Background: Cerebral infraction seriously affects the life quality of patients. Danshensu has been reported to exhibit anti-inflammatory and vascular protective effects. However, the therapeutic function of Danshensu in cerebral vascular injury is still unclear. Methods: Middle cerebral artery occlusion (MCAO) was used to construct the cerebral infraction model. Wound healing and tube formation assays were used to evaluate angiogenesis in vitro. Western blot assay was used to evaluate the activation of the PI3K/Akt/mTOR signaling pathway. The laser Doppler scanner was used to measure the regional cerebral blood flow (rCBF) in the area around the infarction, and the adhesion removal test was used to measure the sensorimotor function. The Modified Neurological Severity Score was performed to evaluate the cognitive functions of mice. Results: Danshensu promoted the proliferation of bEnd.3 cells and angiogenesis in vitro. Danshensu upregulated the expression of VEGF through PI3K/Akt/mTOR signaling pathway in bEnd.3 cells. Danshensu improved rCBF restoration and attenuated the behavioral deficits in mice post-MCAO/R. Conclusion: Danshensu enhances angiogenesis through the PI3K/Akt/mTOR/VEGF signaling pathway in a mouse model of cerebral ischemic injury.

Attention-deficit/hyperactivity disorder has increasingly been conceptualized as a disorder of abnormal brain connectivity. However, far less is known about the structural covariance in different subtypes of this disorder and how those differences may contribute to the symptomology of these subtypes. In this study, we used a combined volumetric-based methodology and structural covariance approach to investigate structural covariance of subcortical brain volume in attention-deficit/hyperactivity disorder-combined and attention-deficit/hyperactivity disorder-inattentive patients. In addition, a linear support vector machine was used to predict patient’s attention-deficit/hyperactivity disorder symptoms. Results showed that compared with TD children, those with attention-deficit/hyperactivity disorder-combined exhibited decreased volume of both the left and right pallidum. Moreover, we found increased right hippocampal volume in attention-deficit/hyperactivity disorder-inattentive children. Furthermore and when compared with the TD group, both attention-deficit/hyperactivity disorder-combined and attention-deficit/hyperactivity disorder-inattentive groups showed greater nonhomologous inter-regional correlations. The abnormal structural covariance network in the attention-deficit/hyperactivity disorder-combined group was located in the left amygdala–left putamen/left pallidum/right pallidum and right pallidum–left pallidum; in the attention-deficit/hyperactivity disorder-inattentive group, this difference was noted in the left hippocampus–left amygdala/left putamen/right putamen and right hippocampus–left amygdala. Additionally, different combinations of abnormalities in subcortical structural covariance were predictive of symptom severity in different attention-deficit/hyperactivity disorder subtypes. Collectively, our findings demonstrated that structural covariance provided valuable diagnostic markers for attention-deficit/hyperactivity disorder subtypes.

Phytopathogens are responsible for the wide spread of plant diseases as well as for great losses of crop yields. At present, the strategies used to control plant disease, including the development of resistant cultivars and chemical control, may become ineffective due to the adaptive evolution of pathogens.

Poaceae plants can locally accumulate iron to suppress pathogen infection. It remains unknown how pathogens overcome host-derived iron stress during their successful infections. Here, we report that Fusarium graminearum (Fg), a destructive fungal pathogen of cereal crops, is challenged by host-derived high-iron stress. Fg infection induces host alkalinization, and the pH-dependent transcription factor FgPacC undergoes a proteolytic cleavage into the functional isoform named FgPacC30 under alkaline host environment. Subsequently FgPacC30 binds to a GCCAR(R = A/G)G element at the promoters of the genes involved in iron uptake and inhibits their expression, leading to adaption of Fg to high-iron stress. Mechanistically, FgPacC30 binds to FgGcn5 protein, a catalytic subunit of Spt-Ada-Gcn5 Acetyltransferase (SAGA) complex, leading to deregulation of histone acetylation at H3K18 and H2BK11, and repression of iron uptake genes. Moreover, we identified a protein kinase FgHal4, which is highly induced by extracellular high-iron stress and protects FgPacC30 against 26S proteasome-dependent degradation by promoting FgPacC30 phosphorylation at Ser2. Collectively, this study uncovers a novel inhibitory mechanism of the SAGA complex by a transcription factor that enables a fungal pathogen to adapt to dynamic microenvironments during infection.

Stroke is one of the leading causes of death and disability worldwide. NLRP3 inflammasome has an essential role in the neuropathology of stroke. Recent studies report that shifting the microglial M1 phenotype to the M2 phenotype protects against ischemic stroke. In the present study, the precise effects of Tranilast, a NLPR3 inflammasome inhibitor, on stroke were evaluated. We established a murine model of distal middle cerebral artery occlusion (dMCAO) and administered Tranilast to dMCAO-induced stroke mice. The NLRP3 level, caspase 1 activity, and infarct volume stroke mice were measured. The sensorimotor function, pro-inflammatory cytokine production, and M1/M2 marker expression were measured. The M1 phenotype was induced by treatment of BV2 microglia with lipopolysacharide and interferon γ, and these BV-2 cells were further treated with Tranilast. The expression of CD16 and CD206 was monitored. dMCAO increased the NLRP3 expression and enhanced caspase 1 activity. Tranilast treatment significantly decreased the infarct volume, improved sensorimotor function, and suppressed the production of inflammatory cytokines in stroke mice. Moreover, Tranilast decreased the M1 marker level while promoting the expression of M2 markers. In summary, our findings suggest that Tranilast ameliorates ischemic stroke through stimulating M2 polarization of microglia.

Attention-deficit/hyperactivity disorder (ADHD) is presumed to be heterogeneous, but the best way to characterize this heterogeneity remains unclear. Although considerable evidence suggests that the 2 different types of ADHD, inattention and combined, have different cognitive and behavioral profiles, and underlying neurobiologies, we currently lack information on whether these subtypes reflect separated brain structure changes. Structural magnetic resonance imaging scans (N = 234), diagnostic, and demographic information were obtained from the ADHD-200 database. Of this sample, 138 were Typically Developing people, 37 were ADHD-Combined, and 59 were ADHD-Inattentive patients. Freesurfer segmentation methods were used to measure cortical thickness, area, and volume, subcortical volume and hipposubfield volume. ADHD-Inattentive patients showed milder clinical symptoms but more serious cognitive injury than ADHD-Combined patients. In addition, dissociable structural brain changes were found in different subtypes of ADHD, particularly in terms of decreased subcortical volume in ADHD-Combined patients compared with Typically Developing people. Clinical symptoms were predominantly related to smaller rh_caudalanteriorcingulate thickness and left-Pallidum volume, whereas verbal IQ injury was correlated strongly with smaller rh_insula area. These findings indicate that there are significant differences in clinical symptoms and gray matter damage between ADHD-Combined and -Inattentive patients. This supports the growing evidence of heterogeneity in the ADHD-Inattentive subtype and the evidence of brain structure differences.

Bacterial wilt (BW) caused by Ralstonia solanacearum (RS) is considered as one of the most destructive plant diseases. An avirulent strain of RS, FJAT1458, is a potential biocontrol agent of BW. In this study, the mechanism of FJAT1458 against BW was evaluated.FJAT1458 was tagged with the red fluorescent protein gene, and the resulting strain was named as FJAT1458-RFP. When FJAT1458-RFP and FJAT91-GFP (a virulent strain of RS labelled with the green fluorescent protein gene), were co-inoculated in potted tomato plants, the colonization of FJAT91-GFP reached an almost undetectable level at 7 days post-inoculation (dpi) in the roots and at 9 dpi in rhizosphere soil. When they were co-inoculated in a hydroponic tomato growing system, numbers of the two strains were similar at 3 dpi in the root tissues; however, FJAT91-GFP was not detected at 9 dpi while FJAT1458-RFP maintained 1.77 × 10The results suggested that the avirulent strain FJAT1458-RFP could suppress colonization of the virulent strain in tomato roots, and induce tomato plant resistance against BW. © 2022 Society of Chemical Industry.

Higher alcohols (HAs) are abundant compounds that provide important flavors in Huangjiu, but they also cause hangover. Previous studies have shown the production of HAs to be related to yeast, but the correlations between HAs and other microorganisms are rarely reported. In this study, we detected changes in levels of HAs and microbial dynamics during the Huangjiu fermentation process. Relationships were characterized using Pearson’s correlation coefficient. The functional core HA-producing bacteria were selected by bidirectional orthogonal partial least squares (O2PLS). The result showed that 2-methyl-1-propanol, phenethyl alcohol and 3-methyl-1-butanol were the principle HAs present at high levels. Lactococcus and Saccharomyces were predominant at the genus level of bacteria and fungi, respectively. A total of 684 correlations between HAs and microorganisms were established. Five genera were screened as functional core HA-producing bacteria. Our findings might provide some new inspiration for controlling the content of HAs, enhancing international prestige and market expansion of Huangjiu.

Biofilm formation is one of most important causes leading to persistent infections. Exopolysaccharides are usually a main component of biofilm matrix. Genes encoding glycosyl hydrolases are often found in gene clusters that are involved in the exopolysaccharide synthesis. It remains elusive about the functions of intracellular glycosyl hydrolase and why a polysaccharide synthesis gene cluster requires a glycosyl hydrolase-encoding gene. Here, we systematically studied the physiologically relevant role of intracellular PslG, a glycosyl hydrolase whose encoding gene is co-transcribed with 15 psl genes, which is responsible for the synthesis of exopolysaccharide PSL, a key biofilm matrix polysaccharide in opportunistic pathogen Pseudomonas aeruginosa. We showed that lack of PslG or its hydrolytic activity in this opportunistic pathogen enhances the signaling function of PSL, changes the relative level of cyclic-di-GMP within daughter cells during cell division and shapes the localization of PSL on bacterial periphery, thus results in long chains of bacterial cells, fast-forming biofilm microcolonies. Our results reveal the important roles of intracellular PslG on the cell fate and biofilm development.

Designing a carbon coated core@shell structure (M@C) is a popular and effective strategy to boost catalytic performance. However, most synthesis technologies for M@C reported up to now are relatively complicated and time consuming (multiple steps for at least a few hours). Herein, a rapid one-step process (4 min only), plasma electrolytic deposition, was developed to fabricate CoMo/CoMoOx@C with a Janus metal/oxide core in a carbon shell, which realized a significantly enhanced hydrogen evolution reaction (HER). More interestingly, this synthesis method is extendable to many other metals. The resulting material can be easily optimized by tuning the metal/oxide core and/or the carbon shell through a wide range of parameters. NiCoMo/NiCoMoOx@C was better than commercial Pt/C in HER catalysis, and NiFeMo/NiFeMoOx was among the best overall water splitting catalysts reported very recently. This work presents a fast and general technique for the design of electrocatalysts with intriguing properties.

Controlling the viscoelastic characteristics of wormlike micelles is of great significance to both basic theory and practical applications. In this article, a novel multi-stimuli responsive wormlike micellar solution was prepared by mixing cationic surfactant 3-hexadecyloxy-2-hydroxypropyltrimethylammonium bromide (R16HTAB) with trans-o-hydroxycinnamic acid (OHCA). Rheological studies, nuclear magnetic resonance (1H-NMR) spectroscopy, UV-vis spectroscopy, and cryogenic-transmission electron microscopy (cryo-TEM) were utilized to investigate the wormlike micellar system's multi-responsive activity. The results showed that the self-assembled structure and viscoelasticity of the mixed system could be regulated by pH, temperature, and light irradiation. With the increasing trans-OHCA concentration, η0 of the mixed solution increases first and then decreases and the turning point is presented at 30 mM trans-OHCA, indicating the transformation of spherical micelles to wormlike micelles, and then to short micelles. The microstructure of the mixed systems could be reversibly altered by adjusting the pH between 6.41 and 3.90, which was ascertained by cryogenic-transmission electron microscopy (cryo-TEM). The relationship of temperature and η0 obeys the Arrhenius law, attributed to the decreasing micellar contour length. η0 of a 40 mM R16HTAB/15 mM OHCA solution sharply increases after UV irradiation, mainly because the cis-isomer could insert into the micelle more easily, and the transition mechanism was studied by UV-vis and 1H NMR. The multi-responsive self-assembled system may open a new vista for building multi-functional aggregates to adapt to various environmental changes.

Building energy consumption prediction has a significant effect on energy control, design optimization, retrofit evaluation, energy price guidance, and prevention and control of COVID-19 in buildings, providing a guarantee for energy efficiency and carbon neutrality. This study reviews 116 research papers on data-driven building energy prediction from the perspective of data and machine learning algorithms and discusses feasible techniques for prediction across time scales, building levels, and energy consumption types in the context of the factors affecting data-driven building energy prediction. The review results revealed that the outdoor dry-bulb temperature is a vital factor affecting building energy consumption. In data-driven building energy consumption prediction, data preprocessing enables prediction across time scales, energy consumption feature extraction enables prediction across energy consumption types, and hyperparameter optimization enables prediction across time scales and building layers.

Perinatal hypoxic-ischemic encephalopathy (HIE) is associated with high neonatal mortality and permanent neurologic deficit. Recent data suggested microRNAs (miRNAs) may have essential functions in the regulation of HIE. This study aims to investigate the functional role of miR-499-5p and the underlying mechanism in regulating neonatal hypoxia-ischemia (HI)-induced brain injury. Dual-luciferase reporter assay and western blotting assay were performed to investigate the relationship between miR-499-5p and C-reactive protein (CRP). TUNEL staining assay was applied to evaluate neuronal cell apoptosis in the hippocampus after administration of miR-499-5p in HIE rat model. Neurobehavioral assays were conducted to evaluate the effect of miR-499-5p on the neurological functions of rat pups with HI-induced brain injury. Our study showed that miR-499-5p regulated CRP expression in L-02 cells and rat HIE model. The miR-499-5p treatment resulted in significant reduction of neuronal cell apoptosis and the infarct size of the brain. Furthermore, administration of miR-499-5p significantly improved spatial learning ability, spatial memory, and locomotor function of rat pups with HIE. Our data demonstrated that miR-499-5p have a neuroprotective effect in HI-induced brain injury in rat pups, which suggests a potential therapeutic application of miR-499-5p in the treatment of neonatal HIE.

Remote collaboration tools for conferencing and presentation are gaining significant popularity during the COVID-19 pandemic period. Most prior work has drawbacks, such as a) limited support for media types, b) lack of interactivity, for example, an efficient replay mechanism, c) large bandwidth consumption for screen sharing tools. In this paper, we propose a general-purpose multimedia collaboration platform-CWcollab. It supports collaboration on general multimedia by using simple messages to represent media controls with an object-prioritized synchronization approach. Thus, CWcollab can not only support fine-grained accurate collaboration, but also rich functionalities such as replay of these collaboration events. The evaluation shows hundreds of kilobytes can be enough to store the events in a collaboration session for accurate replays, compared with hundreds of megabytes of Google Meet.

However, the contribution of the autophagic pathway to fungal development and pathogenicity in the important maize pathogen and mycotoxigenic fungus Fusarium verticillioides is still unknown. In this study, we characterized two autophagy-related proteins, FvAtg4 and FvAtg8. The F. verticillioides deletion mutants ΔFvAtg4 and ΔFvAtg8 were impaired in autophagosome formation, aerial hyphal formation, sexual growth, lipid turnover, pigmentation and fungal virulence. Interestingly, ΔFvAtg4 and ΔFvAtg8 were defective in fumonisin B1 (FB1) synthesis, which may have resulted from decreased intracellular levels of alanine in the mutants. Our results indicate that FvAtg4 and FvAtg8 contribute to F. verticillioides pathogenicity by regulating the autophagic pathway to control lipid turnover, fumonisin biosynthesis, and pigmentation during its infectious cycle.

Diets have important but often complex implications for both environmental quality and nutrition. We establish a production-oriented life cycle model to quantify and compare the farm-to-gate environmental impacts and food nutritional qualities underlying rural and urban diets in China from 1980 to 2019, a period of rapid urbanization and socioeconomic changes. The environmental impacts of rural diets were generally higher than those of urban diets, but this gap reduced after 2000. Environmental and nutritional values varied considerably across the 31 Chinese provinces due to their different food intakes and dietary structures. Dietary changes coinciding with urbanization increased greenhouse gas emissions, eutrophication potential, and nutritional quality, but decreased energy consumption and acidification potential. Based on our results, we propose a new dietary guideline to mitigate environmental impacts and improve nutritional quality.
