During the development of intralogistics systems (ILS), heterogeneous models are created, which represent discipline-specific views, e.g., control software developed by automation engineers or discrete-event simulation models created by simulation engineers. These models represent discipline-specific views on the system but contain overlapping information. Thereby, keeping the information in different development models consistent is challenging and currently requires high manual effort, which highly depends on the developers’ experience. To overcome this challenge, an approach to link heterogeneous model data and identify potential information inconsistencies within and between models automatically is proposed. The concept is evaluated with a use case containing three typical inconsistencies from five representative engineering models applied in ILS development.

The average age of the patients was 60.3 ± 14.1 years. The underlying diseases were diabetes and biliary tract disease, accounting for 38.7% and 22.3%, respectively. The main clinical manifestations were fever (92.9%), abdominal pain (44.7%), and nausea (33.3%). Imaging findings: the proportion of patients with a single lesion was 74.7%, and 67% of the patients had involvement in the right lobe of the liver. The main pathogen was Klebsiella pneumoniae accounted for 74.9% in blood culture and 84.1% in pus culture, mainly extended-spectrum β-lactamase. In 272 strains negative for extended-spectrum β-lactamase (ESBLs), 100% were resistant to ampicillin and less than 50% were sensitive to nitrofurantoin. Only 36 ESBL-positive strains had higher than 80% sensitivity to carbapenems, β-lactamase inhibitor compound, and amikacin. Patients treated with different treatment methods showed significantly different average length of hospital stay (14 [9– 21] vs 13 [8– 18]). Empirical anti-infective therapy: Beta-lactamase complex, carbapenems, cephalosporins, and quinolones were used in 280 (37.6%), 180 (29.7%), 180 (29.7%), and 147 (24.3%) patients, respectively.Conclusion: Patients with community-acquired PLA in this area are mainly males, and the underlying diseases are mainly diabetes and hepatobiliary system disease. The main clinical manifestation is fever, so patients with fever of unknown cause should pay attention to possible liver abscesses. Based on drug sensitivity tests, the empirical use of antibiotics is somewhat unreasonable.Keywords: liver abscess, Klebsiella pneumoniae, epidemiological, community

Simultaneously and independently measuring two-dimensional (2D) displacement, one-dimensional (1D) for out-of-plane measurement and 1D for in-plane measurement, is important for practical applications. Although extrinsic fiber Fabry-Perot interferometers (EFPIs) have been reported for highly precise out-of-plane displacement measurements for many years, simultaneous measurement of 2D displacement with submicron accuracy has not been realized by a monolithic EFPI as far as we know. In this paper, a monolithic dual-cavity EFPI to realize 2D displacement measurement of a target is reported. By quantitatively analyzing and experimentally calibrating the linear dependences of the in-plane displacement sensitivity and interception on the cavity length, the effect of out-of-plane displacement on the in-plane displacement measurement is removed, which is the basis of the 2D displacement detection. 2D random movement detection and the repeatability of the system are investigated experimentally. Within the range of 10m for in-plane displacement, and 352m-358m for cavity length, demodulation errors better than 96nm are achieved. The 2D displacement measurement system based on the dual-cavity EFPI with the advantages of noncontact, high accuracy and compact size is promising to be applied in the 2D acceleration measurement, 2D rotor vibration measurement, 2D precise positioning in semiconductor manufacture, thermal expansion coefficient measurement, and so on.

Nuclear reprogramming of somatic cells into a pluripotent status has the potential to create patient-specific pluripotent stem cells (iPSCs) for regenerative medicine. Currently, however, the epigenetic mechanisms underlying this pluripotent reprogramming are poorly understood. To delineate this epigenetic regulatory network, we utilized a chromatin RNA in situ reverse transcription sequencing (CRIST-seq) approach to identify long noncoding RNAs (lncRNAs) embedded in the 3-dimensional intrachromosomal architecture of stem cell core factor genes. By combining CRIST-seq and RNA-seq, we identified Osilr9 (Oct4-Sox2 interacting lncRNA 9) as a pluripotency-associated lncRNA. Osilr9 expression was associated with the status of stem cell pluripotency in reprogramming. Using shRNA knockdown, we showed that this lncRNA was required for the optimal maintenance of stem cell pluripotency. Overexpression of Osilr9 induced robust activation of endogenous stem cell core factor genes in fibroblasts. Osilr9 participated in the formation of the intrachromosomal looping required for maintenance of pluripotency. After binding to the Oct4 promoter, Osilr9 recruited the DNA demethylase TET1, leading to promoter demethylation. These data demonstrate that Osilr9 is a critical chromatin epigenetic modulator that coordinates the promoter activity of core stem cell factor genes, highlighting the critical role of pluripotency-associated lncRNAs in stem cell pluripotency and reprogramming.

Stable and low contact resistance is the basic requirement of each connector in electrical contact. There are many factors that affect contact resistance and investigated by many previous studies. In this work, the effect of wear debris to electrical contact endurance at different relative humidity is studied by fretting test. The contact resistance evolution curve at different relative humidity indicates that the wear debris is more effective to electrical contact resistance. The composition of the wear debris is analyzed by x-ray photoelectric spectroscopy. It is clearly revealed that wear debris give a sharp rise to contact resistance. In addition, measured results illuminated that the contact resistance is greatly reduced with the wear debris washed. Higher relative humidity can restrain the generation of debris to a certain extent. Physical model of debris formation is established to evaluate ultimate lifetime of contactor.

The highly infectious and serious nature of coronavirus disease 2019 (COVID-19) has highlighted the need for hospital space disinfection technology and the prevention of human exposure to pathogenic environments. This research developed novel chlorine dioxide (ClO2) sterilization technology to reduce bacteria and viruses in the air and on surfaces. A smart sterilization robot system was also developed to spray disinfectants in operating theaters or patients’ rooms, designed according to the results of controlled experiments and the requirements for hospital disinfection. The system was built incorporated a semi-automatic remote-controlled module and an automatic intelligent disinfection function; that is, it could operate independently according to specific epidemic prevention strategies, which were implemented using a combination of Internet of Things (IoT) applications and a gesture recognition function. The elimination of Escherichia coli ( E. coli ) bacteria on sample plates was 99.8 % effective. This paper reviews the evolution of various disinfection technologies and describes a disinfection robot system in detail.

Pine wilt disease is the most devastating pine disease caused by Bursaphelenchus xylophilus. Bursaphelenchus mucronatus is morphologically similar to B. xylophilus and geographically overlaps in its distribution. Although interspecific hybridization of the two nematodes has been performed in vitro, the dynamic regularity of hybrid formation and its risk in forests has not been well evaluated. In this study, a hybrid of B. xylophilus and Bursaphelenchus mucronatus mucronatus was identified in the laboratory and fields by molecular markers. The heterozygosity of ITS-5.8S loci for identification was unstable in the hybrid population, and the allele inherited from B. m. mucronatus was lost over several generations. We also provided evidence that hybrids existed in some new epidemic areas, while old epidemic areas were usually dominated by B. xylophilus. Hybrids could be generated when B. m. mucronatus was invaded by B. xylophilus, and the pathogenicity of the hybrids was similar to that of B. xylophilus. These findings may improve the understanding of the natural hybridization between B. xylophilus and B. m. mucronatus and pathogenic variation in pine wilt disease, providing new insights for future studies on disease detection, transmission, and quarantine.

The reduction of temperature effect is vitally important for high precision sensors. This paper introduces the theoretical analysis and experimental verification of an in-situ compensation method on the temperature coefficient of the scale factor for a single-axis electromagnetic force-balance feedback MEMS accelerometer. A micro thermistor, fabricated together with the MEMS accelerometer, is integrated to in-situ sense the temperature variation of the accelerometer. The thermistor is connected to the voltage-to-current resistance in series, which compensates the variation of the scale factor of the accelerometer that is influenced by temperature variation. The experimental results reveal that the temperature coefficient of the scale factor of the compensated accelerometer is within 6 ppm/°C for the temperature range from 25°C to 50°C, which is reduced by 68 times compared with the uncompensated accelerometer. Benefiting from the compatible fabrication process and the in-situ compensation, the proposed method is promising to be low-cost and high-efficiency.

Protein translation is a critical regulatory event involved in nearly all physiological and pathological processes. Eukaryotic translation initiation factors are dedicated to translation initiation, the most highly regulated stage of protein synthesis. Eukaryotic translation initiation factor 4G2 (eIF4G2, also called p97, NAT1 and DAP5), an eIF4G family member that lacks the binding sites for 5' cap binding protein eIF4E, is widely considered to be a key factor for internal ribosome entry sites (IRESs)-mediated cap-independent translation. However, recent findings demonstrate that eIF4G2 also supports many other translation initiation pathways. In this review, we summarize the role of eIF4G2 in a variety of cap-independent and -dependent translation initiation events. Additionally, we also update recent findings regarding the role of eIF4G2 in apoptosis, cell survival, cell differentiation and embryonic development. These studies reveal an emerging new picture of how eIF4G2 utilizes diverse translational mechanisms to regulate gene expression.

Genomic instability is an important characteristic of cancer, which promotes clonal evolution and tumorigenesis by increasing the frequency of gene destruction and loss of genome integrity. Generally, the maintenance of genomic stability depends significantly on the accurate regulation and timely repair of different genomic scales, ranging from DNA sequence to chromatin higher-order structures to chromosomes. Once irreversible damage and imperfect repair occurred, the resulting genomic instability can lead to a higher risk of tumorigenesis. However, how these factors disrupt genomic stability and their specific tumorigenic mechanisms remain unclear. Inspiringly, numerous studies have confirmed that long noncoding RNAs (lncRNAs), an important regulator of epigenetic inheritance, are functional in such process. Thus, this review aimed to discuss the vital factors that may lead to genomic instability at these multiple genomic scales, with an emphasis on the role of lncRNAs in it.

This work presents a novel gated- diode SCR-incorporated BJT (SIB) structure for ESD protection purpose. A gated diode built in the SIB structure modulates reverse-biased P+ / N-well diode for better junction breakdown control. This device could achieve high TLP second breakdown current /ESD threshold close to that of an SCR, very high holding voltage, and reduced trigger voltage, as well as excellent latchup immunity.

Recurrent spontaneous abortion (RSA) is a highly heterogeneous complication of pregnancy with the underlying mechanisms remaining uncharacterized. Dysregulated decidualization is a critical contributor to the phenotypic alterations related to pregnancy complications. To understand the molecular factors underlying RSA, we explored the role of longnoncoding RNAs (lncRNAs) in the decidual microenvironment where the crosstalk at the fetal-maternal interface occurs. Natural killer (NK) cells perform immune surveillance functions in tumors. The antitumor effects of NK cells are closely related to tumor occurrence and development. However, the molecular factors that determine NK cell antitumor activity remain to be characterized. In the present study, we identified a novel long noncoding RNA (lncRNA), NK cell activity-associated lncRNA 1 (NCAL1), and investigated its function in NK cells. NCAL1 was primarily located in NK cell nuclei, where it functioned by activating Gab2, a scaffold protein with an essential role in immune cells. Gab2 positively regulated the killing activity of NK cells. Mechanistically, NCAL1 upregulated Gab2 epigenetically by binding to the Gab2 promoter, which decreased methylation, recruited the transcription factor Sp1, and increased H3K4me3 and H3K27ac levels in the Gab2 promoter. Furthermore, NCAL1 enhanced the cytotoxicity of NK cells toward tumor cells through the Gab2-PI3K-AKT pathway. Thus, NCAL1 potentiates NK cell cytotoxicity and is a promising therapeutic target to improve NK cell therapy.

Nano- $g$ accelerometers are widely used in space exploration and measurement of the earth’s gravitational field. It is essential to precisely evaluate error effects at high orders such as cross-coupling for applications in a dynamic environment. Nevertheless, it remains challenging to meet the precision requirements using conventional calibration measures. In this article, we propose a method to separate the cross-coupling coefficients of a linear single-axis accelerometer by mounting it on a steadily rotating rate table that is tilted at a fixed deviation angle with respect to the horizontal plane. The gravity component is periodically modulated along the input axis per revolution. Simultaneously, a series of centripetal acceleration is applied along the cross axis in sequence while adjusting the rotation frequency of the rate table by steps. Thus, the cross-coupling coefficient can be separated by its dependence both on the modulated gravity acceleration and the centripetal acceleration. In comparison to the static multipoint angular rotation test on a tilted dividing head, the proposed dynamic modulation method demonstrates improved robustness against corruption from bias drift, with an improved uncertainty. This method to separate the cross-coupling coefficient is suitable for testing high-resolution accelerometers, without requiring high bias stability or sensitive response sustaining at ultralow frequency.

The effect of tempering process on the microstructure and properties of low alloyed cast steel was studied. The results show that tempered at (200~400)°C, the M/A island of the granular bainite decomposes and carbide precipitates. When the tempering temperature rises to (500~600)°C, the M/A island is completely decomposed, and the carbide aggregates and gradually spheroidizes. With the increasing of tempering temperature, the tensile strength increases first and then decreases, elongation and impact energy show a trend of increasing at first, then decreasing and then increasing. The tempering brittleness occurs at 400°C.

High-precision microelectromechanical inertial sensors based on spring-mass structures are of great interests for a wide range of applications, including inertial navigation, disaster warning and resource exploration. Lowering the resonant frequency is essential to further improve the sensitivity of the sensors. However, conventional approaches are facing insurmountable difficulties from size reduction to machining precision. This paper proposed a novel quasi-zero-stiffness mechanism that is compatible with MEMS technologies together with a micromaching approach for adjusting the stiffness precisely. By improving the compliance of a typical spring with a negative-stiffness compensation mechanism induced by axial force, the resonant frequency of the micro spring-mass structure is lowered to 0.7 Hz, which is at least 3 times lower than current state-of-the-art micro structures. Based on this ultra-sensitive micro structure base on the quasi-zero-stiffness mechanism, the micro inertial sensor, with a chip size of a postage stamp, has shown a low self-noise of 0.6 nrad/ $\surd $ Hz at 0.04 Hz and a high long-term stability that are comparable to traditional pendulum inertial sensors. It is the first micro device, to our knowledge, that can successfully measure the tidal tilt signal. [2020-0048]

Acral lentiginous melanoma (ALM) is one of the four major subtypes in cutaneous malignant melanoma (CMM). We aimed to study the incidence and survival of ALM in the United States in recent 10 y and compare the survival between ALM and nonacral CMM. In the meantime, racial disparity and prognostic factors associated with survival were investigated.All the cases of ALM registered in the Surveillance, Epidemiology, and End Results registry from 2006 to 2015 were retrieved, including non-Hispanic whites (NHW), black Americans (blacks), Asian/Pacific Islanders, and Hispanic whites. Age-adjusted incidence was calculated. Clinicopathologic data, including age, gender, race, geographic location, Breslow thickness, ulceration, pathologic staging, sentinel lymph node status, and surgical approach, were collected and analyzed. Melanoma-specific survival (MSS) was analyzed in patients with ALM and CMM. The Kaplan-Meier method was used to estimate the influence of clinicopathologic data on ALM MSS. Only cases with complete staging and active follow-up were included in prognostic factor analysis.A total of 1724 ALM and 87,442 nonacral CMM patients were included in the study. For ALM patients, the age-adjusted incidence rate was 2.0 per million person-years. The proportion of ALM among all melanoma subtypes was greatest in blacks (32.6%). The 5-y MSS rates of ALM were lower than CMM overall (80.6% versus 93.0%, P 0.001, respectively). When controlled by stage, the difference was significant in patients diagnosed at stages I and III. ALM 5-y MSS rates were highest (84.3%) in NHW, intermediate in Asian/Pacific Islanders (76.6%), Hispanic white (72.0%), and lowest in blacks (66.9%). Blacks were elderly, male predominant, located in East and Middle American, and had thicker, more ulcerated, advanced disease as compared with NHWs. When controlled by stage, survival difference was significant between NHWs and blacks in stage I (P = 0.004) and stage III (P = 0.005) patients. Gender, race, sentinel lymph node status, and pathologic stage were identified as independent risk factors in multivariate analysis.The incidence of ALM has been steady in recent 10 y and more prevalent in aged people. ALM is associated with a worse prognosis than CMM. Black Americans have the worst prognosis, and survival difference is significant between NHW and blacks.

Photochromic materials have received extensive attention from scientific researchers due to their novel uses. Though the related mechanism research is getting deeper, the current material preparations are complex and expensive. In this paper, we propose prepare a linen/polyethyleneimine/sodium decadecanate photochromic fabric via layer-by-layer self-assembly method. By building an electron transfer photochromic system on linen fabrics, the fabricated fabric not only presents outstanding reversible color-changing properties (quick color-changes speed, apparently color difference) under UV light (365 nm), but also shows excellent delicate patterns and color preservation ability in visible light environment, which endow it with great potential for application in intelligent devices and UV light detectors. Additionally, the advantageous features of good fatigue resistance (more than 15 times), light weight, comfortable feeling, low-cost and non-toxic, allow this material to be a good candidate in many applied fields.

Photochromic materials have drawn growing interesting due to their capability of interconversion under the action of external optical stimulation, allowing diverse potential applications. Herein, we proposed a novel method of preparing photochromic cotton fabric coated by combined l-(2-hydroxyethyl)-3,3-dimethyl-6-nitrospiro [l(2H)-benzopyran-2,2-indoline] terminated waterborne polyurethane. The synthesized photochromic compounds 1-(2-hydroxyethyl)-3,3-dimethyl-6-nitrospiro[1(2H)-benzopyran-2,2-indoline] (SP-OH), were characterized by FTIR spectra, 1H NMR and ultraviolet (UV) spectrum. The as-prepared spiropyran-terminated water polyurethane (SP-OH/WPU) was characterized via FTIR spectra, ultraviolet (UV) spectrum, SEM and size analysis. The photochromic cotton fabric was featured via FTIR spectra, ultraviolet (UV) spectrum and SEM. In addition, the photochromic properties, thermal stability of as-prepared cotton fabric was tested. The results showed that the photochromic compounds and SP-OH/WPU have been successfully synthesized and the resultant cotton fabric possesses excellent and durable photochromic properties, including noticeable and quick color alteration under UV radiation and fast recovery when exposed to green light. Furthermore, the excellent thermal stability ensured it undergo harsh service environment, which indicated that it is a good candidate for photochromic textile.

Purpose Nuclear factor (erythroid-derived 2)-like 2, also known as NFE2L2 or NRF2, a transcription factor capable of upregulating antioxidant response element (ARE)-mediated expression and cytoprotective proteins, plays critical roles in chemoprevention, inflammation and aging. NRF2 has recently been proposed as a novel target for cancer chemoprevention. The fungicide miconazole has shown promising antiproliferative effects in cancer cells. Materials and methods After miconazole treatment, the p62-KEAP1-NRF2 activation was analyzed by qPCR and Western blot. The nuclear translocation indicating NRF2 activation was further confirmed by immunofluorescence. Finally, the ROS production was detected by CM-H2DCFDA staining. Results We demonstrate in this study that miconazole dramatically increases NRF2 activation in bladder cancer cells, in a dose- and time-dependent manner. Interestingly, levels of expression of p62, a noncanonical pathway that mediates NRF2 activation, appeared to increase in accordance with NRF2. We also investigated levels of the negative regulator kelch-like ECH-associated protein 1 (KEAP1), which is involved in NRF2 activation. As expected, a decrease in KEAP1 expression was found after miconazole exposure. Confirmation of NRF2 nuclear translocation was monitored by immunofluorescence. Miconazole-induced generation of reactive oxygen species (ROS) promoted NRF2 activation. Pretreatment of bladder cancer cells with ROS scavengers abolished NRF2 expression and nuclear translocation, indicating that miconazole activates the noncanonical p62-KEAP1-NRF2 pathway, which is regulated by ROS production. Conclusion Our study elucidates the mechanisms through which miconazole stimulates NRF2 which may contribute to cancer chemopreventive effects.

Background: Patients who received warfarin require constant monitoring by hospital staff. However, social distancing and stay-at-home orders, which were universally adopted strategies to avoid the spread of COVID-19, led to unprecedented challenges. This study aimed to optimize warfarin treatment during the COVID-19 pandemic by determining the role of the Internet clinic and developing a machine learning (ML) model to predict anticoagulation quality.Methods: This retrospective study enrolled patients who received warfarin treatment in the hospital anticoagulation clinic (HAC) and “Internet + Anticoagulation clinic” (IAC) of the Nanjing Drum Tower Hospital between January 2020 and September 2021. The primary outcome was the anticoagulation quality of patients, which was evaluated by both the time in therapeutic range (TTR) and international normalized ratio (INR) variability. Anticoagulation quality and incidence of adverse events were compared between HAC and IAC. Furthermore, five ML algorithms were used to develop the anticoagulation quality prediction model, and the SHAP method was introduced to rank the feature importance.Results: Totally, 241 patients were included, comprising 145 patients in the HAC group and 96 patients in the IAC group. In the HAC group and IAC group, 73.1 and 69.8% (p = 0.576) of patients achieved good anticoagulation quality, with the average TTR being 79.9 ± 20.0% and 80.6 ± 21.1%, respectively. There was no significant difference in the incidence of adverse events between the two groups. Evaluating the five ML models using the test set, the accuracy of the XGBoost model was 0.767, and the area under the receiver operating characteristic curve was 0.808, which showed the best performance. The results of the SHAP method revealed that age, education, hypertension, aspirin, and amiodarone were the top five important features associated with poor anticoagulation quality.Conclusion: The IAC contributed to a novel management method for patients who received warfarin during the COVID-19 pandemic, as effective as HAC and with a low risk of virus transmission. The XGBoost model could accurately select patients at a high risk of poor anticoagulation quality, who could benefit from active intervention.

Numerous long noncoding RNAs (lncRNAs) are abnormally expressed in breast cancer (BC), but the underlying mechanisms remain large unknown. Here, we aimed to investigate the functions and mechanisms of lncRNA cancer susceptibility candidate 9 (CASC9) in BC. Western blotting and quantitative real-time PCR (qRT-PCR) were performed to assess gene and protein expression, respectively. The proliferative and metastatic abilities of BC cells were tested by cell counting kit-8 and transwell assays, respectively. The formation of lymphatic vessels was detected by tube formation assay. Chromatin immunoprecipitation (ChIP) and dual luciferase reporter assays were performed to verify molecular interactions. CASC9 was found to be highly expressed in BC tissues and cell lines, and ectopic overexpression was positively associated with tumor volume, TNM stage, and lymph node metastasis. In addition, CASC9 silencing significantly inhibited the proliferation and invasion of BC cells, as well as BC-associated invasion and formation of lymphatic vessels of human dermal lymphatic endothelial cells. Mechanical studies demonstrated that CASC9 could be transcriptionally activated by STAT3 and elevate SOX4 expression by enhancing the acetylation of its promoter region. Our results illustrated that STAT3-activated CASC9 served as a tumor-promoting gene involved in promoting BC invasion and BC-associated formation of lymphatic vessels by upregulating SOX4 through altering H3K27ac level. This finding elucidated a new underlying network of CASC9 in the metastasis of BC.

Despite the growing impact of smartphone use on countries’ economies, the literature has rarely investigated the link between economic context and smartphone purchase trends. Based on 20,556 smartphones sold from a Cross-Border E-Commerce (CBEC) platform, the study reveals that relationships between GDP per capita and Smartphone Choice Preferences (SCP) as well as Purchase Quantities (PUR) are direct and partially mediated by Price (PRI), Read-Only Memory (ROM), and Random-Access Memory (RAM). That means that the economic context highlighted by the GDP plays a substantial role in smartphone choices and purchases. The study suggests that e-sellers and smartphone brands should adapt their marketing and manufacturing strategies to the countries’ economic contexts to leverage the fearless competition in the smartphone industry.

With the expansion of unmanned aerial vehicle(UAV) applications and increased power, the status of unmanned aerial vehicles in the military field has continued to improve. How to effectively defend against UAV attacks has become a key research issue in various countries. The damage assessment model of the AHEAD in the literature [17] does not consider the systematical error of anti-aircraft artillery, and ignores the dispersion error of the AHEAD explosion-points in the direction of the vertical interception plane, which leads to the inaccuracy of the calculated damage probability. In this paper, the model is modified, the systematical error and the dispersion error of the opening point in the direction of the vertical interception plane are added to the original model, and then the improved model is used to analyze the damage effectiveness of the AHEAD against two typical LSS UAVs. It shows that the AHEAD has good damage efficiency to LSS UAVs. To a certain extent, it has the ability to counter the LSS UAV clusters.

The molecular circuitry that causes stem cells to exit from pluripotency remains largely uncharacterized. Using chromatin RNA in situ reverse transcription sequencing, we identified Peln1 as a novel chromatin RNA component in the promoter complex of Oct4, a stem cell master transcription factor gene. Peln1 was negatively associated with pluripotent status during somatic reprogramming. Peln1 overexpression caused E14 cells to exit from pluripotency, while Peln1 downregulation induced robust reprogramming. Mechanistically, we discovered that Peln1 interacted with the Oct4 promoter and recruited the DNA methyltransferase DNMT3A. By de novo altering the epigenotype in the Oct4 promoter, Peln1 dismantled the intrachromosomal loop that is required for the maintenance of pluripotency. Using RNA reverse transcription-associated trap sequencing, we showed that Peln1 targets multiple pathway genes that are associated with stem cell self-renewal. These findings demonstrate that Peln1 can act as a new epigenetic player and use a trans mechanism to induce an exit from the pluripotent state in stem cells.

Susceptibility to the rubber hand illusion (RHI) varies. To date, however, there is no consensus explanation of this variability. Previous studies, focused on the role of multisensory integration, have searched for neural correlates of the illusion. But those studies have failed to identify a sufficient set of functionally specific neural correlates. Because some evidence suggests that frontal α power is one means of tracking neural instantiations of self, we hypothesized that the higher the frontal α power during the eyes-closed resting state, the more stable the self. As a corollary, we infer that the more stable the self, the less susceptible are participants to a blurring of boundaries—to feeling that the rubber hand belongs to them. Indeed, we found that frontal α amplitude oscillations negatively correlate with susceptibility. Moreover, since lower frequencies often modulate higher frequencies, we explored the possibility that this might be the case for the RHI. Indeed, some evidence suggests that high frontal α power observed in low-RHI participants is modulated by δ frequency oscillations. We conclude that while neural correlates of multisensory integration might be necessary for the RHI, sufficient explanation involves variable intrinsic neural activity that modulates how the brain responds to incompatible sensory stimuli.

Miscarriage, the spontaneous loss of a pregnancy before the fetus achieves viability, is a common complication of pregnancy. Decidualization plays a critical role in the implantation of the embryo. To search for molecular factors underlying miscarriage, we explored the role of long noncoding RNAs (lncRNAs) in the decidual microenvironment, where the molecular crosstalk at the feto–maternal interface occurs. By integrating RNA-seq data from recurrent miscarriage patients and decidualized endometrial stromal cells, we identified H19, a lncRNA that exhibits paternally imprinted monoallelic expression in normal tissues, as the most upregulated lncRNA associated with miscarriage. Aberrant upregulation of H19 lncRNA was observed in decidual tissues derived from patients with spontaneous miscarriage as well as decidualized endometrial stromal cells. The maternally imprinted fetal mitogen Igf2, which is usually reciprocally co-regulated with H19 in the same imprinting cluster, was also upregulated. Notably, both genes underwent loss of imprinting, as H19 and IGF2 were actively transcribed from both parental alleles in decidual tissues. Mechanistically, this loss of imprinting in decidual tissues was associated with the loss of the H3K27m3 suppression marker in the IGF2 promoter, CpG hypomethylation at the central CTCF binding site in the imprinting control center (ICR) that is located between IGF2 and H19, and the loss of CTCF-mediated intrachromosomal looping. These data provide the first evidence that aberrant control of the ICR epigenotype-intrachromosomal looping-H19/IGF2 imprinting pathway may be a critical epigenetic risk factor in the abnormal decidualization related to miscarriage. In electrical discharge machining (EDM), gap control is the key to stable processing; the discharge gap plays a significant role in EDM. To determine the influence of the discharge gap on material removal and melt pool movement, which are two fundamental issues in EDM, high-speed photography and molecular dynamics (MD) simulations were used to study the discharge process. Research results demonstrate that the discharge gap has a significant influence on material removal during the discharge process. A smaller gap width produces more and larger removed materials. The influence mechanism of the gap width on material removal is explained as follows. A smaller gap width produces discharge plasma with a smaller diameter and greater heat flux. Discharge with a greater heat flux generates more material removed during the discharge process. In addition, a smaller gap width and greater heat flux produce a stronger interaction of metal vapor jets, generating a stronger shear force acting on the melt pool. The discharge gap also influences the movement of the melt pool and the final topography of the discharge crater through external pressure acting on the melt pool. Smaller gap width produces greater external pressure acting on the melt pool, generating a bowl-shaped melt pool and a discharge crater with a depression in the center and a bulge around the edge. A larger gap width produces less external pressure acting on the melt pool, generating a flat melt pool and a discharge crater with swelling in the center and a depression around the edge.

Mitophagy is a specialized autophagic pathway responsible for the selective removal of damaged or dysfunctional mitochondria by targeting them to the autophagosome in order to maintain mitochondria quality. The role of mitophagy in tumorigenesis has been conflicting, with the process both supporting tumor cell survival and promoting cell death. Cancer cells may utilize the mitophagy pathway to augment their metabolic requirements and resistance to cell death, thereby leading to increased cell proliferation and invasiveness. This review highlights major regulatory pathways of mitophagy involved in cancer. In particular, we summarize recent progress regarding how nuclear-encoded long non-coding RNAs (lncRNAs) function as novel epigenetic players in the mitochondria of cancer cells, affecting the malignant behavior of tumors by regulating mitophagy. Finally, we discuss the potential application of regulating mitophagy as a new target for cancer therapy.

A new methane formation pathway, which uses methylphosphonate (MPn) as the methane precursor, has been discovered in the upper ocean. Methylphosphonate synthase (MPnS) is a key piece in this pathway to produce MPn from 2-hydroxyethylphosphonate (2-HEP), using an untypical 2-His-1-Gln non-heme iron architecture. Herein, the MPnS reaction mechanism was demonstrated by the density functional calculations to mainly include the substrate hydroxyl deprotonation, the formation of a MPn radical and a formate, and the hydrogen abstraction of formate by MPn radical. The second-shell Lys28' may serve as a proton reservoir activating 2-HEP and regenerating the Fe site. The Fe-bound superoxide radical is a bifunctional species to deprotonate the substrate hydroxyl and abstract the substrate methylene hydrogen. Several alternative mechanisms have been ruled out. Furthermore, the catalytic activity of MPnS was found to be inactivated/reduced by the mutation of Gln152E/Gln152H/Gln152D, rendering a significant evolutionary advantage with an uncommon 2-His-1-Gln triad introduced to the ferrous coordination sphere.

Background: Long non-coding RNAs (lncRNAs) constitute an important component of the regulatory apparatus that controls stem cell pluripotency. However, the specific mechanisms utilized by these lncRNAs in the control of pluripotency are not fully characterized. Methods: We utilized a RNA reverse transcription-associated trap sequencing (RAT-seq) approach to profile the mouse genome-wide interaction targets for lncRNAs that are screened by RNA-seq. Results: We identified Peblr20 (Pou5F1 enhancer binding lncRNA 20) as a novel lncRNA that is associated with stem cell reprogramming. Peblr20 was differentially transcribed in fibroblasts compared to induced pluripotent stem cells (iPSCs). Notably, we found that Peblr20 utilized a trans mechanism to interact with the regulatory elements of multiple stemness genes. Using gain- and loss-of-function experiments, we showed that knockdown of Peblr20 caused iPSCs to exit from pluripotency, while overexpression of Peblr20 activated endogenous Pou5F1 expression. We further showed that Peblr20 promoted pluripotent reprogramming. Mechanistically, we demonstrated that Peblr20 activated endogenous Pou5F1 by binding to the Pou5F1 enhancer in trans, recruiting TET2 demethylase and activating the enhancer-transcribed RNAs. Conclusions: Our data reveal a novel epigenetic mechanism by which a lncRNA controls the fate of stem cells by trans-regulating the Pou5F1 enhancer RNA pathway. We demonstrate the potential for leveraging lncRNA biology to enhance the generation of stem cells for regenerative medicine.

In this paper, we proposed to prepare a spiropyran (SP)-based photochromic cotton fabric with high tri-stimulus response by thiol-ene click chemistry, which has excellent photochromic properties, good durability, and can quickly return to its original state under three different stimulations. The SP target monomer with ene group in the side chain was synthesized, and the cotton fabric was subjected to thiol modification by using 3-mercaptopropyltriethoxysilane. The SP molecule was grafted to the modified cotton fabric by covalent bonding, therefore effectively enhanced its durability in practical daily use. The results of FTIR, NMR, Raman and UV spectra confirmed the chemical composition. SEM images and energy-dispersive X-ray spectroscopy mapping spectra verified the grafting between this SP-based dye and modified cotton fabrics. The test data of color characteristic values indicated that the fabric undergoes significant color changes with fast photochromic response, high fatigue resistance, and maintains impressive reusability after experiencing 20 reversible cycles. The photochromic mechanism of cotton fabrics was attributed to the cleavage of C–O bond in the molecular structure of SP under ultraviolet (UV) irradiation. In addition, the properties related to practical applications including washing fastness and UV resistance have also been studied, proving its great potential in wearable and flexible textile-based sensors.

Long noncoding RNAs (lncRNAs) can regulate the activity of target genes by participating in the organization of chromatin architecture. We have devised a “chromatin-RNA in situ reverse transcription sequencing” (CRIST-seq) approach to profile the lncRNA interaction network in gene regulatory elements by combining the simplicity of RNA biotin labeling with the specificity of the CRISPR/Cas9 system. Using gene-specific gRNAs, we describe a pluripotency-specific lncRNA interacting network in the promoters of Sox2 and Pou5f1, two critical stem cell factors that are required for the maintenance of pluripotency. The promoter-interacting lncRNAs were specifically activated during reprogramming into pluripotency. Knockdown of these lncRNAs caused the stem cells to exit from pluripotency. In contrast, overexpression of the pluripotency-associated lncRNA activated the promoters of core stem cell factor genes and enhanced fibroblast reprogramming into pluripotency. These CRIST-seq data suggest that the Sox2 and Pou5f1 promoters are organized within a unique lncRNA interaction network that determines the fate of pluripotency during reprogramming. This CRIST approach may be broadly used to map lncRNA interaction networks at target loci across the genome.
