Background: Vascular aging, as assessed by structural and functional arterial properties, is an independent predictor of cardiovascular outcomes. We aimed to explore the associations of individual cardiovascular risk factors from childhood to midlife and their accumulation over a 30-year span with vascular aging in midlife. Methods: Using data from the ongoing cohort of Hanzhong Adolescent Hypertension study, 2180 participants aged 6 to 18 years at baseline were followed for over 30 years. Distinct trajectories of systolic blood pressure (SBP), body mass index (BMI), and heart rate from childhood to midlife were identified by group-based trajectory modeling. Vascular aging was assessed by carotid intima media thickness or brachial-ankle pulse wave velocity. Results: We identified 4 distinct SBP trajectories, 3 distinct BMI trajectories, and 2 distinct heart rate trajectories from childhood to midlife. Persistently increasing SBP, high-increasing BMI, and high-stable heart rate were all shown to have a positive association with brachial-ankle pulse wave velocity in midlife. For carotid intima–media thickness, similar associations were observed for persistently increasing SBP and high-increasing body mass index. After further adjustment for SBP, body mass index and heart rate at the time of vascular assessment in 2017, associations were also observed for cardiovascular risk factor trajectories accumulation with brachial-ankle pulse wave velocity (β, 0.656 [95% CI, 0.265–1.047]) and with carotid intima media thickness (β, 0.045 [95% CI, 0.011–0.079]) in adulthood. Conclusions: Longitudinal exposure to individual cardiovascular risk factors from childhood to midlife and cardiovascular risk factor accumulation were associated with an increased risk of vascular aging in midlife. Our study lends support for early targeting of risk factors in order to prevent cardiovascular disease later in life.

The radiance of sky brightness differs principally with wavelength passband. Atmospheric scattering of sunlight causes the radiation in the near-infrared band. The Antarctic is a singular area of the planet, marked by an unparalleled climate and geographical conditions, including the coldest temperatures and driest climate on Earth, which leads it to be the best candidate site for observing in infrared bands. At present, there are still no measurements of night-sky brightness at DOME A. We have developed the Near-Infrared Sky Brightness Monitor (NISBM) in the J, H, and Ks bands for measurements at DOME A. The instruments were installed at DOME A in 2019 and early results of NIR sky brightness from 2019 January–April have been obtained. The variation of sky background brightness with solar elevation and scanning angle is analysed. The zenith sky flux intensity for the early night at DOME A in the J band is in the 600–1100 μJy arcsec−2 range, that in the H band is between 1100 and 2600 μJy arcsec−2, and that in the Ks band is in the range ∼200–900 μJy arcsec−2. This result shows that the sky brightness in J and H bands is close to that of Ali in China and Mauna Kea in the USA. The sky brightness in the Ks band is much better than that in Ali, China and Mauna Kea, USA. This shows that, from our early results, DOME A is a good site for astronomical observation in the Ks band.

Probiotics play an important role in the host and have attracted widespread attention as an alternative to antibiotics. Arbor Acres broilers were used in the present experiment and fed different doses of compound probiotics at 1, 5, and 10g kgWe discovered 425 differentially expressed genes (DEGs; upregulated: 256, downregulated: 169) in the cecal transcriptome study. These DEGs were assigned to fat metabolic pathways, such as the PPAR signaling pathway, according to KEGG analysis. Probiotics downregulated LPL and up-regulated PPARα expression in the cecum. In metabolome analysis of broilers cecum, we screened 86 differential metabolites and performed KEGG enrichment analysis of these metabolites. The KEGG analysis showed that these differentially expressed metabolites were annotated to nucleotide metabolism-related pathways, such as purine metabolism. In the cecum, probiotics upregulated the content of guanine, AMP, 3'-AMP, adenylosuccinate, deoxyguanosine, and ADP-ribose, whereas downregulated the content of 5-hydroxyisourate. Comprehensive transcriptome and metabolome analysis revealed that glycolysis, gluconeogenesis, and glycerophospholipid metabolism pathways were jointly enriched in cecum of broilers fed probiotics-containing diet.This study provides valuable information for studying the regulation and gene metabolism network of probiotics on caecal metabolism in broilers. Currently, the offline manual periodic detection method is a well-established practice in detecting the thermal state of the converter heatsink. This method, however, is huge in maintenance costs. To lower maintenance costs and improve maintenance efficiency in detecting the thermal dissipation state, this paper proposes an intelligent online prediction scheme based on Gauss-Newton iteration method. Firstly, the power loss model of the power module is established according to the characteristics of IGBT and FWD. The power loss of the power device is then calculated in real time with the voltage and current parameters of the converter. Next, the transient thermal model of the heatsink is established based on thermodynamics theory. And the calculation method of steady thermal resistance of heatsink based on Gauss-Newton iteration method is proposed according to the model. The transient thermal impedance data allow for timely prediction of thermal resistance of the heatsink and characterize the thermal state of the heatsink. Finally, with the help of DSP28377D, an experimental platform is built to verify the scheme. Results show that this method can realize intelligent prediction of thermal state online

Background: Promoting bone marrow mesenchymal stem cell (BMSC) osteoblastic differentiation is a promising therapeutic strategy for osteoporosis (OP). The present study demonstrates that miR- 483-5p inhibits the osteogenic differentiation of BMSCs. Therefore, selectively delivering the nanoparticles carrying antagomir-483-5p (miR-483-5p inhibitor) to BMSCs is expected to become an effective treatment drug for OP. Methods: Real-time PCR assays were used to analyze miR-483-5p, ALP and Bglap levels in BMSCs of ovariectomized and aged osteoporotic mice. Immunoglobulin G and poloxamer-188 encapsulated the functional small molecules, and a BMSC-targeting aptamer was employed to confirm the direction of the nanoparticles to selectively and efficiently deliver antagomir-483-5p to BMSCs in vivo. Luciferase assays were used to determine the target genes of miR-483-5p. Western blot assays and immunohistochemistry staining were used to detect the targets in vitro and in vivo. Results: miR-483-5p levels were increased in BMSCs of ovariectomized and aged osteoporotic mice. Inhibiting miR-483-5p levels in BMSCs by antagomir-483-5p in vitro promoted the expression of bone formation markers, such as ALP and Bglap. The FAM-BMSC-aptamer-nanoparticles carrying antagomir- 483-5p were taken up by BMSCs, resulting in stimulation of BMSC osteoblastic differentiation in vitro and osteoporosis prevention in vivo. Furthermore, our research demonstrated that mitogen-activated protein kinase 1 (MAPK1) and SMAD family member 5 (Smad5) were direct targets of miR-483-5p in regulating BMSC osteoblastic differentiation and osteoporosis pathological processes. Conclusions: The important therapeutic role of FAM-BMSC-aptamer-nanoparticles carrying antagomir- 483-5p in osteoporosis was established in our study. These nanoparticles are a novel candidate for the clinical prevention and treatment of osteoporosis. The optimized, targeted drug delivery platform for small molecules will provide new ideas for treating clinical diseases.

Sulfurized polyacrylonitrile (SPAN) represents one of the most promising directions for high-energy-density lithium (Li)-sulfur batteries. However, the practical application of Li||SPAN is currently limited by the insufficient chemical/electrochemical stability of electrode/electrolyte interphase (EEI). Here, a pinned EEI layer is designed for stabilizing a SPAN cathode by regulating the EEI formation mechanism in an advanced LiFSI/ether/fluorinated-ether electrolyte. Computational simulations and experimental investigations reveal that, benefiting from the nonsolvating nature, the fluorinated-ether can not only act as a protective shield to prevent the Li polysulfides dissolution but also, more importantly, endow a diffusion-controlled EEI formation process. It promotes the formation of a uniform, protective, and conductive EEI layer pinning into SPAN surface region, enabling the high loading Li||SPAN batteries with superior cycling stability, wide temperature performance, and high-rate capability. This design strategy opens an avenue for exploring advanced electrolytes for Li||SPAN batteries and guides the interface design for broad types of battery systems.

Implementing quantum algorithms on realistic devices requires translating high-level global operations into sequences of hardware-native logic gates, a process known as quantum compiling. Physical limitations, such as constraints in connectivity and gate alphabets, often result in unacceptable implementation costs. To enable successful near-term applications, it is crucial to optimize compilation by exploiting the capabilities of existing hardware. Here we implement a resource-efficient construction for a quantum version of AND logic that can reduce the compilation overhead, enabling the execution of key quantum circuits. On a high-scalability superconducting quantum processor, we demonstrate low-depth synthesis of high-fidelity generalized Toffoli gates with up to 8 qubits and Grover’s search algorithm in a search space of up to 64 entries. Our experimental demonstration illustrates a scalable and widely applicable approach to implementing quantum algorithms, bringing more meaningful quantum applications on noisy devices within reach.

Silicon-based optical switch is one of the key components for on-chip optical interconnect systems, and mode division multiplexing technology has been employed to boost optical switches’ channel capacity. However, the majority of the proven multimode optical switches have a switching time in the microsecond range, which is insufficient for some applications. In this paper, we design and experimentally demonstrate a high-speed dual-mode 4 × 4 optical switch based on a mode-diversity scheme, composed of four pairs of mode multiplexers and de-multiplexers, and two optimized single-mode 4 × 4 optical switches. Fast switching is enabled based on the carrier dispersion effect. At the same time, we improve the performances of the optical switch by reducing the number of optical switch units used in the 4 × 4 Spanke–Beneš architecture. Its power consumptions are reduced by ∼17%. Its insertion losses are within 8.8 dB in the wavelength range of 1525–1565 nm in the both sates of “through” and “all-cross”, while the optical signal-to-noise ratios are larger than 12.8 dB. Also, 50 Gbps data transmission experiments verify the device’s data transmission functionality.

Clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein 9 (Cas9) system adapted from bacteria is a programmable nuclease-based genome editing tool. The long-lasting effect of gene silencing or correction is beneficial in cancer treatment. Considering the need to broaden the practical application of this technology, highly efficient non-viral vectors are urgently required. We prepared a multifunctional non-viral vector that could actively target tumor cells and deliver CRISPR/Cas9 plasmids into nuclei of cancer cells. Protamine sulfate (PS) which contains nuclear localization sequence was utilized to condense plasmid DNA and facilitate nuclei-targeted delivery. Liposome-coated protein/DNA complex avoided the degradation of nuclease in blood circulation. The obtained PS@Lip/pCas9 was further modified with distearoyl phosphoethanolamine-polyethylene glycol-hyaluronic acid (HA) to endow the vector ability to actively target tumor cell. Results suggested that PS@HA-Lip could deliver CRISPR/Cas9 plasmids into nuclei of tumor cells and induce genome editing effect. With the disruption of MTH1 (mutT homolog1) gene, the growth of non-small cell lung cancer was inhibited. Moreover, cell apoptosis in tumor tissue was promoted, and liver metastasis of non-small cell lung cancer (NSCLC) was reduced. Our study has provided a therapeutic strategy targeting MTH1 gene for NSCLC therapy. STATEMENT OF SIGNIFICANCE: CRISPR/Cas9 as a powerful tool for genome editing has drawn much attention. The long-lasting effect possesses unique advantage in cancer treatment. Non-viral vectors have high loading capacity, high safety and low immunogenicity, playing an important role in CRISPR/Cas9 delivery. In our study, a multifunctional non-viral vector for the efficient delivery of CRISPR/Cas9 plasmid was constructed. With the active targeting ligand and nuclei-targeting component, the cargo was efficiently delivered into cell nuclei and exerted genome editing effect. By using this vector, we successfully inhibited the growth and induced the apoptosis of non-small cell lung cancer by disrupting MTH1 expression with good safety. Our work provided an efficient non-vial vector for CRISPR/Cas9 delivery and explored the possibility for cancer treatment.

Neural precursor cell expressed developmentally downregulated 4-like (NEDD4L), a member of the E3 ubiquitin-protein ligases, encoded by NEDD4L gene, was found to be involved in in salt sensitivity by regulating sodium reabsorption in salt-sensitive rats. The authors aimed to explore the associations of NEDD4L genetic variants with salt sensitivity, blood pressure (BP) changes and hypertension incidence in Chinese adults. Participants from 124 families in Northern China in the Baoji Salt-Sensitive Study Cohort in 2004, who received the chronic salt intake intervention, including a 7-day low-salt diet (3.0 g/day) and a 7-day high-salt diet (18 g/day), were analyzed. Besides, the development of hypertension over 14 years was evaluated. NEDD4L single nucleotide polymorphism (SNP) rs74408486 was shown to be significantly associated with systolic BP (SBP), diastolic BP (DBP) and mean arterial pressure (MAP) responses to low-salt diet, while SNPs rs292449 and rs2288775 were significantly associated with pulse pressure (PP) response to high-salt diet. In addition, SNP rs4149605, rs73450471, and rs482805 were significantly associated with the longitudinal changes in SBP, DBP, MAP, or PP at 14 years of follow-up. SNP rs292449 was significantly associated with hypertension incidence over the 14-year follow-up. Finally, this gene-based analysis found that NEDD4L was significantly associated with longitudinal BP changes and the incidence of hypertension over the 14-year follow-up. This study indicated that gene polymorphism in NEDD4L serve an important function in salt sensitivity, longitudinal BP change and development of hypertension in the Chinese population.

We investigate solutions of the 2d incompressible Euler equations, linearized around steady states which are radially decreasing vortices. Our main goal is to understand the smoothness of what we call the spectral density function associated with the linearized operator, which we hope will be a step towards proving full nonlinear asymptotic stability of radially decreasing vortices. The motivation for considering the spectral density function is that it is not possible to describe the vorticity or the stream function in terms of one modulated profile. There are in fact two profiles, both at the level of the physical vorticity and at the level of the stream function. The spectral density function allows us to identify these profiles, and its smoothness leads to pointwise decay of the stream function which is consistent with the decay estimates first proved in Bedrossian-Coti Zelati-Vicol in [5]., 64 pages

This paper presents a neuromorphic processing system with a spike-driven spiking neural network (SNN) processor design for always-on wearable electrocardiogram (ECG) classification. In the proposed system, the ECG signal is captured by level crossing (LC) sampling, achieving native temporal coding with single-bit data representation, which is directly fed into an SNN in an event-driven manner. A hardware-aware spatio-temporal backpropagation (STBP) is suggested as the training scheme to adapt to the LC-based data representation and to generate lightweight SNN models. Such a training scheme diminishes the firing rate of the network with little plenty of classification accuracy loss, thus reducing the switching activity of the circuits for low-power operation. A specialized SNN processor is designed with the spike-driven processing flow and hierarchical memory access scheme. Validated with field programmable gate arrays (FPGA) and evaluated in 40 nm CMOS technology for application-specific integrated circuit (ASIC) design, the SNN processor can achieve 98.22% classification accuracy on the MIT-BIH database for 5-category classification, with an energy efficiency of 0.75 μJ/classification.

Topological spin textures such as magnetic skyrmions1-7 are promising candidates as information carriers for next-generation memory-in-logic devices8. Antiferromagnetic spin textures9-12, compared to their ferromagnetic counterparts, innately possess high stability with respect to external disturbance13,14 and high-frequency dynamics15-18 compatible with ultrafast information processing. However, deterministic creation and reconfigurable switching of different antiferromagnetic spin textures have not been realized so far. Here, we demonstrate room-temperature deterministic switching between antiferromagnetic domain walls, Bloch bimerons and Néel bimerons in single-crystal hematite (α-Fe2O3)12,19-22. All three topological states are found to be remarkably stable and fully controllable, as confirmed by 1’000 switching cycles. Their dynamics exhibit distinct resonance frequencies and magnetic field dependences. The switching from domain walls to Bloch bimerons requires only one microwave pulse (100 ns) with ultralow energy consumption (1 nJ). When the dynamic excitation wavevector approaches that of static spin textures, and the power lies beyond a threshold, chaotic magnons23-26 are excited, which subsequently nucleate bimerons and finally lead to a deterministic switching, as unveiled by micromagnetic simulations. The progressive switching from Bloch-type to Néel-type bimerons imitates the weighted sum operation in neuromorphic computing27,28. Thus, our work points to the possibility of using spin textures in antiferromagnets for information processing.

Lithium metal batteries suffer from low cycle life. During discharge, parts of the lithium are not stripped reversibly and remain isolated from the current collector. This isolated lithium is trapped in the insulating remaining solid-electrolyte interphase (SEI) shell and contributes to the capacity loss. However, a fundamental understanding of why isolated lithium forms and how it can be mitigated is lacking. In this article, we perform a combined theoretical and experimental study to understand isolated lithium formation during stripping. We derive a thermodynamic consistent model of lithium dissolution and find that the interaction between lithium and SEI leads to locally preferred stripping and isolated lithium formation. Based on a cryogenic transmission electron microscopy (TEM) setup, we reveal that these local effects are particularly pronounced at kinks of lithium whiskers. We find that lithium stripping can be heterogeneous both on a nanoscale and on a larger scale. Cryo TEM observations confirm our theoretical prediction that isolated lithium occurs less at higher stripping current densities. The origin of isolated lithium lies in local effects, such as heterogeneous SEI, stress fields, or the geometric shape of the deposits. The understanding of lithium (Li) nucleation and growth is important to design better electrodes for high-performance batteries. However, the study of Li nucleation process is still limited because of the lack of imaging tools that can provide information of the entire dynamic process. We developed and used an operando reflection interference microscope (RIM) that enables real-time imaging and tracking the Li nucleation dynamics at a single nanoparticle level. This dynamic and operando imaging platform provides us with critical capabilities to continuously monitor and study the Li nucleation process. We find that the formation of initial Li nuclei is not at the exact same time point, and Li nucleation process shows the properties of both progressive and instantaneous nucleation. In addition, the RIM allows us to track the individual Li nucleus’s growth and extract spatially resolved overpotential map. The nonuniform overpotential map indicates that the localized electrochemical environments substantially influence the Li nucleation.

Germanium–polyamide 6 (Ge-PA6) fibers A, B and C containing germanium particles with sizes of D50 = 0.2, 2 and 7 μm were prepared using the melt spinning method. The effect of germanium particle size on the morphologies, mechanical properties, negative air ion (NAI) release and antimicrobial efficiencies of the two kinds of fibers were compared and analyzed. The NAI release and antimicrobial mechanisms of the Ge-PA6 fibers were also discussed. Results indicated that the germanium particles dispersed well inside the PA6 fiber substrate in the range of 1–3 wt%, and smaller particles help the fiber to obtain better diameter evenness and mechanical performance compared with larger ones. The stronger extranuclear electron activity and electrostatic voltage along the C-axis allow the fiber that contains smaller germanium particles to produce more NAIs compared to that containing larger particles. Some 780 and 1587 ions/m3 of NAIs could be produced by fiber A under the circumstance of 35°C and 40 N extrusion force when the germanium concentration was fixed at 3 wt%. The excellent NAI release property of the Ge-PA6 fiber endows it with inspiring antibacterial activity against S. aureus and C. albicans with a 12 h bacteriostasis rate of more than 90% at human body temperature. These results indicated Ge-PA6 fibers have great application potential in apparel and medical textiles.

Background: There are 30%–40% of patients with dilated cardiomyopathy (DCM) having genetic causes, among which Lamin A and C gene (LMNA) mutation is the second most frequent DCM-related mutation, and Lamin A/C may be involved in the pathogenesis of DCM through the regulation of gene transcription or the direct effect of cell structure. Methods: Echocardiography and electrocardiogram were used to diagnose DCM and arrhythmia in a DCM family. Then, linked mutations on LMNA were screened out by high-throughput sequencing and verified by Sanger sequencing in all research individuals. Meanwhile, Human Genome Variation Society (HGVS) and Integrative Genomics Viewer (IGV) were used to analyse the characteristics of the mutated Lamin A/C protein. Finally, mutated-type and wild-type LMNA plasmid was transfected into AC-16 cardiomyocytes with the form of a lentivirus vector, and its effect on nucleus and actin was studied by immunofluorescence detection. Results In this study, we found a new frame-shifted mutation of LMNA (p.Ser414Alafs*66) linked with another point mutation from a DCM family by using High-throughput sequencing, and this deletion mutation led to a truncation of Lamin A/C. By analysing the clinical characteristics of this DCM family, we found that all DCM patients with arrhythmia were carriers of this co-segregation mutation. In the cytological experiment, we found that the mutated-type transfections showed weaker fluorescent intensities on both actin and cell nucleus. Conclusions: A co-segregation mutation of LMNA (Point mutation chr1 156107548 c.1712 G>A and truncated frame-shifted mutation chr1 156106086 c.1240delA) was found from a DCM family, and this type of mutation could participate in the pathogenesis of DCM by affecting the expression of actin.

We report the experimental observation of the spin-wave moirc edge and cavity modes using Brillouin light scattering spectromicroscopy in a nanostructured magnetic moirc lattice consisting of two twisted triangle antidot lattices based on an yttrium iron garnet thin film. Spin-wave moirc edge modes are detected at an optimal twist angle and with a selective excitation frequency. At a given twist angle, the magnetic field acts as an additional degree of freedom for tuning the chiral behavior of the magnon edge modes. Micromagnetic simulations indicate that the edge modes emerge within the original magnonic band gap and at the intersection between a mini flatband and a propagation magnon branch. Our theoretical estimate for the Berry curvature of the magnon-magnon coupling suggests a nontrivial topology for the chiral edge modes and confirms the key role played by the dipolar interaction. Background The new visceral adiposity index (NVAI) was superior to previous obesity indices in predicting cardiovascular diseases among Asians. Nevertheless, the utility of the NVAI for predicting chronic kidney disease is still unclear. The objective of this research was to explore the relationship between the NVAI and subclinical renal damage (SRD) and to investigate whether the NVAI outperforms other common obesity indices in predicting SRD in the Chinese population. Methods Participants in this cross-sectional study were from the Hanzhong Adolescent Hypertension Cohort. The NVAI and seven other common obesity indices were calculated, including body mass index, waist circumference, lipid accumulation product, visceral adiposity index, Chinese visceral adiposity index, a body shape index and metabolic score for visceral fat. Logistic regression models revealed the association between NVAI and SRD. The odds ratio (OR) and the 95% confidence interval (CI) were calculated to show the association between the two variables. The predictive power of eight obesity indices for SRD was evaluated through the receiver operating characteristic curve and area under the curve (AUC). In addition, the net reclassification index (NRI) and integrated discrimination improvement (IDI) were also applied to compare the incremental predictive value for SRD of different obesity indices. Results The median age of the 2358 subjects was 42.00 years. Across NVAI tertiles, the prevalence of SRD was 7.25%, 11.21%, and 21.60%, respectively. After adjusting for confounders, a high level of NVAI remained a risk factor for SRD. The ORs of the middle and top NVAI tertiles for SRD were 1.920 (95% CI: 1.322, 2.787) and 4.129 (95% CI: 2.750, 6.202), respectively. The AUC of the NVAI was 0.666 (95% CI: 0.647, 0.685), which was significantly larger than the AUC of any of the other obesity indicators. Moreover, the NRI and IDI were significantly improved when NVAI was added to the basic model for predicting SRD. Among eight obesity indices, NVAI had the highest NRI (0.392; 95% CI: 0.280, 0.503), and its IDI (0.021; 95% CI: 0.014, 0.027) was second only to that of the body mass index (0.023; 95% CI: 0.014, 0.032). Conclusions NVAI is independently and positively associated with SRD. Among the eight obesity indices, the NVAI shows the strongest predictive power for SRD in the Chinese population. The NVAI may be useful as an effective warning indicator of chronic kidney disease in Chinese adults.
