Introduction: Kidney cancer is one of the most common and lethal urological malignancies. Discovering a biomarker that can predict prognosis and potential drug treatment sensitivity is necessary for managing patients with kidney cancer. SUMOylation is a type of posttranslational modification that could impact many tumor-related pathways through the mediation of SUMOylation substrates. In addition, enzymes that participate in the process of SUMOylation can also influence tumorigenesis and development.Methods: We analyzed the clinical and molecular data which were obtanied from three databases, The Cancer Genome Atlas (TCGA), the National Cancer Institute’s Clinical Proteomic Tumor Analysis Consortium (CPTAC), and ArrayExpress.Results: Through analysis of differentially expressed RNA based on the total TCGA-KIRC cohort, it was found that 29 SUMOylation genes were abnormally expressed, of which 17 genes were upregulated and 12 genes were downregulated in kidney cancer tissues. A SUMOylation risk model was built based on the discovery TCGA cohort and then validated successfully in the validation TCGA cohort, total TCGA cohort, CPTAC cohort, and E-TMAB-1980 cohort. Furthermore, the SUMOylation risk score was analyzed as an independent risk factor in all five cohorts, and a nomogram was constructed. Tumor tissues in different SUMOylation risk groups showed different immune statuses and varying sensitivity to the targeted drug treatment.Discussion: In conclusion, we examined the RNA expression status of SUMOylation genes in kidney cancer tissues and developed and validated a prognostic model for predicting kidney cancer outcomes using three databases and five cohorts. Furthermore, the SUMOylation model can serve as a biomarker for selecting appropriate therapeutic drugs for kidney cancer patients based on their RNA expression.

Background: N6-methyladenosine (m6A) RNA modification has been shown to act fundamental roles in cancer progression. However, the underlying mechanisms by which m6A modification of circRNAs contributes to gastric cancer (GC) remain unknown. Methods: METTL14-mediated m6A modification of circRNAs was identified by an m6A-circRNA epi-transcriptomic microarray and verified by methylated RNA immunoprecipitation (MeRIP) and m6A dot blot. Cell bio-behaviors were measured by molecular oncology experiments in vitro and in vivo. The mechanisms underlying gastric tumorigenesis were conducted by RIP, luciferase gene report, RT-qPCR. and Western blot analyses. The prognostic significance was analyzed by FISH and TCGA dataset. Results: We found that hsa_circ_0030632 (circUGGT2) was a predominant m6A target of METTL14, and METTL14 knockdown (KD) reduced circUGGT2 m6A levels but increased its mRNA levels. circUGGT2 KD impaired cell growth and metastasis in vitro and in vivo, but circUGGT2 overexpression prompted these effects. Furthermore, circUGGT2 was validated to sponge miR-186-3p and upregulate MAP3K9 and could abolish METTL14-caused miR-186-3p upregulation and MAP3K9 downregulation in GC cells. circUGGT2 negatively correlated with miR-186-3p expression and harbored a poor prognosis in patients with GC. Conclusion: Our findings unveil that METTL14-dependent m6A modification of circUGGT2 promotes GC progression by sponging miR-186-3p and upregulating MAP3K9.

The efficacy of the first-line treatment for hypopharyngeal carcinoma (HPC), a predominantly male cancer, at advanced stage is only about 50% without reliable molecular indicators for its prognosis. In this study, HPC biopsy samples collected before and after the first-line treatment are classified into different groups according to treatment responses. We analyze the changes of HPC tumor microenvironment (TME) at the single-cell level in response to the treatment and identify three gene modules associated with advanced HPC prognosis. We estimate cell constitutions based on bulk RNA-seq of our HPC samples and build a binary classifier model based on non-malignant cell subtype abundance in TME, which can be used to accurately identify treatment-resistant advanced HPC patients in time and enlarge the possibility to preserve their laryngeal function. In summary, we provide a useful approach to identify gene modules and a classifier model as reliable indicators to predict treatment responses in HPC.

Understanding the complex pathogenesis in myocardial ischemia/reperfusion (I/R) injury (IRI) is an urgent problem in clinical trials. Increasing pieces of evidence have suggested that miRNAs are involved in the occurrence and development of heart diseases by regulating mitochondria-related gene expression. Mitochondria have been acknowledged as the key triggers of cardiac I/R injury. However, the potential impact of miR-130a on mitochondria remains unclear in myocardial IRI. Exploring the regulatory mechanism of miR-130a on mitochondria may provide a new target for IRI therapy. In the present study, we found that miR-130a significantly increased in acute myocardial infarction (AMI) patients and myocardial I/R rats. MiR-130a could downregulate the viability of cardiomyocytes and the knockdown of miR-130a could protect the viability of cardiomyocytes under hypoxia-reoxygenation (HR). Over-expression of miR-130a resulted in mitochondrial dysfunction. It was evidenced by decreases in mitochondrial ATP production, mitochondrial membrane potential (MMP), and an increase in reactive oxygen species (ROS) production. However, suppression of miR-130a could protect against mitochondrial damage, show elevation of mitochondrial ATP production rate and MMP, and reduce ROS production. We further explored the effect of miR-130a on the mitochondrial quality control (QMC) system by determining mitochondrial-protein-specific proteases and analyzed mitochondrial morphology by fluorescence imaging and electron microscopy, respectively. It was noted that miR-130a could suppress mitochondrial fusion and FUNDC1-mediated mitophagy to accelerate myocardial IRI. Moreover, we investigated the potential miR-130a targeted mitochondria-related genes to understand the regulatory mechanism of miR-130a in the setting of myocardial IRI. It was revealed that miR-130a targeted GJA1, and GJA1 rescued IRI by enhancing ATP production rate and oxidative phosphorylation, meanwhile protecting cell viability, MMP, and activating mitophagy. In addition, the knockdown of miR-130a significantly activated FUNDC1-mediated mitophagy, while the knockdown of GJA1 reversed the relevant response. Collectively, our findings suggest that miR-130a regulates FUNDC1-mediated mitophagy by targeting GJA1 in myocardial IRI.

Background: Oxaprozin is labeled as a Class II drug in the biopharmaceutical classification system, and its poor solubility in the entire gastrointestinal tract may be the main reason for its insufficient oral absorption capacity. Objective: The purpose of this study was to develop an oxaprozin formulation to enhance its oral absorption. Method: Oxaprozin-loaded microemulsions were prepared using the titration method and pseudoternary phase diagram. Characterization experiments were performed on microemulsion preparations, including pH, particle size, shape, zeta potential, and stability (thermodynamic, dilution, and differential scanning calorimetry). Then, the in vitro release of the microemulsion and in vivo pharmacokinetics in rats were evaluated. Results: Several microemulsion formulations were prepared. The optimal formulation was 15% oleoyl macrogolglycerides, 35% Tween 20/isopropanol (Km=2), and 50% distilled water. Its particle size met the requirements, and it had a spherical shape with a negatively charged surface. This microemulsion-loaded drug was applied to in vitro release and in vivo pharmacokinetic experiments at 7.47 mg/mL. In vitro release of the oxaprozin-loaded microemulsion best fit the firstorder model, while the microemulsion preparation had a certain sustained-release effect. In vivo pharmacokinetic experiments indicated that the microemulsion formulation significantly delayed the peak time of the blood concentration and simultaneously prolonged the half-life of drug elimination. Conclusions: The obtained data revealed satisfactory results for this novel microemulsion of oxaprozin, which is very meaningful for clinical trials.

On the basis of the higher order perturbation formulas of g-factors for a 4d1 group in tetragonally compressed octahedra, the g-factors, optical absorption, local structure and their concentration dependences of pentavalent molybdenum in 40PbO-(10-x)Y2 O3 -50P2 O5 :xMoO3 (1 ≤ x ≤ 5 mol%) glass are uniformly investigated. The experimental optical absorption spectra and g factors for Mo5+ at various concentrations x are reasonably regenerated by using the reasonable exponential concentration functions of the cubic field parameter Dq, covalent factor N and relative tetragonal compression ratio ρ. The tetragonal compression ratio ρ due to the Jahn-Teller effect was found in the range of 3.8-4.2%. The decreasing trend of Dq and N and the increasing trend of ρ with concentration can be interpreted as the fact that the variations of MoO3 and Y2 O3 concentrations lead to the modulations of local structure and electron cloud distribution around Mo5+ , associated with the adjustment of the glass network. The concentration dependence of optical basicity is also analyzed for the above glass systems.

In this work, the g factors, d-d transition band, local distortion, and their concentration dependences for impurity V4+ in 20Li2 O-20PbO-45B2 O3 -(15 - x)P2 O5 :V2 O5 (0 ≤ x ≤ 2.5 mol%) glasses are theoretically investigated by using perturbation formulas of g factors for a tetragonally compressed octahedral 3d1 cluster. In the light of the cubic polynomial concentration functions for cubic field parameter Dq , covalency factor N, and relative tetragonal compression ratio ρ, the calculated concentration dependences of d-d transition band and g factors for V4+ show good agreement with the experimental data. With increasing x, N (≈0.7682-0.8165) displays the monotonously increasing trend, whereas ρ (≈6.5-4.2%) and Dq (≈1504.9-1481.1 cm-1 ) exhibit the decreasing tendencies. The above concentration dependences can be ascribed to the modifications of the V4+ -O2- bonding and orbital admixtures around the impurity V4+ due to the effects of V2 O5 doping on the stability of the glass network, the strength of local crystal fields, and the electron cloud distribution.

Background The benefits of automated dispensing cabinets (ADCs) need to be assessed, given the different healthcare practice models. This study aimed to compare the rates of medication errors, including prescription, dispensing, and administrative, before and after using ADCs in intensive care units. Methods The prescription, dispensing, and administrative error data before and after the adoption of ADCs were retrospectively collected from the medication error report system. The severity of medication errors was classified according to the National Coordinating Council for Medication Error Reporting and Prevention guidelines. The study outcome was the rate of medication errors. A descriptive statistical analysis was performed to estimate the rates of medication errors before and after the adoption of ADCs, and Fisher’s exact test was to compare them. Results After the adoption of ADCs in the intensive care units, the rates of prescription and dispensing errors reduced from 3.03 to 1.75 per 100,000 prescriptions and 3.87 to 0 per 100,000 dispensations, respectively. The administrative error rate decreased from 0.046% to 0.026%. The ADCs decreased National Coordinating Council for Medication Error Reporting and Prevention category B and D errors by 75% and category C errors by 43%. Conclusions To improve medication safety, multidisciplinary collaboration and strategies, such as the use of automated dispensing cabinets, education, and training programs from a systems perspective, are warranted.

How terahertz signals perform in the neural system has attracted widespread interest in the life sciences community. Relevant experimental reveals that in animal nerve cells, the myelin sheath of the nerve axon has a higher refractive index than the intracellular and extracellular fluids in the Terahertz-far-infrared (THz-FIR) frequency band. This makes THz-FIR wave transmission possible in nerve fibers. Based on this premise, this article carries out the following work from the theoretical level to investigate the electromagnetic (EM) characteristics of in vivo nerve fibers at the THz-FIR band. First, the EM transmission model of the nerve fibers is established and studied theoretically. The dispersion curves of THz-FIR wave modals transmission in nerve fibers are calculated, which predict that nerve fibers can act as dielectric waveguides for transmitting THz-FIR waves and the THz-FIR waves can transmit at speeds up to 108 m/s. Second, a mode matching algorithm is proposed, which is named RNMMA, to calculate the transmission characteristics of THz-FIR waves at the nodes of Ranvier. The scattering matrix obtained from the proposed algorithm is in good agreement with the results from EM simulation software, which reveals how THz-FIR signals are transmitted forward through the nodes of Ranvier with low loss.

The development and optimization of transition metal-based bifunctional electrocatalysts for both hydrogen evolution reaction (HER) and oxygen evolution reaction (OER) are critical for high-efficient water splitting. Herein, we propose a carbon dots-oriented strategy for fabricating nitrogen-doped carbon dots/fungus-like CoP microsphere composite on Ni foam (CoP-NCDs/NF). Owing to its unique structure and the strong interactions between carbon dots and CoP, the newly developed CoP-NCDs/NF electrode exhibits significantly enhanced HER and OER bifunctional catalytic activities, with overpotentials as low as 103 and 226 mV at a current density of 10 mA cm−2, respectively. Accordingly, the CoP-NCDs/NF displays excellent overall water splitting performance with a low cell voltage of 1.55 V at 10 mA cm−2 and great stability in a two-electrode alkaline electrolyzer. This study provides an alternative pathway on rationally designing and fabricating carbon dots-based materials for high performance water electrolysis and other energy conversion fields.

Efficient transition metal oxide electrocatalysts for the alkaline hydrogen evolution reaction (HER) have received intensive attention to energy conversion but are limited by their sluggish water dissociation and unfavorable hydrogen migration and coupling. Herein, systematic density functional theory (DFT) predicts that on representative NiO, the hydroxylation (OH-) and heterointerface coupled with metallic Cu can respectively reduce the energy barrier of water dissociation and facilitate hydrogen spillover. Motivated by theoretical predictions, we subtly designed a delicate strategy to realize the electrochemical OH- modification in KOH with moderate concentration (HOM-NiO) and to channel rapid hydrogen spillover at the heterointerface of HOM-NiO and Cu, ensuring an enhanced HER kinetic. This HOM-NiO/Cu is systematically investigated by in situ XAS and electrochemical simulations, verifying its extraordinary merits for HER including the enhanced water dissociation, alleviated oxophilicity that is advantageous for consecutive adsorptions of water, and accelerated hydrogen spillover, thereby exhibiting superb HER activity with 33 and 310 mV overpotentials at the current densities of 10 and 1000 mA cm-2 in 1.0 M KOH, outperforming the Pt/C. This study might provide a reasonable strategy for the functionalized design of superior electrocatalysts.

Kidney cancer is one of the most common urological cancers worldwide, and kidney renal clear cell cancer (KIRC) is the major histologic subtype. Our previous study found that von-Hippel Lindau (VHL) gene mutation, the dominant reason for sporadic KIRC and hereditary kidney cancer-VHL syndrome, could affect VHL disease-related cancers development by inducing telomere shortening. However, the prognosis role of telomere-related genes in kidney cancer has not been well discussed. In this study, we obtained the telomere-related genes (TRGs) from TelNet. We obtained the clinical information and TRGs expression status of kidney cancer patients in The Cancer Genome Atlas (TCGA) database, The International Cancer Genome Consortium (ICGC) database, and the Clinical Proteomic Tumor Analysis Consortium (CPTAC) database. Totally 353 TRGs were differential between tumor and normal tissues in the TCGA-KIRC dataset. The total TCGA cohort was divided into discovery and validation TCGA cohorts and then using univariate cox regression, lasso regression, and multivariate cox regression method to conduct data analysis sequentially, ten TRGs (ISG15, RFC2, TRIM15, NEK6, PRKCQ, ATP1A1, ELOVL3, TUBB2B, PLCL1, NR1H3) risk model had been constructed finally. BackgroundA substantial reduction in the number of cardiac implantable electronic device (CIED) implantation was reported in the early stages of the COVID-19 pandemic. None of the studies have yet explored changes in CIED implantation during the following pandemic.ObjectiveTo explore changes in CIED implantation during the COVID-19 pandemic from 2020 to 2021.MethodsFrom 2019 to 2021, 177,263 patients undergone CIED implantation from 1,227 hospitals in China were included in the analysis. Generalized linear models measured the differences in CIED implantation in different periods. The relationship between changes in CIED implantation and COVID-19 cases was assessed by simple linear regression models.ResultsCompared with the pre-COVID-19 period, the monthly CIED implantation decreased by 17.67% (95% CI: 16.62–18.72%, p < 0.001) in 2020. In 2021, the monthly number of CIED implantation increased by 15.60% (95% CI: 14.34–16.85%, p < 0.001) compared with 2020. For every 10-fold increase in the number of COVID-19 cases, the monthly number of pacemaker implantation decreased by 429 in 2021, while it decreased by 676 in 2020. The proportion of CIED implantation in secondary medical centers increased from 52.84% in 2019 to 56.77% in 2021 (p < 0.001). For every 10-fold increase in regional accumulated COVID-19 cases, the proportion of CIED implantation in secondary centers increased by 6.43% (95% CI: 0.47–12.39%, p = 0.036).ConclusionThe impact of the COVID-19 pandemic on the number of CIED implantation is diminishing in China. Improving the ability of secondary medical centers to undertake more operations may be a critical way to relieve the strain on healthcare resources during the epidemic.

The saturable absorber of niobium diselenide (NbSe2), with a wide working bandwidth and excellent nonlinear optical response, was prepared using liquid-phase exfoliation. Its saturation intensity and modulation depth were 5.35 MW/cm2 and 6.3%, respectively. Stable mode-locking with a center wavelength of 1558.7 nm of an erbium-doped fiber laser based on a NbSe2 saturable absorber was successfully achieved. The maximum average output power of the mode-locked laser was 6.93 mW, with a pulse width of 1.3 ps and a repetition rate of 25.31 MHz at a pump power of 550 mW. The results show that NbSe2 is a promising photoelectric modulation material owing to its excellent nonlinear optical properties.

Developing of economic and efficient catalysts is critical for the application of electroreduction of carbon dioxide to highly valuable chemicals. Herein, we present a facile method to synthesize N-doped hieratically porous carbon through pyrolysis of petroleum pitch followed by ammonia etching. We found mesopores are favored formation by removing of asphaltene from petroleum pitch during the carbonation process. Simultaneously, ammonia etching can not only increase the pyridinic-N content, but also upgrade the ratio of meso- to micro- pores of carbon materials. Using the N-doped hieratically porous carbon as catalyst for carbon dioxide electroreduction, the Faradaic efficiency of carbon monoxide reaches 83% at −0.9 V vs. the reversible hydrogen electrode (RHE) in 0.1 M KHCO3. This superior performance is attributed to the synergistic effects of highly pyridinic-N content in conjunction with the hieratically porous architecture, rendering abundant exposed and accessible active sites for electroreduction of CO2. Our work provides a new strategy for the large-scale preparation of high-performance, low-cost catalysts for CO2 electroreduction.

A finite element model (FEM) was firstly developed for new enhanced C-channel connectors (ECs) in steel-concrete-steel sandwich structures (SCSSSs). This developed FEM used the continuum plasticity model (CPM) to simulate the shear fracture of C-channels and bolts used in ECs. Moreover, the FEM also considered the nonlinearities of geometry and mechanical properties of materials as well as complex interactions among different components in the ECs, e.g., channel-concrete, concrete-faceplate, and bolt-faceplate interactions. The FEM was validated by 16 push-out tests on ECs. The validations proved that the developed FEM could simulate well shear behaviours of ECs including shear-slip behaviours, failure modes, and ultimate shear resistances. With this validated FEM, a 51-case parametric study was performed to make an in-depth study on key influencing parameters on shear behaviours of ECs, which included grade and diameter of bolts, gap of bolt hole, prestressing force ratio on bolt, length of C-channel, and grade of concrete. This paper also developed analytical models to estimate the shear strength and shear-slip behaviours of ECs. The validations against test and FE simulations results confirmed the accuracies of these analytical models. Design recommendations were finally given based on these test results and FE parametric studies.

Transition metal oxides (TMOs) with high theoretical capacities are promising anode candidates for sodium ion batteries (SIBs), which yet suffers from inferior cycling stability and rate capability due to large volume change and sluggish transport kinetics during the sodiation/desodiation processes. Herein, a general strategy is developed to fabricate ultrafine TMOs nanoparticles encapsulated in nitrogen-doped carbon nanofibers (uf-TMOs@N-CNFs, M = Fe, Mn, Zn, etc.) as advanced anodes for SIBs. The uf-TMOs@N-CNFs are facilely synthesized via an in-situ electrospinning and subsequent carbonization strategy, among which polyvinylpyrrolidone (PVP) is employed as an effective dispersant to suppress metal agglomeration and control the size of TMOs. Benefiting from the intimate interaction between ultrafine TMOs and conductive N-CNFs substrates, the uf-TMOs@N-CNFs can efficiently mitigate the aggregation and pulverization of TMOs, facilitate electron/ion transfer, and boost pseudocapacitive charge storage, leading to superior sodium storage performance, including satisfactory reversible capacity, excellent rate capability, and durable cycling stability. Interestingly, the obtained uf-TMOs@N-CNFs membranes also exhibit excellent flexibility to serve as self-supported and flexible electrodes, demonstrating great potential for flexible SIBs. The present work provides a general and feasible method to construct robust and flexible electrodes for energy storage devices.

This study aimed to determine F18-Fluorodeoxyglucose Positron Emission Tomography/ Computerised Tomography (FDG-PET/CT) accuracy in nodal staging of head and neck squamous cell carcinoma (HNSCC) and investigate the relationship between increasing standard uptake value max (SUVHistopathological diagnosis was correlated retrospectively with the reported nodal involvement on PET/CT scans. Data was analysed to determine the accuracy, sensitivity, specificity, positive predictive value (PPV) and negative predictive value (NPV). This research explores the intervention effect of the mindfulness acceptance insight commitment (MAIC) training program on the mindfulness level, flow state, and mental health of college swimmers. A sample of 47 college swimmers from a regular university was recruited and randomly divided into two groups before the intervention. Independent variables between groups are psychological training mode (MAIC training/no training), and the independent variable within group was time (pre-test, post-test, and continuity test). The dependent variables are mindfulness level, flow state, and mental health (anxiety, depression, training, and competition satisfaction). Results show that after the intervention of MAIC training, the mindfulness level of athletes’ flow state has been significantly improved, whereas anxiety and depression significantly decreased. In addition, the satisfaction with training and competition significantly improved. In the continuous stage after the intervention, the mindfulness level, flow state, and mental health of athletes are still significantly higher than those in the pre-test. The comparison of the post-test and continuity test show no significant differences in the mindfulness level, flow state, depression, and training and competition satisfaction of athletes. Still, the anxiety level shows an upward trend with a significant difference. This study demonstrates that the MAIC mindfulness training program can significantly improve the mindfulness level, flow state, anxiety, depression, and training and competition satisfaction of college swimmers with a good continuity effect. Thus, the athletes’ sports experience can be improved, and good psychological benefits can be attained.

It was noted that miR-130a could obviously suppress mitochondrial fusion and FUNDC1-mediated mitophagy to accelerate myocardial IRI. Moreover, we investigated the potential miR-130a targetd mitochondria-related genes to understand the regulatory mechanism of miR-130a in the setting of myocardial IRI. It was revealed that miR-130a evidently targeted GJA1, and GJA1 rescued IRI through protecting MMP and activating mitophagy. In addition, knockdown of miR-130a significantly activated FUNDC1-mediated mitophagy, while knockdown of GJA1 reversed the relevant response. Collectively, our findings suggest that miR-130a regulates FUNDC1-mediated mitophagy by targeting GJA1 in myocardial IRI.

Carbon dioxide electroreduction (CO2RR) with renewable energy is of great significance to realize carbon neutralization. Traditional electrolysis devices usually need an ion exchange membrane to eliminate the interference of oxygen generated on the anode. Herein, the novel CuO/CeO2 composite was facilely prepared by anchoring small CuO nanoparticles on the surface of CeO2 nanocubes. In addition, CuO(002) crystal planes were induced to grow on CeO2(200), which was preferable for CO2 adsorption and C-C bond formation. As the catalyst in a membrane-free cell for CO2RR, the Cu+ was stabilized due to strong interactions between copper and ceria to resist the reduction of negative potentials and the oxidation of oxygen from the counter electrode. As a result, a high Faradaic efficiency of 62.2% toward C2 products (ethylene and ethanol) was achieved for the first time in the membrane-free conditions. This work may set off a new upsurge to drive the industrial application of CO2RR through membrane-free electrocatalysis.

The electron paramagnetic resonance (EPR) parameters and local structures for Cu 2+ in (100-2x)TeO 2 -xAg 2 O-xWO 3 (TAW) (7.5 ≤ x ≤ 30 mol %) glasses are quantitatively studied for distinct modifier concentrations x. The octahedral Cu 2+ centers are subject to the medium tetragonal elongations of about 2% along the C 4 axis due to the Jahn-Teller effect. By utilizing only three adjusted coefficients a , b and ω , the quantities ( Dq , k , t and κ) can be suitably characterized by the Fourier type functions, which reasonably account for the experimental concentration dependences of the d-d transition bands and EPR parameters. The calculation results are discussed, and the mechanisms of the above concentration dependences of these quantities are illustrated by the modifications of the local structures and the electron cloud distribution around the Cu 2+ dopant with the variations of the concentration x.
