The Observing Air–Sea Interactions Strategy (OASIS) is a new United Nations Decade of Ocean Science for Sustainable Development programme working to develop a practical, integrated approach for observing air–sea interactions globally for improved Earth system (including ecosystem) forecasts, CO2 uptake assessments called for by the Paris Agreement, and invaluable surface ocean information for decision makers. Our “Theory of Change” relies upon leveraged multi-disciplinary activities, partnerships, and capacity strengthening. Recommendations from >40 OceanObs’19 community papers and a series of workshops have been consolidated into three interlinked Grand Ideas for creating #1: a globally distributed network of mobile air–sea observing platforms built around an expanded array of long-term time-series stations; #2: a satellite network, with high spatial and temporal resolution, optimized for measuring air–sea fluxes; and #3: improved representation of air–sea coupling in a hierarchy of Earth system models. OASIS activities are organized across five Theme Teams: (1) Observing Network Design & Model Improvement; (2) Partnership & Capacity Strengthening; (3) UN Decade OASIS Actions; (4) Best Practices & Interoperability Experiments; and (5) Findable–Accessible–Interoperable–Reusable (FAIR) models, data, and OASIS products. Stakeholders, including researchers, are actively recruited to participate in Theme Teams to help promote a predicted, safe, clean, healthy, resilient, and productive ocean.

OBJECTIVE To determine whether interventions that slow or prevent the development of type 2 diabetes in those at risk reduce the subsequent prevalence of diabetic retinopathy. RESEARCH DESIGN AND METHODS The Diabetes Prevention Program (DPP) randomized subjects at risk for developing type 2 diabetes because of overweight/obesity and dysglycemia to metformin (MET), intensive lifestyle intervention (ILS), or placebo (PLB) to assess the prevention of diabetes. During the DPP and DPP Outcome Study (DPPOS), we performed fundus photography over time on study participants, regardless of their diabetes status. Fundus photographs were graded using the Early Treatment Diabetic Retinopathy Study grading system, with diabetic retinopathy defined as typical lesions of diabetic retinopathy (microaneurysms, exudates, or hemorrhage, or worse) in either eye. RESULTS Despite reduced progression to diabetes in the ILS and MET groups compared with PLB, there was no difference in the prevalence of diabetic retinopathy between treatment groups after 1, 5, 11, or 16 years of follow-up. No treatment group differences in retinopathy were found within prespecified subgroups (baseline age, sex, race/ethnicity, baseline BMI). In addition, there was no difference in the prevalence of diabetic retinopathy between those exposed to metformin and those not exposed to metformin, regardless of treatment group assignment. CONCLUSIONS Interventions that delay or prevent the onset of type 2 diabetes in overweight/obese subjects with dysglycemia who are at risk for diabetes do not reduce the development of diabetic retinopathy for up to 20 years.

Background: Genetic cancer risk assessment is a multidisciplinary approach that enables the invaluable opportunity to recommend measures aimed at decreasing the risk of recurrence and new cancer diagnoses in individuals with inherited cancer predisposition. However, different challenges may limit the uptake of risk-reducing (RR) strategies among patients in underserved settings. This study describes the implementation of RR surgeries or surveillance studies and related barriers among carriers of breast cancer-associated germline pathogenic variants (PV). Methods: PV carriers were identified at two referral cancer centers in Mexico ≥6 months after receiving test results and genetic counseling. From October 2020 to June 2021, participants were invited to answer a multiple-choice survey assessing their uptake rate of RR measures according to NCCN guidelines by type of PV, as well as the challenges that had hindered this uptake. Descriptive statistics and Chi-squared or Fisher’s exact tests were used for data analysis. Results: In total, 162 carriers (median age: 42.5 years [range 21-73]) answered the survey. Most were women (89%), lived in an urban setting (91%), were married/in domestic partnership (72%), had at least high school education (69%), and were employed (51%). Monthly household income was lower than the minimum wage in 25% of cases and most patients had public (53%) or no (32%) healthcare coverage. Overall, 105 (65%) had a personal history of cancer, most commonly unilateral (77%) or bilateral (13%) breast cancer. PV were found in BRCA1/BRCA2 (75%), CHEK2 (11%), PALB2 (6%), ATM (4%), RAD51C (3%), and less frequently (3%) in TP53, NF1 or PTEN. Carriers underwent at least one of the recommended RR surgeries and surveillance studies in 61% and 69% of cases, respectively. Specifically, 52/124 (42%) of female BRCA1/BRCA2, PALB2, PTEN or TP53 PV carriers had undergone RR mastectomy, while 45/77 (58%) of eligible BRCA1/BRCA2 or RAD51C PV carriers had RR salpingo-oophorectomy. As for surveillance studies, carriers performed all, some and none of the recommended examinations according to their PV in 34%, 35% and 31% of cases, respectively. When an annual mammogram was recommended, 69/109 (63%) underwent the study in the previous year; in the case of breast MRI, only 23/103 (22%) had performed it in the last year. Notably, 132 (81%) carriers reported ≥1 challenge that hindered the uptake of RR strategies (median: 2 [range 0-10]). The main barriers were lack of insurance coverage of the recommended interventions (60%), financial limitations (50%), belief that their physician had not requested the studies or procedures (36%), and fear (25%). Uptake of RR strategies was not associated with carriers’ age, education level, employment status, income, place of residence, or type of mutation (high-risk vs moderate- or low-risk PV). Significant associations are reported in the Table. Conclusion: An important proportion of PV carriers in this resource-constrained setting underwent all or some of the recommended RR strategies, with the most cited uptake challenges being financial barriers and lack of endorsement by their physician. Targeted interventions to reduce out-of-pocket expenses and improve patient-physician communication and patients’ understanding of the implications of carrying a genetic mutation are warranted to enhance the uptake of RR surgeries and surveillance strategies among PV carriers. Supplementary Tables 1-2 from Enhanced Antitumor Activity Induced by Adoptive T-Cell Transfer and Adjunctive Use of the Histone Deacetylase Inhibitor LAQ824

Tumors grow in the presence of antigen-specific T cells, suggesting the existence of intrinsic cancer cell escape mechanisms. We hypothesized that a histone deacetylase (HDAC) inhibitor could sensitize tumor cells to immunotherapy because this class of agents has been reported to increase tumor antigen expression and shift gene expression to a proapoptotic milieu in cancer cells. To test this question, we treated B16 murine melanoma with the combination of the HDAC inhibitor LAQ824 and the adoptive transfer of gp100 melanoma antigen-specific pmel-1 T cells. The combined therapy significantly improved antitumor activity through several mechanisms: (a) increase in MHC and tumor-associated antigen expression by tumor cells; (b) decrease in competing endogenous lymphocytes in recipient mice, resulting in a proliferative advantage for the adoptively transferred cells; and (c) improvement in the functional activity of the adoptively transferred lymphocytes. We confirmed the beneficial effects of this HDAC inhibitor as a sensitizer to immunotherapy in a different model of prophylactic prime-boost vaccination with the melanoma antigen tyrosinase-related protein 2, which also showed a significant improvement in antitumor activity against B16 melanoma. In conclusion, the HDAC inhibitor LAQ824 significantly enhances tumor immunotherapy through effects on target tumor cells as well as improving the antitumor activity of tumor antigen-specific lymphocytes. The UN’s Decade of Ocean Science (in particular SDG 4 and 14) calls for programmes that improve scientific knowledge, develop research capacities and transfer marine technological information and expertise across generations. In South Africa, the Department of Science and Innovation’s (DSI) has already taken a significant step forward in such training measures. The current DSI Global Change Grand Challenge programmecalls for platforms that “attract young researchers and retain them by exciting their interest in aspects of global change, while developing their capacity and professional skills in the relevant fields of investigation”. To meet these challenges in the Ocean Sciences, SEAmester – South Africa’s Floating University and a joint initiative between Government and Universities was started in 2016. The strength of SEAmester is that South African postgraduate students combine theoretical classroom learning with the application of this knowledge through ship-based, and more importantly, hands-on research through the Agulhas System Climate Array (ASCA) programme. Now into its 6th year, SEAmester has already made significant progress in ship-based training with over 210 students from 23 South African universities participating in these training cruises. This paper reports the ULTRACAM discovery of dipolar surface spots in two cool magnetic white dwarfs with Balmer emission lines, while a third system exhibits a single spot, similar to the prototype GD 356. The light curves are modeled with simple, circular, isothermal dark spots, yielding relatively large regions with minimum angular radii of 20 deg. For those stars with two light curve minima, the dual spots are likely observed at high inclination (or colatitude), however, identical and antipodal spots cannot simultaneously reproduce both the distinct minima depths and the phases of the light curve maxima. The amplitudes of the multi-band photometric variability reported here are all several times larger than that observed in the prototype GD 356; nevertheless, all DAHe stars with available data appear to have light curve amplitudes that increase toward the blue in correlated ratios. This behavior is consistent with cool spots that produce higher contrasts at shorter wavelengths, with remarkably similar spectral properties given the diversity of magnetic field strengths and rotation rates. These findings support the interpretation that some magnetic white dwarfs generate intrinsic chromospheres as they cool, and that no external source is responsible for the observed temperature inversion. The eighteenth data release of the Sloan Digital Sky Surveys (SDSS) is the first one for SDSS-V, the fifth generation of the survey. SDSS-V comprises three primary scientific programs, or "Mappers": Milky Way Mapper (MWM), Black Hole Mapper (BHM), and Local Volume Mapper (LVM). This data release contains extensive targeting information for the two multi-object spectroscopy programs (MWM and BHM), including input catalogs and selection functions for their numerous scientific objectives. We describe the production of the targeting databases and their calibration- and scientifically-focused components. DR18 also includes ~25,000 new SDSS spectra and supplemental information for X-ray sources identified by eROSITA in its eFEDS field. We present updates to some of the SDSS software pipelines and preview changes anticipated for DR19. We also describe three value-added catalogs (VACs) based on SDSS-IV data that have been published since DR17, and one VAC based on the SDSS-V data in the eFEDS field., Comment: Accepted to ApJS

Two recently discovered white dwarfs, WDJ041246.84$+$754942.26 and WDJ165335.21$-$100116.33, exhibit H$\alpha$ and H$\beta$ Balmer line emission similar to stars in the emerging DAHe class, yet intriguingly have not been found to have detectable magnetic fields. These white dwarfs are assigned the spectral type DAe. We present detailed follow-up of the two known DAe stars using new time-domain spectroscopic observations and analysis of the latest photometric time-series data from TESS and ZTF. We measure the upper magnetic field strength limit of both stars as $B < 0.05$ MG. The DAe white dwarfs exhibit photometric and spectroscopic variability, where in the case of WDJ041246.84$+$754942.26 the strength of the H$\alpha$ and H$\beta$ emission cores varies in anti-phase with its photometric variability over the spin period, which is the same phase relationship seen in DAHe stars. The DAe white dwarfs closely cluster in one region of the Gaia Hertzsprung-Russell diagram together with the DAHe stars. We discuss current theories on non-magnetic and magnetic mechanisms which could explain the characteristics observed in DAe white dwarfs, but additional data are required to unambiguously determine the origin of these stars., Comment: 20 pages, 16 figures. (Abridged) We present an exploration of the internal rotation of GD 278, the first pulsating extremely low-mass white dwarf that shows rotational splittings within its periodogram. We assess the theoretical frequency splittings expected for different rotation profiles and compare them to the observed frequency splittings of GD 278. To this aim, we employ an asteroseismological model representative of the pulsations of this star, obtained by using the LPCODE stellar evolution code. We also derive a rotation profile that results from detailed evolutionary calculations carried out with the MESA stellar evolution code and use it to infer the expected theoretical frequency splittings. We found that the best-fitting solution when assuming linear profiles for the rotation of GD 278 leads to values of the angular velocity at the surface and the center that are only slightly differential, and still compatible with rigid rotation. The values of the angular velocity at the surface and the center for the simple linear rotation profiles and for the rotation profile derived from evolutionary calculations are in very good agreement. Also, the resulting theoretical frequency splittings are compatible with the observed frequency splittings, in general, for both cases. The results obtained from the different approaches followed in this work to derive the internal rotation of GD 278 agree. The fact that they were obtained employing two independent stellar evolution codes gives robustness to our results. Our results suggest only a marginally differential behavior for the internal rotation in GD 278, and considering the uncertainties involved, very compatible with the rigid case, as has been observed previously for white dwarfs and pre-white dwarfs. The rotation periods derived for this star are also in line with the values determined asteroseismologically for white dwarfs and pre-white dwarfs in general., Comment: 9 pages, 6 figures. Segundo por ser un docente ejemplar, lleno de amor, convicción y pasión por compartir los sabores en una universidad que ha sido defensoras de la educación de calidad y calidez, por ser un dirigentes estudiantil y representante de los intereses de la comunidad universitaria. Por esas cualidades y por su afán por acabar con la tiranía somocista entregó su vida por las causas y que sirva este humilde esfuerzo para motivar futuros trabajos que resalten en una obra mayor los valores del profesor Bonilla, el revolucionario que debe ser olvidado. Honor y gloria al mártir de nuestra UNAN-Managua.

Objective: Differences in type 2 diabetes phenotype by age are described, but it is not known whether these differences are seen in a more uniformly defined adult population at a common early stage of care. We characterize age-related clinical and metabolic characteristics of adults with type 2 diabetes on metformin monotherapy, prior to treatment intensification. Research Design and Methods: The Glycemia Reduction Approaches in Diabetes: A Comparative Effectiveness (GRADE) Study enrolled participants with type 2 diabetes duration ≥65 years) using ANOVA and Pearson’s chi-squared tests.. Results: Within the GRADE cohort (n=5,047), we observed significant differences by age with younger adults having greater racial diversity, fewer medications for common comorbidities, lower prevalence of CVD, higher weight and BMI, more pronounced hyperglycemia and diabetic dyslipidemia, with metabolic profile indicating lower insulin sensitivity (1/fasting insulin, HOMA-2S, Matsuda index) and inadequate β cell response (oral disposition index) (p < 0.05 across age categories). Conclusions: Clinical and metabolic characteristics of type 2 diabetes differ by age within the GRADE cohort. Younger adults exhibit more prominent obesity-related characteristics, including higher obesity levels, lower insulin sensitivity and beta cell compensation. Given the increasing burden of type 2 diabetes and complications, particularly among younger populations, these age-related distinctions may inform risk factor management approaches and treatment priorities. Further study will determine whether age-related differences impact response to therapy.

Islet autoimmunity may contribute to β-cell dysfunction in type 2 diabetes (T2D). Its prevalence and clinical significance have not been rigorously determined. In this ancillary study to the Glycemia Reduction Approaches in Diabetes-A Comparative Effectiveness (GRADE) Study, we investigated the prevalence of cellular and humoral islet autoimmunity in patients with T2D duration 4·0±3·0 y, HbA1c 7·5±0·5% on metformin alone. We measured T cell autoreactivity against islet proteins, islet autoantibodies against GAD65, IA2, ZnT8, and β-cell function. Cellular islet autoimmunity was present in 41·3%, humoral islet autoimmunity in 13·5%, and both in 5·3%. β-cell function calculated as iAUC-CG and ∆C-peptide(0– 30)/∆glucose(0–30) from an oral glucose tolerance test was lower among T cell-positives (T+) than T cell-negatives (T-) using two different adjustments for insulin sensitivity (iAUC-CG: 13·2% [95% CI 0·3, 24·4%] or 11·4% [95% CI 0·4, 21·2%] lower; ∆C-peptide(0–30)/∆glucose(0–30)) 19% [95% CI 3·1, 32·3%] or 17·7% [95% CI 2·6, 30·5%] lower). T+ patients had 17% higher HbA1c (95% CI 0·07, 0·28) and 7·7 mg/dL higher fasting plasma glucose levels (95% CI 0·2,15·3) than T- patients. We conclude that islet autoimmunity is much more prevalent in T2D patients than previously reported. T cell-mediated autoimmunity is associated with diminished β-cell function and worse glycemic control.

Antihyperglycemic agents are significant contributors to adverse drug events, responsible for emergency department visits, hospitalizations, and death. Nationally, the rate of serious hypoglycemic events associated with these agents remains high despite widespread efforts to improve drug safety. Transitions of care between healthcare settings can lead to communication challenges between care professionals and increase the risk of adverse drug events. System-based improvements are needed to assure the safe transitions for patients with diabetes who are on antihyperglycemic agents. The objective of this study was to develop a consensus list of requisite elements that should be communicated between care settings during transitions of patients who are prescribed antihyperglycemic agents.The Island Peer Review Organization (IPRO) Hypoglycemia Coalition identified suboptimal transitions of care as a barrier to improving patient safety and quality of diabetes care. The Coalition formed a multidisciplinary Task Force with experts in the field of diabetes care. The Task Force created a draft list of requisite communication elements through literature review and deliberation on monthly conference calls. A blinded iterative Delphi process was subsequently performed to generate a consensus list of requisite communication elements that participating experts agreed were necessary to safely and effectively assume the management of patients with diabetes upon care transitions.The Task Force completed a series of four iterative polls from September 2015 to August 2016, resulting in a final list of 22 requisite communication elements (the Diabetes Management Discharge Communication List), with the elements conceptually categorized into three domains: diagnosis and treatment, factors affecting glycemic control or patient risk, and patient self-management.The Diabetes Management Discharge Communication List provides an initial framework for the development of diabetes-specific resources to improve clinical communication between care settings.

The {\sl TESS} space mission has recently demonstrated its great potential to discover new pulsating white dwarf and pre-white dwarf stars, and to detect periodicities with high precision in already known white-dwarf pulsators. We report the discovery of two new pulsating He-rich atmosphere white dwarfs (DBVs) and present a detailed asteroseismological analysis of three already known DBV stars employing observations collected by the {\sl TESS} mission along with ground-based data. We extracted frequencies from the {\sl TESS} light curves of these DBV stars using a standard pre-whitening procedure to derive the potential pulsation frequencies. All the oscillation frequencies that we found are associated with $g$-mode pulsations with periods spanning from $\sim 190$ s to $\sim 936$ s. We find hints of rotation from frequency triplets in some of the targets, including the two new DBVs. For three targets, we find constant period spacings, which allowed us to infer their stellar masses and constrain the harmonic degree $\ell$ of the modes. We also performed period-to-period fit analyses and found an asteroseismological model for three targets, with stellar masses generally compatible with the spectroscopic masses. Obtaining seismological models allowed us to estimate the seismological distances and compare them with the precise astrometric distances measured with {\it Gaia}. We find a good agreement between the seismic and the astrometric distances for three stars (PG~1351+489, EC~20058$-$5234, and EC~04207$-$4748), although for the other two stars (WD~J152738.4$-$50207 and WD~1708$-$871), the discrepancies are substantial. The high-quality data from the {\sl TESS} mission continue to provide important clues to determine the internal structure of pulsating pre-white dwarf and white dwarf stars through the tools of asteroseismology., Comment: 22 pages, 27 figures, 21 tables. T+ patients had 17% higher HbA1c (95% CI 0.07, 0.28) and 7.7 mg/dL higher fasting plasma glucose levels (95% CI 0.2, 15.3) than T− patients. We conclude that islet autoimmunity is much more prevalent in patients with T2D than previously reported. T-cell–mediated autoimmunity is associated with diminished β-cell function and worse glycemic control.

The COVID-19 pandemic massively changed the context and feasibility of developmental research. This new reality as well as considerations, e.g., about sample diversity and naturalistic settings for developmental research, indicate the need for solutions for online studies. In this article, we present e-Babylab, an open-source browser-based tool for unmoderated online studies specifically targeted at studying young children and babies. e-Babylab offers an intuitive graphical user interface for study creation and management of studies, users, participant data, and stimulus material with no programming skills required. Various kinds of audiovisual media can be presented as stimuli and possible measures include webcam recordings, audio recordings, key presses, mouse-click/touch coordinates, and reaction times. Information pages, consent forms, and participant forms are all customizable. e-Babylab was used with a variety of measures and paradigms in 14 studies with children aged 12 months to 8 years (n = 1516). We briefly summarize some results of these studies to demonstrate data quality and that participants’ engagement and the overall results are comparable in lab and online settings. We then present in more detail one replication of an established preferential-looking paradigm with gaze directions manually coded from the webcam captures. Finally, we discuss useful tips for using e-Babylab and present plans for upgrades.

We present the discovery of J22564-5910, a new type of hot subdwarf (sdB) which shows evidence of gas present in the system and has shallow, multi-peaked hydrogen and helium lines which vary in shape over time. All observational evidence points towards J22564-5910 being observed very shortly after the merger phase that formed it. Using high-resolution, high signal-to-noise spectroscopy, combined with multi-band photometry, Gaia astrometry, and TESS light curves, we aim to interpret these unusual spectral features. The photometry, spectra and light curves are all analyzed, and their results are combined in order to support our interpretation of the observations: the likely presence of a magnetic field combined with gas features around the sdB. Based on the triple-peaked H lines, the magnetic field strength is estimated and, by using the shellspec code, qualitative models of gas configurations are fitted to the observations. All observations can either be explained by a magnetic field of ~650 kG which enables the formation of a centrifugal magnetosphere, or a non-magnetic hot subdwarf surrounded by a circumstellar gas disk/torus. Both scenarios are not mutually exclusive and both can be explained by a recent merger. J22564-5910 is the first object of its kind. It is a rapidly spinning sdB with gas still present in the system. It is the first post-merger star observed this early after the merger event, and as such is very valuable system to test merger theories. If the magnetic field can be confirmed, it is not only the first magnetic sdB, but it hosts the strongest magnetic field ever found in a pre-white dwarf object. Background: Islet autoimmunity may contribute to beta-cell dysfunction in type 2 diabetes (T2D). Its prevalence and clinical significance have not been rigorously determined. We investigated the prevalence of cellular and humoral islet autoimmunity in T2D and their relation to beta-cell function. Methods: The study was nested in the Glycemia Reduction Approaches in Diabetes – A Comparative Effectiveness (GRADE) study. Participants included 419 T2D patients (age 57·4±10·3 y [mean±SD], body mass index 33·6±6·2 kg/m2, diabetes duration 4·0±3·0 y, HbA1c 7·5±0·5%). We measured humoral (autoantibodies against GAD65, IA2, ZnT8) and cellular (T-cell autoreactivity against islet antigens) autoimmunity, beta-cell function (ratio of incremental area under the curve of C-peptide to that of glucose from 0–120 min of an oral glucose tolerance test [iAUC-CG]), and ratio of C-peptide to glucose from 0–30 min (∆C-peptide(0–30)/∆glucose(0–30)), and glycemic parameters. Findings: Cellular islet autoimmunity was present in 41·3%, humoral islet autoimmunity in 13·5%, and both in 5·3%. iAUC-CG and ∆C-peptide(0–30)/∆glucose(0–30) were lower among T-cell-positives than -negatives using two different adjustments for insulin sensitivity (iAUC-CG: 13·2% [95% CI 0·3, 24·4%] or 11·4% [95% CI 0·4, 21·2%] lower; ∆C-peptide(0–30)/∆glucose(0–30)) 19% [95% CI 3·1, 32·3%] or 17·7% [95% CI 2·6, 30·5%] lower). T-cell-positives had 0·17% higher HbA1c (95% CI 0·07,0·28) and 7·7 mg/dL higher fasting plasma glucose levels (95% CI 0·2,15·3) than T-cell-negatives. Interpretation: Islet autoimmunity is highly prevalent in patients with T2D. T-cell-mediated autoimmunity is associated with diminished beta-cell function and worse glycemic control. Research has shown that young children are selective in whom they trust, for example, learning selectively from more reliable sources. To explain what cognitive foundations this capacity may build upon, a recent dual-process account proposed that children recruit different kinds of cognitive strategies, including a) simple heuristics, e.g., favoring protagonists that are overall better or score high on a salient, accessible characteristic, and b) more systematic strategies, e.g., taking into account the protagonists’ individual properties. The present studies investigated the prediction that the more systematic processes require cognitive resources and develop with age. Children and adults were familiarized with two protagonists: The strong-and-shy protagonist scored high on a highly accessible trait (strength), whereas the weak-and-extraverted protagonist scored high on a less accessible trait (extraversion). In test trials, participants chose between these for strength- and extraversion-related tasks. The results were consistent with the prediction of the dual-process account: Older children, and adults under normal conditions, showed a pattern of systematic reasoning, selecting the protagonists with the relevant trait for a given task. Yet, younger children, and adults whose cognitive capacities were burdened with a secondary task, showed a pattern of heuristic reasoning, selecting the strong-and-shy protagonist for all kinds of tasks.

Identifying planets around O-type and B-type stars is inherently difficult; the most massive known planet host has a mass of only about $3M_{\odot}$. However, planetary systems which survive the transformation of their host stars into white dwarfs can be detected via photospheric trace metals, circumstellar dusty and gaseous discs, and transits of planetary debris crossing our line-of-sight. These signatures offer the potential to explore the efficiency of planet formation for host stars with masses up to the core-collapse boundary at $\approx 8M_{\odot}$, a mass regime rarely investigated in planet formation theory. Here, we establish limits on where both major and minor planets must reside around $\approx 6M_{\odot}-8M_{\odot}$ stars in order to survive into the white dwarf phase. For this mass range, we find that intact terrestrial or giant planets need to leave the main sequence beyond approximate minimum star-planet separations of respectively about 3 and 6 au. In these systems, rubble pile minor planets of radii 10, 1.0, and 0.1 km would have been shorn apart by giant branch radiative YORP spin-up if they formed and remained within, respectively, tens, hundreds and thousands of au. These boundary values would help distinguish the nature of the progenitor of metal-pollution in white dwarf atmospheres. We find that planet formation around the highest mass white dwarf progenitors may be feasible, and hence encourage both dedicated planet formation investigations for these systems and spectroscopic analyses of the highest mass white dwarfs., Comment: Accepted for publication in MNRAS

AIMS: We evaluated differences in participants with type 2 diabetes (T2DM) enrolled in the GRADE study at VA vs non-VA sites, focusing on cardiovascular risk factors and rates of diabetes care target achievements. Moderate alcohol consumption has been associated with a lower risk of coronary artery disease (CAD) in the general population but has not been well studied in US veterans. We obtained self-reported alcohol consumption from Million Veteran Program participants. Using electronic health records, CAD events were defined as 1 inpatient or 2 outpatient diagnosis codes for CAD, or 1 code for a coronary procedure. We excluded participants with prevalent CAD (n = 69,995) or incomplete alcohol information (n = 8,449). We used a Cox proportional hazards model to estimate hazard ratios and 95% confidence intervals for CAD, adjusting for age, gender, body mass index, race, smoking, education, and exercise. Among 156,728 participants, the mean age was 65.3 years (standard deviation = 12.1) and 91% were men. There were 6,153 CAD events during a mean follow-up of 2.9 years. Adjusted hazard ratios (95% confidence intervals) for CAD were 1.00 (reference), 1.02 (0.92 to 1.13), 0.83 (0.74 to 0.93), 0.77 (0.67 to 0.87), 0.71 (0.62 to 0.81), 0.62 (0.51 to 0.76), 0.58 (0.46 to 0.74), and 0.95 (0.85 to 1.06) for categories of never drinker; former drinker; current drinkers of ≤0.5 drink/day, >0.5 to 1 drink/day, >1 to 2 drinks/day, >2 to 3 drinks/day, and >3 to 4 drinks/day; and heavy drinkers (>4 drinks/day) or alcohol use disorder, respectively. For a fixed amount of ethanol, intake at ≥3 days/week was associated with lower CAD risk compared with ≤1 day/week. Beverage preference (beer, wine, or liquor) did not influence the alcohol-CAD relation. Our data show a lower risk of CAD with light-to-moderate alcohol consumption among US veterans, and drinking frequency may provide a further reduction in risk.

OBJECTIVE GRADE (Glycemia Reduction Approaches in Diabetes: A Comparative Effectiveness Study) is a 36-center unmasked, parallel treatment group, randomized controlled trial evaluating four diabetes medications added to metformin in people with type 2 diabetes (T2DM). We report baseline characteristics and compare GRADE participants to a National Health and Nutrition Examination Survey (NHANES) cohort. RESEARCH DESIGN AND METHODS Participants were age ≥30 years at the time of diagnosis, with duration of T2DM RESULTS At baseline, GRADE’s 5,047 randomized participants were 57.2 ± 10.0 years of age, 63.6% male, with racial/ethnic breakdown of 65.7% white, 19.8% African American, 3.6% Asian, 2.7% Native American, 7.6% other or unknown, and 18.4% Hispanic/Latino. Duration of diabetes was 4.2 ± 2.8 years, with mean HbA1c of 7.5 ± 0.5% (58 ± 5.3 mmol/mol), BMI of 34.3 ± 6.8 kg/m2, and metformin dose of 1,944 ± 204 mg/day. Among the cohort, 67% reported a history of hypertension, 72% a history of hyperlipidemia, and 6.5% a history of heart attack or stroke. Applying GRADE inclusion criteria to NHANES indicates enrollment of a representative cohort with T2DM on metformin monotherapy (NHANES cohort average age, 57.9 years; mean HbA1c, 7.4% [57 mmol/mol]; BMI, 33.2 kg/m2; duration, 4.2 ± 2.5 years; and 7.2% with a history of cardiovascular disease). CONCLUSIONS The GRADE cohort represents patients with T2DM treated with metformin requiring a second diabetes medication. GRADE will inform decisions about the clinical effectiveness of the addition of four classes of diabetes medications to metformin.

Ultra-compact binaries (UCBs) are systems containing compact or degenerate stars with orbital periods less than one hour. Tens of millions of UCBs are predicted to exist within theGalaxy emitting gravitational waves (GWs) at mHz frequencies. Combining GW searches with electromagnetic (EM) surveys like Gaia and LSST will yield a comprehensive, multimessenger catalog of UCBs in the galaxy. Joint EM and GW observations enable measurements of masses, radii, and orbital dynamics far beyond what can be achieved by independent EM or GW studies. IntroductionSex hormone binding globulin (SHBG) levels are reported to be inversely associated with diabetes risk. It is unknown whether diabetes prevention interventions increase SHBG and whether resultant changes in SHBG affect diabetes risk. The purpose of this analysis was to determine whether intensive lifestyle intervention (ILS) or metformin changed circulating SHBG and if resultant changes influenced diabetes risk in the Diabetes Prevention Program (DPP).Research design and methodsThis is a secondary analysis from the DPP (1996–2001), a randomized trial of ILS or metformin versus placebo on diabetes risk over a mean follow-up of 3.2 years. The DPP was conducted across 27 academic study centers in the USA. Men, premenopausal and postmenopausal women without hormone use in the DPP were evaluated. The DPP included overweight/obese persons with elevated fasting glucose and impaired glucose tolerance. Main outcomes measures were changes in SHBG levels at 1 year and risk of diabetes over 3 years.ResultsILS resulted in significantly higher increases (postmenopausal women: pConclusionsLifestyle intervention may be associated with favorable changes in circulating SHBG, which is largely due to changes in adiposity. Background While adherence to healthful dietary patterns has been associated with a lower risk of coronary artery disease (CAD) in the general population, limited data are available among US veterans. We tested the hypothesis that adherence to Dietary Approach to Stop Hypertension (DASH) food pattern is associated with a lower risk of developing CAD among veterans. Methods and Results We analyzed data on 153 802 participants of the Million Veteran Program enrolled between 2011 and 2016. Information on dietary habits was obtained using a food frequency questionnaire at enrollment. We used electronic health records to assess the development of CAD during follow‐up. Of the 153 802 veterans who provided information on diet and were free of CAD at baseline, the mean age was 64.0 (SD=11.8) years and 90.4% were men. During a mean follow‐up of 2.8 years, 5451 CAD cases occurred. The crude incidence rate of CAD was 14.0, 13.1, 12.6, 12.3, and 11.1 cases per 1000 person‐years across consecutive quintiles of Dietary Approach to Stop Hypertension score. Hazard ratios (95% confidence interval) for CAD were 1.0 (ref), 0.91 (0.84–0.99), 0.87 (0.80–0.95), 0.86 (0.79–0.94), and 0.80 (0.73–0.87) from the lowest to highest quintile of Dietary Approach to Stop Hypertension score controlling for age, sex, body mass index, race, smoking, exercise, alcohol intake, and statin use ( P linear trend, Conclusions Our data are consistent with an inverse association between Dietary Approach to Stop Hypertension diet score and incidence of CAD among US veterans.

The 21st European Workshop on White Dwarfs was held in Austin, TX from July 23rd to 27th of 2018, The Kepler K2 mission has discovered a significant population of white dwarf stars that exhibit photometric variability due to surface inhomogeneities likely related to magnetism. These “spotted” white dwarfs present not only in temperature regimes where we expect convection to dominate white dwarf photospheres, but also where radiation should dominate. We present an exploration of spotted white dwarfs as a function of various physical characteristics, including temperature, magnetic field strength, and rotational period, in order to better understand the origins of these photometric variations.

Stars are stretched by tidal interactions in tight binaries, and changes to their projected areas introduce photometric variations twice per orbit. Hermes et al. (2014, ApJ, 792, 39) utilized measurements of these ellipsoidal variations to constrain the radii of low-mass white dwarfs in eight single-lined spectroscopic binaries. We refine this method here, using Monte Carlo simulations to improve constraints on many orbital and stellar properties of binary systems that exhibit ellipsoidal variations. We analyze the recently discovered tidally distorted white dwarf binary system SDSS J1054-2121 in detail, and also revisit the Hermes et al. (2014) sample. Disagreements in some cases between the observations, ellipsoidal variation model, and Gaia radius constraints suggest that extrinsic errors are present, likely in the surface gravities determined through model atmosphere fits to stellar spectra., Comment: 5 pages, 4 figures. Background:Acne vulgaris is a disease of the pilosebaceous unit characterized by increased sebum production, hyperkeratinization, and immune responses to Propionibacterium acnes (PA). Here, we explore a possible mechanism by which a lipid receptor, G2A, regulates immune responses to a commensal bacterium. Objective:To elucidate the inflammatory properties of G2A in monocytes in response to PA stimulation. Furthermore, our study sought to investigate pathways by which lipids modulate immune responses in response to PA. Methods:Our studies focused on monocytes collected from human peripheral blood mononuclear cells, the monocytic cell line THP-1, and a lab strain of PA. Our studies involved the use of enzyme-linked immunosorbent, Western blot, reverse transcription polymerase chain reaction, small interfering RNA (siRNA), and microarray analysis of human acne lesions in the measurements of inflammatory markers. Results:G2A gene expression is higher in acne lesions compared to normal skin and is inducible by the acne therapeutic, 13-cis-retinoic acid. In vitro, PA induces both the Toll-like receptor 2-dependent expression of G2A as well as the production of the G2A ligand, 9-hydroxyoctadecadienoic acid, from human monocytes. G2A gene knockdown through siRNA enhances PA stimulation of interleukin (IL)-6, IL-8, and IL-1β possibly through increased activation of the ERK1/2 MAP kinase and nuclear factor kappa B p65 pathways. Conclusion:G2A may play a role in quelling inflammatory cytokine response to PA, revealing G2A as a potential attenuator of inflammatory response in a disease associated with a commensal bacterium.

Audience research, this paper suggests, is an excellent field to test the claims of Media Studies 2.0. Moreover, 2.0 claims are a good means to review qualitative audience research itself too. Working from a broad strokes analysis of the theory, politics and method of interpretative research with audiences, it is argued that the new media ecology demands new roles of researchers, and an open approach to 'audiencehood' as practice and innovative research method. The paper ends with a case study of the co-creation project of a research team and a Moroccan-Dutch Internet community-writing team working together on an Internet telenovela.

Defining the specific role of nitric oxide (NO) in the regulation of the immune response against cancer is not a simple task. Despite of being extensively studied, NO, reactive nitrogen species (RNS) and reactive oxygen species (ROS) still maintain their reputation of “double-edge-swords”. However, by examining key issues related to their sources, concentrations and chemical nature and, their locations and neighboring molecules that potentially will be reacting with them, we will have a more precise interpretation of the functional aspects of NO and related RNS in the context of the immune response to tumor cells and pathogenesis of cancer. Variations in the local cellular concentration of the same reactive intermediates induce different outcomes of the immune response. NO and related reactive species trigger defined signal transduction pathways in cancer, and immune-related cells in a concentration-dependent manner. NO bioavailability and NO-dependent responses are strictly functions of the reactivity of ROS with NO-forming RNS. In this chapter, we will examine the basic biology of NO and related species in the context of the immune response to cancer in both their potential role in the pathogenesis of malignancies and also in the control and modulation of the immune response against tumor cells. We will discuss the potential use of NO and related species in the induction of specific anti-cancer activity by the immune system and the modulation of resistance or tolerogenic factors derived from the protective mechanism acquired by the tumor cells in order to evade the anti-tumor immune response .

Acne vulgaris is the most common skin disorder affecting millions of people worldwide and inflammation resulting from the immune response targeting Propionibacterium acnes has a significant role in its pathogenesis. In this study, we have demonstrated that P. acnes is a potent inducer of T helper 17 (Th17) and Th1, but not Th2 responses in human peripheral blood mononuclear cells (PBMCs). P. acnes stimulated expression of key Th17-related genes, including IL-17A, RORα, RORc, IL-17RA, and IL-17RC, and triggered IL-17 secretion from CD4(+), but not from CD8(+) T cells. Supernatants from P. acnes-stimulated PBMCs were sufficient to promote the differentiation of naive CD4(+)CD45RA T cells into Th17 cells. Furthermore, we found that the combination of IL-1β, IL-6, and transforming growth factor-β-neutralizing antibodies completely inhibited P. acnes-induced IL-17 production. Importantly, we showed that IL-17-expressing cells were present in skin biopsies from acne patients but not from normal donors. Finally, vitamin A (all-trans retinoic acid) and vitamin D (1,25-dihydroxyvitamin D3) inhibited P. acnes-induced Th17 differentiation. Together, our data demonstrate that IL-17 is induced by P. acnes and expressed in acne lesions and that both vitamin A and D could be effective tools to modulate Th17-mediated diseases such as acne.

Introduction: Cardiovascular (CV) disease has remained the leading cause of morbidity and mortality worldwide. The American Heart Association (AHA) recently set goals for CV health promotion and disease reduction. There are limited CVD data available to plan prevention strategies in developing countries. Aim: To assess the effect age and gender on the AHA overall CV health score in the Venezuelan population. Methods: In a population-based study in Zulia State, Venezuela, we evaluated 3108 Latino adult men and women on the prevalence of ideal CV health behaviors (nonsmoking, achieving normal weight, enough physical activity, and a healthy diet) and ideal CV health factors (total cholesterol Results: Most (75.7%) of the population had intermediate CV scores (between 3 and 5) while in 10.8% had poor scores (5). Large portions of the population need improvements on diet (97.9%), physical activity (80.9%), weight management (60.2%) and blood pressure (59.6%), while some need interventions to reduce hypercholesterolemia (26.9%), hyperglycemia (23.8%), and smoking (20.8%). The global cardiovascular score significantly decreases with age (p Conclusions: CV health promotion strategies are needed in the majority (∼86%) of the Latino population in Zulia, Venezuela. Prevention strategies focusing on weight reduction through healthy diet and increased physical activity may have significant impact improving overall CV health and reducing obesity related comorbidities, such as hypertension and diabetes, in this population.

To determine the prevalence of hypovitaminosis D (serum 25-hydroxyvitamin D32 ng/mL; HVD) in a population of elderly veterans and conduct a preliminary assessment of the efficacy of supplementation with cholecalciferol in correcting HVD.Randomized, double-blind, placebo-controlled clinical trial.Geriatric clinic at the Bruce W. Carter Veterans Affairs Medical Center, Miami, Florida.Veterans aged 70 and older.Oral cholecalciferol 2,000 IU daily or placebo for 6 months.Serum calcium, 25-hydroxyvitamin D, parathyroid hormone, and 24-hour urinary calcium.Of the 34 participants who completed the study, 62% had HVD at baseline. In the treatment group, mean serum 25-hydroxyvitamin D level rose from 28.4±7.9 ng/mL at baseline to 42.7±10.5 ng/mL at the end of the trial, but levels remained less than 32 ng/mL in three of 17 (18%) of the participants. In the placebo group, the baseline level of 27.7±8.3 ng/mL remained unchanged (28.8±8.7 ng/mL). Supplementation did not alter serum or urinary calcium levels and did not result in any adverse events.These initial observations suggest that, in older veterans, cholecalciferol 2,000 IU daily for 6 months is generally safe and corrects HVD in most but not all individuals.

We identify SDSS J010657.39-100003.3 (hereafter J0106-1000) as the shortest period detached binary white dwarf (WD) system currently known. We targeted J0106-1000 as part of our radial velocity program to search for companions around known extremely low-mass (ELM, ~ 0.2 Msol) WDs using the 6.5m MMT. We detect peak-to-peak radial velocity variations of 740 km/s with an orbital period of 39.1 min. The mass function and optical photometry rule out a main-sequence star companion. Follow-up high-speed photometric observations obtained at the McDonald 2.1m telescope reveal ellipsoidal variations from the distorted primary but no eclipses. This is the first example of a tidally distorted WD. Modeling the lightcurve, we constrain the inclination angle of the system to be 67 +- 13 deg. J0106-1000 contains a pair of WDs (0.17 Msol primary + 0.43 Msol invisible secondary) at a separation of 0.32 Rsol. The two WDs will merge in 37 Myr and most likely form a core He-burning single subdwarf star. J0106-1000 is the shortest timescale merger system currently known. The gravitational wave strain from J0106-1000 is at the detection limit of the Laser Interferometer Space Antenna (LISA). However, accurate ephemeris and orbital period measurements may enable LISA to detect J0106-1000 above the Galactic background noise., Comment: MNRAS Letters, in press

We report the discovery of a new detached, double white dwarf system with an orbital period of 39.8 min. We targeted SDSS J163030.58+423305.8 (hereafter J1630) as part of our radial velocity program to search for companions around low-mass white dwarfs using the 6.5m MMT. We detect peak-to-peak radial velocity variations of 576 km/s. The mass function and optical photometry rule out main-sequence companions. In addition, no milli-second pulsar companions are detected in radio observations. Thus the invisible companion is most likely another white dwarf. Unlike the other 39 min binary SDSS J010657.39-100003.3, follow-up high speed photometric observations of J1630 obtained at the McDonald 2.1m telescope do not show significant ellipsoidal variations, indicating a higher primary mass and smaller radius. The absence of eclipses constrain the inclination angle to 0.3 Msun invisible secondary, at a separation of >0.32 Rsun. The two white dwarfs will merge in less than 31 Myr. Depending on the core composition of the companion, the merger will form either a single core-He burning subdwarf star or a rapidly rotating massive white dwarf.
