resistant liver cancer HepG2/ADM cells, thereby leading to autophagic flux inhibition. Moreover, impairing autophagic flux promoted ADCX-induced apoptotic cell death in HepG2/ADM cells. Interestingly, Akt was overactivated by ADCX treatment, which downregulated CTSB and inhibited autophagic flux. Together, our results provide the first demonstration that an active TCM constituent can overcome multidrug resistance in liver cancer cells via Akt-mediated inhibition of autophagic degradation.

New hybrid films were synthesized by SiO2 doping into phosphomolybdic acid (PMoA)/polyvinyl pyrrolidone (PVPd) composite system, and the effect of SiO2 doping on microstructure and photochromic properties was studied via atomic force microscope (AFM), Fourier transform infrared spectroscopy (FT-IR), ultraviolet–visible spectra (UV–vis) and X-ray photoelectron spectroscopy (XPS). The hybrid film surface with dome projection changed rough from smooth after SiO2 doping. The vibrational spectra of FT-IR verified that component of the heteropolyacids and the polymer did not changed, the vibration bands were expanded and had shifted by a few cm−1, and changed interaction between the acid and the polymer after SiO2 doping. The hybrid film exhibited a good photochromic property, which changed from colorless to blue under UV irradiation. The color change intensity of SiO2/PMoA/PVPd film was 1.48 times stronger than that of PMoA/PVPd under the same condition. XPS results indicated that the amount of PMoA in photo-reductive reaction was increased after SiO2 doping.

Under drip irrigation, the drip discharge has a great influence on soil water movement and crop growth. Studying the soil water movement rules makes great realistic significance to the precise management. Based on soil dynamics theory, we measured the water movement distance of saline soil in Changji Xinjiang through outdoor tests. Then, we built transfer function models between drip discharge and water movement distance under drip irrigation and applied the genetic algorithm to identifying and optimizing model’s parameters. Comparing the soil water movement model’s dynamic change at the same drip discharge, the model’s validity is validated. It can solve the feedback adjusting according to crop root, and the waste of water resource is reduced.
